The behaviors exhibited by first-instarSinea diadema (F.) (Hemiptera: Reduviidae) during intraspecific interactions are described. Five distinct behaviors were characterized: posturing, striking, stalking, stridulating, and beaking. Distinct bouts of grappling were also noted. Interactions between unfed 2-day-old nymphs, unfed 3-day-old nymphs, and unfed 5-day-old nymphs were recorded and analyzed. The behavior of the nymphs changed with the duration of food deprivation. As the period of food deprivation increased from 2 to 5 days, the frequency of posturing decreased as the frequency of beaking increased. During noncannibalistic interactions, posturing or striking by one nymph was most frequently followed by posturing by the other nymph. In contrast, during cannibalistic interactions, posturing or striking was usually followed by beaking. The incidence of grappling increased with the duration of food deprivation. Moreover, grappling always preceded cannibalism. Although first instars were capable of capturing and killing conspecifics of the same age, cannibalism occurred only after 4 days of food deprivation. These results suggest that a decision-making process is involved. During interactions the nymphs can assess the vulnerability of the opponent. Whereas recently emerged nymphs usually forego the risk of attacking conspecifics, the attacks made by nymphs deprived of food are more persistent despite the danger of predation by their opponent.  相似文献

Exapion ulicis (Forster) and Exapion lemovicinum (Hoffmann) (Coleoptera: Curculionoidea: Apionidae) are seed predators of the three gorse species occurring in Brittany (France): Ulex europaeus L., Ulex gallii Planch., and Ulex minor Roth.(Fabaceae). Host‐plant phenology plays a major role in the relationship between apionid weevils and their gorse species, because larvae develop within gorse pods and adults have to wait for pod dehiscence to be released. We monitored flowering and fruiting phenology of gorse species, weevil reproductive behaviour, and egg‐laying patterns in six natural populations in the native area of these gorse species. At each site, U. europaeus, which flowers mainly in spring, was sympatric with one of two autumn flowering gorse species, U. gallii and U. minor. We noticed that E. ulicis laid eggs in spring and was restricted to U. europaeus whereas E. lemovicinum laid eggs in autumn and was restricted to the two autumn‐flowering species U. gallii and U. minor. Therefore, host specificity depended on gorse phenology, and not on geographic proximity. In addition, the infested pod content showed that E. ulicis laid several eggs per pod and suggested that females chose pods with the highest numbers of seeds. In contrast, E. lemovicinum laid a single egg per pod and showed no preference for pods with many seeds. Finally, the impact of seed predation by E. ulicis was higher than that of E. lemovicinum.  相似文献

Genetic variance-covariance structure of larval performance within and among spatio-temporal populations of the widely distributed, polyphagous tiger swallowtail butterfly, Papilio glaucus , is described. Performance traits were assessed for full-sibling families on three host species: Liriodendron tulipifera, Magnolia virginiana and Prunus serotina . Mean performance varied across hosts, indicating these hosts present unique developmental environments. Although full-sibling families significantly differed in plasticity of across-hosts response in three of the five spatio-temporal populations, additive genetic variation was mostly associated with P. serotina or pupal mass. The relative lack of heritable variation in rate and length of larval development on L. tulipifera and M. virginiana was consistent with an earlier study that established host-associated geographic differentiation of P. glaucus populations. Performance appeared relatively independent across hosts and thus genetic constraints cannot be casually invoked to explain persistence of local adaptation and host specialization in the face of extensive gene flow. I promote the hypothesis that gene flow among geographically distant populations is relatively restricted and that previously established, allozyme-based estimates of panmixia are confounded by effects of Pleistocene glaciations. Significant heterogeneity of variance-covariance structure among spatio-temporal P. glaucus populations supports an interpretation of restricted gene flow and relative evolutionary independence. Despite low precision of estimates of genetic parameters, local variance-covariance structure was remarkably consistent with expectations given the presumed evolutionary history of regional populations.  相似文献

