Rats Krushinsky-Molodkina inbred strain (KM) genetically prone to audiogenic seizures were injected with pineal hormone melatonin (50 mg/kg, s.c.) within the period 7th to the 14th posnatal days (PND). The remote effects of this injection adult KM rats consisted in a decrease in the latency and increase in severity of myoclonic seizures produced by audiogenic kindling (20 sound stimuli, 100 dB and 12-15 kHz). As compared to the control, in the cortex and hippocampus of rats of melatonin group, we also found a significant reduction of both total and functional activity of Ca2+/calmodulin-dependent protein kinase II (CAMK II) after audiogenic kindling. On the contrary, melatonin administration within the 1st to 7th PND and the 14th to the 21st PND resulted in a decrease in seizure activity. In the first case, both the total (cortical) and functional (hippocampal) CAMK II activities in melatonin-injected rats were increased as compared to control, whereas in the second case, only a slight increase in Ca2+-independent CAMK II activity in the hippocampus of melatonin-injected rats was observed. Probably, the melatonin administration in the period of early postnatal development changes the features of expression and/or regulation of CAMK II activity, and this could be one of the mechanisms of audiogenic seizure modulation in KM rats.  相似文献

Mosquito biting frequency and meal size are considered to be important parameters in the epidemiology of insect‐vectored diseases such as malaria. Because both parameters are likely to depend on the size and energetic state of adult mosquitoes, the present study investigates the effects of body size and energy state on attack behaviours in the malaria mosquito, Anopheles gambiae. Attack rates are measured as well as total time spent before giving up for individual females when provided with an unobtainable human hand (i.e. mosquitoes are dislodged every time that they land). The factorial design considers two body sizes, small and large, as well as three sugar deprivation states, 0, 1 and 2 days. The results reveal a positive effect of size on attack rate and a nonlinear effect of energy state, where mosquitoes of intermediate energy state show lower attack rates than either 2‐day food‐deprived or nondeprived mosquitoes. Moreover, attack rate is negatively associated with persistence time in nondeprived and 2‐day food‐deprived Anopheles but is unrelated to persistence time in 1‐day food‐deprived mosquitoes, Interestingly, although persistence times are generally inversely related to attack rates, they are not significantly influenced by either energetic or size states.  相似文献

The latency of tonic seizure in response to loud sound (in rats of the Krushinsky–Molodkina strain with audiogenic epilepsy) had been slightly (although statistically significantly) longer after chronic uridine injections (100 mg/kg, i.p., three times a day during 9 or 12 days). The recovery time from the tonic seizure was shorter after 12 days of injections in comparison to the 9-day injection period. At the same time, the intensity of tonic seizures provoked by loud sound did not change after chronic uridine injections. The lack of uridine anticonvulsive effect demonstrated in the audiogenic epilepsy model contradicts the anticonvulsant effects of uridine in experiments with other seizure models, in which the epileptic foci were localized in the forebrain structures.  相似文献

Two experiments were conducted to examine the effects of depriving dairy cows of the ability to feed and lie down for short periods, on behaviour and production. In experiment 1, cows were deprived by confining them in pairs in a pen for 2 or 4 h, and they more frequently exhibited behaviour likely to suggest discomfort - leg stamping, repositioning themselves, shifting their weight between legs and butting. After deprivation, the cows deprived for 2 h made up their lost feeding time within 24 h, but cows deprived for 4 h did not restore their feeding time within the 41-h period of observation. Lying time was not restored in either treatment within the 41-h period. Milk yield was not affected by the treatment. However, in experiment 2, when cows were deprived of feeding and lying for 4 h, during which time their hooves were trimmed (which is likely to be a painful and stressful procedure and result in some discomfort for a period post-trimming) the evidence suggested that milk yield was reduced by approximately 2 l/day for 3 days, with corresponding increases during the subsequent 2 days. Walking speed on returning to the herd was the same as before the treatment. In summary, temporary deprivation of feeding and lying for 2 and 4 h/day induced behaviours that were indicative of discomfort and frustration but had no negative effect on milk production, except when 4 h of deprivation was accompanied by foot trimming.  相似文献

