This work deals with the extraction and back-extraction of a recombinant cutinase using AOT reversed micelles in isooctane. The effect of pH, ionic strength, AOT concentration and temperature on the extraction and back-extraction of the cutinase was investigated. High extraction (97%) of the cutinase was achieved at pH 7.0 with a 50 mM Tris-HCl buffer solution containing 100 mM KCl, but a low activity was detected in the reversed micellar phase. At pH 9.0, cutinase was extracted (75%) to the reversed micelles with higher activity. Cutinase was recovered (50%) from a reversed micellar phase (100 mM AOT/isooctane) into a 50 mM Tris-HCl buffered solution at pH 9.0 with 100 mM KCl, and 20°C. Summary We present a method to test if the proportion of 45,X cases resulting from loss of the maternal chromosome or of cases of 46,X,i(Xq) with the isochromosome of maternal origin is different from 1/2. In an infinite population the frequency distribution of individuals carrying a given number of mutations obeys a set of recursion equations, the equilibrium solution of which describes the mutation-selection balance. Although this solution is well-known in the case of a multiplicative-fitness landscape, where it is assumed that all mutations are deleterious and that each new mutation reduces the fitness of the individual by the same fraction, we are not aware of the existence of an analytical solution for the full dynamics. Using the generating function approach, we present here an explicit analytical solution for the frequency distribution recursion equations valid for all generations and initial conditions.This research was supported by Fundação de Amparo à Pesquisa do Estado de São Paulo (FAPESP), Proj. In order to observe collagen and elastic fibers simultaneously, sections of human aorta, skin, lung, liver, and bladder were stained by Sirius red and analyzed by fluorescence microscopy. In all cases, the fibers of collagen presented the characteristic fluorescent red-orange color that results from the interaction of this extracellular protein with the dye, whereas elastic fibers showed strong green fluorescence (intrinsic fluorescence). Summary A qualitative and quantitative biochemical study of the glycosaminoglycans was performed in tissues constituted predominantly by one type of collagen, or in tissues containing mixtures of different types of collagen. Summary The general ultrastructure of the electrocyte, the basic unit of the electric organs of Electrophorus electricus, is analyzed. Presented herein are detailed observations of the syncytial surface, its fibrillar coat, invaginations of the plasma membrane and synaptic terminals. Using Thiéry's method glycogen granules were identified in the syncytial cytoplasm and inside the synaptic terminals, their size and structure being compatible with the muscular origin of the electric organs, to which the filamentous meshwork found in the cytoplasm may be related. Among the perinuclear-organelles, are dense bodies with crystalline patterns. The reproductive biology of Zeyheria montana was studied through field observations of flower visitors and floral events, controlled manual pollinations, and observations of pollen tube growth and ovule penetration by fluorescence microscopy. Analysis of secretory areas of the nectar chamber was made by flower dissections and histology of serial sections. The flower lasted 6–8 days, but pollen exposure and stigma receptivity occurred only up to the end of the first and second days, respectively. Pollination was effected by several species of hummingbirds, especially Colibri serrirostris. The flowers present a rudimentary, non-functional disc, and secretion of nectar is performed by corolla-borne glandular trichomes. Only hand cross-pollinated and natural-pollinated flowers set fruits. Artificially self-pollinated and non-pollinated flowers dried off after anthesis without presenting any swelling of the ovary. Almost all the ovules in selfed and crossed pistils were penetrated 96 h after pollination. The formation of a skeletal muscle fiber begins with the withdrawal of committed mononucleated precursors from the cell cycle. These myoblasts elongate while aligning with each other, guided by recognition between their membranes. This step is followed by cell fusion and the formation of long striated multinucleated myotubes. We used methyl--cyclodextrin (MCD) in primary cultured chick skeletal muscle cells to deplete membrane cholesterol and investigate its role during myogenesis. MCD promoted a significant increase in the expression of troponin T, enhanced myoblast fusion, and induced the formation of large multinucleated myotubes with nuclei being clustered centrally and not aligned at the cell periphery. MCD myotubes were striated, as indicated by sarcomeric -actinin staining, and microtubule and desmin filament distribution was not altered. Pre-fusion MCD-treated myoblasts formed large aggregates, with cadherin and -catenin being accumulated in cell adhesion contacts. We also found that the membrane microdomain marker GM1 was not present as clusters in the membrane of MCD-treated myoblasts. Summary The possibility