Arbuscular mycorrhizal fungi (AMF) can perform key roles in ecosystem functioning through improving host nutrient acquisition. Nitrogen (N) is an essential nutrient for plant growth, however, anthropogenic N loading (e.g. crop fertilization and deposition from combustion sources) is increasing so that N now threatens ecosystem sustainability around the world by causing terrestrial and aquatic eutrophication and acidification. It is important to better understand the capacity of AMF to directly uptake N from soils and transfer it to host plants because this process may increase N recycling and retention within ecosystems. In addition to understanding the role of AMF in the N cycle in the present day it is important to understand how AMF function may change as global change proceeds. Currently the net effects of N enrichment and elevated temperature predicted with global change on AMF are unknown. In this study, we examined the effects of N enrichment by simulated N‐deposition loading, elevated temperatures expected by future global changes and their interactions on growth and AMF‐mediated N acquisition of switchgrass (Panicum virgatum var. Alamo), an important species for biofuel production. Switchgrass plants were grown in microcosm units that divided mycorrhizal roots from AMF hyphae and organic residues enriched with 15N by compartments separated by an air gap to reduce N diffusion. While AMF did not enhance switchgrass biomass, mycorrhizas significantly increased 15N in shoots and total shoot N. Neither N enrichment nor elevated temperatures influenced this mycorrhizal‐mediated N uptake and transfer. Results from this study can aid in developing sustainable bioethanol and switchgrass production practices that are less reliant on synthetic fertilizers and more dependent on internal N recycling from AMF.

Two experiments were conducted where Cucumis sativus were grown in uncompartmented pots either alone or in symbiosis with Glomus intraradices Schenck and Smith (Experiment 1) or Glomus sp. (Experiment 2) in order to investigate if root colonization by arbuscular mycorrhizal (AM) fungi has an effect on depletion of the soil mineral N pool. All pots were gradually supplied with 31 mg NH4NO3-N kg-1 dry soil from 12–19 days after planting and an additional 50 mg (NH4)2SO4-N kg-1 dry soil (15N-labelled in Experiment 1) was supplied at 21 or 22 days after planting in Experiments 1 and 2, respectively. Dry weight of plant parts, total root length, mycorrhizal colonization rate and soil concentration of NH 4 + and NO 3 - were recorded at five sequential harvest events: 21, 24, 30, 35 and 42 days (Experiment 1) and 22, 25, 28, 31 and 35 days (Experiment 2) after planting. In Experiment 1, plants were also analysed for total content of N and 15N. The mycorrhizal colonization rate increased during time: from 25 to 40% in Experiment 1 and from 50 to 60% in Experiment 2. Plant dry matter accumulation was unaffected by mycorrhizal colonization, except in Experiment 1 where shoot dry weights were slightly increased and in Experiment 2 where root dry weights were slightly decreased compared to non-mycorrhizal control plants. The total root length was similar in the control and mycorrhizal treatments in Experiment 1, while it was decreased (20–30%) by mycorrhizal colonization in the last two harvest in Experiment 2. Mycorrhizal colonization affected the rate of depletion of soil mineral N in Experiment 1, where both NH 4 + and NO 3 - concentrations were markedly lower in the first two harvests, when plants were mycorrhizal. As the root length was similar in mycorrhizal and control treatments, this may indicate that the external AM hyphae contributed to the depletion of the soil mineral N pool. A similar pattern was observed in Experiment 2, although the effect was less pronounced. The 15N enrichment in mycorrhizal plants (Experiment 1) also indicated a faster NH 4 + uptake than in the non-mycorrhizal controls in the first two harvests after application of the 15N-labelled N source. However, the external hyphae and roots seemed to have access to the same N sources as the 15N enrichment and total N content were similar in mycorrhizal and control plants at the end of the experiment. This revised version was published online in June 2006 with corrections to the Cover Date.

