Hemocoelomic injection of 5,800–8,000 cells of Tetrahymena pyriformis strain S per female cockroach resulted in lethargy of the insects within 24 hr and death within 72 hr. Ciliates could be recovered 24–48 hr after injection of these amounts of ciliates. Inoculation of smaller numbers of cells resulted in no apparent ill effects to the insects and ciliates could not be recovered. After recovery from cockroaches and reestablishment in axenic culture, the ciliates were rounded and contained large numbers of lipid droplets which decreased on continued cultivation. The ciliates underwent a sequence of morphologic alterations involving a decrease in length and width to form ovoid cells, an approach to the pyriform shapes (but smaller than normal size), an elongation to an abnormal length/width ratio, and then, after 6 days in culture, a return to the normal shape with usual dimensions.  相似文献

The ability of rumen ciliates to digest chitin is clearly recognized. We investigated the chitinolytic system of the rumen ciliate Eudiplodinium maggii. The ciliates were grown in a selectively faunated sheep. They were isolated from the rumen and purified by sedimentation. A crude enzyme preparation was prepared following incubation of ciliates with antibiotics. This was done in order to reduce their contamination with intracellular bacteria. The activity of particular enzymes was examined by quantification of the products released from specific substrates. It was stated that the optimum conditions for the detected activities varied between 4.5 and 5.5 pH, and 45 and 55 °C. β-N-Acetylglucosaminidase was found as an enzyme of the highest activity (4.2 μmol/l released product per mg protein per h). The activities of endochitinase and exochitinase were almost two times lower than that of β-N-acetylglucosaminidase. Zymographic studies revealed the presence of two endochitinases, two exochitinases and two β-N-acetylglucosaminidases in the examined preparation.  相似文献

Leaves of Quercus robur L. were infected with conidia of Microsphaera alphitoides Griffon and Maublanc. Crude enzyme extracts of healthy and infected tissue, taken during a 7-day experimental period, were assayed, using spectrophotometric and manometric techniques, for glycolate oxidase, NAD- and NADP-glyoxylate reductase activities. After 2 days, extracts from infected tissue contained significantly higher activities of glycolate oxidase and NAD-glyoxylate reductase than extracts from controls, but the activity of NADP-glyoxylate reductase, after an initial rise, was significantly reduced below that in control extracts. The reduction in glycolate oxidase activity may be partly due to a deficiency in flavin mononucleotide in infected tissue. A possible connection between the reduction in flavoprotein enzyme activity, photorespiration and susceptibility to infection is described.  相似文献

Two strains of the anaerobic ciliate Trimyema compressum, isolated from different habitats, were compared. The cytoplasm of the ciliates contained hydrogenosome-like microbodies and methanogenic bacteria; the latter were lost during continued cultivation. In addition both strains harbored a non-methanogenic endosymbiont, which was lost in strain K. The ciliates lacked cytochromes, cytochrome oxidase and catalase but contained superoxide dismutase. Hydrogenase activity could be demonstrated only in strain N. In monoxenic culture strain K needed sterols as growth factors. The cells of both strains reacted similarly with respect to oxygen tolerance (up to 0.5 mg O2/l), inhibition of growth by cyanide and azide, and resistance to antimycin A. Only cells of strain N showed growth inhibition by chloramphenicol. It is concluded that Trimyema compressum is an anaerobic, microaerotolerant organism, its microbodies show more resemblance to hydrogenosomes than to mitochondria.  相似文献

Glycogen phosphorylase and synthase activities were detected in the sonic lysate of rumen ciliates of the genus Entodinium. The ciliate phosphorylase had the following properties. The pH optimum was narrow and centered at pH 5.9. The activity was maximum at 30°C; above 40°C a rapid inactivation occurred. The Km value for glucose-1-phosphate (G-1-P) and for glycogen was 15 mM and 0.069% (w/v), respectively. NaF and ethylenediamine tetraacetic acid had no stimulative effect on the enzyme activity, though adenosine 3′,5′-monophosphate and theophylline activated it. NaHSO3 inhibited the enzyme activity at a concentration of 1 mM. The inhibition of glucose was noncompetitive for G-1-P. Glycolytic intermediates and nucleotides had a minor effect on phosphorylase activity. Glycogen synthase existed in two forms, glucose-6-phosphate dependent and independent forms: the proportion of the latter form increased with the decrease of reserve polysaccharide levels in the ciliates. Correlations between glycolytic enzyme activities included phosphorylase and synthase activities and reserve polysaccharide contents in the ciliates were determined, and a possible regulatory mechanism of polysaccharide synthesis and degradation was discussed.  相似文献

