The German anatomists, M. Ernst and A. Glücksmann, deserve credit for the discovery of widespread cell death in embryonic tissues, including the nervous tissue. In 1934, V. Hamburger described a significant hypoplasia in dorsal root ganglia (DGR) and lateral motor columns, following the extirpation of limb buds in chick embryos. In the early 1940s, Dr. Rita Levi-Montalcini in Turin (Italy) repeated the experiment and suggested that the hypoplasia might result from the death of young differentiated neurons. In a joint reinvestigation, published in 1949, large numbers of degenerating neurons were described in brachial DRG, following wing bud extirpations. In the same embryos, Dr. Levi-Montalcini observed massive neuronal death in cervical and thoracic DRG which had not been affected by the operation. This was the discovery of naturally occurring neuronal death. Long after the discovery of Nerve Growth Factor (NGF) it was recognized that NGF and natural neuronal death are two sides of the same coin: the latter results from an insufficient supply of the former by the target tissues.  相似文献

The remarkable accomplishments in developmental neurobiology within the past 60 years have depended on two things: (i) a succession of original histochemical and immunohistochemical methodologies for identifying pathways in the nervous system with increasing precision and sensitivity, and (ii) the discovery of growth factors for neurons. Growth factors are naturally occurring, essential biological mediators that promote cell growth, differentiation, survival and function in specific nerve cell populations. The discovery of nerve growth factor (NGF) by Rita Levi-Montalcini in the 1950s represents an important milestone in the processes that led to modern cell biology. NGF was the first growth factor identified, for its action on the morphological differentiation of neural-crest-derived nerve cells. Later, its effect on neuronal cells of the peripheral and central nervous systems, and on several non-neuronal cells was also determined. Thus, Levi-Montalcini's work on NGF represents, as acknowledged by the Nobel Prize Assembly in its press release of 13 October 1986, "a fascinating example of how a skilled observer can create a concept out of apparent chaos".  相似文献

Diverse tree structures such as blood vessels, branches of a tree and river basins exist in nature. The constructal law states that the evolution of flow structures in nature has a tendency to facilitate flow. This study suggests a theoretical basis for evaluation of flow facilitation within vascular structure from the perspective of evolution. A novel evolution parameter (Ev) is proposed to quantify the flow capacity of vascular structures. Ev is defined as the ratio of the flow conductance of an evolving structure (configuration with imperfection) to the flow conductance of structure with least imperfection. Attaining higher Ev enables the structure to expedite flow circulation with less energy dissipation. For both Newtonian and non-Newtonian fluids, the evolution parameter was developed as a function of geometrical shape factors in laminar and turbulent fully developed flows. It was found that the non-Newtonian or Newtonian behavior of fluid as well as flow behavior such as laminar or turbulent behavior affects the evolution parameter. Using measured vascular morphometric data of various organs and species, the evolution parameter was calculated. The evolution parameter of the tree structures in biological systems was found to be in the range of 0.95 to 1. The conclusion is that various organs in various species have high capacity to facilitate flow within their respective vascular structures.  相似文献

The nerve growth factor (NGF) is a signaling protein, discovered by Rita Levi-Montalcini in the early 1950's for its effect on growth and differentiation of specific populations of neurons of the peripheral nervous system. Originally identified as neurite outgrowth-stimulating factor, later studies revealed that the purified molecule has a number of target cells in the central nervous system and on nonneuronal cells. Moreover, recent studies showed the potential therapeutic properties of NGF in neuropathies of the central and peripheral nervous system and diseases of the eye and skin. Here I briefly describe the discovery of NGF, the early studies of Rita LeviMontalcini, a pioneer in modern neuroscience, and my scientific and human experience working in her laboratory for over 40 years.  相似文献

Motor skill memory is first encoded online in a fragile form during practice and then converted into a stable form by offline consolidation, which is the behavioral stage critical for successful learning. Praise, a social reward, is thought to boost motor skill learning by increasing motivation, which leads to increased practice. However, the effect of praise on consolidation is unknown. Here, we tested the hypothesis that praise following motor training directly facilitates skill consolidation. Forty-eight healthy participants were trained on a sequential finger-tapping task. Immediately after training, participants were divided into three groups according to whether they received praise for their own training performance, praise for another participant''s performance, or no praise. Participants who received praise for their own performance showed a significantly higher rate of offline improvement relative to other participants when performing a surprise recall test of the learned sequence. On the other hand, the average performance of the novel sequence and randomly-ordered tapping did not differ between the three experimental groups. These results are the first to indicate that praise-related improvements in motor skill memory are not due to a feedback-incentive mechanism, but instead involve direct effects on the offline consolidation process.  相似文献

