Space flight with associated microgravity is complicated by "astronaut's anemia" and other hematologic abnormalities. Altered erythroid differentiation, red cell survival, plasma volume, and progenitor numbers have been reported. We studied the impact of microgravity on engraftable stem cells, culturing marrow cells in rotary wall vessel (RWV) culture chambers mimicking microgravity and in normal gravity nonadherent Teflon bottles. A quantitative competitive engraftment technique was assessed under both conditions in lethally irradiated hosts. We assessed 8-wk engraftable stem cells over a period spanning at least one cell cycle for cytokine (FLT-3 ligand, thrombopoietin [TPO], steel factor)-activated marrow stem cells. Engraftable stem cells were supported out to 56 h under microgravity conditions, and this support was superior to that seen in normal-gravity Teflon bottle cultures out to 40 h, with Teflon bottle culture support superior to RWV from 40 to 56 h. A nadir of stem cell number was seen at 40 h in Teflon and 48 h in RWV, suggesting altered marrow stem cell cycle kinetics under microgravity. This is the first study of engraftable stem cells under microgravity conditions, and the differences between microgravity and normal gravity cultures may present opportunities for unique future stem cell expansion strategies.  相似文献

The goals of this work were to test the feasibility of a continuous plug-flow (PF) bioreactor and to compare the growth in the PF bioreactor to that in a batch bioreactor. A culture of Pseudomonas putida was pumped through a tube made of Teflon with varying residence times. The culture was aerated by pumping of air simultaneously with liquid medium to provide air bubbles along the tubular culture. When the residence time in the PF bioreactor was greater than the time needed to reach the stationary phase in batch mode, the maximum biomass density reached in PF mode was the same as the maximum density reached in the batch bioreactor, and benzoate (the only carbon and energy source) was completely consumed. The drawbacks for practical application of PF were found to be fluctuations of cell concentration in the outflow cultural liquid due to cell aggregation, significant cell adhesion to the inner wall of Teflon tubing, and inadequate aeration.  相似文献

The selective growth of malignant cells on non-adhesive Teflon substrates is reported. Neoplastically transformed cell lines of human and murine origin proliferated on Teflon whereas similarly seeded normal non-tumorigenic cells were unable to undergo cell division and subsequent growth. This substrate should be especially useful in establishing primary cultures from human tumor material.  相似文献

顺-1,2-二羟基-3,5-环己二烯(简称DHCD)是航天业,电子工业,医药业以及精细化工业上重要的手性化合物,利用重组E.coli JM109(pKST11),采用适时监测发酵过程中全细胞甲苯双加氧酶(Toluene dioxygenase,TDO)活性的方法,研究了发酵生产DHCD工艺中的重要影响因子IPTG以及底物苯的供给方式对DHCD产量的影响,研究结果表明:(1)发酵初期利用IPTG诱导TDO的表达,不利于细胞生长,在对数生长中期(6或8h),采用0.5mmol/L IPTG即可诱导出TDO的最高表达。(2)发酵液中的苯对全细胞甲苯双加氧酶(TDO)的活性有抑制作用,而利用液体石蜡作为缓释剂进行两相法发酵则降低了苯的毒害,明显提高了DHCD的产量。当采用传统的通气供苯方法,DHCD的产量仅有7.5g/L;批式添加液体石蜡与苯的混溶物使DHCD的产量提高到22.6g/L,是通气供苯法的3倍;而采用流加的方式添加液体石蜡与苯的混溶物使DHCD的产量进一步提高到36.8g/L,是通气供苯法的5倍,证明通过发酵工艺的优化可以解决苯的毒害与苯作为反应底物在水相中需要一定浓度之间的矛盾,获得较好的转化结果。  相似文献

