A double stain with Magdala red and anilin blue has sometimes given very satisfactory results; but, just as often, has been entirely worthless. The reason for the discrepancy seems to be that stains sold under the name of Magdala red are of various composition, some of them containing no Magdala red at all. The standardized stain phloxine seems to be identical with successful lots of Magdala red and results are rather uniformly successful. Detailed directions for staining with phloxine and anilin blue will be published in a forthcoming number of Stain Technology.  相似文献

Selenocysteine (Sec; U in one-letter code) is the twenty-first naturally occurring amino acid, with a selenium atom that gives this cysteine (Cys) homolog unique biochemical properties, including a high nucleophilicity and significant reactivity with electrophilic agents. This can be used in biotechnological Sec-dependent applications. Here, we describe how Sec can be introduced into a carboxy-terminal tetrapeptide motif (-Gly-Cys-Sec-Gly-COOH, known as a Sel-tag) for recombinant proteins by tailoring the encoding gene to become compatible with the Escherichia coli selenoprotein synthesis machinery. We also describe how the Sel-tag can be used as a basis for efficient one-step protein purification, rapid Sec-targeting protein labeling with electrophilic compounds, or radiolabeling with the positron emitter 11C.  相似文献

Because of an increasing incidence of poisoning with the newer organophosphorus anticholinesterase insecticides, these compounds have been reviewed in terms of their history and pharmacology, relationship with other drugs, factors affecting toxicity, mechanism of action, toxic signs and treatment. The modern organophosphorus pesticide requires metabolic conversion before toxicity develops. Insects have a greater propensity for this conversion than humans. Nevertheless, this conversion does occur in humans and can be potentiated by other drugs. Toxicity also varies with age, sex, route and frequency of administration, and previous exposure. The mechanism of toxicity is inhibition of acetylcholinesterase, causing an intoxicating build-up of acetylcholine. Signs and symptoms consist of the clinical manifestations of unopposed parasympathetic and central activity. Treatment must be initiated early. Respiration must be maintained and the effects of acetylcholine must be counteracted by massive doses of atropine. Metaraminol enhances the antagonistic action of atropine against acetylcholine and may also be given. Once acetylcholinesterase is inactivated, restoration is slow. Recovery can be accelerated by enzyme reactivators like the oxime compounds. Pyridine aldoxime (Pralidoxime, Protopam, P2S and 2-PAM) can be given in combination with atropine and metaraminol (AMP therapy) and may be the treatment of choice.  相似文献

Malignant cells were detected in the pleural effusion of a patient with three separate primary malignancies. These cells were judged by conventional morphologic studies to be poorly differentiated cells, but cytochemical studies showed them to be granulocytic precursor cells. The use of cytochemical or immunochemical techniques may be most practical for the cytodiagnosis of malignant cells in serous effusions.  相似文献

There are essentially three ways in which four species may be related in a phylogenetic tree graph. It is usual to compute for each of these three possibilities the smallest number of mutations that could have brought about the observed distribution of characteristics among the four species. The graph that minimizes this number is then preferred. In fact, the hypothesis that the graph chosen in this way is correct may be accepted with confidence if the minimum is strong in a sense described here. In principle, the theory could be extended to treat sets of more than four species.  相似文献

A prospective survey was carried out to determine the incidence of hypercalcemia in hospitalized patients with lymphoma. Serum calcium determinations were carried out during 440 admissions on 152 patients, most of whom had widespread lymphoma. Forty-one of the 152 patients died, all having had calcium levels recorded on that admission. Two of the 152 patients (1.3%) were found to have hypercalcemia. By comparison hypercalcemia occurred in 6 of 34 patients (17.6%) with multiple myeloma.Hypercalcemia is uncommonly associated with lymphoma but may be seen particularly in centres where many patients with this disease are treated. Hypercalcemia readily responds to therapy and is a reversible cause of morbidity, but when associated with lymphoma it usually indicates widespread disease. The mechanism of hypercalcemia is unknown but there is strong evidence for humoral factors that may or may not be related to parathyroid hormone.  相似文献

This paper describes the experimental and clinical findings of acrylamide intoxication in a human being. It is believed that this is the first such case to be recorded in the medical literature.Acrylamide is widely used as a “chemical grout”. It is pumped into dirt, clay and stone walls of excavations in a liquid state together with a catalyst, and it then polymerizes to make a watertight shield.This chemical is neurotoxic in its non-polymerized form and can be absorbed through the intact skin, mucous membranes and lungs. In spite of warnings with regard to its handling, this worker became careless, and developed a contact dermatitis and a polyneuropathy with bluish cold extremities which dripped perspiration.In six months'' time after his removal from contact with the chemical the patient made a complete clinical recovery. Frozen sections, 15-50 µ thick, are soaked for 5 minutes or longer in a mixture of equal parts of 1.5% aqueous gelatin and 80% alcohol, and teased onto a slide. After allowing excess fluid to evaporate, sections will be moist and can be blotted with filter paper that may require dampening with 95% alcohol. Immersed in 95% alcohol, the remaining gelatin will congeal, anchoring the section to the slide. If necessary, the sections can subsequently be coated with celloidin.  相似文献

