It is crucial to the reproducibility of results and their proper interpretation that the conditions under which experiments are carried out be defined with rigour and consistency, in this review we attempt to clarify the differences and interrelationships among steady, balanced and exponential states of culture growth. Basic thermodynamic concepts are used to introduce the idea of steady-state growth in open, biological systems. The classical, sometimes conflicting, definitions of steady-state and balanced growth are presented, and a consistent terminology is proposed. The conditions under which a culture in balanced growth is also in exponential growth and in steady-state growth are indicated. It is pointed out that steady-state growth always implies both balanced and exponential growth, and examples in which the converse does not hold are described. More complex situations are then characterized and the terminology extended accordingly. This leads to the notion of normal growth and growth that can be synchronous or otherwise unbalanced but still reproducible, and to the condition of approximate steady state manifested by growth in batch culture and by asymmetrically dividing cells, which is analysed in some detail.  相似文献

This study investigates subadult growth spurts in a large sample of anthropoid primates, including humans. Analyses of body mass growth curves show that humans are not unique in the expression of female and male body mass growth spurts. Subadult growth spurts are observed in both New World and Old World anthropoid primates and are more common in males than in females. Allometric analyses of growth spurts indicate that many aspects of primate growth spurts are strongly correlated with species size. Small species tend not to exhibit growth spurts. Although male and female scaling patterns for velocity and size measures are comparable, scaling relations of variables that measure the timing of growth spurts differ by sex. These patterns can he related to sexual differences in life histories. Scaling analyses further show that humans do not depart substantially from patterns that describe other anthropoid primates. Thus, in relative terms, human growth spurts are not exceptional compared to this sample of primates. The long absolute delay in the initiation of the human growth spurt may be of substantial evolutionary importance and serves to distinguish humans from other primates. In essence, humans exhibit growth spurts that are comparable to other primates in many respects. However, human growth spurts are shifted to very late absolute ages. © 1996 Wiley-Liss, Inc.  相似文献

The immunological relatedness of human and porcine growth hormones is examined by means of labelled human growth hormone and guinea pig antiserum. 1) Labelled human growth hormone is found in the precipitate after reaction with antiserum against porcine growth hormone. Parallel dilution curves are obtained with antisera against human and porcine growth hormones. 2) After addition of antiserum against porcine growth hormone, all the radioactivity is eluted from Sephadex G-100 with the void volume. 3) The addition of an excess of porcine hormone displaces labelled human growth hormone from antibodies against human growth hormone to the same extent as an excess of non-labelled human growth hormone does. 4) The standard radioimmunoprecipitation curves for porcine and human growth hormones obtained in the assay system for the human hormone are parallel in slope, provided that the human hormone and our preparation of the porcine hormone are introduced at a proportion of 1 to 560. 5) In a double diffusion test in agarose gel layers, with human and porcine growth hormones diffusing against guinea pig anti-porcine serum, cross reaction is observed. The conclusion is drawn that with guinea pig antisera, human and porcine growth hormones behave immunologically in a similar fashion. Labelled human growth hormone seems to have only such immunodeterminants as are also found in porcine growth hormone.  相似文献

Abstract We have measured the growth rates of some ribosomal mutants of Escherichia coli in different growth media. The mutants are a streptomycin resistant (SmR) mutation in rpsL; a partially streptomycin dependent (SmP) mutation in rpsL; a ribosome ambiguity mutant (ram) in rpsD; a ram mutant in rpsE as well as a mutant defective in tRNA modification, mia A. The data show that the growth rates of all mutants are less inhibited in poor media than they are in rich ones. The translation rates and nonsense suppression levels for each mutant are not significantly different in rich and poor media, which shows that the ribosomal mutant phenotypes are maintained under different growth conditions. These results suggest that the degree of growth inhibition for mutants with altered translation machinery is dependent on the growth conditions. In addition, the data suggest that bacteria are able to physiologically compensate for the loss of growth efficiency in such mutants, particularly, under poor growth conditions.  相似文献

The effects of dispersed phase volume and temperature on the batch growth of Candida lipolytica on gas oil are investigated. Growth parameters are presented for two sets of experiments. The shape of growth curves was basically similar to the system composed of n-hexadecane dissolved in dewaxed gas oil, in spite of the complex nature of the substance. All of the batch growth curves exhibited a linear growth region. The rate of linear growth and its length varied with change in dispersed phase volume. The effect of temperature on growth rate was investigated for temperatures ranging from 23°C to 34°C. The results show a smaller activation energy during linear growth than during the early stages of batch growth. These results are analyzed from the viewpoint of growth models presented previously. The results indicate that growth at drop surfaces is important and that segregation effects may be important.  相似文献

