Responses in flounder (Platichthys flesus) towards benzo [a]pyrene (BaP), 2,3,3',4,4',5-hexachlorobiphenyl (PCB-156), and cadmium (Cd) were investigated in time-course and dose-response studies of selected biomarkers. Measurements of biliary fluorescent BaP metabolites and hepatic concentrations of PCB-156 and cadmium showed that the injected toxicants were rapidly m obilized from the muscle to the liver, but a depot effect was indicated in the highest dose groups of BaP and PCB-156 (12 mg kg-1 bodyweight). Clearest biomarker responses were found in the induction of hepatic cytochrome P450 1A (CYP1A) enzymes as a response towards BaP and PCB-156 exposure. Maximum induction of CYP1A dependent 7-ethoxyresorufin O-deethylase (EROD) activity was observed after 2 and 8 days in BaP and PCB-156-treated flounder, respectively. Positive dose-effect relationships were observed towards both compounds, but the CYP1A induction was more persistent with PCB exposure than with BaP exposure. In Cd-exposed fish, the hepatic level of metallothionein responded more slowly with highest levels observed after 16 days in the time-study. In the combined BaP + Cd treatment, the CYP1A induction was only slightly suppressed. Aspartate aminotransferase in serum appeared to be responsive towards BaP, but also towards the acetone vehicle in controls in the first part of the exposure period. Hematocrit as well as hepatic activities of aldrin epoxidase, glutathione S-transferase, and UDP-glucuronyl transferase were not responsive to any treatm ent in the present study. In general, the results demonstrate that selected biom arkers in flounder are responsive to PAH, PCB, and heavy metal pollutant exposure, indicating the applicability of this species in future environmental pollution monitoring programmes.  相似文献

Pesticides such as chlorpyrifos (CPF) and metals such as copper can impair swimming behavior in fish. However, the impact to swimming behavior from exposure to mixtures of neurotoxicants has received little attention. In the current study, we analyzed spontaneous swimming rates of adult zebrafish (Danio rerio) to investigate in vivo mixture interactions involving two chemical classes. Zebrafish were exposed to the neurotoxicants copper chloride (CuCl, 0.1 μM, 0.25 μM, 0.6 μM, or 6.3, 16, 40 ppb), chlorpyrifos (CPF, 0.1 μM, 0.25 μM, 0.6 μM, or 35, 88, 220 ppb) and binary mixtures for 24 h to better understand the effects of Cu on CPF neurotoxicity. Exposure to CPF increased the number of animals undergoing freeze responses (an anti-predator behavior) and, at the highest CPF dose (0.6 μM), elicited a decrease in zebrafish swimming rates. Interestingly, the addition of Cu caused a reduction in the number of zebrafish in the CPF exposure groups undergoing freeze responses. There was no evidence of additive or synergistic toxicity between Cu and CPF. Although muscle AChE activity was significantly reduced by CPF, there was a relatively poor relationship among muscle AChE concentrations and swimming behavior, suggesting non-muscle AChE mechanisms in the loss of swimming behavior. In summary, we have observed a modulating effect of Cu on CPF swimming impairment that appears to involve both AChE and non-AChE mechanisms. Our study supports the utility of zebrafish in understanding chemical mixture interactions and neurobehavioral injury.  相似文献

Treatment with the herbicide glyphosate led to significantly increased activities of the enzyme gluiathione S-transferase (GST, EC 2.5.1.18) in wheat ( Triticum aestivum L. cv. Kadett and cv. Satu), pea ( Pisum sativum L. ev. Debreceni Világoszöld) and in maize ( Zea mays. L. Pioneer 3839 hybrid) tissues. GST activities in wheat seedlings (cv. Kadett) exposed to 960 μM glyphosate for 4 days were ca 6-fold and 3-fold higher in shoots and roots, respectively, than in the controls. Glyphosate increased the GST activity to a lesser extent in pea and maize than in wheat. In wheat seedlings (cv. Satu) exposed let 120 μM glyphosate gradual increases in the content of non-protein thiols were observed. After 7 days exposure to glyphosate the thiol levels rose to about 360% and 220% of the controls in wheal shoots and roots, respectively. The elevation of thiol content in glyphosate-treated plants was shown to be primarily due increases of glutathione level. These results suggest that the enhanced glutathione metabolism may have a role in the mode of action or degradation of this herbicide.  相似文献