of the histochemical demonstration of the dopa-oxidase reaction in Mycobacterium leprae from the granulomas of untreated cases of human lepromatous leprosy was studied. Fresh frozen sections were taken, fixed for one hour, and incubated with DOPA (3,4-dihydroxyphenylalanine) for two hours at pH 6.8 and at 37° C. The results obtained did not confirm the biochemical phenomena observed in human liver and spleen.Investigation supported by Grant No. DF. Natural killer (NK) cells and cytotoxic T lymphocytes (CTLs) kill target cells by the granule-exocytosis pathway and by the engagement of molecules belonging to the tumor necrosis factor family. The involvement of secretory phospholipase A2 (sPLA2) in the cytotoxic process has been proposed in NK cells. However, its molecular identity and intracellular localization remain unknown, and its mechanism of action is poorly understood. Here, we have readdressed this issue by studying the cytotoxic activity of whole cell extracts of a CTL line. We observed that inactivation of the perforin-granzyme pathway at 37°C in the presence of 1 mM Ca2+ enhanced the ability of CTL extracts to induce apoptosis. This potentiation of cell death was Ca2+-dependent, thermo-resistant, and inhibited by 4-bromophenacyl bromide and scalaradial (two inhibitors of sPLA2). The involvement of an sPLA2 was confirmed by blocking the pro-apoptotic activity of the Ca2+-treated cell extract with an anti-sPLA2 polyclonal antibody. By cell fractionation assays, we showed that the pro-apoptotic sPLA2 was localized in the cytoplasmic fraction but not in perforin-rich granules or plasma membrane fractions. Western blotting analysis revealed the presence of four distinct bands of 56, 29.5, 21, and 15 kDa. The highest molecular weight band was consistent with the expression of a group III sPLA2. Taken together, these data indicate that an apoptosis-inducing sPLA2 is expressed in the cytosol of a CTL cell line and suggest that it plays an effector role in CTL-mediated cytotoxicity. Selective deposition of BaSO4 in the tight junctions (TJs) of frog skins led to profound and reversible functional alterations of these structures, as revealed by changes of tissue conductance (G), clamping current (I), and fluxes of extracellular markers (sulfate (JSO 4) and sucrose (JSUC)). Experiments were performed with nominally Ca2+ -free simple salt solutions on the apical side (usually KCl) and Na2SO4-Ringer on the inner side of skins. The deposition of BaSO4 in the TJs was obtained by diffusion and/or migration through the paracellular path of Ba2+ from the apical solution and SO 4 2– from the inner solution. A brief presence (2 to 6 min) of apical Ba2+ (Ba2+ pulse) is followed (i.e., when Ba2+ is removed from the apical fluid) by a large increase of G, I, JSO 4 and JSUC, above pre-Ba2+ levels. These attain a steady state within 15 to 30 min (overshoot phase), characterizing a conspicuous increase of the paracellular permeability. During the overshoot phase, a second Ba2+ pulse blocks the paracellular route while apical Ba2+ is present, leading to a new and larger overshoot when the Ba2+ pulse is terminated. Addition of apical Ca2+ triggers the resealing of the TJs, resulting in a full recovery of G, I, JSO 4 and JSUC. This Ca2+ -induced recovery persists when apical Ca2+ is removed. The presence of a normal Ca2+ concentration in the inner bathing Ringer does not induce the recovery process. Tissues remain viable after being submitted to the Ba2+ treatment and the subsequent overshoot. Experiments performed in the urinary bladder of Rana catesbeiana and skins and urinary bladders of Bufo marinus indicate that Ba2+ effect can also be elicited in these tissues. The above results seem to report general properties of the TJs. The mechanism of cell death which occurs during Chagas' cardiopathy is disputed. To address this issue we analyzed the molecular pathways implicated in the death of cardiomyocytes during T. cruzi invasion and found that they undergo apoptosis during both in vitro and in vivo infections. However, the death rates and onset were related to the parasite stocks belonging to different biodemes, which can be correlated to the different histological inflammation findings that have already been reported. Our in vitro data provide additional support for this hypothesis since higher levels and earlier apoptosis induction were noted during the interaction with the Dm28c (type I) as compared to the Y and CL stocks (type II). Modifications of the surface carbohydrates of the infected cardiomyocytes were observed and these molecular events may be acting as "eat me" tags for their final engulfment by macrophages and/or other non-professional phagocytes. The analysis of other host cell types showed that the in vitro infection of fibroblasts did not result in host apoptosis even when a highly infective stock was used. Conversely, infected macrophages undergo apoptosis but at a higher degree than cardiomyocytes. Apoptotic intracellular parasites were observed to varied extents depending on the T. cruzi stock, which was related to the parasite invasion and proliferation. In summary, our results