With the imposition of salt stress (0.5 to 3 % NaCl or CaCl2) a decrease in germination rate and accumulation of proline was observed in the root tissue. Both NaCl and CaCl2 solutions induced an increase in the total peroxide content and lipid peroxidation and decrease in catalase, guaiacol peroxidase and superoxide dismutase activities in root tissues suggesting an oxidative stress in the salt sensitive rice cultivar.  相似文献

The effects of increasing relative humidity on the growth and salt tolerance of two melon (Cucumis melo L.) cultivars, Revigal C-8 (salt sensitive) and Galia (salt tolerant) was investigated. One month after germination, the plants were exposed for 15 d to 0 (control) and 80 mM NaCl, under relative humidity (RH), 30 and 70 %. The growth of the whole plant, leaf, stem and root of cv. Revigal C-8 was increased with increasing RH. On the other hand, cv. Galia showed an increase in root growth with increasing RH only under the NaCl treatment. Under salinity, most of the Na+ was withheld in the stems. An increase in RH in the NaCl treatment significantly decreased Na+ and Cl concentrations in leaves of cv. Revigal C-8, while it had no effect on their concentrations in cv. Galia. In both cultivars, increasing RH under NaCl condition significantly decreased water contents in leaves and stems, and increased osmotic potential in roots. The amount of the root exudate of cv. Galia was significantly decreased with increasing RH, while it was not affected in cv. Revigal C-8. Under the NaCl treatment, cv. Galia had significantly higher leaf osmotic potential than cv. Revigal C-8 at both relative humidities and higher amount of root exudate at 30 % RH.  相似文献

盐生植物黄花补血草广泛分布于我国西北地区、东北西部以及华北北部,对改良盐碱土壤具有重要的生态作用。以黄花补血草(Limonium aureum(L.)Hill)为材料,研究分析了不同浓度NaCl胁迫对其种子萌发和幼苗生长产生的抑制效应及作用机制。结果表明:低浓度NaCl(25 mmol/L和50 mmol/L)处理不影响黄花补血草种子萌发和幼苗生长,25 mmol/L NaCl甚至促进了根生长,而高浓度NaCl(100 mmol/L和150 mmol/L)处理明显抑制种子萌发及幼苗生长。利用荧光探针的检测结果表明,NaCl处理的幼苗根中过氧化氢(H2O2)和一氧化氮(NO)含量明显高于对照水平。碘化丙啶(PI)染色结合激光共聚焦显微镜观察及检测相对电导率结果显示,高浓度NaCl处理抑制了幼苗根尖伸长区细胞的伸长生长,增加了细胞膜的通透性,对根细胞造成了明显的伤害。此外,高浓度NaCl处理诱导叶片丙二醛(MDA)含量显著升高。以上结果说明,黄花补血草对低浓度的盐具有一定的耐盐性,但高浓度盐降低了种子的萌发率,使幼苗根中H2O2产生增加,抑制根尖伸长区细胞的伸长生长,对根、叶造成明显氧化损伤,从而抑制黄花补血草幼苗的生长。  相似文献

The effects of 5-aminolevulinic acid (ALA), a key precursor in the biosynthesis of porphyrins such as chlorophyll and heme, on development and salt tolerance of microtubers of two potato (Solanum tuberosum L.) cultivars Jingshi-2 and Zihuabai were examined under in vitro conditions. ALA at 0.3–3 mg/l promoted microtuber formation by increasing the average number, diameter, and fresh weight of microtubers especially under 0.5% NaCl stress conditions, but further increase in ALA concentration resulted in a reduction of microtuber yield irrespective of NaCl stress. Under 1.0% NaCl stress conditions, microtuberization was seriously repressed and could not be restored by the addition of ALA. The accumulation of malondialdehyde in the microtubers treated with 30 mg/l ALA increased by 22% compared to the controls (no salinity), while only a 7% increase was observed when the microtubers were exposed to 0.5% NaCl, indicating that ALA functions as a protectant against oxidative damages of membranes. Under 0.5% NaCl stress conditions, the highest activities of peroxidase and polyphenoloxidase were detected in microtubers treated with ALA at 0.3 and 3 mg/l, being by 73% and by 28% greater than those in the untreated controls, respectively. These results demonstrate that ALA at lower concentrations of 0.3–3 mg/l promotes development and growth of potato microtubers in vitro and enhances protective functions against oxidative stresses, but ALA at 30 mg/l and higher concentrations seems to induce oxidative damage probably through formation and accumulation of photooxidative porphyrins.  相似文献

