Summary The two mesopsammal polychaetes, Hesionides arenaria and Trilobodrilus axi, show a relatively high resistance to cold temperatures in experiments. Great differences, however, between supercooled and frozen individuals exist. In chilling experiments T. axi is much less sensitive (no damage after 5 h at -12° C) than H. arenaria (50% mortality after 1 h at -7° C). The freezing resistance is essentially much less, but identical for the two species (100% mortality at -10° C after 1 h exposure). The differences in the supercooling resistance of the two species are in agreement with their geographic distribution and their seasonal migration behavior in the sandy beach locality.  相似文献

The fine structure of the gametes in six sea urchin species of the Sea of Japan was studied. The sperm in Strongylocentrotus nudus, S. intermedius, Echinocardium cordatum, Scaphechinus mirabilis, Sc. griseus and Echinarachnius parma are species-specific. The conical head and symmetrically disposed ring-shape mitochondrion are common to regular sea urchin sperm cells. S. nudus is characterized by the bulb-shaped head of the sperm; S. intermedius, by a bullet-shaped one. The sperm spearhead and small amount of post-acrosome material are common to irregular sea urchins; the sperm width: length ratio varies for different species, with the highest for Sc. mirabilis. The sperm of Sc. griseus is characterized by two lipid drops in the middle part of sperm. Asymmetrical mitochondrion disposal is usual for E. parma. Actin filaments are found in the postacrosome material in the sperm of heart-shaped sea urchins. The differences in the fine structure of sperm in cosmopolitan species Ech. cordatum inhabiting the Sea of Japan and coastal areas of the Northeast Atlantic may bear record to the complex existence of species Ech. cordatum. The fine structure of sperm is unique for each of the studied families, Strongylocentrotidae, Scutellidae, and Loveniidae. The eggs of all the species are characterized by vitelline and jelly-like membranes. The vitelline membrane is formed by cytoplasm protrusions; the area between them is filled with fibrillar material. The jelly-like membrane is formed by fibrillar material associated with apical parts of microvilli of the vitelline membrane. The irregular sea urchins Sc. griseus, Sc. mirabilis and E. parma are characterized by chromatophores situated in the jelly-like membrane, with the highest abundance in Sc. mirabilis.  相似文献

Female damselflies in the family Calopterygidae have two sperm storage organs: a spherical bursa copulatrix and a tubular spermatheca. Male flies have a peculiar aedeagus with a recurved head with which to remove bursal sperm, and lateral spiny processes to remove spermathecal sperm. The lateral processes differ among species and populations in terms of their width relative to the spermathecal duct: the narrower processes are physically able to access spermathecal sperm, while the wider ones are not. In the present study, sperm storage patterns and aedeagal structures were compared between two calopterygid species with different spermathecal structures –Calopteryx cornelia and Mnais pruinosa– with respect to not only sperm quantity (number) but also sperm quality (viability), by using a recently developed method based on live/dead dual fluorescence. Calopteryx cornelia is a typical spermathecal sperm remover. In this species, viability was similar between bursal and spermathecal sperm. In contrast, in M. pruinosa, the spermatheca was much smaller than the bursa and often contained no sperm. Even when the spermatheca of this species did contain sperm, a high percentage of it was dead. Although the spermatheca of M. pruinosa has such atrophic tendencies, males have nevertheless developed long and spiny lateral processes similar to those of C. cornelia, suggesting the processes have functions other than spermathecal sperm removal. They possibly function as stoppers or guides for manipulating the aedeagal head to remove the sperm mass from the bursa.  相似文献

The evolution of reproductive isolation is a prerequisite in the formation of new species. Although there are numerous studies on ejaculates in lepidopteran insects, ejaculate comparisons among sibling species have not been adequately addressed to understand possible reproductive barriers to hybridization. Here, we examined the interspecific and intraspecific variations of ejaculates in the sibling noctuid moths Helicoverpa armigera and Helicoverpa assulta. We found that there were considerable variations in the number of apyrene and eupyrene sperm and the length of eupyrene sperm. Male pupal mass explained not only a significant proportion of the variation in apyrene sperm number in both H. armigera and H. assulta, but also a significant proportion of the variation in eupyrene sperm number in H. assulta. There was a significant positive relationship between the number of eupyrene sperm and the number of apyrene sperm in both species. No difference in the length of eupyrene sperm was found between them; however, ejaculates of H. armigera had many more eupyrene sperm than H. assulta had. In H. armigera, large males generally mated with large females. The evolutionary consequences of these differences are discussed in this paper.  相似文献

