The susceptibility of members of the Anopheles sinensis Group in Korea to insecticides was evaluated under laboratory conditions using 15 insecticides currently used by local public health centers in Korea. The insecticides included eight pyrethroids, six organophosphates and a pyrazol analogue. Based on their LC50 values, the order of susceptibility of An. sinensis larvae to the insecticides was bifenthrin, chlorfenapyr, α-cypermethrin and λ-cyhalothrin, with values of 0.009, 0.04, 0.06 and 0.08 p.p.m., respectively. The least susceptibility was obtained with fenitrothion, with an LC50 of 7.7 p.p.m. In the comparative resistance test, the resistance ratios (RR) of 14 insecticides were compared to each other using two strains of members of the An. sinensis Group collected in the locality in 2001 and 2008. Anopheles spp. demonstrated higher RR to organophosphates such as fenthion, and low RR for the pyrethroids. Among the organophosphates, fenthion had the highest RR of 33.3 and 270.0 fold differences for LC50 and LC90 values, respectively. Among the pyrethroids, permethrin was observed to have the highest RR of 3.8 and 1.8 fold differences for LC50 and LC90 values, respectively. However, there were no significant differences in susceptibility to chlorfenapyr, chlorpyrifos, deltamethrin and fenitrothion. An. sinensis s. l. was more susceptible to the six insecticides bifenthrin, λ-cyhalothrin, α-cypermethrin, cypermethrin, cyfluthrin and pyridafenthion, showing 0.03, 0.06, 0.3, 0.3, 0.4 and 0.4 fold differences in resistance rates (RR LC50), respectively.  相似文献

The susceptibility of Culex tritaeniorhynchus collected from Gwangju, Jeollabuk Province, Republic of Korea (ROK) to insecticides was evaluated under laboratory conditions using ten insecticides (7 pyrethroids and 3 organophosphates) that are currently applied by local public health centers in the ROK. Based on the values of median lethal concentration (LC50), Cx. tritaeniorhynchus larvae were most susceptible to chlorpyrifos (0.006 ppm), fenitrothion (0.022 ppm), fenthion (0.035 ppm) and bifenthrin (0.038 ppm), and were least susceptible to esbiol (1.722 ppm). In comparative resistance tests, the resistance ratios (RRs) of seven insecticides were compared among each other using two strains of Cx. tritaeniorhynchus that were collected from the same locality during 1992 and 2010. Culex tritaeniorhynchus demonstrated significantly increased RRs to pyrethroids over time, while demonstrating decreased RRs among the organophosphates. Among the pyrethroids, permethrin had the highest RR values of 182.1‐ and 833.3‐fold differences, followed by etofenprox with RRs of 138.4‐ and 224.1‐fold differences in values of LC50 and concentration that produced 90% mortality (LC90), respectively. Culex tritaeniorhynchus strains demonstrated the least amount of change in susceptibility to the organophosphates, chlorpyrifos, fenitrothion and fenthion with 0.020‐, 0.019‐ and 0.001‐fold differences in resistance ratios (RRLC50), respectively.  相似文献

Toxicity of bendiocarb, chlorpyrifos, cyfluthrin, cypermethrin, fenvalerate, hydramethylnon, malathion, propetamphos, propoxur, and pyrethrins against the adult German cockroaches, Blattella germanica (L.), was investigated. At LD50, cyfluthrin was the most toxic insecticide to adult males (0.53 microgram/g), adult females (1.2 micrograms/g), and gravid females (0.85 microgram/g). Malathion was the least toxic insecticide to adult males (464.83 micrograms/g), adult females (335.83 micrograms/g), and gravid females (275.90 micrograms/g). Males and gravid females were generally more sensitive than nongravid females to the insecticides that we tested. In tests with malathion, however, males were more tolerant. The order of toxicity of the insecticide classes varied among the stages of adult German cockroaches. The order of toxicity for males and nongravid females was pyrethroids greater than pyrethrins = organophosphates (except malathion) greater than carbamates = amidinohydrazone. The order of toxicity for gravid females was pyrethroids greater than pyrethrins = organophosphates (except malathion) greater than carbamates greater than amidinohydrazone. These differences in toxicity suggest that sex differences should be considered when determining insecticide toxicity for German cockroaches.  相似文献

