There has been much research studying the effect of homotypic embryonic neural tissue transplantation in the central nervous system of rats and cats, with promising results. The benefits of transplantation are dependent upon the appropriate aged donor. Therefore, it is critical that the exact age of the embryonic tissue be known. For this reason we have developed a method of determining, indirectly, the pregnancy status and embryonic age in the domestic cat (Felis catus) using serum progesterone levels. Serum progesterone levels were monitored in 16 domestic cats during 26 breeding trials. Blood samples were taken prior to and 6 days after natural breeding to determine if ovulation had occurred, indirectly indicating pregnancy status. By knowing the exact date bred and if it resulted in pregnancy, an accurate embryonic age was calculated. A day 6 post-breeding serum progesterone concentration of less than 5.0 ng/ml was considered a negative indication of pregnancy, greater than 5.0 ng/ml a positive indication of pregnancy. Pregnancy was confirmed by abdominal palpation 21 to 26 days after breeding. Serum progesterone levels taken on the sixth day after observed breeding provide an accurate, indirect evaluation of pregnancy in the timed pregnant domestic cat (81%, p = 0.003).  相似文献

We studied copulation solicitation display (CSD) responses to playback in photostimulated female canaries given systemic injections of progesterone. Eight females received injections of 0.1 mg of progesterone dissolved in olive oil during their first breeding cycle and were untreated during their second breeding cycle; eight females received only the oil vehicle during their first breeding cycle and received no treatment during their second breeding cycle. The injections were performed every second day during 15 days, after the onset of nest building. Progesterone treatment resulted in a significant increase of plasma progesterone which in turn provoked an inhibition of females' CSDs and decreased the size of the clutch. During the first breeding cycle, progesterone-treated females had lower CSDs and egg-laying scores than did control females. During the second breeding cycle, when females received no treatment, no differences emerged between the two groups. The suppressive effect of progesterone on female sexual responses was observed as soon as 48 h after the beginning of the treatment. We propose that progesterone plays a key role in mediating the transition from active female courtship behavior to sexual refractoriness in this species. Suppressive effects of progesterone on female sexual behavior have been previously described in lizards as well as in rodents. Our data are consistent with the hypothesis of Godwin et al. (J. Godwin, V. Hartman, M. Grammer, and D. Crews, Horm. Behav. 30, 138-144, 1996) which proposed that the decrease in sexual behavior following plasma progesterone increase represents an evolutionarily conserved mechanism in the regulation of female sexual behavior.  相似文献

The objective of this study was to determine cow factors that influence conception rates after timed artificial insemination (TAI) on a commercial dairy farm. In Trial 1, 197 cows were synchronized by an administration of 25 mg of dinoprost between Days 48 and 54 post partum and again 14 days later. Cows were inseminated 66 and 90 hours after the second treatment. In Trial 2, 186 cows were treated with an Ovsynch protocol consisting of an administration of 0.02 mg of buserelin between Days 62 and 68 post partum, a treatment with 0.75 mg of tiaprost 7 days later and a second treatment with buserelin 48 h later. Cows were bred 16 to 20 hours after the last treatment. Cows with abnormal vaginal discharge at the time of insemination were excluded from AI. Lactation number, milk yield, fat and protein content of milk, signs of endometritis at an examination 14 to 20 days post partum and month of breeding were included as potential factors influencing conception on TAI. Conception rates after timed breeding were 32.0% and 30.6% in Trials 1 and 2, respectively. Logistic regression revealed that neither milk production parameters nor endometritis at post partum examination influenced conception rates in either of the two timed breeding protocols. Only parity showed an effect (P=0.012) in Trial 2. Primiparous cows were more likely to conceive after timed breeding than older cows (43.5 vs. 23.1%). An effect of parity, however, was not observed in Trial 1. It can be concluded that neither milk production nor endometritis at an examination 14 to 20 days post partum influence conception rates after TAI if cows with abnormal vulval discharge at the designated time of AI are excluded from breeding.  相似文献

