Hydroxamic acid levels in Chilean and British wheat seedlings   总被引:7,自引:0,他引:7  
Fifty-two cultivars of Chilean wheats were screened, using high pressure liquid chromatography, for hydroxamic acid levels at the seedling stage when maximal levels occur. DIMBOA levels ranged from 1.4 to 10.9 mmol/kg fr. wt and DIBOA levels ranged from 0 to 1.1 mmol/kg fr. wt. The family of wheat cultivars leading through breeding to the British cultivar Maris Freeman was examined. While DIMBOA and DIBOA levels in the ancestors ranged from 1.6 to 3.8 mmol/kg fr. wt and 0.16 to 0.34 mmol/kg fr. wt, respectively, DIMBOA and DIBOA levels in Maris Freeman were 2.39 and 0.24 mmol/kg fr. wt, respectively. The results are discussed in terms of the possibility of breeding for aphid resistance by making use of hydroxamic acids.  相似文献

The content of hydroxamic acids in wheat plants shows substantial variations between different species and cultivars (1.0–6.3 mmol/kg fr. wt). It also varies with the age of the plant and the organ assayed. The maximum concentration is reached by the fourth day in epicotyls and roots. The amount in leaves at different plant ages is always higher in the younger leaves. Based on feeding and infestation experiments, it is proposed that the distribution of aphids on leaves of different ages is regulated by the hydroxamic acid content.  相似文献

From a previous worldwide screen of wheat cultivars as seedings, five were selected to encompass a wide concentraiton range of the hydroxaminc acid DIMBOA (0.99-8.07 mmol Kg -1 fr. wt). Compared with those in seedings, the concentrations of DIMBOA in flag leaves of the same cultivars were relatively low (0.06-0.87 mmol kg-1 fr. wt). Concentraions of hydroxamic acids in whole ears at anthesis and the early milk stage were even lower, ranging from undetectable to 0.05 mmol kg-1 fr. wt. The three-day mean relaive growth rate (MRGR) was determined for nymphs of Sitobion avenace on the flag leaf (GS 39) and on the ear (GS 60-60) of the five cultivars. No significant differences in MRGR were found between cultivatrs at the same growth stage, although MRGR was higher on ears on than on flag leaces. The results are discussed in realtion to the potential of hydroxamic acids in programmses aimed at breeding mature-plant aphid resistance into wheat cultivars.  相似文献

Secondary metabolites, DIBOA, HBOA, 7-OH-HBOA and BOA, were isolated and quantified from S. dulce (Nees), a native species in Chile belonging to the Acanthaceae family. The highest DIBOA and HBOA contents were determined in leaves (9.25 mmol kg(-1) fr. wt) and root (6.81 mmol kg(-1) fr. wt), respectively. Aglycones, 7-OH-HBOA and HBOA, were isolated together from root extracts of Acanthaceae species. Both, HBOA and 7-OH-HBOA should be direct precursors in the biosynthesis of DIBOA and DIMBOA, respectively.  相似文献

The maize orange leafhopper Cicadulina bipunctata (Hemiptera: Cicadellidae) induces galls characterized by growth stunting and severe swelling of leaf veins on various plants of Poaceae. Previous studies revealed that galls are induced not on feeding site but on distant, newly extended leaves during the feeding, and strongly suggested that some chemicals injected by the leafhopper affect at the leaf primordia. To approach the mechanism underlying gall induction by C. bipunctata, we examined physiological response of plants to feeding by the leafhopper. We performed high-throughput and comprehensive plant hormone analyses using LC-ESI-MS/MS. Galled maize leaves contained higher contents of abscisic acid (ABA) and trans-Zeatin (tZ) and lower contents of gibberellins (GA1 and GA4) than ungalled maize leaves. Leafhopper treatment significantly increased ABA and tZ contents and decreased GA1 and GA4 contents in extending leaves. After the removal of leafhoppers, contents of tZ and gibberellins in extending leaves soon became similar to the control values. ABA content was gradually decreased after the removal of leafhoppers. Such hormonal changes were not observed in leafhopper treatment on leaves of resistant maize variety. Water contents of galled leaves were significantly lower than control leaves, suggesting water stress of galled leaves and possible reason of the increase in ABA content. These results imply that ABA, tZ, and gibberellins are related to gall induction by the leafhopper on susceptible variety of maize.  相似文献

