The taxonomic composition of denitrifying bacteria in soddy podzolic soil was studied by the succession analysis method. This method revealed a significant variation in the taxonomic composition of denitrifying microorganisms in the course of succession. In contrast to succession analysis, the single microbiological analysis of soil samples reflected only the late stage of succession and thus led to an underestimation of the major members of succession. Myxobacteria were found to be the most active denitrifiers at the early stages of succession, whereas bacilli dominated at its late stages. The bacilli were represented by three facultatively anaerobic species, Bacillus cereus, Bac. circulans, and Bac. polymyxa.  相似文献

The seasonal fluctuations in the concentration of cultured denitrifying and N2-fixing bacteria were followed in an ammonium fertilised and a control soil of a Norway spruce forest near Villingen/Black Forest from December 1994 to August 1998. The horizontal distribution of bacteria in three layers was determined by the MPN-method and by molecular probing (colony hybridisation) using specific 0.4–0.7 kb DNA probes for denitrification steps (narG, nirS, nirK and nosZ) and for N2-fixation (nifH). The data showed that highest bacterial counts and higher numbers of denitrifying and N2-fixing bacteria were generally detectable in the upper (= 5 cm) soil layer and that their amount decreased with soil depth. The concentration of these cultured bacteria showed seasonal fluctuations with highest numbers in autumn/winter/early spring and with low counts in summer. Denitrifying and N2-fixing bacteria amounted to less than 10% of the total number of cultured bacteria determined by the MPN-method. Fertilisation with ammonium did not cause a shift in the population of these bacteria. These findings were corroborated by hybridisation experiments with genomic DNA isolated from the different layers. Strongest DNA–DNA hybridisation band intensities were obtained in the upper soil layer and their intensities decreased with soil depth. Soil samples from Villingen assayed in the laboratory produced N2O (in dependence of nitrate and C2H2 added to the vessels) and utilised this gas with higher activities in the assays with the fertilised soil. It is concluded that molecular techniques can successfully be applied for assessing seasonal fluctuations of bacterial populations in soil. Relative abundance of denitrifying and N2-fixing bacteria can be determined from experiments with DNA isolated from soils. Attempts to transform these results to the total population of soil bacteria on a single cell basis are faced with many uncertainties.  相似文献

161 rotifer taxa 46 of which are new records for Estonia were identified in the north-western part of Lake Peipsi. The greatest number of taxa occurred in August (93), followed by September (79). In all, 19 families (Monogononta) are represented with 41 genera. The dominant families are Brachionidae (32 taxa) and Synchaetidae (16). The species of Anuraeopsis, Conochilus, Kellicottia, Keratella, Notholca, Polyarthra and Synchaeta are dominating. The taxonomic composition is analysed in the ecological aspect. Some species of zoogeographic interest, including a supposedly endemic Ploesoma peipsiense, are reported.  相似文献

The root-colonizing actinomycete complexes of genotypically different barley plants grown in soddy podzolic soil were found to contain streptomycetes of the sections Cinereus (series Chromogenes, Achromogenes, and Aureus) and Roseus (series Fradiae), dominant being streptomycetes of the section Cinereus ser. Chromogenes. The abundance and diversity of soil streptomycetes in the barley rhizoplane increased in the order: var. 999-93 < var. Kumir < var. Novichok < var. 889-93. Experiments revealed functional specificity in the root-associated actinomycete complexes of different barley varieties. The actinomycete complex colonizing the barley var. 999-93 roots was distinguished by a wide range of utilizable root exudate metabolites and a low occurrence rate of antagonistic species.  相似文献

A mixed culture containing nitrifying bacteria and denitrifying bacteria was investigated for aerobic simultaneous nitrification and denitrification. A mixture of NaHCO3 and CH3COONa was selected as the appropriate carbon source for cell growth and nitrogen removal, the concentrations of carbon and nitrogen sources were also examined. Ammonia could be oxidized aerobically to nitrite by the mixed culture, and the intermediate nitrite was then reduced to dinitrogen gas. No nitrite was detected during the process. 0.212 g of ammonia/l could be removed in 30 h and nitrate could not be utilized aerobically by the mixed culture. Nitrite could be degraded aerobically as well as anaerobically. Very little ammonia was degraded anaerobically, but the ability to degrade ammonia could be recovered even after oxygen had been supplied for 42 h.  相似文献

