Cell division and cell fate decisions are highly regulated processes that need to be coordinated both spatially and temporally for correct plant growth and development. Gaining a deeper molecular and cellular understanding of these links is especially relevant for plant biology since, unlike in animals, formation of new organs is a process that takes place after embryogenesis and continues throughout the entire plant lifespan. The recent identification of a novel factor, GEM, has provided a molecular framework that coordinates cell division to cell fate in the Arabidopsis epidermis. GEM is an inhibitor of cell division through interacting with CDT1, a DNA replication protein. It also inhibits the expression of the homeobox GLABRA2 (GL2) gene that determines the hair/non-hair fate and the pavement/trichome fate in the root and leaf epidermis, respectively. GEM seems to be crucial in controlling the balance of activating/repressing histone modifications at its target promoters.Key Words: cell division, cell cycle, cell fate, GEM, GLABRA2, CDT1, DNA replication, chromatin, histone methylation, gene expression, root hair, Arabidopsis, plant  相似文献

研究发现植物根毛发育有反馈侧向抑制(lateral inhibition with feedback)和位置决定模式(position-dependent pattern of cell differentiation)两种方式。

The specification of distinct cell fates in multicellular organisms is a fundamental process in developmental biology. The Arabidopsis root epidermis, which consists of root-hair cells and non-hair cells, provides a useful model system for studying cell fate specification. In this tissue, the cell fates are determined by their relative position to the underlying cortical cells, and many genes have been identified that regulate this position-dependent cell fate specification. Recent studies using genetic, molecular, and biochemical approaches have shed new light on this process and revealed a complex network of interacting and interdependent components. In particular, a novel regulatory circuit has recently been identified, which includes a lateral inhibition pathway and a feedback loop that enables intercellular communication and ensures that two distinct cell types arise in an appropriate pattern. This regulatory circuit is also influenced by a positional signaling pathway which includes the SCRAMBLED leucine-rich repeat receptor kinase. The studies of cell fate specification in the Arabidopsis root epidermis provide new insights into the molecular strategies used to define distinct cell types in plants.

Root hairs are tubular outgrowths specifically differentiated from epidermal cells in a differentiation zone. The formation of root hairs greatly increases the surface area of a root and maximizes its ability to absorb water and inorganic nutrients essential for plant growth and development. Root hair development is strictly regulated by intracellular and intercellular signal communications. Cell surface-localized receptor-like protein kinases(RLKs) have been shown to be important components in these cellular processes. In this review, the functions of a number of key RLKs in regulating Arabidopsis root hair development are discussed, especially those involved in root epidermal cell fate determination and root hair tip growth.

The syncytium is a unique plant root organ whose differentiation is induced by plant-parasitic cyst nematodes to create a source of nourishment. Syncytium formation involves the redifferentiation and fusion of hundreds of root cells. The underlying regulatory networks that control this unique change of plant cell fate are not understood. Here, we report that a strong down-regulation of Arabidopsis (Arabidopsis thaliana) microRNA396 (miR396) in cells giving rise to the syncytium coincides with the initiation of the syncytial induction/formation phase and that specific miR396 up-regulation in the developed syncytium marks the beginning of the maintenance phase, when no new cells are incorporated into the syncytium. In addition, our results show that miR396 in fact has a role in the transition from one phase to the other. Expression modulations of miR396 and its Growth-Regulating Factor (GRF) target genes resulted in reduced syncytium size and arrested nematode development. Furthermore, genome-wide expression profiling revealed that the miR396-GRF regulatory system can alter the expression of 44% of the more than 7,000 genes reported to change expression in the Arabidopsis syncytium. Thus, miR396 represents a key regulator for the reprogramming of root cells. As such, this regulatory unit represents a powerful molecular target for the parasitic animal to modulate plant cells and force them into novel developmental pathways.

Pattern formation and morphogenesis require coordination of cell division rates and orientations with developmental signals that specify cell fate. A viable mutation in the TILTED1 locus, which encodes the catalytic subunit of DNA polymerase epsilon of Arabidopsis thaliana, causes a lengthening of the cell cycle by approximately 35% throughout embryo development and alters cell type patterning of the hypophyseal lineage in the root, leading to a displacement of the root pole from its normal position on top of the suspensor. Treatment of preglobular and early globular stages, but not later stage, embryos with the DNA polymerase inhibitor aphidicolin leads to a similar phenotype. The results uncover an interaction between the cell cycle and the processes that determine cell fate during plant embryogenesis.