The localization of β -glucosidase was determined at the tissue level in roots and shoots of rye, wheat and maize seedlings, using an immunohistochemical approach with antibodies directed against purified maize β -glucosidase as the primary antibody. In the roots, the β -glucosidase was found in the epidermis and the underlying cell layer. In the leaves, staining was seen in the epidermis (rye and wheat) and nearby vascular tissue (rye, wheat and maize). In all 3 species, β -glucosidase activity was highest in the coleoptile. Here the enzyme was restricted to the epidermis in wheat and to cells near the vascular tissue in maize, but was found in the whole tissue, except the vascular tissue, in rye. Maize, wheat and rye all contain hydroxamic acid glucosides and results are discussed in relation to a proposed role of β -glucosidase as part of a defense system releasing hydroxamic acid aglucone upon herbivore attack, pathogen penetration or aphid infestation.  相似文献

In a screen of seedlings of a worldwide range of 47 cultivars of Triticum (mainly T. aestivum) the concentration of the hydroxamic acid DIMBOA ranged between 1 and 8 mmol/kg fresh wt. In a bioassay in which alatae of the aphid Sitobion avenae were released among replicated test seedlings, there were highly significant correlations between aphid ‘preference’ and DIMBOA levels in the seedlings. The value of these results in work leading to the production of aphid-resistant cultivars is discussed.  相似文献

The common ice plants (Mesembryanthemum crystallinum) at the stage of five leaf pairs were exposed to cadmium chloride solutions (1, 0.1, and 0.01 mM) under the conditions of water culture. After five days, the partition of cadmium and iron in the plant organs and in the cell structures of the apical root region were investigated. Plant adaptation to excess cadmium in the environment was assessed by an increase in the leaf and root weight, a change in peroxidase activity, and an accumulation of proline. The common ice plant accumulated cadmium mainly in the root system. At a high concentration of cadmium in the nutrient solution (1 mM), its content in the root exceeded 2 g/kg fr wt, while at a concentration of 0.01 mM, it was as low as 10 mg/kg. Dithizone staining of transverse sections of the root apical region showed that, after a 48-h-long exposure of plants to 0.1 mM cadmium chloride, cadmium was localized in the cell walls of endodermis and metaxylem. The level of cadmium in leaves varied from 0.5 to 18 mg/kg fr wt. However, there was only a weak correlation between cadmium accumulation and the extent of a biomass decrease in the leaves of various stories, when cadmium concentration in the medium (1 mM cadmium chloride) was toxic. This fact could be related to a marked efflux of endogenous iron from old leaves into the young ones and to a change in the cadmium/iron ratio in the tissues. Proline accumulation in the third leaf pair and in the roots occurred at a relatively low cadmium content (10–12 mg/kg fr wt) in these organs. Maxima of activity of all three forms of peroxidase, viz., soluble, ionically-bound, and covalently-bound peroxidases, in roots were found at a high accumulation of cadmium in these organs (45 mg/kg fr wt). These maxima exceeded 3–4-fold the activity in aging leaves containing 5 mg cadmium/kg fr wt. A decrease in peroxidase activity in leaves was accompanied by a 3.3-fold decrease in iron content; thus, it could be caused by a deficiency of available iron necessary for the enzyme functioning. It was concluded that the resistance of Mesembryanthemum crystallinum, a halophyte, to excess cadmium content in the medium was achieved by its predominant accumulation in roots, where excess cadmium is compartmentalized in the apoplast and seems to be subjected to detoxification through pectate formation. Moreover, the leaves and, particularly, the roots are characterized by a high activity of the antioxidant systems, such as guaiacol-dependent peroxidases, and an occurrence of proline at modest cadmium concentrations.  相似文献

