Strains of Phytophthora infestans of the same race (P4) isolated from a range of R0 potato varieties differed markedly in their rate of growth on agar but differences in growth rate were not related to variety of origin. Two isolates from each of the varieties Duke of York, Majestic and Arran Consul were grown for six successive subcultures on tubers of these three varieties. The six isolates differed in aggressiveness as shown by their growth on individual varieties. There was a marked difference in growth rate between alternate subcultures which is attributed to the use of too long an incubation period. Analysis of the data for subcultures 1, 3 and 5 revealed a significant interaction between isolates and varieties which reflected the enhanced growth of five of the isolates on the variety from which they had been obtained. Serial subculture on potato tubers of the same variety had no general or specific effect on the aggressiveness of the isolates. It is concluded that strains of P. infestans of the same race may be differentially adapted to the variety on which they are growing in the field. Differential interactions between pathogen biotypes and non-immune hosts are considered to be widespread in host-pathogen systems.  相似文献

The temperature dependence of UV effects was studied for Arctic and temperate isolates of the red macrophytes Palmaria palmata, Coccotylus truncatus and Phycodrys rubens. The effects of daily repeated artificial ultraviolet B and A radiation (UVBR: 280–320?nm, UVAR: 320–400?nm) treatments were examined for all isolates at 6, 12 and 18?°C by measuring growth, optimal quantum yield of PSII (Fv/Fm) and cyclobutane-pyrimidine dimer (CPD) accumulation. Furthermore, possible ecotypic differences in UV sensitivity between Arctic and temperate isolates were evaluated. Large species-specific differences in UV sensitivity were observed for all parameters: the lower subtidal species C. truncatus and P. rubens were highly sensitive to the UV treatments, whereas P. palmata, which predominantly occurs in the upper subtidal zone, was not affected by these treatments. Only minor differences were found between Arctic and temperate isolates, suggesting that no differences in UV sensitivity have evolved in these species. Relative growth rates were temperature-dependent, whereas species-specific UV effects on growth rates were relatively independent of temperature. In contrast, the species-specific decrease in Fv/Fm and its subsequent recovery were temperature-dependent in all species. UV effects on Fv/Fm were lower at 12 and 18?°C compared with 6?°C. In addition, UV effects on Fv/Fm decreased in the course of the experiment at all temperatures, indicating acclimation to the UV treatments. CPDs accumulated during the experiment in both isolates of P. rubens, whereas CPD concentrations remained low for the other two species. CPD accumulation appeared to be independent of temperature. The results suggest that summer temperatures occurring in temperate regions facilitate repair of UV-induced damage and acclimation to UV radiation in these algae compared with Arctic temperatures. Because the differences in UV effects on Fv/Fm, growth and CPD accumulation were relatively small over a broad range of temperatures, it was concluded that the influence of temperature on UV effects is small in these species.  相似文献