Acute toxicity studies of emamectin and spinosad against Helicoverpa armigera revealed that the pest is highly susceptible to both the insecticides. The median lethal dose (LD50) of emamectin is 3.86 × 10−3 µg per larva. The median lethal concentrations (LC50) of emamectin and spinosad were found to be 0.09 and 2.94 ppm, respectively. The discriminating doses were fixed based on the LC95 of the susceptible population of H. armigera as 0.80 ppm for emamectin and 10 ppm for spinosad. Resistance was not observed when the discriminating doses of emamectin and spinosad were applied on field-collected populations of H. armigera from two intensive cotton growing areas, Coimbatore and Madurai, India.  相似文献

The essential oil from leaves of Majorana hortensis Moench (Lamiaceae) was isolated by hydrodistillation with a yield of 1.6% (wt/wt). The insecticidal activity of the oil was evaluated against fourth instars of Spodoptera littoralis Boisduval (Lepidoptera: Noctuidae) and adults of Aphis fabae L. (Hemiptera: Aphididae). The oil showed a remarkable toxic effect against S. littoralis in a topical application assay (LD50 = 2.48 μg per larva) and in a residual film assay (LC50 = 3.14 g/l). The oil of M. hortensis also exhibited a pronounced toxic effect against A. fabae adults with LC50 values of 1.86 and 2.27 g/l in rapid dipping and residual film assays, respectively. Gas chromatography-mass spectrometry analyses of M. hortensis essential oils revealed the presence of 31 compounds and the main components were terpinen-4-ol (30.0%), γ-terpinene (11.3%), and trans -sabinene hydrate (10.8%). Repeated column chromatography of M. hortensis oil on silica gel led to the isolation of two major constituents, which were characterized based on 1H-nuclear magnetic resonance and mass spectrometric data, as terpinen-4-ol and γ-terpinene. These two components were examined for their insecticidal and synergistic activities towards S. littoralis and A. fabae . Terpinen-4-ol and γ-terpinene exhibited a significant insecticidal activity against both insects, but γ-terpinene was more toxic than terpinen-4-ol. When tested in a binary mixture with the synthetic insecticides profenofos and methomyl, it was found that both compounds enhanced the insecticidal activity of these insecticides by two- to threefold. These results show that terpinen-4-ol and γ-terpinene have a synergistic effect on the insecticidal activities of synthetic insecticides profenofos and methomyl.  相似文献

Abstract  Lycoriella ingenua is one of the major pests of cultivated mushrooms, Agaricus bisporus (Lange) Imbach . Insecticide resistance among mushroom sciarid populations has been reported from other countries, and there is a need to determine the toxicity of currently approved and potential pesticides to sustain control of mushroom sciarid populations in Australia. The present study investigated the toxicity of six commercial formulations of insecticides or biopesticides against third instar larvae of L .  ingenua using laboratory bioassays. Insecticide treatments were incorporated into the growing medium for sciarid larvae and the concentration of the pesticide, which killed 90% of the test population (LD90) determined the efficacy of selected insecticides. Triflumuron was the most effective insecticide against L. ingenua with an LD90 of 53.12 mg active ingredient (a.i.)/m2 followed by cyromazine (LD90, 179.68 mg a.i./m2) and diazinon (LD90, 261.72 mg a.i./m2). Abamectin and Bacillus thuringiensis ssp. israelensis were ineffective against L. ingenua . Steinernema feltiae , an entomopathogenic nematode, reduced the number of third instar larvae of L. ingenua only when applied at a higher rate (LD90, 732 422 nematodes/m2) than was recommended on the label.  相似文献

