The influence of nicotianamine (NA) on formation and elongation of adventitious roots in hypocotyls of de-rooted NA-less mutant seedlings of Lycopersicon esculentum Mill, was examined in relation to the iron supply [ferric N-N'-ethylenediaminedi-(2-hydroxyphenylacetate) (FEDDHA), ferric ethylenediaminetetracetate (FeEDTA), ferric N-(2-hydroxyethyl)-ethylenediaminetriacetate (FeHEDTA, Fe-citrate and FeCl3] in the nutrient solution. The initiation of root primordia in hypocotyl cuttings was independent of NA and occurred with about the same frequency in both, mutant and wild-type. In the mutant the development of primordia to adventitious roots was blocked at all iron sources used, except FeEDTA. Addition of NA (5x 10−6 to 2 × 10−5 M ) to the rooting medium resulted in a fast growth of adventitious roots in mutant cuttings with all iron sources tested. Rooting of wild-type cuttings was independent from NA application and iron sources. We suppose that NA is involved in the intracellular transport of iron. Its function is possibly linked with chelation of ferrous iron in the cell.  相似文献

NAD kinase is thought to play an important role in the plant cellular responses to biotic and abiotic stress as one of the isoforms of the enzyme is activated by the Ca2 + –calmodulin (CaM) complex. NAD kinase activity was measured after short‐term NaCl stress applied to isolated cells from Lycopersicon esculentum, var. Volgogradskij (NaCl‐sensitive tomato) and L. pimpinellifolium, acc. PE2 (NaCl‐tolerant species). NAD kinase activity remained constant in the sensitive species, whereas a sharp decrease was observed in the tolerant one. After salt treatment, an induction of the calmodulin gene(s) was observed in the two species, together with a 30–50% decrease in ‘active’ CaM content, i.e. CaM able to activate purified NAD kinase, in L. pimpinellifolium. The decrease in NAD kinase activity could not, however, be fully explained by this decrease in active CaM content. A similar decrease in NAD kinase activity was also recorded with other ionic stresses and exposure to high temperatures, but not in the case of drought, exposure to low temperatures, hormonal (indole‐3‐acetic acid and abscisic acid) or H2O2 treatments. External Ca2 + certainly plays a role in the biochemical mechanism(s) leading to NAD kinase inhibition, while no role could be shown for intracellular Ca2 + . In addition, after salt stress, a modification of the redox state of NAD kinase seems to be responsible for the inhibition of the enzyme.  相似文献

Ferredoxin-dependent glutamate synthase (EC 1.4.7.1) catalyzes an essential step in the pathway of glutamate biosynthesis. Exposing detached tomato ( Lycopersicon esculentum ) leaves for 6 h to 12 g l−1 NaCl resulted in a significant two-fold increase in the activity of ferredoxin-dependent glutamate synthase extracted from the leaves. Western blot studies demonstrated that salt treatment also increased the ferredoxin-dependent glutamate synthase content of the leaves. A similar effect of salt on the concentration of this enzyme was found in the leaves of hydroponically-grown tomato plants. The induction of ferredoxin-dependent glutamate synthase under salt stress may provide the glutamate required for the proline synthesis which is a common response to salt stress.  相似文献

The extreme dwarf d x tomato ( Lycopersicon esculentum Mill.) mutant has very short internodes which were found to contain shorter and fewer epidermal cells. The leaves are highly abnormal. The mutant showed a substantial stem growth response to GA3, without approaching normal stature or morphology. The active gibberellin GA1 and its precursors GA19 and GA20 were identified by coupled gas chromatography-mass spectrometry (GC/MS) in d x shoots. Quantitative GC/MS revealed that GA20 accumulated to far higher levels than normal in stems and leaves of the mutant.  相似文献

Localized wounding is known to induce systemic proteinase inhibitors (PI) in seedlings of tomato (Lycopersicon esculentum L.). Inhibitors of elastase (EC 3.4.21.36) were shown here to be among those systemically induced by wounding, and a simple rapid assay for PI based on elastase was developed. Using this assay, the nature of the systemic signalling system (‘PIIF’) was investigated. Hydraulic signals were shown to be induced in tomato by localized wounds. These signals travelled throughout the plant well within the lag time before appearance of systemic wound-induced PI. A number of correlations were drawn between the occurrence of the hydraulic signals and induction of systemic PI, suggesting that hydraulic signals might be the PIIF, or a component of it. It was shown that systemic hydraulic signals could be triggered, without significant wounding, by excision of a single leaflet through the submerged petiole. These hydraulic signals were similar in both kinetics and magnitude to those induced by localized wounding. However, they did not induce systemic PI. In addition, it was shown that systemic events almost as rapid as wound-induced hydraulic signals could be induced without wounding, under certain environmental conditions. This indicates that rapid hydraulic signals do not provide a specific signal of wounding. These findings demonstrate that hydraulic signals per se are not the PIIF.  相似文献