The serum level of magnesium and calcium was systematically measured in patients with gallstones before and after cholecystectomy. It was found that 60 percent of the operated patients suffered of different digestive syndromes in association with magnesium deficiency, while 40 percent of patients had the same complaints in association of magnesium and calcium deficiency. When magnesium and/or magnesium plus calcium was supplemented these syndromes could be decreased significantly. In the latter case, an optimal ratio of magnesium/calcium is needed in the supplementary therapy.  相似文献

In adult Wistar, KM, and Wag/Rij rats, the threshold of pain sensitivity (tail-flick test) and degree of spasm attack in response to a strong sound were estimated after neonatal administration of Semaks (analog of ACTG4-10 fragment) or after placebo (administration of saline for the control of the effect of neonatal pain stimulation). These neonatal influences did no affect the rates of sensorimotor maturation at an early age (Fox tests), i.e., did not affect directly the physiological activity of rat pups at the age of up to 21 days. In all control rats injected with saline (pain stimulation), the latent periods of audiogenic attacks increased reliably, while their degree decreased. Administration of Semaks "raised" these parameters to the lvl of those in intact animals, i.e., increased the sensitivity to sound. Neonatal administration (per os) of caffeine to KM rats increased reliably the latent period of audiogenic attacks. The thresholds of pain sensitivity in the rats of all strains were significantly lower than in the intact control, just as the level of dopamine in the hippocampus of KM rats. These data are interpreted as an evidence of changes in the development of some brain systems in response to neonatal influences.  相似文献

The magnesium ion-requiring step in fertilization of sea urchins was investigated. When eggs were inseminated in Mg-free sea water, several spermatozoa were found to bind to each egg surface with their reacted acrosomes without elevation of fertilization membrane. The number of binding jelly-treated spermatozoa to an egg did not differ regardless of the presence or virtual absence of magnesium ions. Although fertilization did not occur in Ca, Mg-deficient sea water (CM-deficient SW) even when jelly-treated spermatozoa were employed, some eggs could be fertilized by the addition of magnesium to the CM-deficient SW 60 sec after insemination, when jelly-treated spermatozoa had completely lost their fertilizing capacity in the CM-deficient SW. The acrosomal process of jelly-treated spermatozoa appeared to penetrate the vitelline layer in the CM-deficient SW. DTT- or pancreatin-treated eggs could not be fertilized in the virtual absence of magnesium. Re-fertilization using the fertilized eggs deprived of fertilization membrane did not occur under conditions of magnesium deficiency. These results suggest that external magnesium ions are indispensable at least for the fertilization process following penetration of the vitelline layer by the spermatozoa, such as fusion of the plasma membrane between an egg and a reacted spermatozoon, or the subsequent step(s) such as sperm penetration into egg interior and egg activation which precedes the cortical reaction.  相似文献

Abstract: DBA/2 mice between 21 and 28 days of age are highly susceptible to sound-induced seizures. Drug studies suggest a possible deficit of γ-Aminobutyric acid (GABA)-mediated neurotransmission may be involved. We have measured the whole brain GABA concentration and glutamic acid decar-boxylase activity in DBA/2 mice at various ages before, during, and after the period of maximal susceptibility to audiogenic seizures. Corresponding determinations were carried out on age-matched TO mice, a strain much less susceptible to audiogenic seizures than DBA/2 mice at all ages. No significant differences in GABA concentration or glutamic acid decarboxylase activity were found between strains at any age. The susceptibility of DBA/2 mice to audiogenic seizures does not result from a gross inability to synthesise or store GABA.  相似文献