Summary The effects of increasing concentrations of NaCl and CaCl2 on quince (Cydonia oblonga Mill. BA 29 clone) somatic embryogenesis and adventitious root regeneration were investigated. Leaves collected from in vitro-grown shoots were used as explants and induced for 2d in liquid Murashige and Skoog medium containing 11.3 μM 2,4-dichlorophenoxyacetic acid. Explants were then cultured on semisolid Murashige and Skoog medium enriched with 4.7 μM kinetin and 0.5 μM naphthaleneacetic acid under red light for 25 d and under white light for another 25 d. Two experiments were performed: in the first, NaCl was used at 0,25, 50, 100, and 200 mM in factorial combination with CaCl2 at 3, 9, and 27 mM; in the second, NaCl was applied at 0, 5, 10, 20, 40, and 80 mM in combination with CaCl2 at 0.3, 1.0, and 3.0 mM. Quince leaves revealed the capacity to regenerate somatic embryos and/or adventitious roots. Quantitative and qualitative regeneration from leaves was affected by NaCl treatments: increasing NaCl concentrations, in combination with CaCl2 at 1 mM, led to an increase in the proportion of leaves producing somatic embryos only, and to a decrease of both leaves regenerating roots only and leaves simultaneously producing somatic embryos and adventitious roots. This suggests a beneficial effect of salt stress on the embryogenic process. The regeneration response decreased with increasing salt concentrations and was almost totally inhibited above 50 mM NaCl and 9 mM CaCl2. The presence of CaCl2 in the culture medium apparently mitigated the effects of salt stress, but only when NaCl was applied at 40 mM. NaCl at 5 mM, in the presence of 0.3 or 1 mM CaCl2, was favorable both to somatic embryo and root production. No value of the ratio Na+/Ca2+ was found to be optimal for the regeneration processes.  相似文献

Potato plants grown in vitro were subjected to different salt stresses by providing the salts NaCl, Na2SO4, MgCl2 and MgSO4 in different concentrations up to 300 mM. Salinity greatly affected the survival and the rooting of the plants. Shoot and root growth decreased with increasing salt concentrations. Under mild stress conditions, i.e. in conditions where the plant is able to adapt to the stress, the observed decrease was dependent upon the salt used. Under severe stress conditions, however, the decrease of the shoot and root growth was independent of the nature of the ions.  相似文献

The changes in cell wall peroxidase activity against ferulic acid (FPOD) and lignin level in roots of NaCl-stressed rice seedlings and their correlation with root growth were investigated. Increasing concentrations of NaCl from 50 to 150 mmol L−1 progressively decreases root growth. The reduction of root growth by NaCl is closely correlated with the increase in FPOD activity extracted from the cell wall. In contrast, lignin level was reduced by NaCl. Since proline and ammonium accumulations are associated with root growth inhibition caused by NaCl, we determined the effect of proline or NH4Cl on root growth and FPOD in roots. Exogenous application of NH4Cl or proline markedly inhibited root growth and increased FPOD activity in rice seedlings in the absence of NaCl. An increase in FPOD activity in roots preceded inhibition of root growth caused either by NaCl, NH4Cl, or proline. Our results suggest that cell-wall stiffening catalyzed by FPOD may participate in the regulation of root growth reduction of rice seedlings caused by NaCl.  相似文献

