Ex situ conservation of Bulgarian endemic plant Achillea thracica Velen. was achieved by successful in vitro cultivation of mono-nodal segments on MS-B5 medium supplemented with 1.0 mg/L BA for 20 days and subsequent transferring of regenerated plants on hormone free basal MS-B5 medium for root development and accumulation of leaf biomass. In vitro multiplicated plants were successfully acclimated in a growth chamber with 100% survival. GC–MS analysis of the essential oils resulted in the identification of 30, 10 and 28 compounds in in situ grown, in vitro cultivated and ex vitro adapted plants, respectively, constituting 77.7%, 99.9% and 84.1% of the total oils. The wider variety of compounds was found in the essential oils of in situ and ex vitro adapted plants where santolina alcohol, β-eudesmol, 1,8-cineole, germacrene D, α-cadinol and artemisia alcohol were the principal components comprising 68.7% and 69.3 of the oil, respectively. In vitro cultivated plants consist of mainly 1,8-cineole, germacrene D and artemisia alcohol representing 87% of the oil. Different growth conditions affect the composition of essential oils, suggesting their possible involvement in the process of adaptation and surviving in changing environmental conditions.  相似文献

An investigation was carried out with paclobutrazol (PBZ) in order to study its effect on the photosynthetic and anatomical characteristics of Catharanthus roseus (L.) G. Don. The chlorophyll content and related parameters like net photosynthetic rate, transpiration rate and internal CO2 concentration were measured from control and treated plant samples. The anatomical characteristics, viz., leaf thickness, epidermis and cuticle, palisade layer, spongy layer and vascular bundle were studied. The root alkaloid ajmalicine was extracted and estimated from both treated and control plant roots. Photosynthetic pigment increased under PBZ treatment. PBZ treatment increased the net photosynthetic rate and internal CO2 concentration, but reduced transpiration. PBZ treatment increased the thickness of the leaf, epidermis and cuticle, palisade layer and spongy layer but reduced the diameter of xylem vessels significantly when compared to control, however the phloem elements had shown an increased diameter as compared to control in the PBZ treated plants. There was a significant enhancement in ajmalicine content under PBZ treatment when compared to control plants. The results suggested that the application of PBZ have significant effects on photosynthetic and anatomical responses thus can be used for improving productivity in medicinal plants.  相似文献

Recent studies have provided evidence of a large flux of root-respired CO2 in the transpiration stream of trees. In our study, we investigated the potential impact of this internal CO2 transport on aboveground carbon assimilation and CO2 efflux. To trace the transport of root-respired CO2, we infused a 13C label at the stem base of field-grown Populus deltoides Bartr. ex. Marsh trees. The 13C label was transported to the top of the stem and throughout the crown via the transpiration stream. Up to 17% of the 13C label was assimilated by chlorophyll-containing tissues. Our results provide evidence of a mechanism for recycling respired CO2 within trees. Such a mechanism may have important implications for how plants cope with predicted increases in intensity and frequency of droughts. Here, we speculate on the potential significance of this recycling mechanism within the context of plant responses to climate change and plants currently inhabiting arid environments.  相似文献

We analysed the impact of elevated CO2 on water relations, water use efficiency and photosynthetic gas exchange in barley (Hordeum vulgare L.) under wet and drying soil conditions. Soil moisture was less depleted under elevated compared to ambient [CO2]. Elevated CO2 had no significant effect on the water relations of irrigated plants, except on whole plant hydraulic conductance, which was markedly decreased at elevated compared to ambient CO2 concentrations. The values of relative water content, water potential and osmotic potential were higher under elevated CO2 during the entire drought period. The better water status of water-limited plants grown at elevated CO2 was the result of stomatal control rather than of osmotic adjustment. Despite the low stomatal conductance produced by elevated CO2, net photosynthesis was higher under elevated than ambient CO2 concentrations. With water shortage, photosynthesis was maintained for longer at higher rates under elevated CO2. The reduction of stomatal conductance and therefore transpiration, and the enhancement of carbon assimilation by elevated CO2, increased instantaneous and whole plant water use efficiency in both irrigated and droughted plants. Thus, the metabolism of barley plants grown under elevated CO2 and moderate or mild water deficit conditions is benefited by increased photosynthesis and lower transpiration. The reduction in plant water use results in a marked increase in soil water content which delays the onset and severity of water deficit.  相似文献

