Single larval inoculations of both the citrus and banana races of Radopholus similis from Florida indicate parthenogenetic reproduction is possible. These races normally reproduce amphimictieally. Both races produced all-female populations from single larva inoculations after 80 d and male-female populations after 180 d. The all-female populations produced female progeny with viable eggs, but no spermatozoa were present in the spermatheca, indicating tychoparthenogenesis occurred. Uninseminated females in the latter study produced both male and female progeny. Approximately 95% of the female progeny of the male-female populations were inseminated, indicating that cross-fertilization had occurred. The proportion of males to females was as expected in a normal bisexual population. No intersex males were observed. The mechanism of sex determination is not fully understood but assumed to be enviromnentally induced.  相似文献

Karyotype, host preference, isozyzme patterns, morphometrics, and mating behavior of two burrowing nematode populations from Hawaii, one infecting Anthurium sp. and the second infecting Musa sp., were compared with Radopholus similis and R. citrophilus populations from Florida. The population from Anthurium sp. had five chromosomes (n = 5), and that from Musa sp. had four (n = 4). Neither of the Hawaiian nematode populations persisted in roots of Citrus limon or C. aurantium. Anthurium clarinerivum and A. hookeri were hosts of the burrowing nematode population from anthurium in Hawaii and of R. citrophilus from Florida, whereas the two anthurium species were poor hosts of the population from Musa sp. in Hawaii and R. similis from Florida. The isozyme pattern of the population isolated from anthurium was identical to that of R. citrophigus, whereas the pattern of the population from banana in Hawaii was identical to that of R. similis. Mating behavior between the burrowing nematode population isolated from Anthurium sp. and a Florida population of R. citrophilus supports their close taxonomic relationship. Mating was observed between the population from Anthurium sp. and the Florida population of R. citrophilus but not between the Hawaiian burrowing nematode population isolated from Musa sp. and a Florida population of R. citrophilus. These findings indicate that a previously unidentified population of R. citrophilus which does not parasitize citrus occurs in Hawaii.  相似文献

Analysis of proteins of the banana and citrus race of Radopholus similis was carried out by several different types of polyacrylamide gel electrophoresis. These included standard slab gel, SDS slab gel, gradient slab gel, and two-ditnensional slab gel electrophoresis. A major band difference was detected between the two races by slab gel electrophoresis. However, several other poorly resolved but consistent hands of high molecular weight proteins near the gel origin also were considered as diagnostic. Resolution of protein bands was greatly improved by SDS and gradient slab gel electrophoresis, but no differences could be detected among the proteins resolved between the two rares with these techniques. Two-dimensional gels revealed a large number of proteins, but background staining obscured them hindering interpretation. When nematode races were reared on three different host plants, no differences in protein patterns were detected between them, indicating host preferences does not play a role in determining the types proteins occurring in these nematodes.  相似文献

Some physiological properties of citrus and noncitrus races ofGeotrichum candidum isolated from soils of citrus groves and noncitrus fields in Japan were studied. Of 358 isolated of citrus race, 260 isolates required pyridoxine for growth on asparagine-glucose agar, while 98 isolates could grow in the absence of pyridoxine. On the other hand, of 11 isolates of noncitrus race, 10 isolates could grow without pyridoxine, while 1 isolate absolutely required pyridoxine for growth. The citrus race grew well in autoclaved lemon juice at pH 2.2, while the noncitrus race failed to grow. The radial growth rate of the citrus race on potato-dextrose agar was higher than that of the noncitrus race. The two races grew well within the same range of temperature and pH, the optimal temperature ranging between 25 and 30°C, and optimal pH being 6.0. Both races utilized glucose, galactose, xylose, sorbose, sorbitol and mannitol as carbon sources, but could not utilize arabinose, lactose, maltose, rhamnose and sucrose. Arginine and asparaghine were the best nitrogen sources for both races, but KNO3 could not be utilized.  相似文献

220 populations of Meloidogyne incognita and related forms from 46 countries reproduced by mitotic parthenogenesis (apomixis). Determination of somatic chromosome numbers from oogonia and oocytes revealed the existence of a predominant, possibly triploid race A with 3n = 40 to 46 and a rare, diploid race B with 2n = 32 to 36 chromosomes. There is no correlation between cytological races and the four recognized host races of this species. The characteristic behavior of prophase I chromosomes of maturing oocytes, which results in a prolonged prophase stage, is a unifying feature of all forms of M. incognita and supports monophyletic evolution, distinct from that of other Meloidogyne species. Extensive chromosomal polymorphism detected among populations can be helpful in elucidating the cytological pathway of evolution of the species.  相似文献

