Newcastle disease (Paramyxoviridae) is a highly infectious virus shed in the faeces of infected birds. Non-biting Muscid flies characteristically visit manure and decaying organic material to feed and oviposit, and may contribute to disease transmission. The housefly, Musca domestica (Linnaeus, 1758) (Diptera: Muscidae), has been implicated as a mechanical vector of numerous pathogens. In this study 2000 aerial net-captured houseflies were examined for their ability to harbour Newcastle disease virus (NDV). In an adjacent study, laboratory-reared flies were experimentally exposed to NDV La Sota strain. The virus was detected in the dissected gastrointestinal tract of laboratory-exposed flies for up to 72 h post-exposure, whereas the untreated control flies were negative.  相似文献

Abstract. The role of houseflies as agents in the spread of bacterial diseases has been thoroughly investigated, yet the fate of bacteria ingested by flies has not. We examined the physical location of the bacterial enteropathogen Aeromonas caviae in the midgut of laboratory‐reared adult houseflies. Food ingested by houseflies was separated from the midgut epithelium by a double‐layered peritrophic matrix (PM). The inner PM intimately enveloped the food as fecal pellets (food boluses), while the outer PM appeared as a long continuous tube. In flies fed a suspension of A. caviae, live bacteria were not observed within the inner PM, but were compartmentalized between folds of the PM in the inter‐PM space. Similar observations were made for flies fed a suspension of Serratia liquefaciens and for highly contaminated feral flies. Isolates of both A. caviae and S. liquefaciens were chitinolytic (as demonstrated by clearing zones on chitin agar), but the potential role of bacterial enzymes in the alteration of PM morphology or formation needs further investigation.  相似文献

Breeders of greyhound dogs traditionally feed racing animals and nursing bitches raw meat, and that meat generally is obtained frozen from commercial renderers. Previous studies have shown that the rendered meat is frequently contaminated with enteric bacteria, including Salmonella spp., and that during thawing the rendered meat is exposed to filth flies common in dog kennels. Nursing greyhound pups tend to experience a high morbidity and mortality from intestinal infections, and we attempted to determine in this study whether enterics could be spread to pups through contaminated flies. At intervals during 1995 and 1996, flies were trapped or were net-collected from 10 dog breeding kennels in the region around Abilene, KS. Trapped flies were identified and counted to determine population numbers, and netted flies were cultured in tetrathionate broth and streaked to medium selecting for Salmonella sp. and other lactose-negative Gram (−) bacteria. The relative numbers of different fly species varied with the sampling method, but traps and sweep nets produced similar proportions of the different fly species. Blow flies were twice as likely to be contaminated with enteric bacteria as any other fly. The most common enteric bacteria found were Proteus spp., followed by Providencia spp., Pseudomonas spp., and Salmonella spp. The incidence of Salmonella and Proteus spp. seemed to correlate more with accessibility of flies to dog excrement than to rendered meat. The apparent high incidence of enteric contamination of filth flies clearly implicates them as vectors of enteric diseases in kennels. Received: 7 July 1997 / Accepted: 26 August 1997  相似文献

Various bacterial strains were cultured in a laboratory medium, on chicken breast meat or derived from faeces of orally infected chicks. Suspensions of these bacterial cultures were brought in contact with chicken breast meat. The way in which the bacteria were cultured had little influence on the number of bacteria which attached to the chicken breast meat surface or on as the strength of attachment ( i.e. the easiness of removal of attached bacteria). Only bacteria from the faeces of freshly infected chicks (24 h after inoculation) attached in higher numbers (more than one log10 unit) and these were less easy to remove.  相似文献

Francisellosis, caused by the bacterium Francisella noatunensis, is one of the most severe diseases affecting farmed cod, and has caused great economic loss for the cod farming industry in Norway. We studied the fate of F. noatunensis in the marine environment, focusing on the role of blue mussels. In experimental challenges, waterborne F. noatunensis was rapidly filtered by the blue mussel and transported to the digestive diverticulae. The bacteria passed through the entire digestive system. Intraperitoneal injection of cod with suspensions prepared from faeces collected from challenged mussels resulted in the development of francisellosis in the recipients, demonstrating that some bacteria were alive and infective when shed in mussel faeces. Bacterial clearance from the mussels was relatively fast, and no evidence was found, suggesting that the bacterium is capable of persisting or multiplying in the mussel tissues. A cohabitation experiment with cod and mussels previously exposed to F. noatunensis did not lead to infection in cod. A direct transmission from contaminated mussels to cod was thus not demonstrated; however, faeces particles with infective bacteria may play a role in the transmission of the bacterium in marine food chains.  相似文献