Certain soybean [Glycine max (L.) Merr.] cultivars that are grown in saline nutrient cultures are killed when the inorganic phosphate (Pi) concentration in the substrate exceeds 0.10 mM. To determine the role of Na and Cl on this adverse salinity×Pi interaction, four cultivars, Clark, Clark 63, Lee, and Lee 74 were grown in the greenhouse in nutrient solutions salinized with 1) Cl and NO3 salts to produce treatments with variable amounts of Cl or 2) with NaCl or KCl and CaCl2 to obtain treatments with and without Na. At an osmotic potential of ?0.34 MPa, all salts enhanced Pi uptake and accumulation in the tissue of plants grown in ≧0.12 mM substrate Pi. Leaf Cl concentration was linearly related (r2≥0.9) to the mole fraction (mf) of Cl in the substrate, therefore excess substrate NO3 did not greatly influence leaf Cl accumulation. Foliar injury was only observed on plants grown in saline solutions at high Pi (≥0.12 mM) and was not alleviated when KCl replaced NaCl in the substrate. This indicates that Na did not play a direct role in the salinity×Pi interaction. However, as the mf of Cl increased, severity of injury increased. The severity of injury, and its symptoms, were dependent upon leaf P and Cl concentration. Plants died when Cl and P in their leaves exceeded 800 and 600 mmol kg?1 dry wt, respectively (e.g., Clark 63 grown at mf of Cl=1). The necrotic leaves were beige in color. Leaves that contained P in excess of 600 mmol kg?1 dry wt and Cl between 150–200 mmol kg?1 dry wt, were severely injured and reddish-brown in color (e.g., Clark 63 at mf of Cl=1/4 and Lee 74 Pi grown at mf of Cl=1). When leaf Cl was below 150 mmol kg?1 dry wt, development of reddish-brown coloration in the leaves was sporadic. The adverse salinity×Pi interaction observed on these soybean variaties, therefore, was caused by a synergistic interaction between P and Cl in the leaves.  相似文献

Stress state in plants caused by salinization conditions is characterized by the disturbance of ionic and osmotic homeostasis. The maintenance of the latter is reached by accumulation of osmolytes including free amino acids and soluble sugars in cells. The free amino acid level in the 8-day-old control seedling leaves was higher, than in the roots, whereas the contrary picture was observed in 17-day-old plant tissues. At the same time 8-day-old seedling roots contained more total sugars, than leaves, although the reduced sugar content was nearly a half of the total sugar content. A decrease of both total and reduced sugar levels was observed in 17-day-old seedling tissues. One-day exposure of 7-day-old seedlings to 0.1 M NaCl increased the free amino acid content especially in roots, than in leaves, and the total sugar content in maize leaves, whereas in roots this level remained without changes. The prolongation of salt exposure to 10 days leads to osmolyte content decrease. The seed treatment with Methyure and Ivine intensified accumulation of free amino acids and soluble sugars in the root and leaf tissues under salinization conditions.  相似文献

The carbohydrate composition of the 80% ethanol-insoluble polysaccharides (EIP) from water extracts of ‘Rutgers,’ rin (ripening inhibitor) and nor (non-ripening) tomatoes has been determined. The amount of EIP extracted from ‘Rutgers’ fruit increased from 0.34 to 0.61 mg/g fr. wt during ripening little change occurred in rin or nor fruit. The carbohydrate composition (μg/g fr. wt) of EIP from mature green fruit was: galacturonic acid (48); rhamnose (3); arabinose (20); xylose (48); mannose (31); glucose (139); galactose (51). The most obvious changes that accompanied ripening were a 7.4-fold and 4-fold increase in galacturonic acid and rhamnose content, respectively. These changes were attenuated in the ripening mutants. EIP was fractionated into three major peaks by using DEAE-cellulose ion exchange chromatography. The first peak, which was not retained by the column, contained predominantly glucose and mannose, with lower amounts of galacturonic acid and galactose. The two retained peaks which eluted at 0.1 and 0.2 M sodium chloride contained primarily galacturonic acid, xylose, galactose and arabinose. The galacturonic acid content of these two fractions increased substantially during ripening, whereas the other components decreased. No changes were evident in the ripening mutants. No increase in water-soluble polysaccharides high in galactose content was observed during ripening.  相似文献