Summary The mechanisms by which sperm cells recognize and fuse with the egg and central cell during double fertilization in flowering plants are unknown. To identify membrane surface molecules that might function in fertilization, we immunized mice with isolated sperm ofBrassica campestris and screened the polyclonal sera and monoclonal hybridoma supernatants by immunocytochemistry for binding to isolated sperm cells. We identified three cell lines producing hybridoma supernatants which bind to sperm cell surfaces inB. campestris and further analyzed the properties of one of these, BRSP1. The molecular mass of the epitope to BRSP1 was 54 kDa, and was not glycosylated. Although the antibodies were immunoglobulin M, neither removal of carbohydrates nor competition with antibodies which recognize arabinogalactan decreased binding. BRSP1 recognized sperm ofPlumbago zeylanica, Nicotiana tabacum. Arabidopsis thaliana, andEruca vesicaria and generative cells ofLilium longiflorum and ofNarcissus tazetta but did not recognize sperm ofHelianthus annuus, Gerbera jamesonii, orZea mays. Antibodies to plant sperm allow us to probe sperm membranes for functional components.  相似文献

The acrosome of Macaca fascicularis sperm cannot be distinguished by conventional light microscopy, so determining whether sperm are acrosome-intact or-reacted is difficult. We describe methods for labeling the acrosomal region of sperm with two different probes: fluoresceinated Pisum sativum agglutinin and anti-sperm antiserum. Acrosome-intact sperm are much more heavily labeled in the acrosomal region than are acrosome-reacted sperm, providing a simple means of differentiating the two types of sperm. The two probes detect similar numbers of acrosome-reacted sperm following treatment with the divalent cation ionophore, A23187.  相似文献

Males of many species concurrently produce more than one sperm type, now called sperm heteromorphism. In the Drosophila obscura group, all species examined to date produce multiple sperm types that differ in sperm length. Short sperm types in at least three obscura group species do not participate in fertilization, leading to questions regarding the adaptive value of sperm heteromorphism. The common and pervasive inheritance of this trait in the obscura group, however, may indicate that sperm heteromorphism is phylogenetically constrained and therefore does not reflect an adaptive response to selection pressures. I measured interspecific variation in sperm length and determined the number of sperm types produced in 10 obscura group species. I subsequently tested if interspecific variation in sperm length is significantly associated with phylogeny by using an autoregressive comparative method. All obscura group species examined produce two visually distinct sperm lengths, short and long. Phylogenetic autoregression analyses indicated that 22% of the interspecific variation in long sperm is related to phylogeny, whereas short sperm are not significantly correlated with phylogeny. These results suggest different selection pressures on the two sperm length types; long sperm have evolved in response to fertilization demands and short sperm have been decoupled from these requirements.  相似文献

Cytochrome oxidase was isolated from the midpiece of boar sperm by extraction and fractionation with ammonium sulfate in the presence of cholate. The enzyme was further purified to apparent homogeneity by DEAE-cellulose column chromatography in the presence of minimal amounts of Triton X-100. The purified enzyme exhibited similar oxidized and reduced optical spectra to those of the bovine heart and rat liver cytochrome oxidases. However, the sperm oxidase was found to contain stoichiometrically more subunits I, II, and III than the other, smaller subunits. The sperm oxidase also required phospholipids for its activity, but it was less sensitive to inhibition by KCN. Interestingly, the sperm oxidase was much more acid-stable than the bovine heart and rat liver counterparts. The optimumpH for the sperm oxidase catalyzing the electron transfer between ferrocytochromec and cytochromea was aroundpH 4.8, and those for the bovine heart and rat liver were 6.2 and 6.8, respectively. AtpH 4.5, the sperm oxidase still maintained about 70% enzyme activity, whereas less than 20% activity remained in the heart and liver oxidases. The peculiar properties of sperm cytochrome oxidase may be due to the fact that the well-packed chromosomes in the sperm headpiece do not function, such that the nuclear gene-coded subunits are deficient in the sperm cytochrome oxidase. The finding that the sperm oxidase was more acid-stable is an example of structure-function coordination, and is discussed from the viewpoint of chemiosmotic theory and the unique structure and functions of the sperm mitochondria.  相似文献

