Harvey is generally accepted as the patron saint of cardiology. Some shading is not amiss because a medical history canon is by definition an instantaneous photograph. Therefore De Motu Cordis is set in a historic perspective and portrayed as the result of genuine scientific progress. Nevertheless Harvey symbolises the watershed between ancient medicine and modern medicine based on observational and experimental research. We too seem to witness a transitional zone between modern medicine and a new customised medicine, individualised and integrative, firmly based on a further expanding technology. In vivo and in vitro expressed N-terminal sequence of EWS (EAD) and hsRPB7 (subunit of human RNA polymerase II) were probed for protein–protein interactions using pull-down assays. In result, it was found that the proteins 57Z (residues 1–57 of EAD) and hsRPB7 interact in vitro forming a stable complex. The direct interaction between 57z and hsRPB7 indicate that DHR-related peptides and other small molecules, targeted to N-terminus of EWS might possess therapeutic potentialities as anti-cancer agents to function as inhibitors of EAD-mediated transactivation.  相似文献

Seven essential oils – Rosemary, Wintergreen, Teatree, Patchouli, Palmarosa, Geranium and Eucalyptus were used to determine the in vitro antifungal activity on Phomopsis azadirachtae. This organism is responsible for the destructive die-back disease of neem. The oils mentioned above were screened using the food poisoning technique. All the seven oils showed considerable antifungal activity against P. azadirachtae. After the conduction of the tests using the oils, the results were such that Wintergreen, Geranium and Palmarosa showed complete or 100% inhibition of mycelial growth at a concentration of 2500 ppm. Among the oils tried, Palmarosa proved to be most potent showing complete inhibition of mycelia at 500 ppm. This really suggests that plant sources possess antifungal activity and can be effectively used to treat and manage plant diseases thereby circumventing the fungal spread in our environment.  相似文献

We have previously reported that the catalytic RNA subunit of RNase P of Escherichia coli (M1 RNA) cleaves Drosophila initiator methionine tRNA (tRNA(Met)i) within the mature tRNA sequence to produce specific fragments. This cleavage was dependent on the occurrence of an altered conformation of the tRNA substrate. We call this further cleavage hyperprocessing. In the present paper, to search for another tRNA that can be hyperprocessed in vitro, we used total mature tRNAs from Drosophila as substrates for the in vitro M1 RNA reaction. We found that some tRNAs can be hyperprocessed by M1 RNA and that two such tRNAs are an alanine tRNA and a histidine tRNA. Using mutant substrates of these tRNAs, we also show that the hyperprocessing by M1 RNA is dependent on the occurrence of altered conformations of these tRNAs. The altered conformations were very similar to that of tRNA(Met)i. We show here that M1 RNA can be used as a powerful tool to detect the alternative conformation of tRNAs. The relationship between these hyperprocessing reactions and stability of the tRNA structure will also be discussed.  相似文献

Aim and backgroundTo investigate the feasibility of in vivo rectal dosimetry in image-guided adaptive brachytherapy of cervical cancer.Materials and methodsError of measurement of dose rate in a semiconductor diode probe was investigated depending on the distance and angle in water, and on temperature in a polymethyl methacrylate phantom using an Ir-192 source. Furthermore, the difference between the measured and calculated dose was analysed in the interstitial brachytherapy of 30 cervix cancer patients. The relationship between in vivo measured dose, calculated dose in the point of the diode, calculated maximal dose in the point of the diodes and calculated maximal dose of the rectum were examined.ResultsThe dosimeter measured with 85% accuracy at more than 5 cm from the source, but within a closer distance the accuracy decreased significantly. At 45–90° angle, the device measured with a 15% error. The error increased with the temperature, 22% at 35 °C. In 8 cases (26.7%) the maximal dose was measured in the correct diode. The device measured 73% of the calculated dose in the point of the diode. The maximum of the calculated doses of diodes was 60% of the calculated maximal dose. The in vivo measured dose was 35% of the calculated maximal dose.ConclusionsUnder treatment conditions, the semiconductor diode does not provide reliable measured data. The probe pushes the rectal wall closer to the high dose areas and underestimates the dose of it. Semiconductor probe is not recommended for in vivo dosimetry of the rectum in image-guided brachytherapy of cervical cancer.  相似文献

I analyze the historical background and philosophical considerations of Karl Bühler and his student Karl Popper regarding the crisis of psychology. They share certain Kantian questions and methods for reflection on the state of the art in psychology. Part 1 outlines Bühler's diagnosis and therapy for the crisis in psychology as he perceived it, leading to his famous theory of language. I also show how the Kantian features of Bühler's approach help to deal with objections to his crisis diagnosis and to aspects of his linguistic theory. Part 2 turns to Popper's dissertation, completed in 1928 under Bühler. I analyze Popper's disapproval of Schlick's physicalism in psychology, as well as Popper's attempt to extend Bühler's Kantian strategy to the domain of the psychology of thinking. In conclusion, I indicate how these approaches to the crisis in psychology differ from Thomas Kuhn's notions of crisis and revolution, which are still all too popular in current philosophical discussions of psychology.  相似文献