 Plant ability to withstand acidic soil mineral deficiencies and toxicities can be enhanced by root-arbuscular mycorrhizal fungus (AMF) symbioses. The AMF benefits to plants may be attributed to enhanced plant acquisition of mineral nutrients essential to plant growth and restricted acquisition of toxic elements. Switchgrass (Panicum virgatum L.) was grown in pHCa (soil:10 mM CaCl2, 1 : 1) 4 and 5 soil (Typic Hapludult) inoculated with Glomus clarum, G. diaphanum, G. etunicatum, G. intraradices, Gigaspora albida, Gi. margarita, Gi. rosea, and Acaulospora morrowiae to determine differences among AMF isolates for mineral acquisition. Shoots of mycorrhizal (AM) plants had 6.2-fold P concentration differences when grown in pHCa 4 soil and 2.9-fold in pHCa 5 soil. Acquisition trends for the other mineral nutrients essential for plant growth were similar for AM plants grown in pHCa 4 and 5 soil, and differences among AMF isolates were generally higher for plants grown in pHCa 4 than in pHCa 5 soil. Both declines and increases in shoot concentrations of N, S, K, Ca, Mg, Zn, Cu, and Mn relative to nonmycorrhizal (nonAM) plants were noted for many AM plants. Differences among AM plants for N and Mg concentrations were relatively small (<2-fold) and were large (2- to 9-fold) for the other minerals. Shoot concentrations of mineral nutrients did not relate well to dry matter produced or to percentage root colonization. Except for Mn and one AMF isolate, shoot concentrations of Mn, Fe, B, and Al in AM plants were lower than in nonAM plants, and differences among AM plants for these minerals ranged from a low of 1.8-fold for Fe to as high as 6.9-fold for Mn.  Nitrogen deposition and intentional forest fertilisation with nitrogen are known to affect the species composition of ectomycorrhizal fungal communities. To learn more about the mechanisms responsible for these effects, the relations between fungal growth, nitrogen uptake and nitrogen availability were studied in ectomycorrhizal fungi in axenic cultures and in symbiosis with pine seedlings. Effects of different levels of inorganic nitrogen (NH4) on the mycelial growth of four isolates of Paxillus involutus and two isolates of Suillus bovinus were assessed. With pine seedlings, fungal uptake of 15N-labelled NH4 was studied in short-term incubation experiments (72 h) in microcosms and in long-term incubation experiments (3 months) in pot cultures. For P. involutus growing in symbiosis with pine seedlings, isolates with higher NH4 uptake were affected more negatively at high levels of nitrogen availability than isolates with lower uptake. More NH4 was allocated to shoots of seedlings colonised by a high-uptake isolate, indicating transfer of a larger fraction of assimilated NH4 to the host than with isolates showing lower NH4 uptake rates. Thus low rates of N uptake and N transfer to the host may enable EM fungi avoid stress induced by elevated levels of nitrogen. Seedlings colonised by S. bovinus transferred a larger fraction of the 15N label to the shoots than seedlings colonised by P. involutus. Seedling shoot growth probably constituted a greater carbon sink in pot cultures than in microcosms, since the mycelial growth of P. involutus was more sensitive to high NH4 in pots. There was no homology in mycelial growth rate between pure culture and growth in symbiosis, but N uptake in pure culture corresponded to that during growth in symbiosis. No relationship was found between deposition of antropogenic nitrogen at the sites of origin of the P. involutus isolates and their mycelial growth or uptake of inorganic nitrogen. Clonal cultures of plankton are widely used in laboratory experiments and have contributed greatly to knowledge of microbial systems. However, many physiological characteristics vary drastically between strains of the same species, calling into question our ability to make ecologically relevant inferences about populations based on studying one or a few strains. This study included 19 non-axenic strains of three species of the diatom Pseudo-nitzschia isolated primarily from the mid-Atlantic coastal region of the United States. Toxin (domoic acid) production and growth rates were measured in cultures using different nitrogen sources (NH4+, NO3 and urea) and growth irradiances. The strains exhibited broad differences in growth rate and toxin content even between strains isolated from the same water sample. The influence of bacteria on toxin production was not investigated. Both P. multiseries clones produced toxin, yet preferentially used different nitrogen sources. Only two of nine P. calliantha and two of five P. fraudulenta isolates were toxic and domoic acid content varied by orders of magnitude. All three species had variable intraspecies growth rates on each nitrogen source, but P. fraudulenta strains had the broadest range. Light-limited growth rate and maximum growth rate in P. fraudulenta and P. multiseries varied with species. These findings show the importance of defining intra- and interspecies variability in ecophysiology and toxicity. Ecologically relevant functional diversity in the form of ecotypes or cryptic species appears to be present in the genus Pseudo-nitzschia.  相似文献

