To investigate the evolutionary cost of an immune response, we selected six lines of the mosquito Aedes aegypti for earlier or later pupation and measured the extent to which this selection procedure changed the mosquito's ability to encapsulate and melanize a negatively charged Sephadex bead. After 10 generations of selection, the age at pupation in the two selection regimes differed by about 0.7 days, accompanied by an increase of wing length of the mosquitoes selected for late pupation. Among the mosquitoes that had been selected for early pupation, only 6% had strongly or completely melanized the bead, while among the individuals that had been selected for late pupation, 32% had melanized the bead. Thus, our results suggest a genetic correlation between age at pupation and immunocompetence. As a consequence, mosquitoes that respond to increased intense parasite pressure with more effective immunity are predicted to pay for the increased defense with slower development.  相似文献

The costs and benefits of activating the immune system can reach across generations. Thus, in vertebrates and in several invertebrates, stimulating the immune system of a female can enhance immunity of her offspring or decrease offspring fitness. We evaluated the potential maternally transmitted costs and benefits of the melanization response, an innate immune response of insects that helps to protect mosquitoes from malaria parasites. We manipulated the maternal melanization response of the yellow fever mosquito Aedes aegypti by inoculating female mosquitoes with negatively charged sephadex beads or with immunologically inert glass beads; a control group was not inoculated. In the next generation, we assayed the melanization response and measured three other life-history traits: survival up to emergence, the age at emergence, and body size (estimated as wing length). We found no evidence of fitness costs or benefits for the offspring. A retrospective power analysis found that our experiment would have detected an effect size that is three times smaller than the maternal immune priming effects that have been reported in the literature. We did find a strong correlation between offspring wing length and melanization response. Overall, our findings indicate that trans-generational immune priming in invertebrates cannot be generalized, and that it may depend on the species, the immune challenge, and the environmental conditions.  相似文献

The melanization response against intrathoracically inoculated Brugia pahangi and Dirofilaria immitis microfilariae (mff) isolated from vertebrate host blood was evaluated in both uninfected Aedes aegypti black-eyed Liverpool strain and in mosquitoes harboring a developing B. pahangi infection. The immune response against inoculated mff of either species was significantly reduced by 28-47% in infected as compared with uninfected mosquitoes. Attempts to passively transfer this suppression factor(s) by inoculating naive mosquitoes with 0.1-0.2 microliter of hemolymph from B. pahangi-infected mosquitoes produced equivocal results. The role this parasite-induced immune suppression might play in aiding parasite survival in compatible vectors is discussed.  相似文献

Phospholipase A2 (PLA2) hydrolyzes fatty acids from phospholipids at the sn‐2 position. Two intracellular PLA2s, iPLA2A and iPLA2B, have been found in Spodoptera exigua. Both are calcium‐independent cellular PLA2. Their orthologs have been found in other insects. These two iPLA2s are different in ankyrin motif of N terminal region. The objective of this study was to determine whether Toll/immune deficiency (IMD) signal pathways could mediate cellular immune responses via induction of iPLA2 expression. Both iPLA 2s were expressed in all developmental stages of S. exigua, showing the highest expression in the adult stage. During larval stage, hemocyte is the main tissue showing expression of these iPLA2s. Both iPLA2s exhibited similar expression patterns after immune challenge with different microbial pathogens such as virus, bacteria, and fungi. Promoter component analysis of orthologs encoded in S. frugiperda indicated nuclear factor‐κB‐ and Relish‐responsible elements on their promoters, suggesting their expression in S. exigua under Toll/IMD immune signaling pathways. RNA interference (RNAi) of MyD88 or Pelle under Toll pathway suppressed inducible expression levels of both iPLA2s in response to Gram‐positive bacteria containing Lys‐type peptidoglycan or fungal infection. In contrast, RNAi against Relish under IMD pathway suppressed both iPLA2s in response to infection with Gram‐negative bacteria. Under RNAi conditions, hemocytes significantly lost cellular immune response measured by nodule formation. However, addition of arachidonic acid (a catalytic product of PLA2) rescued such immunosuppression. These results suggest that Toll/IMD signal pathways can mediate cellular immune responses via eicosanoid signaling by inducing iPLA2 expression.  相似文献