The association between latex RNA and latex production was examined using MAK column chromatography techniques. In young untapped trees the introduction of tapping or the treatment of bark with growth regulators resulted in an increase of RNA level and of rRNA/tRNA ratio in the latex. In regularly tapped trees an increase in rRNA but not in tRNA was brought about by increasing the tapping frequency. Treatment with growth regulators had the same effect but essentially only through the related enhancement of latex export from latex vessels. During latex flow, the highest RNA level was registered in latex fractions originating from the most heavily drained areas of bark. Using32P labeling, evidence was obtained that the export of latex results in an enhancement of rRNA migration into the inner latex containing space of the vessels. This is considered as the reason of the generally observed association of high RNA level and of high rRNA/tRNA ratio with high latex yield. It is proposed that in controlling the RNA level and RNA proportions in the latex an important role is played by changes in turgor pressure associated with the loss of latex which may influence the export of RNA from the nucleus through related induction of pressure disequilibrium between the nucleoplasm and the latex cytoplasm.  相似文献

We investigated the role of oyster gonadal TGFβ (og-TGFβ) in the reproduction of Crassostrea gigas, using an in vivo RNA interference approach. We designed double-stranded RNA targeting og-TGFβ, which is specifically expressed in the somatic cells surrounding germ cells in the gonad of both male and female oysters. In vivo injection of this og-TGFβ dsRNA into the gonad led to knock-down phenotypes for both sexes, with significant reduction (77.52% relative to controls) of the gonad area, lowered reproductive effort and germ cell under-proliferation. Interestingly, half of the injected females halted their vitellogenesis, since we were only able to observe pre-vitellogenic oocytes. In addition, apoptotic germ cells and haemocytes infiltrated into the gonad, likely as part of the active resorption of degenerating germ cells. Conversely, males showed a normal phenotype at the cellular level, with spermatids and spermatozoids observed in the gonads of control and injected males. As a result, og-TGFβ appears to play an essential role in C. gigas germ cell development by functioning as an activator of germ cell proliferation in both male and female oysters and vitellogenesis in females.  相似文献

A variety of semi-defined artificial diets were developed and tested for their ability to support the in vitro development of Edovum puttleri. In the most effective diet, 2.6% of E. puttleri pupated. This diet contained high levels of hen egg yolk combined with Manduca sexta larval hemolymph, or with a mixture of M. sexta egg homogenate and larval hemolymph. Egg homogenate alone (without the addition of hemolymph) was not capable of supporting the parasitoid's development. Thus, hemolymph appears to contain unidentified factor(s) important for inducing pupation of the wasp. Addition of M. sexta pupal fat body tissue extract (in place of hemolymph) also promoted pupation of E. puttleri. Gypsy moth (Lymantria dispar) larval hemolymph could not replace M. sexta larval hemolymph. Fractionation irreversibly reduced the growth-promoting effects of M. sexta larval hemolymph. However, the most effective fraction contained components whose molecular weights were 1000 kd. In diets that were devoid of insect materials, the best results were achieved when hen egg yolk, FreAmine, yeast extract, lactalbumin, trehalose, fetal bovine serum and bovine milk were included. This is the first report of an artificial diet for in vitro rearing an eulophid parasitoid from the egg through the pupal stage.  相似文献

Efficient and rapid tissue culture systems were developed for Piper longum, an important medicinal plant, through shoot tip multiplication and direct regeneration. Multiple shoots were induced from shoot tips cultured on agar-based Murashige and Skoog (MS) medium containing 4.44–22.19 M benzyladenine (BA) and 4.64–13.9 M kinetin (K). Maximum number of shoots were induced with 8.9 M BA and 4.64 M K. Adventitious shoot regeneration from leaf segments was achieved on MS containing 3.6–22.19 M BA along with 3.31–12.4 M picloram (P). Shoot differentiation occurred directly from the leaf bases without intermediale callus formation. Maximum shoot buds were obtained on MS medium with 17.76 M BA and 8.28 M P. Elongated shoots were separated and rooted in MS supplemented with 2.46 M indole butyric acid (IBA). Plantlets, thus developed were established in soil.  相似文献

The nature of the explant, seedling age, medium type, plant growth regulators, complex extracts (casein hydrolysate, coconut milk, malt extract and yeast extract) and antioxidants (activated charcoal, ascorbic acid, citric acid and polyvinylpyrrolidone) markedly influenced in vitro propagation of Gymnema sylvestre. A maximum number of shoots (57.2) were induced from 30 day old seedling axillary node explants on Murashige and Skoog (MS) medium containing 6-benzyladenine (1 mg l−1), kinetin (0.5 mg l−1), 1-napthalene acetic acid (0.1 mg l−1), malt extract (100 mg l−1) and citric acid (100 mg l−1). High frequency of rooting was obtained in axillary node explant derived shoots (50%) on half strength MS medium supplemented with IBA (3 mg l−1).
