Isotope screening is a simple test for determining the photosynthetic pathway used by plants. The scope of this work was to classify the photosynthetic type of some herbs and medicinal plants through studies of the carbon isotope composition (δ13C). Also, we propose the use of carbon isotope composition as a tool to control the quality of herbs and medicinal plants. For studies of δ13C, δ13C‰ = [R (sample)/R (standard) − 1] × 10−3, dry leaves powdered in cryogenic mill were analyzed in a mass spectrometer coupled with an elemental analyzer for determining the ratio R = 13CO2/12CO2. In investigation of δ13C of 55 species, 23 botanical families, and 44 species possessed a C3 photosynthetic type. Six species found among the botanical families Euphorbiaceae and Poaceae were C4 plants, and 5 species found among the botanical families Agavaceae, Euphorbiaceae, and Liliaceae possessed CAM-type photosynthesis. Carbon isotope composition of plants can be used as quality control of herbs and medicinal plants, allowing the identification of frauds or contaminations. Also, the information about the photosynthetic type found for these plants can help in introducing and cultivating exotic and wild herbs and medicinal plants.  相似文献

Roots of annual crop plants are a major sink for carbon particularly during early, vegetative growth when up to one-half of all assimilated carbon may be translocated belowground. Flowering marks a particularly important change in resource allocation, especially in determinate species, with considerably less allocation to roots and, depending on environmental conditions, there may be insufficient for maintenance. Studies with 14C indicate the rapid transfer belowground of assimilates with typically 50% translocated in young cereal plants of which 50% is respired; exudation/rhizodeposition is generally <5% of the fixed carbon. Root: total plant mass decreases through the season and is affected by soil and atmospheric conditions. Limited water availability increased the allocation of 13C to roots of wheat grown in columns so that at booting 0.38 of shoot C (ignoring shoot respiration) was belowground compared to 0.31 in well-watered plants. Elevated CO2 (700 mol CO2 mol–1 air) increased the proportion of root:total mass by 55% compared with normal concentration, while increasing the air temperature by a mean of 3 °C decreased the proportion from 0.093 in the cool treatment to 0.055 in the warm treatment.  相似文献

Here we describe a simple method for pulse-labeling tree seedlings with 13CO2(gas), and then apply the method in two related experiments: t (i) comparison of carbon allocation patterns between t Betula papyrifera Marsh. and t Pseudotsuga menziesii (Mirb.) Franco, and t (ii) measurement of one-way belowground carbon transfer from t B. papyrifera to t P. menziesii. Intraspecific carbon allocation patterns and interspecific carbon transfer both influence resource allocation, and consequently development, in mixed communities of t B. papyrifera and t P. menziesii.In preparation for the two experiments, we first identified the appropriate 13CO2(gas) pulse-chase regime for labeling seedlings: a range of pulse (100-mL and 200-mL 99 atom%13 CO2(gas)) and chase (0, 3 and 6 d) treatments were applied to one year-old t B. papyrifera and t P. menziesii seedlings. The amount of 13CO2 fixed immediately after 1.5 h exposure was greatest for both t B. papyrifera (40.8 mg excess 13C) and t P. menziesii (22.9 mg excess 13C) with the 200-mL pulse, but higher 13C loss and high sample variability resulted in little difference in excess13 C content between pulse treatments after 3 d for either species. The average excess 13C root/shoot ratio of t B. papyrifera and t P. menziesii changed from 0.00 immediately following the pulse to 0.61 and 0.87 three and six days later, which reflected translocation of 75% of fixed isotope out of foliage within 3 d following the pulse and continued enrichment in fine roots over 6 d. Based on these results, the 100-mL CO2(gas) and 6-d chase were considered appropriate for the carbon allocation and belowground transfer experiments.In the carbon allocation experiment, we found after 6 d that t B. papyrifera allocated 49% (average 9.5 mg) and t P. menziesii 41% (average 5.8 mg) of fixed isotope to roots, of which over 55% occurred in fine roots in both species. Species differences in isotope allocation patterns paralleled differences in tissue biomass distribution. The greater pulse labeling efficiency of t B. papyrifera compared to t P. menziesii was associated with its two-fold and 13- fold greater leaf and whole seedling net photosynthetic rates, respectively, 53% greater biomass, and 35% greater root/shoot ratio.For the carbon transfer experiment, t B. papyrifera and t P. menziesii were grown together in laboratory rootboxes, with their roots intimately mingled. A pulse of 100 mL13 CO2(gas) was applied to paper birch and one-way transfer to neighboring t P. menziesii was measured after 6 d. Of the excess 13C fixed by t B. papyrifera, 4.7% was transferred to neighboring t P. menziesii, which distributed the isotope evenly between roots and shoots. Of the isotope received by t P. menziesii, we estimated that 93% was taken up through belowground pathways, and the remaining 7% taken up by foliage as13 CO2(gas) respired by t B. papyrifera shoots. These two experiments indicate that t B. papyrifera fixes more total carbon and allocates a greater proportion to its root system than does t P. menziesii, giving it a competitive edge in resource gathering; however, below-ground carbon sharing is of sufficient magnitude that it may help ensure co-existence of the two species in mixed communities.  相似文献

