Comparison of the effects of auxin and fusicoccin on phosphate absorption by aged potato tuber discs
Auxin (IAA, 5 × 10−5 M ) partially prevents the increase in the rate of phosphate uptake during ageing of potato tuber discs ( Solatium tuberosum L. cv. Bintje), whereas fusicoccin (FC, 10−5 M) stimulates it. After the development of enhanced phosphate transport capacity, the response to fusicoccin is greater than with fresh discs. Complementary experiments on K+ (86 Rb) absorption show that FC also slightly enhances the rate of K+ uptake, while IAA has no much effect. It is suggested that IAA acts specifically on the development of a mechanism which occurs during the ageing period, while FC action may be more directly linked to the system of phosphate transport itself. 相似文献

Escherichia coli accumulates K+ by means of multiple uptake systems of which Kup is the major transport system at acidic pH. In cells grown under fermentative conditions at pH 5.5, K+ influx by a wild-type strain upon hyper-osmotic stress at pH 5.5 was accompanied by a marked decrease in H+ efflux, with a 1:1 ratio of K+ to H+ fluxes. This was observed with cells treated with N,N'-dicyclohexylcarbodiimide. Similar results with a mutant defective in Kdp and TrkA but with a functional Kup system but not in a mutant defective in Kdp and Kup but having an active TrkA system suggest that Kup operates as a H+ -K+ -symporter. 相似文献

The influx of Rb+ into the roots of two barley varieties (Hordeum vulgare L. cv. Salve and cv. Ingrid) from a K+-free 86Rb-labelled nutrient solution with 2.0 mM Rb+, was checked at intervals from day 6 to day 18. The control plants were continuously grown in complete nutrient solution containing 5.0 mM K+, while two other groups of plants were grown in K+-free nutrient solution starting on day 6 and between day 6 and day 9, respectively. The pattern of Rb+ influx was similar for both varieties, although their efficiencies in absorbing Rb+ were different. The relationship between Rb+ influx and K+ concentration of the root could be interpreted in terms of negative feedback through allosteric control of uptake across the plasmalemma of the root cells. Hill plots were bimodal, but in the opposite direction. The Hill coefficients, reflecting the minimum number of interacting allosteric binding sites for K+ (Rb+), were low (≤–3.0). It is discussed whether the threshold value, that is the breaking point in the Hill plot, is indicative of a changed efficiency of transporting units for K+ (Rb+) transport to the xylem. Moreover, feedback regulation might be involved in transport of K+ between root and shoot. The variation in K+ concentrations in the roots and shoots of control plants were cyclic but in phase opposition despite an exponential growth. A smooth colony strain, resistant to phages λ and P22, was isolated from sewage and identified as Escherichia coli (strain H). Four temperate phages plaquing on strain H were isolated from sewage. The archetype, HK620, does not plaque on strains C and K12 of E. coli nor on the LT2 strain of Salmonella enterica. Bacterial mutants resistant to a clear plaque mutant of HK620 produce rough colonies. Some are also galactose-negative, a few are histidine auxotrophs, and most show sensitivity to λ. HK620 can transduce a wide variety of auxotrophic mutants of E. coli H to prototrophy. It can recombine with λ but its virions resemble those of P22. 相似文献

Aerobic growth of Escherichia coli sodAsodB and katE mutants lacking cytosolic superoxide dismutases and catalase hydroperoxidase II was inhibited by osmotic upshift to a greater extent than of their wild-type parent strains. The fur mutation leading to an intracellular overload of iron also increased sensitivity of growing E. coli cells to osmotic upshift. Using lacZ fusions, it was shown that expression of antioxidant genes soxS and katE was stimulated by an increase in osmolarity. These data suggest that in aerobically growing E. coli cells, moderate osmotic upshift causes activation of certain antioxidant systems. 相似文献