Two simple models of the ecology of population growth are described: "exponential" growth with "r -selection," and "logistic" growth, with "K- selection." Various methods for estimating the parameters of these models are presented in detail, along with statistical methods of evaluation and comparison. Also briefly discussed are more complex models of population growth sometimes used by demographers and ecologists. The two simpler models of population growth are then applied, by way of illustration, to two episodes of population growth in protohistoric southwest Iran, dating from 4000–2350 B. C. Growth curves of successive leaves of Populus euramericana (Dode) Guinier 'Robusta' have been determined. With ample supply of water and nutrients the growth of a poplar shoot follows a fixed pattern: an initial logarithmic acceleration phase followed by a stationary phase in which leaves of equal size are produced at a constant rate. Analysis of growth curves of leaves enabled the growth curves of leaf primordia to be predicted. These primordial growth curves are compared to the indirectly determined growth curves of primordia by measuring the lengths of successive leaf primordia in the apex during the stationary phase of growth. The increase in length of successive leaves in the acceleration phase of growth continues for a longer period at high than at low irradiace. The relative growth rates of leaf primordia, leaves and internodes are discussed in terms of shoot growth and phyllotaxis.  相似文献

The growth of cells in culture and in vivo is modulated by different effectors, some of which are called growth factors. This designation is given to polypeptides that have the ability to enhance cellular growth. Other important growth regulatory molecules are the growth inhibitory polypeptides. The balance between stimulatory and inhibitory signals is evidently essential for normal control of cell proliferation. Disturbances of cellular growth thus presumably result from quantitative alterations between stimulatory and inhibitory signals that the cells get from their environment via their cell surface receptors. Thus, either enhanced amounts of stimulatory or decreased inhibitory signals can contribute to augmented, cancerous growth. An important growth regulator appears to be transforming growth factor-beta (TGF beta), which has both stimulatory and inhibitory effects on cells. The significance of growth inhibitors in the regulation of cellular growth and differentiation is becoming an important research field of modern biology.  相似文献

Messenger RNA isolated from chicken pituitaries was used to construct a chicken pituitary cDNA library. A chicken growth hormone cDNA clone was isolated using 32P-labeled mammalian growth hormone cDNA probes. The amino acid sequence (derived from the DNA sequence) of the mature form of chicken growth hormone shows 77% homology with that of bovine growth hormone. The chicken growth hormone cDNA clone was used to generate a vector capable of producing chicken growth hormone in Escherichia coli. The recombinant E. coli-derived chicken growth hormone was similar to pituitary chicken growth hormone in several biochemical and immunological properties. The recombinant-derived hormone has been used to establish a sensitive radioimmunoassay for growth hormone determinations made from chicken sera. The chicken growth hormone gene has also been introduced into a retroviral vector capable of establishing productive infections of chicken cells both in in vitro and in vivo. The resulting infections are accompanied by the production of radioimmunoassay-detectable growth hormone. The concentrations of growth hormone in sera of Leghorn chickens infected with the recombinant retrovirus are three- to tenfold higher than in control animals.  相似文献

The vertebrate eye is composed of a variety of tissues that, embryonically, have their derivation from surface ectoderm, neural ectoderm, neural crest, and mesodermal mesenchyme. During development, these different types of cells are subjected to complex processes of induction and suppressive interactions that bring about their final differentiation and arrangement in the fully formed eye. With the changing concept of ocular development, we present a new perspective on the control of morphogenesis at the cellular and molecular levels by growth factors that include fibroblast growth factors, epidermal growth factor, nerve growth factor, platelet-derived growth factor, transforming growth factors, mesodermal growth factors, transferrin, tumor necrosis factor, neuronotrophic factors, angiogenic factors, and antiangiogenic factors. Growth factors, especially transforming growth factor-beta, have a crucial role in directing the migration and developmental patterns of the cranial neural-crest cells that contribute extensively to the structures of the eye. Some growth factors also exert an effect on the developing ocular tissues by influencing the synthesis and degradation of the extracellular matrix. The mRNAs for the growth factors that are involved in the earliest aspects of the growth and differentiation of the fertilized egg are supplied from maternal sources until embryonic tissues are able to synthesize them. Subsequently, the developing eye tissues are exposed to both endogenous and exogenous growth factors that are derived from nonocular tissues as well as from embryonic fluids and the systemic circulation. The early interaction between the surface head ectoderm and the underlying chordamesoderm confers a lens-forming bias on the ectoderm; later, the optic vesicle elicits the final phase of determination and enhances differentiation by the lens. After the blood-ocular barrier is established, the internal milieu of the eye is controlled by the interactions among the intraocular tissues; only those growth factors that selectively cross the barrier or that are synthesized by the ocular tissues can influence further development and differentiation of the cells. An understanding of the tissue interactions that are regulated by growth factors could clarify the precise mechanism of normal and abnormal ocular development.  相似文献