目的分离培养普通卷甲虫肠道中的可培养细菌,筛选有产消化酶活性的细菌,推测其在协助普通卷甲虫消化食物中的作用。方法通过传统分离培养法分离普通卷甲虫肠道中的可培养细菌,利用平板透明圈法筛选产淀粉酶、蛋白酶、纤维素酶和脂肪酶活性的细菌,利用水解圈与菌落直径的比值,比较不同细菌的产消化酶活性。利用SPSS 20.0软件进行统计学分析,组间比较采用单因素方差分析。结果在普通卷甲虫肠道中分离出4个属9种细菌,其中气单胞菌属3种,假芽胞杆菌属和柠檬酸杆菌属各2种,芽胞杆菌属和假单胞菌属各1种。9种细菌中弗氏柠檬酸杆菌、豚鼠气单胞菌、南海假芽胞杆菌等3种细菌可产蛋白酶,嗜水气单胞菌、波特卡伦柠檬酸杆菌、水生气单胞菌、豚鼠气单胞菌和南海假芽胞杆菌等5种细菌可产纤维素酶,嗜水气单胞菌、波特卡伦柠檬酸杆菌、印度芽胞杆菌、水生气单胞菌、嗜盐假芽胞杆菌、豚鼠气单胞菌和南海假芽胞杆菌等7种细菌可产淀粉酶,未筛选到产脂肪酶细菌。统计学分析表明,3种产蛋白酶细菌和5种产纤维素酶细菌的产酶活性差异无统计学意义。而7种产淀粉酶细菌产酶的活力间差异有统计学意义,水生气单胞菌的产淀粉酶活性能力最强。结论普通卷甲虫肠道可培养细菌结构简单,但有消化酶活性的细菌种类多,5种细菌有产2种以上消化酶功能,说明肠道细菌可能在普通卷甲虫食物消化中起着重要作用。  相似文献

House flies are of major concern as vectors of food-borne pathogens to food crops. House flies are common pests on cattle feedlots and dairies, where they develop in and feed on animal waste. By contacting animal waste, house flies can acquire human pathogenic bacteria such as Escherichia coli and Salmonella spp., in addition to other bacteria, viruses, or parasites that may infect humans and animals. The subsequent dispersal of house flies from animal facilities to nearby agricultural fields containing food crops may lead to pre-harvest food contamination with these pathogens. We hypothesized that odors from honeydew, the sugary excreta produced by sucking insects feeding on crops, or molds and fungi growing on honeydew, may attract house flies, thereby increasing the risk of food crop contamination. House fly attraction to honeydew-contaminated plant material was evaluated using a laboratory bioassay. House flies were attracted to the following plant-pest-honeydew combinations: citrus mealybug on squash fruit, pea aphid on faba bean plants, whitefly on navel orange and grapefruit leaves, and combined citrus mealybug and cottony cushion scale on mandarin orange leaves. House flies were not attracted to field-collected samples of lerp psyllids on eucalyptus plants or aphids on crepe myrtle leaves. Fungi associated with field-collected honeydews were isolated and identified for further study as possible emitters of volatiles attractive to house flies. Two fungal species, Aureobasidium pullulans and Cladosporium cladosporioides, were repeatedly isolated from field-collected honeydew samples. Both fungal species were grown in potato dextrose enrichment broth and house fly attraction to volatiles from these fungal cultures was evaluated. House flies were attracted to odors from A. pullulans cultures but not to those of C. cladosporioides. Identification of specific honeydew odors that are attractive to house flies could be valuable for the development of improved house fly baits for management of this pest species.  相似文献

Wild-caught and colonized tampan ticks, Ornithodoros moubata (Murray), were fed on hepatitis B virus (HBV)-positive blood-means in a series of four experiments. Hepatitis B surface antigen (HBsAg) persisted in nymphal and adult ticks for up to 779 days, while the epsmark antigen (HBeAg) persisted in mature nymphs up to 13 days, in adult males up to 11 days and in adult females up to 16 days. HBsAg was transmitted trans-stadially through two moults during the life cycle but transovarial transmission did not occur. The surface antigen was transmitted by two out of fifteen single ticks into 0.4 ml aliquots of HBV-negative blood, although six groups of ticks failed to transmit into 5.5 ml aliquots of blood: this antigen was not transmitted to hamsters. HBsAg was detected in samples of the ticks' coxal and rectal fluid secretions always at the infecting feed and usually at the second feed. HBeAg was only detected in one of two samples of coxal fluid collected at the infecting feed. The results as a whole indicate that no biological multiplication of virus occurs in O.moubata but that mechanical transmission from ticks to man could occur by: (i) contamination of a person when crushing infected ticks; (ii) infection by bite; (iii) contamination with coxal fluid, especially by scratching bites. This is thought to take place among the Kavango tribe in their village huts in north-eastern Namibia where infestations of infected O.moubata occur.  相似文献