Bovine serum albumin (BSA) enhances nicotinic agonist-induced (86)Rb+ efflux from synaptosomal fractions of the mouse thalamus, but how it does so is not understood. The experiments reported here indicated that BSA enhancement of nicotinic acetylcholine receptor function was rapid, reversible, depended on BSA concentration, and occurred at all points of the nicotinic agonist concentration-response curve. We hypothesized that BSA-extractable compounds, such as long-chain fatty acids, were responsible for inhibiting nicotinic responses in the absence of BSA. The hypothesis was tested by applying arachidonic, linolenic, or oleic acids in the absence of BSA after an initial prewash with BSA. All three fatty acids exhibited a rapid, concentration-dependent inhibition of nicotinic-agonist stimulated ion flux. Concentration-response curves produced after 30 s of pre-treatment with arachidonic acid were similar to those seen when BSA was completely absent. The effects of pre-treatment were reversed immediately by the introduction of BSA. Furthermore, no effects of fatty acids were observed when preparations were continuously exposed to BSA or when BSA was continuously absent. These results suggest that the removal of endogenous, inhibitory compounds is largely responsible for the rapid, potentiating action of BSA at nicotinic acetylcholine receptors expressed in the mouse thalamus. 相似文献

The survival of Leishmania, which encounter drastic changes of environment during their life-cycle, requires regulation and control of ionic concentrations within the cell. We analysed the influence of growth stage, ionic composition of the medium, heat and acidic stress on 86Rb+ influx in L. infantum promastigetes. Proliferating promastigotes exibited faster and higher 86Rb+ uptake than stationary cells. Cl− anion did not have any effect, but in the presence of physiological concentration of HCO3−, 86Rb+ uptake was significantly increased. This enhancing effect was only partially inhibited by N,N′-dicyclohexylcarbodiimide (DCCD), a blocker of ion-translocating ATPases. 86Rb+ influx was abolished by N-ethylmaleimide (NEM), indicating a major contribution of plasma membrane transporters. Heat shock and acidic shock notably decreased 86Rb+ influx. Our data provide indirect evidence that an energy-dependent system which brings K+ in, such as K+/H+-ATPase evidenced by Jiang et al. (1994), is active in Leishmania in different environments. Mechanism(s) other than ion-translocating ATPase occur, at least in the presence of HCO3−, and their contribution to K+ pathways varies in different environmental conditions. 相似文献

We characterized two Shiga toxin-producing Escherichia coli (STEC) O86:HNM isolates from a patient with hemolytic uremic syndrome (HUS) or bloody diarrhea. Both of them did not possess the eaeA gene. However, the isolate from a HUS patient carried genetic markers of enteroaggregative E. coli (EAEC) and showed aggregative adherence pattern to HEp-2 cells. The other isolate from bloody diarrhea, which was negative with EAEC markers, was diffusely adhered to HEp-2 cells. The stx2 gene in both E. coli O86:HNM strains was encoded in each infectious phage, which was partially homologous to that of strain EDL933, a STEC O157:H7. These results will help to explain the genotypic divergences of STEC. 相似文献

In this article the existence of A1 adenosine receptors and the absence of A2 adenosine receptors in myelin membranes purified from pig brain white matter are demonstrated. The characterization of (R)-[3H]phenylisopropyladenosine ([3H]R-PIA) binding to purified myelin fractions was performed. The distribution of high- and low-affinity species of the A1 adenosine receptor was different in heavy, medium, and light myelin. The fluidity of myelin subfractions and of pig brain cortical membranes was estimated; the microviscosity of heavy myelin (5.4 poises) and of cortical membranes (5.1 poises) was similar and less than that of medium (7.8 poises) and light (8.2 poises) myelin. It was also demonstrated that the agonist R-PIA modifies the microviscosity of myelin membranes and that the degree of modification depends on the fluidity of the membrane assayed. These results suggest that adenosine receptors may have an important role in the functionality of myelin membranes. 相似文献

Mini-Tn10luxABcam/Ptac-ATS was constructed in order to develop a luciferase-transducing bacteriophage for detecting Escherichia coli O157:H7. The transposon was designed to deliver a 3.6-kb insertion that confers n-decanal-dependent bioluminescence and resistance to chloramphenicol and was constructed using mini-Tn10cam/Ptac-ATS in the plasmid pNK2884 and luxAB from Vibrio harveyi. PhiV10, a temperate bacteriophage infecting common phage types of Escherichia coli O157:H7, was mutagenized as a prophage in E. coli O157:H7 strain R508. PhiV10::luxABcamA1-23 was rescued from the strain by propagating it on a strain lacking the bacteriophage and the vector containing the transposon. The bacteriophage transduced n-decanal-dependent bioluminescence to E. coli O157:H7 strain R508 that was measurable approximately 1 h post infection. 相似文献