Reproductive management of the southern tamandua (Tamandua tetradactyla) should include timed introductions for breeding to minimize aggression and pregnancy monitoring. Since serial blood sampling could cause unnecessary stress, and urinary progesterone metabolites are found in very low concentrations, this study sought to validate progesterone and estradiol enzyme immunoassays for measuring fecal progesterone metabolite (FPM) and fecal estrogen metabolite (FEM) concentrations in two females. Peaks in FEM concentrations coincided with breeding and conception, were 5–6 times higher than baseline concentrations, and were followed by clear luteal phases distinguished by FPM concentrations 5–6 times higher than baseline concentrations. FPM concentrations during the first 30–53 days of gestation overlapped with luteal phase concentrations, thereafter increasing to 8–25 times higher than baseline concentrations. FEM concentrations during the first 41–44 days of gestation remained near basal values for one female, whereas concentrations were 1.8 times higher than baseline for the second. FEM concentrations became elevated for the former by 44 days of gestation and increased further for the latter after 53 days, ultimately averaging four times higher than baseline for both females. The biphasic increase in FPM and FEM concentrations, follicular and luteal phase durations (follicular: 7 ± 1 days, luteal: 25 ± 1 days), total cycle length (41 ± 1 days), and gestation (161–165 days) documented in this study were consistent with previous reports from serum and urine analyses. Monitoring FPM and FEM is a reliable noninvasive method for tracking reproductive cycles and pregnancy in southern tamandua that overcomes the challenges associated with serum or urinary hormone analysis.  相似文献

This study provides endocrine data in relation to behavioral events during the transition of the non-breeding into the breeding season in American bison (Bison bison). Fecal progesterone metabolite patterns (20-oxo-P) were obtained in 13 adult female American bison and hormonal data were correlated with behavioral observations; i.e. copulation, male tending, female tail-up behavior and gestation length. Based on fecal progesterone metabolite patterns, the breeding season started between the middle of July and early August. Predictable short cycles reflected the transition from non-breeding to the breeding season; the luteal phase of these cycles was 4.10+/-0.86 days. Copulations and female tail-up behavior were reliably associated with the hormonally detected ovulation. Male tending behavior was more loosely associated with hormonally detected ovulation. The observed hormonal pattern in the study females indicated that 9 of 10 pregnant cows conceived during the second ovulatory period in the breeding season. One other cow conceived during her third ovulatory period, and one cow did not conceive until later in the breeding season by beginning of October. Gestation duration was on average 266.30+/-1.00 days. In summary, this study confirmed that the bison is a seasonally polyestrous species; the transition from the non-breeding into the breeding season was characterized by short cycles with low progesterone metabolite values.  相似文献

The aim of the study was to show whether it is possible to induce ovulation in the hare by GnRH analogue administration and to carry out an effective artificial insemination (AI). The research was carried out during the breeding and non-breeding season. During the breeding season, plasma progesterone concentrations increased on the 4th day after intramuscular injection of GnRH analogue (buserelin), indicating induced ovulation and corpus luteum development. Prostaglandin F(2)alpha (dinoprost) was an effective luteolytic agent on day 9. During the non-breeding season, the GnRH analogue injection does not cause an increase of progesterone. The 17beta-estradiol concentrations during the breeding and non-breeding season were similar. It was shown that after GnRH analogue administration and artificial insemination with semen diluted in Tris buffer extender 80% females delivered live young (39-43 days after artificial insemination), which proves the effectiveness of inducing ovulation in the hare by means of hormonal stimulation.  相似文献

