本研究表明 ,旱育秧床中土壤好气微生物总量高于对照 ,其中好气性固氮菌、纤维分解菌、氨化细菌、有机磷分解菌、亚硝酸细菌增加了 1 .40、2 0 .67、8.0 4、74.0 0、0 .69倍 ,除脱氢酶外土壤蛋白酶、纤维素酶、磷酸酶、尿酶和蔗糖转化酶活性比对照增加77.5%、51 .1 5%、47.1 2 %、43.59%、6.88% ,这有利于土壤物能转化 ,提高土壤养分的有效性 ,使旱育秧床土壤硝态氮、速效N、P、K比对照增加 2 7.9、2 .0 8、0 .97、0 .7倍 .同时还表明 ,旱育秧白根率高 ,根系氧化能力强 ,低节位分蘖多 ,并表现出强大的生理生化优势 ,淀粉酶、硝酸还原酶、ATP酶以及过氧化物酶、过氧化氢酶、超氧物歧化酶等保护酶含量增加 ,而丙二醛含量和膜透性比对照降低了 7.1 2 %和 43.42 % .  相似文献

Activities of peroxidase, amylase and catalase were analyzed on beech (Fagus orientalis Lipsky) during one year to determine relationship between these enzymes with the period of dormancy and growing season. Results showed that amylase activity was high but catalase and peroxidase activities were low during the growing season. Peroxidase and catalase activities increased from July to November whereas amylase activity started to decrease at the same time, which means that the plants were prepared themselves to be dormant and the hardening was happened. The peroxidase maximum activity was in November, which is an important sign of dormancy and completing hardening in plant. The dormancy released from February along with decrease of catalase activity. At the beginning of growing season, correlation between amylase and catalase was increased. Amylase activity also increased gradually. No significant correlation between amylase-catalase and amylase-peroxidase activities was found at the period of dormancy. In our study, we found that the seasonal activity of enzymes such as peroxidase, catalase, and amylase, also correlation between these enzymes could be an important factor for the detection the period of dormancy and growing season.  相似文献

匙吻鲟仔稚鱼消化酶发育的研究   总被引:3,自引:0,他引:3  
对出膜后0—53d匙吻鲟的酸性蛋白酶、碱性蛋白酶、α-淀粉酶、脂肪酶以及磷酸酶的活性变化进行了测定。匙吻鲟出膜后饲养于室内水泥培育池中,从第3天开始投喂枝角类,之后于第40天将试验鱼转移至池塘。试验材料为受精卵及出膜后第3、第6、第12、第20、第30、第40、第44、第47、第53天仔稚鱼样品。研究发现主要消化酶在出膜时或卵黄期即可检测出活力。碱性蛋白酶和酸性蛋白酶分别在出膜后3d(3DAH)和刚出膜时(0DAH)检测出活力。碱性蛋白酶活力在44DAH达到最大值[(1.96±0.09)U/fish],47DAH出现下降,但在53DAH开始上升,比活力在53DAH达到最大值[(8.84±0.59)U/mg protein]。酸性蛋白酶在44DAH达到最大值[(0.52±0.05)U/fish],比活力在6DAH出现第一个峰值[(2.08±0.09)U/mg protein],并在30DAH出现最小值[(0.83±0.06)U/mg protein]。试验期间碱性蛋白酶活力高于酸性蛋白酶。在12DAH—40DAH期间α-淀粉酶活力相对稳定,并在47DAH达到最大值[(0.42±0.03)U/fish],比活力在12DAH出现一个峰值[(1.18±0.12)U/mg protein],并于47DAH出现最大值[(1.94±0.16)U/mg protein]。发育早期脂肪酶活力较高,活力和比活力分别在30DAH[(0.20±0.02)U/fish]和6DAH[(2.28±0.22)U/mg protein]出现最大值。碱性磷酸酶活力变化趋势与比活力变化趋势相似,但是最大值分别出现在44DAH[(0.08±0.00)U/fish]和30DAH[(1.96±0.15)U/mg protein]。酸性磷酸酶活力在3DAH出现一个峰值[(0.01±0.00)U/fish],之后显著升高,并在44DAH达到最大值[(0.05±0.00)U/fish],其比活分别在30DAH[(1.19±0.10)U/mg protein]和44DAH[(1.10±0.08)U/mg protein]出现两个峰值。结果表明,蛋白酶、α-淀粉酶和磷酸酶随个体发育活力增加,碱性蛋白酶在个体发育早期对蛋白质的消化具有重要作用。养殖环境发生改变时,酸性蛋白酶、α-淀粉酶、碱性磷酸酶和酸性磷酸酶活力在生长减慢时增加,生长加快时降低,而脂肪酶活力则维持稳定。  相似文献

