White light inhibits the conversion of 1-amino-cyclopropane-1-carboxylic acid (ACC) in discs of green leaves of tobacco (Nicotiana tabacum L.) and segments of oat (Avena sativa L.) leaves by from 60 to 90%. Etiolated oat leaves do not show this effect. The general nature of the effect is shown by its presence in both a mono- and a dicotyledon. Since the leaves have been grown and pre-incubated in light, yet can produce from 2 to 9 times as much ethylene in the dark as in the light, it follows that the light inhibition is fully reversible. The inhibition by light is about equal to that exerted in the dark by CoCl2; it can be partly reversed by dithiothreitol and completely by mercaptoethanol. Excised wheat (Triticum aestivum L.) leaves, when subjected to drought stress, increased ethylene production as a result of an increased synthesis of 1-aminocyclopropane-1-carboxylic acid (ACC) and an increased activity of the ethyleneforming enzyme (EFE), which catalyzes the conversion of ACC to ethylene. The rise in EFE activity was maximal within 2 h after the stress period, while rehydration to relieve water stress reduced EFE activity within 3 h to levels similar to those in nonstressed tissue. Pretreatment of the leaves with benzyladenine or indole-3-acetic acid prior to water stress caused further increase in ethylene production and in endogenous ACC level. Conversely, pretreatment of wheat leaves with abscisic acid reduced ethylene production to levels produced by nonstressed leaves; this reduction in ethylene production was accompanied by a decrease in ACC content. However, none of these hormone pretreatments significantly affected the EFE level in stressed or nonstressed leaves. Summary Most of the gibberellin activity detectable in extracts of etiolated wheat leaf tissue occurs in a bound form. There is a rapid increase in extractable gibberellin-like substances following exposure of the tissue to red light with a concomitant fall in the amount of bound gibberellin. Actinomycin-D and AMo 1618 do not inhibit this initial phase of red light stimulated gibberellin production.It is concluded that red light stimulated gibberellin production in etiolated wheat leaf tissue is due to release from a bound form and to synthesis.Holder of a Science Research Council Studentship.  相似文献

Homogenates of etiolated pea (Pisum sativum L.) shoots formed ethylene upon incubation with 1-aminocyclopropane-1-carboxylic acid (ACC). In-vitro ethylene formation was not dependent upon prior treatment of the tissue with indole-3-acetic acid. When homogenates were passed through a Sephadex column, the excluded, high-molecular-weight fraction lost much of its ethylene-synthesizing capacity. This activity was largely restored when a heat-stable, low-molecular-weight factor, which was retarded on the Sephadex column, was added back to the high-molecular-weight fraction. The ethylene-synthesizing system appeared to be associated, at least in part, with the particulate fraction of the pea homogenate. Like ethylene synthesis in vivo, cell-free ethylene formation from ACC was oxygen dependent and inhibited by ethylenediamine tetraacetic acid, n-propyl gallate, cyanide, azide, CoCl3, and incubation at 40°C. It was also inhibited by catalase. In-vitro ethylene synthesis could only be saturated at very high ACC concentrations, if at all. Ethylene production in pea homogenates, and perhaps also in intact tissue, may be the result of the action of an enzyme that needs a heat-stable cofactor and has a very low affinity for its substrate, ACC, or it may be the result of a chemical reaction between ACC and the product of an enzyme reaction. Homogenates of etiolated pea shoots also formed ethylene with 2-keto-4-mercaptomethyl butyrate (KMB) as substrate. Of a large number of amino acids examined, changes in glycine were the only ones which were correlated with the ability of dark-grown barley leaves to synthesise protochlorophyllide, δ-aminolaevulinic acid and chlorophyll on exposure to light. A rapid depletion was found in endogenous glycine in barley leaves after day 7. Illumination of the leaves increased the rate of glycine depletion. Glycine concentrations were high throughout the young leaf. The top and middle leaf sections however, which had maximal chlorophyll synthesising potential exhibited the most pronounced decrease in glycine as the leaf aged. Using glycine-[14C] pulse techniques the half life of glycine in 7 and 14-day-old dark-grown leaves was 3.5 and 4.4 min respectively. Light treatment lengthened the half life to 6.9 and 12.1 min in 7 day and 14-day-old-leaves. Sustained illumination continued to decrease glycine turnover.  相似文献