Dopamine Neurotoxicity: Inhibition of Mitochondrial Respiration   总被引:15,自引:6,他引:9  
Abstract: Dopamine, due to metabolism by monoamine oxidase or autoxidation, can generate toxic products such as hydrogen peroxide, oxygen-derived radicals, semiquinones, and quinones and thus exert its neurotoxic effects. Intracerebroventricular injection of dopamine into rats pretreated with the monoamine oxidase nonselective inhibitor pargyline caused mortality in a dose-dependent manner with LD50 = 90 µg. Norepinephrine was less effective with LD50 = 141 µg. The iron chelator desferrioxamine completely protected against dopamine-induced mortality. In the absence of pargyline more rats survived, indicating that the products of dopamine enzymatic metabolism are not the main contributors to dopamine-induced toxicity. Biochemical analysis of frontal cortex and striatum from rats that received a lethal dose of dopamine did not show any difference from control rats in lipid and protein peroxidation and glutathione reductase and peroxidase activities. Moreover, dopamine significantly reduced the formation of iron-induced malondialdehyde in vitro, thus suggesting that earlier events in cell damage are involved in dopamine toxicity. Indeed, dopamine inhibited mitochondrial NADH dehydrogenase activity with IC50 = 8 µ M , and that of norepinephrine was twice as much (IC50 = 15 µ M ). Dopamine-induced inhibition of NADH dehydrogenase activity was only partially reversed by desferrioxamine, which had no effect on norepinephrine-induced inhibition. These results suggest that catecholamines can cause toxicity not only by inducing an oxidative stress state but also possibly through direct interaction with the mitochondrial electron transport system. The latter was further supported by the ability of ADP to reverse dopamine-induced inhibition of NADH dehydrogenase activity in a dose-dependent manner.  相似文献

Abstract.  Insect myoinhibitory peptides were discovered through their inhibitory activity on visceral muscle contraction. The present study tests the antimyotropic gut properties of three galanin-related myoinhibitory peptides (Mas-MIP II: GWQDLNSAW-NH2; Grb-AST-B1: GWQDLNGGW-NH2; and Grb-AST-B3: AWRDLSGGW-NH2) in adult females of the cockroach Blattella germanica (L.) (Dictyoptera, Blattellidae). The three peptides elicit a strong inhibitory effect on both foregut and hindgut contractions, with ID50 values in all the cases within the nanomolar range. In addition, the modulatory effects of these three peptides on food intake are studied on previously starved female cockroaches. The results show that Grb-AST-B3 is the most active peptide, inhibiting food intake by 60–80% at doses between 15 and 50 µg, followed by Grb-AST-B1 (45% inhibition of food intake at the 50 µg dose), whereas Mas-MIP II is inactive even at the 50 µg dose. The differences between the three peptides may be due to a differential effect of their structure on activity or to a differential degradation. These results show that myomodulatory gut activity in vitro and antifeeding effects do not always correlate.  相似文献

The toxicity of some of the most commonly used insecticides in the organophosphate and pyrethroid classes were investigated against different Chrysoperla carnea (Stephens) (Neuroptera: Chrysopidae) populations collected over three consecutive years (2005-2007). The populations were tested using leaf dip bioassays for residual effects and topical applications to measure the response of larvae that would come into direct contact with field application of insecticides. In leaf dip assays, the LC50 (micrograms per milliliter; 120 h) values for chlorpyrifos and profenofos were in the range of 59.3-1,023 and 180.02-1,118 respectively. The LC50 values for lambda-cyhalthrin, alphamethrin, and deltamethrin were 359.08-2,677, 112.9-923.5, and 47.81-407.03, respectively. The toxicity for the above insecticides in topical application was similar to toxicity in leaf dip assays. The susceptibility of a laboratory population, which was locally developed and designated as (Lab-PK), to deltamethrin was comparable with another susceptible laboratory population. Resistance ratios for five field populations were generally low to medium for deltamethrin, but high to very high for chlorpyrifos, profenofos, lambda-cyhalthrin and alphamethrin compared with the Lab-PK population. Our data also suggested that the five field populations had multiple resistance to two classes of insecticides. The populations showed resistance to two organophosphates tested and to lambda-cyhalthrin and alphamethrin; however, resistance to deltamethrin was only found at two locations. This pattern indicates occurrence of two divergent patterns of resistance within pyrethroids. The resistance to the insecticides was stable across 3 yr, suggesting field selection for general fitness had also taken place in various populations of C. carnea. The broad spectrum of resistance and stability of resistance to insecticides in C. carnea in the current study suggested that it could be a prime candidate for mass releases and compatible with most spray programs.  相似文献