The binding of prantschimgin (PRAN) to matrix metalloproteinase 9 (MMP9) was investigated using multiple techniques. Fluorescence spectroscopy showed that PRAN could quench the MMP9 fluorescence spectra. Changes in the UV/vis and Fourier transform infrared (FTIR) spectra were observed upon ligand binding, along with a significant degree of tryptophan fluorescence quenching on complex formation. The interaction of PRAN with MMP9 has also been studied using molecular docking and molecular dynamics (MD) simulation. The binding models demonstrated aspects of PRAN's conformation, active site interaction, important amino acids and hydrogen bonding. Computational mapping of the possible binding site of PRAN revealed that the ligand is bound in a large hydrophobic cavity of MMP9. The MD simulation results suggested that this ligand can interact with the protein, with little affecting the secondary structure. The results not only lead to a better understanding of interactions between PRAN and MMP9, but also provide useful data about the influence of PRAN on the structural conformation. 1. Numerous studies have reported the effects of learning or experience on parasitoid host preference and location. However, the integration of pre‐imaginal and adult experiences on the subsequent host preference and adult/offspring performance has been rarely tested in host–parasite interactions. 2. We present direct evidence that theses two kinds of experiences affect host preference and related fitness in the polyphagous parasitoid, Scleroderma guani. Two colonies of parasitoids were reared on Monochamus alternatus and Saperda populnea (Cerambycidae: Lamiinae). Individuals from the two colonies were given host‐switching experience for one generation (pre‐imaginal experience) while other individuals were given prior ovipositing experience on the two species, respectively (adult experience). 3. Scleroderma guani females demonstrated that their experiences determined adult behavioural responses and their subsequent performance to hosts. Females maximised both adult fitness (fecundity and longevity) and offspring fitness (survival and sex ratio) when they encountered hosts similar to their maternal hosts. Behavioural plasticity in host choice was affected by adult experience, resulting in improved adult feeding and ovipositing behaviour and further modifying adult fecundity and the offspring sex ratio. There was a positive correlation between oviposition preference and adult fecundity. 4. The results indicated that S. guani exhibited positive preference–performance correlations. This is most likely due to an adaptation to maternal hosts over multiple generations. However, foraging potential of adults to available cues from hosts may be driven quickly by an experience‐induced learning process rather than by natural selection processes shaped over many generations.  相似文献

Vertebrate frugivores often feed on fruits upon or within which insects also feed, yet little information exists on the potential magnitude of interactions between these consumers. The Mediterranean shrub Pistacia terebinthus, the birds that consume its fruits, and the wasps that feed upon its seeds are examined in this study. P. terebinthus produces a highly variable fraction of final-sized red fruits that never become mature (green-colored). Red fruits can be immature, parthenocarpic, aborted, or attacked by wasps, and their pulp is much less nutritious than that of mature fruits. A total of 20 bird species consumed the fruits in the study area. Legitimate dispersers accounted for 39% of the total fruit removal, while pulp eaters and seed predators accounted for the remainder. Birds strongly preferred the mature fruits (only 4% of the fruits consumed were red). The incidence of wasps in the seeds ranged from 0 to 42% of the crop in 1989 and from 0 to 24% in 1990. The influence of avian and insect frugivore guilds on each other appears to be quite low because of the narrow overlap in resource utilization by birds and wasps, and an overall low intensity of wasp seed predation. From an evolutionary perspective, the possible ability of wasps to preclude fruit maturation appears not to be attributable to the present interaction with avian frugivores.  相似文献