GA1, GA8, GA17, GA19, GA20 and GA29 were identified by combined gas chromatography-mass spectrometgry (GC-MS) in immature seeds and pericarp of Lycopersicon esculentum Mill. (tomato). Higher levels of these GAs were present in the seeds than in the pericarp; seeds in addition contained GA15, GA24, GA25, and GA44. Fruits of the Lycopersicon pimpinellifolium Mill. mutant I were smaller and contained lower GA1 concentrations, but higher GA20 concentrations, than those of mutants III and IV. In contrast, differences in fruit size in L. esculentum due to position on the truss did not correlate with GA1 concentration in either the pericarp or seeds.  相似文献

Abstract Etiolated hypocotyls from normal tomato plants show a negative gravitropic response within 20 min of stimulation. In contrast, etiolated hypocotyls from the gravitropic mutant Lazy-l do not reorientate after gravistimulation. Etiolated hypocotyls from both types of plant are positively phototropic, however, Lazy-l seedlings achieve a greater final angle of bending following phototropic stimulation compared to normal plants. Anatomical studies reveal that etiolated hypocotyls from normal plants contain sedimenting amyloplasts located within the endodermal cells. Such sedimenting amyloplasts are absent in Lazy-l tissue. It is hypothesized that the hypocotyl of Lazy-l is agravitropic since it is unable to perceive a gravistimulus.  相似文献

Development of galactomannan hydrolyzing activity was followed in seeds of tomato [ Lycopersicon esculentum (L.) Mill. cv. Toyonishiki] during priming and germination. The activity developed in seeds that were being primed in polyethylene glycol (-0.8 MPa). The activity was detected exclusively in the endosperm portion just adjacent to the radicle tip. Part of the activity remained active after desiccation of the primed seeds. After transfer to water, the activity in the primed seeds immediately began to increase, while in unprimed seeds the beginning of the increase in activity was delayed by about 1 day. In scanning electron microscopy, the inner surface of the cell walls of the micropylar endosperm portion appeared eroded in primed seeds that had been imbibed in water for 16 h (before germination), but not in unprimed seeds imbibed for the same period. These results support the hypothesis that galactomannan hydrolyzing enzyme, which is believed to be responsible for breakdown of tomato endosperm cell walls and hence for the weakening of mechanical restraint against radicle growth, may be involved in the improved germination of primed tomato seeds.  相似文献

Development of galactomannan-hydrolyzing activity, that is involved in the weakening of the mechanical restraint of the endosperm, was followed at pre-germinative stages in tomato ( Lycopersicon esculentum ) seed. Prior to germination the activity developed exclusively in the endosperm portion just adjacent to the radicle tip. In other parts of the endosperm, the activity developed only after germination occurred. Under the conditions where germination was suppressed (far-red light- or ABA-treatment). no activity was detected in the endosperm at the pre-germinative stages. Under the conditions where the inhibition of germination was alleviated (far-red + red or ABA + GA3), the activity developed prior to germination in the endosperm part in front of the radicle tip. Thus, a clear parallel relationship was observed between germinability of the seed and the pre-germinative development of activity in the part of the endosperm portion adjacent to the radicle tip.  相似文献

Broad-band UV-B radiation inhibited hypocotyl elongation in etiolated tomato ( Lycopersicon esculentum Mill. cv. Alisa Craig) seedlings. This inhibition could be elicited by < 3 μmol m−2 s−1 of UV-B radiation provided against a background of white light (> 620 μmol m−2 s−1 between 320 and 800 nm), and was similar in wild-type and phytochrome-1-deficient aurea mutant seedlings. These observations suggest that the effect of UV-B radiation is not mediated by phytochrome. An activity spectrum obtained by delivering 1 μmol m−2 s−1 of monochromatic UV radiation against a while light background (63 μmol m−2 s−1 showed maximum effectiveness around 300 nm, which suggests that DNA or aromatic residues in proteins are not the chromophores mediating UV-B induced inhibition of elongation. Chemicals that affect the normal (photo)chemistry of flavins and possibly pterins (KI, NaN, and phenylacetic acid) largely abolished the inhibitor) effect of broad-hand UV-B radiation when applied to the root zone before irradiation. KI was effective at concentrations < 10−4 M , which have been shown in vitro to be effective in quenching the triplet excited stales of flavins but not fluorescence from pterine or singlet states of flavins. Elimination of blue light or reduction of UV-A, two sources of flavin excitation, promoted hypocotyl elongation, but did not affect the inhibition of elongation evened by UV-B. Kl applied after UV-B irradiation had no effect on the inhibition response. Taken together these findings suggest that the chromophore of the photoreceptor system invoked in UV-B perception by tomato seedlings during de-etiolation may be a flavin.  相似文献

