A simple, nondestructive physical process was developed for routinely isolating the outermost layers from female, male, and sporophyte fronds of Chondrus crispus Stack-house. Yields of pure cuticles from apical segments ranged from 0.74 to 2.35% on a dry weight basis after 5–7 d of culture. These undegraded cuticles were examined by electron microscopy (scanning and transmission electron microscopy), spectroscopy (infrared and X-ray), and chemical means. Cuticles isolated from female or male fronds were characterized by parallel arrays of electron-dense lamellae (typically 6–14) alternating with more electron-transparent regions. The thickness and uniformity of these lamellae provide the physical basis for the iridescence characteristic of C. crispus fronds. Sporophyte fronds are not iridescent. This phenomenon may be explained by the fewer electron-dense cuticular lamellae (usually three to seven) and the fact that these lamellae anastomose freely to form a thin cuticle with a highly irregular substructure. Elements detected by X-ray analysis, in addition to carbon and oxygen, included Mg, Br, S, and Ca in both gametophyte and sporophyte cuticles. Major features of FTIR spectra of all cuticles were absorbances due to proteins. A strong band, indicative of sulfate ester, occurred near 1250 cm?1 in all cuticle preparations. Gametophyte, but not sporophyte, cuticles absorbed at 935, 846, and 800 cm?1 consistent with the presence of kappa and/or iota carrageenan. Amino acid analyses showed that sporophyte and gametophyte cuticles were generally similar in gross composition. All contained proline as the principal residue together with significant amounts of cysteine, methionine, and lysine. Protein contents calculated from these analyses ranged from 37.6 to 44.4% on a dry weight basis as compared to 51.5–56.7% calculated from total nitrogen values. Up to 75% of the cuticle mass was solubilized by sodium dodecyl sulfate-β-mercaptoethanol. Three similar migrating bands were seen in female and male cuticle extracts on sodium dodecyl sulfate–polyacrylamide gel electrophoresis; however, none of the three weaker bands from sporophyte cuticles comigrated with those from gametophytes. Chloroform-methanol extraction removed < 3.3% of the cuticle mass, suggesting that lipids were minor components.  相似文献

Coalescence of developing sporelings of Chondrus crispus Stackhouse was observed. Juvenile tetra-sporophytes showed a higher proportion of coalescence than developing gametophytes. Stages of complete coalescence between different sporelings are illustrated. Coalesced sporelings exhibit vertical and horizontal alignment of cells, as well as “cuticular” continuity and secondary pit connections between adjacent, coalesced sporelings. Ultimately the cells in the center of the coalesced sporelings produce upright, multiaxial fronds that grow more rapidly than fronds of non-coalesced sporelings. Other red algae, such as Gracilaria verrucosa (Hudson) Papenfuss and Gigartina stellata (Stackhouse) Batters also show a similar sequence of sporeling coalescence and enhanced growth. The ecological significance of sporeling coalescence is discussed.  相似文献

Susceptibility of rhodophycean macroalgae to infection by the green endophytes, Acrochaete operculata Correa & Nielsen and A. heteroclada Correct & Nielsen was studied. Cross-infection experiments showed that A. operculata is host specific and developed only in sporophytic fronds of Chondrus crispus Stackh. and Iridaea cordata (Turn.) Bory. Although A. operculata penetrated equally the multilamellar outer cell wall of sporophytic and gametophytic fronds of C. crispus, subsequent development was arrested in the gametophytic fronds. Susceptibility of the sporophytic phase of C. crispus was detected early in the development of the host, at a discoid stage that is structurally distinct from the adult fronds. The evidence strongly suggests that host specificity in A. operculata is determined by cell-wall composition of the hosts, likely the carrageenan fraction. In contrast, A. heteroclada was not host specific, infecting all offered hosts, including carrageenophytes and agarophytes. Germination occurred on the surface of the hosts and led to the development of an epiphytic stage. Subsequent penetration in many cases involved total displacement of cortical tissue in the infected frond.  相似文献

Morphological variability of intertidal Chondrus crispus Stackh. fronds along a small open rocky coast was related to wave exposure and emersion. Cluster analysis revealed two well-defined morphologies: filiform and planiform, named the N morphotype and B morphotype, respectively. We propose a rapid method of classifying fronds based on the morphology of the cross section at half the height on the thallus. The N morphotype is characterized by fewer dichotomies per unit length, a circular cross section with a large inner cortex, and narrow fronds. It is abundant at low intertidal and exposed sites. The B morphotype is characterized by more dichotomies, smaller sizes, a subelliptical or flattened cross section, and broad fronds. It is abundant at high intertidal sites in sheltered areas. Regression analysis revealed a major effect of water movements on frond morphology with respect to tidal level, which was more evident at high intertidal levels. No relationships were observed between morphology and life history phases.  相似文献

