Cell-free extracts of 3–4 days old mats of nitrate-grown Penicillium citrinum catalyze the hydrolytic cleavage of the N-glycosidic bonds of inosine, guanosine and adenosine optimally at pH 4, 0.1 M citrate buffer. The same extracts catalyze the hydrolytic deamination of cytidine at a maximum rate in 0.08 M Tris-acetate buffer pH 6.5, 40°C and 50°C were the most suitable degrees for purine nucleoside hydrolysis and cytidine deamination, respectively. The incubation of the extracts at 60°C, in the absence of cytidine caused a loss in the deaminating activity, while freezing and thawing had no effect on both activities. The deaminating activity seems to be cytidine specific as neither cytosine, adenine, adenosine nor guanosine could be deaminated. Uridine competively inhibited this activity, while ammonia had no effect. The apparent Km value of this enzyme for cytidine was 1.57×10?3M and its Ki value for uridine was 7.8×10?3M. The apparent Km values of the N-glycosidic bond cleaving enzyme for inosine, guanosine and adenosine were 13.3, 14.2 and 20×10?3 M, respectively.  相似文献

The reactions of PtCl2en or cis-Pt(NH3)2Cl2 and their aqua species with adenine and adenosine were studied by means of ion-pair HPLC. From the chromatograms, it was found that the first binding site of Pt(II) was the N(7) site of adenine under both acidic and neutral conditions. The rates of Pt(II) binding at the (N7) site of adenosine and deoxyadenosine were measured. The rate constants, k1, were obtained for the reactions of PtCl2en or cis-Pt(NH3)2Cl2 with adenosine and deoxyadenosine at pH 3 and 7 over the temperature range 9–25 °C. The k1 values were 6.8–7.7 × 10−4 dm3 mol−1 s−1 at 25 °C. For the aqua species, the rate of [cis-Pt(NH3)2ClH2O]+ with adenosine N(7) was measured. The rate constants, k2 which were found to be smaller than those of hydrolysis, kh, were calculated at pH 3 over the temperature range 25–40 °C. The k2 value obtained at 25 °C was 1.1 × 10−2 dm3 mol−1 s−1, 15 time larger than k1. The activation parameters were also calculated.  相似文献

We have used inelastic laser light scattering to study the kinetics of the spontaneous assembly of heads and tails of bacteriophage T4D to form noninfectious tail fiberless particles. For interpretation of the kinetics, it was first necessary to determine the physical properties of the strongly scattering phage parts. For heads, these are D20,w = 3.60 × 10−8cm2/s, 820,w = 1025 S, M = 1.76 × 108. For tail fiberless particles, D20,w = 3.14 × 10−8cm2/s, 820,w = 968 S, and M = 1.95 × 108. The kinetics of the head-tail joining process was followed by measuring the time variation of the homodyne scattering autocorrelation function. This was interpreted as a sum of exponentials whose decay constants were known from the scattering angle and the diffusion coefficients, and whose amplitudes were related to the concentrations of reactants and products. Scattering experiments at 22 °C gave a bimolecular rate constant of 1.02 × 107m−1 s−1, while infectivity assays at 30 °C gave a rate constant of 1.28 × 107. Adjustment of both rate constants to 20 °C, assuming diffusion controlled reaction, gave 0.97 × 107 and 0.98 × 107m−1 s−1, respectively. This rate is about 1500 that predicted by Smoluchowski theory for a diffusion controlled reaction between two spherical particles; the discrepancy is largely explicable from orientational factors.  相似文献

Analkali tolerant α-l-rhamnosidase has been purified to homogeneity from the culture filtrate of a new fungal strain, Fusarium moniliforme MTCC-2088, using concentration by ultrafiltration and cation exchange chromatography on CM cellulose column. The molecular mass of the purified enzyme has been found to be 36.0 kDa using SDS-PAGE analysis. The Km value using p-nitrophenyl-α-l-rhamnopyranoside as the variable substrate in 0.2 M sodium phosphate buffer pH10.5 at50 °C was 0.50 mM. The catalytic rate constant was15.6 s−1giving the values of kcat/Km is 3.12 × 104M−1 s−1. The pH and temperature optima of the enzyme were 10.5 and 50 °C, respectively. The purified enzyme had better stability at 10 °C in basic pH medium. The enzyme derhamnosylated natural glycosides like naringin to prunin, rutin to isoquercitrin and hesperidin to hesperetin glucoside. The purified α-l-rhamnosidase has potential for enhancement of wine aroma.  相似文献

