The central nervous system glutamate receptors are an important target for drug discovery. Herein we report initial investigations into the synthesis and glutamate receptor activity of 1,2,3-triazolyl amino acids. Summary.  Two glutamic acid analogs (1SR,3RS,4RS)- and (1SR,3SR,4SR)-1-amino-4-phosphono cyclopentane-1,3-dicarboxylic acids (APCPD) have been synthesized. Pure E-(diethoxy-phosphoryl)-acrylic acid ethyl ester was obtained from ethyl propiolate, phenol and triethylphosphite. It was used as dienophile in a Diels-Alder reaction. Oxidation and cyclization afforded 3-(ethoxy-carbonyl)-4-(diethoxy-phosphoryl)-cyclopentanone. Bucherer-Bergs reaction and hydrolysis yielded APCPD-III and -IV which are inactive on mGlu1a receptor and antagonists on mGlu2 and mGlu8a receptors. The second-generation asymmetric synthesis of the trans-tris(homoglutamic) acids reported herein proceeds via Strecker reaction of chiral ketimines, obtained from condensation of racemic 2-ethoxycarbonylmethylcyclopentanone and commercially available (S)- and (R)-1-phenylethylamine, respectively. In the key stereodifferentiating step, the cyanide addition leads to mixtures of diastereomeric alpha-amino nitrile-esters, the composition of which is independent of the reaction temperature and the type of the solvent, respectively. Hydrolysis of the alpha-amino nitrile-esters with concentrated H(2)SO(4) yielded diastereomeric mixtures of secondary alpha-amino amido-esters, which after separation were hydrogenolyzed and hydrolyzed each to the enantiomeric trans-1-amino-2-carboxymethylcyclopentanecarboxylic acids. Their configuration was completely established by NMR methods, CD spectra, and X-ray analysis of the trans-1S,2R-configured secondary alpha-amino amido-ester. In receptor binding assays and functional tests, trans-1S,2R-1-amino-2-carboxymethylcyclopentanecarboxylic acid hydrochloride was found to behave as a selective mGluR(2)-antagonist without relevant binding properties at iGluRs.  相似文献

Stomata help plants regulate CO2 absorption and water vapor release in response to various environmental changes, and plants decrease their stomatal apertures and enhance their water status under elevated CO2. Although the bottom‐up effect of elevated CO2 on insect performance has been extensively studied, few reports have considered how insect fitness is altered by elevated CO2‐induced changes in host plant water status. We tested the hypothesis that aphids induce stomatal closure and increase host water potential, which facilitates their passive feeding, and that this induction can be enhanced by elevated CO2. Our results showed that aphid infestation triggered the abscisic acid (ABA) signaling pathway to decrease the stomatal apertures of Medicago truncatula, which consequently decreased leaf transpiration and helped maintain leaf water potential. These effects increased xylem‐feeding time and decreased hemolymph osmolarity, which thereby enhanced phloem‐feeding time and increased aphid abundance. Furthermore, elevated CO2 up‐regulated an ABA‐independent enzyme, carbonic anhydrase, which led to further decrease in stomatal aperture for aphid‐infested plants. Thus, the effects of elevated CO2 and aphid infestation on stomatal closure synergistically improved the water status of the host plant. The results indicate that aphid infestation enhances aphid feeding under ambient CO2 and that this enhancement is increased under elevated CO2.  相似文献

A branched amino acid was synthesized from methyl glucopyranoside; this amino acid presents three amino groups protected by Fmoc and one acid group and can be used in classic peptide synthesis. In parallel, similar azido terminated blocks were synthesized.Successive coupling reaction and deprotection afforded dendrimers with up to 27 azido functional groups. As an example of application, d-mannose and l-fucose residues were linked through CuAAC coupling and resulting glycodendrimers were evaluated in their interaction with DC-SIGN using SPR competition assay.  相似文献