Knowledge of the basic reproductive physiology of snow leopards is required urgently in order to develop a suitable management conditions under captivity. In this study, the long-term monitoring of concentrations of three steroid hormones in fecal matter of three female snow leopards was performed using enzyme immunoassays: (1) estradiol-17β, (2) progesterone and (3) cortisol metabolite. Two of the female animals were housed with a male during the winter breeding season, and copulated around the day the estradiol-17β metabolite peaked subsequently becoming pregnant. The other female was treated in two different ways: (1) first housed with a male in all year round and then (2) in the winter season only. She did not mate with him on the first occasion, but did so latter around when estradiol-17β metabolite peaked, and became pseudopregnant. During pregnancy, progesterone metabolite concentrations increased for 92 or 94 days, with this period being approximately twice as long as in the pseudopregnant case (31, 42, 49 and 53 days). The levels of cortisol metabolite in the pseudopregnant female (1.35 μg/g) were significantly higher than in the pregnant females (0.33 and 0.24 μg/g) (P<0.05). Similarly, during the breeding season, the levels of estradiol-17β metabolite in the pseudopregnant female (2.18 μg/g) were significantly higher than those in the pregnant females (0.81 and 0.85 μg/g) (P<0.05). Unlike cortisol the average levels of estradiol-17β during the breeding season were independent of reproductive success.The hormone levels may also be related to housing conditions and the resulting reproductive success in female leopards. The female housed with a male during the non-breeding season had high levels of cortisol metabolites and low levels of estradiol-17β in the breeding season, and failed to become pregnant. This indicates that housing conditions in snow leopards may be an important factor for normal endocrine secretion and resulting breeding success.  相似文献

Prostaglandin or prostaglandin analogues have been shown to be luteolytic in the pregnant goat. In this study the temporal changes in the plasma concentrations of progesterone during pregnancy and after administration of PGF2a to pregnant goats are described. PGF2a administration to pregnant goats at 30 and 65 days after breeding induced abortion within 34 to 75 hours. These abortions were accompanied by estrus and profuse muco-hemorrhagic discharges. When PGF2a was administered to pregnant goats 140 or 142 days after breeding, premature parturition occurred within 42 to 76 hours. Live kids were delivered in all cases. The plasma levels of progesterone in all pregnant goats showed dramatic decreases within 24 hours after the prostaglandin injections and continued to decrease gradually until abortions or premature parturition. Thereafter, the progesterone levels remained low for several days.  相似文献

The objectives were to determine the effects of one or three timed artificial insemination (AI) before natural service (NS) in lactating dairy cows not observed for detection of estrus on hazard of pregnancy, days nonpregnant, and 21-days cycle pregnancy rate. A total of 1050 lactating Holstein cows were subjected to a double Ovsynch program for their first postpartum AI. On the day of first AI (78 ± 3 days in milk), cows were blocked by parity and randomly assigned to receive either one timed AI (1TAI, n = 533) or three timed AI (3TAI, n = 517) before being exposed to NS. Cows assigned to 1TAI were exposed to bulls 7 days after the first AI. Nonpregnant cows in 3TAI were resynchronized with the Ovsynch protocol supplemented with progesterone twice, with intervals between AI of 42 days, before being exposed to NS 7 days after the third AI. Cows were evaluated for pregnancy 32 days after each timed AI, or every 28 days after being exposed to NS. Pregnant cows were re-examined for pregnancy 28 days later (i.e., 60-day gestation). Exposure to heat stress was categorized based on the first AI being performed during the hot or cool season, according to the temperature-humidity index. Body condition was scored at first AI. All cows were allowed a period of 231 days of breeding, after which nonpregnant cows were censored. Pregnancy to the first AI did not differ between 1TAI and 3TAI on Day 60 after insemination (30.8 vs. 33.5%). Cows receiving 3TAI had a 15% greater hazard of pregnancy and a 17% greater 21-days cycle pregnancy rate than 1TAI and these benefits originated from the first 84 days of breeding. These changes in rate of pregnancy reduced the median and mean days nonpregnant by 9 and 10 d, respectively. Despite the long inter-AI interval in cows subjected to 3TAI, reproductive performance was improved compared with a single timed AI and subsequent exposure to NS. In dairy herds that use a combination of AI and NS, allowing cows additional opportunities to AI before onset of breeding with bulls is expected to improve reproductive performance.  相似文献

Menstrual patterns and progesterone levels were monitored for 5 years from a cohort of 28 female vervet monkeys that were individually caged indoors. Three distinct cycle types (short, normal, and prolonged) were defined according to cycle length. Mean length of the normal cycle (32.5 days) and menses duration (4.8 days) are in agreement with previous reports. Prolonged cycles (> 50 days) contributed 20% of the total, with a decreased incidence during the natural peak breeding period. Weekly progesterone measurements indicated that many prolonged cycles were associated with an extended luteal phase, while others were probably due to lack of ovulation. From these data it would appear that the vervet monkey, although not strongly seasonal, does favor a particular time of year for breeding in a colony housed indoors.  相似文献