Viktor Hamburger was a developmental biologist interested in the ontogenesis of the vertebrate nervous system. A student of Hans Spemann at Freiburg in the 1920s, Hamburger picked up a holistic view of the embryo that precluded him from treating it in a reductionist way; at the same time, he was committed to a materialist and analytical approach that eschewed any form of vitalism or metaphysics. This paper explores how Hamburger walked this thin line between mechanistic reductionism and metaphysical vitalism in light of his work on the factors influencing growth of neurons into limb buds, and the discovery of nerve growth factor, work carried out with Rita Levi-Montalcini and Stanley Cohen.  相似文献

This study investigated the effects of host density and distribution on the patch-leaving behavior of Diadegma semiclausum (Hymenoptera: Ichneumonidae), a solitary endoparasitoid of larval Plutella xylostella (Lepidoptera: Plutellidae). Individual female wasps were released onto an experimental plant infested with host larvae at different densities and distributions, and were allowed to freely leave for an alternative host plant placed upwind of the experimental plant in a wind tunnel. The influence of host density and distribution, as well as within-patch foraging experience, on the parasitoids patch-leaving tendency was analyzed by means of the proportional hazards model. This study aimed to test the predictions of a number of patch-leaving models, including the Marginal Value Theorem, rules of thumb, and incremental or countdown mechanisms. The parasitoids patch-leaving tendency decreased with increased host density, more clustered host distribution, and unsuccessful host encounter as a result of host defense, but increased with successful oviposition. None of the simple rules of thumb such as fixed time, fixed number of hosts parasitized, or fixed giving-up time was employed by this parasitoid. The results agreed with the general predictions of the Marginal Value Theorem that patch residence time and numbers of ovipositions by the parasitoid increased with increasing host density. The decreasing influence of oviposition on the parasitoids patch-leaving tendency, regardless of host density or distribution, was consistent with the prediction of a countdown mechanism.  相似文献

Ion transport through a gramicidin A like channel in the presence of solvent molecules with van der Waals parameters of water has been studied by means of the molecular dynamics simulation technique. It was found that the presence of solvent molecules in the channel has a tendency to equalize the effective masses of the ions through "association" thus giving the experimentally found ion selectivity to the gramicidin A channel.  相似文献

Submicroscopic embryo organization during preimplantation development was analyzed and its carbohydrate metabolism at these stages was studied. In the process of investigation the following regularities were revealed: progressive rearrangement of the nucleus (from compact to reticular); alterations in cytoplasmic structures (in multivesicular bodies, in autophagal vacuoles, in mitochondria, in granular cytoplasmic network, in lamellar structures); complication of intracellular interrelations (by means of microvilli and median bodies at early stages and during subsequent formation of cellular processes, contacts after "thorn--net" pattern, dense, fissure-like contacts and desmosomes). By means of cytochemical and electron histochemical methods, glycogen distribution in embryos at different stages of development was elucidated, and tendency to its decrease with the progress of the development was noted, with its least contents being at the stage of blastocyst.  相似文献

针对毛乌素沙地脆弱农业生态系统生产力实际,根据复合生态系统耦合理论,借助能值理论及分析方法,选择毛乌素沙地偏农区的榆阳区和偏牧区的乌审旗为比较研究对象,对两地近27a的农牧耦合系统初级生产力、次级生产力及系统耦合状况进行了对比分析,探讨性提出了区域农牧系统耦合程度的演变规律。结果表明,榆阳区是偏农型的农牧耦合产业发展模式,乌审旗是偏牧型的牧农耦合产业发展模式;榆阳区有以耗粮型生猪、禽类生产为主的势头,乌审旗有以节粮型草食动物生产为主的势头;榆阳区和乌审旗的初级生产力、次级生产力及A(有效能值利用率)、B(单位低质能产品转化成高质能产品率)、SCD(系统耦合度)值均沿时间序列增大,且SCD值有呈三阶段型增长的规律性。依据研究结果,最后提出了毛乌素沙地农牧生态系统耦合发展的途径,以期能为沙地农牧生态系统稳健发展提供可靠的理论支撑与政策导向。  相似文献