The biosynthetic basis for the high rates of ethylene production by the apical region of etiolated pea (Pisum sativum L.) seedlings was investigated. The ethylene precursor 1-aminocyclopropane-1-carboxylic acid (ACC) was quantified in extracts of various regions of seedlings by measuring isotopic dilution of a 2H-labelled internal standard using selected-ion-monitoring gas chromatography/mass spectrometry. The ACC levels in the apical hook and leaves were much higher than in the expanded internodes of the epicotyl. Shoot-forming tobacco (Nicotiana tabacum var. Wisconsin 38) callus tissues contain significantly lower concentrations of the ethylene precursor 1-aminocyclopropane-1-carboxylic acid compared to non-shoot-forming callus tissues. This difference is evident 1 day after subculture to shoot-forming or non-shoot-forming medium, and is maintained through the first week of growth. The lack of auxin in shoot-forming medium is the probable cause for this difference in ACC concentrations.  相似文献

Application of levulinic acid (LA), a competitive inhibitor of δ-aminolevulinic acid (ALA) dehydratase, to greening plant tissues causes ALA to accumulate at the expense of chlorophyll. 4,6-Dioxoheptanoic acid (DA), which has been reported to be an effective inhibitor of this enzyme in animal systems, has a similar but more powerful effect on ALA and chlorophyll metabolism in greening leaves of Hordeum vulgare L. var. Larker. Both LA and DA also inhibit the uptake of [14C]amino acids into etiolated and greening barley leaves and reduce their incorporation into protein. Treatment of etiolated and greening leaves with these compounds results in the inhibition of 14CO2 evolution from labeled precursors, including amino and organic acids. Inhibition of 14CO2 evolution by these compounds is more effective in greening leaves than in etiolated leaves when [4-14C]ALA or [1-14C]glutamate are employed as precursors. Both LA and DA also inhibit the uptake and increase the incorporation of 32Pi into organophosphorus by etiolated barley leaves. These results indicate that LA and DA have more far-reaching effects upon plant metabolism than was previously believed.  相似文献

The yields of ethylene from endogenous and exogenous 1-aminocyclo-propane-1-carboxylic acid (ACC) in avocado (Persea Americana Mill.) fruit pedicel extracts were very low when assayed by the method of Lizada and Yang (1979 Anal Biochem 100: 140-145). Addition of phenolic compounds, which are present in avocado tissues, to the assay mixture significantly reduced the conversion efficiency of ACC to ethylene. A negative correlation was found between the amount of the plant material in the assay mixture and the conversion efficiency of ACC to ethylene. Removal of phenolic compounds from pedicel extracts by polyvinylpolypyrrolidone, Amberlite XAD-7, and Dowex-50 column chromatography or lead acetate precipitation greatly increased the yields of thylene from ACC in these extracts. The use of polyvinylpolypyrrolidone column chromatography also enabled us to obtain more accurate estimations of endogenous ACC levels in carnation (Dianthus caryophyllus L.) petal extracts. The conversion efficiency of ACC to ethylene could be improved by increasing the concentrations of mercuric chloride and NaOCl in the assay mixture.  相似文献

In order to understand the physiological significance of the in-vitro lipoxygenase (EC 1.13.11.12)-mediated ethylene-forming system (J.F. Bousquet and K.V. Thimann 1984, Proc. Natl. Acad. Sci. USA 81, 1724–1727), its characteristics were compared to those of an in-vivo ethylene-forming system. While oat (Avena sativa L.) leaves, as other plant tissues, preferentially converted only one of the 1-amino-2-ethylcyclopropane-1-carboxylic acid (AEC) isomers to 1-butene, the lipoxygenase system converted all four AEC isomers to 1-butene with nearly equal efficiencies. While the in-vivo ethylene-forming system of oat leaves was saturable with ACC with a Km of 16 M, the lipoxygenase system was not saturated with ACC even at 10 mM. In contrast to the in-vivo results, only 10% of the ACC consumed in the lipoxygenase system was converted to ethylene, indicating that the reaction is not specific for ethylene formation. Increased ACC-dependent ethylene production in oat leaves following pretreatment with linoleic acid has been inferred as evidence of the involvement of lipoxygenase in ethylene production. We found that pretreating oat leaves with linoleic acid resulted in increased ACC uptake and thereby increased ethylene production. A similar effect was observed with oleic acid, which is not a substrate of lipoxygenase. Since linoleic acid hydroperoxide can substitute for lipoxygenase and linoleic acid in this system, it is assumed that the alkoxy radicals generated during the decomposion of linoleic acid hydroperoxide are responsible for the degradation of ACC to ethylene. In excised wheat (Triticum aestivum L.) leaves, water-deficit stress resulted in a rapid increase, followed by a decrease, in ethylene production rates and in the levels of 1-aminocyclopropane-1-carboxylic acid (ACC), the immediate precursor of ethylene. However, the level of N-malonyl-ACC (MACC), the major metabolite of ACC, increased gradually, then leveled off. This increase in MACC was much greater than the decrease in ACC level. The MACC levels were positively correlated with severity of water stress. Once established, the MACC levels did not decrease even after the stressed tissues were rehydrated. Administration of labeled ACC and MACC showed that the conjugation of ACC to MACC was essentially irreversible. Repeated wilting treatments following the first wilting and rehydration cycle resulted in no further increase in ethylene production and in the levels of ACC and MACC. However, when benzyladenine was supplied during the preceding rehydration process, subsequent wilting treatment resulted in a rise in MACC level and a rapid rise followed by a decline in ethylene production rates and in the level of ACC. The magnitude of these increases was, however, smaller in these rewilted tissues than that observed in the first wilting treatment. Since MACC accumulates with water stress and is not appreciably metabolized, the MACC level is a good indicator of the stress history in the detached leaves used.  相似文献