We investigated the influence of an increased inorganic carbon supply in the root medium on NO?3 uptake and assimilation in seedlings of Lycopersicon esculentum (L.) Mill. cv. F144. The seedlings were pre-grown for 4 to 7 days with 0 or 100 mM NaCl in hydroponic culture using 0.2 mM NO?3 (group A) or 0.2 mM NH+4 (group B) as nitrogen source. The nutrient solution for group A plants was aerated with air or with air containing 4 800 μumol mol?1 CO2. Nitrate uptake rate and root and leaf malate contents in these plants were determined. The plants of group B were subdivided into two sets. Plants of one set were transferred either to N-free solution containing 0 or 5 mM NaHCO3, or to a medium containing 2 mM NO?3 and 5 mM NaHCO3. Both sets of group B plants were grown for 12 h in darkness prior to 2 h of illumination, and were assayed for malate content and NO?3 uptake rate (only for plants grown in N-free solution). The second set of group B plants was labeled with 14C by a 1-h pulse of H14CO?3 which was added to a 5 mM NaHCO3 solution containing 0 or 100 mM NaCl and 0 or 2 mM NO?3, and 14C-assimilates were extracted and fractionated. The roots of group B plants growing in carbonated medium accumulated twice as much malate as did control plants. This malate was accumulated only when NO?3 was absent from the root medium. Both a high level of root malate and aeration with CO2-enriched air stimulated NO?3 uptake. Analysis of 14C-assimilates indicated that with no NO?3 in the medium, the 14C was present mainly in organic acids, whereas with NO?3, a large proportion of 14C was incorporated into amino acids. Transport of root-incorporated 14C to the shoot was enhanced by NO?3, while the amino acid fraction was the major 14C-assimilates in the shoot. It is concluded that inorganic carbon fixed through phosphoenolpyruvate carboxylase (EC 4.1.1.31) in roots of tomato plants may have two fates: (a) as a carbon skeleton for amino acid synthesis; and (b) to accumulate, mainly as malate, in the roots, in the absence of a demand for the carbon skeleton. Inorganic carbon fixation in the root provides carbon skeletons for the assimilation of the NH+4 resulting from NO3 reduction, and the subsequent removal of amino acids through the xylem. This ‘removal’ of NO?3 from the cytoplasm of the root cells may in turn increase NO?3 uptake.  相似文献

Abstract. Deuterium-labelled ABA-aldehyde was fed to various tomato genotypes. Normal and notabilis mutant plants incorporated substantial amounts of the label into ABA. In contrast, two ABA-deficient mutants, flacca and sitiens , reduced ABA-aldehyde to a mixture of cis- and trans -ABA alcohol rather than oxidizing it to ABA. It was concluded that ABA-aldehyde is the immediate precursor of ABA in higher plants. It appears that the flacca and sitiens lesions both act to block the last step of the ABA biosynthetic pathway. The mutant gene loci are likely to be involved in coding for different sub-units of the same dehydrogenase enzyme.  相似文献