  • 1.1. The diffusional water permeability (Pd) of rabbit red blood cell (RBC) membrane has been monitored by a doping nuclear magnetic resonance (NMR) technique on control cells and following inhibition with p-chloromercuribenzene sulfonate (PCMBS).
  • 2.2. The values of Pd were around 6.3 × 10−3 cm/sec at 15°C, 7.0 × 10−3cm/sec at 20°C, 8.0 × 10−3 cm/sec at 25°C, 9.1 × 10−3 cm/sec at 30°C and10.7 × 10−3 cm/sec at 37°C.
  • 3.3. Systematic studies on the effects of PCMBS on water diffusion indicated that the maximal inhibition was reached in 15 min at 37°C with 0.5 mM PCMBS.
  • 4.4. The values of maximal inhibition were around 71–74% at all temperatures.

The α-carbonic anhydrase gene from Helicobacter pylori strain 26695 has been cloned and sequenced. The full-length protein appears to be toxic to Escherichia coli, so we prepared a modified form of the gene lacking a part that presumably encodes a cleavable signal peptide. This truncated gene could be expressed in E. coli yielding an active enzyme comprising 229 amino acid residues. The amino acid sequence shows 36% identity with that of the enzyme from Neisseria gonorrhoeae and 28% with that of human carbonic anhydrase II. The H. pylori enzyme was purified by sulfonamide affinity chromatography and its circular dichroism spectrum and denaturation profile in guanidine hydrochloride have been measured. Kinetic parameters for CO2 hydration catalyzed by the H. pylori enzyme at pH 8.9 and 25°C are kcat=2.4×105 s−1, KM=17 mM and kcat/KM=1.4×107 M−1 s−1. The pH dependence of kcat/KM fits with a simple titration curve with pKa=7.5. Thiocyanate yields an uncompetitive inhibition pattern at pH 9 indicating that the maximal rate of CO2 hydration is limited by proton transfer between a zinc-bound water molecule and the reaction medium in analogy to other forms of the enzyme. The 4-nitrophenyl acetate hydrolase activity of the H. pylori enzyme is quite low with an apparent catalytic second-order rate constant, kenz, of 24 M−1 s−1 at pH 8.8 and 25°C. However, with 2-nitrophenyl acetate as substrate a kenz value of 665 M−1 s−1 was obtained under similar conditions.  相似文献

The irreversible thermal denaturation of cytochrome cd1 oxidase from P.aeruginosa as a function of the oxidation-reduction states of its hemes was observed with a differential scanning calorimeter. Upon full reduction of the four hemes, the apparent denaturation temperature decreases by about 10° and the denaturation enthalpy decreases slightly: oxidized, 5.9 cal/gm; reduced, 5.4 cal/gm. At pH 7.5, the first order rate constants for denaturation at 90°C are: reduced, 33 × 10?3s?1; oxidized, 3 × 10?3s?1. Thus, oxidation of the hemes reuults in heat stabilization of the cytochrome oxidase. The activation energy for denaturation of fully reduced oxidase, 53 kcal/mol, is less than that for fully oxidized protein (73 kcal/mol).  相似文献

The title complex undergoes decomposition in acidic aqueous solution resulting in equimolar concentration of aquapentaamminecobalt(III) and hexa- aquacobalt(II). The kinetic studies over the ranges of 0.048 M ⩽ [H+] ⩽ 0.385 M, 25 ⩽ θc ⩽ 41.5°C and at I = 0.5 M reveals that the intricate mechanism involves protonation equilibrium of the title complex, followed by a rate determining bridge cleavage. The further follow-up reaction is a fast electron transfer process to form products. The rate expression derived from the mechanism is kobs = k1K1[H+]/(1 + K1[H+]) where the values of k, and K, are found to be 8.9 × 10−4 s−1 and 3.5 M−1 respectively at 25 °C. The results are compared with that obtained for the decomposition reactions of mononuclear aquaammine complexes of cobalt(III).  相似文献

