Aquatic snails from south western Zimbabwe belonging to theBulinus trunscatus/tropicus complex vary widely in shell formsuggesting that more than one taxon could be present. This possibilitywas investigated by making observations on snail samples from13 populations from the Plumtree area, in respect of allozymevariation (5 polymorphic loci), shell morphology (9 variables),copulatory organ and chromosome number. Comparative data wereobtained from snails from north western Zimbabwe identifieddefinitely as B. tropicus. Analysis of the genetic structurerevealed a high degree of polymorphism (P) ranging from 0.29–0.80among populations from Plumtree and expected heterozygosity(He) from 0.02–0.22. No enzymatic diagnostic loci werefound which could differentiate the different morphs or populationsand discriminant function analysis on the morphological datashowed an overlap of morphs among populations. Snails analyzedfor chromosome number were all diploid (2n = 36). Snails exposedto Schistosoma haematobium mira-cidia were all refractory. Thisinformation supports the view of a single species, B. tropicus,which is differentiated due to migration barriers and whereenvironmental variables might be implicated in the morphometricdivergence. This paper presents the evaluation of genetic diversity in 29populations of wild taxa of theBrassica oleraceaL. group (2n=18)and two cultivars, using RAPDs as molecular markers. In a previouspaper (Lázaro and Aguinagalde,Annals of Botany82: 000–000,1998), 11 isozymes were used for the same purpose. Results obtainedwith the two molecular markers (isozymes and RAPDs) are compared.DNA from ten individuals per population was analysed using sixdifferent primers; the 151 detected bands were polymorphic,11 were common to all species, six to all taxa, only one toevery population; and no bands were shared by every individual.The dendrogram obtained using genetic distances clustersB. oleraceapopulationswithB. bourgeaui, B. alboglabra, B. montanaandB. incana. The life cycle, population dynamics, growth and secondary productionof the land snail C. vindobonensis were studied in northernGreece. Demographic analysis of the populations of C. vindobonensisrevealed that a) three cohorts were present in the field throughoutthe year, b) the reproductive period started in late April-Mayand the newly hatched snails appeared in the beginning of June,and c) increased growth rates were observed during spring andearly summer, but also during autumn for the newly hatched snails. According to von Bertalanffy's method C. vindobonensis needs7 years to attain its maximum size measured in the field. Mortalityrate is very high during the first year of life, while lifeexpectancy is higher during the second year of life and decreasesafterwards. Net reproductive rate (Ro) was equal to 3.1 andthe finite capacity for increase (antilogerc) was equal to 1. Estimated annual secondary production with Hynes' frequencymethod revealed a mean standing crop (B) of 0.99 g/m2/year anda production (P) of 1.3 ± 0.11 g/m2/year. The growth rates of ovotestis and individual accessory sexualorgans (ASO) of Biomphalaria glabrata snails were studied forcontrols and for immature and mature snails infected with Schistosomamansoni. The infection of immature B. glabrata strongly delaysgrowth of the ovotestis and inhibits the development of theaccessory sexual organs. There is no significant differenceup to 2 weeks post infection in the volume of the ovotestisand the ASO between mature infected B. glabrata and controlsnails. From 3 to 4 weeks post infection there was a reductionin the volume of the ovotestis and the ASO of infected matureB. glabrata; then growth of the ovotestis, albumen gland andfemale organs was stopped, but the effect of infection was lessconsistent for the male organs. For a parasite, immature andmature snails have to be considered as two different resourceenvironments, each having at infection time a particular patternof resource allocation, towards growth for juvenile and towardsreproduction for adult snails, changing the possible energyutilization patterns which can be used by the trematode. Surfacing and water-leaving behaviour (quitting) of aquaticpulmonate snails was investigated in the laboratory. Regressioncoefficients of surfacing rate per hour for 16 snails in 200cm3 of water were 1.28 for L. peregra, 1.35 for B. glabrata(albino form), 0.47 for B. glabrata (pigmented form) and 0.17for B. jousseaumei. Disturbance, snail size and conditioningof the water were relatively unimportant factors; water volumeadjusted by depth was also unimportant up to 