Results obtained with graded concentrations of NaCl (20–200 mM) show decrease in the chlorophyll ‘a’ contents of Anabaena with increasing concentration of NaCl except at extremely low concentration of NaCl (5–20 mM). The rate of Hill activity and oxygen evolution are found to be stimulated by lower concentrations of NaCl, but not at higher concentrations of NaCl. Results have demonstrated that the O2 evolution process is relatively more sensitive to NaCl stress than the Hill activity. Further, the results show NaCl induced an increase in the rate of RNO bleaching and loss of total thiol (-SH) contents. Taken together, these results suggest a NaCl-induced general oxidative stress. Results on the effect of oxygen radical quenchers reveal a predominant role of singlet oxygen in the NaCl-induced general oxidative stress as evident from a higher quenching effect of sodium azide than formate and histidine on the rate of RNO bleaching in Anabaena cells. However, the rate of lipid peroxidation and SOD activity show a declining pattern in response to increasing concentrations of NaCl. There is the possibility of a NaCl-induced decrease in the rate of lipid peroxidation when the SOD activity is also lower. But the NaCl-induced decline in the SOD activity does suggest that symptoms of general oxidative stress at elevated levels of NaCl are apparently owing to collapse of intracellular defense of the cells against the toxic oxygen radicals, not because of the higher rate of photosynthetic activity. Lactic acid content and production, as well as lactic dehydrogenaseactivity were studied in pea root tips grown in media salinatedwith NaCl or Na2SO4. Salinity of both types depressed the lacticacid content of the tissue. Lactic dehydrogenase activity linkedto NADH was depressed by high concentrations of NaCl in thegrowth medium, but was stimulated by increasing concentrationsof Na2SO4. There was a trace of NADPH linked activity of theenzyme; this activity was not affected by chloride salinitybut was depressed by sulphate salinity. A salt marsh species, Jaumea carnosa, was used in hydroponic experiments to test the effects of increasing NaCl concentrations on leaf succulence and plant accumulations of K, Ca, Mg, Na and Cl. A nested experimental design was used with four salinity levels. Plants were grown in full Hoagland's solution plus different amounts of NaCl (0.0–1.2 osmoles). Leaf succulence was measured as percent water content as well as vertical elongation of mesophyll cells. There were no corresponding increases in leaf succulence with increasing concentrations of NaCl in the root zone. Plants receiving aerosol spray (40 mg/dm2/day) did not show significant increases in leaf succulence. Leaf succulence was significantly increased when the plants were removed from the NaCl solutions and placed in non-salinized Hoagland's solution. Osmotic concentrations of cell sap in leaf tissues showed significant increases as NaCl concentrations increased in the root zone. The concentrations of K, Ca and Mg were higher in plants grown without NaCl than in those grown with NaCl. The accumulations of K in the root tissues were always higher than those of the shoot tissues. Although there was a two-fold difference in NaCl concentrations at the highest levels, the concentrations of Na in the shoot tissues were relatively similar. The results of the Cl analyses of shoot tissues showed a similar pattern of regulation of uptake. This regulation of salt uptake may be important in preventing injury by limiting accumulations of salt in plant tissues when growing in soils of high osmotic potentials.  相似文献

The in vitro response of kiwifruit (Actinidia deliciosa) to increasing concentrations of boron (B) and NaCl in the culture medium was studied. Kiwifruit shoot cultures were grown in vitro for 12 weeks on an MS medium containing two B concentrations (0.1 and 2 mM) combined with five NaCl concentrations (0, 10, 20, 40 and 80 mM). Kiwifruit produced the longest shoots with 2 mM B when NaCl concentration was 0--20 mM. More shoots were produced with 2 mM B for all NaCl treatments. More shoots were produced with 2 mM B and 10 and 20 mM NaCl. High B concentrations in the culture medium significantly increased shoot proliferation. Explants exhibited a moderate chlorotic appearance with 40 mM NaCl and shoots died with 80 mM NaCl. With 2 mM B, the B concentration of explants was 5--9X greater for the various NaCl treatments compared to the control. Increasing the NaCl concentration from 10 to 80 mM, resulted in higher Na and Cl concentrations in explants for all B treatments, while K and Ca concentrations decreased. Phosphorus concentration in the explants was significantly increased by increasing the NaCl concentration reaching a maximum value at 80 mM NaCl for the two B concentrations.  相似文献