Chondrus crispus Stackh. has been intensely studied, yet no study to date has elucidated its population structure or mating system despite many populations in which there was a haploid bias and lack of male gametophytes. Therefore, 12 nuclear microsatellite loci were identified in this red alga. Microsatellite markers were developed and tested against a panel of specimens collected from two shore levels at two sites in Brittany, France: Pointe de Primel and Pointe de la Jument, Concarneau. Single locus genetic determinism was verified at eight polymorphic loci, as only one band was observed for haploid genotypes, whereas one or two bands were observed for diploids. These markers enabled the detection of unique genotypes within sampled populations, indicating that very few fronds shared the same multilocus genotype. This finding suggests that asexual reproduction was not the prevailing mode of reproduction. In addition, we explored the hierarchical population structure showing that gene flow is restricted at small spatial scales (<50 m) between upper and lower Chondrus‐range populations within a shore. Sexual reproduction predominated in the populations of C. crispus studied, but probably due to fine‐scale spatial substructuring, inbreeding was also significant. In conclusion, this study reveals that fine‐scale genetic variation is of major importance in C. crispus, suggesting that differences between microhabitats should be essential in understanding evolutionary processes in this species.  相似文献

Chondrus crispus Stackhouse alternates between two isomorphic life history phases that differ in cell-wall phycocolloid composition. It has been long hypothesized that the gametophyte, with strong-gelling kappa-type carrageenans, is mechanically superior to the tetrasporophyte, with nongelling lambda-type carrageenans, which could contribute to the observed gametophytic dominance in many wave-swept environments. Standard mechanical tests were performed on distal tissues of C. crispus sampled from a range of environments in Narragansett Bay, Rhode Island, using a tensometer equipped with a video extensometer. Life history phase was by far the most important determinant of mechanical properties, whereas environmental factors had only modest influence (vertical distribution) or no effect (exposure); gametophytic distal tissues were 43% stronger, 21% more extensible, and 21% stiffer than tetrasporophytic distal tissues. However, the superior strength of gametophytic tissues was not evident at the stipe/holdfast junction (where breakage typically occurs), and the two phases were equally susceptible to dislodgment by a given force. The primary ecophysiological role of carrageenans in C. crispus may not be the provision of a structure to resist wave action.  相似文献

Carpospores and tetraspores germinate and the germlings develop similarly. A prostrate holdfast of regular or irregular outline is formed if the spore makes direct contact with the substrate. If not, rhizoids grow from the few-celled germling unilaterally in no apparent relation to light or gravity, forming a prostrate holdfast if they encounter a solid substrate, and, in any case, eventually an erect shoot of different anatomy. Tetrasporophytes have been grown to reproductive maturity from single spore cultures in the laboratory. The ontogeny of the procarp and subsequent postfertilization events are described from field material. Only a single putative male plant was found when plants bearing procarps were numerous at Plymouth. Evidence from this study and from other reported observations places in doubt the expected role of fertilization in the diphasic, cytological alternation of Chondrus.  相似文献

One of the cysteine residues believed to be necessary for reductive light activation is lacking in the only red algal NADP-linked glyceraldehyde-3-P dehydrogenases for which sequences are available, namely Gracilaria verrucosa (Hudson) Papenfuss and Chondrus crispus Stackhouse. Consistent with the mechanism of light modulation proposed for this enzyme, which involves reduction of domain movement-restricting disulfide bonds, it is not reductively activated in Chondrus crispus extracts, and it is not light-activated in whole cells or dithiothreitol (DTT) activated in extracts of the North American species Gracilaria tikvahiae McLachlan. Fructosebisphosphatase and glucose-6-P dehydrogenase, two enzymes for which sequence information from these algae is not yet available, are both activated in crude extracts by DTT treatment, but only fructosebisphosphatase is light-activated in intact Gracilaria.  相似文献

