In order to investigate the potential causal link between the rhythm of activity and body temperature, we simultaneously recorded rectal temperature and total locomotor activity in five clinically healthy female rabbits (blue Vienna breed), 12 week old and mean body weight 2.7 ± 0.3. Animals were housed in individual cages (90?×?50?×?35 cm) under natural 12/12 light/dark cycle. Total locomotor activity was monitored for 15 days by an activity data logger. On day 1, 5, 10, and 15 rectal temperature was recorded every 2 h for a 24-h period. Application of single cosinor method showed a nocturnal daily rhythm of rectal temperature with a range of oscillation of about 1 °C, acrophase after dusk and low robustness value. The daily rhythm of locomotor activity showed its acrophase in the middle of the scotophase and a high robustness value. This information improves the knowledge available on the circadian biology of rabbits useful in the evaluation of physiology of this species.  相似文献

The aim of the current investigation was to study the effect of lithium on circadian rhythms of pineal - testicular hormones by quantitations of pineal and serum serotonin, N-acetylserotonin and melatonin, and serum testosterone at four time points (06.00, 12.00, 18.00 and 24.00) of a 24-hr period under normal photoperiod (L:D), reversed photoperiod (D:L), constant light (L:L) and constant dark phase (D:D) in rats. Circadian rhythms were observed in pineal hormones in all the combinations of photoperiodic regimens, except in constant light, and in testosterone levels in all the photoperiodic combinations. Pineal and serum N-acetylserotonin and melatonin levels were higher than serotonin at night (24.00 hr), in natural L:D cycle, in reversed L:D cycle or similar to normal L:D cycle in constant dark phase, without any change in constant light. In contrast, testosterone level was higher in light phase (12.00 hr through 18.00 hr) than in the dark phase (24.00 hr through 06.00 hr) in normal L:D cycle, in reversed L:D cycle, similar to normal L:D cycle in constant dark (D:D), and reversed to that of the normal L:D cycle in constant light (L:L). Lithium treatment (2 mEq/kg body weight daily for 15 days) suppressed the magnitude of circadian rhythms of pineal and serum serotonin, N-acetylserotonin and melatonin, and testosterone levels by decreasing their levels at four time points of a 24-hr period in natural L:D or reversed D:L cycle and in constant dark (D:D). Pineal indoleamine levels were reduced after lithium treatment even in constant light (L:L). Moreover, lithium abolished the melatonin rhythms in rats exposed to normal (L:D) and reversed L:D (D:L) cycles, and sustained the rhythms in constant dark. But testosterone rhythm was abolished after lithium treatment in normal (L:D)/reversed L:D (D:L) cycle or even in constant light/dark. The findings indicate that the circadian rhythm exists in pineal hormones in alternate light - dark cycle (L:D/D:L) and in constant dark (D:D), but was absent in constant light phase (L:L) in rats. Lithium not only suppresses the circadian rhythms of pineal hormones, but abolishes the pineal melatonin rhythm only in alternate light - dark cycles, but sustains it in constant dark. The testosterone rhythm is abolished after lithium treatment in alternate light - dark cycle and constant light/dark. It is suggested that (a) normal circadian rhythms of pineal hormones are regulated by pulse dark phase in normal rats, (b) lithium abolishes pineal hormonal rhythm only in pulse light but sustains it in constant dark phase, and (c) circadian testosterone rhythm occurs in both pulse light or pulse dark phase in normal rats, and lithium abolishes the rhythm in all the combinations of the photoperiod. The differential responses of circadian rhythms of pineal and testicular hormones to pulse light or pulse dark in normal and lithium recipients are discussed.  相似文献

The aim of this study was to evaluate the circadian rhythm of daily locomotor activity in sea bass exposed to three different artificial lighting regimes including 12/12, 24/0, and 0/24 L/D periods, and scheduled daytime feeding. Daily rhythm of locomotor activity during 12/12 L/D was observed, whereas locomotor activity recorded during 24/0 and 0/24 L/D resulted arrhythmic. Statistical differences in mesor values among the photoperiods and between light and dark phases of each photoperiod were found. During the 12/12 and 0/24 L/D, the fish were active mostly during the light phase. During 24/0 L/D, a phase inversion occurred. The results showed that photoperiod is a dominant synchronizer in sea bass, and that the appetite in this fish follows daily rhythms which does not match the imposed feeding schedule, suggesting the absence or the lability of internal process that couples feeding cycles and biological rhythms.  相似文献

