目的探讨p16和bcl-2表达产物在宫颈上皮瘤样病变及宫颈癌中表达的意义。方法对正常宫颈鳞状上皮、宫颈上皮内瘤变(CIN)和宫颈癌组织共69例,采用免疫组织化学EliVision法,对宫颈癌变过程中p16和bcl-2蛋白进行研究,将结果进行统计分析。结果p16蛋白在CIN中的表达高于正常宫颈上皮(P<0.01),在宫颈癌中的表达也高于正常宫颈上皮(P<0.01),且高于CIN中的表达(P<0.05);bcl-2蛋白在CIN中的表达高于正常宫颈上皮(P<0.01),在宫颈癌中的表达也高于正常宫颈上皮(P<0.01),但与CIN中表达无差异。p16和bcl-2两种蛋白在CIN和宫颈癌中的表达无明显差异。结论p16和bcl-2蛋白的表达与宫颈癌的发生有关,提示这两种蛋白有可能作为高危人群早期筛查的一种免疫组化指标。  相似文献

This study investigates the fluid flow through tissues where lymphatic drainage occurs. Lymphatic drainage requires the use of two valve systems, primary and secondary. Primary valves are located in the initial lymphatics. Overlapping endothelial cells around the circumferential lining of lymphatic capillaries are presumed to act as a unidirectional valve system. Secondary valves are located in the lumen of the collecting lymphatics and act as another unidirectional valve system; these are well studied in contrast to primary valves. We propose a model for the drainage of fluid by the lymphatic system that includes the primary valve system. The analysis in this work incorporates the mechanics of the primary lymphatic valves as well as the fluid flow through the interstitium and that through the walls of the blood capillaries. The model predicts a piecewise linear relation between the drainage flux and the pressure difference between the blood and lymphatic capillaries. The model describes a permeable membrane around a blood capillary, an elastic primary lymphatic valve and the interstitium lying between the two.  相似文献

为了探讨p16基因甲基化及异常表达在宫颈癌中的意义,分别采用甲基化特异性PCR(MSP)方法检测不同病理类型和临床分期的60例宫颈癌组织中p16基因启动子区域5´CpG岛甲基化状态;采用PCR方法检测p16基因外显子1(E1)和外显子2(E2)纯合缺失情况;采用免疫组化的方法分析p16蛋白的表达缺失和减弱情况。结果显示正常对照组织及癌旁p16基因无甲基化,且无E1和E2缺失和p16蛋白表达异常。60例宫颈癌标本的甲基化率为21.67%(13/60);p16 基因缺失率为15.00%(9/60);p16蛋白表达下降或无表达为51.67%(31/60)。可见p16基因蛋白的阳性表达率随着临床分期升高呈明显下降趋势。结果提示p16基因失活在宫颈癌中多见且与病理分级密切相关。p16 基因甲基化在宫颈癌发生中起着一定作用。Abstract: To detect hypermethylation and aberrant expression of the p16 gene in cervical carcinoma (CC), methylation-specific PCR (MSP) was used to determine the methylation status of 5´CpG islands of the p16 gene, loss or decrease of p16 expression was analyzed by immunohistochemistry (IHC), and homozygous deletion of exon 1 (E1) and/or exon 2 (E2) was determined by PCR. in 60 cases of CC at different pathological grades and clinical stages. The results showed absence of methylation and presence of normal expression of the p16 gene in the control and adjacent tissues of CC. Hypermethylation, loss or decrease of expression and deletion of the p16 gene were detected in 21.67%(13/60), 51.67%(31/60) and 15.00%(9/60) of the tumor tissues, respectively. The rate of p16 expression markedly reduced with the increase of clinical stages. Our data suggested that inactivation of the p16 gene was a frequent event and positively correlated with pathological grades in CC, and that methylation of the p16 gene was an important event in carcinogenesis of CC.  相似文献

目的:研究胰腺癌组织中微血管密度(Microvessel density,MVD)和淋巴管密度(Lymphatic vessel density,LVD)的变化、与胰腺癌临床病理的联系。方法:采用免疫组织化学SABC法应用CD34及D2-40分别检测41例胰腺导管腺癌患者配对癌组织内、癌旁及正常胰腺组织中MVD及LVD的表达情况,分析其与肿瘤分化程度、分期和淋巴转移间的相关性;以及癌组织MVD与癌旁组织LVD表达之间的关系。结果:在41例胰腺癌组织配对癌组织及正常胰腺组织平均MVD值分别为46.585±16.935,11.100±4.036,两组之间差别有统计学意义(P=0.000<0.01)。配对癌组织内、癌旁及正常胰腺组织中平均LVD值分别为11.244±4.800,15.829±7.470和13.512±5.139;其中癌旁组织与正常胰腺组织LVD值差别无统计学意义(P=0.060>0.05),癌旁组织与胰腺癌组织的LVD值有显著性差异(P=0.000<0.01)。癌组织内MVD值与癌旁组织LVD值之间有相关性(P=0.025<0.05)。结论:胰腺导管腺癌组织中MVD及LVD与肿瘤分化程度、病理分期及淋巴转移存在关联。胰腺癌组织内MVD值与癌旁组织LVD之间存在相关性。  相似文献

目的:检测c-fos蛋白和c-jun蛋白在宫颈癌、CIN以及宫颈炎组织中的表达,并探讨其意义.方法:免疫组织化学S-P法检测23例宫颈癌、11例CINⅢ、13例CINⅡ、20例CINⅠ、10例宫颈炎组织标本中c-fos和c-jun蛋白的表达.结果:①c-fos蛋白在宫颈癌、CINⅢ、CINⅡ和CINⅠ以及宫颈炎中的表达分别为82.6%(19/23)、65.2%(7/11)、46.2%(6/13)、35.0%(7/20)、0.0%(0/10);②c-jun蛋白在宫颈癌、CINⅢ、CINⅡ、CINⅠ以及宫颈炎中的阳性表达率分别为78.3%(18/23)、54.5%(6/11)、38.5%(5/13)、30.0%(6/20)、0.0%(0/10);③c-fos和c-jun在宫颈癌、宫颈癌前病变、宫颈炎中的表达均有显著差异(H=0.000,P<0.05);经秩相关检验分析,c-fos和c-jun在宫颈炎、宫颈癌前病变、宫颈癌中的表达呈正相关(r=0.581,P=0.000).结论:c-fos和c-jun蛋白的表达可能与宫颈癌前病变、宫颈癌的发生密切相关,两者可能在宫颈癌的发生过程中起一定的协同作用.  相似文献

长期以来人们一直认为基因突变或基因缺失参与肿瘤的形成。近年来众多研究表明,表观遗传修饰对肿瘤的发展也具有非常重要的意义,它的主要表现形式有DNA甲基化、组蛋白修饰、微小RNA调节、染色质重组等。DNA异常甲基化可通过影响染色质结构、癌基因及抑癌基因表达而参与肿瘤的形成。了解目前宫颈癌中DNA甲基化的研究进展不仅有助于宫颈癌的早期诊断,对其分子靶向治疗及预后评估亦显示出良好的应用前景。  相似文献