Oligotrophic bacteria were shown to exist abundantly in all layers of a rendzina forest soil throughout the year. Two-hundred-three oligotrophic bacteria were isolated from forest soil (Aoba, Sendai) at different layers (L, F, H and A layers) throughout the year, and their morphological and physiological characteristics were examined. A high proportion (95%) of the isolated oligotrophs were Gram-negative, non-spore forming bacteria. Based on the cell shape, the isolates were divided into four groups: regular rods, curved/spiral bacteria, irregular rods, and buddin and/or prosthecate bacteria. Each group of bacteria is discussed in relation to the physiological characteristics. Notably oligotrophic bacteria of different cell types showed a marked zonal distribution in respect to profile depth.  相似文献

The study of various factors (soil acidity, the variety of barley plants, and their developmental phases) on the rhizosphere actinomycete complex showed that it is soil acidity that substantially influences the population of rhizosphere actinomycetes. The effect of soil acidity was most likely due to the different tolerance of rhizosphere actinomycetes to high concentrations of the aluminum and hydrogen ions. The developmental phases of barley plants correlated with the population indices of only one genus of actinomycetes, Micromonospora.  相似文献

Bacterial succession in soil was studied for two variants of initiation (moistening and moistening with addition of glucose). To determine the numbers of viable gram-negative bacteria, the modified nalidixic acid method was applied. The numbers of gram-negative bacteria revealed by this method were 2 to 3.5 times higher than those determined by the traditional method. In a developing community, the highest total bacterial numbers were observed on day 7; afterwards their numbers decreased and stabilized at a level exceeding four-to fivefold the initial one. In both experimental variants, the highest numbers of viable gram-negative bacteria were revealed on day 15 (75–85% of the total bacterial numbers). Morphology of these bacteria suggests their classification as cytophagas (chitinophagas) utilizing chitin from the dead fungal mycelium.  相似文献

【背景】深海海域具有高压、低温、无光等环境条件,蕴含着丰富而独特的微生物资源。【目的】从深海沉积物中定向分离、筛选脱氮效率高的好氧脱氮菌株资源,并揭示其脱氮特性,为开发水体脱氮微生物技术提供物质基础。【方法】以东太平洋、南大西洋、西南印度洋共10个站位的深海沉积物为研究材料,在28°C下使用无机氮源连续进行两轮富集培养,然后定性筛选可以脱除氨氮、亚硝态氮和硝态氮的菌株,并通过形态学和16S rRNA基因序列分析进行初步分类鉴定;对优选得到的功能菌株,分别采用以氨氮、亚硝态氮、硝态氮为唯一氮源的培养基定量研究其生长和脱氮性能。【结果】从10份大洋深海沉积物样品中共分离得到49株好氧反硝化菌,其中3株在有氧条件下反硝化效率较高,分别命名为Pseudomonassp.G111、Pseudomonassp.G112和Dietziamaris W023a,其中菌株G111和G112与模式菌株博岑假单胞菌Pseudomonas bauzanensis BZ93T的16S rRNA基因序列相似度为99.2%,菌株W023a与模式菌株海洋迪茨氏菌DietziamarisATCC35013T的16SrRNA基因序列相似度为99.9%。菌株G111、G112和W023a培养48h后,对氨氮的脱除率分别为98.0%、85.2%和97.6%;对亚硝态氮的脱除率分别为71.9%、67.5%和34.7%;对硝态氮的脱除率分别为66.0%、52.6%和56.3%。菌株G111、G112和W023a均为异养硝化-好氧反硝化菌,可通过好氧反硝化作用将亚硝态氮和硝态氮还原为含氮气体,也可通过异养硝化-好氧反硝化作用将氨氮转化为含氮气体。【结论】从深海沉积物中分离筛选得到3株高效好氧反硝化菌,所获得的菌株在水体净化、污水处理、生态系统修复等领域具有应用潜力。  相似文献