14C- and 15N-anthranilic acid are incorporated into the 1,4-benzoxazin-3-ones in maize seedling leaves with low dilution of the isotope. o-Aminophenol and 3-hydroxyanthranilic acid are not incorporated and are probably not intermediates. We report the variation of all 1,4-benzoxazin-3-one derivatives content detectable in maize with plant age in roots and aerial parts. Our results show that the concentration of hydroxamic acids, 2,4-dihydroxy-7-methoxy-1,4-benzoxazin-3-one glucoside (DIMBOA-Glc) and its 8-methoxylated analogue (DIM2BOA-Glc) is high after seed germination and then decreases with plant age. Nevertheless, these compounds continue to be biosynthesised during 6-10 days after germination. Variation in concentration of N-O-methylated DIMBOA-Glc (HDMBOA-Glc) is similar to the one of hydroxamic acids in aerial parts. On the contrary, in roots, its concentration remains relatively stable with plant age. After 10 days, HDMBOA-Glc becomes the main compound in roots. This compound is also present in higher concentration than hydroxamic acids in the oldest leaf of 20-day-old maize. The presence of four other DIMBOA related compounds in maize plants depends on variety, age and tissue. The role of these compounds in plant resistance to aphids is discussed.  相似文献

Extraction and analytical methods have been refined and newones devised to allow precise determinations by GC-EC of thelevels of abscisic acid (ABA) and indol-3ylacetic acid (IAA)in samples of maize root tips as small as 1.0 g fr. wt. Seminalroots of 5-d-old maize seedlings grown in normal (bulk density1200 kg m–3) and compacted (bulk density 1600 kg m–3)sand/garden loam mixtures have been examined. Seminal rootsfrom compacted soil had an average length of about 40% of thatof control roots and were much thicker. The ABA levels in 10mm tips of impeded roots (c. 25–35 ng g–1 fr.wt.)did not differ significantly from those of normal root tipson both a fresh and dry weight basis. The levels in 0–1mm tips were approximately double those in the remaining 1–10mm zones. IAA levels were increased by about 3 times in impededroots (176.3 as compared with 52.4 ng g–1 fr.wt) and itis concluded that this response is likely to be the main causeof the morphological and growth changes brought about by soilcompaction.  相似文献

Expression of the 1,2-dihydroxynaphthalene dioxygenase (nahC) gene from Pseudomonas putida in tobacco transgenic plants produces notable phenotypic and biochemical changes: retarded growth and rooting and earlier flowering; chlorotic and necrotic spots on leaves; and a threefold increase in the total phenolics in the leaves of 6-week-old plants (94.51 g/g fr wt as compared to 33.18 g/g fr wt in the control) and in the phenylalanine-ammonia lyase activity in 4-week-old plants (0.035 U/g fr wt as compared to 0.014 U/g in the control plants of the same age). The transgenic plants expressing the nahC bacterial gene may serve as a model to study the putative functions of dioxygenases and phenol compounds in plant growth, development, and stress responses.  相似文献