Septin-based ring complexes maintain the sperm annulus. Defective annular structures are observed in the sperm of Sept12- and Sept4-null mice. In addition, sperm capacitation, a process required for proper fertilization, is inhibited in Sept4-null mice, implying that the sperm annulus might play a role in controlling sperm capacitation. Hence, we analyzed sperm capacitation of sperm obtained from SEPT12 Ser196 phosphomimetic (S196E), phosphorylation-deficient (S196A), and SEPT4-depleted mutant mice. Capacitation was reduced in the sperm of both the Sept12 S196E- and Sept12 S196A-knock-in mice. The protein levels of septins, namely, SEPT4 and SEPT12, were upregulated, and these proteins were concentrated in the sperm annulus during capacitation. Importantly, the expression of soluble adenylyl cyclase (sAC), a key enzyme that initiates capacitation, was upregulated, and sAC was recruited to the sperm annulus following capacitation stimulation. We further found that SEPT12, SEPT4, and sAC formed a complex and colocalized to the sperm annulus. Additionally, sAC expression was reduced and disappeared in the annulus of the SEPT12 S196E- and S196A-mutant mouse sperm. In the sperm of the SEPT4-knockout mice, sAC did not localize to the annulus. Thus, our data demonstrate that SEPT12 phosphorylation status and SEPT4 activity jointly regulate sAC protein levels and annular localization to induce sperm capacitation.  相似文献

In externally fertilizing species, the gametes of both males and females are exposed to the influences of the environment into which they are released. Sperm are sensitive to abiotic factors such as salinity, but they are also affected by biotic factors such as sperm competition. In this study, the authors compared the performance of sperm of three goby species, the painted goby, Pomatoschistus pictus, the two-spotted goby, Pomatoschistus flavescens, and the sand goby, Pomatoschistus minutus. These species differ in their distributions, with painted goby having the narrowest salinity range and sand goby the widest. Moreover, data from paternity show that the two-spotted goby experiences the least sperm competition, whereas in the sand goby sperm competition is ubiquitous. The authors took sperm samples from dissected males and exposed them to high salinity water (31 PSU) representing the North Sea and low salinity water (6 PSU) representing the brackish Baltic Sea Proper. They then used computer-assisted sperm analysis to measure the proportion of motile sperm and sperm swimming speed 10 min and 20 h after sperm activation. The authors found that sperm performance depended on salinity, but there seemed to be no relationship to the species' geographical distribution in relation to salinity range. The species differed in the proportion of motile sperm, but there was no significant decrease in sperm motility during 20 h. The sand goby was the only species with motile sperm after 72 h.  相似文献

Sperm apical hooks in murine rodents play an important role in sperm competition. Apical hooks are more curved and longer in species with relatively larger testes, that is in species with a higher risk of sperm competition. The sperm can form aggregations, ‘trains’, that can move faster than individual sperm, thus reaching the egg earlier as was observed in Apodemus sylvaticus. The apical hook plays an important role for train formation. This study focuses on the changes in the curvature of sperm apical hooks during the final stages of spermiogenesis and stages before fertilization (sperm-life span). Apical hook curvatures of field mice (A. agrarius and A. sylvaticus) vary significantly between dormant and active sperm. In contrast, there are no significant differences among the stages in the eastern house mouse. Since there are high ranges of angle values in all stages, the mean angles of apical hook curvature are not appropriate for evaluating risk of sperm competiton. The ranges of angle values point to a level of flexibility of the apical hooks. The lengths of sperm hooks in individual species do not change during particular stages. The length and flexibility of the sperm apical hooks are important for the formation of sperm aggregations, thus these sperm characters indicate the risk of sperm competition and the sperm strategies in murine rodents.  相似文献