The potential utility of antioxidant enzymes and lipid peroxidation as indicators of exposure to 2,4-D and azinphosmethyl together with the toxic effects of these compounds in freshwater fish Cyprinus carpio were evaluated. Biochemical parameters were recorded spectrophotometrically in fish liver, which were exposed to a single dose of 2,4-D and azinphosmehtyl (1/3 LC(50)), and their mixture at 1:1 ratio for 24, 48, 72, and 96 h. The most sensitive parameter was glutathione S-transferase (GST) activity, which significantly increased with experimental exposures. Glucose 6-phosphate dehydrogenase activity did not change after 24 and 48 h while there was an elevation after 72 h in all exposure groups. The activity decreased only when these were applied in combination at 96 h. Superoxide dismutase activity increased after azinphosmethyl exposure for 48 and 96 h. 2,4-D decreased the activity after 24 h while the activity remained at the same level with control after 48 h. An elevation was found between 72 and 96 h. Mixture treatment did not changed the activity. Glutathione reductase and catalase enzyme activities, and malondialdehyde levels remained constant in all the treatment groups compared with controls. These results suggest that induction of GST activity may be used as biomarker for the assessment of water pollution in C. carpio.  相似文献

The relationship between a putative metallothionein gene (MT) and exposure to cadmium (Cd) in blood cockles (Anadara granosa) is reported. In a 96-h dose-response experiment, mortality of cockles was found to proportionately increase in the range of 0.2-5.0 mg/l Cd with a calculated LC(50) of 2.94 mg/l. Exposure to 0.25 mg/l Cd for 16 days caused significant increases (P<0.05) in Cd concentrations in whole tissues, gills and hepatopancreas, and the accumulation of Cd in these tissues increased with the duration of exposure. Two cDNA libraries constructed using the hepatopancreas from control and Cd-treated cockles gave titres of 5.62 x 10(5) and 1.94 x 10(5) pfu/microg vector, respectively. A putative MT gene, AnaMT, of 510 nucleotides in length, was isolated from the treated cDNA library using a heterologous probe MT20 from the blue mussel, Mytilus edulis. Northern analyses using AnaMT as a probe indicated low expression of the MT mRNA in control animals. In cockles treated with 0.25 mg/l Cd for 4 days, MT mRNA level increased to approximately 168%, but declined to 108% at day 8. After 12 and 16 days of Cd treatment, expression of the MT gene was 138% and 187%, respectively, compared to the controls. These observations suggest that induction of the MT gene by a sublethal dose of Cd is rapid, occurring within 4 days of treatment.  相似文献

We synthesized information on freshwater fish biodiversity in the Yangtze River basin. We documented 361 species and subspecies that had been recorded and described from the basin. Of these, 177 species are endemic. The basin is usually divided into three parts, i.e. the upper reaches, the middle reaches and the lower reaches. This study indicated that the three reaches approach was not supported by fish distribution patterns. Hydrological alterations are perhaps the largest threat to fish biodiversity in the basin. Fishes in the upper reaches will be seriously affected by the construction of the Three Gorges Dam and other dams, and action should be taken for priority conservation. The most immediate restoration need is reconnection of the Yangtze River with its lakes. The cluster of lakes in the Central Yangtze should be protected to maintain habitats for spawning, feeding and migration of migratory fishes. Our study indicates a need to identify areas of high fish biodiversity and to select nature reserves to mitigate the loss of fish biodiversity in the Yangtze River basin.  相似文献

Diazinon (DZN) is one of the most organophosphate insecticides that widely used in agriculture and industry. Selenium is generally recognized to be a trace mineral of great importance for human health, protecting the cells from the harmful effects of free radicals. Therefore, the present study was carried out to investigate the alterations in biochemical parameters, free radicals and enzyme activities induced by diazinon in male rat serum, and the role of selenium in alleviating the negative effects of DZN. Animals were divided into four groups of seven rats each; the first group was used as control. Groups 2, 3 and 4 were treated with selenium (Se; 200 μg/kg BW), diazinon (DZN; 10 mg/kg BW) and diazinon plus selenium, respectively. Rats were orally administered their respective doses daily for 30 days. Results obtained showed that DZN significantly induced thiobarbituric acid reactive substances (TBARS) and decreased the activities of glutathione S-transferase (GST), superoxide dismutase (SOD), catalase (CAT), glutathione peroxidase (GPx) and glutathione reductase (GR) and the levels of reduced glutathione (GSH) in rat sera. Aminotransferases (AST, ALT), phosphatases (AlP, AcP) and lactate dehydrogenase (LDH) activities were significantly increased while acetylcholinesterase (AChE) activity was decreased due to DZN administration. Also, DZN treatment caused significant perturbations in lipids profile and serum biochemical parameters. On the other hand, Se alone significantly decreased the levels of TBARS, total lipids, cholesterol, urea and creatinine, while increased the activities of antioxidant enzymes and glutathione content, total protein (TP) and albumin. In addition, Se in combination with DZN partially or totally alleviated its toxic effects on the studied parameters. In conclusion, Se has beneficial effects and could be able to antagonize DZN toxicity.  相似文献