Bacteriophages that carry the Shiga toxin gene (stx) represent an additional hazard in cattle manure-based fertilizers in that their survival could lead to toxigenic conversion of Escherichia coli and other bacteria post-composting. A Stx-phage in which the Shiga toxin (stx(2)) gene was inactivated by insertion of a chloramphenicol resistance gene was used in combination with a rifampicin-resistant E. coli host where RecA is constitutively activated so that all infectious phage particles could be enumerated by plaque assay. PCR-based confirmation methods and the additional application of a host enrichment protocol ensured that very low numbers of surviving bacteriophage could be detected and unequivocally identified. Stx-bacteriophage numbers declined rapidly over the first 48 h and none could be detected after 3 days. The host enrichment method was applied after 6 days and no bacteriophages were recovered. While addition of fresh E. coli cells at intervals after the compost temperature had reduced below 40 degrees C demonstrated that E. coli growth could be supported in the compost, Stx-phages or their lysogens were never detected. Here, we demonstrate that composting animal manure for 40 days during which a temperature of >60 degrees C is maintained for at least 5 days is effective at removing both E. coli and a model infectious Stx-encoding bacteriophage. 相似文献

The effects of pH on the growth and the K+ (86 Rb) uptake and K+ content of excised rice ( Oryza sativa L. cv. Dunghan Shali) and wheat ( Triticum aestivum L. cv. GK Szeged) roots were investigated. Rice roots responded to H+ stress with an increased K+ (86 Rb) influx and a decreased K+ content, suggesting an increased exchange between the cytoplasmic K+ pool and the external medium. Under the same experimental conditions wheat did not show any anomalous K+ (86 Rb) influx. Growth of both rice and wheat was relatively insensitive to pH between 4 to 10. 相似文献

目的了解鹤岗市人民医院2007年1月至2009年12月泌尿系病原菌菌株分布,分析抗菌药的敏感情况,为临床合理应用抗菌药提供实验室依据。方法 2007年1月至2009年12月该院泌尿系感染者2375例中段尿标本分离出776株病原菌进行鉴定和体外药物敏感试验,采用法国生物梅里埃公司ATB鉴定系统对菌株进行鉴定,采用纸片扩散法(即K-B法)进行抗菌药敏感性试验。结果在泌尿系感染大肠埃希菌和肠球菌的检出率分别为40.3%和19.7%;革兰阴性菌对亚胺培南敏感性较高,对氨苄西林、头孢唑林耐药率较高。革兰阳性菌对万古霉素全部敏感,对氨苄西林、青霉素耐药率较高。真菌检出率为16.1%,真菌对两性霉素、氟康唑敏感性高。结论泌尿系感染以大肠埃希菌和粪肠球菌为主,耐药呈上升趋势。临床医师应严格依照泌尿道感染致病菌的种类及药敏结果合理应用抗菌药物。 相似文献

Translocation of labeled potassium (K+ ) from the root to the ear and its distribution within the culm during 4, 8 and 12 h of uptake was studied in intact wheat plants ( Triticum aestivum L. cv. Kolibri) 3 and 5 weeks after anthesis at 0.5 and 5.0 m M K+ concentration in the uptake solution. Uptake of labeled K+ into the shoot was proportional to the K+ concentration applied. After 4 h of uptake about 2% and after 12 h about 7% of labeled K+ applied to the roots were taken up into the shoot at both K+ concentrations. After 12 h of uptake only 6% of the total label in the culm had reached the ear, while about 40% of the label was found in the upper three internodes. In spite of an increasing concentration of labeled K+ during 12 h in the uppermost internode (peduncle), translocation of K+ into the rachis was low. The low and uniform K+ content found generally in grain dry weight seems therefore to be due to a controlled K+ supply to the ear. 相似文献