Nitrogen (N) capture by arbuscular mycorrhizal (AM) fungi from organic material is a recently discovered phenomenon. This study investigated the ability of two Glomus species to transfer N from organic material to host plants and examined whether the ability to capture N is related to fungal hyphal growth. Experimental microcosms had two compartments; these contained either a single plant of Plantago lanceolata inoculated with Glomus hoi or Glomus intraradices, or a patch of dried shoot material labelled with (15)N and (13)carbon (C). In one treatment, hyphae, but not roots, were allowed access to the patch; in the other treatment, access by both hyphae and roots was prevented. When allowed, fungi proliferated in the patch and captured N but not C, although G. intraradices transferred more N than G. hoi to the plant. Plants colonized with G. intraradices had a higher concentration of N than controls. Up to one-third of the patch N was captured by the AM fungi and transferred to the plant, while c. 20% of plant N may have been patch derived. These findings indicate that uptake from organic N could be important in AM symbiosis for both plant and fungal partners and that some AM fungi may acquire inorganic N from organic sources.

Arbuscular mycorrhizal fungi (AMF) can improve plant nutrient acquisition, either by directly supplying nutrients to plants or by promoting soil organic matter mineralization, thereby affecting interspecific plant relationships in natural communities. We examined the mechanism by which the addition of P affects interspecific interactions between a C4 grass (Bothriochloa ischaemum, a dominant species in natural grasslands) and a C3 legume (Lespedeza davurica, a subordinate species in natural grasslands) via AMF and plant growth, by continuous 13C and 15N labelling, combined with soil enzyme analyses. The results of 15N labelling revealed that P addition affected the shoot uptake of N via AMF by Bischaemum and Ldavurica differently. Specifically, the addition of P significantly increased the shoot uptake of N via AMF by Bischaemum but significantly decreased that by Ldavurica. Interspecific plant interactions via AMF significantly facilitated the plant N uptake via AMF by B. ischaemum but significantly inhibited that by L. davurica under P-limited soil conditions, whereas the opposite effect was observed in the case of excess P. This was consistent with the impact of interspecific plant interaction via AMF on arbuscular mycorrhizal (AM) benefit for plant growth. Our data indicate that the capability of plant N uptake via AMF is an important mechanism that influences interspecific relationships between C4 grasses and C3 legumes. Moreover, the effect of AMF on the activities of the soil enzymes responsible for N and P mineralization substantially contributed to the consequence of interspecific plant interaction via AMF for plant growth.

Plant mycorrhizal associations influence the accumulation and persistence of soil organic matter and could therefore shape ecosystem biogeochemical responses to global changes that are altering forest composition. For instance, arbuscular mycorrhizal (AM) tree dominance is increasing in temperate forests, and ericoid mycorrhizal (ErM) shrubs can respond positively to canopy disturbances. Yet how shifts in the co-occurrence of trees and shrubs with different mycorrhizal associations will affect soil organic matter pools remains largely unknown. We examine the effects of ErM shrubs on soil carbon and nitrogen stocks and indicators of microbial activity at different depths across gradients of AM versus ectomycorrhizal (EcM) tree dominance in three temperate forest sites. We find that ErM shrubs strongly modulate tree mycorrhizal dominance effects. In surface soils, ErM shrubs increase particulate organic matter accumulation and weaken the positive relationship between soil organic matter stocks and indicators of microbial activity. These effects are strongest under AM trees that lack fungal symbionts that can degrade organic matter. In subsurface soil organic matter pools, by contrast, tree mycorrhizal dominance effects are stronger than those of ErM shrubs. Ectomycorrhizal tree dominance has a negative influence on particulate and mineral-associated soil organic matter pools, and these effects are stronger for nitrogen than for carbon stocks. Our findings suggest that increasing co-occurrence of ErM shrubs and AM trees will enhance particulate organic matter accumulation in surface soils by suppressing microbial activity while having little influence on mineral-associated organic matter in subsurface soils. Our study highlights the importance of considering interactions between co-occurring plant mycorrhizal types, as well as their depth-dependent effects, for projecting changes in soil carbon and nitrogen stocks in response to compositional shifts in temperate forests driven by disturbances and global change.