A feedback control system of the glucose feed rate in a bakers' yeast fed-batch culture was developed by keeping the ethanol concentration constant. A PID controller and on–off controller were applied and discussed with the aid of the porous Teflon tubing method. Experimental results showed the effectiveness of the control system for avoiding the glucose effect and glucose starvation. It was shown that the feedback control system developed hare could achieve a maximum specific growth rate of 0.3 h?1 or a maximum cell yield of 0.5 g cell/g glucose in the fedhyphen;batch culture.  相似文献

When paraffin wax is dispersed in medium as emulsion, some kinds of bacteria and yeasts readily grow on it. This paper presents a study on microbial cell production from solid paraffin. In this study a paraffin wax which contains 91% of normal paraffins ranging from C25 to C37 with the melting point of 62.5°C was used as a substate, but no solvent was used for the dispersion of the wax.

As a result of this study, the following have been found out. (1) Many strains of liquid normal paraffin assimilating bacteria and yeasts can assimilate paraffin wax. (2) Dried cell yields on added hydrocarbons of Corynebacterium hydrocarboclastus S-12-B2 and Candida tropicalis S-315-Y1 are 70% and 56% respectively, when they are cultured by wax emulsion of 0.6% concentration. (3) When nonion surface-active agent (Plysurf A210G) was added as an emulsifing agent, highly concentrated wax emulsion was obtained, but the growth of microorganisms on it was slower. Further investigation is needed to obtain better strains of bacteria and yeasts and also to find out optimum culture conditions.  相似文献

The effects of agitation rate and impeller type on the combined oxygen mass-transfer coefficient (kL a) in four different benchtop bioreactors have been examined. Surface oxygenation of a cell culture medium supplemented with fetal bovine serum and distilled deionized water has been studied by passing air through the bioreactor headspace at approximately one headspace volume per minute. A new ribbon-type impeller design using strips of Teflon has been shown to be superior to conventional impeller designs for oxygen transfer.  相似文献

Inasmuch as cell adhesion and spreading are fundamental properties involved in a variety of vital processes, the dependence of the expression of spontaneous macrophage-mediated cytotoxicity on these attributes was investigated using unique means by which substratum adhesiveness of effector and target cells could be either markedly diminished or varied within a large range. Spontaneous cytotoxicity by activated macrophages against various tumor targets growing in adherent or in suspension culture was unaltered, irrespective of whether interaction took place on plastic, on Teflon, or on plastic coated with varying concentrations of the polymer, poly(HEMA). The findings indicate that whatever the nature is of spontaneous macrophage-mediated target cell killing, it is quite independent of cell adhesion and cell shape considerations.  相似文献

The injection treatment of unilateral recurrent vocal cord nerve paralysis, successfully performed 40 years ago with paraffin, has recently been reintroduced with non-reactive synthetics. Improvement in the voice far and above that which can be achieved by voice therapy or any practicable surgical procedure has been demonstrated by several investigators. The technique is relatively simple, and there are no significant untoward reactions. Judging from experimental and clinical studies with these inert materials when introduced into other parts of the body, adverse long range effects are not likely to occur in the larynx.Eleven of 12 patients treated by injection of silicone or Teflon paste had substantial improvement in vocal quality and strength. Silicone was absorbed slowly from the injection site and the improvement was not enduring; when Teflon was used, it stayed at the site and the improvement was sustained.  相似文献

Summary Hybridoma concentrations were reduced in shake flask and continuous culture by medical-grade PVC and polyurethane samples. Cell viability was unaffected and nutrient uptake rates were increased. No inhibition was observed for silicone, C-Flex or Teflon samples. The inhibition by PVC could be reduced by conditioning the sample with complete medium. The reduction in final cell yield and growth rate may result from extraction of one or more growth factors from the medium.  相似文献

采用石蜡切片法,对天南星科植物羽裂蔓绿绒(Philodendron pitfieri Engl.)水培和土培根的解剖特征进行观察,探讨蔓绿绒适应水生环境的形态学特点,并比较了水培根与土培根的形态解剖学差异.结果表明:水培后根冠不明显,淀粉体较少,多糖类物质较少而土培根根冠体积较大,淀粉体较多,多糖类物质多,土培根PAS反应强烈;水培根表皮厚度比土培根小,水培根根毛少或退化消失;水培根皮层薄壁组织发达,细胞大型,壁薄,细胞间隙中含有溶生性通气组织,土培根皮层未见通气组织.  相似文献

