The interactions of cellular and humoral factors of hemolymph of the American oyster, Crassostrea virginica, and several species of marine cercariae were studied. Attraction of hemocytes to dead but not to living cercariae was observed. Dead cercariae were encapsulated in vitro by oyster hemocytes. The plasma of C. virginica was apparently not toxic to the species of cercariae tested.  相似文献

Perkinsus marinus is a major cause of mortality in eastern oysters along the Gulf of Mexico and Atlantic coasts. It is also well documented that temperature and salinity are the primary environmental factors affecting P. marinus viability and proliferation. However, little is known about the effects of combined sub-optimal temperatures and salinities on P. marinus viability. This in vitro study examined those effects by acclimating P. marinus at three salinities (7, 15, 25 ppt) to 10 °C to represent the lowest temperatures generally reached in the Gulf of Mexico, and to 2 °C to represent the lowest temperatures reached along the mid-Atlantic coasts and by measuring changes in cell viability and density on days 1, 30, 60 and 90 following acclimation. Cell viability and density were also measured in 7 ppt cultures acclimated to each temperature and then transferred to 3.5 ppt. The largest decreases in cell viability occurred only with combined low temperature and salinity, indicating that there is clearly a synergistic effect. The largest decreases in cell viability occurred only with both low temperature and salinity after 30 days (3.5 ppt, 2 °C: 0% viability), 60 days (3.5 ppt, 10 °C: 0% viability) and 90 days (7 ppt, 2 °C: 0.6 ± 0.7%; 7 ppt, 10 °C: 0.2 ± 0.2%).  相似文献

Range expansion and population establishment of individual species can have significant impacts on previously established food webs and predator-prey dynamics. The stone crab (Menippe spp.) is found throughout southwestern North Atlantic waters, from North Carolina through the Gulf of Mexico and the Central American Caribbean, including the Greater Antilles. Recent observations suggest that stone crabs have become better established on certain oyster reefs in North Carolina than in the early 1900s when they we first observed in NC. To assess the predatory impact of stone crabs on oysters, we (1) quantified stone crab densities on subtidal oyster reefs in Pamlico Sound, NC using scuba surveys, and (2) conducted laboratory predation experiments to assess the functional response of stone crabs to varying densities of oysters. We then (3) analyzed previously unpublished functional response data on another important oyster predator, the mud crab Panopeus herbstii. Finally, we (4) compared and contrasted potential predatory impacts of stone, mud and blue crabs (Callinectes sapidus). The functional response data and analyses for both stone crabs and mud crabs were consistent with a type II functional response. Mud crabs, on a m2 basis, inflicted the highest proportional mortality on oysters over a 24 hour period, followed by stone and then blue crabs. Proportional mortality did not vary significantly with oyster size; however, relatively small and large oysters were consumed disproportionately less than medium-sized oysters, likely due to the mechanical inability of stone crabs to handle small oysters, and the inability to crush large oysters. Although stone crabs appear to be established in Pamlico Sound at densities equivalent to densities in other systems such as the U.S. Florida Panhandle, their predatory activities on oysters are not expected to have as significant a negative impact on oyster populations compared to other resident predators such as mud crabs.  相似文献

