The complete nucleotide sequence of Chinese rape mosaic virus has been determined. The virus is a member of the tobamovirus genus of plant virus and is able to infect Arabidopsis thaliana (L.) Heynh systemically. The analysis of the sequence shows a gene array that seems to be characteristic of crucifer tobamoviruses and which is slightly different from the one most frequently found in tobamoviruses. Based on gene organization and on comparisons of sequence homologies between members of the tobamoviruses, a clustering of crucifer tobamoviruses is proposed that groups the presently known crucifer tobamovirus into two viruses with two strains each. A New South Wales isolate (Ca) of capsicum mosaic virus was tested against antisera to it and capsicum tobamovirus isolates from the Netherlands (P8, P11), USA (SL), Argentina (FO) and Sicily (PM). The comparison demonstrated that the four viruses Ca, P8, PM and SL are closely related to each other, forming a series of decreasing relationship to Ca in the above order. FO was related to these but insufficiently to be considered part of the group, and P11 was only slightly related to the others. The literature on serology of tobamoviruses in Capsicum spp. was collated and it is suggested that isolates from Sicily (pepper mild mottle), Australia (capsicum mosaic), The Netherlands (P8, P14) and USA (SL) be considered as strains of a virus distinct from both tobacco mosaic and tomato mosaic viruses and that these isolates all be referred to in future as strains of pepper mild mottle virus.  相似文献

The degree of serological variability among pepper strains of potato virus Y (PVY) was assessed through the analysis of samples of infected pepper collected in three main pepper producing regions of Spain. Samples corresponding to the period 1980–1991 were analysed by ELISA with five different monoclonal antibodies (MAbs) produced against potato strains of the virus. The results obtained show a limited degree of epitope variability among pepper PVY-isolates, since only eight out of 32 possible serological profiles were found. Most isolates are not recognised by a MAb directed towards an epitope reported to be present in all potato-PVY isolates. The overall serological behaviour of pepper isolates with these MAbs places them as closer to the group O, of the three groups into which the potato isolates of PVY have been subdivided.  相似文献

Transgenic pepper plants coexpressing coat proteins (CPs) of cucumber mosaic virus (CMV-Kor) and tomato mosaic virus (ToMV) were produced by Agrobacterium-mediated transformation. To facilitate selection for positive transformants in transgenic peppers carrying an L gene, we developed a simple and effective screening procedure using hypersensitive response upon ToMV challenge inoculation. In this procedure, positive transformants could be clearly differentiated from the nontransformed plants. Transgenic pepper plants expressing the CP genes of both viruses were tested for resistance against CMV-Kor and pepper mild mottle virus (PMMV). In most transgenic plants, viral propagation was substantially retarded when compared to the nontransgenic plants. These experiments demonstrate that our transgenic pepper plants might be a useful marker system for the transgene screening and useful for classical breeding programs of developing virus resistant hot pepper plants.  相似文献

A Tobamovirus Causing Heavy Losses in Protected Pepper Crops in Spain   总被引:4,自引:0,他引:4  
During a four-year (1982–1985) survey of plant viruses infecting pepper cultivars grown under plastic in the Southeastern region of Spain, a tobamovirus was found to be the major disease agent of this crop. The virus produces slight or no symptoms on the leaves, but causes chlorotic mottling, malformation and reduction in size with occasional necrosis on the fruits and was able to infect all commercial pepper cultivars tested, including those resistant to other tobamoviruses, causing a catastrophic disease. The biological and serological characterization of the virus showed that it is very similar to pepper mild mottle virus (PMMV) (Wetter et al. 1984) and therefore we have termed it as Spanish strain of PMMV (PMMV-S). The need of grouping all the so-called “pepper strains” of tobacco mosaic virus (TMV) as a new distinct member of the tobamovirus group with the name of PMMV is emphasized.  相似文献

The partial nucleotide sequence of the 3-terminal region of the Korean isolate of odontoglossum ringspot tobamovirus (ORSV-Cy) from cool-growing Cymbidium was determined. The sequence contained a full length open reading frame (ORF) coding for the viral cell-to-cell movement protein (MP). The ORF was located upstream of the coat protein gene and 105 nucleotides longer than that of tobacco mosaic virus (TMV). The ORF predicts a polypeptide chain of 303 amino acids with a molecular weight of 33573. The ORF contained a similar region of conserved sequence motif of tobamoviruses and putative assembly origin of the viral RNA was located at about 1,100 nucleotides away from the 3 end. The predicted amino acid sequence for the MP gene of ORSV-Cy is more closely related to pepper mild mottle virus (PMMV), TMV-vulgare and TMV-Rakkyo than to tobacco mild green mosaic virus (TMGMV), TMV-L, cowpea strain of TMV (SHMV), and cucumber green mottle mosaic virus (CGMMV).  相似文献