Grassland ecosystems store an estimated 30% of the world's total soil C and are frequently disturbed by wildfires or fire management. Aboveground litter decomposition is one of the main processes that form soil organic matter (SOM). However, during a fire biomass is removed or partially combusted and litter inputs to the soil are substituted with inputs of pyrogenic organic matter (py‐OM). Py‐OM accounts for a more recalcitrant plant input to SOM than fresh litter, and the historical frequency of burning may alter C and N retention of both fresh litter and py‐OM inputs to the soil. We compared the fate of these two forms of plant material by incubating 13C‐ and 15N‐labeled Andropogon gerardii litter and py‐OM at both an annually burned and an infrequently burned tallgrass prairie site for 11 months. We traced litter and py‐OM C and N into uncomplexed and organo‐mineral SOM fractions and CO2 fluxes and determined how fire history affects the fate of these two forms of aboveground biomass. Evidence from CO2 fluxes and SOM C:N ratios indicates that the litter was microbially transformed during decomposition while, besides an initial labile fraction, py‐OM added to SOM largely untransformed by soil microbes. Additionally, at the N‐limited annually burned site, litter N was tightly conserved. Together, these results demonstrate how, although py‐OM may contribute to C and N sequestration in the soil due to its resistance to microbial degradation, a long history of annual removal of fresh litter and input of py‐OM infers N limitation due to the inhibition of microbial decomposition of aboveground plant inputs to the soil. These results provide new insight into how fire may impact plant inputs to the soil, and the effects of py‐OM on SOM formation and ecosystem C and N cycling.

Nutrient enrichment can reduce ecosystem stability, typically measured as temporal stability of a single function, e.g. plant productivity. Moreover, nutrient enrichment can alter plant–soil interactions (e.g. mycorrhizal symbiosis) that determine plant community composition and productivity. Thus, it is likely that nutrient enrichment and interactions between plants and their soil communities co-determine the stability in plant community composition and productivity. Yet our understanding as to how nutrient enrichment affects multiple facets of ecosystem stability, such as functional and compositional stability, and the role of above–belowground interactions are still lacking. We tested how mycorrhizal suppression and phosphorus (P) addition influenced multiple facets of ecosystem stability in a three-year field study in a temperate steppe. Here we focused on the functional and compositional stability of plant community; functional stability is the temporal community variance in primary productivity; compositional stability is represented by compositional resistance, turnover, species extinction and invasion. Community variance was partitioned into population variance defined as community productivity weighted average of the species temporal variance in performance, and species synchrony defined as the degree of temporal positive covariation among species. Compared to treatments with mycorrhizal suppression, the intact AM fungal communities reduced community variance in primary productivity by reducing species synchrony at high levels of P addition. Species synchrony and population variance were linearly associated with community variance with the intact AM fungal communities, while these relationships were decoupled or weakened by mycorrhizal suppression. The intact AM fungal communities promoted the compositional resistance of plant communities by reducing compositional turnover, but this effect was suppressed by P addition. P addition increased the number of species extinctions and thus promoted compositional turnover. Our study shows P addition and AM fungal communities can jointly and independently modify the various components of ecosystem stability in terms of plant community productivity and composition.

Both endophytic and mycorrhizal fungi interact with plants to form symbiosis in which the fungal partners rely on, and sometimes compete for, carbon (C) sources from their hosts. Changes in photosynthesis in host plants caused by atmospheric carbon dioxide (CO2) enrichment may, therefore, influence those mutualistic interactions, potentially modifying plant nutrient acquisition and interactions with other coexisting plant species. However, few studies have so far examined the interactive controls of endophytes and mycorrhizae over plant responses to atmospheric CO2 enrichment. Using Festuca arundinacea Schreb and Plantago lanceolata L. as model plants, we examined the effects of elevated CO2 on mycorrhizae and endophyte (Neotyphodium coenophialum) and plant nitrogen (N) acquisition in two microcosm experiments, and determined whether and how mycorrhizae and endophytes mediate interactions between their host plant species. Endophyte‐free and endophyte‐infected F. arundinacea varieties, P. lanceolata L., and their combination with or without mycorrhizal inocula were grown under ambient (400 μmol mol−1) and elevated CO2 (ambient + 330 μmol mol−1). A 15N isotope tracer was used to quantify the mycorrhiza‐mediated plant acquisition of N from soil. Elevated CO2 stimulated the growth of P. lanceolata greater than F. arundinacea, increasing the shoot biomass ratio of P. lanceolata to F. arundinacea in all the mixtures. Elevated CO2 also increased mycorrhizal root colonization of P. lanceolata, but had no impact on that of F. arundinacea. Mycorrhizae increased the shoot biomass ratio of P. lanceolata to F. arundinacea under elevated CO2. In the absence of endophytes, both elevated CO2 and mycorrhizae enhanced 15N and total N uptake of P. lanceolata but had either no or even negative effects on N acquisition of F. arundinacea, altering N distribution between these two species in the mixture. The presence of endophytes in F. arundinacea, however, reduced the CO2 effect on N acquisition in P. lanceolata, although it did not affect growth responses of their host plants to elevated CO2. These results suggest that mycorrhizal fungi and endophytes might interactively affect the responses of their host plants and their coexisting species to elevated CO2.

