Four physiologically and phenotypically diversified tobacco (Nicotiana tabacum L. cv. Samsun) plantlet variants had been generated by cultivation on media either lacking or containing sucrose (0 and 3 %, m/v) under two different photon flux densities (PFD), 50 µmol m–2 s–1 (LL) and 200 µmol m–2 s–1 (HL). Plantlets were transferred into soil without any pre-acclimation and grown either under PFD of 200 µmol m–2 s–1 or 700 µmol m–2 s–1. Sucrose feeding in vitro resulted in reduced degree and duration of wilting after transfer. The highest readiness for ex vitro acclimation was found in 3 % HL plants, in which changes of photosynthetic apparatus and stress responses were the smallest. On the contrary, the steepest decline of Fv/Fm ratio on the first day after transplantation, doubled chlorophyll content and almost tripled D1/LHC 2 ratio after 7 d of ex vitro growth under 700 µmol m–2 s–1 characterized 0 % HL plants, which had suffered chronic photoinhibition in vitro. Remarkably high abscisic acid content at the end of in vitro cultivation and during acclimation as well as increased synthesis of both D1 and LHC 2 proteins even at the end of analyzed acclimation period were found only in 0 % LL plants. Increase of D1/LHC 2 ratio and chlorophyll contents demonstrate that in vitro developed leaves of all plant variants are able to acclimate to new environment. The most surprising result in the whole study is the drop of D1 protein synthesis in all plants on the 3rd day. Five times decline of photoprotection level of xanthophylls in plants after ex vitro transfer into the same PFD showed stress character of in vitro cultures.  相似文献

AxenicTrentepohlia odorata was cultured at three different NH4Cl levels (3.5 × 10–2, 3.5 × 10–3, 3.5 × 10–4 M) and three different light intensities (48, 76, 122 µmol m–2 s–1). Chloride had no effect on growth over this range of concentration. High light intensity and high NH4Cl concentration enhanced the specific growth rate. The carotenoid content increased under a combination of high light intensity and low N concentration. WhenD. bardawil was exposed to the same combination of growth conditions, there was an increase in its carotenoid content. The light saturation and the light inhibition constants (K s andK i, respectively) for growth, and the saturation constant (K m) for NH4Cl were determined. TheK s andK i values were higher inT. odorata (66.7 and> 122 mol m–2 s–1, respectively) than inD. bardawil (5.1 and 14.7 µmol m–2 s–1, respectively). The adaptation of barley (Hordeum vulgare L. cv. Akcent) plants to low (LI, 50 µmol m–2 s–1) and high (HI, 1000 µmol m–2 s–1) growth irradiances was studied using the simultaneous measurements of the photosynthetic oxygen evolution and chlorophyll a (Chl a) fluorescence at room temperature. If measured under ambient CO2 concentration, neither increase of the oxygen evolution rate (P) nor enhancement of non-radiative dissipation of the absorbed excitation energy within photosystem 2 (PS2) (determined as non-photochemical quenching of Chl a fluorescence, NPQ) were observed for HI plants compared with LI plants. Nevertheless, the HI plants exhibited a significantly higher proportion of QA in oxidised state (estimated from photochemical quenching of Chl a fluorescence, qP), by 49–102 % at irradiances above 200 µmol m–2 s–1 and an about 1.5 fold increase of irradiance-saturated PS2 electron transport rate (ETR) as compared to LI plants. At high CO2 concentration the degree of P stimulation was approximately three times higher for HI than for LI plants, and the irradiance-saturated P values at irradiances of 2 440 and 2 900 µmol m–2 s–1 were by 130 and 150 % higher for HI plants than for LI plants. We suggest that non-assimilatory electron transport dominates in the adaptation of the photosynthetic apparatus of barley grown at high irradiances under ambient CO2 rather than an increased NPQ or an enhancement of irradiance-saturated photosynthesis.  相似文献

Tobacco plantlets were cultured in vitro under high (200 µmol m–2 s–1) or low (60 µmol m–2 s–1) irradiance with or without saccharose in the medium. Light microscopy and image analysis were used to evaluate the effect of these culture conditions on leaf anatomy. Addition of saccharose resulted in thicker leaves (all leaf layers) and larger mesophyll cells under both growth irradiances. Various irradiance affected leaf anatomy differently when plantlets had been cultivated in presence or absence of saccharose in the medium. While under high irradiance in presence of saccharose leaf thickness and number of chloroplasts per cell section were increased, plantlets grown under high irradiance in absence of saccharose had thinner leaves and less chloroplasts per cell section. The changes were more pronounced in palisade parenchyma layer.  相似文献