The susceptibility values for field‐collected (BG‐BG) and laboratory (KS‐BG) strains of German cockroaches (BS‐BG), Blattella germanica (L.) to five synthetic pyrethroid and three organophosphate insecticides were assessed by topical application. Both KS‐BG and BS‐BG strains demonstrated the highest levels of susceptibility to deltamethrin with LD50 values of 0.0035 μg/female and 1.5744 μg/female, respectively. The KS‐BG strain demonstrated the lowest susceptibility level to fenthion with an LD50 value of 0.0698 μg/female, while the BS‐BG field population showed the lowest susceptibility to bifenthrin with 4.6811 μg/female. The BS‐BG field population showed resistance levels of 50 to 640 fold higher against the eight pesticides evaluated compared to the KS‐BG strain. The resistance ratios (RR) of the eight pesticides were compared using populations of B. germanica that were collected from the same locality during 2007 and 2014. The field population of B. germanica collected during 2014 demonstrated significantly high resistance to an organophosphates and all pyrethroids when compared with those collected during 2007, but RR values were <10 except for chlorpyrifos‐methyl and chlorpyrifos with RR values of 74 and 104, respectively.  相似文献

The interrelationship of two extrinsic factors-nutrition and pesticide exposure-was studied in German cockroaches, Blattella germanica (L.). Amount of dietary protein had no effect on susceptibility of male German cockroaches to propoxur and chlorpyrifos; however, the source of protein produced significant changes in insecticide susceptibility. Increased total body lipids, carbohydrates, and uric acid may have resulted in increased tolerance to these insecticides. Three days of food or water deprivation (or both) caused significant differences in susceptibility to chlorpyrifos and propoxur.  相似文献

Introduction. The susceptibility of Anopheles albimanus to organophosphates, carbamates and pyrethroid insecticides was unknown in the Panama communities of Aguas Claras, Pintupo and Puente Bayano, located in the Amerindian Reservation of Madungandi. This region is considered a malaria transmission area, where An. albimanus is the main vector. Objective. The resistance to organophosphate insecticides, carbamates and pyrethroids was evaluated in field populations of the Anopheles albimanus in Panama. Materials and methods. Progeny of An. albimanus collected in three localities in the indigenous Madugandi region were exposed to bioassays of susceptibility to organophosphate insecticides (fenitrothion, malathion and chlorpyrifos), the carbamate (propoxur) and pyrethroids (deltamethrin, lambdacyhalothrin, cyfluthrin and cypermethrin). The protocols were in accordance with those established for adult mosquitoes by World Health Organization. Results. The three strains of the An. albimanus were resistant to the pyrethroid insecticides deltamethrin, lambdacyhalothrin, cyfluthrin and cypermethrin. Susceptibility remained for the organophosphate insecticides fenitrothion, malathion, chlorpyrifos, and the carbamate insecticide propoxur. Conclusion. The results provided important information to the vector control program, contributing to the application of new strategies on the use of insecticides, and thereby lengthening the life of the insecticide in use.  相似文献