Fenoprofen, a compound with analgesic, anti-inflammatory, and antipyretic properties in animals, has been compared with placebo in a double-blind cross-over trial in 60 patients with rheumatoid arthritis. There was a statistically highly significant reduction in pain, duration of morning stiffness, analgesic requirements, and articular index, with increase in grip strength. There was no significant reduction in joint size or temperature. In a subsequent double-blind group-comparative study fenoprofen proved to be as effective as aspirin in relieving the symptoms of rheumatoid arthritis, with strikingly fewer side effects. Almost half of the patients taking aspirin were unable to tolerate the drug in adequate dosage for six months. The remainder were able to take on average only 4 g daily, and at this dose almost half still complained of tinnitus and deafness.Fenoprofen is likely to be useful for patients who cannot tolerate aspirin and other more toxic anti-inflammatory drugs or whose disease is not of sufficient severity to justify their use.  相似文献

Hypertrophic cardiomyopathy (HCM) is increasingly being diagnosed in pregnant women. Women with HCM generally tolerate pregnancy well. The risk is however higher in women who are symptomatic before pregnancy or in those with severe left ventricular outflow tract obstruction. The incidence of arrhythmias does not appear to be increased during pregnancy and maternal mortality is low. Prior to conception, women with HCM should have a risk assessment as well as genetic counselling. During pregnancy beta-blockers should be continued and the judicious use of diuretics may be required to treat symptoms of dyspnoea. A vaginal delivery with regional anaesthesia is usually appropriate. Women should be managed by a specialist multidisciplinary team.  相似文献

Sustained, long-term cognitive workload is associated with variations and decrements in performance. Such fluctuations in vigilance can be a risk factor especially during dangerous attention demanding activities. Functional MRI studies have shown that attentional performance is correlated with BOLD-signals, especially in parietal and prefrontal cortical regions. An interesting question is whether these BOLD-signals could be measured in real-world scenarios, say to warn in a dangerous workplace whenever a subjects'' vigilance is low. Because fMRI lacks the mobility needed for such applications, we tested whether the monitoring of vigilance might be possible using Near-Infrared Spectroscopy (NIRS). NIRS is a highly mobile technique that measures hemodynamics in the surface of the brain. We demonstrate that non-invasive NIRS signals correlate with vigilance. These signals carry enough information to decode subjects'' reaction times at a single trial level.  相似文献

The persistence of highly endemic parasitic, bacterial and viral diseases makes individuals and populations vulnerable to emerging and re-emerging diseases. Evaluating the role of multiple component, often interacting, causes of disease may be impossible with research tools designed to isolate single causes. Similarly, it may not be possible to identify statistically significant treatment effects, even for interventions known to be effective, when multiple morbidities are present. Evidence continues to accumulate that nutritional deficiencies, bacterial, viral and parasitic coinfections accelerate HIV transmission. Inclusion of antiparasitics and other beneficial interventions in HIV-prevention protocols is impeded by reliance on inappropriate methodologies. Lack of full scientific certainty is not a reason for postponing safe, cost-effective measures to prevent irreversible damage.  相似文献

The technique of vital staining with a fluorochrome and subsequent flow cytometry has been employed to investigate the DNA content of living sperm. There have been several reports of chromosomal damage caused by staining cells with a bis-benzimidazole dye. Hoechst 33342, either with or without subsequent flow cytometry. Therefore the use of this technique might be expected to affect adversely the fertilising capacity of inseminated sperm, or to result in the production of congenital deformities in the young. Insemination experiments to date have resulted in the production of more than 400 offspring in four species, including 5 successive generations of rabbits, where stained or sorted sperm were used. All progeny were observed to be normal by anatomical criteria. There was no evidence to suggest that sperm treated in this manner were unable to fertilise oocytes in vivo or that embryonic development was affected.  相似文献

Biotechnological methods can be used for cryptography. Here two different cryptographic approaches based on DNA binary strands are shown. The first approach shows how DNA binary strands can be used for steganography, a technique of encryption by information hiding, to provide rapid encryption and decryption. It is shown that DNA steganography based on DNA binary strands is secure under the assumption that an interceptor has the same technological capabilities as sender and receiver of encrypted messages. The second approach shown here is based on steganography and a method of graphical subtraction of binary gel-images. It can be used to constitute a molecular checksum and can be combined with the first approach to support encryption. DNA cryptography might become of practical relevance in the context of labelling organic and inorganic materials with DNA 'barcodes'.  相似文献