This study aims to elucidate the effects of abscisic acid (ABA) foliar application on plant hardening during ex vitro acclimatization using a forest model species Ulmus minor L. Plant leaves were sprayed with ABA (0, 50 and 100 μM) immediately after ex vitro transfer and twice a week for the first 3 weeks of acclimatization. After this period, parameters related with photosynthesis, hormone levels and oxidative stress were measured to assess plant performance. The results demonstrated that ABA foliar application alleviates the negative shock of ex vitro acclimatization since it reduces the water loss through transpiration, relieving the risk of wilting. Moreover, ABA promoted net CO2 assimilation rate (P N ) and plant dry mater accumulation. ABA treatment increased the antioxidant battery during acclimatization, with more effective results at the concentration of 50 μM ABA. Also, flow cytometry data support that cytosolic compounds, which may increase in response to 50 μM ABA, could also protect DNA from oxidative damage. We propose here that ABA foliar application (immediately after ex vitro transfer), by preventing water loss, enhancing photosynthesis efficiency and the activity of antioxidant enzymes, improves the plants hardening and ability to deal with the ex vitro stresses.  相似文献

Photosynthetic CO2 assimilation, transpiration, ribulose-1,5-bisphosphate carboxylase (RuBPCase), and soluble protein were reduced in leaves of water-deficit (stress) `Valencia' orange (Citrus sinensis [L.] Osbeck). Maximum photosynthetic CO2 assimilation and transpiration, which occurred before midday for both control and stressed plants, was 58 and 40%, respectively, for the stress (−2.0 megapascals leaf water potential) as compared to the control (−0.6 megapascals leaf water potential). As water deficit became more severe in the afternoon, with water potential of −3.1 megapascals for the stressed leaves vs. −1.1 megapascals for control leaves, stressed-leaf transpiration declined and photosynthetic CO2 assimilation rapidly dropped to zero. Water deficit decreased both activation and total activity of RuBPCase. Activation of the enzyme was about 62% (of fully activated enzyme in vitro) for the stress, compared to 80% for the control. Water deficit reduced RuBPCase initial activity by 40% and HCO3/Mg2+-saturated activity by 22%. However, RuBPCase for both stressed and control leaves were similar in Kcat (25 moles CO2 per mole enzyme per second) and Km for CO2 (18.9 micromolar). Concentrations of RuBPCase and soluble protein of stressed leaves averaged 80 and 85%, respectively, of control leaves. Thus, reductions in activation and concentration of RuBPCase in Valencia orange leaves contributed to reductions in enzyme activities during water-deficit periods. Declines in leaf photosynthesis, soluble protein, and RuBPCase activation and concentration due to water deficit were, however, recoverable at 5 days after rewatering.  相似文献

Water deficit (WD) is a growing problem in agriculture. In citrus crops, genetically-determined rootstock characteristics are important factors influencing plant responses to WD. Aquaporins are involved in regulating the water supply to the plant by mediating water flow through the cell membranes. Recent studies support a direct role for aquaporins in plant water relations and demonstrate their involvement in WD tolerance. This study investigates the relationship between photosynthetic and water-balance parameters with aquaporin expression levels and hydraulic conductance of roots (Kr) in conditions of moderate WD in citrus rootstocks. The plant materials used were the rootstocks Poncirus trifoliata (L.) Raf. (PT), Cleopatra mandarin (Citrus reshni Hort ex Tan.) (CM) and 030115 (a hybrid of the two former rootstocks), all grafted with the citrus variety ??Valencia Late?? (C. sinensis (L.) Osb). Plants were irrigated with two differents irrigation doses (normal irrigation and moderate WD) during 70 days and leaf water potential (??s), net CO2 assimilation (ACO2), transpiration, stomatal conductance (gs) and substomatal CO2 concentration (Ci) were measured periodically under both irrigation conditions. Kr and PIP1 and PIP2 gene expression levels in fine roots of control plants and plants subjected to WD on day 43 of the experiment were determined. Under WD conditions, the hybrid 030115 drastically reduced aquaporin expression and Kr, accompanied by a loss of plant vigour but without reducing the net CO2 assimilation (ACO2). PT maintained the same aquaporin expression level and similar Kr under WD as under normal irrigation conditions, but suffered a sharp reduction in ACO2. CM, which has lower Kr and aquaporin expression than PT under both normal irrigation conditions and WD, responded better to water stress conditions than PT. Low aquaporin levels, or down-regulated aquaporin expression, accompanied by decreased plant vigour led to decreased plasma membrane permeability, thereby facilitating water retention in the cells under water stress conditions. This may induce water stress tolerance in citrus rootstocks.  相似文献