Spirulina platensis (= Arthrospira fusiformis) was isolated from Lake Chitu, a saline, alkaline lake in Ethiopia, where it forms an almost unialgal population. Optimum growth conditions were studied in a turbidostat. Cultures grown in modified Zarrouk's medium and exposed to a range of light intensities (20–500 µmol photons m–2s–1) showed a maximum specific growth rate (µmax) of 1.78 d–1. Quantum yield for growth (µ) was 3.8% at the optimum light for growth of 330 µmol photons m–2s–1, and ranged from 2.8 to 9.4%. With increase in irradiance, the chlorophyll a concentration decreased, and the carotenoids/chlorophyll a ratio increased by a factor of 2.4. The phosphorus to carbon ratio (P/C) showed some variation, while the nitrogen to carbon ratio (N/C) remained relatively constant, thus causing fluctuations in the N:P ratio (7–11) of cells. An optimum N:P ratio of about 7 was attained in cells growing at the optimum light for growth. Results from the continuous culture experiments agreed well with maximum values of photosynthetic efficiency given in the literature for natural populations of S. platensis in the soda lakes of East Africa, Lake Arenguade (Ethiopia), and Lake Simbi (Kenya).  相似文献

Laurencia brongniartii is usually found at depths below 4 m, but can be found in shallow subtidal areas in crevices and on the walls of a coral reef in Amami Oshima Island, Kagoshima Prefecture, Japan, where irradiances were significantly lower than those at similar depths in open water. In preparation for the possible cultivation of this species for its antibiotic compounds, the effects of temperature and irradiance on photosynthesis and growth were measured. Photosynthesis and growth rates of L. brongniartii explants were highest at 26 and 28 °C, which closely corresponded to temperatures found during August to late December when it was most abundant. The estimated maximum photosynthesis rate (P max) was 4.41 mol photon m–2 s–1 at 26 °C and 4.07 mol photon m–2 s–1 at 28 °C. Saturating irradiance occurred at 95 mol photon m–2 s–1 at 26 °C and 65 mol photon m–2 s–1 at 28 °C. In contrast, growth experiments at 41.7 mol photon m–2 s–1 caused bleaching of explants and the maximum growth rate observed during the study was 3.02 ± 0.75% day–1 at 28 °C and 25 mol photon m–2 s–1. The difference in the saturating irradiance for photosynthesis and the irradiance that caused bleaching in growth experiments suggests that long-term exposure to high irradiance was detrimental and should be addressed before the initiation of large scale cultivation.  相似文献

The rooting of shoots of micropropagated Rosa hybrida cv. Madame Delbard was conducted on MS medium with 30 kg m–3 sucrose or on hydroponic medium (containing less mineral salts), under higher photosynthetic photon flux density (PPFD) (100 in comparison with 45 µmol m–2 s–1) and flushed by ambient air [AC, 340 µmol(CO2) mol–1] or by CO2-enriched air (EC, 2 500 µmol mol–1) and lower relative humidity (80–90 % vs. 96–99 %). This cultivation led to plantlets with longer roots and adventitious root formation. Net photosynthetic rate and ribulose-1,5-bisphosphate carboxylase/oxygenase (RuBPCO) activities, RuBPCO/phosphoenolpyruvate carboxylase activities ratio, and starch accumulation increased under these conditions. After 14 d, plantlets had functional stomata and could be acclimated on open benches without gradual decrease in relative humidity. The percentage of survival was higher when the rooting took place in EC than in AC. However, the advantage acquired during rooting phase by plantlets cultured in liquid medium was not maintained after 4 weeks of acclimatisation.  相似文献

The growth yield of the PUFA-producing marine microalgaIsochrysis galbana ALII-4 grown in a light limited chemostat, was measured under a wide variety of conditions of incident irradiance (I O ) and dilution rates (D). The experiments were conducted under laboratory conditions at 20 °C under continuous light. D ranged from 0.0024 to 0.0410 h–1 at three intensities of Io (820, 1620 and 3270 µmol photon m–2 s–1) close to those found in outdoor cultures. A maximum efficiency max = 0.616 g mol photon–1 was obtained at I O = 820 µmol photon m–2 s–1 and D = 0.030 h–1 and the maximum capacity of the biomass to metabolize the light harvested was found to be 13.1 µmol photon g–1 s–1. Above this value, a significant drop in the system efficiency was observed. A new approach based in the averaged irradiance is used to assess the photon flux absorbed by the biomass.  相似文献

