Tamarix ramosissima and Tamarix chinensis are invasive weed species in western North America. Previous studies based on single locus DNA sequence data revealed some information about the invasion process, but multilocus markers can provide additional information about levels of introgression and genotype origins. We have developed primers that amplify 10 polymorphic microsatellite loci from T. ramosissima; these primer pairs also successfully amplify polymorphic microsatellites from the closely related T. chinensis, a species that forms hybrids with T. ramosissima in the western USA.  相似文献

Dominance of non-native riparian trees in western USA   总被引:1,自引:0,他引:1  
Concern about spread of non-native riparian trees in the western USA has led to Congressional proposals to accelerate control efforts. Debate over these proposals is frustrated by limited knowledge of non-native species distribution and abundance. We measured abundance of 44 riparian woody plants at 475 randomly selected stream gaging stations in 17 western states. Our sample indicates that Tamarix ramosissima and Elaeagnus angustifolia are already the third and fourth most frequently occurring woody riparian plants in the region. Although many species of Tamarix have been reported in the region, T. ramosissima (here including T. chinensis and hybrids) is by far the most abundant. The frequency of occurrence of T. ramosissima has a strong positive relation with the mean annual minimum temperature, which is consistent with hypothesized frost sensitivity. In contrast the frequency of occurrence of E. angustifolia decreases with increasing minimum temperatures. Based on mean normalized cover, T. ramosissima and E. angustifolia are the second and fifth most dominant woody riparian species in the western USA. The dominance of T. ramosissima has been suspected for decades; the regional ascendance of E. angustifolia, however, has not previously been reported.  相似文献

叶片是植物获取资源的重要器官,研究其功能性状与环境因子的关系,有助于更好地了解植物对环境要素变化的生态适应性。以塔里木盆地北缘不同生境下(绿洲、过渡带、荒漠)典型荒漠植物多枝柽柳(Tamarix ramosissima)为研究对象,分析其叶片结构和生理性状在不同生境间的差异,建立叶片性状与土壤理化因子的相关关系,解析荒漠植物的生态适应性并揭示影响荒漠植物叶片功能性状的关键环境因子。结果表明:(1)多枝柽柳叶片功能性状具有不同程度的变异,其中叶片面积变异幅度最大(40%),比叶面积、可溶性蛋白含量等变异幅度最小(均为15%)。(2)多枝柽柳叶片厚度和叶片面积等结构性状以及叶片可溶性蛋白、脯氨酸、淀粉含量等生理性状在不同生境中均具有显著差异(P<0.05),其中可溶性蛋白、脯氨酸含量均在荒漠生境中达到最高。(3)多枝柽柳叶片部分功能性状之间存在显著的相关关系,其中叶片厚度、淀粉含量等与叶组织密度呈显著负相关关系(P<0.05),而叶片厚度与淀粉含量之间、可溶性糖含量与非结构性碳水化合物含量之间均呈极显著正相关关系(P<0.01)。(4)通过RDA排序分析发现,土壤速效磷含...  相似文献

目前胡杨更新困难,种群处于退化阶段,而与其生态位高度重叠的多枝柽柳却在扩张。为探讨多枝柽柳对胡杨生长的影响,本研究设置了3个水平的水分、盐分梯度,对胡杨进行了单种和混种的盆栽控制实验,通过测定胡杨幼苗的生长和存活状况,分析不同水、盐梯度下多枝柽柳如何影响胡杨生长。结果表明:(1)多枝柽柳伴生降低当年生胡杨幼苗的存活率,随着水分条件改善,胡杨存活率提高,而盐分对存活率没有显著的影响。(2)水分、盐分和伴生模式几个因子对胡杨的生长特征的影响存在显著的交互作用(P<0.05)。各水盐条件下,多枝柽柳会不同程度降低胡杨幼苗的株高、主根长和地上地下生物量,而根冠比增大。混种条件下,胡杨幼苗对水盐的响应更为敏感。(3)相对竞争强度对水盐环境有明显的响应(P<0.05),随水分条件改善,地上、地下相对竞争强度降低。多枝柽柳对胡杨的生长产生不良影响的机制是通过快速消耗土壤的水分,从而导致胡杨幼苗水分亏缺。土壤水分条件是胡杨和多枝柽柳幼苗共存的关键因素,在水分供给不足的情况下,对水分偏好的差异导致胡杨无法通过实生苗进行有效的更新从而加剧胡杨种群的衰退,而多枝柽柳更为耐旱的特性使其逐步占据河岸的生境。我们的研究结果强调了两个树种对环境因子的适应性差异决定了河岸带植被发育过程中植物-土壤的相互作用导致胡杨在演替过程中表现出的衰退现象。  相似文献

