The validity of a chemiosmotic hypothesis for uptake of weak acids as an explanation for the accumulation of auxin by cells has been explored further by comparing the uptake of indole-3-acetic acid (IAA) by 1-mm segments of corn (Zea mays L.) coleoptiles with that of benzoic acid and two neutral indoles, indoleethanol and indoleacetonitrile, which do not ionize. These substances, while structurally related to IAA lack both auxin activity and polar transport. Uptake of IAA and benzoic acid increase with decreasing external pH, whereas the uptake of the two neutral indoles is independent of external pH.Although metabolism of IAA, during 90 min or less, is minimal and without significant effect on its uptake, metabolism of benzoic acid appears responsible for the apparent saturation of benzoic acid uptake at high concentrations. An inhibitor of auxin transport, N-1-naphthylphathalamic acid (NPA), stimulates uptake of IAA but has no effect on uptake of either benzoic acid or the two neutral indoles. Thus, NPA does not affect the driving forces for accumulation of weak acids but probably specifically decreases the flux of the auxin anions relative to undissociated auxin. Since the electrochemical potential of auxin anions is usually higher in than outside cells, blocking the anion flux with NPA would enhance auxin uptake. Azide, which abolishes accumulation of both IAA and benzoic acid, may simply collapse the pH gradient across the plasma membrane.In the absence of NPA, increasing concentrations of auxins or the analogoue -naphthaleneacetic acid (-NAA) exert two opposing effects on the uptake of IAA-depression and stimulation. Stimulation results from saturating the anion flux. With uptake fully stimulated by NPA, however, increasing concentrations of auxins or analogues only depress uptake of [3H]IAA. These results are consistent with more than one path for auxin transport each with a different dependence on concentration. In depressing NPA-stimulated IAA uptake, the effectiveness of -NAAIAA-NAA benzoic acid, a specificity similar to that of an auxin binding site in vitro that has been implicated by others in auxin transport. The induction of synthesis of the secreted enzymes endo-1,4--xylanase (EC 3.2.1.8) and -galactosidase (EC 3.2.1.23) in original and recombinant Penicillium canescens strains has been studied. In all producer strains, the synthesis of these enzymes was induced by arabinose and its metabolite arabitol. The two enzymes differed in the concentration of arabinose required for induction: the synthesis of -galactosidase was most pronounced at 1 mM, whereas maximum synthesis of endo-1,4--xylanase was observed at 5–10 mM. An increase in the number of endo-1,4--xylanase copies in the high-copy-number strain of the fungus suppressed the synthesis of -galactosidase; the synthesis of endo-1,4--xylanase in the high-copy-number recombinant producing -galactosidase was affected to a lesser extent. The amount of enzymes synthesized did not depend on the saccharide used as the sole source of carbon for growing the mycelium prior to its transfer to the inducer-containing medium.  相似文献

Dazu noch einige seltene Besonderheiten.

Summary 1965 a dermatoglyphic investigation of 10 individuals (7 and 3 ) with Thyreoiditis lymphomatosa Hashimoto has been published in this Journal. The now increased series of 21 women confirms the results gained formerly: on the finger tips abundance of whorls and increased quantitative value; on the palm increased number of patterns on the hypothenar, lack of patterns with accessory triradius on the interdigita, tendence of line A to follow a longitudinal course. In addition there are some rare peculiarities.

Summary A histochemical and ultrastructural study was carried out on subcommissura organs from 42 human embryos and fetuses in order to characterize some large granulesTypical granules make their appearance in the rostral hypendymal region of the subcommissural organ (SCO) in fetuses of about 50 mm CRL. Although they appear in other SCO-regions later, the highest number of granules is always located towards the pineal gland.Typical granules are of spherical shape with a diameter of about 2 microns. The various histochemical reactions reveal a reactivity which differentiates the shell of the granules from the granule interior. Nucleoproteins are present in the shell together with phospholipids and/or lipoproteins. The interior of the granules can contain different materials such as glycogen or lipid or neurosecretory substance. Ultrastructural observations show that a granule consists of whorls of endoplasmic reticulum sparsely studded with ribosomes surrounding an interior containing either lipid or lipoprotein inclusions, large amounts of glycogen or simply cytoplasm.It is suggested that the concentric lamellar organelle (CLO) is a morphological entity that might be involved in secretory processes rather than being the secretory granules themselves.This work was supported by a grant from Statens almindelige Videnskabsfond, Copenhagen.  相似文献