The pathogenicity of 20 strains of Listeria was determined with the mouse intravenous bio-assay and the 10-day chick embryo chorioallontoic membrane test. In the former, survival in the spleen was measured and in the latter, the LD100 and LD50. There was good correlation between the results of the two tests. Listeria monocytogenes strains that grow in the mouse spleen had an LD100 of < 125 organisms, while strains in which the numbers of organisms in the mouse spleen remain constant had an LD100 > 125 organisms. Listeria seeligeri, L. innocua, L. welshimeri, L. grayi and L. murrayi did not persist in the spleen and the numbers of organisms used did not kill chick embryos.  相似文献

Abstract The chick embryo model was evaluated as a method to compare virulence between selected strains of Neisseria meningitidis . Inoculation of 13-day-chick embryos via the egg yolk distinguished strains having an LD50 of 103 colony forming units (CFU) or greater (low virulence) from those having an LD50 of approximately 101 or less (high virulence). A strain of serogroup B and a spontaneous nonpiliated strain of group C were found to be of relatively high virulence while a strain of N. lactamica , a serogroup A carrier strain, and certain nongroupable strains were found to be of low virulence. Strains having an LD50 of 102 were not differentiated from either of these. Alternatively, inoculation of the chorioallantoic membrane (CAM) of 9-day-old chick embryos statistically differentiated most strains of N. meningitidis although inoculation via this route was less sensitive.  相似文献

We determined nonlethal physiological effects of hydroprene on the German cockroach, Blattella germanica L., and the effects of hydroprene exposure on German cockroach susceptibility to traditional insecticides. Live and dry weights of German cockroach adults exposed as nymphs to the juvenile hormone analog hydroprene increased significantly (6-35%) in both sexes. The change in weight was affected by hydroprene dose and time after adult emergence. Hydroprene treatment did not affect total body lipids and carbohydrates consistently. However, total body urates of males and females increased significantly. The susceptible and multiresistant German cockroach strains were significantly more susceptible to propoxur at the lower hydroprene application rate (0.697 mg per tub) than the untreated cockroaches. Hydroprene application also significantly lowered the LD50 for chlorpyrifos of the multiresistant strain at the lower hydroprene application rate. However, hydroprene failed to affect chlorpyrifos susceptibility of the susceptible strain.  相似文献

Abstract: In cultured bovine adrenal medullary cells, stimulation of nicotinic receptors by carbachol evoked the Ca2+-dependent exocytotic cosecretion of proadrenomedullin N-terminal 20 peptide (PAMP) (EC50 = 50.1 µ M ) and catecholamines (EC50 = 63.0 µ M ), with the molar ratio of PAMP/catecholamines secreted being equal to the ratio in the cells. Addition of PAMP[1–20]NH2 inhibited carbachol-induced 22Na+ influx via nicotinic receptors (IC50 = 2.5 µ M ) in a noncompetitive manner and thereby reduced carbachol-induced 45Ca2+ influx via voltage-dependent Ca2+ channels (IC50 = 1.0 µ M ) and catecholamine secretion (IC50 = 1.6 µ M ). It did not alter high K+-induced 45Ca2+ influx via voltage-dependent Ca2+ channels or veratridine-induced 22Na+ influx via voltage-dependent Na+ channels. PAMP seems to be a novel antinicotinic peptide cosecreted with catecholamines by a Ca2+-dependent exocytosis in response to nicotinic receptor stimulation.  相似文献

Abstract Virulent and avirulent strains of Aeromonas spp. were identified and virulence quantified using an animal model. Virulence was measured by determining a 50% lethal dose (LD50) 43 h after oral administration of live bacteria. The LD50 of virulent Aeromonas isolates ranged from log10 7.53 (mean) organisms to log10 8.88 (mean). Some isolates were avirulent in this model. Detection of cytotoxic activity in culture supernatants correlated with virulence (Fisher exact test, P = 0.0029). There was no correlation between LD50 and the source of the isolate, β-haemolysis or lipopolysaccharide (LPS) banding profile on SDS-PAGE. In this animal model, virulence was multifactorial in that: (i) bacterial multiplication in the gut was associated with fatal infection; (ii) the increase in bacterial numbers in the gut of mice administered a lethal dose of bacteria was accompanied by accumulation of fluid; and (iii) there was evidence of extraintestinal spread of infection. Protection of suckling mice by rabbit antiserum to Aeromonas cell envelopes was observed.  相似文献