The combined effect of NaCl and P on the growth of lucerne was studied in two hydroponic greenhouse experiments. NaCl concentrations were identical in each experiment (0, 50 and 100 mM NaCl) while external P concentrations were low (viz. 0.002, 0.02 and 0.2 mM measured as 0.006, 0.026 and 0.2 mM, respectively) in one experiment and higher (0.5 and 5.0 mM) in the second. Plant biomass was reduced more by the low P levels than by high concentrations of NaCl. A significant NaCl*P effect was found where external P concentrations were low (0.006–0.2 mM) but there was no difference in plant production between the two P concentrations of 0.5 and 5.0 mM. Shoot and root concentrations of Na and Cl increased significantly with increasing NaCl concentration in both experiments and there were some differences in the concentrations of these ions at different external P levels. At low P, NaCl had no significant effect on shoot concentrations of P; however, root P concentrations tended to decrease with increasing NaCl level. Increasing external P from 0.006 to 0.2 mM led to significant increases in P concentrations in both roots and shoots. At higher P, concentrations of P in both the shoots and the roots did not differ with external NaCl or P conditions. Our results illustrate the complex relationship that exists between NaCl and P at low P levels. We conclude that high or non-limiting concentrations of P (0.2 – 5.0 mM) do not affect lucerne's response to NaCl.  相似文献

Summary Growth sensitivity of four local grapevine (Vitis vinifera) varieties, Ashlamesh, Helwani, Kassofee, and Khoudeiry, were evaluated for salt. They were cultured on DSD1 medium until rooting stage, then they were transferred to a liquid DSD1 medium containing 0, 10, 20, 30, 40, 80, 120, or 150 mM NaCl for 30 d. The shoot length and leaf number of Ashlamesh, Helwani, and Kassofee were significantly increased at 10 and/or 30 mM NaCl, whereas, 150 mM NaCl decreased shoot length of all varieties except Kassofee. The presence of NaCl at 80 mM or higher concentrations decreased the chlorophyll content and root number of all varieties, while 30 mM NaCl increased root number of Kassofec.  相似文献

The effects of salinity (NaCl) stress on growth, cytosolic Ca(2+) gradients and cytosolic pH homeostasis of root hairs of Arabidopsis thaliana are assessed here. Neither cytosolic Ca(2+) nor pH at the hair apex were significantly affected by 20 min exposure of up to 90 mM NaCl or of up to 5 mM extracellular Ca(2+). Exposure to increasing NaCl concentrations, up to 90 mM, for 2 d or 6 d reduced hair extension, and this inhibition was relieved by supplemental extracellular Ca(2+). Such extended salinity stress reduced the magnitude of the Ca(2+) gradient in the apical 12 microm of hairs at all NaCl concentrations tested (up to 90 mM), including NaCl concentrations that did not reduce hair extension. The magnitude of the tip-focused gradient was also reduced in root hairs of plants grown with low (0.5 mM) extracellular Ca(2+) when compared to those in 5 mM extracellular Ca(2+), regardless of the presence of NaCl. Up to 90 mM NaCl did not affect cytosolic pH of root hairs in any of the treatments. It is concluded that NaCl inhibition of root hair extension in the long term may operate via alterations in the tip-focused Ca(2+) gradient that regulates root hair growth. However, NaCl-induced alterations in this gradient do not always lead to detectably altered growth kinetics. Short-term signalling events in response to NaCl may operate by a means other than altering Ca(2+) at the root hair apex. Salinity stress in root hairs does not appear to be mediated by effects on cytosolic pH.  相似文献