应用酶学分析法测定了树麻雀Passer montanus春季的腺胃、肌胃、小肠、大肠、肝脏和胰脏组织中淀粉酶、蛋白酶和纤维素酶的活力.结果表明,不同组织中的消化酶活力差异显著,淀粉酶和蛋白酶以胰脏中活力最高,腺胃次之,纤维素酶均较低;同一组织中不同消化酶的活力差异显著,淀粉酶活性最高,蛋白酶次之,纤维素酶活力最低.这些差异提示消化酶活力大小与器官分化有关,并受食物组成的影响,因此产生了不同的酶活力分布.这是树麻雀长期适应东北地区寒冷环境的生存策略之一.  相似文献

Summary Bacillus caldolyticus, grown at 70°C, produces a highly active extracellular amylase and protease. Both enzymes are formed either within the membrane, or at its inner surface. The activity of both extracellular enzymes was found to decline drastically when brain-heart infusion was omitted from the medium. A simultaneous increase of both enzymes inside the cell was observed. The shifting in extra- and intra-cellular activity was caused by changes in membrane composition due to the increase of anteiso-odd and n-even, and the decrease of iso-odd fatty acids. Membrane composition and enzymic activity could be influenced by the addition of either leucine or iso-leucine as precursors for the synthesis of branched-chain fatty acids: In presence of leucine the anteiso-odd and n-even fatty acids returned to their normal level, while the iso-odd fatty acids increased. Simultaneously the extracellular protease activity increased, and the intracellular activity declined. Growth in amylose-medium supplied with leucine lead to a decrease of both the intra- and extracellular amylase, and changes in the fatty acid composition of the membrane which could not be restored by transfer of the organism to complete media. Addition of iso-leucine first lead to a sharp decrease of extracellular protease and a drastic increase of intracellular protease activity, accompanied by an increase of anteiso-odd and n-even fatty acids, and a decrease of iso-odd compounds. After the second growth in presence of iso-leucine the intra- and extra-cellular protease activity was reversed, and thus showed a return to the starting situation. The reversal is accompanied by the preferential incorporation of fatty acids with a higher melting point into the membrane. Extracellular amylase activity was found to increase after the first growth with iso-leucine, and to decline sharply after the second culture with iso-leucine, together with a very high intracellular amylase activity at that point. Extra- and intra-cellular amylase activity both declined upon growth in complete medium, while the fatty acid distribution remained different from the initial composition.  相似文献

采用动物性饵料和人工饲料培育1~10日龄怀头鲇(Silurus soldatovi)仔稚鱼,分析测定了全鱼酸性、碱性蛋白酶、淀粉酶、脂肪酶以及碱性磷酸酶的活性。结果表明:孵化后3天开口期仔鱼已具有较高的碱性蛋白酶活性,5日龄时碱性蛋白酶比活力达到较高值,8日龄时出现低值,总体变化呈波动上升趋势;酸性蛋白酶活性在1~8日龄处于较低水平,8日龄后开始迅速升高;淀粉酶活性在5日龄左右达到最高值,随后酶活性开始下降至较低水平;脂肪酶活性变化波动较大,表现为双峰型,两个峰值分别出现在3~4日龄和6~8日龄。摄食动物性饵料仔稚鱼消化酶活性和碱性磷酸酶活性均高于摄食人工饲料。在整个早期发育过程中,碱性蛋白酶比酸性蛋白酶活性高,碱性蛋白酶、淀粉酶比活力在约8日龄仔稚鱼转变期明显下降,而酸性蛋白酶活性开始迅速升高,这说明消化酶活性的变化与仔稚鱼发育过程中消化机能转换具有相关性。怀头鲇在10日龄内碱性磷酸酶活性呈上升趋势,表明怀头鲇胃肠道功能的逐步发育完善。  相似文献

