Substance P (SP) effects on the central mechanisms of escape reaction, elicited by threshold electrical stimulation of the ventromedial hypothalamus were investigated in rabbits pretreated with ethanol (0.5 g/kg). SP (30 micrograms/kg) was demonstrated to normalize in 71.4% of cases the excitability of the ventromedial hypothalamus which was decreased by ethanol and restored in 83.3% of cases the facilitatory effects of the midbrain reticular formation in escape reactions. However, SP was ineffective in the restoration of the inhibitory effects of the dorsal hippocamp on the excitability of the ventromedial hypothalamus that was obvious in intact animals. Partial normalizing effect of SP on escape reaction in rabbits after previous ethanol administration can be accounted for by the fact that both undecapeptide and ethanol are similar in their realization of central effects such as an interaction with the same brain neurotransmitters, interference with neuronal enzyme processes and reactions with opiate receptors.  相似文献

Chronic experiments were made on intact rabbits and rabbits with destroyed paraventricular and ventromedial nuclei of the hypothalamus to explore the hydrocortisone-induced inhibition of the stressor response of the pituitary-adrenocortical system. Intravenous injection of hydrocortisone in a dose of 100 micrograms/kg 5 minutes before immobilization stress led to inhibition of corticosteroid elevation induced by immobilization of the animals. Inhibition of the stressor reaction was maximal in intact animals, less in rabbits with destroyed ventromedial nuclei, and further less in animals with destroyed paraventricular nuclei. The paraventricular and ventromedial nuclei of the hypothalamus are necessary for inhibition of the pituitary-adrenocortical system by the feedback mechanisms.  相似文献

The authors studied bioelectric potentials in the area of the hypothalamic ventromedial nucleus and the lateral hypothalamus of rats fed on a standard, a high protein and a high fat diet. On the first 3--6 days after changing from the standard to the high fat and high protein diets, a decrease in the amplitude of electrical activity was recorded in both the areas in question. It was also found that the frequency of electrical activity in the hypothalamic ventromedial nucleus or the lateral hypothalamus rose, after 3 days administration of the high fat or the high protein diet, in correlation to the type of diet, and that, in the frequency spectrum, a change occurred in the proportion of basic frequency in relation to superimposed frequencies distorting it. It was further found that there was a permanent difference between the amplitude of electrical activity in the lateral hypothalamus and the hypothalamic ventromedial nucleus.  相似文献

Many gastrointestinal meal-related signals are transmitted to the central nervous system via the vagus nerve and thereby control changes in meal size. The c-Fos-positive neuron has been used as a marker of neuronal activation after lipid meals to examine the contribution of a selective macronutrient on brain neurocircuit activity. In rats fed Intralipid, the c-Fos-positive neurons were highly stimulated in the nucleus of the solitary tract (NTS) and in the hypothalamus, including the paraventricular nucleus (PVN), arcuate nucleus of the hypothalamus (ARC), and ventromedial hypothalamus at 4 h lipid feeding. However, c-Fos-like immunoreactivity was markedly attenuated in these brain regions when chylomicron formation/secretion was blocked by Pluronic L-81. After lymph was diverted from the lymph cannulated animals, the rats had a lower number of c-Fos-positive cells in the NTS and ARC. In contrast, the rats had higher c-Fos-positive neurons in PVN. The present study also revealed that c-Fos-positive neurons induced by feeding of Intalipid were abolished by CCK type 1 receptor antagonist, Lorglumide. We conclude that the formation and/or secretion of chylomicron are critical steps for initiating neuronal activation in the brain.  相似文献

The effects of estradiol (E2) on the expression of proteins in the pars lateralis of the ventromedial nucleus of the hypothalamus (VMNpl) in ovariectomized rats was studied using 2-dimensional gel electrophoresis followed by RPLC-nanoESI-MS/MS. E2 treatment resulted in the up-regulation of 29 identified proteins. Many of these proteins are implicated in the promotion of neuronal plasticity and signaling.  相似文献

Bilateral lesions of the ventromedial nucleus of the hypothalamus interfered with the estrogenic induction of sexual receptivity in the female rat, but seemingly did not affect the ability of female rats to show lordosis following combined stimulation with estrogen and progesterone. In addition, ventromedial hypothalamic lesions did not affect the ability of females to show male-like sexual activity in response to exogenous androgenic stimulation.  相似文献