Two sets of experiments to determine the effect of mycorrhiza on soybean (Glycine max) growth under saline conditions and to investigate the salt acclimation of mycorrhizal fungi were conducted. In the first experiment, the effect of an arbuscular mycorrhizal (AM) fungus Glomus etunicatum on mineral nutrient, proline and carbohydrate concentrations and growth of soybean. Under different NaCl concentrations (0, 50, 100, 150 and 200mM) was evaluated. Salinity decreased AM colonization. In both the M and nonAM plants shoot and root proline and shoot Na and Zn concentrations were increased under salinity. Soybean plants inoculated with the AM fungus had significantly higher fresh and dry weight, root proline, P, K and Zn but lower shoot proline and Na concentrations compared to the non inoculated plants. In the second experiment, the AM fungus was pre-treated with NaCl (salt acclimation) then was used as inoculum for soybean plants subjected to 100mM NaCl. Root colonization, fresh and dry weight, root proline, P, K and Zn concentrations were greater in soybean plants inoculated with the salt pre-treated fungus, compared to those inoculated with the nonsalt pre-treated fungus. However, for Na, the situation was the opposite. Based on these results, the AM inoculation helps the growth of soybean plants grown in saline conditions. When the AM fungus was pre-treated with NaCl with a gradual increase of concentration, and then exposed to a sudden salt stress, their efficiency was increased. This may be due to the acclimation of the AM fungus to salinity.  相似文献

采用砂培试验,用不同浓度的NaCl和等渗PEG6000(聚乙二醇6000,渗透势约为-0.44MPa)处理生长20d的菊芋幼苗,3d后分别测定其根、茎、叶中的Na^ 、K^ 、Cl^-含量以及叶片SOD、POD活性。结果表明,在NaCl和PEG胁迫下,根、茎、叶的Na^ 、Cl^-含量不断升高,而K^ 含量保持稳定。其中,茎中Na^ 含量高于根和叶。NaCl胁迫下,根、茎、叶的SX、Na值随胁迫强度的增加而递增,茎中SK、Na值小于根和叶。随着NaCl胁迫强度的增加,菊芋幼苗叶片的SOD和POD活性先上升后下降;PEG处理下,SOD活性分别高于对照和等渗NaCl处理31.1%和27.1%;而POD活性却分别低于对照和等渗NaCl处理26.0%和36.1%。  相似文献

In several plant species, oxidative stress has been shown to be one of the causes of damage produced by salinity. In order to assess the implication of oxidative stress in the reported sensitivity of the citrus rootstock Carrizo citrange to salt stress, 5-month-old seedlings were grown with increasing NaCl concentrations added to the watering solution. As an indicator of oxidative damage, malondialdehyde content was measured. The antioxidant capability of the plants was determined by measuring superoxide dismutase, ascorbate peroxidase, catalase and glutathione reductase activities together with the non-enzymatic antioxidant activity. As additional physiological responses to the stress, 1-aminocyclopropane-1-carboxilic acid and proline accumulation were assessed. Data indicate that Carrizo citrange responded to salt-induced oxidative stress by increasing enzymatic and non-enzymatic antioxidant defenses proportionally to the extent of the stress imposed, and that in all plants the malondialdehyde content remained at a moderate level. We suggest that the important deleterious effects reported in Carrizo citrange grown under high NaCl concentrations are mainly due to a cellular intoxication by Cl(-) ions and not to the salt-induced oxidative stress.  相似文献