This study reports the effects of light availability during the acclimatization phase on photosynthetic characteristics of micropropagated plantlets of grapevine (Vitis vinifera L.) and of a chestnut hybrid (Castanea sativa × C. crenata). The plantlets were acclimatized for 4 weeks (grapevine) or 6 weeks (chestnut), under two irradiance treatments, 150 and 300 mol m–2 s–1 after in vitro phases at 50 mol m–2 s–1. For both treatments and both species, leaves formed during acclimatization (so-called `new leaves') showed higher photosynthetic capacity than the leaves formed in vitro either under heterotrophic or during acclimatization (so-called `persistent leaves'), although lower than leaves of young potted plants (so-called `greenhouse leaves'). In grapevine, unlike chestnut, net photosynthesis and biomass production increased significantly with increased light availability. Several parameters associated with chlorophyll a fluorescence indicated photoinhibition symptoms in chestnut leaves growing at 300 mol m–2 s–1. The results taken as a whole suggest that 300 mol m–2 s–1 is the upper threshold for acclimatization of chestnut although grapevine showed a better response than chestnut to an increase in light.  相似文献

The uptake of ammonium, nitrate and phosphate by laboratory-grown young sporophytes of Laminaria abyssalis was measured in a perturbed system (batch mode) at 18 °C and 35 ± 5 µE m–2 s–1 photon flux density. Uptake of all appeared to follow saturation-type nutrient uptake kinetics. The NO inf3 sup– (K s = 14.0 µM, V max = 5.0 µmol h–1 g–1 dry wt) and NH inf4 sup+ (K s = 4.6 µM, V max= 2.0 µmol h–1 g–1 dry wt) were taken up simultaneously, although NH inf4 sup+ was taken up more rapidly. Values of K 3 and V max for phosphate were, respectively, 2.21 µM and 0.83 µmol h–1 g–1 dry wt. Nitrate and phosphate were both consumed in similar rates (V max /Ks 0.37) at low concentrations. NH inf4 sup+ , thus, might be a more efficient form of N fertilizer if artificial enrichment of seawater is used.  相似文献

Photosynthetic acclimation to reduced growth irradiances (650 and 200 µmol m–2 s–1) in Eleusine coracana (L.) Garten, a nicotinamide adenine dinucleotide-malic enzyme (NAD-ME) C4 species and Gomphrena globosa L., a nicotinamide adenine dinucleotide phosphate-malic enzyme (NADP-ME) C4 species were investigated. E. coracana plants acclimated in 4 and 8 d to 650 and 200 µmol m–2 s–1, respectively, whereas G. globosa plants took 8 and 10 d, respectively, to acclimate to the same irradiances. The acclimation to reduced irradiance was achieved in both species by greater partitioning of chlorophyll towards the light-harvesting antennae at the expense of functional components. However, magnitude of increase in the light-harvesting antenna was higher in E. coracana as compared to G. globosa. Superior photosynthetic acclimation to reduced irradiance in G. globosa was due to the smaller change in functions of the cytochrome b 6/f complex, photosystem (PS) 1 and PS2 leading to the higher carbon fixation rates compared to E. coracana.  相似文献

We tested the effect of growing conditions during micropropagation on the fast kinetics of chlorophyll (Chl) fluorescence of Gardenia jasminoides Ellis plantlets during a 4-week acclimation to ex vitro. We studied whether photoautotrophic growing in vitro produced plantlets with less photoinhibition impairment during acclimation. Of the growing conditions stimulating photoautotrophy in vitro, only loose tube caps had a positive effect, whereas low sucrose or sucrose-free content in the medium and high PPFD showed a negative effect. Thus, plantlets cultured with 3 % (m/v) of sucrose were subsequently less photoinhibited throughout acclimation than those cultured with low sucrose (0.5 %) or sucrose-free media. Moreover, at the end of acclimation the former plantlets showed Fv/Fm and Fv/F0 ratios typical of unstressed ex vitro plants as well as a higher Chl content and ratio of Chls to carotenoids. Plantlets cultured at a photosynthetic photon fluence density (PPFD) of 50 µmol m–2 s–1 also showed a better performance at the end of acclimation than those cultured at a higher (110 µmol m–2 s–1) PPFD. Thus except in the case of loose-tube closure, gardenia plantlets cultured in vitro under conventional sucrose concentration and PPFD are the least photoinhibited during acclimation. Nevertheless, significant interactions between the in vitro growing factors were observed at the end of acclimation.  相似文献