A fast-growing normal and a slow-growing gibberellin-deficient mutant of Lycopersicon esculentum (L.) Mill. cv. Moneymaker were used to test the hypothesis that slow-growing plants reduce NO3? in the root to a greater extent than do fast-growing plants. Plants that reduce NO3? in the root may grow more slowly due to the higher energetic and carbon costs associated with root-based NO3? reduction compared to photosynthetically driven shoot NO3? reduction. The plants were grown hydroponically with a complete nutrient solution containing 10 mM NO3? and the biomass production, gas exchange characteristics, root respiratory O2 consumption, nitrate reductase activity and translocation of N in the xylem were measured. The gibberellin-deficient mutants accumulated more total N unit?1 dry weight than did the faster-growing normal plants. There were no significant differences between the genotypes in the rates of photosynthesis expressed on a leaf dry weight basis. The plants differed in the proportion of photosynthetic carbon available to growth due to a greater proportion of daily photo-synthate production being consumed by respiration in the slow-growing genotype. This difference in allocation of carbon was associated with differences in the specific leaf area and specific root length. In addition, a greater leaf weight ratio in the fast-growing than in the slow-growing plants indicates a greater investment of carbon into biomass supporting photosynthetic production in the former. We did not find differences in the activity or distribution of nitrate reductase or in the N composition of the xylem sap between the genotypes. We thus conclude that the growth rate was determined by the efficiency of carbon partitioning and that the site of NO3? reduction and assimilation was not related to the growth rate of these plants.  相似文献

Partitioning of 14C was assessed in sweet chestnut seedlings (Castanea sativa Mill.) grown in ambient and elevated atmospheric [CO2] environments during two vegetative cycles. The seedlings were exposed to 14CO2 atmosphere in both high and low [CO2] environments for a 6-day pulse period under controlled laboratory conditions. Six days after exposure to 14CO2, the plants were harvested, their dry mass and the radioactivity were evaluated. 14C concentration in plant tissues, root-soil system respiratory outputs and soil residues (rhizodeposition) were measured. Root production and rhizodeposition were increased in plants growing in elevated atmospheric [CO2]. When measuring total respiration, i.e. CO2 released from the root/soil system, it is difficult to separate CO2 originating from roots and that coming from the rhizospheric microflora. For this reason a model accounting for kinetics of exudate mineralization was used to estimate respiration of rhizospheric microflora and roots separately. Root activity (respiration and exudation) was increased at the higher atmospheric CO2 concentration. The proportion attributed to root respiration accounted for 70 to 90% of the total respiration. Microbial respiration was related to the amount of organic carbon available in the rhizosphere and showed a seasonal variation dependent upon the balance of root exudation and respiration. Crops of tomorrow are likely to grow under higher levels of atmospheric CO2. Fundamental crop growth processes will be affected and chief among these is carbon allocation. The root to shoot ratio (R:S, defined as dry weight of root biomass divided by dry weight of shoot biomass) depends upon the partitioning of photosynthate which may be influenced by environmental stimuli. Exposure of plant canopies to high CO2 concentration often stimulates the growth of both shoot and root, but the question remains whether elevated atmospheric CO2 concentration will affect roots and shoots of crop plants proportionally. Since elevated CO2 can induce changes in plant structure and function, there may be differences in allocation between root and shoot, at least under some conditions. The effect of elevated atmospheric CO2 on carbon allocation has yet to be fully elucidated, especially in the context of changing resource availability. Herein we review root to shoot allocation as affected by increased concentrations of atmospheric CO2 and provide recommendations for further research. Review of the available literature shows substantial variation in R:S response for crop plants. In many cases (59.5%) R:S increased, in a very few (3.0%) remained unchanged, and in others (37.5%) decreased. The explanation for these differences probably resides in crop type, resource supply, and other experimental factors. Based on stable carbon isotope ratio (δ13C) measurements, photosynthetic pathway types were determined for 61 species in 54 genera and 24 families of flowering plants from the saline meadows of Northeastern China. Of these total vascular plants, 18 species in 17 genera from 6 families were found to have C4 photosynthesis; 43 species in 38 genera from 20 families had C3 photosynthesis. Six dicotyledonous species exhibited C4 pathway, 12 monocotyledonous species were found with C4 photosynthesis. The dicotyledonous C4 species had relative greater mean δ13C value and less total carbon content than both monocotyledonous C4 and C3 species. Most dicotyledonous C4 species were annual forbs and halophytes. Some C4 species had been previously documented, but their δ13C values varied remarkably from those of the present study. Even though there are some fluctuations for the δ13C values of some C4 species, δ13C value was still more reliable for C3 and C4 identification than the use of the enzyme ratio method and of low CO2 compensation concentration.  相似文献