The induction of UV-absorbing compounds known as mycosporine-like amino acids (MAAs) by red, green, blue, and white light (43% ambient radiation greater than 390 nm) was examined in sublittoral Chondrus crispus Stackh. Fresh collections or long-term cultures of sublittoral thalli, collected from Helgoland, North Sea, Germany, and containing no measurable amounts of MAAs, were exposed to filtered natural radiation for up to 40 days. The MAA palythine (λmax 320 nm) was synthesized in thalli in blue light to the same extent observed in control samples in white light. In contrast, thalli in green or red light contained only trace amounts of MAAs. After the growth and synthesis period, the photosynthetic performance of thalli in each treatment, measured as pulse amplitude modulated chlorophyll fluorescence, was assessed after a defined UV dose in the laboratory. Thalli with MAAs were more resistant to UV than those without, and exposure to UV-A+B was more damaging than UV-A in that optimal (Fv/Fm) and effective (φII) quantum yields were lower and a greater proportion of the primary electron acceptor of PSII, Q, became reduced at saturating irradiance. However, blue light-grown thalli were generally more sensitive than white light control samples to UV-A despite having similar amounts of MAAs. The most sensitive thalli were those grown in red light, which had significantly greater reductions in Fv/Fm and φII and greater Q reduction. Growth under UV radiation alone had been shown previously to lead to the synthesis of the MAA shinorine (λmax 334 nm) rather than palythine. In further experiments, we found that preexposure to blue light followed by growth in natural UV-A led to a 7-fold increase in the synthesis of shinorine, compared with growth in UV-A or UV-A+B without blue light pretreatment. We hypothesize that there are two photoreceptors for MAA synthesis in C. crispus, one for blue light and one for UV-A, which can act synergistically. This system would predispose C. crispus to efficiently synthesize UV protective compounds when radiation levels are rising, for example, on a seasonal basis. However, because the UV-B increase associated with artificial ozone reduction will not be accompanied by an increase in blue light, this triggering mechanism will have little additional adaptive value in the face of global change unless a global UV-B increase positively affects water column clarity.  相似文献

The nature and seasonal extent of microbial fouling on Ascophyllum nodosum (L.) LeJol., Fucus vesiculosus L., Polysiphonia lanosa (L.) Tandy and Chondrus crispus Stackh. were observed by scanning electron microscopy. Bacterial filaments and smaller rod and coccoid forms dominated the fouling communities on all species, with pennate diatoms constituting a minor component in contrast to results with plastic substrates on which pennate diatoms dominated and preceded bacterial colonization. The total percent microbial coverage on the surfaces of all four seaweed species was determined by monthly stereological analyses of representative composite micrographs. These showed a simultaneous decline between April and May which could represent the die-off of the cold water bacterial flora when water temperature increased past the threshold for obligate psychrophiles. Microbial colonization patterns were directly correlated (P = 0.005) with maximum coverage in April and November–December and reduced levels from May to October. Significant inverse (P < 0.041) correlations between total percent coverage and water temperature indicate distinct seasonal cycles, however, the patterns of dominance by filamentous bacteria and rod and coccoid forms were markedly different. Total coverage patterns of both rhodophytes showed no apparent seasonal cycle and were not related to water temperature. Rod and coccoid bacteria were apparently suppressed year round on P. lanosa relative to the other species. These interspecific differences in seasonal fouling patterns are discussed in light of possible modes of regulation, especially algal antibiosis.  相似文献

The induction and protective role of the UV-absorbing compounds known as mycosporine-like amino acids (MAAs) were examined in sublittoral Chondrus crispus Stackh. transplanted for 2 weeks in the spring and summer to shallow water under three irradiance conditions: PAR (photosynthetically active radiation; 400–700 nm), PAR + UVA (PAR + 320– 400 nm), PAR + UVA + UVB (PAR + UVA + 280– 320 nm). Sublittoral thalli collected around Helgoland, North Sea, Germany, from 6 m below the mean low water of spring tides contained less than 0.1 mg·g−1 dry weight (DW) total MAAs, whereas eulittoral samples contained over 1 mg·g−1 DW. Transplantation to shallow water led to the immediate synthesis of three MAAs in the following temporal order: shinorine (λmax 334 nm), asterina (λmax 330 nm), and palythine (λmax 320 nm), with the shinorine content peaking and then declining after 2 days (exposure to 100 mol photons·m−2). Maximum total MAA content (2 mg·g−1 DW) also occurred after 2 days of induction, exceeding the content normally found in eulittoral samples. Furthermore, the relative proportion of the different MAAs at this time was different than that in eulittoral samples. After 2 days the total content declined to the eulittoral value, with palythine as the principal MAA. Similar data were obtained for all treatments, indicating that MAA synthesis in C. crispus was induced by PAR and not especially stimulated by UV radiation. The ability of photosystem II (PSII) to resist damage by UVB was tested periodically during the acclimation period by exposing samples to a defined UVB dose in the lab. Changes in chlorophyll fluorescence (Fv/Fm and effective quantum yield, φII) indicated that PSII function was inhibited during the initial stage of acclimation but gradually improved with time. No difference among screening treatments was detected except in spring for the samples acclimating to PAR + UVA + UVB. In this treatment Fv/Fm and φII were significantly lower than in the other treatments. During the first week of each experiment, growth rates were also significantly reduced by UVB. The reductions occurred despite maximum MAA content, indicating an incomplete protection of photosynthetic and growth-related processes.  相似文献