Digestive cell lysosomes in mussels are known to respond to individual organic chemicals and metals after experimental exposure under laboratory conditions but reports dealing with the response to mixtures of pollutants are scarce. The aim of the present investigation was to compare the lysosomal responses elicited by exposure to a model organic chemical compound (benzo(a)pyrene, B[a]P), a model toxic metal (Cd) and their combination (B[a]P+Cd) under controlled laboratory conditions. Dimethylsulfoxide (DMSO) was used as vehicle to dissolve organic chemicals into seawater. Control mussels were either kept untreated in clean seawater or treated with DMSO. Digestive glands were excised on Day 21. beta-Glucuronidase activity was demonstrated in 8 mum cryotome sections. Lysosomal volume, surface and numerical densities (Vv, Sv and Nv), and surface-to-volume ratio (S/V) were quantified by image analysis. Lysosomal enlargement was evident in digestive cells of mussels exposed to either Cd, B[a]P or B[a]P+Cd. Such enlargement was more marked after exposure to B[a]P+Cd than to B[a]P, but did not reach the levels recorded after Cd exposure. It seems therefore that the presence of B[a]P reduced to some extent the effects of Cd on digestive cell lysosomes in mussels.  相似文献

Summary Data on factors determining activity cycles of nearctic fishes are scarce overall, and nonexistent for Coregoniae (family Salmonidae), a group with closely related and somewhat better known palaearctic species. We studied effects of photoperiodicity, light intensity and temperature on diel locomotor activity of lake whitefish Coregonus clupeaformis (Mitchill). Under LD 12:12, a day-active pattern was evident. Under continuous light (LL) and continuous dark (DD), rhythmicity was greatly suppressed or absent, respectively. Day-time activity levels were positively correlated with illuminance over the range tested (0.005 to 500 lux). At the three temperatures tested, activity was lowest at 7° C, highest at 12° C (a reported temperature optimum), and intermediate at 17° C. Our results suggest that level and diel pattern of lake whitefish activity are under overriding external control, and we find similarities with published data on the European species Coregonus lavaretus.  相似文献

The effect of age and environmental parameters on growth, cholinesterase (ChE), and glutathione S-transferase (GST) activities were assessed in juvenile Apporectodea caliginosa earthworms. Earthworms were maintained in three types of soil- loam, sandy, and clay- at a range of moisture contents (15-30%), and temperatures (5-20 C). Earthworm age (1-3 months) had no significant affect on ChE activity. Growth rates were influenced by all environmental parameters tested and these effects were interrelated. Optimum conditions for growth appeared to be in loam or sandy soil with 25-30% moisture at 10-15 C. The GST activity was influenced by soil temperature and activity was significantly higher at 15 C than at 5 C and 10 C. Soil type also influenced GST activity and this influence was dependent on moisture content. In sandy soil GST activity was significantly lower at 30% moisture than at lower moisture contents, in loam soil GST activity was significantly higher at 15% moisture than at higher moisture contents, while in clay soil GST activity was not affected by moisture content. These results indicate that in field experiments when evaluating GST activity soil temperature and soil type need to be consistent between control and 'contaminated sites'. ChE activity was only affected by temperature, so this should be considered when comparing control and treatedareas.  相似文献