Hypotheses on the evolution of the karyotypes of 8 chromosome races (2n = 4, 6, 8, 10, 12, 16-two forms, 26) within theOrnithogalum tenuifolium complex are discussed. Four of the karyotypes are strictly bimodal: 2n = 8 (6 long and two short chromosomes), 2n = 10 (6 long and 4 short chromosomes), 2n = 12 (6 long and 6 short chromosomes) and 2n = 16 (12 long and 4 short chromosomes). The hypotheses are tested by means of measurements of nuclear DNA content, studies of meiosis and pollen fertility of hybrids, and comparisons of karyotype morphology. The results indicate that the E. African 2n = 12 chromosome race is the most primitive and has given rise to the other chromosome races. The 2n = 6 race is found to have a significantly higher fitness than the 2n = 12 race.  相似文献

A comparison of the karyotypes of races D (2n=8), E (2n=10), B (2n=12) and C (2n=16) of B. lineariloba suggests that these races have in common a basic set of four chromosome pairs, and that the higher chromosome number races are related to race D by successive chromosome addition. — A study of meiosis in B × C and A1 × B hybrids supports this contention and elucidates the homologies of the additional chromosomes. — Meiotic pairing in hybrids between A and C is very complex. At present it can only be stated that there are extensive interchromosomal homologies between the two races. — Two phyletic schemes of the relationships of the races are considered. The second, which is favoured, involves successive chromosome addition, with the quasidiploid race E (2n = 10) giving rise to race B by diploidisation of the univalent chromosomes. This scheme is supported by features of univalent behaviour in the various races and their hybrids. — The ecogeographic distribution pattern of the races shows replacement of D by E by B by C as the species extends into more arid and more harsh environments. This replacement is also associated with increasing vigour. — It seems most likely that the addition chromosomes are derived from a race A (2n=4) source since they are added always by twos, and each addition increases both vigour and drought tolerance. Race A is the most vigorous and one of the most drought tolerant of the five races.It is suggested that the evolution of the races can be related to the increasing aridity of the Late Pleistocene and Recent geological epochs.  相似文献

Analysis of genetic variation between the banana and the citrus races of Radopholus similis by starch gel eleclrophoresis demonstrated that 7 of 16 enzyme-encoding loci could be used for their diagnostic separation. The two races are closely related arid share approximately 75% of the enzymes evaluated. The level of dissimilarities o1 inherited bands indicates that no gene flow occurs between the races. Aldolase, α + β esterase, glucose-6-phosphate dehydrogenase, isocitrate dehydrogenase, lactate dehydrogenase, malate dehydrogenase, and phosphoglucose isomerase are diagnostic markers of the races.  相似文献

Root gall induction and egg production by the four recognized host races and two cytological races of Meloidogyne incognita were compared on cotton Gossypium hirsutum cvs. Deltapine 16 (root-knot susceptible) and Auburn 634 (highly resistant). The 12 nematode populations included in the study were from various parts of the world. No population increases occurred on the highly resistant cultivar. After 45 days, populations of host races 1 and 2 induced slight root galling on both cuhivars with only limited reproduction. Host race 4 populations induced moderate root galling with higher reproduction on Deltapine 16 than that of race 1 or race 2 populations. Host race 3 populations induced severe root galling with population density increases of 7-30-fold. In a complementary study, 24 cotton cultivars or breeding lines were compared for suitability as hosts for a typical population of M. incognita race 3. The poorest hosts, ''Aubnru 623,'' ''Auburn 634,'' and ''McNair 220,'' yielded fewer eggs after 45 days than were added initially. The best hosts - ''M-8.'' ''DES 24-8,'' ''McNair 235,'' and ''Coker 20l'' - yielded > 5 times as many eggs as were added initially.  相似文献