This investigation evaluates hypotheses that seek to explain temporal retardation or prolongation of human ontogeny. Current hypotheses that address this issue are poorly defined and conflate several distinct theoretical positions. A model that predicts homogeneity in the extension of human growth periods is evaluated. This model is contrasted with two alternatives. The first alternative predicts heterogeneity in the extension of human growth periods. The second anticipates that human growth prolongation is the result of the uniquely derived “insertion” of a human childhood period into an ancestral ontogenetic trajectory. Allometric analyses of body mass growth data from 21 species of anthropoid primates suggest that human female and male ontogenies often depart from patterns established by other primates, but these departures are not uniformly exceptional. Comparisons imply that derived changes in human growth are heterogeneous. Relative to interspecific expectations, early growth periods are much prolonged, but later growth periods are actually reduced. Moreover, the attributes of early growth periods, including growth rates, timing of growth events, and size-for-age, are highly variable across primates. Low correlations among growth periods suggest independence among growth phases. These analyses highlight minimal distinctions between competing models (heterogeneous extension and insertion hypotheses) that attempt to explain human growth prolongation. More important, the present study facilitates refinements of causal models that have been proposed to explain human growth prolongation. Specifically, human growth prolongation may be related to derived changes in patterns of brain development. Alternatively, metabolic factors may have exerted influences on human ontogeny. However, models that predict long growth periods as a byproduct of metabolic factors do not adequately explain temporal retardation of human ontogeny. Summary The coronar growth of a cidaroid and an aulodont echinoid are investigated by means of tetracycline labelling. The results are compared with earlier investigations on a stirodont and on a camarodont echinoid in order to evaluate the general features of coronar growth. In all echinoids new coronar plates are added at the apical end of the corona throughout the life cycle. The plates are shifted towards the peristome and they grow peripherically.In cidaroids ambulacral (A-) plates are detached from the firm corona. They are transformed into scales covering the peristomial field. The interambulacral (IA-) plates, however, are partially reabsorbed at the peristomial margin. In this manner the oldest solitary interambulacral plates are lost. The subsequent plates are arranged in pairs. The cidaroids thus show interradial growth even at their peristomial margin. This is unique to echinoids.In non-cidaroids there is a perignathic girdle made up of paired ambulacral auricles with interambulacral ridges in between. In some species the ridge is a solitary element. Therefore interradial growth cannot occur in the peristomial margin. In other species the ridge consists of several elements, but it also grows as a whole. Slight resorption of calcite occurs in places at the peristomial margin. In other places, however, calcite is added onto the peristomial edge. In non-cidaroids, therefore, the widening of the peristome is achieved solely by means of lateral growth in the plates bordering the peristome. The shift of the coronar plates from apicad to orad in noncidaroids is a relative shift only.In all echinoids the coronar plates are arranged in meridional columns. All plates grow up to the peristome. Their growth rates are relatively uniform towards the adambulacral sutures (which run between A- and IA-columns). Their growth rates towards the perradius and the interradius respectively are high in younger plates which are positioned above the ambitus, and decrease rapidly in plates located below the ambitus. Near the peristome the interradiad and perradiad growth rates are always considerably lower than adradiad growth rates. Perradial and interradial growth serve to adjust the plates in size and shape to their respective position in the corona.  相似文献

The kinetics of Bacillus thuringiensis growth and its assimilation of nutrient substances were studied under the conditions of batch cultivation in a complex medium containing yeast extract and in a chemically defined medium with amino acids. The growth of B. thuringiensis can be divided into five phases: exponential growth; decelerated growth; stationary phase when protein crystals are formed; stationary phase when spores are formed; lysis of sporangia releasing spores. The first phase may in turn be subdivided into three stages according to changes in the specific growth rate and substrate assimilation: a high specific growth rate and no glucose assimilation; an abrupt drop in mu and the beginning of intensive glucose assimilation from the medium; a new rise in the specific growth rate. As follows from the results of studying the kinetics of B. thuringiensis growth in a chemically defined medium, the above changes in the exponential growth phase are due to the fact that the culture assimilates yeast extract components in the complex medium or amino acids in the chemically defined medium during this phase, and then starts to assimilate glucose and ammonium in the following phases of growth.  相似文献

The adhesion of chick embryo sensory neurons to glass coverslips was examined with interference reflection optics. On untreated glass, adhesive contacts are common only beneath growth cones and are small. On polylysine-treated glass growth cones are highly spread, microspikes reach treat lengths and extensive adhesive contacts underlie growth cones, microspikes and nerve fibers. Veils, expanded from the growth cone, are adherent to the substratum either centrally or laterally, while the extending edge of the cell margin is non-adherent. Linear adhesions are frequent beneath microspikes and pass centrally beneath the growth cone margin. The distribution of linear adhesions resembles that of microfilament bundles seen within whole mounts of growth cones. Adhesive contacts stabilize extensions of the growth cone margin and may influence the organization of the microfilamentous network within the growth cone. Regulation of microfilament organization by adhesion may influence microfilament functions in growth cone mobility and the assembly of neurite structures.  相似文献