At pH 8.0 and 25°C α1-proteinase inhibitor and α2-macroglobulin bind human pancreatic elastase with rate constants of 4.7·105 M−1·s−1 and 6.4·106 M−1·s−1, respectively. The corresponding delay times of elastase inhibition in plasma are 0.4 s and 0.2 s, respectively, indicating that both inhibitors may act as physiological antielastases. Elastin impairs the elastase inhibitory capacity of α1-proteinase inhibitor and α2-macroglobulin. In presence of human elastin, the former behaves like a slow-binding elastase inhibitor, with a rate constant of about 260 M−1·s−1. In contrast, α2-macroglobulin is a fast-binding inhibitor of elastin-bound elastase, but only one of its two sites is functioning in presence of elastin.  相似文献

A kinetic study of the oxidation of (hydroxyethyl)ferrocene (HEF) by [2-pyridylmethylbis(2-ethyl-thioethyl)ainine]copper(II) (Cu(pmas)2+) is reported, with the objective of documenting the influence of the two thioether sulfur ligands on the electron transfer rate. Both reactants exhibit a first-order dependence at pH 6, I = 0.1 M(NaNO3); k(25°C) = 1.3 × 104M−1sec−1, ΔH3 = 10.1 kcal/mole, ΔS3 = −6 eu. The apparent Cu(pmas)2+/+ self-exchange electron transfer rate constant calculated from this reaction on the basis of relative Marcus theory (4.7 × 101M−1 sec−1) agrees well with previous findings on ferrocytochrome c, Fe(CN)64−, and Ru(NH3)5py2+ oxidations. Spectrophotometric titrations of Cu(pmas)2+ and Cu(tmpa)2+ (tmpa = tris(2-pyridylmethyl)amine) with azide ion showed that both Cu(pmas)N3)+ (Kf1 = 3.1 × 103M−1) and Cu(pmas)(N3)2 (Kf2 = 3.5 × 101M−1) but Cu(tmpa)(N3)+ (Kf = 6.6 × 102M−1) are formed up to 0.15 M N3 (25°C, pH 6, I = 0.2 M), suggesting that a thioether sulfur atom is displaced in the uptake of a second N3 ion by Cu(pmas)(N3)+. The effect of thioether sulfur displacement by azide ion on the HEF-Cu(pmas)2+ reaction rate may be understood entirely through the tendency of N3 to shift the position of the redox equilibrium towards the reactant side, without invoking any special role for the sulfur ligand in promoting electron transfer reactivity.  相似文献

All-inorganic n-i-p perovskite solar cells (PSCs) using doped Spiro-OMeTAD as hole transport material (HTM) suffer from photothermal stability due to ionic diffusion and radical-induced degradation by the dopants. In this article, dopant-free starlike molecule (N2, N2-bis(4-(bis(4-methoxyphenyl)amino)phenyl)-N5,N5-bis(4-methoxyphenyl)pyridine-2,5-diamine (BD)) is synthesized to engineer the stacking properties and delivered higher hole mobility than doped Spiro-OMeTAD (3.2 × 10−4 versus 1.76 × 10−4 cm2 V−1 s−1) as dopant-free HTM. Starlike BD HTM has a twisted acceptor unit and strong dipole, forming crystalline and ordered packing film to ensure intramolecular charge transfer and improve mobility. The BD CsPbI3 PSCs deliver the maximum efficiency of 19.19%, which is the highest performance for all-inorganic PSCs based on dopant-free HTMs. Meanwhile, the ordered molecules-packing blocks the migration channel of I ions to metal electrodes and improves the device stability. BD-based devices maintain more than 93% and 80% of the initial efficiency after 85 °C storage for 35 days and maximum power point (MPP) tracking at 85 °C for 1000 h, respectively.  相似文献

The hydrolysis of the ester 2,4-dinitrophenyl- ethyl methylphosphonate has been examined by both stop-flow spectrophotometric and pH-stat techniques. These reactions have been carried out in the presence of several nucleophiles including simple non-labile (w.r.t. substitution) mono-aquo metal ion complexes. Comparison of reaction rates of the metal complexes with sterically hindered organic nucleophiles has led to the conclusion that the metal ions function predominantly as general base catalysts in dilute aqueous solution. Reaction rates for the various nucleophiles studied are tabulated together with solvolysis constants for hydroxide ion and water at 35 °C and I=0.1 mol dm−3 (KNO3). These later two values are respectively 32.7 mol−1 dm3 s−1 and 1.37 x 10−4 s−1. A Brönsted β value of 0.52 for the phosphonate ester studied has also been derived.  相似文献