Production of exotoxins by Aeromonas spp. at 5 degrees C   总被引:1,自引:0,他引:1  
The ability of 60 strains of Aeromonas to produce enterotoxin and haemolysin after cultivation at 5 degrees C for 7-10 d was investigated. The strains were isolated from lamb meat, offal, carcasses and faeces, and had previously been tested for their ability to produce these exotoxins at 37 degrees C. The results showed that some strains of Aeromonas hydrophila and A. sobria were capable of producing enterotoxin and haemolysin at 5 degrees C, but none of the A. caviae strains tested produced these two factors. Of the 30 A. hydrophila strains investigated 25 and 27 were enterotoxigenic and haemolytic respectively. Likewise, of the 24 A. sobria strains investigated 16 and 18 were enterotoxigenic and haemolytic respectively. The results indicate that certain strains of Aeromonas species, in particular A. hydrophila and A. sobria, are of potential public health significance in meats stored at refrigeration temperature.  相似文献

The Western flower thrips (Frankliniella occidentalis), a polyphagous insect with global distribution, has a permanent association with a near Erwinia species TAC bacterium in its hindgut. Since this bacterium is able to grow outside the thrips, it is a facultative symbiont that is not completely dependent on the host. In this study we address the question of how the association is maintained and how bacteria are transmitted to newly hatched thrips larvae. Bacteria are passed on to new thrips via the food source. No evidence was found for vertical transmission from mother to offspring via the egg. Gut bacteria show unlimited growth during the larval (feeding) stages, and in the second instar stage 100% of the larvae become infected with high numbers of bacteria. In the prepupal and pupal stage, the number of bacteria declines, but increases again during the adult phase. A method to rear aposymbiotic (bacteria-free) thrips is described which enables studies on the impact of bacteria on the fitness of thrips.  相似文献

In many industrialized countries, the incidence of campylobacteriosis exceeds that of salmonellosis. Campylobacter bacteria are transmitted to humans mainly in food, especially poultry meat products. Total prevention of Campylobacter colonization in broiler flocks is the best way to reduce (or eliminate) the contamination of poultry products. The aim of this study was to establish the sources and routes of contamination of broilers at the farm level. Molecular typing methods (DNA macrorestriction pulsed-field gel electrophoresis and analysis of gene polymorphism by PCR-restriction fragment length polymorphism) were used to characterize isolates collected from seven broiler farms. The relative genomic diversity of Campylobacter coli and Campylobacter jejuni was determined. Analysis of the similarity among 116 defined genotypes was used to determine clusters within the two species. Furthermore, evidence of recombination suggested that there were genomic rearrangements within the Campylobacter populations. Recovery of related clusters from different broiler farms showed that some Campylobacter strains might be specifically adapted to poultry. Analysis of the Campylobacter cluster distribution on three broiler farms showed that soil in the area around the poultry house was a potential source of Campylobacter contamination. The broilers were infected by Campylobacter spp. between days 15 and 36 during rearing, and the type of contamination changed during the rearing period. A study of the effect of sanitary barriers showed that the chickens stayed Campylobacter spp. free until they had access to the open area. They were then rapidly colonized by the Campylobacter strains isolated from the soil.  相似文献

Electric fly killers (EFKs) are commonly used to control flying insects that enter food establishments. For establishment of the incidence of pathogen-bearing insects in food establishments, insect samples obtained from EFK trays could be used. The principal difficulty with this approach is that the survival time of microorganisms on or within insect corpses after electrocution is unknown. This study determined the survival of Serratia marcescens (as a representative of the enteric bacteria) within houseflies following their electrocution by a commercial EFK. S. marcescens was successfully ingested by houseflies and survived on and within the corpses after electrocution for up to 5 weeks. Maximal levels of bacteria were recovered 24 h postelectrocution. The study also demonstrates the ability of ingested S. marcescens to out-compete resident microbial flora within houseflies. The findings are intended to pave the way for further research to determine the incidence of pathogen-laden flying insects in food establishments. The ability of motile Aeromonas spp. to survive in drinking water (mineral and tap water) and in sea water was experimentally tested. Clinically isolated cytotoxic strains of A. hydrophila, A. caviae and A. sobria were selected for this study. After contamination of water samples, the survival of Aeromonas strains was studied for at least three months using viable counts. The results obtained show that the survival of the Aeromonas spp. varies considerably depending on species and water type. For all three species, the survival time was longest in mineral water, where viable bacteria of each strain were still detected after 100 d. Moreover, A hydrophila and A. caviae also re-grew on the first day. In tap water all strains showed marked survival, although to a lesser extent than in mineral water. Aeromonas cells showed a rapid decline in sea water (90% reduction in viable cells after about two d) and thus seem to be more sensitive to saline/marine stress than chlorination.  相似文献