A preliminary study was conducted using the stable isotope 13C to pulse label the cover crop phacelia (Phacelia tanacetifolia) to examine its decomposition in soil, under field conditions. Plants were grown, in pots, in the greenhouse and after four weeks of growth were labelled with 13CO2 six times, at 1–2 week intervals. A single chamber was placed over the pots, and 13CO2 was generated, inside the chamber, by injecting lactic acid into sodium carbonate (99 atom % 13C). For calculating the quantity of Na2CO3 required, a target enrichment of 5 atom% 13C within the shoots of plants, assuming no respiration losses, was used. When harvested, at flowering, the mean enrichment of the shoot material was 3.0466 atom% 13C, or 1.9654 atom% excess 13C. To assess uniformity of labelling within plants, the shoot of a single plant was divided into leaves and stem from three sections of equal length. Ninety-three percent of this plant's dry matter had a 13C enrichment within 20 % of the weighted mean. At a field site with sandy soil, 13C labelled shoot and root material were combined and mixed with soil (0–15 cm). The soil was sampled 16 and 179 days later to determine the recovery of the added excess 13C in soil total C. The recoveries in soil (0–30 cm) were, respectively, 78 and 40 % at 16 and 179 days; there was appreciable variation associated with the recovery data from day 16, much less so at day 179. Methodological procedures for (i) enhancing the uniformity of labelling with 13C within plants, and (ii) minimising variability in the recovery of 13C from soil are suggested. ei]R Merckx  相似文献

The late-glacial climatic oscillation, 12-10 ka BP, is characterised in ice core oxygen isotope profiles by a rapid and abrupt return to glacial climate. Recent work has shown that associated with this cooling was a drop in atmospheric CO2 concentration of ca. 50 ppm. In this paper, the impact of these environmental changes on 13C discrimination is reported, based on measurements made on a continuous sequence of fossil Salix herbacea leaves from a single site. The plant responses were interpreted using an integrated model of stomatal conductance, CO2 assimilation and intercellular CO2 concentration, influenced by external environmental factors. According to the model, temperature exerts a marked influence on 13C discrimination by leaves and the pattern of 13C changes recorded by the fossil leaves is consistent with other palaeotemperature curves for 12-10 ka BP, particularly the deuterium isotope record from Alaskan Salix woods, which generally reflects ocean temperatures. The gas exchange model correctly accounts for these changes and so permits the reconstruction of ancient rates of leaf CO2 uptake and loss of water vapour in response to the abrupt late-glacial changes in global climate and CO2. The approach provides the required physiological underpinning for extracting quantitative estimates of past temperatures and for contributing an ecophysiological explanation for changes in 13C discrimination in the fossil record.  相似文献