Lake Averno sediment was used to isolate the facultative anaerobic bacteria having the potential for H2 production. Twenty-five out of 35 isolates recovered from the sediment sample produced hydrogen under anaerobic conditions from glucose with yields ranging from 0.1 to 0.49 mol H2/mol glucose. Identification based on 16S rRNA gene sequence analysis revealed that most of them belong to the Firmicutes group, with a prevalence of the Paenibacillus polymyxa species. Seven distinct genomic fingerprints among the 11 P. polymyxa isolates were obtained using the random amplified polymorphic DNA (RAPD) technique. Glucose fermentation by P. polymyxa isolates was investigated. Glucose was totally consumed after 3 days of fermentation. The fermentation products were hydrogen (0.18–0.47 mol H2/mol glucose), ethanol (0.1–0.5 mol ethanol/mol glucose), and 2,3-butanediol (0.1 mol 2,3-butanediol/mol glucose). Lower amounts of acetic, butyric, formic, lactic, and propionic acids were detected. All metabolic data concerning P. polymyxa isolates were analyzed by cluster analysis to reveal similarities and/or differences with clustering based on RAPD profiles. Despite the high metabolic similarity among almost all P. paenibacillus isolates, results of cluster analyses of metabolic and genetic data do not match completely.  相似文献

Aggressiveness in four isolates of Fusarium head blight (FHB) species (F. culmorum, F. solani, F. verticillioides and F. equiesti) was studied in vitro on six wheat cultivars using a modified Petri-dish test. Results showed differences between cultivars inoculated with FHB isolates and control for three aggressiveness criteria: germination rate reduction, standardised area under disease progress curve (AUDPCstandard), and coleoptile length reduction. Regarding AUDPCstandard and Petri-dish aggressiveness index, significant differences were detected among fungal isolates. The other two aggressiveness criteria: germination rate reduction and coleoptile length reduction did not distinguish between FHB isolates. The Petri-dish test was repeatable and stable method to assess aggressiveness of four FHB species for all tested wheat cultivars. The current study confirmed the suitability of in vitro modified Petri-dish method to be used as fast and reliable test to analyse aggressiveness in FHB species.  相似文献

The pathogenicity and growth rate in vivo were assessed on 27 isolates of Pythium spp. recovered from cavity spot lesions on carrots grown in various parts of northwest France. Polyacrylamide gel electrophoresis of isoesterases was used to identify the Pythium spp. involved. Slow-growing isolates were more aggressive than fast-growing ones when inoculated on carrot tap roots. Isoesterase patterns identified the slow-growing isolates as P. violae and P. sulcatum; P. ultimum and P. intermedium were identified among the less aggressive fast-growing isolate group, in which some isolates were also classed as P. sylvaticum or P. irregulare, which have similar electrophoretic profiles. The incidence of Pythium spp. associated with the disease in France is discussed in regard to cavity spot in other countries.  相似文献

The effect of three different nitrogen sources on the growth of external ectomycorrhizal mycelium was studied in Perspex micorocosms. Nonsterile peat was used as substrate. Five different fungal isolates growing in symbiosis with pine seedlings were investigated: two isolates of Paxillus involutus, one of Suillus bovinus and two unidentified ectomycorrhizal fungi isolated from ectomycorrhizal root tips. Three different nitrogen sources were used: ammonium as (NH4)2SO4, nitrate as NaNO3 and a complete nutrient solution (Ingestad 1979), and three different nitrogen concentrations, 1, 2 or 4 mg N/g dry wt. of peat. The mycelial growth of all fungi was found to be negatively affected by the nitrogen amendments, although the sensitivity to nitrogen varied between the isolates. One of the unidentified isolates was extremely sensitive and growth was completely inhibited by all nitrogen treatments. In contrast, the growth of one of the P. involutus isolates was only slightly reduced by the nitrogen amendments. The different nitrogen sources all reduced growth, and since no significant difference was found between the nitrogen sources or between the different nitrogen concentrations the results were pooled to give one value that summarized the effect of nitrogen on mycelial growth. Thus, the mycelial growth of one of the two P. involutus isolates was reduced to approximately 80% of the growth in the control, the other P. involutus and one of the unidentified fungi, vgk 2 89.10, were reduced to 40–50% of the control growth, S. bovinus to 30% of the control and the most sensitive fungus, the unidentified isolate vg 1 87.10, was reduced to 3% of the growth in the control treatment. In all experiments, the shoot to root ratio generally increased, mainly as a result of increased shoot growth.  相似文献