Hybrids between female Trichinella spiralis and male Trichinella britovi were constructed. Then, hybrid genotype was characterized by DNA markers including mitochondrial cytochrome c oxidase subunit I (CO I) gene, the gene encoding the 43-kDa excretory–secretory (ES) protein, and genomic DNA fragments specific for T. spiralis and T. britovi identified from random amplified polymorphism DNA (RAPD). PCR-restriction fragment length polymorphism (PCR-RFLP) analysis of the mitochondrial CO I gene revealed that all hybrids carried a T. spiralis pattern. The same analysis of the gene encoding the 43-kDa ES protein showed that each hybrid carried both T. spiralis and T. britovi gene type simultaneously. In the analysis of genomic DNA using RAPD-derived PCR primers, some hybrids carried T. spiralis and T. britovi-specific RAPD markers, while others carried the RAPD marker of T. spiralis only.  相似文献

Summary In cultivated beet no useful level of resistance of the beet cyst nematode (BCN) Heterodera schachtii Schm. has been found, unlike the situation in wild species of the section Procumbentes. Stable introgression of resistance genes from the wild species into Beta vulgaris has not been achieved, but resistant monosomic additions (2n =18 + 1), diploids of B. vulgaris with an extra alien chromosome carrying the resistance locus, have been obtained. Here we describe a new series of resistant monosomic fragment addition material of B. patellaris chromosome 1 (pat-1). We further describe the cloning of a single-copy DNA marker that specifically hybridizes with a monosomic addition fragment of approximately 8 Mb (AN5-90) carrying the BCN resistance locus. This marker and another fragment-specific, single-copy DNA marker probably flank the BCN locus on the addition fragment present in the AN5-203 material, which is approximately 19 Mb in size. Furthermore, several specific repetitive DNA markers have been isolated, one of which hybridizes to AN5-90 and also to DNA from a smaller DNA segment of Beta procumbens, present in line B883, carrying a BCN resistance locus introgressed into the B. vulgaris genome. This suggests that the specific repetitive marker is closely linked to the BCN locus.  相似文献

During 1993, potted plants ofTamarix ramosissimaC. F. Ledebour (saltcedar) from the United States of America andT. gallicaL. from France were placed underTamarix gallicatrees at Sète and Vendémian, southern France, and the natural enemies that attacked these plants were recorded. The most promising natural enemies found were three species ofPsectrosema(Diptera: Cecidomyiidae);Hypophyes pallidulus(Gravenhorst),Corimalia tamarisci(Gyllenhal), andConiatus tamarisciF. (Coleoptera: Curculionidae);Stylosomusprob. tamaricis (Herrich-Schaffer) (Coleoptera: Chrysomelidae); andAgdistis tamaricis(Zeller) (Lepidoptera: Pterophoridae). The host specificity of these insects was studied in a field test during 1994. Of the 11 plant species tested, none of the economically important plants and neitherTamarix aphylla(L.) nor any of the nine genera of plants outside the family Tamaricaceae were attacked. The above mentioned natural enemies seem to be specific feeders on saltcedar and are recommended as prospective agents for biological control ofT. ramosissimain the United States.  相似文献

Natural hybridization and introgression is recorded between the newt species Triturus vulgaris and T. montandoni in Slovakia. To confirm a hybrid status of two putative hybrids, morphological and Randomly Amplified Polymorphic DNA (RAPD) markers specific for T. vulgaris and T. montandoni were used. The individuals found in Zbojská (Veporské vrchy Mts, Slovakia; analyzed morphologically and genetically) and Zubrohlava (Oravská kotlina basin, Slovakia; analyzed only morphologically) possessed markers of both species. Segregation of RAPD markers together with a model-based Bayesian analysis revealed that the specimen from Zbojská belonged to later generation hybrids.  相似文献