An experiment with rats was conducted to determine whether magnesium retention is increased and calcium utilization is altered by a marginal zinc deficiency and whether increased oxidative stress induced by a marginal copper deficiency exacerbated responses to a marginal zinc deficiency. Weanling rats were assigned to six groups of ten with dietary treatment variables of low zinc (5 mg/kg for 2 weeks and 8 mg/kg for 7 weeks), low copper (1.5 mg/kg), adequate zinc (15 mg/kg), and adequate copper (6 mg/kg). Two groups of rats were fed the adequate-zinc diet with low or adequate copper and pair-fed with corresponding rats fed the low-zinc diet. When compared to the pair-fed rats, marginal zinc deficiency significantly decreased the urinary excretion of magnesium and calcium, increased the concentrations of magnesium and calcium in the tibia, increased the concentration of magnesium in the kidney, and increased the urinary excretion of helical peptide (bone breakdown product). Marginal copper deficiency decreased extracellular superoxide dismutase and glutathione, which suggests increased oxidative stress. None of the variables responding to the marginal zinc deficiency were significantly altered by the marginal copper deficiency. The findings in the present experiment suggest that increased magnesium retention and impaired calcium utilization are indicators of marginal zinc deficiency. (1) The genetic response of the mouse spermatogonial stem cell to a high dose of X-rays given in two unequal fractions 24 h apart can be dependent upon the order in which the two fractions are given. When 1000 R was administered as 100 R followed by 900 R the recovered translocation yield (22%) was similar to that which can be obtained by extrapolation from lower doses and also to that of a 500 + 500 R 24 h fractionation. By contrast, when the 900 R preceded the 100 R the response was much lower (7.4%), yet still greater than that produced by a single 1000 R treatment (4.5%). The same order of effectiveness was observed for length of sterile period. (2) The sub-additive translocation yields previously obtained with 800 R treatments given in fractions of 500 R and 300 R at intervals of 3-12 days were found to be maintained with intervals up to at least 15 days but additivity was regained by the end of the third week. Sterile period data indicated that with these intervals the germinal epithelium had recovered sufficiently from the first fraction for spermatogenesis to restart before the second fraction was given. (3) It is concluded from the two experiments that (a) 24 h after a radiation exposure the surviving stem cells are more sensitive than formerly both to killing and genetic damage, (b) at this time they are no longer heterogeneous in their radiosensitivities, so that increasing yields of genetic damage may be obtained with increasing dose i.e. there is no fall in yield at higher doses, (c) the change in sensitivity could be a consequence of a synchronization to a sensitive stage in a cell cycle, or to a transitional phase preparatory to entering a different cell cycle. (d) to achieve rapid repopulation of the germinal epithelium the surviving stem cells are stimulated to enter a shorter cell cycle and this is the cause of the sub-additive translocation yields with fractionation intervals of 3-15 days, (e) the recommencement of spermatogenesis is associated with the reestablishment of the heterogeneity in radiosensitivity among the stem cells. Strains of the plague microbe, antigen complete and defective by fraction I and mouse toxin had the same in vitro susceptibility to ofloxacin (MIC 0.08 mg/L). The drug was superior in its activity to pefloxacin and especially nalidixic acid. In the experiments with albino mice (prophylaxis, 5 days) the ofloxacin efficacy was lower when the infection was due to the plague microbe strains deprived of the ability to produce fraction I and mouse toxin, evident from a statistically significant increase of the drug ED50 and a decrease of the animal survival percentage. When used in the doses corresponding to the human average daily doses, ofloxacin provided effective animal protection (80 to 100 per cent survival) after the prophylaxis for 7 days and the treatment of the plague infection irrespective of the strains, complete or antigen changed. However, when the infection is due to the antigen changed strain, ofloxacin should be used in the maximum daily doses at least for 7 days.  相似文献

The effect of chronic irradiation on T and B cell numbers and function was studied in mice. Cobalt 60 gamma radiation at 6 R/hour reduced the numbers of anti-SRBC PFC in the spleen, with minimal levels recorded after total exposures of 1000-2000 R. Recovery was incomplete after 1000 R, reaching only 40-50 per cent of normal in four months and remaining at that level for the animal's lifetime. The long-term deficiency in PFC formation was not due to a quantitative lack of T or B cells since normal cell numbers were observed in the spleen 60-144 days after 1000 R. Adoptive transfer studies with combinations of bone marrow and thymus cells, or of splenic T and B cells, from normal and irradiated mice, revealed functional defects in both cell compartments during the first two months. Normal and near normal function of T and B cells occurred 100 days postirradiation, a time when the splenic in vivo response was still only 50 per cent of the controls. The latter observation suggests that the microenvironment of the chronically irradiated spleen alters factors regulating T and B cell interactions in response to a T-dependent antigen.  相似文献