Zusammenfassung Das vegetative Nervensystem entwickelt in allen Abschnitten des membranösen Labyrinths einen intramuralen Plexus. Die marklosen Nervenfasern dieses Plexus entstammen den Gefäßnerven, beim Hund auch den intramuralen Ganglienzellen. Letztere lassen sich den beiden Dogielschen Zelltypen zuordnen und besitzen häufig auffallend randständige Kerne.Das subepitheliale Neurofibrillengeflecht bildet die letzte Endausbreitung des intramuralen Nervensystems im Innenohr. Durch spärliche Anastomosen zwischen den Neurofibrillen kommt ein weitmaschiges Netzwerk zustande.Im Vestibularapparat des Hundes kommen direkt unter dem Epithel besonderes gebaute Kapillare-Gefäßknäuel, Glomeruli vasculosi vor. Besondere, vermutlich dem vasoregulatorischen System zugehörige Nervenfasern versorgen die Glomeruli vasculosi.Der morphologische Charakter des im Plexus marginalis vorhandenen Endgeflechtes der Neurofibrillen zeugt sehr wahrscheinlich für seine Zugehörigkeit zum vegetativen Nervensystem.  相似文献

Preincubation of lymphocytes with des-Tyr1--endorphin (DTE) inhibits the reaction between some HLA alloantisera and their corresponding antigens. One HLA-A2-specific antiserum was found which could detect a subtype of the HLA-A2 antigen on DTE-treated lymphocytes from some donors. Comparison with the HLA-A2 subtypes as defined by a combination of cytotoxic T lymphocyte typing and biochemistry showed a complete correlation with the previously described HLA-A2.3 subtype.  相似文献

We describe a novel compound ciliary structure (g-cilium) from the food groove of the lobate ctenophore Mnemiopsis mccradyi. G-cilia are small, flat compound ciliary organelles that are oriented with their tips pointing toward the mouth. Typically three to four rows of g-cilia line the inner surface of the tentacular groove, which together with the transport groove, make up the food groove. G-cilium cells are 11.4 m long and 4.2 m wide at the g-cilium base. The g-cilium itself is 3.4 m long and tapers to a flat, sharp tip. G-cilia are not motile but are surrounded by many hundreds of smaller, actively motile cilia that beat with orally-directed effective strokes. G-cilia contain 50 conventional `9+2' cilia embedded in a fibrous core that arises from the cell body. In addition, g-cilia contain mitochondria, thousands of small membrane-bounded vesicles and rod bacteria. G-cilia basal bodies are anchored by large, strongly-banded rootlets that extend approximately the entire length of the cell. G-cilia may have organizational, sensory and/or secretory function within the feeding apparatus. Their placement strongly suggests that they play critical roles in feeding. They may enhance the efficiency of prey capture and so contribute to M. mccradyi's well-known voracious appetite. By enhancing prey capture they probably play a critical role in the capacity of this organism to follow prey dynamics, so contributing to dense blooms in mid-late summer in coastal regions.  相似文献

Pattern classification using connectionist (i.e., neural network) models is viewed within a statistical framework. A connectionist network's subjective beliefs about its statistical environment are derived. This belief structure is the network's subjective probability distribution. Stimulus classification is interpreted as computing the most probable response for a given stimulus with respect to the subjective probability distribution. Given the subjective probability distribution, learning algorithms can be analyzed and designed using maximum likelihood estimation techniques, and statistical tests can be developed to evaluate and compare network architectures. The framework is applicable to many connectionist networks including those of Hopfield (1982, 1984), Cohen and Grossberg (1983), Anderson et al. (1977), and Rumelhart et al. (1986b).  相似文献