Light inhibits while carbon dioxide enhances the conversion of 1-aminocyclopropane-1-carboxylic acid (ACC) to ethylene in oat ( Avena sativa L. cv. Victory) leaf segments. The possibility that the light inhibition is mediated through changes of carbon dioxide has been investigated. The level of CO2 increases or decreases in the sealed incubation vial in darkness or in light, respectively, which can apparently account for the differences in ACC-dependent ethylene production between the dark and light treatments. However, although the evolution of ethylene from ACC in the dark is reduced upon depletion of CO2, the difference between light and dark is still very noticeable. Moreover, the production of the ethylene in CO2-free air in the dark was still higher than in the light, where the concentration of CO2 was 0.01%. It is proposed that the light effect on the conversion of ACC to ethylene is composed of two distinguishable components: one CO2-mediated and the other CO2-independent.  相似文献

Etiolated barley seedlings lose the ability to produce chlorophyll and soluble protein on exposure to light with increasing age. Similarly, the production of δ-aminolaevulinic acid-dehydratase and succinyl-CoA synthetase is decreased in older etiolated leaves exposed to light. The rate of protochlorophyllide652 regeneration decreased well before the rates of exogenous δ-aminolaevulinic acid conversion to protochlorophyllide632 was affected by ageing. Ethylene production in plant tissues declines rapidly following induction, and this decline is due to a rapid decrease in the activity of 1-aminocyclopropane-1-carboxylic acid (ACC) synthase, a key enzyme in ethylene biosynthesis. To study the nature of the rapid turnover of ACC synthase in vivo, proteins in wounded ripening tomato (Lycopersicon esculentum) fruit discs were radiolabeled with [35S]methionine, followed by a chase with nonradioactive methionine. Periodically, the radioactive ACC synthase was isolated with an immunoaffinity gel and analyzed. ACC synthase protein decayed rapidly in vivo with an apparent half-life of about 58 min. This value for protein turnover in vivo is similar to that previously reported for activity half-life in vivo and substrate-dependent enzyme inactivation in vitro. Carbonylcyanide-m-chlorophenylhydrazone and 2,4-dinitrophenol, potent uncouplers of oxidative phosphorylation, strongly inhibited the rapid decay of ACC synthase protein in the tissue. Degradation of this enzyme protein was moderately inhibited by the administration of aminooxyacetic acid, a competitive inhibitor of ACC synthase with respect to its substrate S-adenosyl-l-methionine, α,α′-dipyridyl, and phenylmethanesulfonyl fluoride or leupeptin, serine protease inhibitors. These results support the notion that the substrate S-adenosyl-l-methionine participates in the rapid inactivation of the enzyme in vivo and suggest that some ATP-dependent processes, such as the ubiquitin-requiring pathway, are involved in the degradation of ACC synthase proteins.  相似文献

Of the different hormones tested, cytokinins stimulated nitrate-induced nitrate reductase (NR) activity in the dark. The optimal stimulation was obtained at 16 hr and this was sensitive to tungstate, 6-methylpurine and cycloheximide. The cytokinin stimulation of NR activity was further enhanced by brief irradiation with red light, but this effect was not noticed when leaves were exposed to far-red light. Both kinetin and red light, when given together, or given with a darkness interruption, stimulated the NR activity more than with either of them alone.  相似文献