The occurrence of three endophytic green algae within Chondrus crispus Stackh. is reported. Two of them are new to science and are described as Acrochaete heteroclada Correa and Nielsen sp. nov. and A. operculata Correa and Nielsen sp. nov. The algae were studied in unialgal culture and in association with the host following infection of C. crispus under laboratory conditions. The experimental infection showed A. heteroclada to be initially epiphytic, with endophytic filaments growing into the cortex of the host during late infection. A. operculata is entirely endophytic when associated with the host. Phaeophila dendroides (Crouan frat.) Batters behaves as an epiphyte at 15 and 24°C but penetrates the host tissues at 20°C. For all three species, a close physical association with the host is established only when zoospores settle and germinate on the surface of C. crispus.  相似文献

Inferring how the Pleistocene climate oscillations have repopulated the extant population structure of Chondrus crispus Stackh. in the North Atlantic Ocean is important both for our understanding of the glacial episode promoting diversification and for the conservation and development of marine organisms. C. crispus is an ecologically and commercially important red seaweed with broad distributions in the North Atlantic. Here, we employed both partial mtDNA Cox1 and nrDNA internal transcribed spacer region 2 (ITS2) sequences to explore the genetic structure of 17 C. crispus populations from this area. Twenty‐eight and 30 haplotypes were inferred from these two markers, respectively. Analysis of molecular variance (AMOVA) and of the population statistic ΦST not only revealed significant genetic structure within C. crispus populations but also detected significant levels of genetic subdivision among and within populations in the North Atlantic. On the basis of high haplotype diversity and the presence of endemic haplotypes, we postulate that C. crispus had survived in Pleistocene glacial refugia in the northeast Atlantic, such as the English Channel and the northwestern Iberian Peninsula. We also hypothesize that C. crispus from the English Channel refugium repopulated most of northeastern Europe and recolonized northeastern North America in the Late Pleistocene. The observed phylogeographic pattern of C. crispus populations is in agreement with a scenario in which severe Quaternary glaciations influenced the genetic structure of North Atlantic marine organisms with contiguous population expansion and locally restricted gene flow coupled with a transatlantic dispersal in the Late Pleistocene.  相似文献

The existence of a phenomenon in phosphorus (P) nutrition comparable to the “Neish effect” in nitrogen (N) nutrition (an inverse relation between seawater N enrichment and carrageenan content) was investigated in the temperate red alga Chondrus crispus Stackhouse. Plants were preconditioned for 17 d and then cultured under varying enrichments of P (0, 3, 6, 10, 15 μM P·wk?1) and a constant N enrichment (53.5 μM N·wk?1) for 5 wk. Tissue total P, tissue total N, and carrageenan contents were then determined. Identical experiments were performed using C. crispus collected during the fall, winter, spring, and summer seasons. The procedure was repeated using material collected during the following fall season and cultured under constant P (6 μM P·wk?1) and varying N enrichments (0, 3, 6, 10, 25 μM N·wk?1). In the fall (P) experiment, carrageenan content was the highest [53.1 ± 0.3% DW (dry weight)], and tissue total P content was the lowest (1.71 ± 0.27 mg P·g DW?1) in plants that received no P enrichment. Carrageenan content was stable (46.1 ± 1.8% DW) for plants given enrichments of 3 μM P·wk?1 and greater. Thus, a decrease in carrageenan content, concomitant with an increase in tissue total P content, was observed, but only at tissue total P levels below 2 mg P·g DW?1. As these levels were always higher than 2 mg P·g DW?1 in the winter, spring, and summer experiments, carrageenan content remained constant within each season at 46.2 ± 1.3, 43.1 m 0.7, and 44.5 ± 0.6% DW, respectively. Nitrogen enrichment of plants collected in the fall did not affect carrageenan content, which was stable at 49.3 ± 0.9% DW. When these plants were compared with those of the previous fall experiment (6 μM P·wk?1 and 53.5 μM N·wk?1), a slight increase in carrageenan content was noted. Thus, at sufficiently high concentration, N also decreased carrageenan content in C. crispus. Phosphorus nutrition had no significant effect on photosynthesis versus irradiance parameters (Pmax, α, Rd, Ic, and Ik), the contents of the photosynthetic pigments chlorophyll-a, phycoerythrin (PE), phycocyanin (PC), and allophycocyanin (APC), and the ratios PE:APC and PC:APC. In contrast, N nutrition affected both Pmaxand the photosynthetic pigment contents. The data indicate that N limitation reduces the number of phycobilisomes but not their size. The greater reduction in phycobiliprotein than chlorophyll-acontent corroborates the natural bleaching phenomenon regularly observed in C. crispus populations during summer when N levels are generally low in seawater. These results suggest that C. crispus in the temperate waters of the Bay of Fundy may experience N limitation, but P limitation is unlikely.  相似文献