This paper presents data on the taxonomic composition and dynamics of numerical density of meroplankton in the Sea of Azov. Studies performed in June 2003–2005 revealed benthic invertebrate larvae of 26 taxa: Polychaeta, 7; Cirripedia, 1; Decapoda, 5; Gastropoda, 5; and Bivalvia, 8. Meroplankton was dominated by larvae of species that are able to endure considerable concentrations of pollution by labile sulfides in bottom sediments and eutrophication: Mytilaster lineatus, Cerastoderma sp., Abra ovata (Philippi), Hydrobia acuta (Gastropoda), and Amphibalanus improvisus (Cirripedia). The spatial and temporal variations in the structure of dominant species and numerical abundance of meroplankton in the Sea of Azov are likely to be associated with water temperature fluctuations, pelagic predators, and eutrophication. Under anthropogenic stress and predation pressure, most larvae of benthic invertebrates are apparently unable to complete metamorphosis and contribute to recruitment to parental populations. Thus, the numerical density of meroplankton in the Sea of Azov can significantly vary, even over one month.  相似文献

【目的】探索叶际微生物协同植物削减大气氮氧化物的机制,了解叶际可培养好氧反硝化细菌的存在及多样性,获得高效的叶际好氧反硝化细菌资源。【方法】采用富集培养结合格里斯试剂检测、溴百里酚蓝(bromothymol blue, BTB)培养基筛选的方法从景观植物叶际分离筛选好氧反硝化细菌,对好氧反硝化细菌的16S rRNA基因序列进行系统发育分析,并选取其中一株高效好氧反硝化细菌进行脱氮性能研究。【结果】从6种景观植物石楠、女贞、木樨、樟树、卫矛冬青、荷花玉兰的叶际中分离到好氧反硝化细菌13株,经16S rRNA基因序列分析发现,13株细菌分别属于4门7科7属,其中4株为肠杆菌属(Enterobacter),3株为无色杆菌属(Achromobacter),2株为假单胞菌属(Pseudomonas),其余4株分别属于鞘氨醇杆菌属(Sphingobacterium)、不动杆菌属(Acinetobacter)、微杆菌属(Microbacterium)和假节杆菌属(Pseudarthrobacter)。定量分析发现菌株SF的反硝化效果较好。通过单因素试验和响应面设计试验,对菌株SF的脱氮性能进行了一系列研究,探究了碳源、温度、初始pH、碳氮比和转速等因素对菌株SF脱氮效果的影响。结果表明,菌株SF的最佳脱氮条件:碳源为葡萄糖,初始pH值为7.5,碳氮比为9.7,转速180 r/min,温度为33.5 ℃。在此条件下,当初始硝酸盐浓度为361 mg/L时,72 h总氮去除率可达到93.3%。【结论】景观植物叶际中存在较多种类的可培养好氧反硝化细菌,丰富了叶际氮循环相关微生物的类型,为探索叶际微生物协同削减大气氮氧化物的机制奠定了基础。通过高效脱氮菌株的筛选,为进一步应用微生物协同植物削减空气氮氧化物污染提供了候选菌株。  相似文献