Reserpine (0.1 mg/kg/day) was administered to rats from 11 through 30 days of age. During and after administration of reserpine, concentrations of catecholamines, epinephrine and norepinephrine, in the brain were estimated. Levels of catecholamines were about 30 per cent of normal during the period of reserpine administration. Approximately 3 weeks were required for these levels to return to normal. When animals were 95-100 days of age, they were deprived of food and were trained to press a bar for food. When the rate of responding became stable, the animals were subjected to three successive extinctions at daily intervals and the increase in response rate after the onset of each extinction was determined. This extinction-induced increase in response rate was greater for previously reserpinized animals than controls during the second and third extinctions, but not the first. These findings are interpreted as a decreased ability of the animals, reserpinized during infancy, to learn to respond discriminatively during non-reinforcement (extinction). Thus, an effect of reserpine administration during infancy on a type of behaviour in the adult has been demonstrated. This occurs after the catecholamine-depleting effect of the reserpine has been fully dissipated.  相似文献

Adult male rats were given prednisolone (Urbason solubile = 6-methyl-prednisolone) in doses of 4 mg/100 g body weight for 10, 20 or 30 days and its effect on the calcium, phosphorus and magnesium content of their femurs was studied. At all the given intervals a statistically significant decrease in the magnesium content of the bone was found; the calcium content was significantly reduced only in the first phase of the experiment (at 10 days).  相似文献

A series of nine experiments were done to obtain further evidence that boron might be involved in major mineral metabolism (Ca, P, and Mg), thus indicating that boron is an essential nutrient for animals. Eight factorially arranged experiments of 6–10 wk durations were done with weanling Sprague-Dawley male rats. One factorially arranged experiment was done with weanling spontaneously hypertensive rats. The variables in each experiment were dietary boron supplements of 0 and 3 μg/g, and dietary magnesium supplements of either 200 (Experiments 1–3) or 100 (Experiments 4–9) and 400 μg/g. In Experiments 7 and 9, a third variable was dietary manganese supplements of 25 and 50 μg/g. Methionine status was varied throughout the series of experiments by supplementing the casein-based diet with methionine and arginine. Findings were obtained indicating that the severity of magnesium deprivation and the methionine status of the rat strongly influence the extent and nature of the interaction between magnesium and boron, and the response to boron deprivation. When magnesium deprivation was severe enough to cause typical signs of deficiency, a significant interaction between boron and magnesium was found. Generally, the interaction was characterized by the deprivation of one of the elements making the deficiency signs of the other more marked. The interaction was most evident when the diet was not supplemented with methionine and especially when the diet contained luxuriant arginine. Signs of boron deprivation were also more marked and consistent when the diet contained marginal methionine and luxuriant arginine. Among the signs of boron deprivation exhibited by rats fed marginal methionine were depressed growth and bone magnesium concentration, and elevated spleen wt/body wt and kidney wt/body wt ratios. Because the boron supplement of 3 μg/g did not make the dietary intake of this element unusual, it seems likely that the response of the rats to dietary boron in the present study were manifestations of physiological, not pharmacological, actions, and support the hypothesis that boron is an essential nutrient for the rat. The importance of cobalt on the anaerobic conversion of methanol under thermophilic conditions was studied in three parallel lab-scale UASB-reactors and in cobalt-limited enriched cultures. Reactors R1, R2, and R3 were fed with methanol in a bicarbonate-buffered medium, supplied with iron and macronutrients: in R1 all metals were supplied (control), R2 was cobalt deprived, and in R3 all metals were deprived. In the 136 days of continuous experiment, a drop in performance was observed over the last 30 days. Particularly in R3, both methanol removal and methane formation dropped by 7.1% and 13.7%, respectively, compared to the control reactor, R1. When the medium was cobalt-deprived, acetate was not produced and, as a consequence, the enriched consortium lost its capacity to degrade acetate, indicating that the acetotrophic microorganisms were washed out. The addition of 0.5 microM of cobalt to a cobalt-deprived enrichment culture led to acetate accumulation. The results obtained in this study indicate that the mixed consortium requires a proper amount of cobalt, and its addition to a concentration of 0.1 microM leads to the highest methanol conversion rate, with methane as the sole end product from methanol.  相似文献

