不同活力玉米种子胚萌发期间热激蛋白的合成   总被引:8,自引:0,他引:8  
玉米(Zea mays L.)种子在萌发期间热激处理(42℃)时蛋白质合成率低于对照(25℃);高活力种子胚热激蛋白的合成率最高。在42℃热激处理时玉米种胚子合成的热激蛋白的分子量分别为73、65、62、54、18kD等5种。高活力种胚合成的热激蛋白在最上高于低活力种子,高活力种胚合成的特异性热激蛋白56kD可以作为衡量种子活力的指标。双向电泳表明高低活力种子间热激蛋白的合成有更多质上的差异。  相似文献

随着花生种子萌发率和活力指数的下降,胚轴DNA开始合成的时间推迟,其合成水平也降低。但DNA合成都是先于吸胀的12h(高活力胚)或18h(低活力胚)出现一个峰,然后再持续上升。腐胺预处理明显地促进老化胚轴萌发早期(12~2dh)的DNA合成。钙离子预处理则有一定抑制作用,但两种预处理均能提高种子的活力指数,并能促进吸胀30h以后的DNA合成。  相似文献

利用PvLEA-18蛋白抗体,分析小麦种子在萌发过程中类PvLEA-18表达情况。Western blot表明在小麦干种胚中未检测到类PvLEA-18蛋白,而在种子萌发不同时期(12h,24h,36h)的胚和盾片组织中检测到类PvLEA-18蛋白的表达。其分子量分别约为81kD和70kD。种子萌发24h后,81kD蛋白消失,70kD的类PvLEA-18表达量也降低。  相似文献

为比较不同浓度盐胁迫下小麦种子萌发前期胚蛋白表达情况,以"晋麦-47"为试验材料。选取经0、0.1和0.2mol/L处理过的种子,采用2-DE技术对3个浓度下胚蛋白含量变化进行研究。通过PDQuest图像分析软件分析盐胁迫下蛋白含量的变化,检测到正常的种子胚有121个点,0.1 mol/L NaCl胁迫下有32个蛋白点丰度下调,21个蛋白点丰度上调;在0.2 mol/LNaCl胁迫下,检测到46个蛋白质点丰度下调,16个蛋白点的丰度上调。研究结果表明,盐胁迫对小麦种子萌发时期胚中部分蛋白含量和表达产生不同程度的抑制与激活。  相似文献

西洋参(Panax Quinquefolium L.)种胚需经形态后熟和生理后熟后才能萌发.经过激素(GA100ppm+KT50ppm+BA20ppm)处理的西洋参种子,在后熟过程中鲜重增加,β—淀粉酶活性增强,过氧化物酶活性保持较高水平.在生理后熟期,可溶性蛋白量逐渐增多.激素处理加速了胚体内物质的分解和合成,促使胚迅速发育和分化,缩短了种胚完成形态和生理后熟的时间,使种子提早一年萌发.  相似文献

休眠、层积和萌发的梣叶槭种子中抑制物质的含量不同。休眠的种子中抑制物质的含量最多,层积后的种子抑制物质活性明显减弱,萌发的种子中依然存有少量的抑制物质。抑制物质在种子内的分布也不同。子叶内较多,胚中较少。种子萌发时,胚内的抑制物质可能向子叶转移。  相似文献

将不同胚龄的白及种子接种到不同培养基中进行培养,对白及种子萌发率、萌发时间、丛生芽增殖、生根等方面进行了研究。结果表明:白及种子萌发率与其胚龄及有胚率成正相关,萌发时间则与胚龄及有胚率成负相关;胚龄16周的种子在1 g/L花宝1号 2 g/L花宝2号的培养基上萌发率可达84%;胚龄等于或大于20周的种子萌发率不受培养基成分的影响,均可达100%,萌发时间只需7 d;丛生芽增殖的最佳培养基为1/2 MS 4.0 mg/L6-BA 0.2 mg/L NAA 100 g/L CM,其增殖倍数达4.41倍;诱导生根较好的培养基为1/2 MS 0.2 mg/L NAA,生根率达90%。  相似文献