The newt, Cynops pyrrhogaster, exhibits physiological polyspermic fertilization, in which several sperm enter an egg before egg activation. An intracellular Ca2+ increase occurs as a Ca2+ wave at each sperm entry site in the polyspermic egg. Some Ca2+ waves are preceded by a transient spike-like Ca2+ increase, probably caused by a tryptic protease in the sperm acrosome at the contact of sperm on the egg surface. The following Ca2+ wave was induced by a sperm factor derived from sperm cytoplasm after sperm–egg membrane fusion. The Ca2+ increase in the isolated, cell-free cytoplasm indicates that the endoplasmic reticulum is the major Ca2+ store for the Ca2+ wave. We previously demonstrated that citrate synthase in the sperm cytoplasm is a major sperm factor for egg activation in newt fertilization. In the present study, we found that the activation by the sperm factor as well as by fertilizing sperm was prevented by an inhibitor of citrate synthase, palmitoyl CoA, and that an injection of acetyl-CoA or oxaloacetate caused egg activation, indicating that the citrate synthase activity is necessary for egg activation at fertilization. In the frog, Xenopus laevis, which exhibits monospermic fertilization, we were unable to activate the eggs with either the homologous sperm extract or the Cynops sperm extract, indicating that Xenopus sperm lack the sperm factor for egg activation and that their eggs are insensitive to the newt sperm factor. The mechanism of egg activation in the monospermy of frog eggs is quite different from that in the physiological polyspermy of newt eggs.  相似文献

Synopsis Sturgeon gametes differ from those of most fish in that the sperm possess acrosomes that undergo exocytosis and filament formation while the eggs possess numerous micropyles. Acipenser transmontanus eggs are encased by multilayered envelopes that consist of outer adhesive jelly coats and three structured layers interior to the jelly. The glycoprotein jelly layer only becomes adhesive upon exposure to freshwater. The layer interior to the jelly, layer 3, is the other carbohydrate-containing component of the egg envelope. This layer consists of a water-insoluble glycoprotein that, upon freshwater exposure, is hydrolyzed by a trypsinlike protease to yield a water-soluble, lower molecular weight carbohydrate-containing component. This component can be identified in the surrounding medium when unfertilized eggs are incubated in freshwater. This egg water component elicits acrosome reactions only in homologous sperm. The A. transmontanus sperm acrosome reaction is a Ca++ and/or Mg++ dependent event that includes the formation of a 10 μ long fertilization filament. A. transmontanus fertilization can occur at low sperm per egg ratios; however, crossfertilization of A. transmontanus eggs with lake sturgeon, A. fluvescens, sperm results in a very low number of fertilized eggs, even at high sperm per egg ratios. The morphological, physiological, and biochemical phenomenon reviewed in this paper are related to the environment in which they occur. Also, the possible role of the acrosome and the presence of numerous micropyles are discussed.  相似文献

The sperm head morphology and tail length of two species of Australian rock rats, Zyzomys argurus and Zyzomys pedunculatus, are presented. In Z. argurus the sperm head has an apical hook together with two ventral processes extending from the upper concave surface that are largely composed of cytoskeletal material, and the sperm tail is about 135 µm in length. By contrast, in Z. pedunculatus the sperm head is paddle‐shaped with the nucleus capped by an acrosome that has a large apical segment and is surrounded by a thin layer of cytoskeletal material, and the sperm tail is only around 85 µm in length. Since the structure of the spermatozoon of Z. argurus is similar to that of most of the old endemic Australian rodents it is presumed to be the ancestral condition within the Zyzomys genus with that of Z. pedunculatus being highly derived and showing convergence with the sperm structure in some other orders of mammals.  相似文献

Crosses between Drosophila mojavensis and D. arizonensis produce fertile females, but the males from the cross ♂ D. mojavensis × ♀ D. arizonensis are sterile. The chromosomal basis of sperm immotility was studied in these hybrids. Interspecific crossing-over was avoided by crossing hybrid males to pure-species females, and chromosomal identification in backcross progeny was possible by means of electrophoretic markers. The main findings are as follows. The Y-chromosome and two autosomes are involved in the determination of sperm motility. The other autosomes, with the exception of the sixth which was not tested, appear to have no effect. The effect of the D. arizonensis X-chromosome was not examined, but it is established that the D. mojavensis X-chromosome has no effect on sperm motility in males carrying the D. arizonensis Y-chromosome and any combination of autosomes. The Y-chromosome and the two autosomes interact with each other in a simple and predictable way, so that certain combinations of these chromosomes always produce motile sperm and others immotile sperm. Thus, asymmetrical male hybrid sterility may have a simple genetic basis. In contrast to ethological isolation, the genetic basis for this postmating isolating mechanism does not appear to vary among conspecific populations, an observation which suggests that postmating isolation antedates ethological isolation in these species.  相似文献