Twenty one-day-old Phaseolus vulgaris 'Saxa'plants were cultured in a growth chamber and the plants supplied with either a complete or a Mg-free nutrient solution. From 6 days after transfer to the Mg-free solution, the rate of increase of the area of the second trifoliate leaf was considerably reduced; by day 11 the sucrose concentration in the first trifoliate leaf had increased 6. 2-fold at the end of the dark period and 4. 6-fold after the light period as compared with the control plants. Corresponding starch concentrations increased 6. 6-fold and 2. 9-fold respectively. After days 5 to 6 the assimilation rates declined in the first trifoliate leaf of the plants showing deficiency, in comparison with the plants fully supplied with nutrients; respiration increased during darkness. The reduction in net assimilation rate was to a great extent reversible after resupply of magnesium.
The reduction of magnesium concentration in the deficient plants was much more marked in the expanding leaves than in the mature primary leaves and roots. Sucrose and starch accumulation did not occur when the first trifoliate leaf was partially shaded, although magnesium concentration, as in the unshaded leaves, was reduced to 13% of that of the control plants. The consequences of magnesium deficiency in the expanding first trifoliate leaf are discussed in terms of the possibility of sink limitation.  相似文献

Dibrachys boarmiae was recently found as a parasitoid of the pistachio twig borer moth, Kermania pistaciella, an important pistachio pest in Iran. Several biological and behavioural aspects of the wasp were studied under controlled conditions using three different lepidopterous host species, K. pistaciella, Sitotroga cerealella and Galleria mellonella. Development time from egg to adult decreases from 54 days at 15°C to 12.6 days at 32.5°C. The thermal constant was estimated 270°D, and the lower development threshold was at 9.9°C. Longevity of females that had the opportunity to oviposit and access to food (honey) was on average 23 days, and 134 offspring were produced during 13 days of oviposition. Ninety percent of ovipositions occur during the first 10 days of the parasitoid's life, although no pre-oviposition period was found. If females were deprived of food, longevity as well as fecundity, were drastically reduced to 8 days and only 79 offspring respectively. Females were able to mate immediately after emergence and the sex ratio of the progeny was strongly female biased on all examined hosts, especially on the largest host G. mellonella (0.08 male). When females were kept with a male throughout their life, the total number of progeny decreased to 92 wasps on average but the sex ratio was unaffected. The parasitoid significantly preferred to attack the pre-pupal stage, and this influenced the size of clutches allocated to different host stages. The parasitoid laid larger clutches on bigger hosts.  相似文献

Innate immune and inflammatory responses mediated by Toll like receptors (TLRs) have been implicated in myocardial ischemia/reperfusion (I/R) injury. This study examined the role of TLR3 in myocardial injury induced by two models, namely, myocardial infarction (MI) and I/R. First, we examined the role of TLR3 in MI. TLR3 deficient (TLR3−/−) and wild type (WT) mice were subjected to MI induced by permanent ligation of the left anterior descending (LAD) coronary artery for 21 days. Cardiac function was measured by echocardiography. Next, we examined whether TLR3 contributes to myocardial I/R injury. TLR3−/− and WT mice were subjected to myocardial ischemia (45 min) followed by reperfusion for up to 3 days. Cardiac function and myocardial infarct size were examined. We also examined the effect of TLR3 deficiency on I/R-induced myocardial apoptosis and inflammatory cytokine production. TLR3−/− mice showed significant attenuation of cardiac dysfunction after MI or I/R. Myocardial infarct size and myocardial apoptosis induced by I/R injury were significantly attenuated in TLR3−/− mice. TLR3 deficiency increases B-cell lymphoma 2 (BCL2) levels and attenuates I/R-increased Fas, Fas ligand or CD95L (FasL), Fas-Associated protein with Death Domain (FADD), Bax and Bak levels in the myocardium. TLR3 deficiency also attenuates I/R-induced myocardial nuclear factor KappaB (NF-κB) binding activity, Tumor necrosis factor alpha (TNF-α) and Interleukin-1 beta (IL-1β) production as well as I/R-induced infiltration of neutrophils and macrophages into the myocardium. TLR3 plays an important role in myocardial injury induced by MI or I/R. The mechanisms involve activation of apoptotic signaling and NF-κB binding activity. Modulation of TLR3 may be an effective approach for ameliorating heart injury in heart attack patients.  相似文献