The red alga Stictosiphonia hookeri is epilithic in shaded habitats of the upper intertidal zone from 30 to 55° S. Thalli of this species from Argentina, Chile, South Africa and Australia, usually without reproductive structures when collected, all developed tetrasporangia in culture. Although good vegetative growth occurred in all nine isolates at 20–25 °C, 12:12 light: dark cycle, 10–30 µmol photons m–2 s–1, none reproduced in these conditions except one isolate from Australia. At 15 °C the four South African (34 °S) isolates developed tetrasporangial stichidia, and three completed a Polysiphonia-type life history. Gametophytes were unisexual or bisexual. At 15 °C one isolate from Chile (36 °S) formed tetrasporangia, but sporelings were not viable. At 10 °C isolates from Argentina and Chile (53 °S and 54 °S) formed tetrasporangia; however, only the Chile isolate completed a Polysiphonia-type life history with unisexual gametophytes. The temperature required to induce sporogenesis correlates with the range of water and air temperatures in the natural habitats of each isolate. In irradiances >50 µmol m–2 s–1 the thalli became yellow- brown within two weeks because of phycobiliprotein loss, but this did not impair growth or reproduction. The Argentina and Chile isolates were resistant to freezing in seawater for at least two days, showing no cell damage. The protein cuticle of the outer cell wall is repeatedly shed in culture. This may serve to minimize the attachment of epiphytes in the field.  相似文献

We collected the ephemeral macrophyte Ruppia drepanensis Tin. ex Guss. from the athalassic shallow lake Fuente de Piedra (Málaga. Southern Spain). This lake, situated in an endorheic basin, shows great seasonal changes in depth and Total Dissolved Solids (TDS).Dissolved oxygen evolution studied in the laboratory at 17 different photon flux densities (PFD) showed a maximum rate of photosynthesis of 0.55 mg C g dry wt–1 h–1, a light compensation point at 86 µE m–2 s–1 and a saturation point at 333 µE m–2 s–1. A moderate photoinhibition (\ = 1.68 10–4) was found above 695 µE m–2 s–1.Estimates of pigment concentrations revealed 10 times more carotenoids than chlorophyll.The adaptation of the plants to high irradiances and to the particular features of their hypersaline environment are discussed.  相似文献

Total in vitro activity of RuBPCO (ribulose-1,5-bisphosphate carboxylase/oxygenase) enzyme was assayed spectrophotometrically by the continuous measurement of 3-phosphoglycerate-dependent NADH oxidation in a coupled enzyme system. RuBPCO activities were found in the ranges 1.01–2.76 and 1.23–3.10 µmol(CO2) m−2 s− 1 in current Norway spruce needles growing in ambient (AC) and elevated (EC) CO2 concentration, respectively. RuBPCO activity in AC needles from the upper layer (U) was 11–15 % higher compared to those from the middle (M) layer, and even 44–56 % higher compared to the lower (L) layer of spruce crown. Over the vegetation season, we observed a highly significant decrease of RuBPCO activity in the EC-U needles from 3.10 (July) to 1.60 (October) µmol(CO2) m−2 s−1 as a consequence of downward feedback regulation. Moreover, this down-regulation was not caused by a non-specific decrease in total leaf nitrogen content.The work forms a part of the research supported by grants no. In vitro cultures of Nephrolepis exaltata and Cordyline fruticosa were stored at 5°, 9° or 13°C, at a low irradiance (3–5 mol m–2 s–1) or in darkness. Prior to storage the cultures were subjected to 18°, 21°, 24° or 27°C and 15, 30 or 45 mol m–2 s–1 in a factorial combination.The optimal storage conditions for Nephrolepis were 9°C in complete darkness. These cultures were still transferable to a peat/perlite mixture at the end of the experimental period of 36 months.The optimal storage conditions for Cordyline were 13°C and a low light level (±3–5 mol m-2 s-1). When the pre-storage conditions were normal growth room conditions (24°C and 30 mol m-2 s-1), in vitro cultures could be stored for 18 months. With the most favourable pre-storage treatment (18°C and 15 mol m-2 s-1) some cultures still had green shoots after 36 months of storage, but did not survive transfer to peat/perlite.Pre-conditioning before storage was most favourable for Nephrolepis, and not that important, but still favourable, for Cordyline. There was an interaction between pre-storage temperature and pre-storage irradiance. Grain sorghum [Sorghum bicolor (L.) Moench. cvs. TX430 and KS82] was grown in a Haynie very fine sandy loam (coarse-silty, mixed, superactive, calcareous, mesic Mollic Udifluvents) under constant 47 % shade or full irradiance in a greenhouse under two watering regimes to see the combined and individual effects of low irradiance (LI) and low water (LW) on the sorghum genotypes. Under the high-irradiance (HI) and high-water (HW) treatment (control) and the LI-HW treatment, TX430 grew taller than KS82. Both LI and LW reduced several times the fresh and dry masses. Under the control conditions, TX430 reached its maximum net photosynthetic rate (P Nmax) of 28.93 mol m–2 s–1 at a photosynthetic photon flux density (PPFD) of 1 707 mol m–2 s–1, and KS82 reached its P Nmax of 28.32 mol m–2 s–1 at a PPFD of 2 973 mol m–2 s–1. The fact that TX430 had P Nmax under a lower PPFD than KS82 may relate to its taller growth under LI conditions. Hence genotypes of sorghum might be selected for low irradiance using curves relating P N to PPFD.  相似文献