外源氮素(N)输入陆地生态系统后会引起植物和土壤各碳库的变化,但是对不同化学形态氮素的长期输入如何影响光合碳在植物组织、土壤、土壤呼吸中的分配及转运知之甚少,尤其是对于氮输入引起光合碳分配变化进而作用于植物和土壤碳库的机制的认识还非常匮乏。基于在青藏高原矮嵩草草甸开展的不同化学形态氮素添加的长期实验,利用~(13)C示踪方法揭示了光合碳在植物地上、地下组织的分配,及其随时间在土壤中的滞留和随土壤呼吸的释放。研究结果表明,外源氮素添加10年后,与对照未添加氮素处理相比,氨态氮处理下的地上生物量增加了49.5%,氨态氮处理下的地下生物量增加了111.3%。土壤中滞留的~(13)C整体呈下降趋势,氨态氮处理下的土壤碳库显著高于硝态氮处理下的值。不同处理下的土壤呼吸中~(13)C的滞留量随时间呈指数衰减的变化趋势,其中,硝态氮处理下的~(13)C衰减最快。~(13)C同位素标记后第1天测定植物茎和叶内的~(13)C约占刚刚标定完茎和叶内~(13)C的80%,不同处理之间没有显著性差异。直至标记后的第30天,茎和叶内~(13)C的滞留量约占初始量的30%。硝态氮处理下的值在第21天和第30天显著低于对照和氨态氮处理下的值,表明硝态氮处理下,植物光合固定的碳在短期内迅速输入地下组织和土壤中。这些结果从机理上阐明了植物光合碳分配对不同化学形态氮素长期输入的响应,进而影响到土壤呼吸CO_2的释放,以及对土壤碳库动态的贡献。加深了对高寒草甸土壤有机碳库稳定性维持机制的认识,能够为高寒草地的科学管理以及资源的可持续利用提供理论指导。  相似文献

Biomass allocation is an important plant trait that responds plastically to environmental heterogeneities. However, the effects on this trait of pollutants owing to human activities remain largely unknown. In this study, we investigated the response of biomass allocation of Phragmites australis to petroleum pollution by a 13CO2 pulse-labelling technique. Our data show that plant biomass significantly decreased under petroleum pollution, but the root–shoot ratio for both plant biomass and 13C increased with increasing petroleum concentration, suggesting that plants could increase biomass allocation to roots in petroleum-polluted soil. Furthermore, assimilated 13C was found to be significantly higher in soil, microbial biomass and soil respiration after soils were polluted by petroleum. These results suggested that the carbon released from roots is rapidly turned over by soil microbes under petroleum pollution. This study found that plants can modulate biomass allocation in response to petroleum pollution.  相似文献

This study examines the effect of elevated atmospheric carbon dioxide [CO2] (+340 ppm, 13C-depleted) and/or elevated air temperature (2.8–3.5°C) on the rate and δ13C of soil respiration. The study was conducted in a boreal Norway spruce forest using temperature-controlled whole-tree chambers and 13C as a marker for root respiration. The δ13C of needle carbohydrates was followed after the onset of the CO2 treatment in August 2001 and during a 2.5-week period in the summer of 2002. Averaged over the growing seasons of 2002 and 2003, we observed a 48% and 62% increase, respectively, in soil respiration in response to elevated [CO2], but no response to elevated air temperature. The percentage increase in response to elevated [CO2] varied seasonally (between 10% and 190% relative to the control), but the absolute increase varied less (39 ± 11 mg C m−2 h−1; mean ± SD). Data on δ13C of soil respiration indicate that this increase in soil respiration rate resulted from increased root/rhizosphere respiration of recently fixed carbon. Our results support the hypothesis that root/rhizosphere respiration is sensitive to variation in substrate availability.  相似文献