The grasshopper Podisma sapporensis consists of two main chromosome races in Hokkaido. The western group of populations of P. sapporensis, belonging to the XO race, has a diploid number of chromosomes 2n = 23 in the male and 2n = 24 in the female (sex determination XO male/XX female). The eastern group of populations of this species, belonging to the XY race, differs from the western one as a result of Robertsonian translocation between the originally acrocentric X chromosome and M5 autosome in homozygous state, having resulted in the forming of chromosome sex determination neo-XY male/neo-XX female (2n = 22). These races are geographically isolated by the mountainous system consisting of the Mts Daisetsu and Hidaka range, occupying the central part of the island. The hybrid zones between the races have not so far been discovered. Various levels of polymorphism for the pericentric inversions and C-banding variation exist in different chromosomes throughout populations in both chromosome races. In some solitary populations (the population at the summit of Mt Yotei, populations in the vicinity of Naganuma, Oketo, and Tanno) pericentric inversions are fixed in some pairs of chromosomes, which enables marking of the discrete karyomorphes. In the Mt Daisengen population all chromosomes are two-armed as a result of fixing the pericentric inversions. These facts contradict karyotypical conservatism of the tribe Podismini. The level of diversity of P. sapporensis karyotypes could provide a new perspective on the evolutionary process of different karyotype in Orthoptera. The considerable occurrence of polymorphism in chromosomes suggests that karyotypic diversification is undergoing in P. sapporensis. The authors also proposed that P. sapporensis would be divided into four chromosome subraces in the XO chromosome race and two chromosome subraces in the XY race, on the basis of karyotypic features. These races may have been established by fundamental climatic changes during the glacial epoch.  相似文献

Summary Pachytene chromosome morphology was compared in nine races ofRicinus communis L. (2n = 20), using pollen mother cells (PMCs) and light microscopy. Of the ten bivalents, only the two possessing nucleolar organizing regions (NORs), chromosomes 2 and 7, exhibit structural variations among the races. The NORs are located in the short arms of these two chromosomes. Most of the observed structural variations affect these short arms, which are similar morphologically and consist largely of heterochromatic segments. The PMCs contain a single nucleolus and this is associated with the NOR of each of the two chromosomes at a particular frequency in each race. In eight races, a nucleolar constriction (NC) is present in either chromosome 2 or chromosome 7. In these races, the nucleolus is associated with the chromosome possessing an NC at a frequency of 100% and with the chromosome lacking an NC at a frequency ranging between 5.6 and 100%, depending upon the race. No microscopically visible NC is present in the ninth race. In this race, the nucleolus is associated with both chromosomes 2 and 7 at a frequency of 100%. The association of the nucleolus with a chromosome possessing an NC is at the NC and with a chromosome lacking an NC is at the terminal heterochromatic segment of the short arm. Several interpretations are offered to account for the variations in frequency of association between the nucleolus and each of the nucleolar organizing chromosomes. It is suggested that the two non-linked NORs have evolved through some intragenomic changes rather than polyploidy, that this species is highly intolerant to structural variations other than those occurring in or near the NORs, and that structural variations in the nucleolar organizing chromosomes are not associated with racial variations in plant phenotype.Paper of the Journal Series, New Jersey Agricultural Experiment Station  相似文献

Heterochromatin distribution and differentiation in metaphase chromosomes of two morphologically identical Drosophila races, D. nasuta nasuta and D. n. albomicana, have been studied by C- and N-banding methods. — The total heterochromatin values differ only slightly between these races. However, homologous chromosomes of the two Drosophila forms show striking differences in the size of heterochromatin regions and there is an alternating pattern in D. n. nasuta and D. n. albomicana of chromosomes which contain more, or respectively less heterochromatin than their counterparts in the other race. — Three different N-banding patterns could be obtained depending on the conditions of the method employed: One banding pattern occurs which corresponds to the C-banding pattern. Another pattern is the reverse of the C-band pattern; the euchromatic chromosome regions and the centromeres are stained whereas the pericentric heterochromatin regions remain unstained. In the Y chromosomes of both races and in chromosome 4 of D. n. albomicana, however, the heterochromatin is further differentiated. In the third N-banding pattern only the centromeres are deeply stained. Furthermore, between the races, subtle staining differences in the pericentric heterochromatin regions can be observed as verified in F1 hybrids. On the basis of C- and N-banding results specific aspects of chromosomal differences between D. n. nasuta and D. n. albomicana are discussed.Dedicated to Prof. W. According to published reports from 1906 to 1968, leprosis nearly destroyed the Florida citrus industry prior to 1925. This was supported with photographs showing typical leprosis symptoms on citrus leaves, fruit, and twigs. Support for the past occurrence of citrus leprosis in Florida includes: (1) presence of twig lesions in affected orange blocks in addition to lesions on fruits and leaves and corresponding absence of similar lesions on grapefruit; (2) yield reduction and die-back on infected trees; and (3) spread of the disease between 1906 and 1925. Transmission electron microscopy (TEM) examination of tissue samples from leprosis-like injuries to orange and grapefruit leaves from Florida in 1997, and fruits from grapefruit and sweet orange varieties from Texas in 1999 and 2000 did not contain leprosis-like viral particles or viroplasm inclusions. In contrast, leprosis viroplasm inclusions were readily identified by TEM within green non-senescent tissues surrounding leprosis lesions in two of every three orange leaf samples and half of the fruit samples obtained from Piracicaba, Brazil. Symptoms of leprosis were not seen in any of the 24,555 orange trees examined across Florida during 2001 and 2002. The authors conclude that citrus leprosis no longer exists in Florida nor occurs in Texas citrus based on: (1) lack of leprosis symptoms on leaves, fruit, and twigs of sweet orange citrus varieties surveyed in Florida: (2) failure to find virus particles or viroplasm inclusion bodies in suspect samples from both Florida and Texas examined by TEM; (3) absence of documented reports by others on the presence of characteristic leprosis symptoms in Florida; (4) lack of its documented occurrence in dooryard trees or abandoned or minimal pesticide citrus orchard sites in Florida. In view of the serious threat to citrus in the U.S., every effort must be taken to quarantine the importation of both citrus and woody ornamental plants that serve as hosts for Brevipalpus phoenicis (Geijskes), B. californicus (Banks), and B. obovatus Donnadieu (Acari: Tenuipalpidae) from countries where citrus leprosis occurs.  相似文献