The Eastern oyster, Crassostrea virginica, may improve water quality by filtering large quantities of particulate matter (both organic and inorganic) and nutrients from the overlying water column. Additionally, oyster reefs alter hydrodynamic conditions, further increasing the removal of particulate matter from the water column. This study examined the effects of small-scale oyster additions on sediment loading, chlorophyll a, nutrient concentrations, and flow in small tidal creeks. Two reefs were established in Hewletts Creek, New Hanover County, North Carolina. Total suspended solids (TSS), chlorophyll a, and ammonium were measured upstream and downstream of each created reef and in an adjacent control channel that lacked a reef. Data were collected monthly during ebb tides over a 10-month period between September 2000 and June 2001. In the first month after initial reef placement, mean TSS concentrations downstream of reef placement were slightly lower than those upstream of the reef. Although not statistically significant, TSS concentrations downstream of the reefs were less than upstream concentrations for five out of nine and five out of seven post-reef sampling months for the upland and the lower creek sites, respectively. Chlorophyll a concentrations were not significantly affected by initial reef placement (2×3 m), but were reduced substantially after reef enlargement (3×4 m) in one of the experimental creeks. Reef placement resulted in significant increases in ammonium concentrations downstream of the transplanted-reefs. In addition, deposition of feces and pseudofeces by the oysters resulted in accumulation of finer-grained materials in the treated channel relative to the control channels. Oyster filtration was most effective three hours following high tide, when the ratio of flow discharge to reef surface area was the highest. This work demonstrates that small oyster reefs established and maintained in some small tributary channels can reduce TSS and chlorophyll a concentrations and that the magnitude of the effect may vary over the course of the tidal cycle.  相似文献

Lamellar transformations of cytosomes, DOPA decarboxylase and lateral ciliary activity were measured in neuronal components and/or ctenidia of the oyster, Crassostrea virginica (Gmelin). Prolonged exposure to light, anoxic conditions, EGTA (a calcium chelator), and exogenous perfusion of dopamine (a cilioinhibitory neurotransmitter) increases the frequency of lamellar type cytosomes. This is accompanied by decreases in enzymatic activity and a lateral cilioinhibition. Prolonged periods of darkness, electrical stimulation of the branchial nerve and exogenous perfusion of serotonin (a cilioexcitatory neurotransmitter) reduces the conversion of lamellar type configurations, increases enzymatic activity and produces a lateral cilioexcitation. Salinity changes that produce an increase to higher acclimation values result in a gradual decrease in the number of lamellar configurations with a subsequent increase in enzymatic activity and lateral cilioexcitation. The magnitude of the lateral cilioexcitation-cilioinhibition and decarboxylase activity is directly related to the number of lamellar conversions.  相似文献

Combined effects of acclimation temperature (12, 20 and 28 °C) and exposure to a toxic metal cadmium (Cd, 50 μg L−1) on haemolymph parameters related to immune defense and metal transport were studied in a model marine bivalve, Crassostrea virginica. Acclimation to elevated temperatures resulted in higher plasma protein concentrations and increased Cd levels in oyster haemolymph plasma and haemocytes. Cd accumulation in haemocytes was linear over the 45 days of Cd exposure and accumulation rates were 0.10, 0.53 and 0.56 μg Cd g−1 dry mass at 12, 20 and 28 °C, respectively. Percentage of blood Cd burden associated with haemocytes increased with increasing temperatures from 13–20% at 12 °C to 26–47% at 20 and 28 °C suggesting a higher role for cellular Cd transport at elevated temperatures. Cd levels in gills and hepatopancreas were positively correlated with Cd concentration in haemocytes, but accumulation rates were considerably faster, so that after 45 days of exposure Cd levels in gills and hepatopancreas were >10–20 times higher than in haemocytes. As a result of slow Cd accumulation possibly reflecting fast haemocyte turnover rates and/or exocytosis of Cd-containing granules, haemocytes in Cd-exposed oysters did not reach threshold Cd burdens required to trigger apoptosis. This suggests that haemocyte viability is not likely to contribute to immunosuppression in the environmentally relevant Cd range. In contrast, elevated temperature (28 °C) resulted in a significant increase in the percentage of apoptotic haemocytes compared to 12 or 20 °C supporting the notion that 28 °C is physiologically stressful for C. virginica. Overall, our study demonstrates strong effects of environmental temperature on haemocyte viability and other important blood parameters such as plasma protein content and metal transport capability which may mask potential Cd effects at environmentally relevant exposure levels.  相似文献

