The roles of preconditioning and coiling upon entrance into anhydrobiosis by Aphelenchus avenae were tested via video-assisted analysis at 252°C. Fourth-stage juveniles or young adults of A. avenae were individually placed on 5% agar containing 0.8 M sucrose. Nematodes became quiescent within 3 hr, then gradually resumed a low level of activity and assumed a coiled posture. High desiccation survival rate was recorded when nematodes were incubated on agar for more than 6 hr; the survival rates were 0%, 3%, 73%, and 92% for 0, 2, 6, and 12 hr on agar, respectively. All nematodes placed on agar for 24 hr or more revived after rehydration following desiccation. Once nematodes were on agar for a sufficient time, no difference in desiccation survival was observed between nematodes taking a coiled posture and those uncoiled artificially. Based on these results, exposure to osmotic stress for 6 hr can prepare A. aveae physiologically for anhydrobiosis, but coiling does not appear to be a physiological requirement for desiccation in survival. 相似文献

The effect of methyl bromide (MB) was tested on active and anhydrobiotic Aphelenchus avenae. A. avenae was induced into anhydrobiosis by three different techniques. Both active and anhydrobiotic nematodes were subjected to 3,000 μ1 MB/liter air for 14 periods from 0 to 82 h. Anhydrobiotic nematodes were more resistant to fumigation than active nematodes, regardless of the technique used to induce anhydrobiosis. The percent survival decreased with increasing MB exposures (μ1 MB × h). For an LD₉₅ of 45,000-54,000 μ1/1 × h were required for active nematodes and >279,000 μ1/1 × h for anhydrobiotic nematodes. 相似文献

The onset of molting in all stages of Hemicycliophora arenaria was preceded by the appearance of numerous, discrete globular structures which were termed "molting bodies" because they were present in the hypodermis only during the production of the new cuticle. In all parasitic stages the molt commenced with the separation of the cuticle from the hypodermis from which the new sheath and cuticle were differentiated. Following completion of the new sheath and cuticle most of the old outer covering was apparently absorbed before ecdysis. Electronmicrographs of body wall cross sections in molting L4 male specimens revealed the final molt to be a double molt in which an additional sixth cuticle was produced. Since both a new sheath and cuticle were produced during the molt of each stage, the sheath must be considered as an integral part of the cuticle and not as a residual cuticle or the result of an incomplete additional molt. Molting in Aphelenchus avenae and Hirschmanniella gracilis was less complex and "molting bodies" were not observed. After cuticle separation the hypodermis gave rise to a new trilaminate zone, the future cortex, and (later) the matrix and striated basal layers. 相似文献

Ultrathin sections of all parasitic stages of Hemicycliophora arenaria revealed two major divisions in the body covering. The outermost was a seven-layered sheath and the innermost a five-layered cuticle comprising three zones; an outer trilaminate cortex, a fibrillar matrix and a striated basal layer. The body covering of the nonparasitic males also exhibited two major divisions: the outer, a relatively thin four-layered sheath and the inner, a six-layered cuticle consisting of three zones; an outer trilaminate cortex, a two-layered matrix and a striated basal layer. The cuticles of all stages of Aphelenchus avenae were similar, consisting of five layers divisible into three zones; an outer trilaminate cortex, a fibrillar matrix and a striated basal layer. Starving Aphelenchus avenae survived 3-4 weeks in microaerobic and anaerobic environments, but Caenorhabditis sp. survived less than 80 hr. Aerobically, both nematodes metabolize neutral lipid reserves: there was no microaerobic ( <5% O₂) or anaerobic neutral lipid catabolism. Early in anaerobiosis both nematodes utilized endogenous glycogen. Caenorhabditis sp. depleted the glycogen and died. A. avenae under oxygen stress longer than 120 hr entered cryptobiosis, during which there was neither measurable O₂ uptake nor glycogen or neutral lipid utilization, Only when re-aerated, did A. avenae recover and resume "''normal" metabolism. 相似文献