东北刺人参(Oplopanax elatus Nakai)种子透水性良好,休眠后萌发不受其影响。种皮和胚乳的水提取物中存在萌发抑制物质,胚乳中提取物对白菜种子萌发的抑制效果比种皮更明显。种子自然脱落时胚尚未分化完全,处于心形胚阶段。种子需要先温暖层积以完成胚的分化与生长,然后转入低温层积完成生理后熟。同批种子胚的发育不完全同步,变温层积处理7个月有极少数种子萌发,连续变温层积处理17个月大部分种子萌发。不同年份受气候条件影响,种子产量和发芽率差异较大。种子耐贮性较强,贮藏2年的种子生活力变化不大,仍具有较高的萌发潜力。  相似文献

一氧化氮供体硝普钠(Sodium nitroprusside,SNP)能明显地促进渗透胁迫下小麦(Triticum aestivum L.)种子萌发、胚根和胚芽伸长,提高萌发过程中淀粉酶和内肽酶的活力,加速贮藏物质的降解;胁迫解除后,仍能使种子维持较高的活力.此外,SNP还能显著诱导渗透胁迫下CAT、APX活力的上升和脯氨酸含量积累,抑制LOX活力,从而提高渗透胁迫下小麦种子萌发过程中抗氧化能力.进一步研究还发现,SNP诱导切胚半粒小麦种子萌发早期(6h)的淀粉酶活力上升可能与GA3无直接关系.  相似文献

Aerated hydration (AH) treatments of cauliflower seeds for 12 h (12AH) and 28 h (28AH) at 20 degrees C resulted in improved or reduced storage potential of low or high vigour seeds, respectively. Seeds were stored at their initial seed moisture content (mean 5.5% mc) or at 12% mc at 10 degrees C for 12 months and at 20 degrees C for 4 months. The improved longevity of low vigour seeds was associated with increased K(i) (initial seed viability) and a reduced rate of deterioration (1/sigma) whereas the K(i) of high vigour seeds fell after 28AH and the rate of deterioration increased such that the time to lose one probit of viability decreased from 28.7 to 5.3 months at 10 degrees C and from 10.4 to 1.2 months at 20 degrees C. The improved K(i) of low vigour seeds could be explained by the reduction in the extent of deterioration after AH, as indicated by the increase in germination after cotrolled deterioration (CD), and the possible activation of metabolic repair during treatment. In contrast the reduced germination after CD of AH-treated high vigour seeds was indicative of deterioration as a result of treatment. Both high and low vigour seeds contained constitutive levels of ss-tubulin which increased during AH treatment, the increase being greater in high vigour seeds. High vigour seeds also showed an increase in the proportion of nuclear DNA present as 4C DNA, from 3% (untreated seeds) to 26% (28AH), indicative of germination advancement from the G(1) to G(2) phase of the cell cycle during treatment. This higher proportion of 4C DNA is correlated with the increased sensitivity of seeds to drying and/or storage after AH, leading to their reduced K(i) and storage potential. In contrast, there was little change in %4C in low vigour seeds. Priming in polyethylene glycol (PEG, -1.0 MPa) for 5 d or 13 d also improved the longevity of low vigour seeds stored at their initial and 12% mc at 10 degrees C for 8 months, as reflected in their laboratory and CD germination. In this case, however, the improved longevity of the low vigour seeds following 13 d priming was associated with an increase in 4C DNA from 4% (dry control) to 56% after treatment. The germination of both untreated and primed high vigour seeds remained high throughout the storage period. Increases in the rate of germination (decreased mean germination time) observed after all AH and PEG treatments were not consistently associated with an increase in the proportion of nuclei containing 4C DNA.  相似文献