The red pigment produced by a marine Pseudomonas sp. which causes abnormal development and mortality in developing embryos of the American oyster, Crassostrea virginica, was analyzed. A comparative study of a nonpigmented and two pigmented mutants of the red parental strain indicated that virulence was associated and varied with pigmentation. The use of sonicated cells supported lysing of the pseudomonad cells as the most probable means of pigment release. Crude pigment extracted from the red parental strain and its yellow mutant was toxic to developing oyster embryos. Neither the “pigment” extracted from the white mutant nor dimethyl sulfoxide, used for dissolving the extract, was toxic. Three pigment fractions were demonstrated by thin-layer chromatography even after purification. Studies indicate that only the fraction corresponding to R1 0.41 was necessary for virulence. The virulent pigment fraction was identified as belonging to the prodiginine group.  相似文献

Five families of gynogenetic diploid Pacific oyster (Crassostrea gigas) were induced by inhibiting the second polar body in meiotic cell division of eggs fertilized with UV-irradiated sperm. Segregation patterns of eight microsatellite loci were investigated in the gynogenetic diploid offspring; the proportion of heterozygous progeny was used to estimate microsatellite-centromere (M-C) distances. Mendelian inheritance was confirmed for the eight loci by examining the genotypic segregation in the control crosses. Three of the eight microsatellite loci showed the existence of null alleles in four control crosses. All gynogenetic offspring only possessed the alleles of the mother, indicating 100% success level for the five families. The M-C recombination frequency estimates ranged from 0.62 to 0.77 (0.72 mean), comparable to those in the oyster based on allozyme markers and suggesting that meiotic gynogenesis does not appear to be a very efficient inbreeding method in the oyster. Recombination frequencies observed were often higher than the theoretical maximum of 0.67, indicating the existence of positive interference after a single chiasma formation in some chromosomes. Information on the positions of centromeres in relation to the microsatellite loci will represent a contribution toward assembly of genetic maps in C. gigas.  相似文献

A study has been made of the processes of extra- and intracellular digestion in the Pacific oyster, Crassostrea gigas (Thunberg) over two 24-h cycles in winter and summer.The results show that the digestive processes are discontinuous and related to the tidal cycle. Variations in tidal height resulting from a diurnal inequality of the tide affect both the relative dissolution of the crystalline style and the relative lengths of the constituent phases of the intracellular digestive process in the digestive diverticula. On a seasonal basis the style is present for a greater length of time in winter and, conversely, remains dissolved longer in summer. A seasonal variation in the structure of the digestive tubules has also been found.The results confirm conclusions reached earlier that the processes of extracellular digestion in the stomach and intracellular digestion in the digestive diverticula of intertidal bivalves are both discontinuous, alternate, and irrevocably interrelated since breakdown of the digestive cells of the digestive diverticula following intracellular digestion results in the dissolution of the crystalline style. The arrival of food in the stomach has a minimal effect upon the style. Moreover, the cytological structure of the digestive diverticula of C. gigas undergoes a series of synchronized changes related to the tidal cycle.  相似文献

The interaction is reported of selected chemical elements (cadmium, calcium, copper, iron, magnesium, manganese, strontium, and zinc) in cultured sea water, with soft tissues, prismatic calcite of the right valve, and foliated calcite of right and left valves of genetically similar American oysters, Crassostrea virginica (Gmelin) grown in a natural habitat and in two environmentally controlled experimental systems (flow-through and recycle). The addition of trace elements as algal nutrients in ambient sea water was reflected in higher concentrations of trace metals in shells and soft tissues of oysters grown in experimental systems. Calcium was relatively uniformly distributed in major regions of valves from the three habitats, even though its concentration fluctuated widely in sea water in experimental systems. Magnesium and strontium were most concentrated in valves of oysters grown in the recycle system (magnesium in the prismatic layer of the shell and strontium in the foliated calcite). Iron was uniformly distributed. Cadmium, copper, manganese, and zinc were most concentrated in the prismatic calcite of valves from the flow-through system. In soft tissues, calcium was more concentrated in oysters from experimental systems than in those from the natural habitat. Manganese was about equally distributed in soft tissues from the three habitats, whereas copper and iron were more concentrated in soft tissues in experimental systems than in the natural habitat, and were many times more concentrated in soft tissues than in valves from all three habitats. As concentrations of magnesium, strontium, mangenese, zinc, and cadmium increased in valves in experimental systems, pigmentation of valves decreased. The study confirmed the capacity of oysters to concentrate several elements in their valves as concentration of these elements increased in ambient sea water and disclosed the heterogeneous distribution of these elements in major regions of the valves.  相似文献