THE COMPOSITION OF ISOLATED NUCLEI AND CELL PREPARATIONS FROM TISSUES OF CALF, BEEF, HORSE, AND FOWL WAS STUDIED WITH RESPECT TO THE FOLLOWING COMPONENTS: 1. Liver and kidney arginase, catalase, and uricase; pancreatic lipase and amylase; cardiac muscle myoglobin; erythrocyte hemoglobin; intestinal alkaline phospharase. These are referred to as "special" components in view of their characteristically restricted distribution reflecting the differentiated nature of the tissues in question. 2. Esterase, beta-glucuronidase, alkaline and nucleotide phosphatases, adenosine deaminase, guanase, and nucleoside phosphorylase. These are enzymes of general distribution. The differences in nuclear composition noted with respect to the "special" components, together with the broad variability in nuclear activity found for enzymes of general distribution, led to the conclusion that nuclei are differentiated structures. The following distribution was observed: 1. "Special" components: Hemoglobin was found to be present in fowl and goose erythrocyte nuclei, but myoglobin was entirely absent from heart muscle nuclei; of the special enzymes listed, only catalase and arginase appeared to be concentrated in some of the nuclei. There was no significant nuclear concentration of lipase, amylase, uricase, or alkaline phosphatase. No simple relationship was found between the concentration of a special enzyme in a tissue and its activity in the corresponding nuclei. For example, arginase activity, which is high in mammalian liver and in fowl kidney, was found in liver, not kidney, nuclei. Similarly, catalase activity was demonstrated only in mammalian liver nuclei, although, in mammals, both liver and kidney are rich sources of this enzyme. 2. Enzymes of general distribution fell into three classes: (a) Those present in low concentrations, if at all, in the nuclei-alkaline phosphatase, the nucleotide phosphatases) and beta-glucuronidase. (b) Those present in nuclei in varying concentrations-esterase. (c) Those present in high proportions in most nuclei-adenosine deaminase, nucleoside phosphorylase, and guanase. The exceptionally low nuclear activity of intestinal mucosa with respect to these enzymes was discussed in relation to physiological considerations. The response of nuclei to changes in physiological state was demonstrated by experiments on starvation. The outstanding aspect of this response was a change in nuclear enzymatic activity opposing that observed in the cytoplasm. A comparison of fetal and adult mucosa cells led to the following tentative interpretation of the observed intracellular enzyme distribution: In cells tending to moribundity, as in those subjected to starvation, relative nuclear enzymatic activity falls. The occurrence of special enzymes in nuclei was considered in terms of differentiation, and the high nuclear concentration of the nucleoside-specific enzymes was interpreted in terms of general nuclear metabolic activity.  相似文献

目的研究复合微生态制剂对幼刺参体壁营养成分、几种消化酶和免疫酶活力的影响。方法在封闭式循环系统中投喂不同的微生态制剂进行30 d刺参养殖实验。结果投喂液态复合微生态制剂组和粉状复合微生态制剂组的刺参体壁的粗脂肪、总糖、粗蛋白含量最高,两组与对照组相比差异有统计学意义(Ps0.05)。添加微生态制剂的3个实验组刺参肠道蛋白酶、淀粉酶活力均比对照组高,且差异有统计学意义(Ps0.05)。实验组刺参的肠道、体壁、体液过氧化氢酶活力均比对照组高,其中投喂液态复合微生态制剂实验组活力最高。实验组刺参组织的酸性磷酸酶和碱性磷酸酶活力也明显高于对照组(与对照组相比差异有统计学意义,Ps0.05)。结论微生态制剂可以有效改善幼刺参体壁营养成分,促进机体消化活力,提高刺参免疫力。  相似文献