Reproduction of Aphelenchus avenae, reared on Rhizoctonia solani growing on steamed wheat seeds and Caenorhabditis sp., reared on a mixed bacterial culture grown on oatmeal, was significantly reduced at 5% oxygen and inhibited at 4% oxygen and below. Aeration ranging from atmospheric air (21%) to 10% oxygen had no effect on reproduction. Close interval (5 days or less) fluctuations, between high and low oxygen concentrations, inhibited population buildup of Hemicycliophora arenaria on tomato in soil, and of A. avenae and Caenorhabditis sp. in vitro. In soil tests with H. arenaria exposed to 12 hr of nitrogen every three days (in air) inhibited the rate of buildup compared to controls maintained in continuous air. With the in vitro studies, as little as 4 hr nitrogen every 3 days (stored in air) significantly influenced the population numbers. 相似文献

In greenhouse experiments, massive application of the fungivorous nematode, Aphelenchus avenae, in summer at 26-33 C (1 x l0⁵ nematodes/500 cm³ autoclaved soil) or in autumn at 18-23 C (5 x 10⁴ nematodes/500 cm³ autoclaved soil) suppressed pre-emergence damping-off of cucumber seedlings due to Rhizoctonia solani AG-4 by 67% or 87%, respectively. Application of 2 x l0⁵ A. avenae to sterilized soil infested with R. solani caused leafminer-like symptom on the cotyledons, which did not occur in mixed inoculations with the entomopathogenic nematode, Steinernema carpocapsae. When 1 x 10⁶ A. avenae were applied 3 days before inoculation with 100 Meloidogyne incognita juveniles, gall numbers on tomato roots were reduced to 50% of controls. Gall numbers also were suppressed by S. carpocapsae (str. All). Reduction in gall numbers was no greater with mixed application of A. avenae and S. carpocapsae than with application of single species, even though twice the number of nematodes were added in the former case. These nematodes were positively attracted to tomato root tips. Aphelenchus avenae suppressed infection of the turnip moth, Agrotis segetum, but not the common cutworm, Spodoptera litura, by S. carpocapsae. 相似文献

Carbamate and Organophosphorus Nematicides: Acetylcholinesterase inhibition
and Effects on Dispersal
The sensitivities of acetylcholinesterases (ACHE) from the fungus-feeder Aphelenchus avenae and the plant-parasitic species Helicotylenchus dihystera and Pratylenchus penetrans and the housefly, Musca domestica, were compared using a radiometric assay which utilized H³ acetylcholine as a substrate. Nematode ACHE were generally less sensitive to inhibition by organophosphorns and carbamate pesticides than were ACHE from the housefly. ACHE from the plant-parasitic species and A. avenae were generally similar in sensitivity. In soil, carbamates were more toxic than the organophosphorus pesticides to A. avenae. All pesticides tested affected nematode movement, but fenamiphos was more inhibitory than others. The effects on dispersal of nematodes may be an important mechanism in control by some nematicides. 相似文献

Exposure to carbofuran and fenamiphos for 72 hours reduced the numbers of active Aphelenchus avenae in aqueous suspension by > 75%. When nematicides were removed, many A. avenae exposed to carbofuran resumed normal movement but A. avenae treated with fenamiphos did not recover. Acetylcholinesterase (AChE) activity was suppressed by > 95% in nematodes treated with carbofuran or fenamiphos. However, 48 hours after treated nematodes had been placed in water, AChE activity in carbofuran treated populations was 98% of the levels in control nematodes. Nematodes that had been treated with fenamiphos showed only slight AChE recovery. The antidotes, atropine sulfate and 2-PAM, were largely ineffective in counteracting the toxic effects of the nematicides. 相似文献

The sensitivities of acetylcholinesterases (ACHE) from the nematode Aphelenchus avenae and the house fly Musca domestica to various pesticides were compared using a colorimetric assay. ACHE from A. avenae were generally less sensitive than ACHE from M. domestica to inhibition by organophosphorous and carbamate pesticides. Carbamates were somewhat more inhibiting than organophosphorous pesticides to nematode ACHE. In vivo tests with concentrations of various pesticides up to 500 ppm in sand caused less than 100% mortality of nematodes. 相似文献