Rates of assimilation and transpiration in Eucalyptus pauciflora Sieb. ex Spreng were measured at various ambient partial pressures of CO2 and various irradiances and were used to estimate leaf conductance and intercellular partial pressure of CO2. The responses of leaf conductance and rate of assimilation to change in intercellular partial pressure of CO2 were expressed in terms of feedback. They are small in the sense that their combined effect was to reduce disturbances in intercellular partial pressure of CO2 by 30% only. The magnitude of the feedback had no influence on the system as affected by irradiance, because the direct responses of conductance and rate of assimilation to changes in irradiance in the range 0.25 to 2 millieinsteins per meter per second were such that intercellular partial pressure was maintained almost constant.  相似文献

Summary Nodal cuttings of Eucalyptus camaldulensis L. plantlets were cultured photoautotrophically (sugar-free nutrient medium and with enriched CO2 and high photosynthetic photon flux) in a scaled-up vessel (volume 4.0 liters) under forced ventilation (SV-treatment). After 28 d of culture, physiological aspects of the plantlets were compared with plantlets grown photomixotrophically (20 g l−1 sucrose in the medium) in a Magenta vessel (volume 0.4 liters) under natural ventilation (control). In the SV-treatment net photosynthetic rates were enhanced, normal stomatal closing and opening were observed, and the epicuticular leaf-wax content was significantly higher than the control. The anatomical study showed well-organized palisade and spongy mesophyll layers of SV leaves. The SV-treatment also allowed in vitro acclimatization, and after transplanting ex vitro, the transpiration rate and the percent water loss was lower than those of the control and thus the SV plantlets acclimatized easily ex vitro.  相似文献

Unlike C-3 plants, cacti possess a crassulacean acid metabolism (CAM) physiology that can alter the pattern of carbon uptake and affect plant growth under artificial environmental conditions, especially in tissue culture. In vitro-derived plantlets of Coryphantha minima grew 7-fold larger than plants cultured under similar ex vitro conditions. Growth regulators incorporated into the culture media during shoot proliferation stage of micropropagation had a strong influence on this increased growth. Other important factors that contributed to increased growth under in vitro conditions were high relative humidity and sugar in the culture medium. An analysis of gas exchange and daily fluctuations of malic acid levels revealed an increase in net photosynthetic rate, in terms of carbon assimilation, by in vitro plants compared with that of ex vitro plants. This stimulated photosynthesis in the presence of an external carbon source was unexpected but apparently true for cacti exhibiting CAM physiology. Unlike CAM plants grown in ex vitro conditions, net CO2 uptake by in vitro-cultured cacti occurred continuously in the light as well as the dark. Once regenerated, cacti were transferred to ex vitro conditions where the normal CAM pathway resumed with a concomitant reduction in growth and CO2 uptake. These results showed that growth of cacti can be considerably accelerated by in vitro culture. This revised version was published online in June 2006 with corrections to the Cover Date.  相似文献

Rates of CO2 assimilation and leaf conductances to CO2 transfer were measured in plants of Zea mays during a period of 14 days in which the plants were not rewatered, and leaf water potential decreased from −0.5 to −8.0 bar. At any given ambient partial pressure of CO2, water stress reduced rate of assimilation and leaf conductance similarly, so that intercellular partial pressure of CO2 remained almost constant. At normal ambient partial pressure of CO2, the intercellular partial pressure of CO2 was estimated to be 95 microbars. This is the same as had been estimated in plants of Zea mays grown with various levels of nitrogen supply, phosphate supply and irradiance, and in plants of Zea mays examined at different irradiances.