Activities of hydrolytic enzymes (esterase, lipase, phosphatase, protease, 1,3-β-glucanase, chitinase and amylase) were demonstrated in washings and extracts of ungerminated and germinated conidia of Erysiphe graminis f. sp. hordei , Marchal. No cellulase and only very little xylanase activity was detected. Considerable amounts, 65% of total conidia proteins can be extracted by shaking the conidia when the extraction medium contained non-ionic detergent only. Different extraction procedures were used to determine enzyme distribution between exo- and endoconidial compartments and to evaluate the enzyme extraction efficiency from conidia.
The possible physiological roles of the hydrolytic enzymes in the ungerminated as well as in the germinated conidia were discussed.  相似文献

目的 测定不同生态环境中铜绿丽金龟幼虫肠道细菌产消化酶活性。方法 采用平板透明圈法和分光光度计法。结果 平板透明圈法测得废弃菜园和花生田中铜绿丽金龟幼虫肠道细菌产蛋白酶和淀粉酶活性差异无统计学意义(F=1.089、0.4963,P>0.05),而细菌的产纤维素酶活性有显著差异,其中花生田铜绿丽金龟幼虫肠道中分离到的粘质沙雷菌产酶活性显著高于其他菌株;分光光度计测得的不同生态环境中铜绿丽金龟幼虫肠道细菌的产蛋白酶、淀粉酶和纤维素酶活性差异均有统计学意义(F=461.565、42.349、18.7673,P<0.05)。从变异系数来看,分光光度计法测得的蛋白酶、淀粉酶和纤维素酶的变异程度较低,而脂肪酶测定时平板透明圈法的变异系数较低。结论 平板透明圈法和分光光度计法均可用于铜绿丽金龟幼虫肠道细菌产消化酶活性测定。通过分析研究比较,分光光度计法适于测定细菌的产蛋白酶、淀粉酶和纤维素酶活性,平板透明圈法适于测定细菌的产脂肪酶活性。  相似文献

在一个水稻生长季节,采用田间采样与室内分析的方法,利用氯仿熏蒸培养法、苯酚钠比色法、TTC比色法、高锰酸钾滴定法和茚三酮比色法测定和研究了稻鸭与稻鱼生态系统稻田土壤微生物量N(MBN)和土壤脲酶、脱氢酶、过氧化氢酶和蛋白酶活性的动态变化,及其与土壤养分和水稻吸N量的相关性.结果表明,(1)随着水稻的生长,土壤微生物量N表现为先上升随后下降,于成熟期有所回升趋势;土壤微生物量N与土壤速效N、全N和全P不相关;由于水稻与微生物对养分的竞争,土壤微生物量N与水稻吸N量不相关;稻田养鸭,养鱼显著地提高了土壤微生物量N.(2)土壤脲酶、脱氢酶和蛋白酶活性都表现出先上升后下降趋势,而过氧化氢酶活性变化不大;稻鸭与稻鱼共作显著地提高了土壤脲酶、脱氢酶和蛋白酶活性,而对过氧化氢酶活性影响不大;线性相关分析表明,在一个稻季内,土壤脲酶和脱氢酶活性与土壤速效N负相关,土壤脲酶、脱氢酶、过氧化氢酶和蛋白酶活性与土壤全N和全P不相关;土壤脲酶、脱氢酶和蛋白酶活性与水稻吸N量呈显著相关.(3)稻田土壤微生物量N与土壤酶不相关.  相似文献