We investigated the daily rhythm of the response to noradrenaline injections in Djungarian hamsters (Phodopus sungorus sungorus) at neutral ambient temperature, under long photoperiod (L:D 12:12) and after four weeks of acclimation to cold (10ºC) and short photoperiod (L:D 8:16). Animals were injected with noradrenaline (0.6 mg/kg) every four hours. Body temperature and gross motor activity were measured with MiniMitter transmitters implanted into abdominal cavity. Additionally, we measured body weight and food intake prior to, and after acclimation. After four weeks of acclimation, the experiment was performed under LD cycle and then repeated during one-day of constant light (LL) and constant darkness (DD). In animals acclimated to L:D 12:12 and ambient temperature of 25ºC, noradrenaline injections caused short-lasting increase in body temperature followed by marked decrease. There was no significant difference in the magnitude of the reaction between light and dark phase of the day. After acclimation to cold and L:D 8:16, under LD conditions, we recorded significant differences between the responses to the noradrenaline injections during light and dark phase of the day. Post-injection increase was higher during the day than during the night while following noradrenaline-induced hypothermia was much more pronounced in darkness. In experiments performed after acclimation to cold and short photoperiod but during one day of LL and DD regimes, these differences were attenuated. Data presented here indicate that in cold acclimated hamsters, the response to exogenous noradrenaline depends on the time of injection and it exhibits clear daily rhythm. The rhythmicity is altered under LL and DD regimes. It seems that post-injection increase in body temperature elicits following hypothermia. This hypothermia might be of a great ecological importance. Reasonable lowering of body temperature would be a protective mechanism, allowing for energy charge restoration.  相似文献

The ability to predict and adjust physiology and behavior to recurring environmental events has been necessary for survival on Earth. Recent discoveries revealed that not only changes in irradiance but also light spectral composition can stimulate the suprachiasmatic nucleus (SCN), ensuring the body’s synchronization to the environment. Therefore, using a lighting system that modulates spectral composition during the day using combined red-green-blue (RGB) lights, we evaluated the effect of variations in light spectral composition on the rest-activity rhythm of rodents. Male Wistar rats (n = 17) were gestated and raised under different lighting conditions and exposed to a long photoperiod (16 h light: 8 h dark). The difference between groups was the presence of variations in light spectral composition during the day (RGB-v) to simulate daily changes in natural light, or not (RGB-f). After weaning, spontaneous motor activity was recorded continuously for rhythm evaluation. Our results indicated that animals under RGB-v did not present a reactive peak of activity after the beginning of the light phase, suggesting that this group successfully detected the variations aimed at mimicking daily alterations of natural light. Furthermore, RGB-v animals exhibited an earlier activity acrophase in comparison to animals under RGB-f (RGB-v = 12:16 – “hh:mm”, RGB-f = 13:02; p < 0.001), which might have been due to the capability to predict the beginning of the dark phase when exposed to variations in light spectrum. However, this earlier activity acrophase can be also explained by the blue-light peak that occurred in RGB-v. The spectral and waveform analysis of daily patterns of motor activity revealed that rats in the RGB-v group were better entrained to a circadian rhythm throughout the experiment. RGB-v showed higher interdaily stability (IS) values (29.75 ± 6.5, n = 9) than did RGB-f (t(15) = 2.74, p = 0.015). Besides, the highest power content (PC) on the first harmonic (circadian) was reached earlier in the RGB-v group. The circadianity index (CI) of the whole period was higher in the RGB-v group (t(15) = 3.47, p = 0.003). Thus, we could consider that locomotor activity rhythm was entrained to the light-dark cycle in the RGB-v rats earlier compared to the RGB-f rats. Our results provide additional evidence for the effect of variations in light spectral composition on the rest-activity pattern of nocturnal rodents. This suggests that these animals might predict the arrival of the activity phase by its advanced acrophase when exposed to RGB-v, demonstrating a better synchronization to a 24-h rhythm.  相似文献