The source of nitrogen in the spores of arbuscular mycorrhizal (AM) fungi was quantified by a 15N-labeling technique. N was applied as coated urea to the soil and in solution to plant shoots. Soil-applied fertilizer had a significant effect on spore % 15N (P<0.01), with a 24–75% contribution to spore N. Fertilizer applied to either alfalfa shoots or bahia grass shoots had little effect on spore % 15N, accounting for 0–14% or 1–9% of spore N, respectively. These results indicate that AM fungi obtain spore N mostly from the soil. The small amount of spore N originating from shoot-applied N may have been obtained via root exudation. C and N mineralisation kinetics obtained in laboratory incubations during decomposition of crop residues under non-limiting nitrogen conditions were simulated using a simple dynamic model. This model includes three compartments: the residues, microbial biomass and humified organic matter. Seven parameters are used to describe the C and N fluxes. The decomposed C is either mineralised as CO2 or assimilated by the soil microflora, microbial decay producing both C humification and secondary C mineralisation. The N dynamics are governed by the C rates and the C:N ratio of the compartments which remain constant in the absence of nitrogen limitation. The model was parameterised using apparent C and N mineralisation kinetics obtained for 27 different residues (organs of oilseed rape plants) that exhibited very wide variations in chemical composition and nitrogen content. Except for the C:N ratio of the residues and the soil organic matter, the other five parameters of the model were obtained by non-linear fitting and by minimising the differences between observed and simulated values of CO2 and mineral N. Three parameters, namely the decomposition rate constant of the residues, the biomass C:N ratio and humification rate, were strongly correlated with the residues C:N ratio. Hyperbolic relationships were established between these parameters and the residues C:N ratio. In contrast, the other two parameters, i.e. the decay rate of the microbial biomass and the assimilation yield of residue-C by the microbial biomass, were not correlated to the residues C:N ratio and were, therefore, fixed in the model. The model thus parameterised against the residue C:N ratio as a unique criterion, was then evaluated on a set of 48 residues. An independent validation was obtained by taking into account 21 residues which had not been used for the parameterisation. The kinetics of apparent C and N mineralisation were reasonably well simulated by the model. The model tended to over-estimate carbon mineralisation which could limit its use for C predictions, but the kinetics of N immobilisation or mineralisation due to decomposition of residues in soil were well predicted. The model indicated that the C:N ratio of decomposers increased with the residue C:N ratio. Higher humification was predicted for substrates with lower C:N ratios. This simple dynamic model effectively predicts N evolution during crop residue decomposition in soil.

Forests in northeastern North America are influenced by varying climatic and biotic factors; however, there is concern that rapid changes in these factors may lead to important changes in ecosystem processes such as decomposition. Climate change (especially warming) is predicted to increase rates of decomposition in northern latitudes. Warming in winter may result in complex effects including decreased levels of snow cover and an increased incidence of soil freezing that will effect decomposition. Along with these changes in climate, moose densities have also been increasing in this region, likely affecting nutrient dynamics. We measured decomposition and N release from 15N‐labeled sugar maple leaf litter and moose feces over 20 months in reference and snow removal treatment (to induce soil freezing) plots in two separate experiments at the Hubbard Brook Experimental Forest in New Hampshire, USA. Snow removal/soil freezing decreased decomposition of maple litter, but stimulated N transfer to soil and microbial biomass. Feces decomposed more rapidly than maple litter, and feces N moved into the mineral soil more than N derived from litter, likely due to the lower C : N ratio of feces. Feces decomposition was not affected by the snow removal treatment. Total microbial biomass (measured as microbial N and C) was not significantly affected by the treatments in either the litter or feces plots. These results suggest that increases in soil freezing and/or large herbivore populations, increase the transfer rate of N from plant detritus or digested plants into the mineral soil. Such changes suggest that altering the spatial and temporal patterns of soil freezing and moose density have important implications for ecosystem N cycling.