Ethane was used as tracer gas to assess the likelihood of thermo-osmotically induced mass-flow in the aerenchyma of Avicennia marina seedlings without pneumatophores. Ethane movement was measured in darkness and with illumination at approximately 600 µ mol m–2s–1 provided to the leaves and stem, with the expectation that leaf warming under illumination would provide for thermo-osmotic flow. In some seedlings the flow increased with illumination, and in others it either decreased or remained unchanged. Overall, there was no statistically significant difference in the conductance to ethane between darkened illuminated plants, and the rates of ethane movement were consistent with an average diffusive conductance to oxygen down the stem of 0.22 × 10–19m3 s–1. it was concluded that there was no evidence for thermo-osmotically induced in this case.  相似文献

Various ecophysiological investigations on carnivorous plants in wet soils are presented. Radial oxygen loss from roots of Droseraceae to an anoxic medium was relatively low 0.02 – 0.07 mol(O2) m– 2 s–1 in the apical zone, while values of about one order of magnitude greater were found in both Sarracenia rubra roots and Genlisea violacea traps. Aerobic respiration rates were in the range of 1.6 – 5.6 mol kg–1 (f.m.) s–1 for apical root segments of seven carnivorous plant species and 0.4 – 1.1 mol kg–1 (f.m.) s–1 for Genlisea traps. The rate of anaerobic fermentation in roots of two Drosera species was only 5 – 14 % of the aerobic respiration. Neither 0.2 mM NaN3 nor 0.5 mM KCN influenced respiration rate of roots and traps. In all species, the proportion of cyanide-resistant respiration was high and amounted to 65 – 89 % of the total value. Mean rates of water exudation from excised roots of 12 species ranged between 0.4 – 336 mm 3 kg–1 (f.m.) s–1 with the highest values being found in the Droseraceae. Exudation from roots was insensitive to respiration inhibitors. No significant difference was found between exudation rates from roots growing in situ in anoxic soil and those kept in an aerated aquatic medium. Carnivorous plant roots appear to be physiologically very active and well adapted to endure permanent soil anoxia.  相似文献

Three eulittoral algae(Ulva lactuca, Porphyra umbilicalis, Chondrus crispus) and one sublittoral alga(Laminaria saccharina) from Helgoland (North Sea) were cultivated in a flow-through system at different temperatures, irradiances and daylengths. In regard to temperature there was a broad optimum at 10–15° C, except inP. umbilicalis, which grew fastest at 10 °C. A growth peak at this temperature was also found in four of 17 other North Sea macroalgae, for which the growth/temperature response was studied, whereas 13 of these species exhibited a growth optimum at 15 °C, or a broad optimum at 10–15 °C. Growth was light-saturated inU. lactuca, L. saccharina andC. crispus at photon flux densities above 70 µE m–2s–1, but inP. umbilicalis above 30 µE m–2s–1. Growth rate did not decrease notably in the eulittoral species after one week in relatively strong light (250 µE m–2s–1), but by about 50 % in the case of the sublittoralL. saccharina, as compared with growth under weak light conditions (30 µE m–2s–1). In contrast, chlorophyll content decreased in the sublittoral as well as in the eulittoral species, and the greatest change in pigment content occurred in the range 30–70 µE m–2s–1. Growth rate increased continuously up to photoperiods of 24 h light per day inL. saccharina andC. crispus, whereas daylength saturation occurred at photoperiods of more than 16 h light per day inU. lactuca andP. umbilicalis.  相似文献