The aim of the present study was to monitor the daily rhythm of rectal and cutaneous temperatures together with the changes in the serum levels of mitochondrial uncoupling protein 1 (UCP1) in equine and goat species. On five gelding horses and five female goat rectal and cutaneous temperatures were recorded at 4 h intervals for a 48 h period. At the same time points blood samples were collected from each animal. Daily rhythms of rectal and cutaneous temperatures were observed in both species, in both day of monitoring. Mesor value of rectal temperature was statistically higher of about 4 °C than cutaneous temperature, and acrophase was postponed of about 2 h in both day of monitoring and in both species. UCP1 did not show daily rhythmicity in horses and goats. We can speculate that the thermogenesis due to thermal system is auxiliary to keeping the body temperature daily rhythm.  相似文献

Marine and estuarine crabs brood attached eggs, which hatch synchronously releasing larvae at precise times relative to environmental cycles. The subtidal crab Dyspanopeus sayi has a circadian rhythm, in which larvae are released within the 4-h interval after the time of ambient sunset. Previous studies demonstrated that the rhythm can be entrained by the light:dark cycle. Since subtidal crabs are also exposed to temperature fluctuations, an unstudied question was whether the circadian rhythm could be entrained by the diel temperature cycle. To answer this question, ovigerous D. sayi were entrained in darkness to 2.5, 5, and 10 °C temperature cycles that were reverse in phase from the ambient temperature cycle. After entrainment, larval release times were monitored in constant conditions of temperature and darkness with a time-lapse video system. The effectiveness of a temperature cycle to shift the timing of larval release increased as the magnitude of the temperature cycle increased and as crabs were exposed to increasing numbers of entrainment cycles. However, entrainment to a 10 °C cycle only lasted 2 days in constant conditions. When crabs were entrained to a light:dark vs. a 10 °C temperature cycle, the light:dark cycle was dominant for entrainment. Nevertheless, ovigerous crabs do sense temperature cycles and in areas where daylight is too low for entrainment, temperature cycles can be used to regulate the time of larval release.  相似文献

The chronobiological basis of the daily dynamics of thermopreference was tested in adults of Triatoma infestans, continuously registering the preferred temperature of insects released over a temperature gradient for 13 days. We found that the thermopreference in T. infestans is a dynamic process that depends on the time of the day and the post-feeding time. When submitted to a 12:12 h light/darkness cycle (L/D), the preferred temperature reached the highest and the lowest values at the end of the light and dark phases, respectively. This daily rhythm persisted under constant conditions of illumination (D/D and L/L), suggesting the existence of an internal oscillator controlling this behaviour. Statistical analysis revealed that the thermopreference of insects kept under L/D exhibited a ca. 24 h periodicity, while insects kept in D/D and L/L showed free-running periods of tau((D/D))=23.35 and tau((L/L))=27.35 h, respectively. The persistence of a cyclic pattern of thermopreference under constant conditions, and free-running periods, close to, but different from 24 h, demonstrate the existence of an endogenous control of the thermopreference in this species. The biological relevance of these results is discussed in the light of the hypothesis that both the length of time elapsed since feeding and the time of the day modify thermopreference in these bugs. The gradual decline in preferred temperature following feeding may be associated with energy conservation during starvation. The rhythmic modulation of thermopreference may be associated with the daily rhythm of locomotion activity shown by these bugs.  相似文献

To assess the accuracy of infrared methodologies for daily rhythm monitoring of skin temperature, five clinically healthy Italian Saddle gelding horses, and five not pregnant and not lactating Camosciata goats, were monitored every 4 h over a 48 h period. The horses were housed in individual boxes, while the goats in two indoor pens, under natural photoperiod and natural environmental temperature. In each animal, skin temperature was recorded with the use of a digital infrared camera and a non-contact infrared thermometer, in five regions: neck, shoulder, ribs, flank and croup. Recorded values were compared with the well-established daily rhythm of rectal temperature. Rectal temperature was recorded at the same time by means of a digital thermometer. In horses, a lower value of skin temperature was recorded using the infrared thermometer for the croup region compared to shoulder and flank; a lower value of skin temperature was recorded using thermography for the croup region compared to the shoulder. In goats, a lower value of skin temperature was recorded using the infrared thermometer for the croup region compared to the flank. In both species, higher values of rectal temperature were observed, compared to the temperature recorded at the skin regions using the other two methodologies. Cosinor rhythmometry showed a daily rhythm of rectal and skin temperature recorded using both methodologies in all the examined regions. General linear model (GLM) showed statistically significant effect of breed on all rhythmic parameters; of day of monitoring on amplitude; of site of recording (rectal vs skin regions) on mesor, amplitude and acrophase; and no effect of methodologies used. The results of this study show the differences in rhythmicity of various body regions temperature and their differences in comparison with daily rhythm rectal temperature. The use of infrared methodologies was inaccurate in assessing body core temperature, but its use could be considered for the evaluation of inflammation in the different body sites.  相似文献