50 mm depth. Importantfactors controlling surfacing appeared to be population density,time of year, temperature, water volume adjusted by perimeterand dissolved oxygen concentration. The response to the latterwas not clear and both B. glabrata and L. peregra orientatedalmost as well to a nitrogen/water interface as to an air/waterinterface. Water-quitting regressed on volume, which had beenadjusted by perimeter variation, revealed a curvilinear relationshipfor B. glabrata, with maximum quitting at 1 snail per 7cm3 (1snail per 14cm of climbable perimeter). L. peregra showed anegative exponential curve, with maximum quitting at 1 snailper 3 cm3 of water (1 snail per 8 cm of climbable perimeter).These data could be relevant to design and management of irrigationchannels where bilharzia might occur. To estimate the geographical extent of introgression, we studiedthe genetic structure of sympatric and allopatric populationsof hybridizing freshwater snail species Viviparus ater and V.contectus in central Europe. Six allozyme loci which were variablein Lake Garda, Italy in a previous study (five nearly diagnosticloci between the two species and one highly polymorphic locusin V. contectus) were analyzed from ten sympatric locationsand four allopatric populations each for the two species. Presumablyintrogressed genes (low allele frequencies) were found fromat least one locus in seven out of the ten sympatric sites.These seven sites covered most of northern Italy. The data indicatethat introgression has occurred from Viviparus contectus toV. ater and vice versa. Therefore, there is a possibility ofwidespread introgression or mosaic zones in nature. However,we cannot rule out that the observed patterns are due to theshared ancestry. V. ater possessed low genetic variation (thejackknifed mean of Wright's FST±S.E. over four loci was0.041±0.004). On the other hand, V. contectus showedhigh genetic differentiation (the jackknifed mean of FST±S.E. over six loci was 0.546±0.166). Although introgressionmay have caused evolutionary changes in V. ater and V. contectus,it was not strong enough to level out the genetic differencesbetween the two species, which may have originated from isolationamong populations in V. contectus and a past bottleneck eventin V. ater. Changes in population of Bulinus globosus, B. forskalii andLymnaea natalensis were studied in four different locationsnamed Sites A, B, C. and D in Bo town Sierra Leone. The snailpopulations and oxygen concentrations of water increased inthe rainy season. The biological oxygen demand did not havesignificant correlation with changes in snail populations inSites B and D. B. globosus was absent from Site A while B. forskaliiwas the predominant snail in Site C. Pascopyrum smithii (C3) andBouteloua gracilis (C4) are importantforage grasses native to the Colorado shortgrass steppe. Thisstudy investigated photosynthetic responses of these grassesto long-term CO2enrichment and temperature in relation to leafnonstructural carbohydrate (TNC) and [N]. Glasshouse-grown seedlingswere transferred to growth chambers and grown for 49 d at twoCO2concentrations (380 and 750 µmol mol-1) at 20 and 35°C, and two additional temperatures (25 and 30 °C) at750 µmol mol-1CO2. Leaf CO2exchange rate (CER) was measuredat a plant's respective growth temperature and at two CO2concentrationsof approx. 380 and 700 µmol mol-1. Long-term CO2enrichmentstimulated CER in both species, although the response was greaterin the C3,P. smithii . Doubling the [CO2] from 380 to 750 µmolmol-1stimulated CER ofP. smithii slightly more in plants grownand measured at 30 °C compared to plants grown at 20, 25or 35 °C. CO2-enriched plants sometimes exhibited lowerCER when compared to ambient-grown controls measured at thesame [CO2], indicating photosynthetic acclimation to CO2growthregime. InP. smithii , such reductions in CER were associatedwith increases in TNC and specific leaf mass, reductions inleaf [N] and, in one instance, a reduction in leaf conductancecompared to controls. InB. gracilis , photosynthetic acclimationwas observed more often, but significant changes in leaf metabolitelevels from growth at different [CO2] were generally less evident.Temperatures considered optimal for growth (C3: 20 °C; C4:35 °C) sometimes led to CO2-induced accumulations of TNCin both species, with starch accumulating in the leaves of bothspecies, and fructans accumulating only inP. smithii. Photosynthesisof both species is likely to be enhanced in future CO2-enrichedand warmer environments, although responses will sometimes beattenuated by acclimation. Random