Summary Mesophyl protoplasts of two genotypes of cultivated tomato (Lycopersicon esculentum Mill.) and one of its wild relative species (Lycopersicon peruvianum Mill.) were fused by using electrofusion and polyethyleneglycol-induced fusion. Forty-three fertile tetraploid somatic hybrid plants, each deriving from separate calli, were recovered from both fusion procedures. Electrofusion appeared more efficient than chemical fusion for the production of somatic hybrids. These plants appeared morphologically similar, whatever the fusion procedure and tomato genotype. They had intermediate leaf, inflorescence, and flower morphology. After self-pollination, the hybrids set fruit of intermediate size and color. The hybrid nature of these plants was confirmed by isoelectric focusing of the Rubisco small subunits used as nuclear markers. L. esculentum and L. peruvianum were distinguished by means of two chloroplast markers: CF1-ATPase subunit as analyzed by isoelectro-focusing and ct DNA restriction patterns. All hybrids displayed both ct markers of only one parent with no biased transmission. Mitochondrial (mt) DNAs were prepared from flower buds by using miniaturized CsCl gradients. Preliminary analysis indicated that mt genomes from the hybrids all differed from those of both parents. mt DNA Sall restriction enzyme analysis revealed that all but two hybrids contained one novel fragment of 13.5 kb. Gene mapping experiments showed that the mt apocytochrome b and ATPase subunit 9 homologies in the somatic hybrid mt DNA resembled L. esculentum and L. peruvianum, respectively; the mt nad5 probe distinguished at least four distinct patterns in the hybrids. These results indicated that mt DNA rearrangements involving intergenomic recombinations occurred through protoplast fusion. A greater mt DNA polymorphism was induced with chemical fusion than with electrofusion.  相似文献

F2 progeny (105 individuals) from the cross Jo4002 x Sv3402 were used to identify DNA markers associated with palmitic-acid content in spring turnip rape (Brassica rapa ssp. oleifera). QTL mapping and ANOVA analysis of 140 markers exposed one linkage group with a locus controlling palmitic-acid content (LOD score 27), and one RAPD (random amplified polymorphic DNA) marker, OPB-11a, closely linked (1.4 cM) to this locus. Palmitic-acid content in the 62 F2 plants with the visible allele of marker OPB-11a was 8.45 ±3.15%, while that in the 24 plants without it was 4.59 ±0.97%. As oleic-acid concentration is affected by a locus on the same linkage group as the palmitic-acid locus, this locus probably controls the chain elongation from palmitic acid to oleic acid (through stearic acid). Marker OPB-11a may be used in future breeding programs of spring turnip rape to simplify and hasten the selection for palmitic-acid content.  相似文献

Simmondsia chinensis (Link) Schneider, a multipurpose and monogeneric dioecious shrub from arid zones, has emerged as a cash crop all over the globe. Its seed propagation poses severe problems due to its male-biased population: the male:female ratio is 5:1. Investigations have been carried out to generate a sex-specific Inter-simple sequence repeat (ISSR) marker for the early detection of male and female plants. Of the 42 primers analysed with a bulk sample of pooled male DNA and a bulk sample of pooled female DNA, only one primer, UBC-807, produced a unique ~1,200 base-pair fragment in the male DNA. To validate this observation, this primer was re-tested with individual male and female samples from eight cultivars. A similar unique ~1,200 bp fragment was present in the male individuals of all eight cultivars and completely absent in the female individuals tested. This is the first report of the use of ISSR markers to ascertain sex in physiologically mature S. chinensis plants.  相似文献

Many Oreochromis species utilized in aquaculture were extensively introduced outside their native range in Africa. Given their recent evolutionary radiation, these species hybridize easily, posing a threat to the integrity of local adaptation. The objective of this work was to study the genetic diversity of the Mozambique tilapia (Oreochromis mossambicus) in its native range, southern Africa, and provide a method for identifying hybrids with genetic markers. We genotyped the mitochondrial DNA (mtDNA) control region (385 bp) of wild and farmed O. mossambicus, wild and farmed O. niloticus and morphologic wild hybrids. These data were complemented with published sequences of parapatric and sympatric Oreochromis taxa. Phylogeographic analysis showed the presence of two O. mossambicus lineages, the southernmost representing a recent Holocene radiation. Hybridization of O. mossambicus was indicated by the presence of O. niloticus and O. mortimeri–andersonii mtDNA specimens in the Limpopo basin and of O. karongae mtDNA in specimens from Malawi. We also genotyped seven suspected hybrid individuals from the Limpopo River, and 137 wild and farmed Mozambique and Nile tilapia samples with five microsatellite markers. Factorial Component Analysis, Bayesian clustering and assignment analyses consistently delineated an O. mossambicus and an O. niloticus group, with the putative hybrids positioned in between. Different levels of hybridization were detected by the Bayesian assignment. The complex nature of hybridization and introgression between cichlid species raises major concerns for the long-term integrity of Mozambique tilapia.  相似文献