Although the ability of plant-parasitic nematodes to survive in a dehydrated or anhydrobiotic state for long periods of time has been well documented, the ability of free-living nematodes has not. Here we report on the survival of a free-living nematode, Panagrolaimus sp., from Armenia in the anhydrobiotic state for 8.7 years. This Panagrolaimus sp. can be cultured and maintained readily and may provide a good system for studying anhydrobiosis in nematodes. 相似文献

The influence of soil temperature and moisture on Meloidogyne incognita (Kofoid and White) Chitwood was examined in relation to hatching and survival of second-stage juveniles (J2). Nematodes were cultured on cotton (Gossypium hirsutum L. cv. Acala SJ2) under field conditions to provide populations similar to those found in the field in late autumn. Egg masses were placed in a temperature range (9-12 C and 21 C), and hatch was measured over a period equivalent to 20 degree days > 10 C (DD10). Hatch occurred below the reported 18 C activity threshold, was restricted below 12 C, and was inhibited below 10 C. Soil moisture influence on hatch was measured by placing egg masses in Hesperia sandy loam and subjecting them to suction pressures ranging from -1.1 bars to -4 .5 bars. Suction potentials of less than -2 bars reduced hatch and less than -3 bars inhibited hatch. J2 were placed in sandy loam soil with soil moisture near field capacity, and their motility was measured over a period of 500 DD10. In the absence of a host, more than 90% of J2 became nonmotile over this period. 相似文献

Disc-electrophoretic separation of soluble proteins from whole nematode homogenates yielded band profiles useful for distinguishing selected species of Meloidogyne and Ditylenchus, and the genera Heterodera, and Aphelenchus. Certain protein bands were common to all the species of Meloidogyne, whereas other bands were specific. Meloidogyne spp. and Heterodera glycines shared some protein similarities, but other genera differed distinctly. In a series of microcosm experiments with an arable, sandy loam soil amended with sugarbeet leaf, the short-term (8 weeks) dynamics of numbers of nematodes were measured in untreated soil and in γ-irradiated soil inoculated with either a field population of soil microorganisms and nematodes or a mixed population of laboratory-propagated bacterivorous nematode species. Sugarbeet leaf stimulated an increase in bacterivorous Rhabditidae, Cephalobidae, and a lab-cultivated Panagrolaimus sp. Differences were observed between the growth rates of the nematode population in untreated and γ-irradiated soils, which were caused by two nematophagous fungi, Arthrobotrys oligospora and Dactylaria sp. These fungi lowered the increase in nematode numbers due to the organic enrichment in the untreated soil. We estimated the annually produced bacterivous nematodes to consume 50 kg carbon and 10 kg nitrogen per ha, per year, in the upper, plowed 25 cm of arable soil. 相似文献

