We studied genetic differentiation of two charr species, Dolly Varden Salvelinus malma malma Walbaum and resident lacustrine charr Salvelinus sp., which sympatrically inhabit Nachikinskoe Lake (the Bol'shaya River basin) in southwestern Kamchatka Peninsula. Using restriction analysis (RFLP), three mitochondrial DNA fragments (ND1/ND2, ND5/ND6, and Cytb/D loop) amplified in polymerase chain reaction (PCR) were compared. The divergence of the mtDNA sequences between Salvelinus sp. and S. malma malma was 2.8%; Salvelinus sp. and S. taranetzi, 0.36%; Salvelinus sp. and S. krogiusae, 0.21%; Salvelinus sp. and S. alpinus, 3.0%. These results point to reproductive isolation of charrs in Nachikinskoe Lake and support the earlier suggestion on a close relationship between Salvelinus sp., S. taranetzi, and S. krogiusae. 相似文献

In previously identified sympatric char forms (species) from Kronotsky Lake (Kamchatka Peninsula) and a number of other chars from the genus Salvelinus, nucleotide sequences of the mtDNA cytochrome b gene were determined and phylogenetic analysis of these forms was performed. The divergence of the cytochrome b gene in the Kronotsky char group (0.48%) coincided with the interpopulation difference estimates in northern Dolly Varden, which indicates a relatively recent divergence of the chars from Kronotsky Lake. Haplotypes of each form of the Kronotsky Lake chars are divided into two groups, which belonged to two different phylogenetic mtDNA lineages of northern Dolly Varden from Chukotka and Kamchatka. The formation of the Kronotsky char forms proved to be unrelated to the observed variability of the mtDNA cytochrome b gene. 相似文献

Chars of the genus Salvelinus, inhabiting lakes and lake-river systems, comprise morphological and ecological forms whose taxonomic status is under dispute. In the present work, we have examined genetic variation and divergence in various chars from the Kronotsky lake basin: the lacustrine chars (white, nose, and longhead) and riverine Dolly Varden Salvelinus malma. The study was conducted using analysis of allozyme and microsatellite loci, myogens, RAPD, and restriction analysis of two mtDNA segments. The estimates of heterozygosity at allozyme and microsatellite loci were similar to the corresponding parameters in populations of northern Dolly Varden and Arctic char. Heterozygote deficit was recorded in both samples of separate forms, and in the total sample of all chars from Kronotsky Lake. For allozyme and microsatellite loci, appreciable genetic differentiation among the samples of different char forms was found, which was comparable to that among the spatially isolated populations of northern Dolly Varden. This result indicates reproductive isolation among the char forms examined. However, this isolation is not complete, because no fixed differences between the forms by any of the genetic systems analyzed were found. Using the isozyme analysis, genetic variations in six species of the genus Oxytropis DC. (Fabaceae) from Kamchatka was assessed. It was demonstrated that diploid species from the section Arctobia were characterized by a low level of variations typical of endemic plant species. At the same time, polyploid species from the Orobia section demonstrated very high values of the heterozygosity parameters (H o varied from 0.200 to 0.274). It has been suggested that the level of polymorphism of the oxytropes from Kamchatka was shaped as a result of the interaction of a number of factors, among which the most important are the ecological confinedness of the species, the specific features of the reproductive system, and gene drift. In the species of Orobia section, it is also the presence of the polyploid genome. 相似文献

Resident (lacustrine) chars, inhabiting many lakes of Kamchatka, Chukotka, and continental coast of the Sea of Okhotsk, are of particular interest for understanding the mechanisms of speciation and evolution of Salvelinus chars. Since one of these, the char from Dal'nee Lake (Paratunka River basin, southeastern Kamchatka), is substantially different from lacustrine-riverine and anadromous chars from the Paratunka River basin in several morphological traits, hemoglobin spectrum, ecology, and reproduction, it is considered an isolated species Salvelinus krogiusae. The karyotype of the resident char from Dal'nee Lake was shown to be variable in most individuals, containing 76 to 80 chromosomes in different cells, with NF = 98. This suggests that the variability of the chromosome number in this char form is explained by Robertsonian translocations. In females and males, cells with respectively 2n = 78 and 2n = 77 prevailed. Heteromorphic sex chromosomes were not found in the karyotype of this char species. Nucleolar-organizing regions (NORs) were mostly detected in the telomeric regions of short arms of large submetacentric chromosomes. These features of the Krogius char karyotype demonstrate that this species is isolated from other Kamchatka chars, being similar to Taranetz char S. taranetzi from Chukotka Peninsula, which is in good agreement with morphological data. 相似文献