Hot pepper and tomato fields in the main growing areas in the Rift Valley and the west of Ethiopia were surveyed for virus infections in 1994. A total of 286 samples from hot pepper and 222 samples from tomato plants and associated Datura stramonium L. and Nicandra physalodes Gaertn. weeds with symptoms suggestive of virus infections were collected and analysed using electron microscopy, serology and test plant reactions. Potato virus Y (PVY), Ethiopian pepper mottle virus (EPMV), pepper veinal mottle virus (PVMV) and tomato mosaic virus (ToMV) were detected in hot pepper samples while tomato samples were shown to be infected with tomato mild mottle virus (TMMV), PVY and ToMV. The most widespread and predominant viruses which also occurred frequently in mixed infections were PVY and EPMV in hot pepper and PVY and TMMV in tomato. TMMV was also found in many samples of D. stramonium and N. physalodes. ToMV was identified in only few samples from both crops in the Rift Valley by its characteristic particle morphology, serological properties and symptomatology. PVMV was found in hot pepper samples only from western Ethiopia, but no natural infection of tomato with this virus was revealed. This is the first report on the natural occurrence of TMMV in tomato, D. stramonium and N. physalodes, as well as of ToMV in hot pepper and tomato in Ethiopia.  相似文献

Bell Pepper Mottle Virus, a Distinct Tobamovirus Infecting Pepper   总被引:1,自引:0,他引:1  
Bell pepper mottle virus (BPeMV) can be distinguished by symptomatology and host range from other tobamoviruses but a reliable identification needs serological tests. The relationships of BPeMV to tobacco mosaic virus (TMV), Odontoglossum ringspot virus (ORSV), tobacco mild green mosaic virus (TMGMV), and pepper mild mottle virus (PMMV) were investigated using precipitin drop tests on slides, immunodiffusion gel tests, double antibody sandwich enzyme-linked immunosorbent assay (ELISA), and indirectELISA using enzyme-linked goat anti-rabbit globulins for the determination of antiserum titers and serological differentiation indices (SDI). Comparisons of SDIs and amino acid composition data demonstrated that BPeMV is a new species of the tobamovirusgroup. BPeMV, ORSV, PMMV, and TMGMV form a cluster within the genus (group) and could be considered as a subgenus of tobamoviruses.  相似文献

Pythium catenulatum, P. diclinum and P. paroecandrum, as well as asexual forms with filamentous sporangia were isolated from irrigation water in the Poniente region of Almería (south-eastern Spain). The taxonomic and morphological details of these fungal isolates are described. P. catenulatum and P. diclinumare new reports to Spain. The first Citrus tristeza virus (CTV) genomes completely sequenced (19.3-kb positive-sense RNA), from four biologically distinct isolates, are unexpectedly divergent in nucleotide sequence (up to 60% divergence). Understanding of whether these large sequence differences resulted from recent evolution is important for the design of disease management strategies, particularly the use of genetically engineered mild (essentially symptomless)-strain cross protection and RNA-mediated transgenic resistance. The complete sequence of a mild isolate (T30) which has been endemic in Florida for about a century was found to be nearly identical to the genomic sequence of a mild isolate (T385) from Spain. Moreover, samples of sequences of other isolates from distinct geographic locations, maintained in different citrus hosts and also separated in time (B252 from Taiwan, B272 from Colombia, and B354 from California), were nearly identical to the T30 sequence. The sequence differences between these isolates were within or near the range of variability of the T30 population. A possible explanation for these results is that the parents of isolates T30, T385, B252, B272, and B354 have a common origin, probably Asia, and have changed little since they were dispersed throughout the world by the movement of citrus. Considering that the nucleotide divergence among the other known CTV genomes is much greater than those expected for strains of the same virus, the remarkable similarity of these five isolates indicates a high degree of evolutionary stasis in some CTV populations.  相似文献

A virus causing ‘eyespot’ leaf symptoms in groundnut plants was transmitted by sap-inoculation and by Aphis craccivora in the non-persistent manner. It infected 16 of 72 species from five of 12 families and was easily propagated in Arachis hypogaea and Physalis floridana. The virus has particles c. 13 × 755 nm and is serologically closely related to soybean mosaic and pepper veinal mottle viruses, and more distantly to four other potyviruses. The virus differs in host range, in vitro properties and serological properties from previously described strains of soybean mosaic and pepper veinal mottle viruses. It seems to be a distinct member of the potyvirus group and we propose the name groundnut eyespot virus.  相似文献