Sapropelic ciliates from anoxic mud samples were enriched and cultivated in monoculture together with natural food bacteria growing on cellulose. The ciliates lacked cytochrome oxidase and contained bluish fluorescent endosymbionts. One of the anaerobic ciliates, Trimyema compressum, contained methanogenic bacteria as was shown by methane formation. During continued cultivation, T. compressum gradually lost its endosymbionts. With SEM microscopy no episymbiotic bacteria could be detected.From enrichment cultures of T. compressum, anaerobic bacteria were isolated in pure culture. One of the strains, a Bacteroides spec., proved capable of serving as food bacteria, thus allowing establishment of monoxenic T. compressum cultures. These cultures exhibited a requirement for sterols as growth factors. The doubling time of this ciliate was 13 h at 28°C. The highest yield obtained was 2100 cells/ml.Dedicated to Holger W. Peroxidase and polyphenol oxidase activities and peroxidase isozyme patterns were determined at different stages of hypocotyls of mung bean infected with Rhizoctonia solani. The effect of ethephon (2-chloroethyl phosphonic acid) has been studied. Peroxidase activity increased at 24 h after inoculation as compared with controls followed by a decline later. It increased at 120 h. Polyphenol oxidase activities increased after inoculation. Ethephon treatment increased the resistance to Rhizoctonia solani and enhanced peroxidase and polyphenol oxidase activities. Peroxidase isozyme pattern was found to change as a result of inoculation and ethephon treatment. The results indicated that ethephon-induced resistance was related to peroxidase and polyphenol oxidase activities.  相似文献

This study examined the effect of two natural toxins (a venom from the parasitic wasp Habrobracon hebetor and destruxin A from the entomopathogenic fungus Metarhizium anisopliae), and one pathogen (the entomopathogenic fungus Isaria fumosorosea) on the activity of basic digestive enzymes in the midgut of the cockroach Periplaneta americana. Simultaneously, the role of adipokinetic hormones (AKH) in the digestive processes was evaluated. The results showed that all tested toxins/pathogens elicited stress responses when applied into the cockroach body, as documented by an increase of AKH level in the central nervous system. The venom from H. hebetor showed no effect on digestive enzyme activities in the ceca and midgut in vitro. In addition, infection by I. fumosorosea caused a decrease in activity of all enzymes in the midgut and a variable decrease in activity in the ceca; application of AKHs did not reverse the inhibition. Destruxin A inhibited the activity of all enzymes in the midgut but none in the ceca in vitro; application of AKHs did reverse this inhibition, and no differences between both cockroach AKHs were found. Overall, the results demonstrated the variable effect of the tested toxins/pathogens on the digestive processes of cockroaches as well as the variable ability of AKH to counteract these effects.  相似文献

Ciliates are some of the most abundant gut fauna in wild chimpanzees. However, their presence in captive populations is usually low presumably due to anti‐helmintic prophylaxis or feeding on low fibre diet. We studied a semi‐captive colony of chimpanzees at the Sweetwaters Sanctuary in Kenya subject to routine prophylactic dose of albendazole to clear chimpanzees of parasitic helminths. Fresh faecal samples from known individuals were collected before and subsequently after prophylaxis. The samples were fixed in 10% formalin and examined by the sedimentation method. Troglodytella abrassarti had 42.5% prevalence whereas other ciliates had 65% prevalence. The prevalence of the T. abrassarti and other ciliates significantly declined immediately after prophylaxis and then rose slowly thereafter. Our results suggest that ciliates are susceptible to anti‐helmintic prophylaxis and that treatment may eliminate ciliates or inhibit their proliferation within the host subsequently lowering their prevalence in the population. Variation in prevalence was not influenced by the age of the host. However, a steady recovery of ciliate prevalence was lower for male compared to female hosts. Our results imply that the intervals between prophylactic regimes could be prolonged differently for males and females to increase the prevalence of ciliates in captive populations.  相似文献