Cerebellar granule cells (CGC) at different stages of maturation in vitro (1 or 6 DIV), were treated with 25–35 and acetyl-L-carnitine arginine amide (ST857) in presence of 25 mM KC1 in the culture medium, and neuronal viability was assessed. Three days of treatment slightly modified the survival of 1 DIV-treated cells, which degenerate and die five days later -amyloid matching. Similarly, a significative neurotoxic effect was observed on 6 DIV treated-cells after 5 days of exposure to the peptide, while the death occurred within 8 days. ST857 coincubated with 25–35 was able to rescue neurons from 25–35-induced neurotoxicity. We also studied the changes in Ca2+ homeostasis following glutamate stimulation, in control and -amyloid treated single cells, either in presence or in absence of ST857. 25–35 did not affect basal [Ca2+]i, while modified glutamate-induced [Ca2+]i increase, causing a sustained plateau phase of [Ca2+]i, that persisted after the removal of the agonist. ST857 pretreatment completely reverted this effect suggesting that, in CGC chronically treated with 25–35, ST857 could protect the cells by neurotoxic insults of the peptide likely interfering with the cellular mechanisms involved in the control of Ca2+ homeostasis.  相似文献

There are important genetic influences on the tendency to dizygotic (DZ) twinning and it is a plausible hypothesis that these reside in one or more of the genes coding for the major reproductive hormones. We used Southern analysis of DNA from 50 young (<32) mothers of DZ twins, who also had a family history of DZ twinning, and 50 controls, to examine allele frequencies of five restriction fragment length polymorphisms (RFLPs) in four hormone genes coding for follicle stimulating hormone (FSH), chorionic gonadotropin (CGB), inhibin B and gonadotropin releasing hormone (GnRH). Comparison of allele frequencies revealed no significant differences between DZ twin mothers and controls. However this does not rule out the role of these genes in the hereditary tendency of multiple ovulation in humans, since absence of linkage disequilibrium does not imply absence of linkage.  相似文献

Mannans of the yeast Saccharomyces cerevisiae have been implicated as containing the allergens to which bakers and brewers are sensitive and also the antigen recognized by patients with Crohn's disease. A fraction of S. cerevisiae mannan, Sc500, having high affinity for antibodies in Crohn's patients has been characterized by NMR spectroscopy followed by fragmentation using alkaline elimination, partial acid hydrolysis and acetolysis. The released oligosaccharides were separated by gel filtration on a Biogel P4 column and analyzed by fluorescence labeling, HPLC and methylation analysis. The relationship between structure and antigen activity was measured by competitive ELISA. The antigenic activity of the original high molecular weight mannan could be ascribed to terminal Man13Man12 sequences which are rarely found in human glycoproteins but were over-represented in Sc500 compared to other yeast mannans.  相似文献

Two intracellular -glucosidases (E.C. 3.2.1.21) were purified from the filamentous fungus Neurospora crassa, mutant cell-1 (FGSC no. 4335) and characterized. The extent of purification were 2.55- and 28.89-fold for -glucosidase A and -glucosidase B, respectively. -Glucosidase A was a dimeric protein, and B a monomeric protein, with molecular masses of 178 and 106 kDa, respectively. Both isoenzymes were glycoproteins with relatively high carbohydrate contents (-glucosidase A, 29.2%; -glucosidase B, 34.2%). The isoelectric points determined by IEF were 6.27 and 4.72, respectively. pH optima for activity were determined to be 5.0 and 5.5, and temperature optima to be 55 and 60 °C, for -glucosidases A and B, respectively. Both purified -glucosidases. especially -glucosidase B, showed relatively high stability against pH and temperature. Both enzymes were stable in the pH range of 5.0–9.0. The activities were completely retained up to 48 h at temperatures below 40 °C. At higher temperatures, enzymes were relatively unstable and lost their activities at 60 °C after 24 h. Both -glucosidases were highly activated by CuCl2, and inhibited by SnCl2 and KMnO4. Hg2+ and Ag+ also inhibited severely -glucosidase B. The K m and V max values of the isoenzymes against cellobiose as substrate were 1.50 mM and 12.2mol min–1 mg–1 for -glucosidase A and 2.76 mM and 143.5 mol min–1 mg–1 for -glucosidase B.  相似文献