Pinealectomized and control groups of Single Comb White Leghorn pullets were housed in individual laying cages within an experimental room maintained at a temperature of 22 ± 2°C. Hourly feed intake data were collected on the birds subjected to single 3-h changes in the duration of light or dark periods at either auroral (lights-on) or vespertine (lights-off) time of a 14L:10D lighting cycle. Feed intake reached an acrophase at either the 12th or 13th hour of the light period then declined until the onset of darkness. Vespertine changes in the light cycle were more effective in shifting the intake acrophase than the auroral changes. This observation was consistent irrespective of the direction of the change. When the laying hens were subjected to a 26-h lighting rhythm, the strength of cyclic light as a zeitgeber for feed intake rhythm was again demonstrated. The feed intake rhythm developed a 26-h duration with an acrophase consistently 22–23 hours post-vespertine. Pinealectomy did not effect the ability of hens to adjust to new lighting regimens.  相似文献

Daily increments were demonstrated in the sagitta otoliths of fast- and slow- growing Atlantic salmon parr, Salmo salar L., when held under natural photoperiod and temperature. Otolith increments continued to be deposited at a daily rate when fish were held under constant light and/or temperature and on single or multiple feeding regimes. However abnormally short photoperiods of 6L: 6D induced two increments per day. The results suggest that an endogenous rhythm, synchronized lo light/dark transitions within a 24 h period, controls otolith increment deposition.  相似文献

Animals secrete glucocorticoids to deal with daily stressors. Studies have found that supplemental melatonin decreases glucocorticoid metabolite levels in stressed animals. We determined the effect of light interference (LI) and supplemental melatonin on (1) body mass, (2) food intake and (3) glucocorticoid metabolite levels of the striped mouse (Rhabdomys pumilio). Experiment was split into three phases: 8 L: 16 D; 8 L: 16 D with a 15 min light interruption every 4 h; and 8 L: 16 D with a 15 min light interruption every 4 h and melatonin (0.2 μg/ml) added to the water. Body mass was significantly different between phases with lowest body mass (89.17 ± 6.56 g) occurring during standard 8 L: 16 D. LI and melatonin significantly increased body mass. LI increased and melatonin decreased glucocorticoid metabolite levels. LI significantly increased and melatonin significantly decreased assimilation efficiencies possibly due to changes in energetic demands.  相似文献