The present study investigates the interaction of the second generation photosensitizer Foscan® with plasma albumin and lipoproteins. Spectroscopic studies indicated the presence of monomeric and aggregated Foscan® species upon addition to plasma protein solutions. Kinetics of Foscan® disaggregation in albumin-enriched solutions were very sensitive to the protein concentration and incubation temperature. Kinetic analysis demonstrated that two types of Foscan® aggregated species could be involved in disaggregation: dimers with a rate constant of k1 = (2.30 ± 0.15) × 10−3 s−1 and higher aggregates with rate constants varying from (0.55 ± 0.04) × 10−3 s−1 for the lowest to the (0.17 ± 0.02) × 10−3 s−1 for the highest albumin concentration. Disaggregation considerably increased with the temperature rise from 15 °C to 37 °C. Compared to albumin, Foscan® disaggregation kinetics in the presence of lipoproteins displayed poorer dependency on lipoprotein concentrations and smaller variations in disaggregation rate constants. Gel-filtration chromatography analysis of Foscan® in albumin solutions demonstrated the presence of aggregated fraction of free, non-bound to protein Foscan® and monomeric Foscan®, bound to protein.  相似文献

Frequent off-flavor events caused by geosmin and 2-methylisoborneol (MIB) have attracted research on the main producers, cyanobacteria. This study evaluated the effects of light and temperature on the odor production of MIB-producing Pseudanabaena sp. Lauterborn and geosmin-producing Anabaena ucrainica (Schhorb.) Watanabe. The maximum MIB production and lowest growth rate (indicated by the chlorophyll a (Chl a)) were observed at 35 °C compared with that at 10 °C and 25 °C. Cultures grown under a light intensity of 60 μmol photons m−2 s−1 demonstrated the highest MIB production and minimum growth rate, whereas the minimum MIB production and maximum growth rate were obtained under 10 μmol photons m−2 s−1. Similar patterns were observed for geosmin production. A. ucrainica had the highest geosmin production and lowest Chl a concentration under 10 °C and 60 μmol photons m−2 s−1. Moreover, greater proportions of geosmin and MIB were released into extracellular under growth-inhibiting temperatures and light intensities. An inverse correlation between odor production and the cell growth rate was suggested, and this relationship may reflect the competition for substrates of odor and Chl a synthesis. Thus, the accumulation of geosmin and MIB was probably the result of decreased cellular metabolic activity in cyanobacteria.  相似文献

The kinetics of rapid CO substitution by PPh3 in Co4(CO)12 and Rh4(CO)12 have been examined by stopped-flow and low temperature FT-IR methods. In Co4(CO)12 rapid (kobs ∼ 1.8 s−1) substitution of CO occurs after a 1–15 s induction period at 28 °C in C6H5Cl solvent by a catalytic process. Addition of PPh3 to Rh4(CO)12 yields Rh4(CO)11(PPh3) according to a predominantly second order rate law k1[Rh4- (CO)12] + k2[Rh4(CO)12][PPh3] with k1 = 25 ± 11 s−1 and k2 = 2.97 ± 0.27 X 104 M−1 s−1 at 28 °C. Substitution of a second CO ligand also occurs rapidly with k1 = 0.15 ± 0.09 s−1 and k2 = 6.54 ± 0.07 X 102 M−1 s−1 at 28 °C. The reactivity of Rh4(CO)12 toward associative substitution is 104– 1011 faster than for the Co and Ir analogues, In Rh4(CO)11(PPh3) the increase in CO substitution rates over Co and Rh analogues is 102–107. The ordering of associative substitution rates Co << Rh >>> Ir in these clusters exaggerates the trend seen in mononuclear metal complexes.  相似文献