Chromosomes from ten human male fibroblast metaphases were completely reconstructed from electron micrographs of serially sectioned material. Chromosome centromere positions were determined by finding the three-dimensional coordinates of the centromere midpoint. The data set showed the identity of nine chromosome types (chromosomes 1, 2, 3, 6, 9, 16, 17, 18 and the Y chromosome) preserved as they are positioned in vivo. The results indicate that there is (1) no significant association of the homologous chromosomes examined, (2) a significant tendency for a central location of the Y chromosome and of chromosome 18, (3) a significant tendency for a peripheral location of chromosome 6, (4) no significant tendency for homologous chromosomes to reorganize as metaphase advances and (5) no significant differential condensation across the metaphase plate. Therefore, the only organization pattern observed for the centromeres of the homologous chromosomes studied is some sorting by size across the metaphase plate. These results may be typical of dividing cell types. Different chromosome arrangements are found in some non-dividing cell types (e.g. mammalian brain cells). The different distributions of chromosomes in different cell types can be considered as forms of nuclear differentiation. It is postulated that nuclear differentiation may be related to cell differentiation.  相似文献

Within the framework of discussion about the informative value of the method of coherent analysis, results of successful clinical application of this method of EEG processing in different morbid conditions are reviewed. Not denying the theses proposed by the opponent about the imperfection of methodological maintenance of the EEG coherent analysis and incompatibility of the results received with different software, the ways of overcoming these difficulties are specified. First of all, it is offered to unify procedural conditions of the technique. The thesis of the opponent that calculation errors are influenced by hypothetical "noise" (all signals of the environment) is regarded skeptically, as it could not be found in patients with "zero" coherence with the diagnosis of brain death. Some doubts are cast upon the prospect of replacement of the coherent analysis with an artificial envelope of the modulated groups of waves in assessment of synchronous processes of the brain. It is concluded that all technological and methodological problems of application of the EEG coherent analysis existing now can be overcome.  相似文献

Human homogamy may be caused in part by individuals' preference for phenotypic similarities. Two types of preference can result in homogamy: individuals may prefer someone who is similar to themselves (self-referent phenotype matching) or to their parents (a sexual-imprinting-like mechanism). In order to examine these possibilities, we compare faces of couples and their family members in two ways. First, "perceived" similarity between a pair of faces is quantified as similarity ratings given to the pair. Second, "physical" similarity between two groups of faces is evaluated on the basis of correlations in principal component scores generated from facial measurements. Our results demonstrate a tendency to homogamy in facial characteristics and suggest that the tendency is due primarily to self-referent phenotype matching. Nevertheless, the presence of a sexual-imprinting-like effect is also partially indicated: whether individuals are involved in facial homogamy may be affected by their relationship with their parents during childhood.  相似文献

Caudal nasal deviation, manifested by a "crooked tip," asymmetric nostrils, and a deviated columella, is one of the most challenging deformities encountered in rhinoplasty. This entity is often ignored by rhinoplasty surgeons, on the basis of the assumption that correction of other segments of the deviated nose will improve the caudal nose. Failure to correct this imperfection (or, occasionally, deformity) invariably produces suboptimal results. The nasal structures involved in caudal nasal deviation, namely, the septum, the lower lateral cartilages, and the anterior nasal spine, must be evaluated for identification of the anatomical blocks that have a causative role in caudal nasal deviation. The specific structures with abnormalities related to this deformity are discussed, as are techniques for the correction of the deformities. These techniques significantly augment the surgeon's repertoire of methods for addressing the subtleties of caudal nasal deviation correction and achieving predictable results.  相似文献