To examine the role of opioid neurons on luteinizing hormone (LH) secretion in the female rabbit, we determined LH release at timed intervals after naloxone administration to rabbits aged 25-150 days. The LH response to naloxone (10 mg/kg) was not significantly elevated until day 43 when LH rose 76-113% above basal levels at 40-80 min. In 56-day-old females the corresponding increase was 160% at 15 min and in 65- to 67-day-olds it was 154%. From 70 to 80 days of age the LH response was blunted and no significant elevations could be elicited. By contrast, naloxone-induced LH increases were again evident when rabbits were older than 100 days. At all ages no significant change in FSH concentrations was observed. In the adult females, naloxone at 2.5, 5, and 10 mg/kg caused increases in LH secretion which occasionally were high enough to induce ovulation as exemplified by elevated serum progesterone 4 days later. These data suggest that opioid peptides may be involved in the prepubertal rise in LH and in the normal inhibition of adult secretion in the female rabbit.  相似文献

The Vancouver Island marmot (Marmota vancouverensis; VIM) is one of North America's most endangered species with fewer than 150 individuals remaining in the wild. A captive breeding program was established across four facilities in Canada as an insurance population and source of animals for reintroduction to the wild. The purpose of this study was to gather information about the basic reproductive biology and behavior of this species, which is essential to improve captive breeding programs. Regular fecal samples were obtained from adult female (n = 14) and male (n = 10) marmots, 2 years of age and older, over 1-3 breeding seasons (2-3 months duration posthibernation) for steroid hormone analysis. Enzyme immunoassays were validated for quantifying fecal testosterone metabolite concentrations for males, and fecal estrogen and progesterone metabolite concentrations for females. Results indicated that fecal progesterone metabolite concentrations can be used to monitor ovulation and pregnancy. Behavioral monitoring through infrared video surveillance was conducted in four breeding pairs over a 2-year period (n = 7 behavioral profiles). Breeding behaviors correlated strongly with changes in reproductive endocrine profiles. A high frequency of play behavior or "wrestling" was observed in conjunction with breeding activity before an elevation in progesterone metabolite concentrations. Impending parturition was associated with increased aggression and exclusion of the male from the maternal nestbox as well as an increase in nesting activity. Observational data combined with hormonal analysis suggest that female VIMs are induced ovulators and that multiple breeding attempts may be required for ovulation and conception. Gestation appears to be approximately 34 days from peak breeding activity (32 days from estimated ovulation). Fecal testosterone concentrations suggest that testicular activity is seasonal with the reproductive activity occurring immediately posthibernation. Monitoring breeding behavior is a useful means of indicating estrus, conception and pregnancy, which can also be supported by the hormonal analysis of daily fecal samples of individual animals.  相似文献

There is limited information available on the oestrous cycle of female southern hairy-nosed wombats (Lasiorhinus latifrons). This is mainly due to an extremely poor breeding success in captivity and the difficulty in routine recapturing of these cryptic, semi-fossorial animals in the wild. The aim of this study was to characterise the oestrous cycle of this species by monitoring peripheral plasma concentrations of progesterone and oestradiol, assessing changes in vaginal cytology, pouch condition and the urogenital sinus. Eight adult female wombats were monitored during the breeding season (July-December) over 2 years (2002-2003). Samples were collected up to three times a week. Vaginal smears contained several cell types, categorised by morphology, as either superficial epithelial cells or parabasal-intermediate cells. Leucocytes were also counted. Plasma progesterone profiles showed a mean oestrous cycle length of 36.33+/-0.67 days with a peak progesterone concentration of 139.53+/-10.62nmol/L. Levels of oestradiol peaked at a mean level of 467.33+/-44.32pmol/L on average 5 days before a rise in plasma progesterone values. The proportion of epithelial cells in vaginal smears varied throughout the cycle, with a high percentage of superficial epithelial cells observed during the follicular phase. During periods when progesterone concentrations were high, a greater percentage of parabasal-intermediate cells was observed. In conclusion, this study has characterised the oestrous cycle of the southern hairy-nosed wombat and confirmed that changes in vaginal smears together with pouch and urogenital sinus details could be used to determine signs of oestrus in this species.  相似文献