Abstract A Spanish strain of pepper mild mottle virus (PMMV-S) (A lonso et al. 1989) can be differentiated from the Italian PMMV (W etter et al. 1984) by the responses of Capsicum spp. with resistance genes to tobamoviruses, by radioimmunoassay and by the electrophoretic mobility of their viral particles. Moreover, the analysis of the electrophoretic mobility of the viral particles in agarose gels and that of the viral coat proteins in polyacrylamide-urea gels are reliable and rapid techniques for distinguishing PMMV isolates from other members of the tobamovirus group and thus can be used for diagnostic purposes. These results support the proposal of grouping these pepper viruses, including the "pepper strains" of TMV, as a new tobamovirus subgroup (PARES 1985, BETTI et al. 1987, WETTER et al. 1987, ALONSO et al. 1989).  相似文献

Two viruses were isolated from commercial groundnut plants showing mild mottle (MM) and severe mottle (SM) symptoms in the Rayalaseema area of Andhra Pradesh State, India. The host range of both the isolates was restricted to Fabaceae. The physical properties of both isolates were: DEP 10-6 to 10-7. TIP 70–85 C and LIV 5–8 days. Partially purified virus preparations contained slightly flexuous filamentous particles. In ELISA and gel diffusion tests both isolates reacted similarly with anusera to the carlaviruses cowpea mild mottle, cassia mild mosaic and potato M. Particle morphology. sedimentation coefficient, and estimated sizes of the coat proteins and RNAs gave additional evidence that the viruses were carlaviruses. Bemisia labaci transmitted only the MM isolate and the two isolates also differed in electrophoretic mobility of intact particles and amino acid composition of the coat proteins. The isolates are identified as distinct strains of cowpea mild mottle virus.  相似文献

The tissue blot immunobinding assay (TBIA) is widely used for the detection and localization of plant viruses in various plant tissues. The basic experimental procedures of TBIA sampling and blotting were simplified using commercially available micropipette tips. This method was termed the ring-blot immunobinding assay (R-BIA), as the blot on the membrane forms a ring shape. The detection efficacy of R-BIA was tested for two chili pepper viruses, pepper mild mottle tobamovirus (PMMoV) and pepper mottle potyvirus (PepMoV), following the optimized serological procedures of TBIA (length of the incubation period and BSA concentration, and primary and secondary antibodies). Sensitivity of the R-BIA was about 1 ng/ml of purified PMMoV in pepper leaf sap from a healthy pepper plant. R-BIA also showed high specificity in the detection of PMMoV and PepMoV. Moreover, the modified sampling and blotting procedures were simpler and more reliable than other TBIA methods (such as whole-leaf blotting and crushed-leaf blotting), suggesting that the R-BIA may be used for medium- to large-scale detection of plant viruses in laboratories with minimal facilities.  相似文献

Eleven isolates of phleum mottle virus, each from a different grass species, were grouped into five distinct strains: phleum mottle (PM), holcus transitory mottle (HTM), festuca mottle (FM), cocksfoot mild mosaic (CMM) and brome stem-leaf mottle (BSM). Their interrelationship was expressed three-dimensionally using serological differentiation indices. When 105 species of Gramineae were inoculated with each strain, eighty-one in forty genera became infected. These included festucoid, chloridoid, panicoid and andropogonoid species. Electron microscopy was more reliable than back-inoculation for detecting infection in plants that remained symptomless. Annual species were usually more severely affected than perennials. PM, HTM, FM, CMM and BSM did not appear to differ in host range, but sometimes differed in the symptoms induced in a given host. Each was symptomless in some hosts, caused a mild or severe mottle in others and mottling plus necrosis in a few. HTM was symptomless in most, and BSM in fewest species: BSM caused necrosis in most and HTM in fewest hosts.  相似文献

Infectious hematopoietic necrosis virus (IHNV) contains 3 major genogroups in North America with discreet geographic ranges designated as upper (U), middle (M), and lower (L). A comprehensive genotyping of 237 IHNV isolates from hatchery and wild salmonids in California revealed 25 different sequence types (a to y) all in the L genogroup; specifically, the genogroup contained 14 sequence types that were unique to individual isolates as well as 11 sequence types representing 2 or more identical isolates. The most evident trend was the phylogenetic and geographical division of the L genogroup into 2 distinct subgroups designated as LI and LII. Isolates within Subgroup LI were primarily found within waterways linked to southern Oregon and northern California coastal rivers. Isolates in Subgroup LII were concentrated within inland valley watersheds that included the Sacramento River, San Joaquin River, and their tributaries. The temporal and spatial patterns of virus occurrence suggested that infections among adult Chinook salmon in the hatchery or that spawn in the river are a major source of virus potentially infecting other migrating or resident salmonids in California. Serum neutralization results of the California isolates of IHNV corroborated a temporal trend of sequence divergence; specifically, 2 progressive shifts in which more recent virus isolates represent new serotypes. A comparison of the estimates of divergence rates for Subgroup LI (1 x 10(-5) mutations per nucleotide site per year) indicated stasis similar to that observed in the U genogroup, while the Subgroup LII rate (1 x 10(-3) mutations per nucleotide site per year) suggested a more active evolution similar to that of the M genogroup.  相似文献