The affect misattribution procedure (AMP) was proposed as a technique to measure an implicit attitude to a prime image [1]. In the AMP, neutral symbols (e.g., a Chinese pictograph, called the target) are presented, following an emotional stimulus (known as the prime). Participants often misattribute the positive or negative affect of the priming images to the targets in spite of receiving an instruction to ignore the primes. The AMP effect has been investigated using behavioral measures; however, it is difficult to identify when the AMP effect occurs in emotional processing—whether the effect may occur in the earlier attention allocation stage or in the later evaluation stage. In this study, we examined the neural correlates of affect misattribution, using event-related potential (ERP) dividing the participants into two groups based on their tendency toward affect misattribution. The ERP results showed that the amplitude of P2 was larger for the prime at the parietal location in participants showing a low tendency to misattribution than for those showing a high tendency, while the effect of judging neutral targets amiss according to the primes was reflected in the late processing of targets (LPP). In addition, the topographic pattern analysis revealed that EPN-like component to targets was correlated with the difference of AMP tendency as well as P2 to primes and LPP to targets. Taken together, the mechanism of the affective misattribution was closely related to the attention allocation processing. Our findings provide neural evidence that evaluations of neutral targets are misattributed to emotional primes.  相似文献

Biological individuality is a major topic of discussion in biology and philosophy of biology. Recently, several objections have been raised against traditional accounts of biological individuality, including the objections of monism (the tendency to focus on a single individuality criterion and/or a single biological field), theory-centrism (the tendency to discuss only theory-based individuation), ahistoricity (the tendency to neglect what biologists of the past and historians of biology have said about biological individuality), disciplinary isolationism (the tendency to isolate biological individuality from other scientific and philosophical domains that have investigated individuality), and the multiplication of conceptual uncertainties (the lack of a precise definition of “biological individual” and related terms). In this introduction, I will examine the current philosophical landscape about biological individuality, and show how the contributions gathered in this special issue address these five objections. Overall, the aim of this issue is to offer a more diverse, unifying, and scientifically informed conception of what a biological individual is.  相似文献

To estimate the space-radiation effects separately from other space-environmental effects such as microgravity, frozen human lymphoblastoid TK6 cells were sent to the "Kibo" module of the International Space Station (ISS), preserved under frozen condition during the mission and finally recovered to Earth (after a total of 134 days flight, 72 mSv). Biological assays were performed on the cells recovered to Earth. We observed a tendency of increase (2.3-fold) in thymidine kinase deficient (TK(-)) mutations over the ground control. Loss of heterozygosity (LOH) analysis on the mutants also demonstrated a tendency of increase in proportion of the large deletion (beyond the TK locus) events, 6/41 in the in-flight samples and 1/17 in the ground control. Furthermore, in-flight samples exhibited 48% of the ground-control level in TK(-) mutation frequency upon exposure to a subsequent 2 Gy dose of X-rays, suggesting a tendency of radioadaptation when compared with the ground-control samples. The tendency of radioadaptation was also supported by the post-flight assays on DNA double-strand break repair: a 1.8- and 1.7-fold higher efficiency of in-flight samples compared to ground control via non-homologous end-joining and homologous recombination, respectively. These observations suggest that this system can be used as a biodosimeter, because DNA damage generated by space radiation is considered to be accumulated in the cells preserved frozen during the mission, Furthermore, this system is also suggested to be applicable for evaluating various cellular responses to low-dose space radiation, providing a better understanding of biological space-radiation effects as well as estimation of health influences of future space explores.  相似文献

The responsibility to return "value" to those who support basic research is an obligatory part of accepting funds to support the research. This reality should, but now does not, impact planning and execution of all basic research from its earliest stages. Universities are becoming ever more important in their role in the accelerating quest of a national goal of transition to a "knowledge-based economy." As such, the complex organizational format of a university, laden with entrenched procedures of questionable utility, should be adjusted to identify the means to commercialize the small subset of projects that appear suitable for further development. Of special concern is the growing tendency to encourage academic "innovators" to develop spin-out companies "on the side." While seductive in perceived simplicity, this is a difficult step and we believe that most such individuals are ill-suited to these activities. Not because of technical ability but because of lack of relevant management experience. We attempt to address that situation through a brief listing of some reasons why people "do research" and outline phases (steps) in moving from concept to application, including an overview of start-up and funding early-stage spin-outs. A discussion of the limits to applying results of basic research to enhancing sperm fertility in commodity and companion animals and humans is provided. Hurdles are so daunting that there is concern as to why anyone would attempt to translate basic observations into practical solutions; which in turn raises the question of why funding agencies should fund basic studies in the first place.  相似文献