The protective effects of egg-yolk antibodies obtained from hens immunized with fimbrial antigens from a local strain (Escherichia coli K88+ MB, Manitoba, Canada) of K88+ piliated enterotoxigenic E. coli (ETEC) were evaluated in 3- and 21-day-old piglets in which ETEC diarrhea was induced and also in early-weaned piglets in a commercial farm. The results demonstrated that the E. coli K88+ MB-induced diarrhea in 3-day-old piglets was cured 24 h after treating with egg-yolk antibodies while those treated with egg-yolk powder from conventional hens continued to have diarrhea and 62.5% of them died of severe diarrhea. For 21-day-old weaned piglets, those fed egg-yolk antibodies had transient diarrhea, positive body weight gains and 100% survival during the period of the experiment, whereas control piglets that were treated with placebo had severe diarrhea and dehydration and some died within 48 h after infection. In the field trial, the incidence and severity of diarrhea of 14-18-day-old weaned piglets fed egg-yolk antibodies were much lower than in those fed a commercial diet containing an antibiotic. These results indicate that the neonatal and early-weaned piglets that received the egg-yolk antibodies were protected against ETEC infection. 相似文献

Interactive effects of K+ and N (principally NH4+) on plant growth and ion uptake were investigated using hydroponically grown rice (Oryza sativa L. cv. M202) seedlings by varying the availability of NH4+ or NO3? and K+ during an 18d growth period, a 3d pretreatment period and during flux measurements. Plants grew best in media containing 100 mmol m?3 NH4+ and 200mmolm?3 K+ (N100/K200), followed by N2/K200 < N100/K2 < N2/K2. 86Rb+(K+) fluxes were increased by exposure to N during the 18 d growth period and the 3 d of pretreatment, but decreased by the presence of NH4+ during flux measurements. This inhibition was a function of prior N/K provision and the [NH4+]0 present during flux determinations. NH4+ was least inhibitory to 86Rb+(K+) influx in high-N/low-K plants. Pretreatments with K+ failed to stimulate NH4+ uptake, and the presence of K+ in the uptake solutions reduced NH4+ fluxes only in high-N/low-K plants. 相似文献

Seedlings of eleven varieties of barley (Hordeum vulgare L.) showed differences in utilization of K+ from a full nutrient solution containing 3.0 mM K+. The K+ content of both roots and shoots was proportional to the fresh weights and dry weights after a week in the nutrient solution. The K+ use-efficiency ratio, which indicates the efficiency of nutrient utilization (mg dry weight produced per mg K+ absorbed), differed significantly among the varieties. There was no correlation between influx of Rb+ and the content of K+. It is suggested that there are wide varietal differences in such genetically-determined properties as ion influx and efflux and net ion transport to the shoot. Further-more, the influx of Rb+ was closely linked to transpiration, probably due to a variety-specific non-metabolic part of Rb+ influx. Varietal differences in influx of Rb+ were more pronounced in high-K+ roots than in low-K+ roots with maximum rate of Rb+ uptake, but the rank of varieties was the same in each case. – Criteria for the selection of K+ use-efficient varieties of barley are discussed. 相似文献

目的 探讨水-甘油通道蛋白(glycerol protein ficilitator,GlpF)的生理功能及对细菌生长繁殖的影响.方法 将大肠埃希菌接种于等渗透压的液体培养基(1 IM)培养,18h后,突然改变培养液的渗透压,在30、60、120 min时间点检测细菌的A600nm值,RT-PCR分析其GlpF的表达.结果 突然改变渗透压后,大肠埃希菌数量均有不同程度的减少.在1/2 IM组、1/4 IM组细菌的A600nm值与其各自对照组相比,变化并不明显,但其细菌GlpF表达量明显降低.在高渗组,2 IM组的A600nm值与等渗组相比变化不大,而其GlpF表达量明显高于等渗组.结论 在环境渗透压突然改变时,细菌可以通过调控GlpF的表达来实现对细菌细胞内外水份的调节,以维持胞内环境稳定. 相似文献