The distribution of GABA-immunoreactive (IR) cells was studied by immunohistochemistry in conjunction with highly specific antiserum GABA in the cat hypothalamus. Colchicine pretreatment made it possible to visualize a large number of labeled cells in the medial preoptic and dorsal hypothalamic areas. In contrast, the ventromedial and anterior hypothalamic nuclei contained only a few labeled cells, and the paraventricular and supraoptic nuclei were devoid of them. A very dense network of GABA-IR presumptive terminals was seen in the ventrolateral posterior hypothalamus where labeled cells could be recognized. The possibility of an involvement of the GABAergic neuronal system in the regulation of sleep-waking cycle is discussed.  相似文献

In experiments on rabbits, the action was studied of vasoactive peptides angiotensin-II, bradykinin and lysyl-vasopressin, injected into the brain ventricles, on a passive defensive reaction to electrical stimulation of the ventromedial hypothalamus. It was found that angiotensin-II and lysyl-vasopressin had a pronounced inhibitory influence, while bradykinin had a facilitatory effect on this reaction. Electrical stimulation of the ventromedial hypothalamus predominantly facilitated reactions of individual neurones of the sensorimotor cortex to angiotensin-II and bradykinin. Intraperitoneal injection of P-substance to rats in conditions of an acute conflict situation producing an emotional stress, brought about a significant diminution of sudden death incidence from cardiovascular insufficiency.  相似文献

The Zigmond method was made use of to study tyrosine hydroxylase activity in cervical, stellate and nodosal ganglia of rabbits predisposed and resistant to stress. Experimental acute emotional stress was induced by concurrent stimulation of the ventromedial nuclei of the hypothalamus and different skin areas. Experiments have shown that changes in metabolic activity of the sympathetic and parasympathetic systems were phasic in nature throughout 3 hours of the experiment. The changes in tyrosine hydroxylase activity in nodosal and stellate ganglia of rabbits predisposed to stress differs from those in stress-resistant animals. Stress-predisposed rabbits manifested a decrease in the activity of tyrosine hydroxylase in stellate and nodosal ganglia.  相似文献

Leptin, secreted by white adipocytes, has profound feeding, metabolic, and neuroendocrine effects. Leptin acts on the brain, but specific anatomical sites and pathways responsible for mediating these effects are still unclear. We have systematically examined the distribution of leptin receptor containing neurons in the porcine hypothalamus by means of immunohistochemical staining methods. Leptin receptor immunoreactivity (OBR-IR) was observed in both the preoptic area and anterior hypothalamic area. No immunoreactive structures were found in the median eminence. Only single, small neurons were observed in the arcuate nucleus. The most abundant OBR-IR cell bodies were located in the supraoptic nucleus. In the paraventricular nucleus, OBR-IR neurons were moderate in number. Single, dispersed neurons were found in the ventromedial nucleus. These findings indicate that there are distinct OBR-IR neuronal populations in the porcine hypothalamus and leptin not only plays an integrative role in feeding behavior, but also in neuroendocrine activity.  相似文献

Employing immunohistochemistry techniques, we examined the c-fos expression in different hypothalamic areas, when plasma glucose levels were modified by the administration of insulin and 2-deoxyglucose (2-DG) respectively. Subsequently, the hypoglycemia produced by an injection of insulin significantly increased feeding concomitant to higher c-fos expression in the arcuate nucleus (ARC), paraventricular nucleus (PVN), dorsomedial hypothalamus (DMH) and lateral hypothalamus (LH), while no statistical changes in the ventromedial hypothalamus (VMH) were found. Also, the glucopenia induced by 2-DG administration produced similar stimulatory effects on appetite and the neuronal activity affecting all the hypothalamic areas studied, including the VMH. The peripheral blockade of the orexigenic hormone ghrelin with a specific antibody (AGA) significantly decreased food intake as induced from acute hypoglycemia and glucopenia. Curiously, the conjoint AGA and insulin or 2-DG administration produced a differential effect on the hypothalamic neurons analyzed, by increasing the number of c-fos positive neurons in the ARC, PVN and DMH, but not in the VMH and LH. This outcome suggests an interactive effect of the glucostatic pathways involving these two areas with the ghrelin signaling.  相似文献