A recently completed study of sperm transfer and spermathecal filling in Culicoides melleus (Ceratopogonidae) provided evidence that the spermathecae create an incoming current by fluid absorption. This current, rather than sperm motility, was thought to accomplish transfer of spermatozoa from spermatophore to spermathecae. This review discusses the approach used to assess the role of sperm motility, as applied critically to available observations in other lower Diptera. Aedes aegypti (Culicidae), Simulium decorum (Simuliidae) and Plecia nearctica (Bibionidae) are considered. The results suggest that sperm motility very probably does not contribute to spermathecal filling in these species, and that fluid absorbtion is the more likely mechanism. Certain physical properties of the female reproductive systems are discussed in the light of this conclusion.  相似文献

Dasyurids are a diverse group of Australian native carnivores and insectivores that contains several threatened species. Despite successful cryopreservation of sperm from several marsupials, only 3% postthaw motility is reported for dasyurid marsupials. This study examined sperm preservation in the fat-tailed dunnart (Sminthopsis crassicaudata), an experimental model, with supplementary observations on the eastern quoll (Dasyurus viverrinus) and northern quoll (Dasyurus hallucatus). In S. crassicaudata, a toxicity trial demonstrated that incubation with up to 40% glycerol did not reduce sperm viability, suggesting that glycerol is not toxic to dasyurids. On the basis of this finding, S. crassicaudata, D. viverrinus, and D. hallucatus sperm were extended to a final concentration of 20% or 40% glycerol in Tris-citrate fructose and frozen in liquid nitrogen vapor. Postthaw sperm from all three species were nonmotile, and vital staining (SYBR14 and propidium iodide) indicated that sperm were nonviable. However, there was no evidence suggesting disruption of normal gross morphology or loss of acrosomal integrity when assessed by Bryan's staining. After freeze drying, Bryan's staining indicated that approximately 80% of S. crassicaudata sperm had normal acrosomes and no head loss. Despite being nonviable, terminal deoxynucleotidyl transferase–mediated dUTP nick-end labeling showed that S. crassicaudata sperm frozen in 40% glycerol or freeze-dried had no DNA damage compared with fresh controls. This study has described a method for preservation of the dasyurid sperm nuclei, but continued studies are required to achieve viable motile sperm and establish tools for the long-term storage of dasyurid sperm.  相似文献

The mature sperm of A. perniciosus are organized into bundles, about 350 μm long by 9–10 μm wide. Each bundle contains 32 sperm enclosed by a common sheath. The sperm contains an elongated ‘central core’, representing nuclear material, surrounded by a spiral microtubular sheath and cytoplasm. The electron-dense nuclear material is localized in the more pointed half of the sperm. The spiral microtubular sheath is composed of 30— 100 microtubules (depending on the cross-sectional level), situated parallel to the longitudinal axis of the sperm. On the basis of this ultrastructural organization, the motility of the sperm and sperm bundle as a whole is discussed. The sperm of A. perniciosus provide strong evidence that the microtubules arranged asymmetrically represent the elements directly involved in sperm motility.  相似文献

The sperm cell morphology and spermatogenesis of Halacaroides antoniazziae Pepato Tiago and da Rocha 2011 and Acaromantis vespucioi Pepato and Tiago 2004 was investigated. Halacaroides sperm cells have a complete acrosomal complex, dense tubules crossing the cytoplasm and modified mitochondria. Mature sperm cells are surrounded by two kinds of secretions. Inside the ejaculatory duct, they lie upon a centre composed of a secretion structured as heaps of elongated bodies. Acaromantis spermatozoa are spindle shaped and lack an acrosomal complex. The plasmalemma is deeply folded; the cytoplasm is very reduced and devoid of organelles. A single kind of globular secretion was found. The sperm mass is surrounded by two layers of amorphous secretions. These species share a peripheral pattern of nuclear condensation during spermatogenesis, a possible apomorphy for most halacarids, and no special adaptation to the interstitial environment could be related to their sperm cell morphology.  相似文献