To assess the possible physiological function of chlorogenic acid (CGA, 3-O-caffeoylquinic acid) in vivo , we characterized the free radical scavenging properties of pure phenylpropanoids and leaf extracts against two free radicals, superoxide and the 2,2'-azinobis(3-ethylbenzothiazoline-6-sulphonic acid) (ABTS) radical cation. CGA was found to be a highly efficient scavenger of these free radicals, surpassing the activity of all other phenylpropanoids tested, as well as the 'classical' antioxidant ascorbate. Seasonal differences in the leaf content of CGA were examined in field populations of the broadleaf evergreen Mahonia repens growing in different light environments. Leaves of fully sun-exposed plants contained significantly more (74 ± 10 mmol m–2) CGA in winter than leaves from plants growing under deeply shaded conditions (17 ± 2 mmol m–2). Sun-acclimated, but not shade-acclimated, leaves also produced high levels of anthocyanins in winter, suggesting a simultaneous increase in carbon flow through the phenylpropanoid and flavonoid pathways in response to high light and seasonal low temperature stress. In summer, high light-acclimated leaves contained ≈ 2-fold less CGA than in winter, whereas CGA levels were similar between seasons in shaded leaves. Consistent with the strong scavenging capacity of CGA measured in vitro , a linear correlation was observed between CGA content and the antioxidant activity of leaf extracts in both scavenging assays. On the basis of these results, we propose that CGA is a powerful hydrogen-donating antioxidant that may play an important role in mitigating the effects of oxidative stress in plants.  相似文献

From the pollen of Zea mays, three brassinosteroids, castasterone, typhasterol and teasterone, were identified by GC/MS and/or 1H NMR. Their concentrations in the pollen were shown by GC/SIM to be about 120 μg/kg fr.wt. (castasterone), 6.6 μg/kg fr.wt. (typhasterol) and 4.1 μg/kg fr.wt. (teasterone). It was also found that the anther contained a fairly large amount of brassinosteroids by a bioassay.  相似文献

The inclusion of sub-lethal amounts ofthe herbicide atrazine [2-chloro-4-(ethylamino)-6-(isopropylamino)-s-triazine] in the nutrient solution supplied to maize and barley increased the growth of the root and shoot and the uptake of nitrate. The activities of nitrate and nitrite reductases, glutamine synthetase and glutamate synthase were enhanced and the amino acid and nitrate contents of the xylem sap increased. All these effects of atrazine were found only in plants grown with nitrate as the nitrogen source. The uptake of 15NO3? and its incorporation into protein in the root and shoot of maize and barley seedlings was significantly greater in the atrazine treated plants. However, a stimulation in the incorporation of leucine-[14C] into TCA-precipitable protein of detached leaves from 7-day-old barley seedlings was obtained only in the absence of a supply of combined nitrogen either in the culture medium or in the in vitro incubation mixture containing the labelled amino acid.  相似文献

Ferritins from maize, pea, and soya bean seeds were purified. They contain two polypeptides of 28 and 26.5 kDa. The molecular weight of native pea seed ferritin has been estimated to be 540,000. Pea and maize seed ferritins were compared by reverse phase high performance liquid chromatography, amino acid composition, and two-dimensional gel electrophoresis. They are very similar, although four isoforms of the 28-kDa polypeptide from the pea were observed in contrast to a unique polypeptide in maize. No isoforms of the 26.5-kDa polypeptide were detected. Rabbit antibodies were produced in response to pea seed ferritin. It was shown by Western blot analysis that ferritins of the three plants analyzed share immunological determinants. However, horse spleen ferritin was not recognized by the phytoferritin antibodies. Antibodies were also used to demonstrate that ferritins are not uniformly distributed in different pea organs from 30-day-old iron-unloaded plants. The protein was more abundant in flowers than in fruits and roots, and was not detected in leaves.  相似文献

In the present study, we isolated novel tocochromanols from green leaves of Kalanchoe daigremontiana and primary leaves of etiolated seedlings of Phaseolus coccineus that were identified as β-, γ-, and δ-tocomonoenols with unsaturation at the terminal isoprene unit of the side chain. The content of γ-tocomonoenol in leaves of etiolated bean increased gradually with the age of seedlings, reaching 50% of the γ-tocopherol level in 40-day-old plants. The content of this compound in leaves was increased by short illumination of etiolated plants and by addition of homogentisic acid, a biosynthetic precursor of tocopherols. These data indicated that γ-tocomonoenol is synthesized de novo from homogentisic acid and tetrahydro-geranylgeraniol diphosphate, a phytol precursor. Based on these results, a biosynthetic pathway of tocomonoenols is proposed.  相似文献