Thirteen girls with grade III-V vesicoureteric reflux were treated by endoscopic injection of Teflon paste behind the intravesical ureter. Fourteen of the 18 treated ureters showed complete absence of reflux after one injection of Teflon. Three ureters required a second injection of Teflon for successful treatment of the reflux. One ureter with grade IV reflux was converted to grade II reflux. Properly carried out, this procedure corrects reflux. It takes less than 15 minutes, may be done as a day procedure, and avoids open surgery. There have been no complications.  相似文献

Fluoroplastics (Aclar and Teflon FEP) were tested for their suitability as coverslip material for nerve tissue cultures. Coverslips prepared from these plastics have a higher transparency over the near-ultraviolet, visible, and infrared regions of the spectrum than glass coverslips, possess exceptional physical and chemical stability, and provide service-ability over a wide temperature range ≥ -320 to 390 F. For nerve tissue culture, circular 22 mm coverslips were cut from 5 mil sheets of Aclar 22A and 33C, and Teflon FEP 100A, cleaned in concentrated HNO3, rinsed, sterilized in 85% ethyl alcohol, dried, and coated with collagen. Peripheral and central nervous system cultures from fetal rats and newborn mice were set up on these as well as on glass coverslips, and carried in Maximow double coverslip assemblies. Over a 2 mo period of cultivation, neuronal maturation, tissue organization and fiber myelination occurred on plastic coverslips in essentially the same manner as in sister cultures maintained on glass coverslips. Cultures fixed and embedded in Epon for electron microscopy directly on Aclar or Teflon 100A coverslips were easily separated from the coverslip. Some difficulty was encountered with retraction of the collagen coat from Teflon 100A coverslips, however, and Aclar 22A coverslips were warped by some of the reagents used in embedding for electron microscopy. Aclar 33C was not adversely affected by these reagents, and provided the most suitable base for collagenization and culture maintenance. A variety of cells grew satisfactorily on either the bare or the collagenized surface of this plastic, and separated easily from it following embedment in Epon.  相似文献

Abstract A Rhodococcus strain possessing a capsule, but no fimbriae, was isolated from pond water by adsorption to Teflon. The strain was hydrophobic, as shown by partitioning between dodecane and buffer. A high emulsifying activity was found in the culture supernatant, from which a polysaccharide was isolated. This contained glucuronic acid, glucose, galactose and rhamnose in a molecular ratio of 1:1:1:2. One acetate residue was found per repeating unit. The polysaccharide molecules formed clusters, which disaggregated on the addition of sodium dodecyl sulphate (SDS). Rabbit antibodies against this polysaccharide aggregated the bacterial cells. Thus, it can be concluded that this polysaccharide at least contributes to the cell surface hydrophobicity, thereby mediating in the adsorption of cells to inert hydrophobic surfaces.  相似文献

This paper is a further research on the origin of gynogenic structures of rice which have been induced by the method of unpollinated ovary culture in our laboratory for several years. Three cultivars were used as materials. The culture medium and technique were basically the same as in previously reported experiments. Samples were fixed, stained and sectioned by paraffin method. All the four components of embryo sac could be induced to division, however, with different frequency and results. Most of the gynogenic proembryos were originated from the synergids. The processes of synergid apogamy are described in detail. The egge cell, in contrast, usually vacuolated considerably and underwent free nuclear division, leading to coenocyte formation. The problem of why and how synergid apogamy takes place in such high frequency in ovary culture is discussed. The anomalous behavior of the egg cell is discussed in relation to the physiological as well as the phylogenetic aspect.  相似文献