While plant litters are the main source of soil organic matter (SOM) in forests, the controllers and pathways to stable SOM formation remain unclear. Here, we address how litter type (13C/15N‐labeled needles vs. fine roots) and placement‐depth (O vs. A horizon) affect in situ C and N dynamics in a temperate forest soil after 5 years. Litter type rather than placement‐depth controlled soil C and N retention after 5 years in situ, with belowground fine root inputs greatly enhancing soil C (x1.4) and N (x1.2) retention compared with aboveground needles. While the proportions of added needle and fine root‐derived C and N recovered into stable SOM fractions were similar, they followed different transformation pathways into stable SOM fractions: fine root transfer was slower than for needles, but proportionally more of the remaining needle‐derived C and N was transferred into stable SOM fractions. The stoichiometry of litter‐derived C vs. N within individual SOM fractions revealed the presence at least two pools of different turnover times (per SOM fraction) and emphasized the role of N‐rich compounds for long‐term persistence. Finally, a regression approach suggested that models may underestimate soil C retention from litter with fast decomposition rates.

Nitrogen (N) inputs to ecosystems have increased worldwide, often leading to large changes in plant community structure and reducing plant diversity. Yet, the interaction of increased N availability with other factors that determine plant community composition, are still poorly understood. Here, we test whether the impact of N addition on plant communities depends on the presence of arbuscular mycorrhizal fungi (AMF). AMF are widespread plant symbionts that facilitate growth of many plant species. We hypothesize that AM fungi reduce the negative impact of N addition on plant communities by supporting growth of species that are sensitive to N enrichment. We established experimental grassland microcosms consisting of 18 plant species. These microcosms were subjected to high and low N supply and were inoculated with AMF or remained nonmycorrhizal. Both N addition and AMF had a big impact on plant community composition, but with opposite effects. N addition induced a 2.8‐fold increase in grass biomass and reduced legume biomass. Grasses dominated the microcosms at high N supply, especially when AMF were absent. In contrast, AMF enhanced biomass of all legumes species (on average 6.8‐fold) and reduced the relative abundance of grasses. The proportion of legume biomass out of total shoot biomass at high N supply was 19% with AMF and only 3% without AMF. Our results show that responses of plant communities to N enrichment depend on AMF and that AMF can reduce the negative impact of increased N availability on plant community structure by reducing grass dominance.

We examined potential large-scale controls over the distribution of arbuscular mycorrhizal (AM) fungi and their host plants. Specifically, we tested the hypothesis that AM fungi should be more prevalent in biomes where nutrients are primarily present in mineral, and not organic, forms. Values of percentage root length colonized (%RLC) by AM fungi, AM abundance, and host plant availability were compiled or calculated from published studies to determine biome-level means. Altogether, 151 geographic locations and nine biomes were represented. Percent RLC differed marginally significantly among biomes and was greatest in savannas. AM abundance (defined as total standing root length colonized by AM fungi) varied 63-fold, with lowest values in boreal forests and highest values in temperate grasslands. Biomes did not differ significantly in the percentage of plant species that host AM fungi, averaging 75%. Contrary to the hypothesis, %RLC, AM abundance, and host plant availability were not related to the size, influx, or turnover rate of soil organic matter pools. Instead, AM abundance was positively correlated with standing stocks of fine roots. The global pool of AM biomass within roots might approach 1.4 Pg dry weight. We note that regions harboring the largest stocks of AM fungi are also particularly vulnerable to anthropogenic nitrogen deposition, which could potentially alter global distributions of AM fungi in the near future.