After leaves of Phaseolus vulgaris L. and Eucalyptus pauciflora Sieb. ex Spreng had been exposed to high irradiance in an atmosphere of CO2-free N2 with 10 millibars O2, rates of assimilation and leaf conductances measured in standard conditions had decreased in similar proportions, so that intercellular partial pressure of CO2 remained almost unchanged. As the conductance of each epidermis that had not been directly irradiated had declined as much as that in the opposite, irradiated surface it was hypothesized that conductance may have been influenced by photoinhibition within the mesophyll tissue.

Transpiration-induced changes in the photosynthetic capacity of leaves   总被引:18,自引:0,他引:18
Thomas D. Sharkey 《Planta》1984,160(2):143-150
High transpiration rates were found to affect the photosynthetic capacity of Xanthium strumarium L. leaves in a manner analagous to that of low soil water potential. The effect was also looked for and found in Gossypium hirsutum L., Agathis robusta (C. Moore ex Muell.) Bailey, Eucalyptus microcarpa Maiden, Larrea divaricata Cav., the wilty flacca tomato mutant (Lycopersicon esculentum (L.) Mill.) and Scrophularia desertorum (Munz) Shaw. Two methods were used to distinguish between effects on stomatal conductance, which can lower assimilation by reducing CO2 availability, and effects on the photosynthetic capacity of the mesophyll. First, the response of assimilation to intercellular CO2 pressure (C i) was compared under conditions of high and low transpiration. Second, in addition to estimating C i using the usual Ohm's law analogy, C i was measured directly using the closed-loop technique of T.D. Sharkey, K. Imai, G.D. Farquhar and I.R. Cowan (1982, Plant Physiol, 60, 657–659). Transpiration stress responses of Xanthium strumarium were compared with soil drought effects. Both stresses reduced photosynthesis at high C i but not at low C i; transpiration stress increased the quantum requirement of photosynthesis. Transpiration stress could be induced in small sections of leaves. Total transpiration from the plant did not influence the photosynthetic capacity of a leaf kept under constant conditions, indicating that water deficits develop over small areas within the leaf. The effect of high transpiration on photosynthesis was reversed approximately half-way by returning the plants to low-transpiration conditions. This reversal occurred as fast as measurements could be made (5 min), but little further recovery was observed in subsequent hours.Abbreviations and symbols A photosynthetic CO2 assimilation rate - C a ambient CO2 partial pressure - C i partial pressure of CO2 inside the leaf - VPD leaf-to-air water-vapor pressure difference This research was begun while the author was a Postdoctoral Research Fellow at the Australian National University, Canberra  相似文献

In the study reported here, two different photoinhibitory phenomena were compared within a single plant species. Bean plants were grown in three different light intensities to simulate sun and shade environments. The effects of photoinhibitory treatments on in vivo CO2 assimilation rates and in vitro chloroplast electron transport reactions were investigated and the extent to which carbon metabolism served to prevent photoinhibition was characterized. It was shown that the photoinhibition which follows exposure of intact leaflets of low light-grown bean plants to high light intensity in normal air is essentially similar to that which occurs when leaflets of plants grown in full sunlight are illuminated in the absence of CO2 at low O2 partial pressures.

Aleurocanthus woglumi (Ashby, 1915) is an important agricultural pest that causes yield losses of 20–80% in citrus plants by removing plant nutrients while feeding and allowing the formation of sooty mold. The objective of this study was to evaluate physiological changes in citrus plants in response to A. woglumi infestation under field conditions. The experiment was conducted in a citrus orchard in Paço do Lumiar, Maranhão, Brazil. Thirty-two citrus plants were used, including eight of each of the following varieties: Tahiti lime, Tanjaroa tangerine, Nissey tangerine, and Ponkan tangerine. Four random plants with A. woglumi infestation and four plants free from this pest were selected from each variety. The physiological parameters evaluated were photochemical efficiency and gas exchange. Regarding photochemical efficiency, infested plants presented photoinhibition damage, with a performance index of 4.22. The gas exchange parameters of infested plants changed, with reductions in photosynthetic CO2 assimilation of 69.7% (Tahiti), 64% (Tanjaroa), 68.8% (Nissey) and 63.3% (Ponkan). Plants infested with A. woglumi also presented physiological changes; their photosynthetic CO2 assimilation, stomatal conductance, instantaneous transpiration, and performance indexes were affected. The infested citrus plants showed photoinhibition of photosystem II. The photosynthetic CO2 assimilation decreased approximately 70% in Tahiti lime, Tanjaroa tangerine, Nissey tangerine, and Ponkan tangerine plants infested with A. woglumi.  相似文献