Pulmonary involvement is common in sarcoidosis, an immune-mediated inflammatory disorder that is characterized by non-caseating granulomas in tissue. Sarcoid patients with advanced pulmonary disease, especially end-stage pulmonary fibrosis, risk developing pulmonary hypertension (World Health Organization group III pulmonary hypertension secondary to hypoxic lung disease). Increased levels of endothelin (ET)-1 in plasma and bronchoalveolar lavage of some sarcoid patients suggest that ET-1 may be driving pulmonary fibrosis and sarcoidosis-associated pulmonary hypertension. Although a relationship between raised levels of ET-1 and clinical phenotype is yet to be identified, early evidence from studies of ET-1 blockade with drugs such as bosentan is encouraging. Such therapy possibly could be combined with standard anti-inflammatory agents to improve outcome.  相似文献

The intrinsic brightness of fluorescent proteins has been taken advantage of to develop a technology of whole-body imaging of tumors and gene expression in mouse internal organs. Stable transformation with fluorescent protein genes can be effected using retroviral vectors containing a selectable marker such as neomycin resistance. The cells that stably express fluorescent proteins can then be transplanted into appropriate mouse models. For whole-body imaging, nude mice are very appropriate. If wild-type mice are used, then hair must be removed by shaving or depilation. The instruments used can range from a simple LED flashlight and appropriate excitation and emission filters to sophisticated equipment such as the Olympus OV100 with a wide range of magnification, enabling both macroimaging and microimaging. It is crucial that proper filters be used such that background autofluorescence is minimal. Fluorescent protein-based imaging technology can be used for whole-body imaging of fluorescent cells on essentially all organs. The timeline for these experiments varies from 2 days to 2 months.  相似文献

Immunotherapy of cancer with dendritic-cell-based vaccines   总被引:32,自引:0,他引:32  
 Animal studies have shown that vaccination with genetically modified tumor cells or with dendritic cells (DC) pulsed with tumor antigens are potent strategies to elicit protective immunity in tumor-bearing animals, more potent than “conventional” strategies that have been tested in clinical settings with limited success. While both vaccination strategies are forms of cell therapy requiring complex and costly ex vivo manipulations of the patient’s cells, current protocols using dendritic cells are considerably simpler and would be more widely available. Vaccination with defined tumor antigens presented by DC has obvious appeal. However, in view of the expected emergence of antigen-loss variants as well as natural immunovariation, effective vaccine formulations must contain mixtures of commonly, if not universally, expressed tumor antigens. When, or even if, such common tumor antigens will be identified cannot be, predicted, however. Thus, for the foreseeable future, vaccination with total-tumor-derived material as source of tumor antigens may be preferable to using defined tumor antigens. Vaccination with undefined tumor-derived antigens will be limited, however, by the availability of sufficient tumor tissue for antigen preparation. Because the mRNA content of single cells can be amplified, tumor mRNA, or corresponding cDNA libraries, offer an unlimited source of tumor antigens. DC transfected with tumor RNA were shown to engender potent antitumor immunity in animal studies. Thus, immunotherapy using autologous DC loaded with unfractionated tumor-derived antigens in the form of RNA emerges as a potentially powerful and broadly useful vaccination strategy for cancer patients. A procedure is described by which muscle fibers can be electrophysiologically studied, intracellulary marked with horseradish peroxidase (HRP), and stained for acetylcholinesterase (AChE) activity to identify nerve endings. Following electrophysiological characterization, 4% HRP is injected through the recording microelectrode with small depolarizing pulses. Muscles are fixed, washed, and stained with a Koelle procedure for AChE modified by extending the incubation period to 15 hours to identify small nerve endings. The HRP is visualized with 3,3'-diaminobenzidine tetrahydrochloride or benzidine dihydrochloride. Stained muscles are embedded in Epon and sectioned. Lucifer yellow dye is also demonstrated to be an excellent intracellular marker of muscle fibers but it cannot be combined with AChE staining.  相似文献

Nerve fibers, in organs fixed with Bouin's fluid, are usually refractive to the Davenport silver technic. The axons, however, can be successfully stained if the sections, on slides, are given a preliminary treatment with concentrated pyridine (1 hour), then a 24-hour bath of ammoniated alcohol (99 cc. 80% alcohol, 1 cc 28% ammonium hydroxide) and an interval in 40% aqueous silver nitrate (6-8 hours) before being immersed in the acidified alcoholic silver solution of Davenport. Following the silvering, reduction and toning of the axons, according to the procedure of Davenport, the surrounding non-nervous tissue elements can be counterstained with a combination of either azocarmine, light green and orange G, or azocarmine, aniline blue and orange G.  相似文献