The effect of gamma irradiation on the melanotic encapsulation response of Aedes aegypti black eye Liverpool strain against inoculated Dirofilaria immitis microfilariae (mff) was assessed at 1, 2, 3, and 6 days postinoculation (PI). Mosquitoes received 6000 rad from a 137Cs source (Shepard Mark I irradiator) at 3 days postemergence and were inoculated with 15-20 mff 24 hr later. These mosquitoes were compared to nonirradiated controls that also were inoculated with 15-20 mff at 3 days postemergence. The immune response was significantly reduced in irradiated mosquitoes as compared with controls at all days PI. Although the response was significantly inhibited compared with controls, irradiated mosquitoes were still capable of eliciting a response against 69% of recovered mff at 6 days PI. External gamma irradiation did not significantly affect the proliferation of hemocytes associated with the melanotic encapsulation response of A. aegypti. The number of circulating hemocytes increased in irradiated mosquitoes in response to inoculated mff in a manner similar to nonirradiated, inoculated controls. Hemocyte monophenol oxidase activity, however, was significantly reduced in gamma-irradiated mosquitoes at 12 hr PI as compared with controls. The reduced immunological capacity of irradiated mosquitoes might be related to an interference with gene activity required for the synthesis or activation of enzymes that are directly or indirectly involved in the biochemical processes associated with the production of melanotic substances that sequester mff.  相似文献

Salivary gland homogenates from adult Aedes aegypti lyse Micrococcus lysodeikticus cells. The bacteriolytic factor is present in a cell type common to both male and female mosquitoes, as well as in the crop of sugar-feeding mosquitoes. A spontaneous recessive sex-linked mutant short wing has been discovered in the mosquito Aedes aegypti. It is situated less than one cross-over unit from the sex determining locus. In homozygous females, flight is impaired and the survival and fecundity is markedly subnormal. Two possible uses of this gene for genetic control operations are envisaged: (a) to provide automatic sexing of males for release and (b) enhancement of the population control potential of other available genetics systems.  相似文献

In this study, we designed a new tent trap; the BioDiVector (BDV) tent trap, consisting of two rectangular tents that use human bait without endangering the technical personnel. The daily activity pattern of Aedes aegypti and Aedes albopictus in intra, peri, and extradomiciliary sites was studied in an endemic area of dengue in southern Mexico by using the BDV tent trap. Totals of 3,128 individuals of Ae. aegypti and 833 Ae. albopictus were captured. More Ae. aegypti males than females were caught, while the opposite was true with Ae. albopictus. The activity of both mosquito species was affected by the interaction between the collection site and time of day. In general, more individuals of both mosquito species were captured at the extradomicillary sites than at the peri and intradomicillary sites. Mosquitoes showed two peaks of activity, one in the morning and the other in the afternoon, but in general this only occurred at the extradomicillary sites, whereas no peak of activity was observed at the intra and peridomicillary sites. Overall, Ae. aegypti had a higher indirect biting rate than Ae. albopictus. Finally, due to its efficiency, simplicity, and low cost, we suggest the use of this innovative tool for entomological surveillance, bionomics and vector incrimination studies in geographical areas where dengue and other arboviruses are present.  相似文献

Ultrastructural and lectin-binding studies have established that the melanotic encapsulation reaction of Aedes aegypti Liverpool strain against inoculated Dirofilaria immitis microfilariae (mff) is a hemocyte-mediated reaction. Total hemocyte counts from mff-inoculated (= immune-activated), saline-inoculated, and uninoculated female A. aegypti were determined using a hemocoel perfusion technique. Total hemocyte populations in uninoculated mosquitoes were significantly larger in younger mosquitoes, but no significant change was noted as mosquitoes aged beyond 14 days. Hemocyte populations in immune-activated mosquitoes increased from 1 to 3 days postinoculation (PI) and decreased on days 4 and 5 PI. Hemocyte populations at 1 to 4 days PI were significantly elevated in mff-inoculated A. aegypti as compared with saline-inoculated controls. Saline-inoculated mosquitoes displayed little change in total hemocyte numbers from 1 to 5 days PI, and their hemocyte populations were similar to those seen in uninoculated insects of the same age. Experiments involving the inoculation of [3H]thymidine along with mff or saline alone and studies involving the administration of colchicine suggest that increased hemocyte populations in immune-activated A. aegypti are a result of mitotic division of circulating blood cells.  相似文献