This work investigated how calcium regulates the ethylene biosynthesis in the fruits of wild-type tomato (Lycopersicon esculentum L.) and their ethylene receptor never-ripe (Nr) mutants. In Nr tomato, the ethylene perception was blocked. When both materials were treated with calcium, the content of 1-aminocyclopropane-1-carboxylic acid (ACC)/malonyl-ACC and the activity of ACC oxidase (ACO) in tomato fruit discs increased, whereas the production of ethylene, content of malondialdehyde, and membrane permeability decreased. Calcium treatment did not affect the activity of ACC synthase, which is the first committed step in the ethylene biosynthesis pathway. The expression of LeACO1 in mature green fruit was inhibited significantly by calcium treatment in wild-type and Nr tomatoes, but the expression of LeACS2, the key ACC synthase gene in ethylene synthesis during tomato fruit maturing, was not affected. These results revealed that the effect of calcium on ethylene biosynthesis in tomato mature green fruit was independent of ethylene perception. The results also revealed that the targeting step of calcium preventing ethylene production was located at the ACC conversion to ethylene, by means of inhibiting ACC availability for ACO through enhancing cell membrane integrity and by means of preventing LeACO1 gene expression. UV-B radiation inhibits hypocotyl elongation in etiolated tomato (Lycopersicon esculentum Mill. cv. Alisa Craig) seedlings acting through a photoreceptor system with peak apparent effectiveness around 300 nm. In order lo further characterize the response and gain insight into its potential ecological significance, the time-course of inhibition was measured and compared with the time-course of flavonoid accumulation in the same seedlings. When a background of strong (> 620 μmol m?2 s?1) white light (WL) was supplemented with low irradiance UV-B (~ 3 μmol m?2 s?1). substantial (~ 50%) inhibition of elongation occurred within 3 h of the light treatment. The magnitude of UV-B-induced elongation inhibition was similar in wild type (WT) and au-mutant seedlings, in spite of the large differences between genotypes in rate and temporal pattern of elongation. In comparison to the effect of UV-B on elongation, induction of flavonoid accumulation in WT and au seedlings undergoing de-etiolation was a much slower response. Several UV-absorbing compounds appeared to be specifically induced by light, and some of them accumulated faster under the WL + UV-B treatment than under WL alone. However, there was little or no delectable effect of WL on flavonoid levels until up to 3 h of treatment, and the specific UV-B effect was measurable only after 6 h of continuous treatment. Indeed. UV-B-screening properties of crude alcoholic extracts were not different between WL and WL + UV-B treatments until after 9 or 24 h. When the light treatments were applied to seedlings that were just breaking through the soil surface. UV-B was found to consistently retard seedling emergence. These results suggest that the rapid inhibition of elongation in de-etiolating seedlings is an evolved response lo UV-B, which may serve to minimize seedling exposure to sunlight until protective pigmentation responses (triggered by WL and UV-B) have taken place in the seedlings epidermis.  相似文献

An approach commonly employed to assess the potential role of the enzyme polygalacturonase (PG, EC 3.2.1.15) in tomato fruit cell-wall pectin metabolism includes correlating levels of extractable PG with changes in specific characteristics of cell wall pectins, most notably solubility and molecular weight. Since information on these features of pectins is generally derived from analyses of subfractions of isolated cell wall, assurance of inactivation of the various isoforms of wall-associated PG is imperative. In the present study, cell wall prepared from ripe tomato (Lycopersicon esculentum Mill. cv. Rutgers) fruit was examined for the presence of active PG and for the ability of phenolic solvents to inactivate the enzyme. Using pectin solubility and Mr (relative molecular mass) changes as criteria for the presence of wall-associated PG activity, pectins from phenol-treated and nonphenol-treated (enzymically active) cell wall from ripe fruit incubated in 50 mM Na-acetate, 50 mM cyclohexanetrans-1,2-diamine tetraacetic acid (CDTA), pH 6.5 (outside the catalytic range of PG), were of similar Mr and exhibited no change in size with incubation time. Wall prepared without exposure to the phenolic protein-denaturants exhibited extensive pectin solubilization and depolymerization when incubated in 50 mM Na-acetate, 50 mM CDTA at pH 4.5, indicating the presence of active PG. Based on the changes in the Mr of pectins solubilized in 50 mM Na-acetate, 50 mM CDTA, pH 4.5, active PG was also detected in wall exposed during isolation to phenolacetic acid-water (PAW, 2:1:1, w/v/v), a solvent commonly employed as an enzyme denaturant. Although the depolymerization of pectins in PAW-treated wall was extensive, oligouronides constituted minor reaction products. Interestingly, PAW-treated wall did not exhibit PG-mediated pectin release when incubated under conditions (30 mM Na-acetate, 150 mM NaCl, pH 4.5) in which nonphenol-treated cell wall exhibited high autolytic activity. In an alternative protocol designed to inactivate PG, cell wall was exposed to Tris-buffered phenol (BP). In contrast to pectins released from PAW-treated wall, pectins solubilized from BP-treated wall at pH 4.5 were indistinguishable in Mr from those recovered from BP-treated wall at pH 6.5 Even when incubated at pH 4.5 at 34°C, conditions under which pectins from PAW-treated wall underwent more rapid and extensive depolymerization, pectins from BP-treated wall exhibited no change in Mr, providing evidence that active PG was not present in these wall preparations. The implications of this study in interpreting the solubility and Mr of pectin in cell wall from ripening fruit are discussed.  相似文献