amplified polymorphic DNA (RAPD) analysis was used tostudy variation among and within selectedIxora (Rubiaceae) populationsand mutants. Six populations of I. congesta yielded identicalbanding patterns suggesting genetic uniformity of this species.However, six populations of I. coccinea varieties (three red-flowered,two yellow-flowered and one red-flowered wild-type) exhibitedinfraspecific differences in RAPD profiles. Small and largeleaves of an atavistic mutant cultivar of I. coccinea were alsosubjected to RAPD analysis. An extra band was amplified in thelarge leaves that was absent in small leaves, suggesting thatthe phenotypic alteration in this taxon is due to genetic mutationrather than epigenetic changes. Similarly, an extra band wasdetected in the white sectors of I. Variegated compared to thegreen sectors, suggesting that the shoot apical meristems ofthis cultivar exist as a genetic chimera. Background and Aims For rare endemics or endangered plantspecies that reproduce both sexually and vegetatively it iscritical to understand the extent of clonality because assessmentof clonal extent and distribution has important ecological andevolutionary consequences with conservation implications. Asurvey was undertaken to understand clonal effects on fine-scalegenetic structure (FSGS) in two populations (one from a disturbedand the other from an undisturbed locality) of Echinosophorakoreensis, an endangered small shrub belonging to a monotypicgenus in central Korea that reproduces both sexually and vegetativelyvia rhizomes. • Methods Using inter-simple sequence repeats (ISSRs) asgenetic markers, the spatial distribution of individuals wasevaluated using Ripley's L(d)-statistics and quantified thespatial scale of clonal spread and spatial distribution of ISSRgenotypes using spatial autocorrelation analysis techniques(join-count statistics and kinship coefficient, Fij) for totalsamples and samples excluding clones. • Key Results A high degree of differentiation betweenpopulations was observed (ST(g) = 0·184, P < 0·001).Ripley's L(d)-statistics revealed a near random distributionof individuals in a disturbed population, whereas significantaggregation of individuals was found in an undisturbed site.The join-count statistics revealed that most clones significantlyaggregate at 6-m interplant distance. The Sp statistic reflectingpatterns of correlograms revealed a strong pattern of FSGS forall four data sets (Sp = 0·072–0·154), butthese patterns were not significantly different from each other.At small interplant distances (2 m), however, jackknifed 95% CIs revealed that the total samples exhibited significantlyhigher Fij values than the same samples excluding clones. • Conclusion The strong FSGS from genets is consistentwith two biological and ecological traits of E. koreensis: bee-pollinationand limited seed dispersal. Furthermore, potential clone matesover repeated generations would contribute to the observed highFij values among genets at short distance. To ensure long-termex situ genetic variability of the endangered E. koreensis,individuals located at distances of 10–12 m should becollected across entire populations of E. koreensis.  相似文献

Background and Aims The desert legume genus Ammopiptanthuscomprises two currently endangered species, A. mongolicus andA. nanus. Genetic variability and genetic differentiation betweenthe two species and within each species were examined. • Methods Inter-simple sequence repeat (ISSR) marker datawere obtained and analysed with respect to genetic diversity,structure and gene flow. • Key Results Despite the morphological similarity betweenA. mongolicus and A. nanus, the two species are geneticallydistinct from each other, indicated by 63 % species-specificbands. Low genetic variability was detected for both populationlevel (Shannon indices of diversity Hpop = 0·106, percentageof polymorphic loci P = 18·55 % for A. mongolicus; Hpop= 0·070, P = 12·24 % for A. nanus) and specieslevel (Hsp = 0·1832, P = 39·39 % for A. mongolicus;Hsp = 0·1026, P = 25·89 % for A. nanus). Moderategenetic differentiation was found based on different measures(AMOVA ST and Hickory B) in both A. mongolicus (0·3743–0·3744)and A. nanus (0·2162–0·2369). • Conclusions The significant genetic difference betweenthe two species might be due to a possible vicariant evolutionaryevent from a single common ancestor through the fragmentationof their common ancestor's range. 本文应用ISSR分子标记技术对中国桃蛀螟Conogethes punctiferalis 11个地理种群的基因组DNA进行了遗传多样性和遗传分化分析。从34条引物中筛选出6条用于ISSR多态性分析, 共扩增出211条带, 其中209条具多态性, 总的多态性条带百分率为99.05%。11个种群的遗传距离在0.0059~0.0237之间, Nei氏基因多样性指数为0.1750, Shannon信息指数为0.2966, 遗传分化系数为0.053, 基因流高达8.8724。结果提示中国桃蛀螟地理种群因基因流水平较高而种群遗传分化水平保持较低。  相似文献