Microbial life in the harsh conditions of Antarctica's cold desert may be considered an analogue of potential life on early Mars. In order to explore the development and survival of this epilithic and endolithic form of microbial life, our most sophisticated, state-of-the-art visualization technologies have to be used to their full potential. The study of any ecosystem requires a knowledge of its components and the processes that take place within it. If we are to understand the structure and function of each component of the microecosystems that inhabit lithic substrates, we need to be able to quantify and identify the microorganisms present in each lithobiontic ecological niche and to accurately characterize the mineralogical features of these hidden microhabitats. Once we have established the techniques that will allow us to observe and identify these microorganisms and mineral substrates in situ, and have confirmed the presence of water, the following questions can be addressed: How are the microorganisms organized in the fissures or cavities? Which microorganisms are present and how many are there? Additional questions that logically follow include: What are the existing water relationships in the microhabitat and what effects do the microorganisms have on the mineral composition? Mechanical and chemical changes in minerals and mineralization of microbial cells can give rise to physical and/or chemical traces (biomarkers) and to microbial fossil formation. In this report, we describe the detection of chains of magnetite within the Martian meteorite ALH84001, as an example of the potential use of SEM-BSE in the search for plausible traces of life on early Mars. Aluminium has the potential to be neurotoxic in humans and animals, and is present in many manufactured foods and medicines and is also added to drinking water for purification purposes. Therefore, the present study was carried out to investigate (1) the alterations in biochemical parameters, free radicals and enzyme activities induced by aluminium chloride (AlCl3) in plasma and different tissues of male rats, and (2) the role of vitamin E (VE) and selenium in alleviating the negative effects of aluminium. VE plays an important role as an antioxidant and is consequently expected to protect tissues from damage caused by reactive oxygen metabolites. Selenium is also generally recognized to be a trace mineral of great importance for human health, protecting the cells from the harmful effects of free radicals. Seven rats per group were assigned to one of six treatment groups: 0 mg VE, 0 mg Se and 0 mg AlCl3/kg body weight (BW) (control); 100 mg VE/kg BW; 200 microg Se kg BW; 34 mg AlCl3/kg BW (1/25 LD50); 34 mg AlCl3 plus 100 mg VE/kg BW; 34 mg AlCl3 plus 200 microg Se/kg BW. Rats were orally administered their respective doses every other day for 30 days. Evaluations were made for lipid peroxidation, enzyme activities and biochemical parameters. Results obtained showed that AlCl3 significantly (p<0.05) induced free radicals (thiobarbituric acid-reactive substances) and decreased the activity of glutathione S-transferase (GST) and the levels of sulphydryl groups (SH groups) in rat plasma, liver, brain, testes and kidney. Aspartate aminotransferase, alanine aminotransferase, alkaline phosphatase, acid phosphatase, and phosphorylase activities were significantly decreased in liver and testes due to AlCl3 administration, while the activities of these enzymes were significantly increased in plasma. In addition, plasma, liver, testes and brain lactate dehydrogenase activities were significantly increased. On the contrary, the activity of acetylcholinesterase was significantly decreased in brain and plasma. Al treatment caused a significant decrease in plasma total protein (TP), albumin and total lipids (TL), and increased the concentrations of glucose, urea, creatinine, bilirubin and cholesterol. VE or Se alone significantly decreased the levels of free radicals, TL, cholesterol, urea and bilirubin, and increased the activity of GST, and SH groups, TP and albumin, while the rest of the tested parameters were not affected. VE or Se in combination with Al partially or totally alleviated its toxic effects on the studied parameters. In conclusion, VE and Se have beneficial effects and could be able to antagonize Al toxicity.  相似文献

Oreochromis niloticus, weighing 36.45 ± 1.12 g were exposed to 10%, 20% and 30% of the LC50 of CdCl2 which represents treatments (T1)1.68, (T2)3.36 and (T3)5.03 mg/l, respectively, for a period of 10, 20 and 30 days. It was found that, compared to a control group reading of 0.19 ± 0.03 μg/g dry weight, accumulation of Cd in the gills was significantly (p < 0.05) increased in samples ranging between 7.64 ± 0.86 and 61.73 ± 0.82 μg/g dry weight from T1 at 10 days to T3 at 30 days. The accumulation of Cd in the liver, meanwhile, was also observed to significantly increase (p < 0.05) with increasing time and concentrations with results ranging between 3.21 ± 0.12 and 181.61 ± 1.32 compared to the control group results of 0.29 ± 0.04 μg/g dry weight. Although muscles exhibited lower levels of accumulation than the gills and liver they still showed the same pattern of increase compared to the control group, with a significant difference ranging between 0.32 ± 0.02 and 2.16 ± 0.08 compared to the control group results of 0.03 ± 0.001 μg/g dry weight. Also, haematological parameters such as red blood cells (RBCs), haemoglobin (Hb) and haematocrit (Hct) were reduced in fish exposed to Cd at all periods, with significant differences (p < 0.05). Plasma glucose concentration showed a significant increase. Total protein levels of fish showed a significant reduction (p > 0.05) for all exposed treatments. Also, the total lipid level increased significantly as fish were exposed to increasing cadmium concentrations, compared to control fish. Finally, the activities of aspartate aminotransferase (AST IU/l) and alanine aminotransferase (ALT IU/l) showed a significant increase (p < 0.05) with increasing time and concentrations.  相似文献