Summary The epidermal tissues of marine mussels can accumulate amino acids from surrounding sea water. In the present study, gill tissue isolated from the California coastal mussel,Mytilus californianus, was used in conjunction with intact, actively pumping mussels to study epidermal transport processes. There appeared to be at least four pathways for this uptake: i) a -neutral pathway which transports taurine; ii) an -acidic pathway specific for substrates such as aspartate; iii) an -neutral pathway having a general specificity for this class of compound, but which also accepts the basic amino acid, lysine; and iv) a second -neutral pathway, also of broad specificity, capable of accepting the imino acid, proline, as a substrate. Replacement of Na in sea water with choline reduced uptake of leucine, taurine, aspartate, and proline by more than 95%, and reduced lysine uptake by 75%, suggesting that Na-independent pathways play no significant role in epidermal transport in the gill. Isolated gill tissue was used to estimate the maximum transport capacities (J max's) of the pathways, which ranged from approximately 5 to 25 mol/(g·hr). Apparent Michaelis constants (K t *'s) of the epidermal transporters were estimated using a convection-diffusion model introduced previously (Wright and Secomb, Am J Physiol 247:R346–R355, 1984). TheseK t *'s ranged from 1 to 5 M. A contextualizing image (CI) is a powerful central image of a dream which appears to contextualize (provide a picture-context for) the dreamer's emotion. For instance, dreamers who have experienced any serious traumatic event sometimes dream, I was overwhelmed by a tidal wave. This appears to picture their feeling of terror and/or vulnerability.A scoring system for CIs is examined here and is applied to dreams and daydreams supplied by 40 students. Two raters scoring dreams on a blind basis showed good inter-rater reliability. Recent dreams were shown to have more as well as more intense CIs than recent daydreams; likewise, dreams that stand out had more intense CIs than daydreams that stand out. Students with thin boundaries had more and more intense CIs than students with thick boundaries in their recent dreams and nightmare, but not so clearly in dreams and nightmares that stand out. The emotions judged as contextualized by the powerful images tended towards fear/terror and helplessness/vulnerability in dreams (especially in dreams that stand out) whereas emotions contextualized by images in daydreams showed a wide range with no clusters.  相似文献

Conclusion The Mauritanian ruling class, imposed upon the masses in 1960, continued to be assisted and advised by French cooperants techniques in the post-colonial period working closely with international finance capital, and have been promising their subjects that the construction of their society depended on the successful operation of multinational cooperation, represented in the country by MIFERMA and SOMIMA. By the 1970's it became clear to the majority of the Mauritanian people that these foreign firms were carrying out the systematic pillage of their resources rather than stimulating the development of their country; and that the rapid growth of the extractive sector was part of a radical process of peripherization, which has woven the Mauritanian economy and society into the capitalist center. Moreover, external economic aid pushed the neo-colonial state into the debt trap. Accordingly, Bonte observed that while production apparently increased and while private capital certainly flowed into the country, it flowed out in even larger quantities. These flows led to a persistent deficit in the balance of payment and to a growing debt [95]. This trend has been worsening in recent years. Without loans from European, American, and conservative Arab regimes, the neo-colonial state would have collapsed financially. Indeed, from 1970 to 1975, foreign loans provided between 75 and 90 percent of the funds for the governmental development programs [96].The social conditions of the people drastically worsened in the 1970's. For instance, in 1976 a government official admitted that only 15 percent of the total population received medical attention. Moreover, the activities of the multinational corporations have destroyed the supportive communal structures of the rural masses which, in the past, have minimized the consequences of drought and other economic crises. When the 1969–74 drought occurred, innumerable droughtstricken nomads and subsistence agriculturalists found themselves helpless and completely destitute. Confronted with such as state of affairs, they had no alternative but to flee to the mushrooming slums surrounding the urban centers, there to lead mutilated and mutilating lives.For these masses neither Mauritanianization nor nationalization have prevented their growing immiseration. The extractive activities, whether under the aegis of multinational corporations or state-owned companies, which remain dependent on finance capital and its market, cannot resolve the fundamental problems of the Mauritanian people. On the contrary, the operations of these foreigndominated companies negate the possibility of a liberating, diversified industrial process that relies on, assesses and takes into account the primary needs of the people. Only an industry that results in large employment effects generated in proportion to the amount of capital invested and whose management is assumed to a large degree by the producers themselves, can achieve deep development of a country like Mauritania. Extractive industry controlled by multinational corporations cannot generate positive linkage effects [97] to the local economic sectors powerful enough to effectuate their development within a well-planned, culturally integrated, and economically fruitful national social economy. A neo-colonial state of the Mauritanian type — ruled by a class that owes its ascension to power to a retreating colonialism — formed as a mediating mechanism for prolonging the foreign exploitation of its own resources — cannot resolve the basic problems of the people. In short, when the iron ore of Mauritania is worked out, that country will go back to the desert [98]. However, this observation by Amin is apocalyptic, pessimistic, and dangerously deterministic. Another, more viable, more human, alternative is for the northwestern African peoples to extricate themselves from the world capitalist system, and its local comprador bourgeois allies. Summary Glycoproteins rich in mannosyl or glucosyl residues were analyzed in the subcommissural organ (SCO) and the pineal organ of the sheep (Ovis aries). By use of concanavalin A labelled with fluorescein isothiocyanate, fluorescent material was found both in ependymal and hypendymal cells of the SCO. In the pineal organ, either isolated or grouped parenchymal cells showed a marked fluorescence. These cells may correspond to ependymal elements also called interstitial cells or supporting cells. In addition, scarce slender, fluorescent processes were observed in the pineal parenchyma. The techniques of electrophoresis and electrotransfer on nitrocellulose paper have been applied to analyze the glycopeptide content of the SCO and the pineal organ in comparison to cerebellar and cerebral fractions solubilized by use of Triton X 100. Approximately 30 different concanavalin A-reactive glycopeptides were revealed in each fraction. In the SCO extract four glycopeptides (30, 54, 72, 100 kd) might correspond to subunits of the glycoprotein(s) characteristically stored in the ependymal cells of the SCO. In addition, two glycopeptides (32/33, 115 kd) are specific to the pineal organ extract. The possible similarity of the concanavalin A-reactive material in both organs is discussed and a putative secretory activity of the pineal ependymal cells is postulated.  相似文献