The carbon isotope composition of the halophyte Mesembryanthemum crystallinum L. (Aizoaceae) changes when plants are exposed to environmental stress and when they shift from C3 to crassulacean acid metabolism (CAM). We examined the coupling between carbon isotope composition and photosynthetic pathway by subjecting plants of different ages to salinity and humidity treatments. Whole shoot 13C values became less negative in plants that were exposed to 400 mM NaCl in the hydroponic solution. The isotopic change had two components: a direct NaCl effect that was greatest in plants still operating in the C3 mode and decreased proportionally with increasing levels of dark fixation, and a second component related to the degree of CAM expression. Ignoring the presumably diffusion-related NaCl effect on carbon isotope ratios results in an overestimation of nocturnal CO2 gain in comparison to an isotope versus nocturnal CO2 gain calibration established previously for C3 and CAM species grown under well-watered conditions. It is widely taken for granted that the shift to CAM in M. crystallinum is partially under developmental control and that CAM is inevitably expressed in mature plants. Plants, cultivated under non-saline conditions and high relative humidity (RH) for up to 63 days, maintained diel CO2 gas-exchange patterns and 13C values typical of C3 plants. However, a weak CAM gas-exchange pattern and an increase in 13C value were observed in non-salt-treated plants grown at reduced RH. These observations are consistent with environmental control rather than developmental control of the induction of CAM in mature M. crystallinum under non-saline conditions.  相似文献

Carbon dioxide is released from the soil to the atmosphere in heterotrophic respiration when the dead organic matter is used for substrates for soil micro-organisms and soil animals. Respiration of roots and mycorrhiza is another major source of carbon dioxide in soil CO2 efflux. The partitioning of these two fluxes is essential for understanding the carbon balance of forest ecosystems and for modelling the carbon cycle within these ecosystems. In this study, we determined the carbon balance of three common tree species in boreal forest zone, Scots pine, Norway spruce, and Silver birch with gas exchange measurements conducted in laboratory in controlled temperature and light conditions. We also studied the allocation pattern of assimilated carbon with 14C pulse labelling experiment. The photosynthetic light responses of the tree species were substantially different. The maximum photosynthetic capacity (P max) was 2.21 μg CO2 s−1 g−1 in Scots pine, 1.22 μg CO2 s−1 g−1 in Norway spruce and 3.01 μg CO2 s−1 g−1 in Silver birch seedlings. According to the pulse labelling experiments, 43–75% of the assimilated carbon remained in the aboveground parts of the seedlings. The amount of carbon allocated to root and rhizosphere respiration was about 9–26%, and the amount of carbon allocated to root and ectomycorrhizal biomass about 13–21% of the total assimilated CO2. The 14CO2 pulse reached the root system within few hours after the labelling and most of the pulse had passed the root system after 48 h. The transport rate of carbon from shoot to roots was fastest in Silver birch seedlings.  相似文献

A model for rhizodeposition and root respiration was developed and parameterised based on 14C pulse labelling of Lolium perenne. The plants were grown in a two-compartment chamber on a loamy Haplic Luvisol under controlled laboratory conditions. The dynamics of 14CO2 efflux from the soil and 14C content in shoots, roots, micro-organisms, dissolved organic carbon (DOC) and soil were measured during the first 11 days after labelling. Modelled parameters were estimated by fitting on measured 14C dynamics in the different pools. The model and the measured 14C dynamics in all pools corresponded well (r 2=0.977). The model describes well 14CO2 efflux from the soil and 14C dynamics in shoots, roots and soil, but predicts unsatisfactorily the 14C content in micro-organisms and DOC. The model also allows for division of the total 14CO2 efflux from the soil in 14CO2 derived from root respiration and 14CO2 derived from rhizomicrobial respiration by use of exudates and root residues. Root respiration and rhizomicrobial respiration amounted for 7.6% and 6.0% of total assimilated C, respectively, which accounts for 56% and 44% of root-derived 14CO2 efflux from the soil planted with 43-day-old Lolium perenne, respectively. The sensitivity analysis has shown that root respiration rate affected the curve of 14CO2 efflux from the soil mainly during the first day after labelling. The changes in the exudation rate influenced the 14CO2 efflux later than first 24 h after labelling.  相似文献

The possibility of measuring the rates of light and dark CO2 assimilation using 13C carbonate was demonstrated on Lake Kichier (Marii El). The application of methods utilizing the stable 13C and the radioactive 14C isotopes resulted in comparable values of the rates of light and dark CO2 fixation. Due to its absolute environmental safety, the method with 13C mineral carbon can be recommended as an alternative to radioisotope methods for qualitative measurements of CO2 fixation rates in aquatic ecosystems.  相似文献