The shore crab Carcinus maenas (L.) reported hitherto not to express endogenous circatidal rhythmicity in winter, is shown not to lose the ability to express such rhythmicity. Crabs maintained in constant reduced salinity in winter exhibit circatidal and circadian rhythms similar to the normal endogenous rhythms of summer caught crabs.In sinusoidal changes of salinity of tidal periodicity, reductions of salinity and increases to ambient sea water induced increased locomotor activity. The former were purely exogenous responses but the latter were also observed to entrain the underlying endogenously controlled circatidal pattern of behaviour.The occurrence of separate exogenous and endogenous responses to different phases of imposed salinity cycles has implications when seeking to understand rhythmic locomotor activity of crabs on the shore and in the search for components of the underlying physiological clock mechanism.  相似文献

A generic index (GI) utilizing epilithic diatom assemblages as abioindicator of water pollution in subtropical rivers in Taiwan was applied toastudy of a tropical river. Seven sites on the River Tsanwun were sampledseasonally from August 1995 to June 1998. Although there is a marked differencein water chemistry between tropical and subtropical rivers in Taiwan, thedominant genera of their diatom assemblages are quite similar. The six generaused for the calculation of GI in the subtropical rivers were also the maincomponents of diatom assemblages in the River Tsanwun. The calculated valuesdisplayed a close correlation with water quality, evaluated on the basis ofphysical and chemical variables. A strong correlation was found between this GIand other diatom-based indices of water quality.  相似文献

Changes in water temperature and salinity are responsible for a variety of physiological stress responses in aquatic organisms. Stress induced by these factors was recently associated with enhanced reactive oxygen species (ROS) generation, which caused oxidative damage. In the present study, we investigated the time-related effects of changes in water temperature and salinity on mRNA expression and the activities of antioxidant enzymes (SOD and CAT) and lipid peroxidation (LPO) in the gills and digestive glands of the ark shell, Scapharca broughtonii. To investigate physiological responses, hydrogen peroxide (H2O2), lysozyme activity, aspartate aminotransferase (AspAT), and alanine aminotransferase (AlaAT) were measured in the hemolymph. Water temperature and salinity changes significantly increased antioxidant enzyme mRNA expression and activity in the digestive glands and gills in a time-dependent manner. H2O2 concentrations increased significantly in the high-temperature and hyposalinity treatments. LPO, AspAT and AlaAT levels also increased significantly in a time-dependent manner, while lysozyme activity decreased. These results suggest that antioxidant enzymes play important roles in reducing oxidative stress in ark shells exposed to changes in water temperature and salinity.  相似文献

Barley seedlings grown on filter paper moistened with 1mM Cd showed 50% root growth inhibition within 24h of exposure. The amount of cadmium after 24h Cd treatment was highest in the first 2mm-long apical root segment, while it was slightly higher in the fourth segment, 6-8mm behind the root tip, after 48h. In recovery experiments, when Cd-treated plants were transferred onto filter paper moistened with distilled water, a large amount of Cd was localised in the apoplast and considerable cell death was detected even though root growth was renewed. This indicates that cell death is likely an active physiological process that contributes to the removal of Cd from the root during root growth recovery. Elevated lipid peroxidation and thiol contents were detected in all individual segments of Cd-treated barley root. On the other hand, proline accumulation was disturbed during Cd stress, showing a significant decrease in all of the studied segments except the first. Cd-induced alteration in the expression of genes involved in metal signalling and detoxification and in drought and oxidative stress responses indicates that Cd-induced water and oxidative stress is responsible for the root growth inhibition, probably through an accelerated differentiation of root tissues.  相似文献
