Analysis of the sialic acid fraction obtained by mild acid hydrolysis of the Cuvierian tubules of the sea cucumber Holothuria forskali Della Chiaje revealed the presence of F-glycolylneuraminic acid and fucopyranosyl-(1 → 4)-N-glycolylneuraminic acid. Furthermore, methylation analysis of a pronase-digest of the tubules demonstrated that all N-glycolylneuraminic acid residues are substituted at C-4, explaining the earlier reported resistance of sialic acid in tubules to cleavage by neuraminidase.  相似文献

Three species of gorgonians (Primnoa pacifica, Calcigorgia japonica, and Thouarella superba) are reported as new records for the northwestern Sea of Japan; one of them (Calcigorgia japonica sp. n.) has been described as a new species. Variations of colony morphology in some species are described; the taxonomical value of specific characters is discussed. The geographical range of Primnoa pacifica in the northwestern Pacific is shown to be wider than was considered earlier. Variations of sclerite morphology (number of marginal plates, presence of additional basal plates, and number of marginal spines on the basal plate) are regarded as intracolonial or intraspecific variability. The distribution range of Thouarella superba described earlier from the Aleutian Islands (Nutting, 1912) was shown to extend into the low-boreal part of the boreal Pacific. An examined specimen of Paragorgia sp. differed from all known species and appeared similar only to P. regalis Nutting, 1912 reported from the eastern coast of Japan. The most distinctive characters of Calcigorgia japonica sp. n. were the shape of polyp, with a thin lower part looking like folded podia (in fixed specimens) and the presence of two types of clubs scattered among the sclerites of the polyp.  相似文献

Simultaneous oxidation-reduction potential versus absorbance titrations on intact mitochondria have allowed us to measure the individual spectral and thermodynamic properties of cytochromes a and a3 in the presence or absence of added ligands. In coupled pigeon heart mitochondria, saturated with CO, the reduced a3CO compound can be fully oxidized by ferricyanide under conditions of high phosphate potential, i.e., on addition of ATP. These ATP-induced effects are reversed by the presence of oligomycin or uncouplers with complete restoration of the reduced a3CO compound. This observation has permitted us to examine the spectral and thermodynamic properties of the a3CO compound in some detail. The data indicate that, in the presence of ATP, the midpoint potential of the a3CO compound varies as a function of CO concentration in a thermodynamically predictable fashion. The ATP-induced changes on the a3CO compound are directly analogous to the changes reported earlier for cytochrome a3 in the absence of added ligands.  相似文献

The amphibian chytrid fungus Batrachochytrium dendrobatidis (Bd) is a purported agent of decline and extinction of many amphibian populations worldwide. Its occurrence remains poorly documented in many tropical regions, including the Guiana Shield, despite the area’s high amphibian diversity. We conducted a comprehensive assessment of Bd in French Guiana in order to (1) determine its geographical distribution, (2) test variation of Bd prevalence among species in French Guiana and compare it to earlier reported values in other South American anuran species (http://www.bd-maps.net; 123 species from 15 genera) to define sentinel species for future work, (3) track changes in prevalence through time and (4) determine if Bd presence had a negative effect on one selected species. We tested the presence of Bd in 14 species at 11 sites for a total of 1053 samples (306 in 2009 and 747 in 2012). At least one Bd-positive individual was found at eight out of 11 sites, suggesting a wide distribution of Bd in French Guiana. The pathogen was not uniformly distributed among the studied amphibian hosts, with Dendrobatidae species displaying the highest prevalence (12.4%) as compared to Bufonidae (2.6 %) and Hylidae (1.5%). In contrast to earlier reported values, we found highest prevalence for three Dendrobatidae species and two of them displayed an increase in Bd prevalence from 2009 to 2012. Those three species might be the sentinel species of choice for French Guiana. For Dendrobates tinctorius, of key conservation value in the Guiana Shield, smaller female individuals were more likely to be infected, suggesting either that frogs can outgrow their chytrid infections or that the disease induces developmental stress limiting growth. Generally, our study supports the idea that Bd is more widespread than previously thought and occurs at remote places in the lowland forest of the Guiana shield.  相似文献