Plasma progesterone concentrations were recorded during one breeding season in 19 lesser mouse lemur females living in different social conditions. The oestrous cycle length and the progesterone profile mainly depended on the social environment of the female. For totally isolated females, the oestrous cycle lasted 38 +/- 5.7 days and included a 25-30-days spontaneous luteal phase with a progesterone peak about 100 ng/ml between the 20th and 25th days after oestrus, and a prolonged preovulatory period of 10-15 days which could be considered equivalent to the follicular phase of a menstrual cycle. When females were able to communicate through olfactory, visual and auditory signals, the oestrous cycle was significantly lengthened (53.7 +/- 5.9 days). When females had tactile contacts, the oestrous cycle was further lengthened (62.7 +/- 0.8 days). This lengthening of the oestrous cycle was related to an extension of the luteal phase associated with a decrease in progesterone concentrations during this period. In females maintained with one male (paired) or with males and females (heterosexually grouped), large individual variations were shown in cycle lengths or in progesterone concentrations. In these females, cycle lengths and progesterone concentrations were inversely correlated to plasma cortisol concentrations.  相似文献

Immature female mink, 8 weeks of age in July, were treated with implants releasing melatonin. Mating, which induced ovulation, took place during the normal breeding season in the following March. Circulating prolactin and progesterone concentrations did not undergo the expected gestational increases, and no embryos implanted. A similar absence of gestational changes in prolactin and progesterone values ensued in primiparous mink treated with the melatonin implant 2-3 days after the second of 2 matings. Administration of exogenous sheep prolactin (0.5 mg/day) by minipump induced precocious elevation of progesterone concentrations in mated mink. Prolactin administration overcame the effects of melatonin, in that the corpora lutea were activated and embryos implanted, but exogenous prolactin resulted in degeneration of implanted embryos both in the presence and absence of chronic melatonin. The results suggest that melatonin has a single effect in alteration of gestation in mink; i.e. the prevention of prolactin secretion. Hyperprolactinaemia may inhibit embryo development in this species.  相似文献

Vaginal cytology was used to monitor ovarian cycles, two pregnancies, and three pseudopregnancies. Vaginal smears were collected two or three times per week from three adult females; smears plus blood samples were collected once per week from a fourth, adolescent female. Mean cycle lengths, based on intervals between onset of leukocyte infusions, were 11.9 ± 4.9 days (n = 43 cycles), 10.8 ± 5.1 days (n = 49), and 12.3 ± 6.3 days (n = 7) for the three females. Weekly hormone data from the adolescent female revealed a correlation between serum estradiol and percent anuclear cells, suggesting that these cells may be indicative of estrus. The fourth female experienced two sustained, 6-week increases in serum progesterone, one spontaneous and the other following follicle-stimulating hormone (FSH) administration. Leukocyte infusions continued during these periods of increased progesterone secretion. Progesterone, oestradiol-17 beta and LH were measured in plasma from 6 non-pregnant, captive, female muskoxen during the 1984 and 1985 breeding seasons. Jugular blood samples were taken on an alternating 3/4-day schedule in 1984 and daily or at 4-h intervals over oestrus, via indwelling jugular cannulae, for 6 weeks in 1985. Oestrous cycle length was 19.6 +/- 0.96 (s.d.) days (n = 19) and did not vary between the first and subsequent cycles of the season. Progesterone was lowest at oestrus (less than or equal to 0.1 ng/ml), began to rise on Days 4-5, peaked on Days 10-12 (mean = 2.6 ng/ml) and returned to baseline 2-5 days before the next oestrus. A small rise in progesterone before the first cycle of the breeding season was observed on 7 of 12 occasions. Oestradiol-17 beta was significantly higher (P less than 0.001) 1-4 days before, or coincident with, oestrus. The average duration of the LH peak was 24.6 h (n = 7) and coincided with observations of behavioural oestrus. In one animal behavioural oestrus and an LH peak preceded a small progesterone rise at the beginning of the breeding season. The temporal relationship of these three hormones during the muskox oestrous cycle is very similar to that seen in domestic ruminants.  相似文献