The aim of the present study was to evaluate the inhibitory effect of Enterococcus faecium and Lactococcus lactis subsp. lactis isolated from faeces of healthy dogs on (i) lag phase, (ii) growth rate, and (iii) aflatoxin B1 production by Aspergillus section Flavi on in vitro assays. Thirteen lactic acid bacteria (LAB) isolates were used as antagonist microorganisms. Antagonistic activity was assayed against four potentially aflatoxigenic Aspergillus section Flavi isolates: A. flavus (AF210 and AF281), A. parasiticus (AP245) and A. parasiticus (NRRL 2999). In general, the longest lag phases of Aspergillus isolates were obtained with E. faecium GJ40. Respecting the growth rate, no significant reduction was found in this parameter in the interaction assays with A. flavus and antagonist isolates respecting the control. While in A. parasiticus a significant reduction in growth rate was only observed in the interaction among reference strain and E. faecium MF5 isolate (p < 0.05). In general, AFB1 production was reduced by most of the LAB isolates assayed, except for E. faecium GJ18, GJ20, MF3 and MF4. This study provides the first data about the antiaflatoxigenic activity of autochthonous LAB isolated from dog faeces.  相似文献

Eighteen indigenous fungal isolates has been successfully isolated from samples of used motor oil, top five centimetres of soil and drainage water contaminated with used motor oil. All of the pure fungal isolates obtained were identified, characterized and subjected to preliminary screening by evaluating the average growth rate of each fungal isolates on minimal media containing 1% (v/v) used motor oil. Trichoderma asperellum strain TUB F-1067 (SA4), Trichoderma asperellum strain Tr48 (SA5), Trichoderma asperellum strain TUB F-756 (SA6), Penicillium species (P1), and Aspergillus species (P9) were further selected for their hydrocarbon biodegradation potential. Among these five fungal isolates selected, P1 strain presented a significant degree of degradation by degrading almost all of the n-alkanes (n-C-15 to n-C-23 range) present in the used motor oil, thus of greater potential in degrading the aliphatic hydrocarbon compounds of used motor oil. Single-spored isolates of Pyrenophora associated with spot and net type net blotch of barley were compared using total DNA banding patterns, morphological and cultural characteristics, symptomatology and mating studies. Isolates of spot and net type net blotch were found to vary regarding conidium length and cultural growth rate. Mating studies among and between ascospore, spot and net type isolates proved unsuccessful under the conditions studied. Total DNA polymorphisms of the net spot and ascospore isolates digested with the restriction enzymes HpaII and HaeIII showed that the isolates have similar banding patterns. Random amplified polymorphic DNA (RAPD) showed that the banding patterns of the spot and net type isolates were again similar but were distinct from outgroups such as P. semeniperda and P. triticirepentis. The homology in DNA banding patterns of local isolates indicated that the difference in conidium length is insufficient to separate them as two species. It is concluded that spot and net type isolates occurring in South Africa belong to P. teres. Therefore spot type lesions are caused by P. teres f. sp. maculata and not by P. japonica as reported previously.  相似文献

This study showed the impact on germination, mycelial growth and aflatoxin B1 accumulation when interacting Aspergillus aflatoxigenic strains with Kluyveromyces isolates and the effect of water activity on this relationship. Isolates Y14 and Y16 reduced the percentage of germination of all Aspergillus strains and decrease germ tube elongation rate at majority of water activity assayed. Similarly they produced an increase of germination lag phase and lag phase of growth beside decreased growth rate of all Aspergillus strains. At water activities 0.994, 0.982, 0.955 and 0.937, no aflatoxins were produced in paired cultures with isolates Y25, Y22, Y16, and Y14, and Kluyveromyces isolates Y14 and Y16 impact both growth and aflatoxin accumulation at wide range of water activity.  相似文献