Mitotic chromosome numbers are reported from 25 vascular plant taxa, endemic to the Balearic Islands that are poorly known cytogenetically. The chromosome numbers ofAnthyllis vulneraria subsp.balearica (2n=12),Cymbalaria fragilis (2n=56), andPolygonum romanum subsp.balearicum (2n=40) were determined for the first time. A new chromosome number was found in several populations ofAnthyllis hystrix (2n=70) suggesting that this species is decaploid, in contrast to an earlier work reporting a higher ploidy level (2n=12x=84). The new chromosome number 2n=32 was reported inHypericum hircinum subsp.cambessedesii. It is suggested that the previous count (2n=40) could be explained by the presence of anomalous pentaploid cells in some tissues, contrating with the presence of a regular tetraploid complement (2n=32). Cytogenetic observations suggest thatSibthorpia africana has a diploid chromosome complement of 2n=18, with 0–2 accessory chromosomes. Accessory chromosomes are also reported forPhlomis italica, being the first record of B chromosomes in this genus. Chromosomal instability was found inGalium crespianum andG. friedichii species, with three numbers 2n=44, 55 and 66. Two cytotypes differing in ploidy level were documented within single plants. It is suggested that both species share a regular complement of 2n=44 and that the past hybridization events and formation of regenerating roots from the typical rootstock ofG. crespianum andG. friedrichii could be involved in the genesis of chromosome variants through partial endopolyploidy and concomitant somatic segregation.  相似文献

The ATP-hydrolyzing activity of F1-ATPase purified from potato tubers mitochondria was stimulated 2- and 3.5-fold by anions, chloride and bicarbonate, respectively, and 5.5- and 6.5-fold by detergents, octyl glucoside and lauryl dimethylamine oxide (LDAO), respectively. The maximal specific activity of the activated F1, 129 μmol/min per mg protein is the highest activity of plant mitochondrial F1 hitherto reported and exceeds several-fold values reported earlier. In the absence of activators F1-catalyzed ATP hydrolysis exhibits non-linear double-reciprocal plots of [ATP]−1 vs v−1 indicative of negative cooperativity, while in the presence of activators, linear plots are observed. It is suggested that the activators reduce the cooperativity originating from the interaction between different subunits of the enzyme.  相似文献

The two diastereoisomers, (2S,4R)-4-hydroxyglutamic acid and (2S,4S)-4-hydroxyglutamic acid, occur in characteristic concentration ratios in Phlox species. The second of these compounds is the principal free amino acid in the green parts of the plants. The presence of (2S,4R)-4-hydroxyglutamic acid in plants is reported for the first tiine. No other 4-substituted acidic amino acids were detected in the Phlox species analysed, although special attention was paid to the possible presence of 4-hydroxy-4-methylglutamic acids which have previously been reported in plants. It was found, however, that both diastereoisomers of (2S)-4-hydroxy-4-methylglutamic acid co-exist in Ledenbergia roseoaenea and also in Pandanus veitchii. Although the presence of 3,4-dihydroxyglutamic acids in green parts of Lepidium sativum and Rheum rhaponticum has been previously reported, we were not able to detect or isolate any of the possible diastereoisomers from the green parts or seeds of these plants. We did isolate glutathione which was found to have some properties in common with those reported for the dihydroxy compounds.  相似文献