As a key nitrogen (N) source, soil amino acids play an important role in plant N nutrition. However, how amino acids differentially influence the N use strategies of native and invasive plants remains unclear. We performed a potted experiment using five pairs of native and invasive plant congeners, which were subject to 23 N treatments (i.e., 20 protein primary amino acids, nitrate, ammonium, and control), each with 10 replicates. We determined their growth, biomass allocation, N use efficiency, and the growth advantage of plant invaders over their natives. Native and invasive plants used the same 18 amino acid N sources (i.e., a similar amino acid economics spectrum). The growth of plant invaders was invariably better than the growth of native plants, and this superior growth of invaders was linked to their higher root biomass allocation and greater N use efficiency. Additionally, invasive plants had a greater growth advantage on amino acid N than on inorganic N, so was this advantage greater on neutral amino acids than on acidic amino acids. These findings suggest that the differences in amino acid use strategies between invasive and native congeners could help to explain plant invasiveness, as indicated by a growth advantage.  相似文献

Based on a split-and-mix strategy, a library of trimeric Polyamide Amino Acids (PAA) incorporating four different amino acids (Lys, Ala, Arg, and Phe) has been prepared. Screening of the batches for HIV TAR RNA binding in a fluorescent assay allowed the identification of several components that interact with TAR RNA at a micromolar concentration, with a good TAR versus tRNA specificity. Some of these compounds compete efficiently with the association of TAR and Tat protein. In cell cultures, these compounds display a moderate antiviral activity, associated nevertheless with some toxicity. Overall, these results confirm that this new family can be a basis for the design of novel RNA targeting drugs.  相似文献

Stomata play a critical ecological role as an interface between the plant and its environment. Although the guard‐cell pair is highly conserved in land plants, the development and patterning of surrounding epidermal cells follow predictable pathways in different taxa that are increasingly well understood following recent advances in the developmental genetics of the plant epidermis in model taxa. Similarly, other aspects of leaf development and evolution are benefiting from a molecular–genetic approach. Applying this understanding to extinct taxa known only from fossils requires use of extensive comparative morphological data to infer ‘fossil fingerprints’ of developmental evolution (a ‘palaeo‐evo‐devo’ perspective). The seed‐plant order Bennettitales, which flourished through the Mesozoic but became extinct in the Late Cretaceous, displayed a consistent and highly unusual combination of epidermal traits, despite their diverse leaf morphology. Based on morphological evidence (including possession of flower‐like structures), bennettites are widely inferred to be closely related to angiosperms and hence inform our understanding of early angiosperm evolution. Fossil bennettites – even purely vegetative material – can be readily identified by a combination of epidermal features, including distinctive cuticular guard‐cell thickenings, lobed abaxial epidermal cells (‘puzzle cells’), transverse orientation of stomata perpendicular to the leaf axis, and a pair of lateral subsidiary cells adjacent to each guard‐cell pair (termed paracytic stomata). Here, we review these traits and compare them with analogous features in living taxa, aiming to identify homologous – and hence phylogenetically informative – character states and to increase understanding of developmental mechanisms in land plants. We propose a range of models addressing different aspects of the bennettite epidermis. The lobed abaxial epidermal cells indicate adaxial–abaxial leaf polarity and associated differentiated mesophyll that could have optimised photosynthesis. The typical transverse orientation of the stomata probably resulted from leaf expansion similar to that of a broad‐leaved monocot such as Lapageria, but radically different from that of broad‐leafed eudicots such as Arabidopsis. Finally, the developmental origin of the paired lateral subsidiary cells – whether they are mesogene cells derived from the same cell lineage as the guard‐mother cell, as in some eudicots, or perigene cells derived from an adjacent cell lineage, as in grasses – represents an unusually lineage‐specific and well‐characterised developmental trait. We identify a close similarity between the paracytic stomata of Bennettitales and the ‘living fossil’ Gnetum, strongly indicating that (as in Gnetum) the pair of lateral subsidiary cells of bennettites are both mesogene cells. Together, these features allow us to infer development in this diverse and relatively derived lineage that co‐existed with the earliest recognisable angiosperms, and suggest that the use of these characters in phylogeny reconstruction requires revision.  相似文献