Seedlings of Chrysanthemum, cultivar ‘Puma Sunny’, were grown under a range of shading regimes (natural full sunlight, 55, 25, and 15% of full sunlight) for 18 days. Here, we characterized effects of varying light regimes on plant morphology, photosynthesis, chlorophyll fluorescence, anatomical traits, and chloroplast ultrastructure. We showed that leaf color was yellowish-green under full sunlight. Leaf area, internode length, and petiole length of plants were the largest under 15% irradiance. Net photosynthetic rate, water-use efficiency, PSII quantum efficiency, and starch grain were reduced with decreasing irradiance from 100 to 15%. Heavy shading resulted in the partial closure of PSII reaction centers and the CO2 assimilation was restricted. The results showed the leaves of plants were thinner under 25 and 15% irradiance with loose palisade tissue and irregularly arranged spongy mesophyll cells, while the plants grown under full sunlight showed the most compact leaf palisade parenchyma. Irradiance lesser than 25% of full sunlight reduced carbon assimilation and led to limited plant growth. Approximately 55% irradiance was suggested to be the optimal for Chrysanthemum morifolium.  相似文献

Hydrostatic pressures varying from 0 to 6.0 bar were applied to roots of intact Capsicum annuum L. cv. California Wonder plants growing in nutrient solution and the rates of transpiration, and net CO2 assimilation, apparent compensation point and leaf water potential measured. Increasing the pressure on the roots of plants with roots in solution with either -0.5 or -5.0 bar osmotic potential with 1 bar increments resulted in a decrease in transpiration. With the application of 1 or 2 bar pressure the rate of transpiration returned to near or above the original rate. An application of 3 or 4 bar pressure reduced the rate of transpiration of all plants. The transpiration of plants with roots in solution with -0.5 bar osmotic potential remained at the reduced rate for as long as these pressures were maintained. The transpiration of plants with roots in solution with -5.0 bar was only temporarily suppressed at these pressures. Changing the applied pressure from 3 or 4 bar to 0 resulted in a rapid increase in transpiration which lasted approximately 15 minutes. This was followed by a decrease in transpiration to a rate lower than before the pressure was applied. The pattern of response was similar for plants at low or high light intensity or at normal or low CO2 concentrations. When leaf diffusive resistance was 6.0 s cm?1 or greater, changes in net CO2 assimilation were similar to those of transpiration. The apparent CO2 compensation point increased as pressure was applied and decreased with a release in pressure. Leaf water potential increased with an increase in pressure and decreased with a decrease in pressure. The changes in leaf water potential were frequently but not always proportional to changes in pressure. It is postulated that the respouses noted were due to changes in resistance to flow of water from xylem terminals through the mesophyll cells and stomatal cavities to the atmosphere.  相似文献

The objective of this study was to determine the response of nitrogen metabolism to drought and recovery upon rewatering in barley (Hordeum vulgare L.) plants under ambient (350 μmol mol−1) and elevated (700 μmol mol−1) CO2 conditions. Barley plants of the cv. Iranis were subjected to drought stress for 9, 13, or 16 days. The effects of drought under each CO2 condition were analysed at the end of each drought period, and recovery was analysed 3 days after rewatering 13-day droughted plants. Soil and plant water status, protein content, maximum (NRmax) and actual (NRact) nitrate reductase, glutamine synthetase (GS), and aminant (NADH-GDH) and deaminant (NAD-GDH) glutamate dehydrogenase activities were analysed. Elevated CO2 concentration led to reduced water consumption, delayed onset of drought stress, and improved plant water status. Moreover, in irrigated plants, elevated CO2 produced marked changes in plant nitrogen metabolism. Nitrate reduction and ammonia assimilation were higher at elevated than at ambient CO2, which in turn yielded higher protein content. Droughted plants showed changes in water status and in foliar nitrogen metabolism. Leaf water potential (Ψw) and nitrogen assimilation rates decreased after the onset of water deprivation. NRact and NRmax activity declined rapidly in response to drought. Similarly, drought decreased GS whereas NAD-GDH rose. Moreover, protein content fell dramatically in parallel with decreased leaf Ψw. In contrast, elevated CO2 reduced the water stress effect on both nitrate reduction and ammonia assimilation coincident with a less-steep decrease in Ψw. On the other hand, Ψw practically reached control levels after 3 days of rewatering. In parallel with the recovery of plant water status, nitrogen metabolism was also restored. Thus, both NRact and NRmax activities were restored to about 75-90% of control levels when water supply was restored; the GS activity reached 80-90% of control values; and GDH activities and protein content were similar to those of control plants. The recovery was always faster and slightly higher in plants grown under elevated CO2 conditions compared to those grown in ambient CO2, but midday Ψw dropped to similar values under both CO2 conditions. The results suggest that elevated CO2 improves nitrogen metabolism in droughted plants by maintaining better water status and enhanced photosynthesis performance, allowing superior nitrate reduction and ammonia assimilation. Ultimately, elevated CO2 mitigates many of the effects of drought on nitrogen metabolism and allows more rapid recovery following water stress.  相似文献