生物脱氮具有经济高效、绿色环保的特点,在废水处理中具有广阔前景;目前在废水处理工艺中脱氮效果较好的微生物大部分为嗜温菌,这类菌在低温条件下的脱氮效率会受到强烈抑制,而耐冷脱氮菌可有效抵抗低温环境并进行高效脱氮,逐渐引起了研究者们的重视。其次,纳米颗粒(nanoparticles,NPs)在生物学、农学、医药等领域的应用日渐广泛,但在生产、储存和使用含NPs产品的过程中,不可避免地会向水和土壤环境释放NPs,在水环境中,NPs的大量积累会阻碍微生物的脱氮过程,也对废水处理提出了新的挑战,成为公众日益关注的环境问题和研究热点。当前,有部分研究已关注于耐冷菌的脱氮过程、NPs对耐冷菌脱氮过程的毒害作用及减毒措施。基于此,本篇文章将阐述耐冷菌的耐冷机理与脱氮过程,阐明NPs对耐冷菌脱氮过程的毒害作用与减毒调控措施,为低温环境下采用微生物处理含有NPs的氮污染废水提供理论依据。  相似文献

Sequence divergence in the ribosomal genes of known strains and isolates of aquatic denitrifying bacteria was investigated using restriction fragment length polymorphism (RFLP) analysis. The same cultures were characterized for their homology with antibody and gene probes for nitrite reductase (NiR), a key enzyme in the denitrification pathway, and for amplification with a set of polymerase chain reaction primers designed to amplify a portion of the NiR gene. The NiR probes were developed from Pseudomonas stutzeri (ATCC 14405) and several P. stutzeri strains were included in the analyses. The RFLP analysis clustered most of the P. stutzeri strains together, but detected considerable diversity within this group. Isolates from three aquatic environments exhibited within —and among — habitat diversity by RFLP. Hybridization with the NiR probes and amplification with the NiR primers were not correlated with the clustering of strains by rDNA RFLP analysis. The relationships among strains deduced from ribosomal DNA RFLP reflect heterogeneity within the P. stutzeri group and among other pseudomonads, and the patterns differ from those inferred from specificity of the NiR probes.Abbreviations NiR Nitrite reductase - PCR polymerase chain reaction - RFLP restriction fragment length polymorphism  相似文献

异养硝化-好氧反硝化(heterotrophic nitrifying-aerobic denitrification,HN-AD)菌的发现打破了传统的脱氮理论,可以在有氧条件下同时进行硝化和反硝化,成为近年来的研究热点。HN-AD细菌在海洋氮循环中发挥着重要作用。本文对海洋环境中HN-AD菌的多样性和部分已知氮代谢途径及相关酶系进行了介绍,分析了盐度、碳氮比、溶解氧、pH等环境因素对HN-AD菌脱氮效果的影响,对其工艺和技术应用、前景和发展方向进行了综述和展望。  相似文献

在实验室条件下,研究了农药污染对水稻田土壤反硝化细菌(Denitrfying bacteria,DNB)种群数量及其反硝化活性的影响。结果表明,紫色稻田土壤、黄松稻田土壤和红壤稻田土的DNB种群数量范围分别为59.04×104~157.59×104、42.89×104~108.97×104和32.14×104~75.30×104cfu·g-1,稻田土DNB种群数量和土壤NO3^-的消耗量之间具有正相关性。在1kg干土中加入1mg丁草胺或呋喃丹,能刺激DNB的生长及其反硝化活性,在1kg干土中加入5mg多菌灵、10mg丁草胺或呋喃丹,对DNB的生长及其反硝化活性有明显的抑制作用,丁草胺和呋喃丹施后7d,多菌灵施后14d,对水稻田土壤的DNB种群数量和反硝化活性抑制作用达到最大,然后逐渐减轻,最后呈现一定的促进作用。  相似文献

Scanning electron microscopy revealed that the average cell size of bacteria associated with the digestive tract of soil millipedes was 0.65 μm in diameter, 1.36 μm in length, and 0.60 μm3 in volume. An example of millipedes illustrated that the intestinal tract bacteria of soil invertebrates share the following features: (1) a high density level in this habitat; (2) existence mostly in the form of vegetative cells; (3) a cell size significantly smaller than that of bacteria functioning in soil; (4) a cell size closer to the lower limits of the size range characteristic for bacterial cultures grown in laboratory media. All this suggests that the bacterial community of the digestive tract differs from the typical soil community not only in composition but also in a higher level of physiological activity.  相似文献