猕猴桃(Actinidia deliciosa C.F.Liang etA.R.Ferguson. cv. Qinm ei)果实采后的软化过程表现为两个明显的阶段,第一阶段软化较快,此时对软化起主要作用的阶段性专一酶是淀粉酶;第二阶段软化速度变慢,此时起主要作用的阶段性专一酶是多聚半乳糖醛酸酶和纤维素酶。乙烯形成酶(EFE)的活性高峰出现在两个软化阶段之间,它所引起的乙烯释放对软化有促进作用,因此EFE也是与果实软化有关的阶段性专一酶。但是,果胶甲酯酶(PME)的活性变化与果实的软化无相关关系,过氧化物酶(POX)、过氧化氢酶(CAT)和SOD的活性高峰出现在果实完全软化以后,因此不是果实软化的阶段性专一酶  相似文献

多数研究表明外生菌根真菌能够促进植物养分吸收并提高植物生长,但是对其发生的原因研究较少。本文在室内控制条件下,研究了真菌菌丝分泌N、P相关胞外酶及其受土壤有机碳(胡敏酸)和无机碳(碳酸钙)添加的影响,结果表明:1)3种真菌——松乳菇(Lactarius deliciosus)、变色红菇(Russula integra)、铆钉菇(Gomphidius viscidus)菌丝均能够分泌酸性磷酸酶和蛋白酶,而且多数情况下,MMN培养基培养14 d时,各个酶活性较高,而不同菌的胞外酶活性存在较大的差异,平均值来看铆钉菇酸性磷酸酶活性最低而蛋白酶活性最高,其它2个真菌菌丝的胞外酶活性差异不大;2)添加胡敏酸后,3种菌丝的酸性磷酸酶活性都是随着胡敏酸添加量的增加而逐渐增加;但蛋白酶活性存在差异:松乳菇的蛋白酶活性随着胡敏酸添加量的增加而逐渐增加;变色红菇的蛋白酶活性对胡敏酸不敏感,受其影响不大;铆钉菇的蛋白酶活力在少量的胡敏酸作用下最强,但浓度过高反而抑制其蛋白酶的活性。3)添加碳酸钙后,总体来看,3种菌丝胞外酸性磷酸酶和蛋白酶活性都是添加少量碳酸钙时酶活性最强,随着浓度的增加(如0.1 g),其酶活性开始受到抑制。综上所述,真菌菌丝能够分泌酸性磷酸酶和蛋白酶,这可能是因为这些外生菌根真菌能够促进植物养分吸收和快速生长的原因;有机碳和无机碳的加入可以直接影响真菌菌丝胞外酶的分泌,进而影响土壤内有机磷和有机氮化合物的分解,显示其在土壤碳循环中的作用。  相似文献

Various microbial groups are well known to produce a range of extracellular enzymes and other secondary metabolites. However, the occurrence and importance of investment in such activities have received relatively limited attention in studies of Antarctic soil microbiota. In order to examine extracellular enzyme production in this chronically low-temperature environment, fungi were isolated from ornithogenic, pristine and human-impacted soils collected from the Fildes Peninsula, King George Island, Antarctica during the austral summer in February 2007. Twenty-eight isolates of psychrophilic and psychrotolerant fungi were obtained and screened at a culture temperature of 4°C for activity of extracellular hydrolase enzymes (amylase, cellulase, protease), using R2A agar plates supplemented with (a) starch for amylase activity, (b) carboxymethyl cellulose and trypan blue for cellulase activity or (c) skim milk for protease activity. Sixteen isolates showed activity for amylase, 23 for cellulase and 21 for protease. One isolate showed significant activity across all three enzyme types, and a further 10 isolates showed significant activity for at least two of the enzymes. No clear associations were apparent between the fungal taxa isolated and the type of source soil, or in the balance of production of different extracellular enzymes between the different soil habitats sampled. Investment in extracellular enzyme production is clearly an important element of the survival strategy of these fungi in maritime Antarctic soils.  相似文献