Gene duplication provides essential material for functional divergence of proteins and hence allows organisms to adapt to changing environments. Following duplication events, redundant paralogs may undergo different evolutionary paths via processes known as nonfunctionalization, neofunctionalization, or subfunctionalization. Studies of adaptive evolution at the molecular level have progressed rapidly by computationally analyzing nucleotide substitution patterns but such studies are limited by the absence of information relating to alterations of function of the encoded enzymes. In this respect, evolution of the Papilio polyxenes cytochrome P450 monooxygenases (P450s) responsible for the adaptation of this insect to furanocoumarin-containing host plants provides an excellent model for elucidating the evolutionary fate of duplicated genes. Evidence from sequence and functional analysis in combination with molecular modeling indicates that the paralogous CYP6B1 and CYP6B3 genes in P. polyxenes have probably evolved via subfunctionalization after the duplication event by which they arose. Both enzymes have been under independent purifying selection as evidenced by the low dN/dS ratio in both the coding region and substrate recognition sites. Both enzymes have maintained their ability to metabolize linear and angular furanocoumarins albeit at different efficiencies. Comparisons of molecular models developed for the CYP6B3 and CYP6B1 proteins highlight differences in their binding modes that account for their different activities toward linear and angular furanocoumarins. That P. polyxenes maintains these 2 furanocoumarin-metabolizing loci with somewhat different activities and expression patterns provides this species with the potential to acquire P450s with novel functions while maintaining those most critical to its exclusive feeding on its current range of host plants.  相似文献

The spatial scale of genetic diversity among patches of a host plant could affect the likelihood of pathogen adaptation to the host. If host patches are genetically distinct, pathogen adaptation to local host genotypes may occur. To study this issue, we focused on the ecological and genetic interactions between two rust fungi, Puccinia seymouriana and P. sparganioides, and the clonal prairie grass, Spartina pectinata. In a field transplant experiment, disease levels differed among plants from different patches, suggesting variation in resistance. Over a 4.5-km scale, disease levels were not higher on plants transplanted back into their source patch as opposed to other locations, providing no evidence for local adaptation in the pathogen. However, on the spatial scales examined (ranging from 0.2 km to 120 km), there was no relationship between the physical distance separating host patches and their similarity in isozyme banding patterns, implying that plants from more distant patches are not necessarily more genetically distinct than plants from nearby patches. Plants derived from the most distant location had, on average, the lowest mean number of pustules at the end of the summer, suggesting the need for reciprocal transplant studies to be performed on a larger spatial scale.  相似文献

The role of pathologic auto‐antibodies against myelin oligodendrocyte glycoprotein (MOG) in multiple sclerosis is a highly controversial matter. As the use of animal models may enable to unravel the molecular mechanisms of the human disorder, numerous studies on multiple sclerosis are carried out using experimental autoimmune encephalomyelitis (EAE). In particular, the most extensively used EAE model is obtained by immunizing C57BL/6 mice with the immunodominant peptide MOG(35–55). In this scenario, we analyzed the anti‐MOG antibody response in this model using the recombinant refolded extracellular domain of the protein, MOG(1–117). To assess the presence of a B‐cell intramolecular epitope spreading mechanism, we tested also five synthetic peptides mapping the 1–117 sequence of MOG, including MOG(35–55). For this purpose, we cloned, expressed in Escherichia coli and on‐column refolded MOG(1–117), and we applied an optimized microwave‐assisted solid‐phase synthetic strategy to obtain the designed peptide sequences. Subsequently, we set up a solid‐phase immunoenzymatic assay testing both naïve and EAE mice sera and using MOG protein and peptides as antigenic probes. The results obtained disclose an intense IgG antibody response against both the recombinant protein and the immunizing peptide, while no response was observed against the other synthetic fragments, thus excluding the presence of an intramolecular epitope spreading mechanism. Furthermore, as the properly refolded recombinant probe is able to bind antibodies with greater efficiency compared with MOG(35–55), we hypothesize the presence of both linear and conformational epitopes on MOG(35–55) sequence. Fluoride-resistant acid phosphatase (FRAP) activity as characterized in rat and mouse was studied in sensory ganglion and spinal cord of several mammals, using both the Gomori lead-ion capture and azo-dye coupling methods. FRAP was specifically localized to small- and medium-diameter primary afferent neurons and inner substantia gelatinosa of all nonrodent animals studied, including rabbit, cat, dog, monkey, cow, and human. In rabbit, sciatic nerve transection resulted in depletion of enzymatic activity in ipsilateral spinal cord dorsal horn in a pattern corresponding to the distribution of central terminals of the nerve. Further analysis of the substrate specificity and pH dependence of FRAP was carried out primarily in rat sensory ganglion and spinal cord; the enzyme was found to hydrolyze a wide variety of phosphomonoesters in a relatively nonselective manner at both pH 5 and pH 7, including 5′-nucleotides, phosphorylated amino acids, and several exogenous compounds.