Isolates of Phytophthora cactorum resistant to the systemic fungicide metalaxyl were obtained by exposing them to sequentially increased concentrations of metalaxyl. A linear relationship was observed between the concentrations of metalaxyl and percentage inhibition of mycelial growth of P. cactorum. The stability of metalaxyl-resistant isolates 150R and 250R was confirmed after six serial transfers on corn meal agar without fungicide. The in vitro metalaxyl-resistant isolate (Ph10) was less aggressive on apple rootstocks compared with the Ph07 isolated from metalaxyl-treated trees and the Ph03 isolated from untreated trees. Metalaxyl-resistant and sensitive isolates remained sensitive to the chemically unrelated fungicide fosetyl-Al at high concentration (600 μg/ml), to mancozeb, and to a mixture of metalaxyl + mancozeb. Significant differences in resistance to metalaxyl existed among P. cactorum field isolates.  相似文献

Twenty-three provenances of Gliricidia sepium and eleven isolines of Leucaena leucocephala were examined at a low and at high phosphate levels (20 and 80 mg P kg-1 soil) for growth, phosphate (P) uptake and use efficiency. Large differences in growth at the low P level, and in growth response to the higher P rate occurred among L. leucocephala isolines and G. sepium provenances. Shoot dry weight at low P varied from 1.30 to 3.01 g plant-1 for L. leucocephala and from 1.44 to 3.07 g plant-1 for G. sepium.Leucaena isolines had only half the root weight of G. sepium provenances yet produced approximately 90% of the shoot weight of the corresponding G. sepium treatments, i.e. more than a 2-fold difference in root/shoot ratios. Total P in shoots of G. sepium was some 85% greater than of the respective L. leucocephala isolines in corresponding treatments. Physiological phosphate use efficiency (g shoot/mg P in shoots) (PPUE) was not a simple reciprocal relation, being markedly lower at higher shoot % P and content. However, for the same shoot P both species produced the same shoot weight. Nevertheless, there were apparent genotypic differences within species in the root development, shoot P and PPUE.In another study, the numbers of rhizobia in the rhizosphere of L. leucocephala, nodulation, N2 fixation at five different levels of P were determined. The numbers of rhizobia in the rhizosphere of inoculated L. leucocephala during the first two weeks were lower when P was added but later became similar to those without added P. Nodules formed earlier than inoculated plants fertilized with P and in greater numbers (4- to 5-fold) and dry weights than in those without P. However, the percentage of N2 derived from fixation did not change with increasing levels of P application. These results suggest that the observed P effect did not operate via stimulated growth of rhizobia in the rhizosphere, nor through increased N2 fixation rate. The major effect appeared to be due to effects via plant growth.  相似文献

胡杨(Populus euphratica)是塔里木河流域荒漠河岸林的建群种,水分和氮素是限制胡杨幼苗的存活及早期生长的主要因子。利用~(15)N同位素示踪技术分析水和氮素的交互作用对胡杨幼苗不同生长阶段氮素的吸收分配利用及幼苗生长的影响,进一步探究氮素对胡杨实生苗早期形态建成的作用及对干旱胁迫的缓解效应,以期提高幼苗的存活率。实验以一年生胡杨实生幼苗为研究对象,采用温室内盆栽实验,设置4个干旱处理(D_1、D_2、D_3、D_4,土壤相对含水量为:20%—25%、40%—45%、60%—65%、80%—85%)和3种氮素水平(N_0、N_1、N_2:0、3、6 g/盆)测定胡杨幼苗的生长指标和各部分的Ndff、分配率及利用率。结果表明:胡杨幼苗在土壤相对含水量60%—65%(D_3)、氮素添加量3 g/盆(N_1)时,其生长表现为最佳状态;干旱胁迫下,不同氮素添加量对胡杨幼苗各部分的Ndff值存在显著差异,N_2低于N_1;随干旱胁迫减弱(D_3、D_4),植株在生长早期(25 d)根部吸收的~(15)N优先向地上部分转运,生长后期(75 d)植株Ndff最高,其中以根系中Ndff最高;不同生长期幼苗各部分的~(15)N分配存在显著差异,根系~(15)N分配率较高,但不同氮量处理间差异不显著;随生长期的推移,植株对~(15)NH_4~(15)NO_3的利用率表现为粗根最大,各处理中D_3N_1处理均显著高于其他处理。结论:轻度干旱胁迫下添加适量的氮素能够增强植株对氮素的吸收征调能力,优化水资源获取以维持生存的重要机制。  相似文献