A sequence of a D-loop segment of mitochondrial DNA with the length of 558 bp was determined for 224 specimens of char (genus Salvelinus) from the Kronotskoe river-lake system, and a sequence of the cytochrome b gene with the length of 1015 bp was determined for 212 specimens of the same kind. On a joint sequence with the length of 1573 bp (1?C558 bp of D-loop and 559?C1573 bp of cytochrome b), 39 variable sites and 43 different haplotypes were found. Nucleotide sequences of two (405- and 1050-bp) regions of mitochondrial DNA (mtDNA) cytochrome c gene were established in chars of the genus Salvelinus from Russian Far East and Siberia. Based on the divergence and phylogenetic analysis of nucleotide sequences of the mtDNA cytochrome c gene, S. laecomaenis was shown to carry the most ancient mitochondrial lineage, which is close to the ancestral one. The archaic mtDNA of S. levanidovi occupied an isolated position on the phylogenetic trees. The mtDNA lineage of the southern S. malma was close to the S. alpinus-S. malma complex group. Within the S. alpinus-S. malma complex, three groups of mtDNA types having particular geographic distributions were distinguished. The Kolyma-Chukotka group includes lake S. taranetzi, S. boganidae and S. elgyticus from Chukotka, lake chars from Kolyma. The Okhotsk group is represented by northern S. malma, lake chars from northern Sea of Okhotsk, and anadromous S. taranetzi. The Siberian group is close to the Okhotsk one and consists of Taimyr and Baikal region chars as well as Arctic char from Finland. The divergence of char mitochondrial lineages was dated to the Pliocene-Pleistocene. 相似文献

The populations of mykizha Parasalmo (O.) mykiss from western and eastern coasts of Kamchatka were studied by restriction analysis of a fragment of fish mitochondrial genome that included the control region and the region of the cytochrome b gene (cytb). The restriction patterns obtained with five enzymes (MspI; Tru1I; RsaI; BsuRI; DdeI) were identical in all studied individuals. Sequencing of the cytb gene showed high similarity between all samples (99.6-100%). In general, the geographical group of mykiss from Kamchatka is monophyletic with low genetic divergence at the population level. Shantarian mykiss originates most likely from that native to Kamchatka. 相似文献

A parasitological study was performed of chars of the genus Salvelinus inhabiting Lake Kronotskoe (Kamchatka Peninsula)—S. malma, S. albus, S. schmidtii, and S. kronocius, as well as of juvenile Salvelinus spp. Twenty-three species of parasites, including six species new for the lake, Hennequya zschokkei, Protteocephalus longicollis, Diphyllobothrium dendriticum, Crepidostomum sp., Echinorhynchus salmonis, and Paracanthobdella livanowi, were found. With consideration of published data, in chars of this water body, 28 species of parasites were recorded, including seven species (N. cf. pungitius, B. luciopercae, Crepidostomum sp., Cr. fausti, Cr. cf. cooperi, Eubothrium crassium, and Proteocephalus sp.), whose presence or species identification in the lake ecosystem need confirmation. Two species (N. rutili and Diphyllobothrium sp.) are removed from the list. Parasites common for all species of chars were revealed. They include Myxobolus arcticus, E. salvelini, D. ditretum, Crepidostotum sp., Cr. farionis, Cr. metoecus, Cystidicola farionis, Cucullanus truttae, Philonema oncorhynchi, and Salmincola carpionis. Cluster analysis of the fauna of parasites of different species of chars demonstrated considerable differences in infestation, which indicates differences between them in preference for food items and occupied biotopes and thereby supports the ecological differentiation of chars in the basin of Lake Kronotskoe. S. albus and S. kronocius are most similar in parasitofauna, which is determined by their predation; S. malma as a benthophage is infected by the same species of parasites, but considerably less intensively. Extremely high indices of population numbers of some parasite species are considered as a manifestation of the Krebs cycle in parasites under the conditions of an isolated lake. 相似文献