The visualization of FRAP-like activity in several nonrodent species is discussed with reference to previous work indicating its presence only in mouse and rat. Technical factors are considered that limit the applicability of the lead-ion histochemical method in demonstration of FRAP and in efforts at functional characterization of the enzyme, especially in light of its ability to hydrolyze a broad spectrum of substrates over a wide pH range. Alternative interpretations of the expression of acid phosphatase activity in a select class of small sensory ganglion cells are suggested, including several possible nonsynaptic roles of FRAP in the peripheral nervous system.  相似文献

We investigated the physiology that underlies the influence of arbuscular mycorrhizal (AM) colonization on outcomes of interactions between plants. We grew Medicago truncatula A17 and its AM‐defective mutant dmi1 in intragenotypic (two plants per pot of the same genotype, x2) or intergenotypic (one plant of each genotype, 1 + 1) combinations, inoculated or not with Rhizophagus irregularis (formerly Glomus intraradices) or Gigaspora margarita. We measured plant growth, colonization, contributions of AM and direct P uptake pathways using 32P, and expression of plant Pi transporter genes at two levels of P supply. A17 (x2) responded positively to inoculation only at low P. The response was enhanced with 1 + 1 even at high P where colonization in A17 was reduced. With R. irregularis P uptake by the AM pathway was unaffected by P supply, whereas with G. margarita, the AM pathway was lower at high P, and direct uptake higher. Gene expression varied and was unrelated to P uptake through the two pathways. There was no evidence of plant control of P uptake via R. irregularis at high P but there was via G. margarita. Importantly, growth responses of plant genotypes grown alone did not predict outcomes of intergenotypic interactions.  相似文献

Activity-dependent synaptic plasticity has been thought to be a cellular basis of memory and learning. The late phase of long-term potentiation (L-LTP), distinct from the early phase, lasts for up to 6 h and requires de novo synthesis of mRNA and protein. Many LTP-related genes are enhanced in the hippocampus during pentyrenetetrazol (PTZ)- and kainate (KA)-mediated neural activation. In this study, mice were administered intraperitoneal injections of PTZ 10 times, once every 48 h, and showed an increase in seizure indexes. Genes related to plasticity were efficiently induced in the mouse hippocampus. We used a PCR-based cDNA subtraction method to isolate genes that are expressed in the hippocampus of repeatedly PTZ-treated mice. One of these genes, neural activity-related RING finger protein (NARF), encodes a new protein containing a RING finger, B-box zinc finger, coiled-coil (RBCC domain) and beta-propeller (NHL) domain, and is predominantly expressed in the brain, especially in the hippocampus. In addition, KA up-regulated the expression of NARF mRNA in the hippocampus. This increase correlated with the activity of the NMDA receptor. By analysis using GFP-fused NARF, the protein was found to localize in the cytoplasm. Enhanced green fluorescent protein-fused NARF was also localized in the neurites and growth cones in neuronal differentiated P19 cells. The C-terminal beta-propeller domain of NARF interacts with myosin V, which is one of the most abundant myosin isoforms in neurons. The NARF protein increases in hippocampal and cerebellar neurons after PTZ-induced seizure. These observations indicated that NARF expression is enhanced by seizure-related neural activities, and NARF may contribute to the alteration of neural cellular mechanisms along with myosin V.  相似文献

