通过油菜子叶外植体-农杆菌共培养法将苏云金杆菌杀虫蛋白基因导入甘蓝型油菜,获得抗虫的转基因植株。带有1 ̄2mm子叶柄的油菜子叶经农杆菌感染后,共培养2 ̄3天,然后转移到附加15mg/L卡那霉素的MS选择培养基上筛选转化愈伤组织及不定芽。卡那霉素抗性苗相继在含20 ̄50mg/L卡那霉素的选择培养基上继代培养,再转移到含25mg/L卡那霉素的生根培养基上诱导生根。以苏云金杆菌杀虫蛋白基因为探针,进行1  相似文献

胡杨(Populus euphratica)是唯一能在沙漠里生长的高大乔木树种, 建立其转基因体系可为胡杨抗逆分子机制与应用技术研究提供基本方法。通过研究农杆菌介导的胡杨转GUS基因的技术体系得出以下结论: (1) 胡杨再生植株体系, 叶片在附加1.0 mmol.L-1 6-BA和5.0 mmol.L-1 NAA的1/2MS培养基上不定芽诱导率较高; (2) 转基因体系, 胡杨叶片在含100 mmol.L-1 乙酰丁香酮的OD600值为0.4-0.6的根癌农杆菌菌液中浸染15分钟, 共培养2天, GUS基因转化效率较高; (3) 转基因植株抗生素筛选, 转GUS基因胡杨叶片用300 mg.L-1头孢霉素抑制农杆菌生长, 在含9 mg.L-1 G418的培养基上诱导不定芽以获得转基因的抗性植株。  相似文献

以诸葛菜下胚轴和子叶为材料,在附加BA和NAA的MS培养基上诱导芽再生,在1/2MS培养基上诱导生根,获得完整再生植株,建立了诸葛菜组织培养高频再生体系,再用很癌农杆菌介导转化诸葛菜下胚轴和子叶,在附加一定量的氨苄青霉素、头孢霉素和卡那霉素的相应培养基上进行筛选,并培养再生成苗,获得完整抗性再生植株,移植到盛有土壤的花盆中均可存活,生长正常。将再生植株叶片,进行GUS、NPTⅡ酶活性测定和Southernblot分子杂交,证实外源基因已稳定整合到植物基因组中,并高效表达。  相似文献

反义磷脂酶Dγ(Anti-PLDγ)基因转化美洲黑杨G2的研究   总被引:5,自引:0,他引:5  
通过农杆菌介导法将反义磷脂酶DT(Anti-PLDγ)基因转入美洲黑杨G2中以提高其耐盐性。本研究建立了美洲黑杨G2组培再生体系,确立了美洲黑杨G2叶片分化最适宜培养基和生根培养基,进行了卡那霉素(选择性抗生素)敏感性试验,改良了传统的农杆菌介导转化法。经诱导不定芽及生根阶段卡那霉素(选择性抗生素)连续筛选,获得了21株卡那霉素抗性植株,抗性植株经PCR及PCR-Southern杂交检测,有13株均呈阳性,证明Anti-PLDγ基因成功整合到美洲黑杨G2基因组中。耐盐性实验表明,4株转基因植株抗NaCl能力比对照有不同程度提高。  相似文献

洋桔梗是国际上十分流行的盆花和切花种类。以洋桔梗‘圣剑’无菌苗叶片为外植体,研究了6-BA与NAA不同浓度组合对其不定芽再生的影响,并分别比较了不同浓度IBA和NAA诱导其生根的效果,测定了该品种在不定芽再生时对卡那霉素(Km)的敏感性。结果表明:MS+0.5 mg·L-16-BA+0.01 mg·L-1NAA为不定芽再生最适培养基,不定芽再生率达91%;1/2 MS+0.2 mg·L-1IBA为不定根再生的最适培养基,生根率达89%;抑制叶片不定芽再生的Km最低浓度为25 mg·L-1。建立了‘圣剑’洋桔梗植株高频再生体系,并确定了其对卡那霉素的敏感性,为该品种的基因工程研究奠定了基础。  相似文献

以苹果优良矮化砧木‘JM7’ (Malus prunifolia×M. pumila ‘Malling 9’)为试材, 研究了基本培养基对试管苗增殖生长的影响、蔗糖浓度对试管苗生根的影响及基本培养基、细胞分裂素种类和浓度对离体叶片不定梢再生的影响。结果表明: 基本培养基MS比QL显著提高增殖梢数, 但QL比MS更有利于获得健壮生长的绿苗。3%蔗糖浓度比2%的不定根发生速度快。叶片不定梢再生最适宜的基本培养基是QL。在QL培养基上, 6-BA和TDZ对离体叶片不定梢再生率的影响无显著差异, 但6-BA诱导产生的不定芽在不定梢诱导培养基上可直接伸长生长形成不定梢, 而TDZ诱导产生的不定芽需转移到不加TDZ而加低浓度6-BA的培养基上形成伸长生长的不定梢。  相似文献

采用诸葛菜子叶为材料。在MS+BA 3mg/l,+NAA O.2mg/L培养基上诱导芽的再生,1/2 MS+IBA 0.03mg/L.上诱导生根,获得完整再生植株,建立了诸葛莱组织培养高频再生系统,其再生率可达100%。采用土壤杆菌介导转化诸葛菜子叶.在附加一定量的氨苄青霉索、头孢霉素和卡那霉素的相应培养基上进行筛选,进而培养出再生成苗。获得完整植株,经GUS和NPTI酶活测定,以及Southern blot分析.证实为转基因植株。转化子叶的芽再生率为51%.获得完整转基因植株的转化率为5.53%。在国内外首次建立了以根癌土壤杆菌(Agrobacterium tumefaciens)为载体,诸葛菜子叶为受体的遗传操作系统。  相似文献

In experiments on Black Sea skates (Raja clavata), the potential of the receptor epithelium of the ampullae of Lorenzini and spike activity of single nerve fibers connected to them were investigated during electrical and temperature stimulation. Usually the potential within the canal was between 0 and –2 mV, and the input resistance of the ampulla 250–400 k. Heating of the region of the receptor epithelium was accompanied by a negative wave of potential, an increase in input resistance, and inhibition of spike activity. With worsening of the animal's condition the transepithelial potential became positive (up to +10 mV) but the input resistance of the ampulla during stimulation with a positive current was nonlinear in some cases: a regenerative spike of positive polarity appeared in the channel. During heating, the spike response was sometimes reversed in sign. It is suggested that fluctuations of the transepithelial potential and spike responses to temperature stimulation reflect changes in the potential difference on the basal membrane of the receptor cells, which is described by a relationship of the Nernst's or Goldman's equation type.I. Evolution of living organisms is closely connected with evolution of structure of the system of regulations and its mechanisms. The functional ground of regulations is chemical signalization. As early as in unicellular organisms there is a set of signal mechanisms providing their life activity and orientation in space and time. Subsequent evolution of ways of chemical signalization followed the way of development of delivery pathways of chemical signal and development of mechanisms of its regulation. The mechanism of chemical regulation of the signal interaction is discussed by the example of the specialized system of transduction of signal from neuron to neuron, of effect of hormone on the epithelial cell and modulation of this effect. These mechanisms are considered as the most important ways of the fine and precise adaptation of chemical signalization underlying functioning of physiological systems and organs of the living organism  相似文献