Under alkaline conditions and in the presence of reductant, NADH or NADPH, and molecular oxygen, hemin catalyzes the regiospecific para-hydroxylation of aniline to form p-aminophenol [P.A. Adams, D.A. Baldwin, and M.C. Berman, J. Chem. Soc. (Lond) Chem. Commun. 856 (1979); P.A. Adams and M.C. Berman, J. Inorg. Biochem.17, 1 (1982)]. The reaction has now been studied in the presence of H2O2 and alkyl hydroperoxides and in the absence of oxygen and reductant. Results indicate that the H2O2? and alkyl hydroperoxide-supported processes proceed via different mechanisms involving, on the one hand, the hydroperoxide anion (HO2?) and on the other, the undissociated alkyl hydroperoxide molecule (ROOH). The addition of superoxide dismutase to the reaction had no effect, unlike the NADH/O2 supported reaction where the enzyme completely inhibits reaction. Similarities between the hemin-mediated peroxide-supported reactions reported here, and the cytochrome P-450-mediated peroxide-supported reactions reinforce our earlier contentions that the alkaline hemin system appears to be a good model for the in vivo activation of oxygen by hemoproteins.  相似文献

The following structure of the O-polysaccharide (O-antigen) of Salmonella enterica O13 was established by chemical analyses along with 2D 1H and 13C NMR spectroscopy:→2)-α-l-Fucp-(1→2)-β-d-Galp-(1→3)-α-d-GalpNAc-(1→3)-α-d-GlcpNAc-(1→The O-antigen of S. enterica O13 was found to be closely related to that of Escherichia coli O127, which differs only in the presence of a GalNAc residue in place of the GlcNAc residue and O-acetylation. The location of the O-acetyl groups in the E. coli O127 polysaccharide was determined. The structures of the O-polysaccharides studied are in agreement with the DNA sequence of the O-antigen gene clusters of S. enterica O13 and E. coli O127 reported earlier.  相似文献

The importance of courtship song rhythms in Drosophila to molecular and behaviour genetics led to an attempt to verify them. Two songs considered rhythmic by one of the two workers who first reported this behaviour were found to be arrhythmic because the analysis used earlier was inappropriate for identifying song cycles. Using time series analyses by spectral analysis and autocorrelation, no fly showed the sinusoidal pattern in its song claimed by the earlier workers. These findings require recent research concerned with Drosophila song rhythms to be re-evaluated.  相似文献

A ‘lignin’ fraction isolated by the Björkman method from the marine brown alga Fucus vesiculosus has been examined by 13C NMR and degradative analysis, and shown to consist of polyphloroglucinols identical to those characterized earlier from Fucus. Four lignans, four lignan glucosides, one flavonoid and two caffeoyl glycosides of 3,4-dihydroxyphenethyl alcohol were identified variously in leaves of Forsythia suspensa, F. viridissima and F. koreana. A moderately halophilic Gram-negative bacterium, strain DL7, was isolated from saltern sediment of Dalian, China. Phylogenetic analysis based on 16S rRNA gene sequences indicated that this isolate belongs to the genusHalomonas. Based on the taxonomic and DNA sequence in addition to high DNA-DNA homologies, we concluded that this strain was similar with the type strain ofHalomonas ventosae. Ectoine, one of the representative compatible solutes, was mainly detected in the cells as a result of1H- and13C-NMR measurements when grown in the presence of wide concentration ranges of NaCl. The results showed that higher amount of ectoine was synthesized in a shorter incubation time compared with those of other strains reported earlier.  相似文献

The O-specific polysaccharide was isolated by mild acid degradation of the lipopolysaccharide of Yersinia pseudotuberculosis O:4a and studied by NMR spectroscopy, including 2D ROESY and 1H, 13C HMBC experiments. The following structure of the pentasaccharide repeating unit of the polysaccharide was established, which differs from the structure reported earlier [Gorshkova, R. P. et al., Bioorg. Khim. 1983, 9, 1401-1407] in the linkage modes between the monosaccharides: where Tyv stands for 3,6-dideoxy-d-arabino-hexose (tyvelose). The structure of the Y. pseudotuberculosis O:4a antigen resembles that of Y. pseudotuberculosis O:2c, which differs in the presence of abequose (3,6-dideoxy-d-xylo-hexose) in place of tyvelose only.  相似文献