The levels of aminopeptidase activity in the serum and hemolymph cells of the American oyster, Crassostrea virginica, have been determined to be 2.89 ± 2.25 and 0.62 ± 0.39 Sigma units/ml, respectively. Low levels of aminopeptidase activity have been demonstrated cytochemically within cytoplasmic granules, i.e., secondary phagosomes, of circulating hemolymph cells. Exposure of whole hemolymph to heat-killed Bacillus megaterium or to sterile sea water results in a significant increase in cellular aminopeptidase activity, with the level of activity being the highest in cells that had been exposed to bacteria. The level of aminopeptidase activity in serum is unaltered in similarly challenged whole hemolymph. It is concluded that aminopeptidase synthesized in cells during phagocytosis or as a result of stimulation by exposure to sea water is not released into the serum but is retained intracellularly. Our studies suggest that it is the intracellular and not the serum aminopeptidases whith are of primary importance in the degradation of B. megaterium.  相似文献

Changes in sperm features during the movement phase are especially interesting to study in external fertilization species whose sperm duration movement is long because this implies a significant adaptation of moving cells to the external medium. This study describes the changes in tetraploid Pacific oyster sperm characteristics in relation to time post activation.Sperm individually collected on three tetraploid males were activated in seawater. Their features were analysed over a 24 h period and compared to a sperm pool collected on three diploid males as a reference. The percentage of motile spermatozoa, the intracellular ATP content, and the fine structure of spermatozoa were studied in relation to time post activation. Furthermore, the fertilisation capacity of sperm individually collected on five diploid males was assessed after 1 and 24 h post activation.A forward progressive movement was maintained for at least a 20 h duration. Compared to diploid males, the percentage of motile spermatozoa was lower in tetraploid males. The intracellular ATP concentration was higher in spermatozoa from tetraploid males than in spermatozoa from diploid males. A decrease in ATP content was observed in the first 6 h post activation and severe alterations were observed in sperm morphology after 24 h. Then, a lower fertilisation capacity of sperm from diploid males was observed at the end of the movement phase.The cessation of Pacific oyster sperm motility was unlikely caused by ATP consumption as ATP concentration was still high at the end of sperm movement but rather caused by drastic changes in sperm morphology. Compared to sperm collected on diploid males, the lower quality of sperm from tetraploid males was emphasized by a shorter movement duration and deeper morphological alterations at the end of the movement phase.  相似文献

Cadmium, zinc and copper, accumulated from polluted habitats by the oysters Ostrea edulis and Crassostrea gigas, were studied. Three distinct low molecular weight (less than 3000 daltons) zinc complexes were separated from highly contaminated C. gigas, one of which may be a complex with the amino acid taurine. Only one of these zinc complexes was present in less contaminated specimens of O. edulis. On gel permeation chromatography of oyster extracts, copper was eluted together with amino acids (principally taurine) and the betaine homarine. No evidence of metallothionein type proteins was found.  相似文献

Embryos of the oyster, Crassostrea gigas (Thunberg), can take up [14C]glycine from micromolar concentrations, but unfertilized eggs do not. Activation of uptake mechanisms coincides with the first cell division of the fertilized egg. Uptake of glycine is inhibited by ouabain, an inhibitor of the sodium pump. This study, when combined with earlier work, demonstrates that bivalves utilize dissolved organic nutrients throughout their life span, from fertilized egg to adulthood.  相似文献