Background and Aims Plant growth, the increase of organ dimensions over time, and development, the change in plant structure, are often studied as two separate processes. However, there is structural and functional evidence that these two processes are strongly related. The aim of this study was to investigate the co-ordination between growth and development using mango trees, which have well-defined developmental stages.Methods Developmental stages, determined in an expert way, and organ sizes, determined from objective measurements, were collected during the vegetative growth and flowering phases of two cultivars of mango, Mangifera indica. For a given cultivar and growth unit type (either vegetative or flowering), a multistage model based on absolute growth rate sequences deduced from the measurements was first built, and then growth stages deduced from the model were compared with developmental stages.Key Results Strong matches were obtained between growth stages and developmental stages, leading to a consistent definition of integrative developmental growth stages. The growth stages highlighted growth asynchronisms between two topologically connected organs, namely the vegetative axis and its leaves.Conclusions Integrative developmental growth stages emphasize that developmental stages are closely related to organ growth rates. The results are discussed in terms of the possible physiological processes underlying these stages, including plant hydraulics, biomechanics and carbohydrate partitioning.  相似文献

The developmental mechanisms by which growth is coordinated among developing organs are largely unknown and yet are essential to generate a correctly proportioned adult. In particular, such coordinating mechanisms must be able to accommodate perturbations in the growth of individual organs caused by environmental or developmental stress. By autonomously slowing the growth of the developing wing discs within Drosophila larvae, we show that growing organs are able to signal localized growth perturbation to the other organs in the body and slow their growth also. Growth rate is so tightly coordinated among organs that they all show approximately the same reduction in growth rate as the developing wings, thereby maintaining their correct size relationship relative to one another throughout development. Further, we show that the systemic growth effects of localized growth-perturbation are mediated by ecdysone. Application of ecdysone to larvae with growth-perturbed wing discs rescues the growth rate of other organs in the body, indicating that ecdysone is limiting for their growth, and disrupts the coordination of their growth with growth of the wing discs. Collectively our data demonstrate the existence of a novel growth-coordinating mechanism in Drosophila that synchronizes growth among organs in response to localized growth perturbation.  相似文献

Five-week-old female mice supplemented with estradiol and progesterone are able to respond to epidermal growth factor (EGF) and EGF-like growth factors (alpha-transforming growth factor [alpha-TGF] and crude mammary-derived growth factor) with local lobuloalveolar development when these growth factors are directly introduced into the mammary glands via slow-release cholesterol-based pellets. Contralateral glands receiving pellets containing only cholesterol showed no growth response. The local growth effect is maximal at 4-5 days of exposure to hormones and growth factors. The glands appear to be more sensitive to alpha-TGF than EGF, since local development is seen with one-fifth the level of the former vs. the latter growth factor and can be seen even in the absence of the systemic estrogen/progesterone supplement.  相似文献

The three-stage desk calculation of the von Bertalanffy equation to describe growth in height and weight with age in the elephant is compared with a new approach to calculating the three coefficients in the function by a computer. The two methods give different results with respect to the weight/age calculations. Theoretical von Bertalanffy equations calculated by both methods to describe growth in height and weight with age in the African elephant in Zambia are compared with previously published equations for the elephant in East Africa. Details are given of growth in height in two known-age African elephants, a female ‘Diksie’ and a male ‘Kartoum’. Theoretical growth in height curves for the female African and Asiatic elephant are compared. The coefficients K and / for growth in height are not transferable to the growth in weight equations. Inherent inaccuracies in the calculation of the coefficients in the von Bertalanffy equation are discussed, and it is concluded that in animals which have a long life-span such as the elephant, the equation serves as a purely empirical representation of weight-at-age data and that there is little biological significance in the parameters it contains. The computer-calculated curves give the best fit to the data. The regression of log age on log shoulder height from 2–20 years of age provides a more realistic approach to comparative growth studies. The increase in adrenal weight with age is linear. Tusk growth in relation to age and sex in Zambia is compared with East Africa. It is concluded that the tusks in Zambia are smaller and are more difficult to sex correctly than their East African counterparts, possibly a consequence of the Zambian elephant having a greater degree of tusk wear. Allometric growth is described with emphasis on the estimation of body weight from shoulder height. The most reliable estimates are obtained from a purely empirical representation of the data, a semilog plot of log body weight on shoulder height.  相似文献