Summary Measurements of potassium activity with a potassium-sensitive microelectrode have been made in the cells of the stomatal complex, and in epidermal cells, ofCommelina communis L., as a function of stomatal aperture. The estimated osmotic effects of the changing accumulation of potassium salts in the guard cell have been compared with the previous estimates of the osmotic changes required to open/close the pore. The results suggest that a significant fraction of the osmotic pressure of the guard cells, particularly when closed, is contributed by solutes other than potassium salts. The degree of potassium accumulation may determine the aperture of wide-open stomata, but the potassium changes in the early stages of opening are much too small to account for the osmotic changes required. The difference in potassium contents of intact and isolated guard cells is close to that required to overcome the previously estimated effect of subsidiary cell turgor on the water relations of the guard cell. In some tissue (but not in all) much more K is lost from epidermal cells than appears in other cells of the complex as the stomata open, and extracellular storage would be required.  相似文献

Two-dimensional (2D) gel electrophoresis of immunoprecipitated HLA-DR antigens from eight homozygous typing cells (HTC) expressing the HLA-DRw8 specificity revealed a clustering of polymorphic chain patterns into distinct electrophoretic variants. The variant patterns correlate with three discrete HLA-D clusters that are defined in the mixed leukocyte culture reaction (MLR) using DRw8-positive HTC. These HLA-D clusters have been provisionally designated Dw8.1, detected primarily in Caucasoids, Dw8.2, detected primarily in American Indians, and Dw8.3, detected predominantly in Orientals. All three HLA-Dw8.1 cell lines express a single DR-locus product as defined by immunoprecipitation with a DR-specific monoclonal antibody, P4.1. This DR chain is identical among the Dw8.1 cell lines and different from the DR chains of the Dw8.2 and Dw8.3 cell lines. Two separate Dw8.2 HTC express a shared DR chain that is slightly more basic than the 8.1 DR molecule; interestingly, one of these lines also expresses an additional DR-like chain not found in the other cells. Thus, the two lines defining the Dw8.2 cluster share one distinct class 11 molecule, but differ in another and therefore are not biochemically HLA-identical. Cells from the Dw8.3 cluster are likewise distinct from all other Dw8 clusters. One additional DRw8-positive HTC has been analyzed and found to be distinct from the Dw8.1, 8.2 and 8.3 clusters by both MLR and 2D gels. lmmunoprecipitates using monoclonal antibody 1B5 [anti-DR and anti-DQ(DS)] identify additional polymorphic class II variants among the cell lines tested. These data indicate that HLA-DRw8 is a public serologic specificity present on class II molecules expressed on multiple distinct haplotypes. These haplotypes differ from each other in expression of polymorphic class II molecules encoded by at least two HLA loci. They also differ in HLA-D, even though they all type as HLA-DRw8 homozygous. In Dw8.2, variation in expressed chains is not reflected in variation in HLA-D, indicating that MLR, as well as serologic typing, does not detect the full degree of allelic polymorphism within HLA.  相似文献