A population of diploid potato (Solanum tuberosum) was used for the genetic analysis and mapping of a locus for resistance to the potato cyst nematode Globodera rostochiensis, introgressed from the wild potato species Solanum vernei. Resistance tests of 108 genotypes of a F1 population revealed the presence of a single locus with a dominant allele for resistance to G. rostochiensis pathotype Ro1. This locus, designated GroV1, was located on chromosome 5 with RFLP markers. Fine-mapping was performed with RAPD and SCAR markers. The GroV1 locus was found in the same region of the potato genome as the S. tuberosum ssp. andigena H1 nematode resistance locus. Both resistance loci could not excluded to be allelic. Tamarisk (Tamarix ramosissima Ledeb., T. chinensis Lour.), a pernicious invader of riparian areas in the western U.S., is often considered to be allelopathic by virtue of an ability to exude salt from its leaves. However, there is no evidence demonstrating allelopathy or even salinization of soils under tamarisk. We collected soil samples from beneath and just outside of tamarisk canopies at 12 sites along a 110-km reach of Fort Peck Reservoir in northeastern Montana. Samples were analyzed for electrical conductivity (EC), pH and concentration of several nutrients. Plants of western wheatgrass (Agropyron smithii Rydb.), the dominant herbaceous native plant in habitats invaded by tamarisk, were grown in the soil samples as a bioassay. We used principal components analysis (PCA) to explore how intercorrelated soil variables were affected by a tamarisk canopy. EC averaged more than twice as high under a tamarisk canopy than in the open. However, western wheatgrass plants growing in tamarisk-affected soil were 73% larger than those in unaffected soil. PCA analysis indicated that soil beneath a tamarisk canopy had higher values along a gradient of increasing EC and total inorganic N and decreasing pH and NH 4 + /NO 3 ratio. This same gradient was also associated with increasing growth of the bioassay. We hypothesize that this fertilization was due to leaf secretions and/or leaf fall, possibly by increased input of nitrogenous organic compounds or increased P solubility. Our study provided no support for an allelopathic effect of tamarisk.  相似文献

Throughout its native range, the Yellowstone cutthroat trout (YCT), Oncorhynchus clarkii bouvieri, is declining dramatically in both abundance and distribution as a result of introgression with introduced rainbow trout (RBT), O. mykiss. We sampled over 1,200 trout from the South Fork of the Snake River (SFSR) watershed, in southeastern Idaho and western Wyoming, and measured the extent of introgression of RBT genes into native gene pools of YCT using seven species-specific, co-dominant nuclear genetic markers. We also used mitochondrial DNA (mtDNA) haplotype differences between the two parental trout species to determine the directionality of the hybridization. We found low levels of RBT introgression (only 7% of sampled individuals had one or more RBT alleles) into YCT gene pools, with the majority of hybrids (78%) occurring in mainstem localities of the SFSR and in lower elevation reaches of certain tributaries. Hybridization was bidirectional with respect to mtDNA haplotype, but the majority of hybrids (75%) had YCT maternal haplotypes, indicative of the greater proportion (90%) of YCT-genotypes in the SFSR watershed. The primary factor influencing the geographic distribution of RBT introgressed individuals was fluvial distance from localities of stocking origin. To a lesser extent, elevation, also influenced the distribution of hybrid genotypes, with several entire tributaries and all upper elevation reaches within tributaries harboring only YCT-genotypes. Important management implications of the study suggest targeting particular tributaries and upper reaches within tributaries for YCT protection and exclusion of RBT hybrid colonization.  相似文献

Breeding resistant apple plants is an alternative way to control fungal pathogens reducing the environmental impact due to the use of pesticides. The breeding of apple cultivars resistant to Venturia inaequalis could be much improved by marker-assisted selection. A molecular marker closely linked to the resistance locus called Vf could replace selection based on infection studies. To find such molecular markers, DNA of progenies from crossings of a resistant and a susceptible apple tree was subject to bulked segregant analysis. Two markers were found with a genetic distance of 10.6% and 19.7% recombination frequency to the Vf locus.  相似文献