Hemocytes from adult, female Aedes aegypti, intrathoracically inoculated with microfilariae (mf) of the nematode Dirofilaria immitis, were compared to saline-inoculated and uninoculated controls using sodium dodecyl sulfate-polyacrylamide gel electrophoresis (SDS-PAGE), 125I-labeling, and wheat germ agglutinin (WGA) binding techniques. Activation of wound healing and/or melanotic encapsulation responses by the inoculation of saline or mf into the host hemocoel induced alterations in the hemocyte activity of these mosquitoes. Protein assays of whole hemocyte lysates revealed that hemocytes from saline- and mf-inoculated mosquitoes had higher protein concentrations than uninoculated controls. Many polypeptides were seen within all three hemocytes preparations when stained with silver nitrate, but there was an overall increase in protein synthesis in hemocytes from inoculated mosquitoes. In addition, a 200-kDa polypeptide was uniquely expressed in hemocytes from inoculated mosquitoes. There were several prominent surface proteins labeled with 125I, and several of these increased dramatically in intensity during wound healing and/or a melanotic encapsulation response. Similar results were seen in two-dimensional separations. A set of basic polypeptides comigrated with an acidic polypeptide resulting in a surface protein of approximately 80-90 kDa that increased in inoculated mosquitoes. Hemocytes from inoculated mosquitoes exhibited a group of three acidic polypeptides, whereas hemocytes from uninoculated mosquitoes exhibited only one of these protein fragments. Three surface polypeptides bound 125I-labeled WGA, and binding of WGA to hemocyte surface polypeptides was successfully inhibited by the incubation of cells with the lectin and its competing sugar.  相似文献

The neurotrophic cytokines ciliary neurotrophic factor and leukemia inhibitory factor (LIF) play a key role in neuronal and oligodendrocyte survival and as protective factors in neuroinflammation. To further elucidate the potential of endogenous LIF in modulating neuroinflammation, we studied myelin oligodendrocyte glycoprotein-induced experimental autoimmune encephalomyelitis in LIF knockout mice (LIF(-/-) mice). In the late phase of active myelin oligodendrocyte glycoprotein-induced experimental autoimmune encephalomyelitis, LIF(-/-) mice exhibited a markedly milder disease course. The inflammatory infiltrate in LIF(-/-) mice was characterized by an increase in neutrophilic granulocytes early and fewer infiltrating macrophages associated with less demyelination later in the disease. In good correlation with an effect of endogenous LIF on the immune response, we found an Ag-specific T cell-priming defect with impaired IFN-gamma production in LIF(-/-) mice. On the molecular level, the altered recruitment of inflammatory cells is associated with distinct patterns of chemokine production in LIF(-/-) mice with an increase of CXCL1 early and a decrease of CCL2, CCL3, and CXCL10 later in the disease. These data reveal that endogenous LIF is an immunologically active molecule in neuroinflammation. This establishes a link between LIF and the immune system which was not observed in the ciliary neurotrophic factor knockout mouse.  相似文献

ABSTRACT. The ATP analogues adenylylimidodiphosphate and adenylylmethylenediphosphate are 3–5-fold more effective than ATP as gorging stimulants for Aedes aegypti. This increased potency is not due to the fact that the two analogues are not hydrolysed by the mosquito salivary apyrase, but most likely to their greater affinity to the mosquito gustatory receptor protein. The analogues 2'd ATP and 3'd ATP are about half as potent as ATP, while 2',3'-dideoxyadenosine triphosphate is 10-fold more potent than ATP in evoking the gorging response. It is proposed that removal of both hydroxyl groups eliminates binding of the stimulant at the ribose moiety, thus allowing the molecule greater freedom to rotate and bind more effectively to its two other binding sites at the amino group on the purine and at the terminal phosphate. Our data demonstrate that ATP activates the gorging response of Ae.aegypti merely by binding to its receptor protein and is not required as an exogenous source of energy. Gorging response to ATP is competitively inhibited by novobiocin.  相似文献