ABSTRACT

The diurnal and circadian profiles of pineal indoles, except melatonin, are poorly characterized in birds. Moreover, there are no data on the effect of sudden changes in the light–dark cycle on these profiles. Therefore, we investigated the diurnal (Experiment I) and circadian variation (Experiment II) of nine pineal indoles (tryptophan, 5-hydroxytryptophan, serotonin, N-acetylserotonin, melatonin, 5-hydroxyindole acetic acid, 5-methoxytryptophol, 5-methoxyindole acetic acid, 5-methoxytryptamine) in geese, as well as the changes in the profiles of these substances in geese subjected to a reversed light–dark cycle (Experiment III). For the first 12 weeks of life, all geese were kept under a diurnal cycle of 12 h of light and 12 h of darkness (12L:12D). In Experiment I (n = 48), they were kept under these conditions for another 14 days before being sacrificed at 2-h intervals for sampling of the pineal glands. In Experiment II, the geese (n = 48) were divided into three groups (12L:12D, 24L:0D, 0L:24D) for 10 days before sampling at 6-h intervals. In Experiment III, 24 geese were exposed to a reversed light–dark cycle before sampling at 14:00 and 02:00 on the first, second and third days after light–dark cycle reversal. To determine the content of the indoles in the goose pineals, HPLC with fluorescence detection was used. We found that, with the exception of tryptophan, all the investigated indoles showed statistically significant diurnal variation. When geese were kept in constant darkness, most of the indoles continued to show this variation, but when geese were kept in constant light, the indoles did not show significant variation. When the light–dark cycle was reversed (12L:12D to 12D:12L), the profiles of NAS, melatonin, 5-MTAM and 5-MTOL reflected the new cycle within 2 days. The content of serotonin in geese in 12L:12D was higher than that observed in other birds under these conditions, which suggests that this compound may play a special role in the pineal physiology of this species. In conclusion, our results show that the daily variations in the metabolism of melatonin-synthesis–related indoles in the goose pineal gland are generated endogenously and controlled by environmental light conditions, as in other birds. However, comparison of the results obtained with the goose to those obtained with other species (chicken, duck) unambiguously shows that the profiles of pineal indoles differ markedly between species, in both the quantitative proportions of the compounds and the characteristics of the diurnal changes. These findings provide strong arguments for the need for comparative studies.  相似文献

Photoperiod (=day length) is the vital factor for the regulation of behavioral and physiological activities in many avian species. This study investigated the seasonal cycles of testicular growth and secondary sexual characteristics of Indian weaver bird under natural day length (NDL) and the effects of duration and intensity of light on photoperiodic induction. In the first experiment, groups of birds (n = 7 each) were exposed to under NDL in April 2008 and May 2009 for 8 and 12 months, respectively. In second and third experiment, birds (n = 6 each group) were exposed to different photoperiods (11.5L:12.5D, 12L:12D, 13L:11D, and 15L:9D) at the same (500 lux) light intensity, and to 13L:11D at different light intensities (10-, 50-, 500-, and 800-lux). Observations on testis size, molt, and plumage score were recorded 2-week (molt and plumage) or at 4-week intervals (testes). Both the NDL groups showed similar seasonal cycles of testicular growth-regression and secondary sexual characteristics. Second and third experiments suggest that the photoperiodic induction was depending upon duration and intensity of the light. Birds showed testicular growth-regression cycle followed by molt and plumage color change only under 13L:11D and 15L:9D and only 500- and 800-lux under 13L:11D photoperiod but not under 11.5L:12.5D and 12L:12D and 10- and 50-lux light intensities. Pre- and post-nuptial molting on body feathers were progressed with gonadal stimulation–maturation and regression cycle under 13L:11D and 15L:9D. Results under different light–dark cycles suggest that day length of about 12 h or more and above the threshold level of light intensity are essential for the induction of photoperiodic responses.  相似文献

Abstract

The Locomotory activity of the Plains Garter snake was determined under L/D: 12/12 conditions at five constant temperatures and three light intensities during the light period. The snakes were diurnal at low temperatures with nocturnal activity increasing in various amounts at higher temperatures. The different light intensities had relatively small effects on the activity rhythm.

Activity was recorded under four constant light conditions at five constant temperatures and the characteristics of the free‐running rhythm measured. A comparison of the characteristics of the free‐running rhythm to Aschoff's circadian rule indicates that this snake is an exception to this rule.

Increase light intensity decreased total activity under all conditions. Under a L/D: 12/12 cycle the decrease in activity was more pronounced during the dark period than the light period.

It is suggested that crepuscular or nocturnal activity shown by snakes at high temperatures may be an effect the temperature level has on the biological clock and activity controlling mechanisms rather than temperature selection by the snake.  相似文献

Daily rhythms of body core temperature and liver function were recorded in goats maintained under various schedules of lighting and feeding. Concentration of urea in the blood was used as an index of digestion-driven hepatic activity, whereas concentration of cholesterol served as an index of autonomous hepatic activity. Body temperature exhibited robust circadian rhythmicity in the presence and absence of a light-dark cycle and/or a feeding regime. The rhythm was more responsive to shifts in feeding time than to shifts in the light-dark cycle. Urea concentration in the blood exhibited daily rhythmicity only in the presence of a daily feeding regime and, therefore, was driven by ingestive and digestive processes. The rhythm of cholesterol concentration persisted in the presence or absence of a light-dark cycle and/or a feeding regime, except when the feeding time was shifted under constant light. However, the cholesterol rhythm did not respond either to shifts in the light-dark cycle or, more importantly, to shifts in feeding time. Thus, based on this index of hepatic function, the liver cannot be identified as the site of the putative food-entrainable pacemaker.  相似文献