The aim of the currrent investigation was to evaluate (a) the toxicity of three pyrethroids (deltamethrin, lambda-cyhalothrin, and tetramethrin); (b) the effect of these insecticides on the locomotor activity; and (c) the repellent effect of N,N-diethyl-m-toluamide (DEET) on two deltamethrin-resistant strains of Triatoma infestans from Argentina (El Chorro and La Toma), and one susceptible strain. The resistance ratios (RRs) obtained for the La Toma strain were: > 10,769, 50.7, and > 5.2 for deltamethrin, lambda-cyhalothrin, and tetramethrin respectively. The RRs for the El Chorro strain were: > 10,769, 85.8, and > 5.2 for deltamethrin, lambda-cyhalothrin, and tetramethrin respectively. The hyperactivity usually caused by the three pyrethroids was in both the deltamethrin-resistant strains compared to the susceptible reference strain. No differences were observed in the repellent effect of DEET between the three groups. These results indicate that the deltamethrin-resistant insects have a cross resistance to lambda-cyhalothrin and tetramethrin, and are also resistant to the first symptom of pyrethroid poisoning (hyperactivity). However, the sensorial process related to DEET repellency does not appear to be altered.  相似文献

Abstract:  To investigate fluctuation in susceptibility to insecticides, natural populations of Plutella xylostella were collected from the same field in the region of Multan, Pakistan, in late summer, mid-winter and early spring. After bulking up for a generation in the laboratory, the populations were examined against pyrethroids and organophosphates as well as newer insecticides (spinosad, indoxacarb and emamectin). Each population showed a broad range of variation in susceptibility to all chemicals. Comparison between populations collected in different times of years in 2003 and 2004 showed variation in susceptibility to organophosphates and pyrethroids. In winter, susceptibility to organophosphates increased, whereas it decreased in relation to pyrethroids. However, susceptibility to the newer insecticides was unchanged within the season. The results suggest that the fluctuation observed in susceptibility could be either associated with fitness costs of organophosphate and pyrethroids or cessation of insecticides. These results might have important implications in resistance management. We propose a strategy for application of insecticides in relation to P. xylostella management in Pakistan.  相似文献

采用浸液生测法研究了斜纹夜蛾Spodoptera litura巴基斯坦抗性种群中酶抑制剂[胡椒基丁醚(PBO)和脱叶膦(DEF)]对丙溴磷、灭多威、硫双灭多威、氯氰菊酯、氯氟氰菊酯、联苯菊酯、茚虫威和多杀菌素等杀虫剂的增效作用。结果表明:PPO和DEF对氨基甲酸酯杀虫剂灭多威和硫双灭多威均具有增效作用,但对有机磷杀虫剂丙溴磷不具有增效作用。两种抑制剂对氯氰菊酯均产生增效作用,但对联苯菊酯没有增效作用。PPO 和DEF增加了氯氟氰菊酯对Multan种群的毒性,但没有增加其对Mailsi种群的毒性。DEF对多杀菌素具有增效作用,但PBO对其没有增效作用。PBO和DEF对氨基甲酸酯杀虫剂、拟除虫菊酯杀虫剂、茚虫威和多杀菌素具有明显的增效作用,这说明细胞色素P450单加氧酶和酯酶的解毒作用至少部分参与了斜纹夜蛾对这些杀虫剂的抗性过程。不过,两种增效剂对杀虫剂增效作用范围有限,暗示对于斜纹夜蛾巴基斯坦种群而言,其他的机制(如靶位点不敏感、表皮穿透作用降低)可能是更重要的抗性机制。  相似文献