本文研究了光照、pH、温度对菹草光合作用的影响。在一定温度条件下,菹草的净产氧量与一定范围的光照强度呈直线相关。菹草的光补偿点随温度的上升而上升。在菹草自然生活的环境中,温度低于30℃时,升温有利于菹草的光合作用。高pH(PH>10.0)下碳源缺乏对菹草的光合作用影响较大。高pH与强光照射的协同作用严重影响菹草的光合作用。水温与氮、磷营养盐不足并非夏季自然水体中菹草死亡的主要原因。而不良光照(水表层光抑制,中、下层光饥饿)和高pH下缺乏光合碳源的协同作用便可能导致菹草夏季死亡。  相似文献

Little is known about the dynamics and the ecological interactions among ramets (fronds) from populations of clonal red seaweeds. Small ramets are very difficult to tag, so their growth cannot be monitored directly. The temporal variation of the relationship between stand biomass and ramet density offers information on ramet performance. We calculated this relationship for an intertidal population of Pterocladiella capillacea (Gmelin) Santelices et Hommersand (Gelidiales) from Baja California, Mexico. Biomass and density were positively correlated on an annual basis, indicating that biomass accumulated without involving self-thinning among ramets. This contrasts with nonclonal seaweeds, for which self-thinning among individuals occurs during growth, but agrees with other clonal red seaweeds, such as Chondrus crispus Stackhouse and Mazzaella cornucopiae (Postels et Ruprecht) Hommersand (both Gigartinales). The growth pattern for these members of the Gelidiales and of the Gigartinales holds despite differences in holdfast morphology and ramet branching degree and despite differences in the capacity of coalescence during early stages, known only for the Gigartinales. The positive slope for the dynamic biomass–density relationship, on a bilogarithmic scale, was statistically steeper for M. cornucopiae than for P. capillacea and for C. crispus. This suggests that the addition of new ramets during the growth season may be relatively more beneficial for biomass accumulation rates for M. cornucopiae. This would be expected for high-intertidal species subjected to strong abiotic stress, for which ramet crowding constitutes a key protection. Pterocladiella capillacea occurs at the mid-intertidal zone and C. crispus at the subtidal zone, so ramets would be relatively less important in that respect.  相似文献

应用光学显微镜、扫描电镜和透射电镜对安徽大学校内水池中眼子菜科植物菹草的花粉形态进行了观察和研究.结果表明花粉粒球形至近球形,花粉大小为21.0-29.0 μm,平均为24.5 μm.无萌发孔.光学显微镜下,花粉外壁纹饰为网状,外壁厚约4.1μm,两层明显,外层较内层厚.在扫描电镜下花粉表面具粗网状纹饰,网脊窄.在透射电镜下,花粉外壁为三层组成,即覆盖层、柱状层和基层.外壁内层不明显.覆盖层不连续,为半覆盖层;柱状层小柱发达;基层较厚.同时研究了菹草花粉的地理分布及其与生态因子的关系.根据菹草植物赖以生存的生态因子,得出菹草花粉分布区的主要生态因子,包括地理位置、海拔高度、年降水量、年积温及生境,为利用地层中眼子菜科化石花粉重建古气候、古环境及气候变迁提供了现代孢粉学资料和依据.  相似文献

采用rbcL序列分析结合传统形态观察的方法 ,对我国福建省平潭县和漳浦县、山东省青岛市及辽宁省大连市采集的角叉菜属样品进行了研究。新鲜或硅胶干燥样品被用于总DNA提取并用于PCR反应 ,PCR扩增产物回收后直接用于测序反应 ,序列数据经ClustalX1 .8软件排序后 ,采用MEGA2 .1软件根据Kimura’s双因子法计算遗传距离 ,并应用邻接法 (neighbor - joint ing)和最大简约法 (MaximumParsimony)构建系统树 ,Bootstrap法 (5 0 0次重复 )评估分支可信度。研究表明 :确认中国存在Chondrusocellatus、Chondrusnipponicus和Chondrusarmatus三种角叉菜 ;原报道产于我国东南沿海的角叉菜Chondrusocellatus应为Grateloupiaimbricata ;原报道产于大连小平岛的角叉菜Chondrusyendoi应为Mazzaellajaponica。  相似文献