运用16种酶蛋白编码的23个遗传座位对突尼斯非洲跳鼠(Jaculus jaculus)和埃及跳鼠(J. orientalis)自然群体的遗传变异和分化进行了电泳分析。结果表明,与其他啮齿动物等哺乳动物的相关数据比较,发现这两个种群体的遗传变异水平较低。非洲跳鼠群体的观测杂合度 (Hobs) 为0.08—0.19,多态座位百分比(P)为26.2%—45.2%,每个座位的平均等位基因数(A)为1.1—1.4;埃及跳鼠的Hobs为0.10—0.15,P为29.3%—44.1%,A为1.1—1.7。两个种群体各自的遗传分化程度较低(非洲跳鼠和埃及跳鼠的Fst分别为0.0017和0.0019)。而两个种群体间的Fst为0.607(P<0.05),表明两个种之间高度的遗传分化。本研究支持这两个种分类地位的合法性,并强调了地理因素(环境类型和生物气候阶段)对两个种遗传结构的影响。  相似文献

Bot. 38: 67–76 The permeabilities of the amines, ammonia (NH3), methylamine(CH3NH2) and ethylamine (CH3CH2NH2) in the giant-celled charophyteChara corallina (C. australis) R.Br. have been measured andcompared. The permeabilities were corrected for uptake fluxesof the amine cations. Based on net uptake rates, the permeabilityof ammonia was 6?4?0?93 µm s–1 (n = 38). The permeabilitiesof methylamine and ethylamine were measured in net and exchangeflux experiments. The permeabilities of methylamine were notsignificantly different in net and exchange experiments, norto that of ammonia (Pmethylamine = 6?0?0?49 µm s–1(n = 44)). In net flux experiments the apparent permeabilityof ethylamine was slightly greater than that of ammonia andmethylamine (Pethylamine, net = 8?4?1?2 µm s–1 (n= 40)) but the permeability of ethylamine based on exchangeflux data was significantly higher (Pethylamine, exchange =14?1?2 µm s–1 (n = 20)). Methylamine can be validlyused as an ammonium analogue in permeability studies in Chara. The plasmalemma of Chara has acid and alkaline bands; littlediffusion of uncharged amines would occur across the acid bands.The actual permeability of amines across the alkaline bandsis probably about twice the values quoted above on a whole cellbasis i.e. the permeability of ammonia across the permeablepart of the plasmalemma is probably about 12 µm s–1. This study investigated how CO2and temperature affect dry weight(d.wt) accumulation, total nonstructural carbohydrate (TNC)concentration, and partitioning of C and N among organs of twoimportant grasses of the shortgrass steppe,Pascopyrum smithiiRydb. (C3) andBouteloua gracilis(H.B.K.) Lag. ex Steud. (C4).Treatment combinations comprised two temperatures (20 and 35°C)at two concentrations of CO2(380 and 750 µmol mol-1),and two additional temperatures of 25 and 30°C at 750 µmolmol-1CO2. Plants were maintained under favourable nutrient andsoil moisture and harvested following 21, 35, and 49d of treatment.CO2-induced growth enhancements were greatest at temperaturesconsidered favourable for growth of these grasses. Comparedto growth at 380 µmol mol-1CO2, final d.wt of CO2-enrichedP.smithiiincreased 84% at 20°C, but only 4% at 35°C. Finald.wt ofB. graciliswas unaffected by CO2at 20°C, but wasenhanced by 28% at 35°C. Root:shoot ratios remained relativelyconstant across CO2levels, but increased inP. smithiiwith reductionin temperature. These partitioning results were adequately explainedby the theory of balanced root and shoot activity. Favourablegrowth temperatures led to CO2-induced accumulations of TNCin leaves of both species, and in stems ofP. smithii, whichgenerally reflected responses of above-ground d.wt partitioningto CO2. However, CO2-induced decreases in plant tissue N concentrationswere more evident forP. smithii. Roots of CO2-enrichedP. smithiihadgreater total N content at 20°C, an allocation of N below-groundthat may be an especially important adaptation for C3plants.Tissue N contents ofB. graciliswere unaffected by CO2. Resultssuggest CO2enrichment may lead to reduced N requirements forgrowth in C3plants and lower shoot N concentration, especiallyat favourable growth temperatures. The effects of dissolved Pb, Cd and Hg on growth and survivalof adult Biomphalaria glabrata Say. uninfected and infectedwith Schistosoma mansoni Sambon, were examined. Pb at concentrationsfrom 0.25–100 µM, Cd from 0.075–0.25 µMand Hg from 0.25–1 µM significantly reduced growthand survival of normal snails. With each metal the effects increasedwith increasing concentration. The LC 25 at 2 wks exposure was82 ± 19, 0.22 ± 0.04 and 0.94 ± 0.13 µM(x ± S.E.) for Pb, Cd and Hg, respectively. Snails exposedto heavy metals continued to be reproductively active, but theegg production was highly variable and no significant effectof heavy metal exposure was demonstrated. Infection also decreasedsurvival and growth of snail hosts and infected individualsexposed to heavy metals displayed the greatest mortality. TheLC 25 for infected snails at 2 wks exposure was 8 ± 3,0.9 ± 0.02 and 0.29 ± 0.08 for Pb, Cd and Hg respectively.A