One experiment was conducted to determine whether the treatment with artificial long days and exogenous melatonin can induce reproductive activity during spring (seasonal anoestrus) in Mediterranean goats that are in daily contact with bucks and whether this treatment causes a variation in the reactivation of the reproductive activity in the normal breeding season. The experiment started on 4 November 2005 and finished on 27 October 2006. Thirty-four adult and barren does were used, distributed into two groups balanced according to their live weight (LW) and body condition score (BCS). Seventeen females were exposed to long days (16 h of light/day) from 14 November 2005 to 20 February 2006. On 20 February, they received one s.c. melatonin implant (LD-M group) and were exposed to natural photoperiodic changes in an open shed. The other females during the experiment were placed in an open shed under natural photoperiod and remained as the control group (C group). The C and LD-M groups were keeping in contact with males during the whole experiment. During the experiment, the LW, BCS and plasma progesterone concentrations were measured weekly, oestrous activity was tested daily using entire aproned bucks, and ovulation rate was evaluated by laparoscopy 7 days after positive identification of the oestrus. A clear treatment-time interaction was observed for plasma progesterone concentrations (P < 0.001), with a period of high progesterone concentrations during the natural seasonal anoestrus in the LD-M group. Although 94.1% of females in the LD-M group presented ovarian activity during this period, no female in group C did. Resumption of ovarian activity in the subsequent natural breeding season was 2 weeks later in the LD-M group in comparison with group C (P < 0.05). We can conclude that in Mediterranean goat breeding systems, when females are in daily contact with bucks, the treatment with 3 months of long days and melatonin implant at the end of the light photoperiodic treatment can induce ovarian and oestrous activity during the seasonal anoestrus. Finally, this treatment causes a short delay in the subsequent reactivation of ovarian activity in the natural breeding season.  相似文献

Although numbers of giant anteaters within North American facilities have been steadily increasing for the last 15 years, the population now exhibits an unstable age distribution with genetically valuable individuals nearing reproductive senescence. Contributing to this issue is the Association of Zoos and Aquariums (AZA) described lack of standardization of breeding introduction practices and high risk of female injury occurring during such pairings. This report describes the development of a successful breeding protocol at Cleveland Metroparks Zoo based on hormone monitoring and efficient communication between science and animal management teams that minimizes risk of female injury. By training a female giant anteater for urine sample and body weight data collection, staff members accurately predicted estrus, and timed breeding introductions to facilitate positive interactions between the male and female. Such training also allowed for careful monitoring of two pregnancies through parturition (169–184 days from breeding) and post‐partum return to estrus (114–129 days from parturition). Urinary hormone monitoring revealed a sharp progestogen increase averaging >five‐fold over basal levels (0.52 ± 0.05 ng/mg creatinine) which was sustained throughout the second half of pregnancy. Mean regular estrous cycle length (n = 14 cycles), was calculated as 46.17 ± 1.39 days, measured as days between estrogen peaks of mean concentration 2.27 ± 0.19 ng/mg creatinine. This report summarizes impressive collaborative efforts among multiple zoological departments to achieve extensive hormonal and body weight monitoring from a female giant anteater, adding valuable information on reproductive parameters, and specifics for novel hormone assay techniques.  相似文献

A study was undertaken to identify urinary estrogen and progesterone metabolites in the female Indian rhinoceros (Rhinoceros unicornis). Measurements of these metabolites were then used to monitor ovarian function and establish normal levels and patterns of steroid excretion during the estrous cycle and pregnancy. Urine samples were analyzed for estrone sulfate and pregnanediol-3-glucuronide (PDG) by direct radioimmunoassays. Both hormones produced discrete profiles reflecting ovarian activity in nonconceptive cycles. The estrous cycle was observed to be 48 days (range 39–64) with a mean follicular phase of 14.8 days (range 13–19), followed by a mean luteal phase of 19 days (range 17–21). Of the single gestation monitored, PDG levels rose above luteal phase levels by the third month after breeding and remained elevated throughout gestation. The combined estrogen and progesterone metabolite profiles present a complete evaluation of ovarian steriod production in the mature female Indian rhinoceros.  相似文献