In the two Drosophila races D. n. nasuta and D. n. albomicans the nucleolar-organizer regions (NORs) have been localized on metaphase chromosomes by a combined acid treatment and silver-staining technique. In both Drosophila races one NOR is present on the heterochromatic Y chromosomes and another on the microchromosomes. Such a NOR distribution has not yet been reported in Drosophila. It is suggested that this distribution has evolved from an original type with NORs on the X and Y chromosomes.  相似文献

Generalist plant pathogens may have wide host ranges, but many exhibit varying degrees of host specialization, with multiple pathogen races that have narrower host ranges. These races are often genetically distinct, with each race causing highest disease incidence on its host of origin. We examined host specialization in the seed pathogen Pyrenophora semeniperda by reciprocally inoculating pathogen strains from Bromus tectorum and from four other winter annual grass weeds (Bromus diandrus, Bromus rubens, Bromus arvensis and Taeniatherum caput-medusae) onto dormant seeds of B. tectorum and each alternate host. We found that host species varied in resistance and pathogen strains varied in aggressiveness, but there was no evidence for host specialization. Most variation in aggressiveness was among strains within populations and was expressed similarly on both hosts, resulting in a positive correlation between strain-level disease incidence on B. tectorum and on the alternate host. In spite of this lack of host specialization, we detected weak but significant population genetic structure as a function of host species using two neutral marker systems that yielded similar results. This genetic structure is most likely due to founder effects, as the pathogen is known to be dispersed with host seeds. All host species were highly susceptible to their own pathogen races. Tolerance to infection (i.e., the ability to germinate even when infected and thereby avoid seed mortality) increased as a function of seed germination rate, which in turn increased as dormancy was lost. Pyrenophora semeniperda apparently does not require host specialization to fully exploit these winter annual grass species, which share many life history features that make them ideal hosts for this pathogen.  相似文献

由尖孢镰孢菌古巴专化型Fusarium oxysporum f. sp. cubense, Foc引起的香蕉枯萎病是香蕉生产上的毁灭性病害,自1996年以来已对我国华南地区香蕉生产造成了严重危害。传统上香蕉枯萎病菌生理小种的鉴定主要采用人工接种鉴别寄主尔后测定病菌致病性的方法,但实验周期长,且受季节影响。以来自澳大利亚的香蕉枯萎病菌生理小种1号(BW1)、2号(Race 2)、3号(Race 3)以及亚热带4号(BW4)为对照,对分离自我国华南地区主要香蕉产区(广东、广西、海南、福建等省区)的14株香蕉枯萎病菌的单孢菌株进行致病性测定,并结合热带4号小种(TR4)和亚热带4号小种(ST4)的分子特异检测方法,确定其生理小种类型;同时,利用ITS、TEF-1α、IGS、histone H3、β-tubulin等 5个主要用于镰孢菌系统发育学研究的基因,研究不同地区不同来源的Foc菌株之间的亲缘关系及其与非病原尖孢镰孢菌的关系,并评价这5个基因在香蕉枯萎病菌生理小种鉴定上的应用价值。研究结果表明:(1)来源于我国华南地区的4号小种主要为热带4号小种;(2)TEF-1α、IGS、histone H3等3个基因片段能够将Foc中不同生理小种的菌株划分成不同的系统发育谱系,与致病性测定的结果具有对应关系,也能较好地反映尖孢镰孢菌种内菌株的亲缘关系,可用于香蕉枯萎病菌生理小种鉴定;(3)我国Foc 1号生理小种的遗传多样性高于4号生理小种,Foc 1号生理小种的菌系与来自香蕉果实上的非病原尖孢镰孢菌的亲缘关系比其与Foc 4号生理小种的菌系的亲缘关系更近。  相似文献

