Given the rapid growth of artificial intelligence (AI) applications in radiotherapy and the related transformations toward the data-driven healthcare domain, this article summarizes the need and usage of the FAIR (Findable, Accessible, Interoperable, Reusable) data principles in radiotherapy. This work introduces the FAIR data concept, presents practical and relevant use cases and the future role of the different parties involved. The goal of this article is to provide guidance and potential applications of FAIR to various radiotherapy stakeholders, focusing on the central role of medical physicists.  相似文献

Leishmania major and Leishmania donovani cells freshly isolated from infected animals divided slowly as axenic promastigotes but the addition of biopterin in the culture medium greatly enhanced their growth. However, when cells were subjected to serial passages and adapted to culture, this growth-promoting effect of biopterin was no longer observed. Genetic analysis of these culture-adapted Leishmania cells demonstrated that the genes coding for the pterin reductase PTR1 or for the biopterin transporter BT1 were over-expressed. This suggests that Leishmania cells adapted to culture were more efficient in utilizing biopterin, an essential growth factor in Leishmania.  相似文献

Pick up any textbook with ‘Microbiology’ in the title and observe the scant to nonexistent treatment of animal cells in culture. Viruses do not suffer from such an exclusion. Chapters abound regaling the molecular niceties of the complex dances their components undergo while infecting bacteria or animal cells in culture. Indeed, it is normally in the latter sense only that any recognition is given to the existence of animal cells in culture. Such views pervade even the minds of those who initiate and direct activities of societies whoseraison d’être is the growth and exploitation of animal cells in culture. The author of this editorial was appointed to a Chair in Microbiology at the University of Surrey in 1983 and has worked on the growth and use of animal cells in culture continuously since 1970; he, therefore, had to reconcile his position in the university with his chosen calling. In this he was helped by undergraduates too numerous to mention who struggled mightily with essays which asked the question; are animal cells in culture microorganisms? What follows is a digest of their efforts and the views of the author.  相似文献

The scale-up of stirred tank bioreactors from 0·02 m3 to a 0·3 m3 commercial plant is discussed for hybridoma suspension cultures. Schemes for dissolved oxygen control with sparged air in serum containing media are described, as well as mechanical breakage of foam in small and large bioreactors. Porous metal spargers (180–200 × 10−6 m) were found to produce foams which were hard to control. Aeration with larger (≥ 0·001 m) multihole spargers is recommended.

Combined cell damage due to foam formation and control, and possible damage at mechanical seals or submerged bearings, were found to have no measurable effect on cell growth relative to roller bottle production. Hybridomas are shown to withstand significant impeller tip speed ( > 1 m s−1) and fluid turbulence as evidenced by impeller Reynolds numbers in excess of 105. The size of the energy-dissipating terminal eddies was calculated to be greater than ten-fold that of the hybridoma cells. The specific fluid turnover rate was employed as the scale-up criterion.  相似文献

Species vulnerability to pesticides depends on physiological sensitivity, the potential to recover, and the ecological context. We assessed the vulnerability of the mosquito Culex pipiens to a repeated treatment with thiacloprid in outdoor microcosms with and without antagonists (competitive and predatory invertebrates). Microcosms were treated repeatedly (three times) with thiacloprid at a concentration of 0.1, 1, or 10 µg/liter. In microcosms without antagonists, the abundance of Cx. pipiens larvae decreased moderately after the second and the third exposures to 10 µg/liter thiacloprid. In microcosms with antagonists, the abundance of Cx. pipiens larvae declined to approximately zero in the control group and the low concentration treatments during the five weeks of observation. By contrast, the abundance of Cx. pipiens larvae temporarily increased at 10 µg/liter thiacloprid after the second and third contamination. We explained this positive effect on the development of Cx. pipiens because of the decrease in competition due to the elimination of sensitive antagonists combined with the high recovery potential of Cx. pipiens. Based on these results, natural antagonists must be supported for the sustainable control of mosquitoes.  相似文献

Novel coronavirus disease 2019 (COVID-19) pandemic is the most recent health care crisis without specific prophylactic or therapeutic drugs. Antimalarial drug chloroquine (CHL) and its safer derivative hydroxychloroquine (HCHL) have been proposed to be repurposed to treat SARS coronavirus-2 (SARS-CoV-2), the causative agent of COVID-19. CHL/HCHL have anti-inflammatory activity and are used to treat rheumatoid arthritis, osteoarthritis and lupus. Although, CHL/HCHL have an anti-viral activity against several viruses in cell-cultures, the anti-viral activity in-vivo is questionable. Repurposing of CHL/HCHL to treat SARS-CoV-2 infection is appealing. However, there is empirical evidence from animal studies with other viruses suggesting that CHL/HCHL may have an untoward paradoxical effect. One thus cannot exclude the possibility that CHL may increase the severity of the disease and prove deleterious both for the patients and public health efforts to contain the highly contagious and explosive spread of SARS-CoV-2.  相似文献