Sex pheromonal activity was evaluated for propionates of 4 alcohols having a 6,6-dimethyl-bicyclo[3.1.1]heptane skeleton by behavioral assay with the male American cockroach. Their activities were compared with the corresponding acetates. The propionates were active at 20 μg or 500 μg, demonstrating stronger activities than the corresponding acetates. The male to female response ratio (M/F ratio) in EAG was also evaluated for the acetates and propionates, as well as for germacrene-D and camphor. The order of the behavioral activity was represented by that of the ratio. Sex pheromonal activity of (+)-verbanyl propionate (7) and germacrene-D were compared. Stronger activity was consequently observed in 7 at 5 μg dose, supporting the M/F ratio result.  相似文献

The inhibition of K+ uptake through the plasma membrane resulting from injury caused by cutting, or from application of polyamines (PAs), has been investigated in root segments of maize (Zea mays L.) and pea (Pisum sativum L.). It was found, for both treatments, that K+ uptake recovered if the segments were washed for 2 h. The K+ uptake inhibited by cutting and that inhibited by spermidine treatment were stimulated to the same extent by fusicoccin. In addition, there was a correlation between the extent of the recovery of K+ uptake caused by washing and the distribution, along the root axis, of both PAs and the activities of enzymes responsible for PA degradation. In apical segments of maize, where the PA content and the activity of the degradative enzyme polyamine oxidase (EC 1.5.3.3) were higher than in the more distal segments, the recovery of K+ uptake caused by washing was also higher. On the other hand, the opposite trend was observed in root segments of pea, where the PA content and the activity of the degradative enzyme diamine oxidase (EC 1.4.3.6) were higher in distal segments in which K+ uptake was greatly stimulated by washing. The effect of the amine-oxidase inhibitor, aminoguanidine, indicates that the degradation products of PAs are involved in the mechanism of inhibition of K+ uptake by PAs. Allergen extracts from dust mites and cockroaches commonly found in Korean homes were used to evaluate their enzymatic activity as they are believed to influence allergenicity. Allergen extracts were prepared from 3 dust mite species (Dermatophagoides farinae, D. pteronyssinus, and Tyrophagus putrescentiae) and 3 cockroach species (Blattella germanica, Periplaneta americana, and P. fuliginosa) maintained in the Korea National Arthropods of Medical Importance Resource Bank. Proteins were extracted in PBS after homogenization using liquid nitrogen. The activities of various enzymes were investigated using the API Zym system. No significant difference in phosphatase, lipase, or glycosidase activity was observed among the 6 allergen extracts, but much difference was observed in protease activity. Protease activity was assessed in more detail by gelatin zymography and the EnzChek assay. Extract from T. putrescentiae showed the highest protease activity, followed by those of the cockroach extracts. Extracts from D. farinae and D. pteronyssinus showed only weak protease activity. Gelatinolytic activity was detected mainly in a 30-kDa protein in D. farinae, a 28-kDa protein in D. pteronyssinus, a > 26-kDa protein in T. putrescentiae, a > 20-kDa protein in B. germanica, and a > 23-kDa protein in P. americana and P. fuliginosa. The information on various enzymatic activities obtained in this study may be useful for future studies. In particular, the strong protease activity found in cockroach extracts could contribute to sensitization to cockroach allergens, which is known to be associated with the development of asthma.  相似文献

Mutants showing negative oxidase-reaction have been isolated from Pseudomonas aeruginosa following mutagenesis with N-methyl-N-nitro-N-nitrosoguanidine. These mutants were compared to the wild type cells with respect to their respiratory activities and cytochrome contents. They exhibit lower respiration rates and contain much less cytochrome c's which are responsible for their weak or negative oxidase-reaction in these mutants. This is supported in part from an initial linear relationship observed between the measured oxidase activities and the lower cytochrome c contents in these mutants. Further evidence comes from analyzing oxidase-negative cells of P. syringae, in which low cytochrome c contents similar to these oxidase mutants account for negative oxidase activities. Cytochrome o was the sole oxidase found among these mutants as well as in the wild type cell, suggesting that cytochrome c+o complex is responsible for the tetramethyl-p-phenylenediamine-oxidase activity in these mutants as the case in the wild-type cells. From the spectral characteristics it seems that all mutants contain about the same amount and type of terminal oxidase as that of the wild-type cells. (1) The distributions of four oxidative enzymes were studied in crude brain fractions. (2) Freeze-thaw cycle treatment and frozen storage of homogenate fractions gave apparent enhancement of cytochrome oxidase and NADH cytochrome c reductase activities. (3) Deoxycholate released cytochrome oxidase and NADH cytochrome c reductase activities from low-speed precipitates. The NADH diaphorase was enhanced to a small degree while NADPH cytochrome c reductase was not affected by deoxycholate. (4) Distilled water coupled with a single homogenization released trapped soluble enzymes and mitochondria and gave nearly maximal cytochrome oxidase activity as judged by deoxycholate treatment. The total distilled water activity of NADH cytochrome c reductase was much less than that of deoxycholate-stimulated fractions. The activities of other enzymes were not markedly affected by distilled water although their distribution was changed.  相似文献