significant interaction between heavy metal exposure and infectionwas apparent. Infected snails were not reproductively active.Cercarial shedding by infected snails was significantly reducedin the presence of heavy metals and by 6 wks shedding had ceasedat the highest metal concentrations. Random amplified polymorphic DNA (RAPD) profiling revealed geneticdiversity among three populations ofLicuala glabra var. glabra,an understorey palm growing in the dipterocarp forests of Terengganu,Malaysia. The Bukit Bauk population had individuals withoutaerial stems. The Bukit Besi individuals had stem heights to30 cm, while those of the Rasau Kerteh population had stemsup to 4 m in height. Southern blot analysis confirmed that anapparent monomorphic band generated by the primer OPU-17 amongthe three different populations corresponds to the same locus.Six primers were used to generate 87 RAPD bands that formed2-state character raw data for estimating genetic distancesusing the simple matching coefficient of similarity. The phenogramgenerated by UPGMA of the RAPD data grouped the individualsfrom the three populations under three clusters. This was inagreement with the morphological grouping. The infraspecificL. glabra var.selangorensis Becc. was grouped closer to theL. glabra var. glabra individuals than to the outgroupL. scortechiniiBecc. Principal component analysis supported the observationfrom the phenogram. The results show discrete genetic structuringin the populations studied and provide useful information forfuture conservation measures. Marine coccolithophorids, Emiliania huxleyi, Gephyrocapsa oceanicaand Helladosphaera sp. were found to require selenium for theirgrowth. The optimum concen trations were 1–10 nM for SeO2and SeO32– and above 1 µM for SeO24– Duringthe depletion of selenium algal growth was strongly suppressedaccompanied with the decrease in net photosynthesis. Species in the genus Epioblasma have specialized life historyrequirements and represent the most endangered genus of freshwatermussels (Unionidae) in the world. A genetic characterizationof extant populations of the oyster mussel E. capsaeformis andtan riffleshell E. florentina walkeri sensu late was conductedto assess taxonomic validity and to resolve conservation issuesfor recovery planning. These mussel species exhibit pronouncedphenotypic variation, but were difficult to characterize phylogeneticallyusing DNA sequences. Monophyletic lineages, congruent with phenotypicvariation among species, were obtained only after extensiveanalysis of combined mitochondrial (1396 bp of 16S, cytochrome-b,and ND1) and nuclear (515 bp of ITS-1) DNA sequences. Incontrast, analysis of variation at 10 hypervariable DNA microsatelliteloci showed moderately to highly diverged populations basedon FST and RST values, which ranged from 0.12 to 0.39 and 0.15to 0.71, respectively. Quantitative variation between specieswas observed in fish-host specificity, with transformation successof glochidia of E. capsaeformis significantly greater (P<0.05)on greenside darter Etheostoma blennioides, and that of E. f.walkeri significantly greater (P<0.05) on fantail darterEtheostoma flabellare. Lengths of glochidia differed significantly(P<0.001) among species and populations, with mean sizesranging from 241 to 272 µm. The texture and colourof the mantle-pad of E. capsaeformis sensu stricto is smoothand bluish-white, whereas that of E. f. walkeri is pustuledand brown, with tan mottling. Based on extensive molecular,morphological and life history data, the population of E. capsaeformisfrom the Duck River, Tennessee, USA is proposed as a separatespecies, and the population of E. f. walkeri from Indian Creek,upper Clinch River, Virginia, USA is proposed as a distinctsubspecies. RAPD variation was examined in nine populations of Campanulamicrodonta Koidz., endemic to the Izu Islands, Japan. Ninety-eightbands were obtained for all populations, 94% of which were polymorphicat least within a population. Shannon's H values were calculated;these have frequently been used in RAPD studies to estimategenetic diversity. The values within populations did not correlatewith the allozyme gene diversity estimated by a previous studyor with distance from the Japanese mainland. The possible reasonsfor this discrepancy are different selection regimes betweenthe two markers, higher RAPD mutation rates, and each marker'sdifferent coverage of genomes. Cluster analysis of genetic similaritiessuggested that colonization of each island probably occurredonce, except for Miyake Island, where immigration has occurredat least twice.