Wax deposits on leaf surfaces ofin vitro-grown plantlets,in vitro plantlets treated with polyethylene glycol and greenhouse-grown seedlings from five cultivars of date palm (Phoenix dactylifera L.) were extracted and quantified. Significant variations among treatments and cultivars were obtained. Greenhouse-grown plants had the greatest wax deposits followed by the acclimatized plantlets.In vitro plantlets had an average of 15% of the wax of greenhouse plants. Cultivar and plant age differences had a significant effect on the quantity of wax deposits. Greenhouse seedlings of Majhool, Deglet Noor and Khadraoui (cultivars grown under irrigation) had less wax accumulation than Zahidi and Sayer, dryland cultivars.The increase in wax deposition as a result of polyethylene glycol treatment, explains in part, the decreased water loss observed in acclimatized plantlets when transferredex vitro. Zusammenfassung Die autoradiographischen Untersuchungen dienten der Bestimmung der DNS-Synthese-Dauer in den Zellen der Imaginalanlagen der Hinterflügel vonEphestia kühniella Z.Zu diesem Zweck mu\te das Doppelmarkierungsverfahren herangezogen werden. Raupen in den verschiedenen Stadien des letzten Raupenstadiums erhielten eine 1. Injektion von3H-Thymidin und 1 1/2 oder 6 Std später eine 2. Injektion von14C-Thymidin. Die DNS-Synthese-Phase dauert während der eigentlichen Wachstumsphase der Raupen vom 1.–7. Tag nach der letzten Raupenhäutung 12 Std. Im Vorpuppenstadium dauert die DNS-Synthese dagegen nur 6 Std. Diese Ergebnisse wurden in verschiedenen Versuchen mit unterschiedlichen Bedingungen bestätigt.Es wird vermutet, da\ der Wechsel von der langen zur kurzen DNS-Synthese-Dauer durch das Hormon Ecdyson verursacht wird.Verschiedene Möglichkeiten der Zellproliferation in den Imaginalanlagen werden diskutiert. Hierbei werden die erhaltenen Ergebnisse mit den schon bekannten, von Zellzählungen gewonnenen, Vorstellungen verglichen.Die G2-Phase erwies sich als variabel in ihrer Dauer. Sie kann von einer bis auf mehrere Stunden verlängert werden. Die Vermutung wird geäu\ert, da\ die Wachstumsordnung und die Differenzierungsprozesse der Imaginalanlagen der Hinterflügel durch Variation der G2-Phase kontrolliert werden kann.
Autoradiographic determination of the of DNA synthesis duration in the wing imaginal discs ofEphestia kühniella Z.
Summary These experiments were carried out to determine the duration of DNA-synthesis in the cells of the developing wing discs ofEphestia kuehniella Z.For this purpose the double labeling technique was applied. Larvae of the last larval instar were injected first with3H-thymidine and than 1 1/2 or 6 hours later with14C-thymidine. During the growth phase from the 1st to the 7th day after the last larval moult, S-phase duration was found to be about 12 hours. In the prepupal stage, however, S-phase duration was measured to be 5.8 hours. These results were verified in different experimental conditions.It is suggested that the shift from long to short duration of the S-phase is controlled by Ecdyson.Cell proliferation within the wing discs is discussed using these data and the known cell counts. G2-phase was found to be variable lasting from 1 to several hours. It is suggested that the growth pattern and the differentiation of the imaginal discs can be controlled by variing G2-phases.