The activation parameters were evaluated, for X = Cl: ΔH2 = 106.3 ± 0.4 kJ mol−1 and ΔS2 = 40.2 ± 1.7 J K−1 mol, and for X = Br: ΔH2 = 91.6 ± 0.4 kJ mol−1 and ΔS2 = 0.4 ± 1.7 J K−1 mol−1. In comparison with myoglobin molecule as a reference, we have studied the autoxidation rate of human oxyhemoglobin (HbO2) as a function of its concentration in 0.1 M buffer at 35°C and in the presence of 1 mM EDTA. At pH 6.5, HbA showed a biphasic autoxidation reaction that can be described completely by a first-order rate equation containing two rate constants — kf, for fast autoxidation of the α-chain, and ks, for slow autoxidation of the β-chain, respectively. When tetrameric HbO2 was dissociated into αβ-dimers by dilution, the value of kf increased markedly to an extent comparable with the autoxidation rate of horse heart oxymyoglobin (MbO2). The rate constant ks, on the other hand, was found to remain at an almost constant value over the whole concentration range from 1.0 × 10−3 M to 3.2 × 10−6 M in heme. At pH 8.5 and pH 10.0, however, the autoxidation of HbO2 was monophasic, and no enhancement in the rate was observed by diluting hemoglobin solutions. Taking into consideration the effects of 2,3-diphosphoglyceric acid and chloride anion on the autoxidation rate of HbO2, we have characterized the differential susceptibility of the α- and β-chains to the autoxidation reaction in aqueous solution.  相似文献

Activated white cells use oxidants generated by the heme enzyme myeloperoxidase to kill invading pathogens. This enzyme utilizes H2O2 and Cl, Br, or SCN to generate the oxidants HOCl, HOBr, and HOSCN, respectively. Whereas controlled production of these species is vital in maintaining good health, their uncontrolled or inappropriate formation (as occurs at sites of inflammation) can cause host tissue damage that has been associated with multiple inflammatory pathologies including cardiovascular diseases and cancer. Previous studies have reported that sulfur-containing species are major targets for HOCl but as the reactions are fast the only physiologically relevant kinetic data available have been extrapolated from data measured at high pH (>10). In this study these values have been determined at pH 7.4 using a newly developed competition kinetic approach that employs a fluorescently tagged methionine derivative as the competitive substrate (k(HOCl + Fmoc-Met), 1.5×108 M−1 s−1). This assay was validated using the known k(HOCl + NADH) value and has allowed revised k values for the reactions of HOCl with Cys, N-acetylcysteine, and glutathione to be determined as 3.6×108, 2.9×107, and 1.24×108 M−1 s−1, respectively. Similar experiments with methionine derivatives yielded k values of 3.4×107 M−1 s−1 for Met and 1.7×108 M−1 s−1 for N-acetylmethionine. The k values determined here for the reaction of HOCl with thiols are up to 10-fold higher than those previously determined and further emphasize the critical importance of reactions of HOCl with thiol targets in biological systems.  相似文献

The reactions of NO2 with both oxidized and reduced cytochrome c at pH 7.2 and 7.4, respectively, and with N-acetyltyrosine amide and N-acetyltryptophan amide at pH 7.3 were studied by pulse radiolysis at 23 °C. NO2 oxidizes N-acetyltyrosine amide and N-acetyltryptophan amide with rate constants of (3.1±0.3)×105 and (1.1±0.1)×106 M−1 s−1, respectively. With iron(III)cytochrome c, the reaction involves only its amino acids, because no changes in the visible spectrum of cytochrome c are observed. The second-order rate constant is (5.8±0.7)×106 M−1 s−1 at pH 7.2. NO2 oxidizes iron(II)cytochrome c with a second-order rate constant of (6.6±0.5)×107 M−1 s−1 at pH 7.4; formation of iron(III)cytochrome c is quantitative. Based on these rate constants, we propose that the reaction with iron(II)cytochrome c proceeds via a mechanism in which 90% of NO2 oxidizes the iron center directly—most probably via reaction at the solvent-accessible heme edge—whereas 10% oxidizes the amino acid residues to the corresponding radicals, which, in turn, oxidize iron(II). Iron(II)cytochrome c is also oxidized by peroxynitrite in the presence of CO2 to iron(III)cytochrome c, with a yield of ~60% relative to peroxynitrite. Our results indicate that, in vivo, NO2 will attack preferentially the reduced form of cytochrome c; protein damage is expected to be marginal, the consequence of formation of amino acid radicals on iron(III)cytochrome c.  相似文献