Convergence is widely regarded as compelling evidence for adaptation, often being portrayed as evidence that phenotypic outcomes are predictable from ecology, overriding contingencies of history. However, repeated outcomes may be very rare unless adaptive landscapes are simple, structured by strong ecological and functional constraints. One such constraint may be a limitation on body size because performance often scales with size, allowing species to adapt to challenging functions by modifying only size. When size is constrained, species might adapt by changing shape; convergent shapes may therefore be common when size is limiting and functions are challenging. We examine the roles of size and diet as determinants of jaw shape in Sciuridae. As expected, size and diet have significant interdependent effects on jaw shape and ecomorphological convergence is rare, typically involving demanding diets and limiting sizes. More surprising is morphological without ecological convergence, which is equally common between and within dietary classes. Those cases, like rare ecomorphological convergence, may be consequences of evolving on an adaptive landscape shaped by many‐to‐many relationships between ecology and function, many‐to‐one relationships between form and performance, and one‐to‐many relationships between functionally versatile morphologies and ecology. On complex adaptive landscapes, ecological selection can yield different outcomes.  相似文献

Cattleya coccinea and C. brevipedunculata (Orchidaceae) are closely related species distinguished primarily by geographical distribution, vegetative morphology and flowering period. Both species inhabit high‐elevation regions in south‐eastern Brazil, but are traditionally associated with different habitats, located in cloudy forests and campos rupestres (rocky fields), respectively. We used morphometrics and genetic variation of microsatellite markers to test the occurrence of a hybrid zone between these species located in Parque Estadual do Ibitipoca (PEI), Brazil. Morphological data reveal a continuum of variation between the putative taxa, influenced mainly by characters of leaf, pseudobulb and peduncle. However, genetic data do not support the occurrence of hybridization and introgression in PEI, showing that it is a pure population of C. brevipedunculata. Differences in vegetative characters among individuals from cloudy forests and campos rupestres suggest that morphological variation may be related to phenotypic plasticity in response to environmental light fluctuations, an unknown situation for this species. These results highlight the inconsistency of morphology for the identification of hybrids and the role of vegetative characters as a possible complicating factor for the taxonomy of these species, as they are subject to environmental influence.  相似文献

1 Competitive interactions among two specialist predators, Laricobius nigrinus and Sasajiscymnus (Pseudoscymnus) tsugae, and a generalist predator, Harmonia axyridis Pallas, of hemlock woolly adelgid, Adelges tsugae were evaluated in hemlock stands in south‐western Virginia. The two specialist predators are part of a biological control program for A. tsugae, and the potential for competition among these species and previously established generalist predators in the field is unknown. 2 Adult predators were evaluated in branch cages during spring and summer at two field sites infested with A. tsugae. Using females only in 2003 and sexual pairs in 2004, predator survival and net reproduction were examined, as well as their feeding and impact on A. tsugae when present alone and in conspecific and heterospecific groupings. 3 Predator survival was not affected by the presence of additional predators. Total feeding was greater for all species when placed in predator groupings, suggesting that interactions do not significantly interfere with feeding activity. Net reproduction per predator was negatively affected by conspecifics, but unaffected by heterospecifics, indicating that direct or indirect intraspecific interference may occur. In spring, L. nigrinus showed the greatest impact on A. tsugae, and H. axyridis had the greatest impact during summer. 4 These results suggest that it would be beneficial to utilize multiple predator species combinations over single species when implementing biological control for A. tsugae. Low‐density releases are also recommended to reduce intraspecific interference.  相似文献

AIMS: To investigate the occurrence and extent of Saccharomyces cerevisiae and Oenococcus oeni interactions. METHODS AND RESULTS: Interactions between S. cerevisiae and O. oeni were investigated by double-layer and well-plate assays showing the occurrence of specific interactions for each yeast-malolactic bacteria (MLB) coupling. Heat and protease treatments of synthetic grape juice fermented by the S. cerevisiae strain F63 indicated that the inhibitory activity exerted by this yeast on O. oeni is due to a proteinaceous factor(s) which exerts either bacteriostatic or bactericidal effect depending on concentration and affects malolactic fermentation in natural grape juice and wine. CONCLUSIONS: A proteinaceous factor(s) produced by a S. cerevisiae wine strain able to inhibit O. oeni growth and malic acid fermentation was characterized. SIGNIFICANCE AND IMPACT OF THE STUDY: The individuation, characterization and exploitation of yeast proteinaceous factor(s) exerting inhibitory activity on MLB may offer new opportunities for the management of malolactic fermentation.  相似文献