The activities of several stage specific enzymes (SSE) and the changes of some compounds during softening of “Qinmei” kiwifruit (Actinidia deliciosa) were studied. The resuits showed that there were two phases of kiwifruit softening. Firstly the rapid softening phase was coincided with starch degradation (r= 0. 99) and the increase of amylase activity. It suggested that amylase was the key SSE for softening at this phase. In the second phase, the rate of softening was decreased, the contents of water insoluble pectin and cellulose apparently were reduced and the activities of polygalacturoase (PG) and cellulase were markedly increased, which indicated the PG and cellulase were the key SSE in the second softening phase. Besides, ethylene forming enzyme (EFE) activity and ethylene production were at their peaks between the two phases as they might play the part as triggers of the PG and cellulase activities. The experiments also showed that the activity of pectin methylesterase (PME) was not related to the softening of kiwifruit. The maximum activities of peroxidase (POX), catalase (CAT) and superoxide dismutase (SOD) were found after the softening stages. It seemed that they were not the key SSE for the softening of kiwifruit.  相似文献

In homogenates of Tetrahymena pyriformis, five hydrolases — phosphatase, ribonuclease, deoxyribonuclease, proteinase, amylase — with acid pH optima were found. Over 75% of their activity is sedimentable with a centrifugal force of 250,000 g. min. Only 17% of the acid phosphatase and ribonuclease is active when assayed in the presence of 0.25 M sucrose at 0°. Exposure to a lowered osmotic pressure, freezing and thawing, and incubation at temperatures over 0° result in activation of the latent phosphatase and ribonuclease. After isopycnic centrifugation in a sucrose density gradient the hydrolases show a broad distribution which differs greatly from those of enzymes associated with mitochondria (succinate dehydrogenase) or with peroxisomes (catalase). The results are interpreted as evidence that the five acid hydrolases studied are localized in lysosomes which represent a distinct population of subcellular particles in Tetrahymena.  相似文献

Enzymatic activity, i.e. respiratory activity, dehydrogenase activity, phosphatase activity, caseinian protease activity, BAA protease activity and urease activity, was determined to investigate the process of biochemical transformations and to select enzymatic indices of maturity of composts prepared from feathers and lignocellulose wastes (bark, straw). Composting was conducted for 7 months, with periodic determinations of activity of the enzymes. The study revealed significant differences in the enzymatic activity, related with the duration of composting and with the substrate composition of the composts. Generally, composts enriched with straw were characterised by higher enzymatic activity than composts without any addition of straw. It was found that the activity of such enzymes as cellulase and protease, towards the end of the period of composting decreased and stabilised. The enzymes enumerated can be taken into consideration in estimation of the maturity of composts prepared from feathers and lignocellulose wastes.  相似文献

Inclusion of cadmium as cadmium chloride and nickel as nickel chloride in media differentially induced dry matter mobilisation from cotyledons of germinating pigeon pea seeds. Cadmium depressed the activities of total amylase, protease, acid phosphatase and peroxidase in germinating seeds. The activities of these enzymes were stimulated at the lower concentrations of nickel and suppressed at the higher ones. This dual response of hydrolytic enzymes to cadmium and nickel is postulated to account for the promotory and inhibitory effects of these heavy metals on dry matter mobilisation.  相似文献

The activity of 10 enzymes separated by acrylamide disc gel electrophoresis of leaf and stem extracts from Dianthus grown under summer and winter conditions was studied. While banding was constant and highly reproducible under each environment, differences between the 3 cultivars and between the tissues were evident. No significant differences in the isozyme patterns of glutamate dehydrogenase, 6-phosphogluconate dehydrogenase, glucose-6-phosphate dehydrogenase, malate dehydrogenase, and catalase were observed between the 2 environments. Loss of activity was observed under winter conditions with amylase and lactate dehydrogenase and loss of certain isozymic components was evident with acid phosphatase and esterase. Prominent changes were observed in peroxidase isozymes, the hardy cultivars developing additional isozymic components under winter conditions. Only minor changes in the total protein banding were seen. The enzymes showed considerable stability in those tissues killed by the freezing conditions.  相似文献