Plant materials labelled with 13C can be used to trace litter decomposition and root carbon flow, but only if the isotope is uniformly distributed in the plant. We postulated that if 13CO2 were applied at regular intervals, in direct proportion to the rate of photosynthesis, then the abundance of 13C would be uniform among plant parts. To test this hypothesis, wheat plants were grown in the greenhouse, and exposed weekly to 13CO2 for six hours in a closed chamber. A constant dose of 13CO2 (about 33 atom%) was injected whenever CO2 concentration fell below a prescribed limit, so that 13CO2 was added in proportion to photosynthetic rate. Wheat exposed for 13 weeks (starting 11 days after seeding) had reasonably consistent 13C abundance among plant parts: grain = 3.41, chaff = 3.41, stem = 3.65, and root = 3.50 atom%. The `leaf' fraction had slightly higher abundance (3.99 atom%), perhaps because recently-fixed 13C was not translocated from senescing tissue. Exposing plants only during early stages of the growing season increased differences among plant parts. The approach offers a practical way to label plants with 13C.  相似文献

Rhizodeposition, i.e. the release of carbon into the soil by growing roots, is an important part of the terrestrial carbon cycle. However thein situ nature and dynamics of root-derived carbon in the soil are still poorly understood. Here we made an investigation of the latter in laboratory experiments using13CO2 pulse chase labelling of wheat (Triticum aestivum L.). We analyzed the kinetics of13C-labelled carbon and more specially13C carbohydrates in the rhizosphere. Wheat seedlings-soil mesocosms were exposed to13CO2 for 5 hours in controlled chambers and sampled repeatedly during two weeks for13C/C analysis of organic carbon. After a two-step separation of the soil from the roots, the amount of total organic13C was determined by isotope ratio mass spectrometry as well as the amounts of13C in arabinose, fructose, fucose, glucose, galactose, mannose, rhamnose and xylose. The amount and isotopic ratio of monosaccharides were obtained by capillary gas chromatography coupled with isotope ratio mass spectrometry (GC/C/IRMS) after trimethyl-silyl derivatization. Two fractions were analyzed : total (hydrolysable) and soluble monomeric (water extractable) soil sugars. The amount of organic13C found in the soil, expressed as a percentage of the total photosynthetically fixed13C at the end of the labelling period, reached 16% in the day following labelling and stabilised at 9% after one week. We concluded that glucose under the form of polymers was the dominant moietie of rhizodeposits. Soluble glucose and fructose were also present. But after 2 days, these soluble sugars had disappeared. Forty percent of the root-derived carbon was in the form of neutral sugars, and exhibited a time-increasing signature of microbial sugars. The composition of rhizospheric sugars rapidly tended towards that of bulk soil organic matter.  相似文献

The aim of this work was to examine the response of wheat plants to a doubling of the atmospheric CO2 concentration on: (1) carbon and nitrogen partitioning in the plant; (2) carbon release by the roots; and (3) the subsequent N uptake by the plants. The experiment was performed in controlled laboratory conditions by exposing fast-growing spring wheat plants, during 28 days, to a 14CO2 concentration of 350 or 700 L L–1 at two levels of soil nitrogen fertilization. Doubling CO2 availability increased total plant production by 34% for both N treatment. In the N-fertilized soil, the CO2 enrichment resulted in an increase in dry mass production of 41% in the shoots and 23% in the roots; without N fertilization this figure was 33% and 37%, respectively. In the N-fertilized soil, the CO2 increase enhanced the total N uptake by 14% and lowered the N concentration in the shoots by 23%. The N concentration in the roots was unchanged. In the N-fertilized soil, doubling CO2 availability increased N uptake by 32% but did not change the N concentrations, in either shoots or roots. The CO2 enrichment increased total root-derived carbon by 12% with N fertilization, and by 24% without N fertilization. Between 85 and 90% of the total root derived-14C came from respiration, leaving only 10 to 15% in the soil as organic 14C. However, when total root-derived 14C was expressed as a function of root dry weight, these differences were only slightly significant. Thus, it appears that the enhanced carbon release from the living roots in response to increased atmospheric CO2, is not due to a modification of the activity of the roots, but is a result of the increased size of the root system. The increase of root dry mass also resulted in a stimulation of the soil N mineralization related to the doubling atmospheric CO2 concentration.