 Two sex-linked random amplified polymorphic DNA (RAPD) markers identified from Actinidia chinensis were converted into sequence-characterised amplified regions (SCARs) for the large-scale screening of Actinidia breeding populations. Initial SCAR primers converted one RAPD (SmX) into a dominant marker, but the other (SmY), which was potentially more useful because of its linkage to the male determining ‘Y’ locus, failed to retain polymorphism. This difficulty was overcome by cloning and sequencing the alternate ‘allele’ from female plants, and then designing ‘allele’-specific primers that utilised nucleotide differences between the sexes. Using a quick squash-blot method of DNA extraction, the SCAR primers were tested in 120 A. chinensis plants to determine their gender. The system is now in use for large-scale screening of seedling populations in the Actinidia breeding programme. The sex-linked SCAR primers also functioned with plants from some other geographically separate accessions of A. chinensis and with plants in the closely related polyploid species A. deliciosa, but did not amplify a sex-linked band in more distantly related species of Actinidia. Hybridization is regarded as a rapid mechanism for increasing genetic variation that can potentially enhance invasiveness. Tamarix hybrids appear to be the dominant genotypes in their invasions. Exotic Tamarix are declared invasive in South Africa and the exotic T. chinensis and T. ramosissima are known to hybridize between themselves, and with the indigenous T. usneoides. However, until now, it was not known which species or hybrid is the most prevalent in the invasion. With a biocontrol programme being considered as a way of suppressing the alien Tamarix populations, it is important to document the population genetic dynamics of all species in the region. This investigation sought to identify Tamarix species in southern Africa and their hybrids, describe their population structure, and reveal the geographic origin of the invasive species. To achieve this, nuclear Internal Transcribed Spacer (ITS) sequence data and the multilocus Amplified Fragment Length Polymorphisms (AFLPs) markers were used. Phylogenetic analysis and population genetic structure confirmed the presence of three species in South Africa (T. chinensis, T. ramosissima and T. usneoides) with their hybrids. The indigenous T. usneoides is clearly genetically distant from the alien species T. chinensis and T. ramosissima. Interestingly, the Tamarix infestation in South Africa is dominated (64.7 %) by hybrids between T. chinensis and T. ramosissima. The exotic species match their counterparts from their places of origin in Eurasia, as well as those forming part of the invasion in the US.  相似文献

Populus nigra is considered a rare and threatened tree species in Switzerland because of dramatic habitat loss owing to river regulations during the last two centuries and because of potential gene introgression from non-indigenous P. deltoides through planted P. x canadensis hybrids. The significance of introgression as an endangerment to P. nigra, however, is controversial. The aims of the present study were (1) to assess how abundant P. nigra trees are in Switzerland and (2) to assess potential gene introgression. We present data from a molecular survey of 1372 putative P. nigra trees from Switzerland, using both chloroplast DNA and nuclear DNA markers. The results show that P. nigra is more abundant in Switzerland than hitherto thought. Furthermore, we detected a low frequency of gene introgression.  相似文献

To explore the roles of plasticity and genetic variation in the response to spatial and temporal climate variation, we established a common garden consisting of paired collections of native and introduced riparian trees sampled along a latitudinal gradient. The garden in Fort Collins, Colorado (latitude 40.6°N), included 681 native plains cottonwood (Populus deltoides subsp. monilifera) and introduced saltcedar (Tamarix ramosissima, T. chinensis and hybrids) collected from 15 sites at 29.2–47.6°N in the central United States. In the common garden both species showed latitudinal variation in fall, but not spring, leaf phenology, suggesting that the latitudinal gradient in fall phenology observed in the field results at least in part from inherited variation in the critical photoperiod, while the latitudinal gradient in spring phenology observed in the field is largely a plastic response to the temperature gradient. Populations from higher latitudes exhibited earlier bud set and leaf senescence. Cold hardiness varied latitudinally in both fall and spring for both species. For cottonwood, cold hardiness began earlier and ended later in northern than in southern populations. For saltcedar northern populations were hardier throughout the cold season than southern populations. Although cottonwood was hardier than saltcedar in midwinter, the reverse was true in late fall and early spring. The latitudinal variation in fall phenology and cold hardiness of saltcedar appears to have developed as a result of multiple introductions of genetically distinct populations, hybridization and natural selection in the 150 years since introduction.  相似文献