To reveal genetic differentiation and origin of sympatric Lake Kronotskoe charr morphs, we sequenced a fragment of control region (558-bp) and Cytochrome B gene (1015-bp) (combined length 1573-bp) in 232 specimens from the lake and, as an outgroup, 57 specimens from the Kronotskaya River below water-falls (anadromous malma) and 77 specimens from other large water bodies from the Kamchatka. A total of 52 nucleotide positions were polymorphic, and these defined 67 haplotypes. Our data does not allow to estimate genetic differentiation among different lake-dwelling morphs, but strongly supports the monophyletic origin of all morphs within the lake. To estimate genetic differentiation and population structure, an extensive study of microsatellite variation has to be conducted. 相似文献

Genetic variation in several populations of Parasalmo (Onchorhyncus) mykiss from Kamchatka was examined on the basis of data obtained by the author and results from literature. Three (sSOD-1*, LDH-C*, and EST-1*) out of 44 protein-coding loci were highly polymorphic. Low-frequency alternative alleles occurred at sAAT-1,2*, LDH-A2*, EST-5*, IDDH-1,2*, and sMDH-1,2*. The results of the present work and comparison with evidence on North American species indicated that, in the Kamchatka part of the range, P. mykiss is represented by several populations carrying unique alleles and forming a genetically independent group. Gene exchange between North American and Western Kamchatka populations is mainly determined by straying in the feeding stock of the American coastal form entering the Sea of Okhotsk. The genetic divergence and mean heterozygosities in the Kamchatka populations were low (D magnitude of 0.0002-0.0275; HS magnitude of 0.011-0.0371). The difference between the Western Kamchatka populations from the North American coastal form was so small (D magnitude of 0.0109-0.0241) that these forms clustered together. Genetic divergence of the Kamchatka populations and the inland North American P. mykiss is an order of magnitude higher (D magnitude of 0.1973-0.2367). 相似文献

Restriction enzyme analysis was employed in studying the mitochondrial DNA (mtDNA) ATPase6/ND4L region in several Northeast Asian populations of resident lake char of the genus Salvelinus. On evidence of mitotypes, genetic similarity was assumed for populations of neiva (Ueginskoe Lake), lake resident char from the northern coast of the Sea of Okhotsk (Mak-Mak Lake and Elekchanskie lakes), and dolly varden. Mitotype AAAA proved to be common for these populations. Lake char populations of the Juliet and Maxi lakes (the basin of the Kolyma river) had mitotype DBAA, which is similar to mitotype DBAB observed earlier in Taranetz char S. taranetzi from Chukotka. The divergence between mitotypes AAA and DBAA was estimated at 0.3%. Different origins were assumed for the lake resident char populations from the basins of the Sea of Okhotsk and of the Kolyma River, the former originating in the Pacific and the latter, in the Arctic basins. 相似文献

Smelts of the genus Osmerus, O. eperlanus and O. mordax dentes, inhabiting the basins of the Atlantic, Arctic, and Pacific oceans were investigated using RFLP analysis of the mitochondrial DNA segments A8/A6/COIII/ND3 and ND3/ND4, and sequencing of the cytb and COI genes (mtDNA), and intron 1 of the rpS7 gene (nDNA). A total of 14 samples from the populations from most part of the Russian range were examined. The mean values of haplotype and nucleotide diversity constituted 0.5997 +/- 0.11264 and 0.003201 for O. m. dentex, and 0.3086 +/- 0.09892 and 0.000431 for O. eperlanus, respectively. The high level of interspecific diversity (12.94%) along with the low level of intraspercific diversity (0.049% for O. m. dentex, and 0.001% for O. eperlanus was observed. The dendrograms (UPGMA, NJ, MP, and BA) constructed using the data of RFLP analysis of mtDNA, along with the sequencing data of mitochondrial and nuclear genes were congruent. The representatives of O. eperlanus and O. m. dentex formed steady clusters in accordance with their species affiliation, albeit without subdivision into local populations depending on their geographic locality. 相似文献