Two types of polyethylenimine-coated glass microbeads (13–44 μm) were synthesized and used for the immobilization of glucose oxidase from Aspergillus niger and catalase from A. niger and beef liver. The two types of beads were distinguishable by differences in their surface topography. Immobilizations were performed by adsorption followed by treatment with glutaraldehyde. The immobilized-enzyme activities per unit support of all of the enzymes tested were compared with and found to be superior to the immobilized activities attainable on aminopropyl-activated glass microbeads. When enzyme was present in less than saturating amounts, the coated beads were able to remove 100% of the glucose oxidase activity initially present in the immobilization solution, with 78–87% of that activity expressed on the support surface. Bound glucose oxidase was more stable to thermal inactivation than native enzyme.  相似文献

Phenol oxidase activity was detected for the first time in a number of strains belonging to various Azospirillum species. Both extracellular and intracellular activities of laccase, Mn-peroxidase, lignin peroxidase, and tyrosinase were observed. Extracellular enzymes were found to have higher activity. We investigated chilling-induced changes in ethylene levels in Arabidopsis to find plants with distinct patterns of ethylene production in the cold-related biosynthetic pathway. The sensitive mutants identified here includedchs1-2,chs4-2, andchs6-2. Among these, plants of thechs4-2 mutant produced more ethylene than did the wild type after both were transferred from 4°C or 10°C to 22°C. This mutant also showed less freezing tolerance and more electrolyte leakage than the wild-type plants. Our results suggest a relationship between ethylene biosynthesis and chilling sensitivity in the mutant To determine which of the enzymes involved in ethylene biosynthesis were induced by chilling, we tested the activities of ACC synthase and ACC oxidase in both mutant and wild-type plants, and found greater activity by ACC synthase as well as a higher ACC content in the mutants after all the plants were transferred from 10°C to 22°C. However, ACC oxidase activity did not differ between mutant and wild-type plants in response to chilling treatment Therefore, we conclude thatchs4-2 mutants produce more ethylene than do other mutants or the wild type during their recovery from chilling conditions. Furthermore, we believe that ACC synthase is the key enzyme involved in this response.  相似文献

Peroxidases (EC 1.11.1.7) from Populus x euramericana were investigated during the dormant and growing seasons using histochemical and biochemical methods. The activities of syringaldazine oxidase and p-paraphenylenediamine-pyrocatechol oxidase in sections of branches were maximal during spring in both phloem and young xylem. Cytoplasmic and cell-wall peroxidase activities from different lignified tissues were estimated in vitro. Pronounced differences were noticed between fractions isolated during spring and winter. Gel electrophoresis showed the presence of an anionic fast-migrating isoperoxidase group with a high syringaldazine-oxidase activity. The isoenzymes of this group were different in winter and in spring. Mitochondria from Orobanche were analysed for the activities of aconitate hydratase, isocitrate dehydrogenase, succinate dehydro-genase, fumarate hydratase, malate dehydrogenase, NADH oxidase, substrate-cytochrome c oxidoreductases, glutamate dehydrogenase, aminotransferases, ATPase and “malic” enzyme. The specific activities of isocitrate dehydrogenase, NADH oxidase, substrate-cytochrome c oxidoreductases and glutamate dehydrogenase in the mitochondria) fraction from parasite tissue compared favourably with those reported for most of the mitochondria from growing and storage tissues. Succinate dehydrogenase, fumarate hydratase and aspartate aminotransferase were of intermediate activity, while aconitate hydratase and malate dehydrogenase had rather low activity, and “malic” enzyme had very low activity in comparison with other preparations. The relevance of these findings in relation to mitochondrial metabolism in the parasite is discussed. No evidence was obtained to suggest any basic abnormality in the biochemical properties of the mitochondria from Orobanche centua which may be correlated with its obligatorily parasitic existence.  相似文献