A technique Is described for rapid detection of S-pha?e cells of tumor tissues in smear specimens using bromodeoxyuridine (BrdU) immunostaining. Mouse NR-S1 tumors and human tumor specimens were prepared for smear cytology after incubation in RPMI 1640 culture medium containing 200 μM BrdU at 37 °C under 3 atm for 1 hr. Samples were fixed in 70% ethanol for 30 min and used immediately or air dried for 30 min. Samples were then denatured in either 4 N HC1 or 0.07 N NaOH to prepare partially single-stranded DMA. Fixation with air drying for 30 min followed by 30 min in 70% ethanol and 1 min denaturation with 0.07 N NaOH resulted in satisfactory staining quality. Cultured tumor specimens were processed for routine paraffin sections after smears were made for cytology. The labeling indices of the smear specimens and of the paraffin sections gave similar results. This technique should be useful in evaluating the cell proliferative potential of tumor tissue in smear cytology without processing paraffin sections.  相似文献

The characteristics of a monoclonal antibody produced against osteoclast-like multinucleated cells (MNCs) formed in rat bone marrow cultures were examined immunohistochemically and biochemically. The in vitro immunization was performed using as immunogen the MNCs from rat bone marrow cell culture, which revealed many characteristics of osteoclasts. After screening and cloning of hybridomas, the monoclonal antibody HOK 1 was obtained. This antibody reacted weakly with stromal cells and intensely with both MNCs and their putative migratory traces on culture dishes. Immunofluorescent examination of paraffin sections revealed intense reactivity on the epithelium of the choroid plexus, the ileum and the proximal-convoluted tubules of the kidney, and also on bone cells such as osteocytes, osteoblasts, and osteoclasts. Western blotting using purified rat osteopontin verified that the antigen recognized by HOK 1 was osteopontin. Positive HOK 1 immunoreactivity was further observed in the resorption lacunae formed by a culture of MNCs on human tooth slices and on the surface of osteoclasts. The present data suggested that osteopontin is preferentially present on the resorption lacunae in resorbing calcified matrices and that osteoclasts under a specific state might trap this protein on their cell surface.  相似文献

One cervical cancer cell line, C9, carrying human papillomavirus type 18 (HPV18) genes that is one of the major etiologic oncoviruses for cervical cancer was characterized. This cell line was further characterized for its capacity related to the epithelial cell proliferation, stratification and differentiation in reconstituted artificial epithelial tissue. Thein vitro construction of three dimensional artificial cervical epithelial tissue has been engineered using C9 epithelial cancer cells, human foreskin fibroblasts and a matrix made of type I collagen by organotypic culture of epithelial cells. The morphology of paraffin embedded artificial tissue was examined by histochemical staining. The artificial epithelial tissues were well developed having multilayer. However, the tissue morphology was similar to the cervical tissue having displasia induced by HPV infection. The characteristics of the artificial tissues were examined by determining the expression of specific marker proteins. In the C9 derived artificial tissues, the expression of EGF receptor, an epithelial proliferation marker proteins for stratum basale was observed up to the stratum spinosum. Another epithelial proliferation marker for stratum spinosum, cytokeratins 5/6/18, were observed well over the stratum spinosum. For the differentiation markers, the expression of involucrin and filaggrin were observed while the terminal differentiation marker, cytokeratins 10/13 were not detected at all. Therefore the reconstituted artificial epithelial tissues expressed the same types of differentiation marker proteins that are expressed in normal human cervical epithelial tissues but lacked the final differentiation capacity representing characteristics of C9 cell line as a cancer tissue derived cell line. Expression of HPV18 E6 oncoprotein was also observed in this artificial cervical epithelial tissue though the intensity of the staining was weak. Thus this artificial cervical epithelial tissue though the intensity of the staining was weak. Thus this artificial epithelial tissue could be used as a useful model system to examine the relationship between HPV-induced cervical oncogenesis and epithelial cell differentiation.  相似文献