show that during T. cruzi infection, the extent of apoptosis varies according to the host cell type and the parasite stocks. Summary The localization of (Na+-K+) ATPase in the intact electrocyte of the electric organ of Electrophorus electricus (L.) and its subcellular fractions was investigated by biochemical and cytochemical methods. The distribution of AChE activity in the subcellular fractions was also comparatively analysed with this enzyme serving as a marker of the innervated membranes of the electrocyte. After application of cytochemical method of Farquhar and Palade to glutaraldehyde-fixed tissue, reaction was observed only at the membranes of vesicles localized at the periphery of the electrocyte. Previously fixed electrocytes, incubated in Ernst's medium showed reaction only at the vesicles whereas in unfixed tissue reaction also appeared at other membranes (surface and invaginations) of the anterior and posterior faces. This reaction was significantly inhibited in the presence of ouabain or in the absence of K+. Data on 34 patients with progressive pulmonary paracoccidioi domycosis seen in the State of Rio Grande do Sul (Brazil) are reviewed. Clinical manifestations were similar to those of a prolonged or a recurrent undifferentiated respiratory infection. Roentgenographic findings were also non-characteristic. Summary The authors review and comment on Brazilian literature on the observations of South American Blastomycotic lesion in perineal, anal and/or rectal localization.They relate a case of a patient presenting a perineal lesion two years after the excision of a tumor caused byP. brasiliensis in the brain.They comment onMackinnon's pathogenic theory of South American Blastomycosis that presumes that lungs are the portal of entry ofP. brasiliensis by inhalation of its spores. Thus, mucocutaneous lesions are secondary to a primary pulmonary localization by hematogenous route.

Buoyant density in CsCl, melting temperature, and G + C base content of the DNA from four species of Akodon (Rodentia, Cricetidae) were determined. The buoyant density values of 1.699–1.701 g/cm3 were in accordance with the data reported for other cricetids. No satellite bands were seen in neutral CsCl. The T m values determined in 1 × SSC ranged from 86.2 to 87.0 C, which corresponds to G + C contents of 41.2–43.2%. Kritskyia n. g. is proposed for K. moraveci n. sp. from the urinary bladder and ureters of Rhamdia quelen (Quoy & Gaimard, 1824), a pimelodid fish from the reservoir of the Hydroelectric Power Station of Passo Fundo, Rio Grande do Sul State, Brazil. The new genus is most closely related to Acolpenteron Fischthal & Allison, 1940, from which it differs by having a male copulatory complex composed of a coiled cirrus and a bipartite accessory piece, a sinistral vagina, and an antero-ventrally concave haptor which possesses 14 ventrally directed marginal hooks, but lacks an anterior rim and 4A hooks.Research fellow Conselho Nacional de Desenvolvimento Científico e Tecnológico, CNPq, Brasil.  相似文献

Summary The subcommissural organ (SCO), Reissner's fiber (RF) and its massa caudalis of lamprey larvae (Geotria australis) were investigated immunocytochemically by use of an antiserum raised against bovine RF as primary antibody. The affinities of RF and massa caudalis for Ricinus communis agglutinin I (RCA) with and without previous acid hydrolysis, concanavalin A (Con A), wheat-germ agglutinin (WGA), aldehyde fuchsin, and PAS reaction were also studied.SCO and massa caudalis were strongly immunoreactive, whereas RF proper was distinctly negative. RF did not react with Con A and RCA. Only the periphery of RF was WGA-positive. RCA showed affinity for RF only after acid hydrolysis. RF was homogeneously stained by the aldehyde-fuchsin and PAS-methods. At variance with RF proper, the periphery of the massa caudalis reacted with RCA without previous acid hydrolysis, but its core was WGA-positive and reacted with RCA only after hydrolysis. It is suggested that (i) RF has a coat of glycoproteins containing sialic acid as terminal residue, whereas the massa caudalis possesses a coat with galactose as terminal residue; (ii) in RF proper and the massa caudalis the spatial arrangement of glycoproteins might be different.Routine transmission electron-microscopic observations indicate that in larvae of Geotria australis an open communication exists between the ampulla caudalis and blood capillaries via large cavities or lacunae.Supported by Grant I 38259 from the Stiftung Volkswagenwerk, Federal Republic of Germany, Grant S-85-39 from the Dirección de Investigaciones, Universidad Austral de Chile, and Grant 6027 from Fondo Nacional de Desarrollo Científico y Tecnológico, Chile  相似文献

This work reports on the continuous extraction of a recombinant cutinase with reversed micelles using a perforated rotating disc contactor. Intracellular cutinase was directly extracted at 20°C with 100 mM AOT in isooctane from complex biological media of Escherichia coli disrupted cells. The optimal conditions for the direct extraction of the enzyme from media containing cell debris led to an extraction yield of 54.4%.  相似文献