Irregularity of the rhythm of breathing, the breathing rate, blood pressure and heart rate were studied in rats under acute emotional stress induced by non-periodic stimulation of the skin and ventromedial hypothalamus. The irregularity of the rhythm of breathing was substantially increased during stimulation of the hypothalamus up to short-term respiratory arrests in animals predisposed to emotional stress. Disturbances of the rhythm of breathing may be one of objective prognostic criteria of the animals' survival or lethality under emotional stress.  相似文献

Projections into rat ventromedial hypothalamus were studied with retrograde transport of horseradish peroxidase (HRP). Following injection of HRP into ventromedial hypothalamus, labeled neurons were found in cortical and medial amygdaloid nuclei, ipsilateral mediodorsalis thalamus (MD), dorsal raphe nucleus, and contralateral sensorimotor cortex. The anorexigenic gut hormones oxyntomodulin (OXM) and glucagon-like peptide-1 (GLP-1) are thought to physiologically regulate appetite and food intake. Using manganese-enhanced magnetic resonance imaging, we have shown distinct patterns of neuronal activation in the hypothalamus in response to intraperitoneal injections into fasted mice of 900 and 5400 nmol/kg OXM or 900 nmol/kg GLP-1. Administration of OXM at either dose resulted in a reduced rate of signal enhancement, reflecting a reduction in neuronal activity, in the arcuate, paraventricular, and supraoptic nuclei of the hypothalamus. Conversely, GLP-1 caused a reduction in signal enhancement in the paraventricular nucleus only and an increase in the ventromedial hypothalamic nucleus. Our data show that these two apparently similar peptides generate distinct patterns of activation within the hypothalamus, suggesting that GLP-1 and OXM may act via different hypothalamic pathways.  相似文献

[14C] deoxyglucose-autoradiography of mouse brain after food deprivation showed no specific changes in the ratio of autoradiographic optical density of the lateral nucleus of the hypothalamus (LH) versus ventromedial nucleus of the hypothalamus (VMH). The results show that even if there were differences in functional activity between these hypothalamic nuclei during conditions of hunger and satiety, they are not reflected as autoradiographically detectable in relative deoxyglucose uptake.  相似文献

Changes of unit activity in different nuclei of the posterior hypothalamus induced by intravenous oxytocin injections were studied in acute experiments on Wistar rats anaesthetized with nembutal. Influence of nonapeptide was dose-dependent. Latencies of various neuronal reactions were different. Brain‐derived neurotrophic factor (BDNF), corticotropin‐releasing factor (CRF), and hypothalamic neuronal histamine are anorexigenic substances within the hypothalamus. This study examined the interactions among BDNF, CRF, and histamine during the regulation of feeding behavior in rodents. Food intake was measured after treatment with BDNF, α‐fluoromethyl histidine (FMH; a specific suicide inhibitor of histidine decarboxylase that depletes hypothalamic neuronal histamine), or CRF antagonist. We measured food intake in wild‐type mice and mice with targeted disruption of the histamine H1 receptor (H1KO mice) after central BDNF infusion. Furthermore, we investigated CRF content and histamine turnover in the hypothalamus after BDNF treatment, and conversely, BDNF content in the hypothalamus after histamine treatment. We used immunohistochemical staining for histamine H1 receptors (H1‐R) in BDNF neurons. BDNF‐induced feeding suppression was partially attenuated in rats pre‐treated with FMH or a CRF antagonist, and in H1KO mice. BDNF treatment increased CRF content and histamine turnover in the hypothalamus. Histamine increased BDNF content in the hypothalamus. Immunohistochemical analysis revealed that H1‐Rs were expressed on BDNF neurons in the ventromedial nucleus of the hypothalamus. These results indicate that CRF and hypothalamic neuronal histamine mediate the suppressive effects of BDNF on feeding behavior and body weight.  相似文献

Experiments were conducted on male rats; a study was made of the activity of pituitary-adrenal cortex system under conditions of electrolyte destruction of various hypothalamic areas. Pharmacological analysis pointed to the irregular distribution in the hypothalamus of the cholinergic and adrenergic systems controlling the secretion of ACTH-glucocorticoids. As suggested, the cholinergic systems are situated in the mammillary region, alpha-adrenergic ones--in the anterior or the posterior regions of the hypothalamus, and beta-adrenergic receptors--in the ventromedial nuclei of the hypothalamus. The latter possibly play an inhibitory role.  相似文献