Summary Chemical evolution on the primitive earth must have involved the condensation of-amino acids to peptides under a variety of conditions. Subjecting a mixture of methane, ammonia, and water to an electric discharge in the presence of free amino acids yields small peptides. The dehydration-condensation may have taken place via ammonium cyanide, the hydrogen cyanide tetramer, or aminonitriles. We previously reported a combinatorial strategy for designing alpha-helical proteins by assigning only the binary patterning of polar or nonpolar residues [Kamtekar, S., Schiffer, J. M., Xiong, H. Y., Babik, J. M., and Hecht, M. H. (1993) Science 262, 1680-1685]. Here we describe the finding that approximately half of the proteins in the original collection display some level of cooperativity in their thermal denaturation profiles. Many are monomeric in solution, demonstrating that the observed cooperativity is not merely a consequence of oligomerization. These findings demonstrate that although the combinatorial nature of the design strategy precludes explicit design of side-chain packing, binary patterning incorporates sufficient sequence information to generate de novo proteins with cooperatively folded structures. As binary partitioning of polar and nonpolar amino acids is an intrinsic part of the genetic code, these findings may bear on the early evolution of native proteins.  相似文献

The overall morphology of a plant is largely determined by developmental decisions taken within or near the terminally positioned apical meristems of shoots and roots. The spatial separation of these developmental centers emphasizes the need for long-distance signaling. The same signaling events may simultaneously coordinate differentiation within meristems and in the connecting vascular tissues. Recent genetic and molecular analyses not only confirm the proposed role of auxin as a coordinating signal across the plant, but also implicate auxin as a patterning signal in embryo and meristem organization.  相似文献

Protein sequences can be represented as binary patterns of polar (○) and nonpolar (?) amino acids. These binary sequence patterns are categorized into two classes: Class A patterns match the structural repeat of an idealized amphiphilic α-helix (3.6 residues per turn), and class B patterns match the structural repeat of an idealized amphiphilic β-strand (2 residues per turn). The difference between these two classes of sequence patterns has led to a strategy for de novo protein design based on binary patterning of polar and nonpolar amino acids. Here we ask whether similar binary patterning is incorporated in the sequences and structures of natural proteins. Analysis of the Protein Data Bank demonstrates the following. (1) Class A sequence patterns occur considerably more frequently in the sequences of natural proteins than would be expected at random, but class B patterns occur less often than expected. (2) Each pattern is found predominantly in the secondary structure expected from the binary strategy for protein design. Thus, class A patterns are found more frequently in α-helices than in β-strands, and class B patterns are found more frequently in β-strands than in α-helices. (3) Among the α-helices of natural proteins, the most commonly used binary patterns are indeed the class A patterns. (4) Among all β-strands in the database, the most commonly used binary patterns are not the expected class B patterns. (5) However, for solvent-exposed β-strands, the correlation is striking: All β-strands in the database that contain the class B patterns are exposed to solvent. (6) The bias of class A patterns for α-structure over β-structure and the bias of class B patterns for β-structure over α-structure are significant, not merely when compared to other binary patterns of polar (○) and nonpolar (?) amino acids, but also when compared to the full range of sequences in the database. The implications for the design of novel proteins are discussed.  相似文献

Affinity chromatography was used to study an interaction of aspartic proteinases with immobilized aromatic amino acids and their derivatives. The following ligands were used: L-tyrosine, 3-iodo-L-tyrosine, 3,5-diiodo-L-tyrosine, L-phenylalanine, p-iodo-L-phenylalanine and N-acetyl-L-phenylalanine. With the exception of the last one, ligands were coupled directly to divinyl sulfone activated Sepharose 4B. For the preparation of immobilized N-acetyl-L-phenylalanine, divinyl sulfone activated Sepharose 4-B with linked ethylene diamine was used. Porcine pepsin was used for the evaluation of the capacity of the prepared affinity carriers. The capacity of the immobilized amino acid derivatives significantly increased in comparison with the non-derivatized amino acids. The prepared immobilized ligands were further used for the separation of human pepsinogens.  相似文献