Bivalve molluscs, in common with consumers in general, use behavioral and physiological mechanisms to balance metabolic requirements with available nutrients. This study considered how the Pacific oyster, Crassostrea gigas, meets the demands of growth and maintenance, measured in terms of carbon and nitrogen, in a variable food environment. Stoichiometry theory helped to evaluate: a) whether feeding behaviour modifies the intake of C and N given seasonal variability in food quality: b) how rates of metabolism and excretion, and C and N growth efficiencies, respond to mismatch between nutrient intake and the oysters' needs. Two field experiments in the Port Stephens estuary, near Sydney, Australia, measured feeding behaviour, metabolic and growth rates relative to seasonal changes in food supply. In a laboratory experiment, relationships between physiological rates and growth were measured to test a model of growth as a function of absorption of C and N. Potential metabolic targets for compensation were the C/N ratios of body tissues, maintenance and/or of soft tissue added as growth. C/N of whole soft issues varied little during the year (mean 5.4). In July (a time of low food availability of poor quality) growth was negligible and the C/N (maintenance) target was 6.7. In March (abundant food of high quality) growth was rapid with a high N-demand; the C/N of growth was 3.9. In November (medium food quality) there was an enhanced C-demand for glycogen storage; the C/N of growth was 7.9. Feeding behaviour changed the balance between C and N intake across months, primarily due to changes in the selection efficiency for nitrogen, which was highest at low filtration rates on particles of high C/N ratio. Nitrogen intake was favoured over C in July. In November, C-intake increased relative to N. In March, when abundant food nitrogen coincided with a high demand for growth, feeding behaviour was neutral with respect to C/N ratios. In all cases C/N of absorbed matter was greater than the C/N of growth. Growth efficiencies for carbon declined with increased C/N of ingested matter due to higher metabolic increments (SDA) when feeding on lower food quality; the metabolic costs of growth did not vary. In contrast, growth efficiencies for nitrogen did not alter with C/N for ingested matter, due in part to increased nitrogen losses, relative to tissue nitrogen content, when feeding on low C/N food. Nitrogen was therefore conserved metabolically relative to C. Both feeding and metabolic processes contributed to compensation for the mismatch between seasonally variable food quality and the demands of growth.  相似文献

The involvement of algal chemical cues in the pre-ingestive selection of food particles in Crassostrea gigas was studied using a new approach. Live cells of two microalgal species, Nitzschia closterium and Tetraselmis suesica, were separately entrapped in small alginate microcapsules using an emulsification/internal gelation method. Microcapsule size was adjusted to be within the range of particles ingested by oysters. Using this technique, about 80% of microcapsules had a diameter ranging from 21 to 100 μm. The monitoring of entrapped algae showed that phytoplankton cells remained alive and maintained an active growth for at least 24 days. In particle selection bioassays, adult C. gigas were fed a mixture of microcapsules containing the above algae species as well as control empty alginate microcapsules. The comparison of the proportions of each microcapsule type in the diet and in pseudofeces revealed that those containing T. suesica were significantly ingested while those containing N. closterium were preferentially rejected. Since microcapsule material (alginate matrix) prevented physical contacts between algae cells and oyster feeding organs, this study clearly demonstrate that extracellular metabolites produced by microalgae play a crucial role in the pre-ingestive selection of particles in suspension-feeding bivalves.  相似文献

Oysters Crassostrea gigas were placed at water supply canals of three shrimp farms in Guasave, Mexico where WSSV outbreaks occur. Animals were sampled through April-August and September-December to detect WSSV DNA.By using three different PCR protocols, only oysters from a farm undergoing a WSSV outbreak were found WSSV-positive in gills and digestive gland. Two WSSV amplicons were sequenced and they corresponded over 99% to WSSV genome segments. Results showed that oysters can capture WSSV particles suspended in water. Susceptibility of oysters to WSSV infection and their role as a carrier remain to be determined.  相似文献