NaCl-induced changes in the accumulation of message for the 70 kDa subunit of the tonoplast H+-ATPase and plasma membrane H+-ATPase were studied in hydroponically grown plants of Lycopersicon esculentum Mill. cv. Large Cherry Red. There was increased accumulation of message for the 70 kDa (catalytic) subunit of the tonoplast H+-ATPase in expanded leaves of tomato plants 24 h after final NaCl concentrations were attained. This was a tissue-specific response; levels of this message were not elevated in roots or in young, unexpanded leaves. The NaCl-induced accumulation of this message was transient in the expanded leaves and returned to control levels within 7 days. The temporal and spatial patterns of NaCl-induced accumulation of message for the plasma membrane H+-ATPase differed from the patterns associated with the 70 kDa subunit of the tonoplast H+-ATPase. NaCl-induced accumulation of the plasma membrane H+-ATPase message occurred in both roots and expanded leaves. Initially accumulation of the plasma membrane H+-ATPase message was greater in root tissue than in expanded leaves, but increased to higher levels in expanded leaves after 7 days. These results suggest that increased expression of the tonoplast H+-ATPase is an early response to salinity stress and may be associated with survival mechanisms, rather than with long-term adaptive processes.  相似文献

Soil solarization is a non-chemical disinfestation technique that frequently promotes plant growth in the absence of known major pathogens, a phenomenon termed increased growth response (IGR). The effect of solarization on plant nutrients and their role in the IGR was studied with tomato plants grown in solarized or non-solarized (control) sandy soil, under controlled conditions. Solarization considerably increased the soil concentrations of water extractable N, K, Ca, Mg and Na at most sites, whereas Cl and DTPA extractable Mn, Zn, Fe and Cu were decreased by the treatment. Plant growth and specific leaf area were enhanced in solarized as well as in N-supplemented control soil. In tomato plants grown in solarized soil, concentrations of most nutrients in the xylem sap, including N, were increased compared to the control, whereas Cl and SO4 levels decreased. The most significant increase in leaf nutrient concentration caused by soil solarization was recorded for N. Furthermore, leaf N concentration was highly and positively correlated with shoot growth. The concentration of Cu increased in leaves from the solarization vs. the control treatment, whereas that of SO4 and Cl decreased, the latter presumably below the critical toxicity level. The correlation between shoot growth and leaf concentration was positive for Cu and inverse for Cl and SO4. In conclusion, we found that soil solarization significantly affects nutrient composition in tomato plants, and provided strong evidence that N, and eventually also Cl, play a major role in IGR.  相似文献

The photo-inhibition of Lycopersicon esculentum Mill, hypocotyl growth induced by UV radiation may be mediated by both phytochrome and UV-absorbing receptors. The inhibition of growth induced by continuous irradiation with high fluence rate UV radiation is similar in the au mutant, which is severely deficient in spectrophoto metrically and immunochemically detectable phytochrome, and in the isogenic wild type. Parallel irradiation with 692 nm light, which is equivalent to UV radiation for the phytochrome system in our experimental conditions, induced at high photon fluence rates a significant increase in hypocotyl growth in the au mutant. The same light treatments inhibited the hypocotyl growth of the wild type. The responses of water-grown seedlings and chlorophyll-free seedlings (streptomycin and norflurazon treated seedlings) were compared. Water-grown and chlorophyll-free seedlings responded similarly to UV radiation. The presence of chlorophyll correlates with a significant increase in hypocotyl growth of au mutants irradiated with 692 nm light. These results support the conclusion that UV-induced inhibition of growth in the au mutant is independent of phytochrome.  相似文献

The uniflora (uf) mutant of tomato (Lycopersicon esculentum Mill.) is known to produce solitary, normal, fertile flowers instead of inflorescences. Histological and SEM studies revealed that this unusual reproductive structure resulted from the inability of the plant to produce an inflorescence and not from post-initiation abortion processes affecting young flower buds. Development prior to floral transition was apparently not affected by the mutation since rates of germination and leaf initiation were identical in both uf and the Ailsa Craig (AC) initial cultivar. However, the time of flowering of the mutant was always delayed as compared to AC. In uf, environmental conditions markedly influenced flowering time which occurred early in all individuals in summer, but was strongly delayed during winter, with less than 20% plants reaching flowering before having initiated 40 leaves. Defoliation treatments stimulated floral transition in uf plants since 100% flowering occurred whatever the season and since the time of floral transition was usually advanced in comparison to the non-defoliated control plants. Similarly, compared to intact uf plants, flowering of terminal meristem of cuttings and upper axillary bud of decapitated plants was promoted. The involvement of correlative influences and assimilate availability in the control of flowering in tomato is suggested by these findings.  相似文献