Soil populations of plant-parasitic nematodes were monitored bimonthly for 18 months in irrigated and nonirrigated corn plantings using four production systems: conventional and minimum tillage with crop residue returned and minimum tillage with 60% or 90% of previous corn crop residue removed. Populations of Meloidogyne incognita, Scutellonema brachyurum, Pratylenchus scribneri, and Paratrichodorus christiei varied among the tillage, nematicide, and irrigation treatments. Meloidogyne incognita and P. christiei populations were not significantly affected by tillage method, but S. brachyurum populations were highest during April 1981 and 1982 in minimum tillage treatments where crop debris was not removed. In contrast, S. brachyurum populations were lowest during the same period in minimum tillage plots where 90% of previous crop debris had been removed or where residues were incorporated with conventional tillage. Populations of P. scribneri were lowest in minimum tillage during August 1981 and April 1982. Regardless of tillage system, corn yields in all nonirrigated plots were increased during 1982 by application of carbofuran (2.24 kg a.i./ha). The behavior of a California isolate of the predaceous nematode, Odontopharynx longicaudata de Man, was studied in water agar culture. When feeding on an Acrobeloides sp. the predator completed its life cycle in 13 to 14 days at 25 C. Optimum temperature for reproduction of the predator was 25 C, few individuals survived at 10 C, and 30 C was lethal. Males were necessary for reproduction, and at 25 C the sex ratio was about 1:1. All postembryonic stages were voracious feeders. A single female predator consumed 30 individuals of another Acrobeloides sp. in 1.5 days. Juveniles must feed in order to complete their development. Three modes of feeding were observed depending on the prey selected. A high degree of prey selectivity occurred; 6 of 17 nematode prey species were readily consumed by the predator, but there was little or no feeding on the remaining 11 species. Predation percentage varied with prey species. Consumption of Anguina pacificae and the two Acrobeloides spp. was almost 100%, consumption of A. amsinckiae, Pratylenchus vulnus, and Trichodorus sp. was 70-78%. Difference in final predator population densities was obtained after feeding on the two species of Acrobeloides. Soils with specific suppressiveness to plant-parasitic nematodes are of interest to define the mechanisms that regulate population density. Suppressive soils prevent nematodes from establishing and from causing disease, and they diminish disease severity after initial nematode damage in continuous culturing of a host. A range of non-specific and specific soil treatments, followed by infestation with a target nematode, have been employed to identify nematode-suppressive soils. Biocidal treatments, soil transfer tests, and baiting approaches together with observations of the plant-parasitic nematode in the root zone of susceptible host plants have improved the understanding of nematode-suppressive soils. Techniques to demonstrate specific soil suppressiveness against plant-parasitic nematodes are compared in this review. The overlap of studies on soil suppressiveness with recent advances in soil health and quality is briefly discussed. The emphasis is on methods (or criteria) used to detect and identify soils that maintain specific soil suppressiveness to plant-parasitic nematodes. While biocidal treatments can detect general and specific soil suppressiveness, soil transfer studies, by definition, apply only to specific soil suppressiveness. Finally, potential strategies to exploit suppressive soils are presented. 相似文献

A drip irrigation delivery system was used to infest field sites with the plant-parasitic root-knot nematodes, Meloidogyne incognita. Juvenile or egg inocula passed through the system without blockage of emitters or harm to the nematodes. Field sites so infested were available for experimentation. Delivery of approximately 5 x 10⁴ to 10⁵ juveniles or 10⁵ to 3 x 10⁵ eggs per emitter through the drip system resulted in heavy root galling of tomatoes planted next to the drip emitters. Nematodes feeding on bacteria (Acrobeloides sp.) and on fungi (Deladenus durus) also were successfully applied through the drip system. This method has potential for uniformly infesting experimental sites with plant-parasitic or entomogenous nematodes and for manipulation of nematode community structure for soil ecological studies. 相似文献

Aphelenchoides fragariae was isolated from the phylloclades of the ornamental plant Ruscus hypophyllum (Liliaceae). Rotylenchus buxophilus, Scutellonema brachyurum, and Meloidogyne were identified as the most common plant-parasitic nematodes in the soil near the roots. The pathology and life history of A. fragariae were closely related to the climate. To our knowledge, this is the first report of R. hypophyllum as a host of plant-parasitic nematodes. 相似文献

Various fluorescent conjugated lectins have been used for the detection of glycoconjugates on nematode surfaces under light microscopy. Several problems have been experienced with these reagents including penetration of the cuticle by fluorescent lectins, non-glycoconjugate specificity, strong nematode autofluorescence at the emission wavelength of the fluorescent dye, and prevention of persistent visualization due to rapid quenching of the fluorescent components. Gold-conjugated reagents combined with silver enhancement alleviated these difficulties when working with three phytonematode species (Heterodera avenae, H. latipons, and Meloidogyne javanica) and two entomopathogenic species (Steinernema carpocapsae and S. glaseri) under light-microscopy visualization of binding by fluorescent lectins and neoglycoproteins.