1-Aminocyclopropane-1-carboxylic acid (ACC) is aerobically oxidized in plant tissues to form ethylene by ethylene-forming enzyme (EFE). The effect of substrate (ACC and oxygen) concentrations on ethylene production rate by plant tissues was investigated. The Km value for O2 in ethylene production varied greatly depending on the internal ACC content. When ACC levels in the tissue were low (below its Km value), the concentration of O2 giving half-maximal ethylene production rate ([S]0.5) ranged between 5 and 7%, and was similar among different tissues. As the concentration of ACC was increased (greater than its Km value), [S]0.5 for O2 decreased markedly. In contrast, the Km value for ACC was not much dependent on O2 concentration, but varied greatly among different plant tissues, ranging from 8 micromolar in apple (Malus sylvestris Mill.) tissue to 120 micromolar in etiolated wheat (Triticum aestivum) leaf. Such a great variation was thought to be due to the different compartmentation of ACC within the cells in different tissues. These kinetic data are consistent with the view that EFE follows an ordered binding mechanism in which EFE binds first to O2 and then to ACC.  相似文献

It has been proposed that 1-aminocyclopropane-1-carboxylic acid (ACC) oxidase catalyzes the oxidation of ACC to ethylene via N-hydroxyl-ACC as an intermediate. However, due to its chemical instability the putative intermediate has never been isolated. Here, we have shown that a purified recombinant ACC oxidase can utilize alpha-aminoisobutyric acid (AIB), an analog of ACC, as an alternative substrate, converting AIB into CO2, acetone, and ammonia. We chemically synthesized the putative intermediate compound, N-hydroxyl-AIB (HAIB), and tested whether it serves as an intermediate in the oxidation of AIB. When [1-(14)C]AIB was incubated with ACC oxidase in the presence of excess unlabeled HAIB as a trap, no labeled HAIB was detected. By comparing the acetone production rates employing HAIB and AIB as substrates, the conversion of HAIB to acetone was found to be much slower than that of using AIB as substrate. Based on these observations, we conclude that ACC oxidase does not catalyze via the N-hydroxylation of its amino acid substrate. ACC oxidase also catalyzes the oxidation of other amino acids, with preference for the D-enantiomers, indicating a stereoselectivity of the enzyme.  相似文献

In Cymbidium flowers, emasculation by removal of the pollinia and the anther cap leads within 24 hours to red coloration of the labellum (lip). Lip coloration, being the first sign of senescence in these flowers, has been ascribed to the action of ethylene in the lip. When a small incision in the base of the lip is made prior to emasculation, or when the lip is excised and placed in water within 10 to 15 hours after emasculation, coloration is considerably delayed. This indicates that a coloration-associated factor is moving in or out of the lip. Measurements of ethylene production of excised flower parts, isolated at different times after emasculation, showed an increase only in the central column; the other flower parts, including the lip, did not show a measurable change. In contrast, in situ measurements of the ethylene production of the central column and the remaining portion of the flower revealed a simultaneous increase in all the flower parts following emasculation. Similarly, application of radiolabeled 1-aminocyclopropane-1-carboxylic acid (ACC) to the top of the central column in situ leads to the production of radiolabeled ethylene by all the flower parts. In addition, the ethylene production of isolated lips, measured immediately after excision, was initially high but ceased within 10 to 15 minutes. Treatment of the central column in situ with ethylene or ethephon did not stimulate ACC production but did stimulate lip coloration and this was accompanied by an increased internal ethylene concentration in the lip. The data indicate that endogenously produced as well as applied ACC is rapidly translocated from the site of production or application to all the other flower parts where it is immediately converted into ethylene. By excision of a flower organ, the influx of ACC is prevented, causing a rapid decrease in ethylene production. In addition, it was found that ethylene may also be translocated in physiologically significant amounts within the flower. The roles of ACC and ethylene as mobile senescence or wilting factors in emasculation- and pollination-induced senescence is discussed.  相似文献

1-Aminocyclopropane-1-carboxylic Acid (ACC), the immediate precursor of ethylene is routinely assayed by converting it into ethylene with NaOCl, and the ethylene liberated is then determined by gas chromatography (MCC Lizada, SF Yang 1979 Anal Biochem 100: 140-145). However, certain materials which may be present in crude plant extracts or in enzyme reaction mixtures interfere with this assay procedure. Mono, and di-alkyl amines cause poor yields of ethylene from ACC. Ethanol in the presence of NH3 or amines but in the absence of ACC can produce ethylene under the assay procedure. The characteristics of these interfering reactions were studied and precautions to avoid these problems are suggested. Recovery of ACC during its extraction and purification from plant extracts were tested and are discussed.  相似文献