Abstract The red turpentine beetle (RTB), Dendroctonus valens LeConte, as a destructive invasive pest, has become one of the most economically important forest pest in China. Effective control measures are desperately needed. Entomopathogenic fungi, such as Beauveria bassiana, have shown great potential for the management of some bark beetle species. In this study, 12 isolates of B. bassiana from bark beetle were examined for biological characteristics and virulence, to assess their potential as biocontrol agents for RTB. There were significant differences (at P= 0.05) in colony growth rate, conidial yield, conidial germination, tolerance to UV light and extracellular proteases activity among the tested B. bassiana isolates. Isolates, including Bb1801, Bb1906, Bb789 and Bb773, exhibited the best characteristics, because they have faster hyphal growth rate, higher spore production and faster spore germination, higher UV tolerance and protease (Pr1) production. The results of a pathogenicity test of B. bassiana on RTB larvae showed that most isolates of B. bassiana have demonstrated high efficacy and the highest virulent isolate was Bb1801, which killed 100% of the treated insects and had a median lethal time (LT50) of 4.60 days at a concentration of 1×107 conidia/mL. Therefore, isolate Bb1801 has a great potential for sustainable control of RTB in the forest. The correlation between biological characteristics and virulence of the fungal isolates is discussed and the possibility of combination of entomopathogenic fungi with semiochemicals, as one of the promising strategy for RTB control, is considered.  相似文献

Twenty-five Pythium isolates comprising five species viz., Pythium aphanidermatum, P. deliense, P. graminicola, P. heterothallicum and P. ultimum from different geographical locations of Tamil Nadu (Coimbatore, 4; Cuddalore, 6; Dindigul, 1; Dharmapuri, 1; Erode, 1; Madurai, 1; Namakkal, 7; Thanjavur, 1; Theni, 1; Thirunelveli, 1 and Vellore, 1) isolated from chilli crop were analysed with randomly amplified polymorphic DNA (RAPD) markers. Morphological and molecular characteristics of these different species were correlated with the RAPD. Polymerase chain reaction amplification of total genomic DNA with six random primers generated unique banding patterns depending on the primer and the isolate. The isolate I17 produced identical banding patterns, while other isolates produced dissimilar bands within the particular species, indicating the genetic diversity among the isolates within a species. Morphological characters were also different from each other even in isolate I17 which shared identical bands. Cluster analysis showed minimum and maximum per cent similarities among the tested Pythium species which ranged from 49 to 89%, respectively. RAPD markers were better suited for differentiating isolates within a species rather than species.  相似文献

Carbon isotope discrimination (A), leaf conductance (gs), photosynthetic capacity, and plant growth were measured in well-watered, glasshouse-grown potato plants of clones from a cross made between diploid Solanum tuberosum and Solanum vernei. Clones showed significant differences (P < 0.001) in gs, Δ, stomatal density, root growth, and total dry matter production. Carbon isotope discrimination of genotypes was positively correlated (P < 0.001) with gs. There was no correlation between gs and stomatal density indicating that differences in gs reflected differences in stomatal aperture. Differences in rooting characteristics or in root/shoot ratio did not contribute to differences in gs or A. Genotypic differences in photosynthetic capacity were not statistically significant, and there was no correlation between A and photosynthetic capacity. Total dry matter production and A were positively correlated (P < 0.001) when differences in the time of plant emergence were included in the regression model. It is concluded that differences in A among potato genotypes is largely determined by gs, but confounding of gs and photosynthetic capacity reduces genotypic variation in A compared with that in gs. Total dry matter production is largely determined by processes other than carbon assimilation rate per unit area in individual leaves. Effective use of A as a character for selection in plant breeding depends on elucidating the effects that differences in stomatal characteristics have on crop production both in well-watered and in water-limited crops.  相似文献