Five muskmelon (Cucumis melo L.) cultivar seedlots from a commercialsource and freshly produced seeds of two cultivars, when artificiallyaged, were found to differ in their viability and vigour asdetermined by germination tests. Furthermore, the various commercialseedlots without ageing also exhibit a range of deteriorationlevels. Low vigour seeds had higher respiratory quotient valuesthan the high vigour seeds as a result of a higher level ofCO2 production. This high level of CO2 evolution in low vigourseeds may have been due to anaerobic respiration. Levels of acetaldehyde and ethanol produced by imbibing seedswere negatively associated with seed viability and vigour. After6 h of imbibition low vigour seeds produced significantly moreethanol and acetaldehyde than high vigour seeds. After 24 hof imbibition, ethanol continued to accumulate in the commercialseedlots up to 10-fold the amount produced after 6 h of imbibition,whereas, acetaldehyde levels increased less. However, in thefreshly produced, artificially aged seeds (except the most extremeageing), ethanol levels were reduced and no acetaldehyde productioncould be detected, indicating re-utilization of ethanol. It is suggested that ethanol production in the first hours ofimbibition can be used as a reliable index to predict germinationin muskmelon seedlots. Exposing muskmelon (Cucumis melo L.) cultivar seedlots to N2atmosphere created totally anaerobic conditions which stimulatedethanol production and accumulation in both high and low vigourseeds. However, accumulation of ethanol was consistently higherin the low vigour seeds than in the high vigour ones. In addition,CO2 production under N2 and in air suggests the presence ofan apparent ‘Pasteur effect’ in the low vigour seedsbut not in the high vigour seeds. Acetaldehyde production underN2 was very low and did not seem to be associated with seedvigour, probably because of its nature as an intermediate inethanol production. The fast shift toward ethanol may be dueto the fact that alcohol dehydrogenase, the enzyme which catalyzesthe conversion of acetaldehyde to ethanol, exists in sufficientamounts in the imbibing seeds so that it is not a limiting factorin the conversion to ethanol. Ethylene Production in Pea and Cocklebur Seeds of Differing Vigour   总被引:1,自引:0,他引:1  
Relationships between seed vigour and ethylene (C2H4) productionwere studied using C2H4-responsive fatty cocklebur seeds (Xanthiumpennsyhanicum Wallr.) and C2H4-insensitive starchy pea seeds(Pisum sativum L. cv. Alaska), which had been harvested in differentyears and subjected to different storage conditions. In bothspecies, the seeds with the highest vigour evolved the largestamounts of C2H4 during a period of water imbibition. The reductionof C2H4 production in cocklebur seeds occurred concomitantlywith the reduction in the growth potentials of both axial andcotyledonary tissues. Similarly, the activity of ACC-C2H4 conversionincreased with soaking, and was greater in seeds of high vigourcompared with those of low vigour. However, the change in ACCcontent in pea seeds differed from that in cocklebur seeds.That is, pea seeds with high vigour accumulated less ACC duringan imbibition period than those with low vigour. From theseresults it was suggested that the inferior C2H4 production bylow vigour pea seeds is mainly attributable to low ACC-C2H4conversion, whereas that by low vigour cocklebur seeds is dueto the shortage of ACC supply in addition to the reduced ACC-C2H4conversion. However, germination of deteriorated cocklebur seedswas not restored by exposure to ACC or C2H4, suggesting thatthe loss of seed vigour reduces the responsiveness of seedsto C2H4 as well as C2H4 production. 以采自安徽(AH)、山东(SD)及江苏(JS)的6个青檀(PteroceltistatarinowiMaxim.)种源种子为材料,测定各种源种子营养成分含量及种子活力。6个种源种子的蛋白质、可溶性糖、淀粉及粗脂肪含量存在较大差异。在种子品质上,6个种源的种子千粒重、浸泡液电导率及简化种子活力指数(SVIS)也存在显著差异。种子中各营养成分含量的高低与SVIS的大小密切相关,复相关系数达08223,以蛋白质含量对SVIS的影响最大,淀粉和粗脂肪含量影响次之,可溶性糖含量影响最小。采用模糊聚类分析法,以种子千粒重、电导率及SVIS为评价指标,可将6个种源的种子品质划分为三大类:第一类包括SD1、AH1和JS13个种源,种子品质较好;第二类仅为AH3种源,品质中等;第三类包括AH2和SD22个种源,品质较差。  相似文献

在低温吸胀阶段,经PVA(聚乙烯醇)和PEG(聚乙二醇6000)预处理的大豆胚轴蛋白质合成和ATP含量均比对照高。在萌发阶段,胚轴生长增快,蛋白质合成明显加快,ATP迅速被消耗,而对照胚轴则相反。试验结果表明,预处理大豆种子萌发和生长与其蛋白质合成、ATP水平和消耗能力有密切关系。  相似文献