Cat scratch disease is the most common zoonotic infection caused by Bartonella bacteria. Among the many mammals infected with Bartonella spp., cats represent a large reservoir for human infection, as they are the main reservoir for Bartonella henselae, Bartonella clarridgeiae and Bartonella koehlerae. Bartonella spp. are vector‐borne bacteria, and transmission of B. henselae by cat fleas occurs mainly through infected flea faeces, although new potential vectors (ticks and biting flies) have been identified. Dogs are also infected with various Bartonella species and share with humans many of the clinical signs induced by these infections. Although the role of dogs as source of human infection is not yet clearly established, they represent epidemiological sentinels for human exposure. Present knowledge on the aetiology, clinical features and epidemiological characteristics of bartonellosis is presented.  相似文献

The kinetics of anti-listeriolysin O (ALLO) production in the goats experimentally infected with pathogenic Listeria monocytogenes vis-à-vis isolation of the pathogen was studied. In the experimentally infected goats the pathogen was recovered during days 1–13, 2–14, and 4–5 post-inoculation (PI) in faecal, nasal and haemocultures, respectively. All the animals were culture negative on day 16 PI. Antibodies to LLO appeared by days 10–14 PI, peaked between days 11 and 25 days PI and persisted by end of study (day 80 PI) at a detectable level. Isolation of the pathogen was also attempted from meat and blood of 113 slaughtered goats besides testing of serum samples for ALLO by LLO-based indirect ELISA. L. monocytogenes was isolated from 7.08 and 1.77% of meat (113) and blood (113) samples of goat, respectively. Antibodies against LLO were detected in 21.68% serum samples collected from the slaughtered goats.  相似文献

In 2008, the centennial of the discovery of Toxoplasma gondii was celebrated. However, toxoplasmosis is still seen as a neglected and underreported disease, despite having a disease burden similar to that of salmonellosis and campylobacteriosis. Human vaccines are not available and current antiparasitic treatment is disappointing. This has led to an urge to focus more on prevention. Food, soil or water contaminated with oocysts from cat faeces and undercooked meat from infected intermediate hosts are important routes of infection. Oocyst contamination is difficult to control, whereas in Western countries, the control of T. gondii in meat should be feasible. Here, we discuss strategies aimed at developing a Toxoplasma-safe meat chain.  相似文献

Tests in Britain on samples of basic and commercial Brassica oleracea seed between 1976 and 1978 showed that many lots were infected with Alternaria brassicicola. A. brassicae was uncommon in basic seed in these years and in commercial seed harvested in 1976 and 1977 but was frequent in seed harvested in 1978. Most affected seeds were contaminated by surface-borne spores and mycelium of A. brassicicola but many were internally infected by the fungus situated within the seed-coat and in some seeds in the embryo tissues. Superficial contamination by the fungus declined rapidly after 2 yr in cabbage seeds stored at 10 °C, 50% r.h. but internal infection persisted for up to 12 yr. In some samples, internal infection was commonly associated with small shrivelled seeds. Surface contaminated and internally infected seeds transmitted the disease but seedling infection was more closely correlated with the latter.  相似文献

The regular use of antimicrobials in livestock production selects for antimicrobial resistance. The potential impact of this practice on human health needs to be studied in more detail, including the role of the environment for the persistence and transmission of antimicrobial-resistant bacteria. During an investigation of a pig farm and its surroundings in Brandenburg, Germany, we detected abundant cephalosporin- and fluoroquinolone-resistant Escherichia coli in pig faeces, sedimented dust, and house flies (Musca domestica). Genome sequencing of E. coli isolates revealed large phylogenetic diversity and plasmid-borne extended-spectrum beta lactamase (ESBL) genes CTX-M-1 in multiple strains. [Correction added on 28 February 2023, after first online publication: In the preceding sentence, ‘and TEM-1’ was previously included but has been deleted in this version.] Close genomic relationships indicated frequent transmission of antimicrobial-resistant E. coli between pigs from different herds and across buildings of the farm and suggested dust and flies as vectors for dissemination of faecal pathogens. Strikingly, we repeatedly recovered E. coli from flies collected up to 2 km away from the source, whose genome sequences were identical or closely related to those from pig faeces isolates, indicating the fly-associated transport of diverse ESBL-producing E. coli from the pig farm into urban habitation areas. The observed proximity of contaminated flies to human households poses a risk of transmission of antimicrobial-resistant enteric pathogens from livestock to man.  相似文献