Influence of elevated carbon dioxide on water relations of soybeans   总被引:8,自引:1,他引:7       下载免费PDF全文
Soybean (Glycine max L. Merrill cv `Bragg') plants were grown in pots at six elevated atmospheric CO2 concentrations and two watering regimes in open top field chambers to characterize leaf xylem potential, stomatal resistance and conductance, transpiration, and carbohydrate contents of the leaves in response to CO2 enrichment and water stress conditions. Groups of plants at each CO2 concentration were subjected to water stress by withholding irrigation for 4 days during the pod-filling stage.

Under well watered conditions, the stomatal conductance of the plants decreased with increasing CO2 concentration. Therefore, although leaf area per plant was greater in the high CO2 treatments, the rate of water loss per plant decreased with CO2 enrichment. After 4 days without irrigation, plants in lower CO2 treatments showed greater leaf tissue damage, lower leaf water potential, and higher stomatal resistance than high CO2 plants. Stomatal closure occurred at lower leaf water potentials for the low CO2 grown plants than the high CO2 grown plants. Significantly greater starch concentrations were found in leaves of high CO2 plants, and the reductions in leaf starch and increases in soluble sugars due to water stress were greater for low CO2 plants. The results showed that even though greater growth was observed at high atmospheric CO2 concentrations, lower rates of water use delayed and, thereby, prevented the onset of severe water stress under conditions of low moisture availability.

In vivo CO2 fixation activity and in vitro phosphoenolpyruvate carboxylase activity were demonstrated in effective and ineffective nodules of alfalfa (Medicago sativa L.) and in the nodules of four other legume species. Phosphoenolpyruvate carboxylase activity was greatly reduced in nodules from both host and bacterially conditioned ineffective alfalfa nodules as compared to effective alfalfa nodules.

Forage harvest and nitrate application reduced both in vivo and in vitro CO2 fixation activity. By day 11, forage harvest resulted in a 42% decline in in vitro nodule phosphoenolpyruvate carboxylase activity while treatment with either 40 or 80 kilograms nitrogen per hectare reduced activity by 65%. In vitro specific activity of phosphoenolpyruvate carboxylase and glutamate synthase were positively correlated with each other and both were positively correlated with acetylene reduction activity.

The distribution of radioactivity in the nodules of control plants (unharvested, 0 kilograms nitrogen per hectare) averaged 73% into the organic acid and 27% into the amino acid fraction. In nodules from harvested plants treated with nitrate, near equal distribution of radioactivity was observed in the organic acid (52%) and amino acid (48%) fractions by day 8. Recovery to control distribution occurred only in those nodules whose in vitro phosphoenolpyruvate carboxylase activity recovered.

The results demonstrate that CO2 fixation is correlated with nitrogen fixation in alfalfa nodules. The maximum rate of CO2 fixation for attached and detached alfalfa nodules at low CO2 concentrations (0.13-0.38% CO2) were 18.3 and 4.9 nanomoles per hour per milligram dry weight, respectively. Nodule CO2 fixation was estimated to provide 25% of the carbon required for assimilation of symbiotically fixed nitrogen in alfalfa.