Entrainment experiments have been carried out with geographically widely separated populations of the sand beach isopod Eurydice pulchra Leach subjected to periods of simulated tidal agitation imposed concurrently with a 24-h light: dark (L: D) cycle. Circatidal swimming rhythms of greatest amplitude were induced when agitation was applied with the subjective timing, within the L: D cycle, of local spring high tides. This occurred in a normal L: D regime and also when the L: D regime was phase shifted through 90°. Animals previously maintained in constant darkness (D: D) and subsequently exposed to simulated tidal disturbance at various times in constant darkness were unable to modulate the amplitude of circatidal swimming activity. Isopods previously maintained in a normal L: D cycle and subsequently subjected to artificial tidal agitation in constant darkness were, however, able to modulate circatidal activity. This indicates that E. pulchra is capable of detecting tidal agitation and daily light cues and using them in conjunction with its circadian “clock” to modulate its endogenous circatidal rhythmicity. The free-running semilunar rhythm of swimming activity entrained only when the timing of agitation within the day/night cycle mimicked the pattern of local spring high tides. Agitation with the timing of neap high tides entrained no free-running circa-semilunar activity pattern.  相似文献

Rectal temperature of 10 female adult horses was recorded every 2 h for 10 consecutive days under a natural winter photoperiod (9 h of light and 15 h of darkness per day). A robust daily rhythm of body temperature was observed in all animals. The rhythm had a mean level of 38.3°C and a range of excursion of 1.0°C. Temperature started its daily ascent at dawn each day and reached a maximum 14 hours later. Body temperature of 5 of the horses was studied for 10 more days under constant illumination. The rhythm persisted under this condition, although with a slightly longer period of 24.2 h, which confirms the endogenous nature of the rhythm. Despite the fact that the body size of the horse is several orders of magnitude greater than that of rodents, the various parameters of the body temperature rhythm of the horse are similar to those of several species of rodents previously studied.  相似文献

Abstract

A study was made on the developmental rhythms of Ephestia kuehniella during different photoperiods: constant lighting (L/L), under conditions of 12 hours light per day (L/D) and in constant darkness (D/D). Observations were made every 2 hrs, for 3 successive 24‐hr periods, of the number of wandering larvae emerging from food, the number of pupations and of imaginai moults. Emergence of wandering larvae is rhythmic only under L/D conditions, and in addition this is also affected by the density of the larvae. Rhythm is least apparent with greatest density of the larvae. Pupation in E. kuehniella is a non‐rhythmic process with each of the photoperiods examined, whereas the imaginai moult is characterized by an endogenous (existing under D/D conditions) rhythm. Emergence of imagines is non‐rhythmic under L/L, and also when pupae under L/D fail to receive the final period of darkness. The rhythm of the imaginai moult is possibly controlled by the biological clock, which stops under conditions of constant light.  相似文献

In rodents, the alternation of light and dark is the main synchronizer of circadian rhythms. The entrainment abilities of the LD cycle could be estimated by experimental modifications of the photoperiod and by following the subsequent temporal distribution of a circadian rhythm. The rate of reentrainment of a rhythm is determined by the nature of the studied variable, by the direction (advance or delay) and the magnitude (or value) of the phase shift. In rodents, core body temperature and motor activity are known to be well synchronized with each other under L:D 12:12 and under constant conditions (LL or DD). There are clear evidences that the circadian pattern of motor activity is generated by two oscillators, one from dusk signal and the other from dawn signal. Whether the circadian rhythms of body temperature and motor activity are generated by a common circadian mechanism or controlled by separate ones still remains unknown. The purpose of this review is to summarize the results obtained on the circadian rhythms of body temperature and motor activity throughout the daily cycle in order to clarify the relationships between these two functions.  相似文献