Mosquitoes (Diptera: Culicidae) are major vectors of numerous infectious agents. Compounds in mosquito saliva not only facilitate blood-feeding, but may also have an impact upon the immune system of vertebrate hosts. Consequently, the exposure to mosquito saliva may influence pathogen transmission, establishment and disease development. Using two medically important vector mosquitoes, Aedes aegypti (L.) and Culex quinquefasciatus Say, we examined the effects of mosquito saliva on immune cells of host mice. After antigen-specific or non-specific stimulation, murine splenocyte proliferation and production of both Th1 and Th2 cytokines were significantly reduced in the presence of salivary gland extract (SGE) from Ae. aegypti, but not SGE from Cx. quinquefasciatus. T cell populations were highly susceptible to this suppression, showing increased mortality and reduced division rates - judged by flow cytometric analyses. Evidently these two culicine mosquitoes differ in their host immunomodulatory activities.  相似文献

A new system for deployment of fungus‐impregnated black cloths was tested against Aedes aegypti. A “PET trap” was placed in a test chamber to evaluate attractiveness to female A. aegypti with black cloths covered in adhesive film or adhesive film only for 24 and 48 hr. Traps with fungus (Metarhizium anisopliae and Beauveria bassiana)‐impregnated black cloths were tested against female mosquitoes for different time periods (3 h to 48 hr) in the chambers. Traps were then tested under intradomicile conditions against sucrose and blood‐fed insects. Experiments were carried out to ascertain the minimum number of PET traps need to be deployed per test room and to test the effect of different periods of exposure to traps. Exposing the insects for 24 and 48 hr to a PET trap with adhesive film + black cloth resulted in higher rates of trapped mosquitoes (38.6% and 68%, respectively) when compared with adhesive film only (6% and 12.6%, respectively). Both fungal species were effective at reducing survival rates when mosquitoes were exposed to traps for 24 hr or 48 hr. Lower exposure times did not significantly alter survival rates when compared to controls. The results showed that five traps or three traps per room were equally effective in reducing mosquito survival rates when testing both fungal species. The results for sucrose‐fed insects showed significant reductions in survival when exposed to M. ansiopliae or B. bassiana for 24, 48 or 120 hr when compared to control survival, with the lowest survival rates seen following 48‐ or 120‐hr exposures. Survival of blood‐fed mosquitoes exposed to fungus‐impregnated traps for 48 hr was not significantly different to the controls; however, longer exposure times significantly reduced survival rates. PET traps could be an effective system for deploying fungus‐impregnated cloths in residences, facilitating cooperation of volunteers and reducing distribution time.  相似文献

A novel IFN-like molecule, limitin, was recently identified and revealed to suppress B lymphopoiesis through the IFN-alphabeta receptor, although it lacked growth suppression on myeloid and erythroid progenitors. Here we have studied diverse effects of limitin on T lymphocytes and compared limitin with previously known IFNs. Like IFN-alpha and -beta, limitin modified immunity in the following responses. It suppressed mitogen- and Ag-induced T cell proliferation through inhibiting the responsiveness to exogenous IL-2 rather than suppressing the production of IL-2. In contrast, limitin enhanced cytotoxic T lymphocyte activity associated with the perforin-granzyme pathway. To evaluate the effect of limitin in vivo, a lethal graft-versus-host disease assay was established. Limitin-treatment of host mice resulted in the enhancement of graft-versus-host disease. Limitin did not influence thymocyte development either in fetal thymus organ cultures or in newborn mice injected with limitin-Ig, suggesting that limitin is distinguishable from IFN-alpha and -beta. From these findings, it can be speculated that the human homolog of limitin may be applicable for clinical usage because of its IFN-like activities with low adverse effects on, for example, T lymphopoiesis, erythropoiesis, and myelopoiesis.  相似文献