Two-month-old healthy seedlings of a true mangrove, Bruguiera parviflora, raised from propagules in normal nursery conditions were subjected to varying concentrations of NaCl for 45 d under hydroponic culture conditions to investigate the defence potentials of antioxidative enzymes against NaCl stress imposed oxidative stress. Changes in the activities of the antioxidative enzymes catalase (CAT), ascorbate peroxidase (APX), guaiacol peroxidase (POX), glutathione reductase (GR) and superoxide dismutase (SOD) were assayed in leaves to monitor the temporal regulation. Among the oxidative stress triggered chemicals, the level of H2O2 was significantly increased while total ascorbate and total glutathione content decreased. The ratio of reduced to oxidized glutathiones, however, increased due to decreased levels of oxidized glutathione in the leaf tissue. Among the five antioxidative enzymes monitored, the APX, POX, GR and SOD specific activities were significantly enhanced at high concentration (400 mM NaCl), while the catalase activities declined, suggesting both up and downregulations of antioxidative enzymes occurred due to NaCl imposed osmotic and ionic stress. Analysis of the stress induced alterations in the isoforms of CAT, APX, POX, GR and SOD revealed differential regulations of the isoforms of these enzymes. In B. parviflora one isoform of each of Mn-SOD and Cu/Zn-SOD while three isoforms of Fe-SOD were observed by activity staining gel. Of these, only Mn-SOD and Fe-SOD2 content was preferentially elevated by NaCl treatment, whereas isoforms of Cu/Zn-SOD, Fe-SOD1 and Fe-SOD3 remained unchanged. Similarly, out of the six isoforms of POX, the POX-1,-2,-3 and -6 were enhanced due to salt stress but the levels of POX-4 and -5 remained same as in control plants suggesting preferential upregulation of selective POX isoforms. Activity staining gel revealed only one prominent band of APX and this band increased with increased salt concentration. Similarly, two isoforms of GR (GR1 and GR2) were visualized on activity staining gel and both these isoforms increased upon salt stress. In this mangrove four CAT-isoforms were identified, among which the prominent CAT-2 isoform level was maximally reduced again suggesting differential downregulation of CAT isoforms by NaCl stress. The results presented in this communication are the first report on the resolutions of isoforms APX, POX and GR out of five antioxidative enzymes studied in the leaf tissue of a true mangrove. The differential changes in the levels of the isoforms due to NaCl stress may be useful as markers for recognizing salt tolerance in mangroves. Further, detailed analysis of the isoforms of these antioxidative enzymes is required for using the various isoforms as salt stress markers. Our results indicate that the overproduction of H2O2 by NaCl treatment functions as a signal of salt stress and causes upregulation of APX, POX, GR and deactivations of CAT in B. parviflora. The concentrations of malondialdehyde, a product of lipid peroxidation and lipoxygenase activity remained unchanged in leaves treated with different concentrations of NaCl, which again suggests that the elevated levels of the antioxidant enzymes protect the plants against the activated oxygen species thus avoiding lipid peroxidation during salt stress.  相似文献

It has been shown that salicylic acid (SA) acts as an endogenous signal molecule responsible for inducing abiotic stress tolerance in plants. The effect of SA and sodium chloride (NaCl) on growth, metabolite accumulation, oxidative stress and enzymatic and non-enzymatic antioxidant responses on common bean plants (Phaseolus vulgaris, cv. F-15) was studied. Results revealed that either SA or NaCl decrease, shoot, root and total plant dry weights. SA treatments decreased the contents of proline, and reduced forms of ascorbate and glutathione, however, the content of soluble sugars (TSS), thiobarbituric acid-reactive substances (TBARs) and oxidized ascorbate remained unaffected. On the other hand, salinity significantly reduced the levels of endogenous SA but increased the content of proline, soluble sugars, TBARs, ascorbate and glutathione, as well as all increasing the levels of antioxidant enzyme activities assayed, except CAT. The application of SA improved the response of common bean plants to salinity by increasing plant dry weight and decreasing the content of organic solutes (proline and TSS) and damage to the membrane (TBARs). Moreover, SA application under saline conditions decreased the levels of antioxidant enzyme activities POX, APX and MDHAR which could indicate successful acclimatization of these plants to saline conditions.  相似文献

Self-rooted olive ( Olea europaea L.) plants were grown in hydroponics at various NaCl concentrations (from 0 to 200m M ) for 28 to 32 days followed by 28 to 30 days of relief from salinity over two growing seasons. Olive leaves accumulated both glucose and mannitol during the period of salinity stress. The concentrations of fructose, myo -inositol, galactose, galactinol, sucrose, raffinose, and stachyose were not significantly affected by salinity. Starch content was decreased by salinity. The mannitol/glucose and mannitol/soluble carbohydrates ratios increased as the external NaCl concentration was increased, but returned to the control levels during the relief period. The increase in mannitol or glucose molar concentrations, expressed on a leaf tissue water basis, was partially due to a reduction in leaf tissue water content under salinity stress. However, an increase in mannitol concentration was also observed when expressed on a dry weight basis. The accumulation of mannitol in leaf tissue preceded any reduction in leaf area rate or net assimilation rate. The increase in leaf mannitol or glucose concentration was positively correlated with the increasing level of salinity at the root zone, but not with the accumulation of Na+ in the shoot. The role of mannitol. a potential osmoregulator in leaf mesophyll during salinity stress, is discussed in relation to the complex carbohydrate composition of olive leaves.  相似文献