Shigella flexneri is the major cause of bacterial shigellosis in developing countries. S. flexneri is divided into at least 19 serotypes, the majority of which are modifications of the same basic O-antigen by glucosylation and/or O-acetylation of its sugar residues by phage encoded serotype-converting genes. Recently, a plasmid encoded phosphoethanolamine (PEtN) modification of the O-antigen has been reported, which is responsible for the presence of the MASF IV-1 determinant and results in conversion of traditional serotypes X, 4a and Y to novel serotypes Xv, 4av and Yv, respectively. In this study, we characterized 19 serotype Yv strains isolated in China. A variant of the O-antigen phosphoethanolamine transferase gene opt (formerly called lpt-O) carried by a pSFxv_2-like plasmid was found in serotype Yv strains, which specifies the phosphorylation pattern on the O-antigen of this serotype. For the majority of the O-antigen units, the PEtN modification occurs on RhaIII, while for a minority, modifications occur on both RhaII and RhaIII. Serotype-specific gene detection and PFGE analysis suggested that these serotype Yv isolates were originated from serotypes Y, Xv and 2a by acquisition of an opt-carrying plasmid and/or inactivation of serotype-specific gene gtrII or gtrX. These data, combined with those of serotypes Xv and 4av reported earlier, demonstrate that the plasmid-encoded PEtN modification is an important serotype conversion mechanism in S. flexneri, in addition to glucosylation and O-acetylation.  相似文献

The 1H and 13C NMR spectral analysis of the glucoside renifolin, isolated from Pyrola renifolia, demonstrated the so far unknown binding site of the glucose moiety to be at C-8 and hence its structure as 8-β-D-glucosyloxy-2,7-dimethyl-1,4-dihydronaphthalen-5-ol. The earlier reported structure for the glucoside pirolatin, isolated from Pyrola japonica, was also reconfirmed by the 13C NMR spectral analysis.  相似文献

Vestibular schwannomas (VSs) are benign tumors arising from eighth cranial nerve and most often occur sporadically in individuals of middle age group. Sporadic VSs are rarely reported in the young population. In this study, we evaluated clinical behaviors of 12 young sporadic VSs by the statistical comparison with a matched series of 145 adult cases. We found that young tumors were characterized by an earlier onset of initial symptom, shorter duration from the first symptom to diagnosis, and larger tumor size than adult ones. Standard sequencing demonstrated the presence of NF2 mutations in eight tumors. All NF2 mutations identified were truncating mutations (nonsense, frameshift, and splicing-site mutations). Earlier formation of VSs in young patients was evidenced by the high incidence of NF2 mutations (66.7 %) far beyond our previous study in the adult case series (34.5 %). Furthermore, young tumors exhibited deficient merlin or heightened phosphorylated merlin that was subsequently demonstrated to be well correlated with increased tumor size. Finally, we compared protein levels of four pathogenesis-related molecules between young and adult group but there was no significant difference. These results led us to suggest that high frequency of NF2 mutations may play a critical role in early tumorigenesis of young VSs. Moreover, merlin deficiency or phosphorylation status of merlin was involved in their earlier development. Further study remains to fully understand the mechanism for the rapid growth of young VSs.  相似文献

A series of dipeptides were synthesized generally in good yields with carbamoylmethyl (Cam) esters as acyl donors in the presence of a cysteine protease, papain, immobilized on Celite. Several segment condensations were also achieved generally in high yields without danger of racemization and formation of the secondary-hydrolysis product. Moreover, partial sequences of some bioactive peptides were prepared through segment condensations, and aimed-at peptides were obtained generally in high yields without the racemization of C-terminal residues of the carboxyl components. Thus, the superiority of the Cam ester in the kinetically controlled peptide synthesis was once again ascertained in couplings mediated by the cysteine protease as in those catalyzed by the serine proteases reported earlier.  相似文献