Experiments were conducted to study the influence of sowing seasons and drying methods on the seed vigour of two spring soybean (Glycine max (L.) Merr.) cultivars. Two cultivars, ‘Huachun18’ and ‘Huachun 14’, were sown in three seasons viz., spring, summer and autumn and the harvested seeds were dried using three different methods. The results showed that soybean sown in spring had a higher number of branches per plant, pods per branch and seed weight, and consequently resulted in higher seed yields than that of soybean sown in autumn or summer seasons. Seeds sown in the autumn season had the lowest values of electrical conductivity during seed imbibitions, higher peroxidase (POD) activity in germinated seedlings and lower contamination by the seed-borne fungi on the MS medium, which indirectly improved the seed vigour, which was followed by summer sown seeds. Seeds sown during the spring season resulted in poor seed vigour. In addition, the effect of drying methods on the seed vigour was also clarified. Seeds that hung for four days before threshing and then air-dried had the poorest seed vigour which was determined by germination, electrical conductivity, POD activity and seed borne fungal growth. There was no difference in seed vigour between other methods, i.e. seeds threshed directly at harvest and then air-dried on a bamboo sifter or concrete floor. These results indicated that autumn sowing soybean and the drying method in which seeds were threshed directly at harvest and then air-dried on a bamboo sifter resulted in higher seed vigour.  相似文献

Two seed-coat forms (the presence or absence of papillae) andtwo hairiness forms (medium or densely hairy) of Spergula arvensisare differently distributed through the British Isles, theirrelative frequencies being correlated with latitude and altitude.The proportions of the non-papillate and densely hairy formsincrease with increasing distance from Kent to the north-north-westand also with increasing altitude. A possible explanation is that the diverse forms indicate selectivedifferences. Experiments have been carried out to test thishypothesis and the evidence supports it. The non-papillate formproduces a lower proportion of fertile capsules than the papillateform when grown at a high temperature and low humidity. Alsothe non-papillate seeds germinate more readily than the papillateseeds at low temperatures, whereas at higher temperatures thereverse is true. It is suggested that hybrid vigour, fluctuating selection intensities,and migration may all play a part in maintaining the observeddistribution of the various forms.  相似文献

Putrescine, spermidine and spermine of high vigour, low vigour and non-viable (classes 1, 2 and 3 respectively) seeds of Oryza sativa increased with loss of viability. The largest concentration of spermine was found in non-viable embryos. Spermine was absent in the husks of all the three categories of seeds. Arginine decarboxylase was greatest in high vigoured seeds and its activity gradually declined with loss of viability. However, diamine oxidase and polyamine oxidase activities gradually increased with the loss of viability of the seeds while DNA, RNA and protein contents decreased. The total content of polyamines increased on kinetin treatment but declined on ABA treatment. DNA, RNA and protein followed the same trend as polyamines. The polyamine contents increased by ca 3- and 4-fold, respectively, in high vigoured and low vigoured seeds on 10?4 M kinetin treatment. The activity of ADC followed the same change as that of the polyamines in both cases, but the reverse was observed for the activities of diamine and polyamine oxidases.  相似文献

Both low and high vigour seed lots were more than 90% viablewhen germinated at 25 °C. The tissue weights and the contentsof mineral ions and glycinebetaine in the ungerminated embryoswere the same for each lot. The release of K+ and phosphateions from isolated embryos into aqueous media were also thesame. These similarities remained for at least 36 h of germination.At 48 h the coleoptiles of the low vigour embryos were about20% shorter and their fresh weight was about 30% less than thoseof the high vigour embryos. These differences were reduced againto about 10% at 72 h. In contrast to these differences, H+ efflux and 36Rb+ uptakeby isolated embryos were up to 50% slower in low vigour embryoscompared with the high vigour ones. The difference was maximalat 24 h of germination and appeared to depend upon the anionpresented and the pH of the incubation medium. The H+/36Rb+exchange ratio was not, however, changed in low vigour embryoscompared with high vigour ones.  相似文献