Two isolates of Pratylenchus coffeae were collected from coffee roots (in Marília, São Paulo State, Brazil) and Aglaonema (in Rio de Janeiro City, Rio de Janeiro State, Brazil) and maintained in the laboratory on alfalfa callus. Twenty-four plants were tested in the greenhouse to characterize the host preference of these isolates. The host ranges of the isolates differed from each other and, interestingly, coffee, banana, and citrus were not among the better hosts of either isolate. Rather, sorghum, maize, rice, millet, okra, melon, eggplant, and lettuce were the best hosts of the Marília isolate. Poor hosts included French marigold, Rangpur lime, banana, sesame, peanut, sunflower, cotton, French bean, onion, and small onion. The best hosts of the Rio de Janeiro isolate were sesame, soybean, sorghum, castor oil plant, watermelon, squash, eggplant, and melon; the poorest hosts were French marigold, coffee, Rangpur lime, banana, sunflower, peanut, maize, millet, French bean, cotton, onion, sweet pepper, lettuce, okra, and small onion. These isolates have important molecular and morphological differences, suggesting host preference is linked to these characteristics.  相似文献

To evaluate the potential benefits and risks associated with releasing Semielacher petiolatus Girault (Hymenoptera: Eulophidae) in a classical biological control project directed against the citrus leafminer Phyllocnistis citrella Stainton (Lepidoptera: Gracillariidae) in Florida, we evaluated the ability of S. petiolatus females to discriminate between hosts previously parasitized by Ageniaspis citricola Logvinovskaya (Hymenoptera: Encyrtidae) and unparasitized hosts. In laboratory trials, S. petiolatus females did not discriminate between hosts previously parasitized and oviposited and fed on each host category equally. Hatch rate of S. petiolatus eggs on hosts previously parasitized by A. citricola was normal and development time was not different. However, mortality of immatures was significantly higher on previously parasitized hosts when compared to hosts that were not parasitized by A. citricola, and size of adult females reared on hosts previously parasitized was reduced. The relative survivorship of S. petiolatus adults compared with adults of P. citrella and another parasitoid, A. citricola, under three relative humidities (RHs) (55, 76, and 95% at 28 °C) indicated that S. petiolatus survives longer than A. citricola at all RHs tested, but did not survive as long as the citrus leafminer. Finally, the leafminer Liriomyza trifolii Burgess (Diptera: Agromyzidae) was evaluated as a possible host for S. petiolatus but no progeny were produced in choice and no-choice tests. The lack of discrimination raises the concern that S. petiolatus could disrupt the efficacy of A. citricola, which is already established in Florida, without providing substantial reduction of citrus leafminer populations during early spring.  相似文献

During 1945-8, 223 British collections of yellow rust were determined on Gassner & Straib's differential hosts, with the addition of the wheat variety Wilma. Physiologic races 2, 3, 5, 6, 7, 8 and 17 were obtained from wheat, race 46 from barley, race 33 from Hordeum murinum and race 28 from Agropyron repens. Three new races, 6x from wheat, M from H. marinum and G from Dactylis glomerata were also isolated. High temperatures increase the susceptibility of some differential hosts to certain wheat races, but decrease that of others. Biotypes of races 2, 6 and 8, differing from the 'type' cultures of those races, were isolated.
Of the commoner wheat races, nos. 6 and 8, occurring on many wheats, were widespread, but races 5 and 7, on fewer varieties, were confined to the north of Britain, and races 2 and 3, on very few varieties, to the south. Certain grasses are partially susceptible to some of the cereal races. The uredospores of race 2 germinate less well than those of races 5 or 8, other wheat races being intermediate. The optimum temperature for spore germination of race G is 22-5 C, that of all other races 10–13oC.
Twelve wheat varieties were inoculated in the field with each of the eight wheat races, and some varieties developed field resistance to certain races. The races attacking a variety the most severely in the field inoculations were usually the races isolated from that variety in collections received.  相似文献
