We describe a methodology to investigate the potential of given microalgae species for biodiesel production by characterizing their productivity in terms of both biomass and lipids. A multi-step approach was used: determination of biological needs for macronutrients (nitrate, phosphate and sulphate), determination of maximum biomass productivity (the “light-limited” regime), scaling-up of biomass production in photobioreactors, including a theoretical framework to predict corresponding productivities, and investigation of how nitrate starvation protocol affects cell biochemical composition and triggers triacylglycerol (TAG) accumulation. The methodology was applied to two freshwater strains, Chlorella vulgaris and Neochloris oleoabundans, and one seawater diatom strain, Cylindrotheca closterium. The highest total lipid content was achieved with N. oleoabundans (25-37% of DW), while the highest TAG content was found in C. vulgaris (11-14% of DW). These two species showed similar TAG productivities.  相似文献

Abstract The effects of artificial and solar UV-B radiation on the gravitactic (formerly called geotactic) orientation of the freshwater dinoflagellate Peridinium gatunense were measured under artificial UV-B radiation and in a temperature-controlled growth chamber under solar radiation in Portugal. Circular histograms of gravitaxis show the impairement of orientation after UV irradiation. The degree of orientation, quantified using the Rayleigh test and top quadrant summation, decreased as the exposure time to the radiation prolonged. The effects of artifical UV-B radiation on orientation are stronger than those of solar radiation, probably because the radiation source emits higher fluence rates below 300 nm than found in solar radiation. After UV radiation, the gravitactic orientation under artificially increased acceleration at 2 g was drastically affected.  相似文献

Exposure to artificial light at night (ALAN) has been reported to be associated with various pathological changes including sleep deprivation, circadian rhythm disruption, and melatonin suppression with increase in various cancers such as breast or prostate cancers. In this study, we sought to elucidate the association between ALAN and prostate cancer in 27 districts within Gwangju City and urban and rural areas from South Jeolla Province in South Korea. We analyzed the correlation between ALAN and the incidence of a range of cancers by Poisson regression analysis, after adjustment for confounding risk factors, such as smoking, drinking, obesity, stress, air pollution (particulate matter <10 μm in diameter), urbanization (proportion of urbanized area), and the cancer screening rate. Interestingly, the incidence of prostate cancer was significantly associated with ALAN (risk ratio = 1.02, p = 0.0369) and urbanization (risk ratio = 1.06, p = 0.0055). In particular, comparing the prostate cancer incidence at 25% and 75% level of ALAN, the risk ratio was 1.726 (12.6 over 7.3, respectively). No significant association was observed between ALAN and other cancers, including stomach, esophageal, liver, pancreatic, laryngeal, lung and tracheal, bladder, and brain and central nervous system cancers, as well as lymphoma and multiple myeloma. In conclusion, this study shows that a high incidence of prostate cancer may be independently associated with light pollution and urbanization, which represent significant factors in the rapid process of industrialization of South Korea.  相似文献

CO2 sequestration by cyanobacteria and green algae are receiving increased attention in alleviating the impact of increasing CO2 in the atmosphere. They, in addition to CO2 capture, can produce renewable energy carriers such as carbon free energy hydrogen, bioethanol, biodiesel and other valuable biomolecules. Biological fixation of CO2 are greatly affected by the characteristics of the microbial strains, their tolerance to temperature and the CO2 present in the flue gas including SOX, NOX. However, there are additional factors like the availability of light, pH, O2 removal, suitable design of the photobioreactor, culture density and the proper agitation of the reactor that will affect significantly the CO2 sequestration process. Present paper deals with the photobioreactors of different geometry available for biomass production. It also focuses on the hybrid types of reactors (integrating two reactors) which can be used for overcoming the bottlenecks of a single photobioreactor.  相似文献

Nannochloropsis sp. was grown in a Modular FlatPanel Photobioreactor (MFPP) consisting of sixalveolar panels each with 20.5 L culture volume and3.4 m2 illuminated surface area. The panelsformed a closely-packed unit with illuminationprovided by banks of fluorescent tubes placed betweenthe panels. The whole unit was contained in athermoregulated cabinet. Continuous illumination ofone side of the panels with 115 molphoton m-2 s-1 attained a mean volumetricproductivity of 0.61 g (d. wt) L-1 24 h-1,increasing to 0.97 g (d. wt) L-1 24 h-1 whenthe same irradiance was provided on both sides of thepanels. With 230 mol photon m-2 s-1 onone side of the panel, a mean productivity of 0.85 g(d. wt) L-1 24 h-1 was achieved, whichreached 1.45 g (d. wt) L-1 24 h-1 when bothsides were illuminated. Increasing the amount of lightprovided to the culture (either by increasingirradiance or the illuminated surface area) decreasedpigment and enhanced the total fatty acid content, butdid not change significantly the content ofeicosapentaenoic acid. A MFPP of the presentdimensions could produce sufficient microalgae tosupport a hatchery producing 6 million sea breamfingerlings annually.  相似文献

Obesity is a common disorder with many complications. Although chronodisruption plays a role in obesity, few epidemiological studies have investigated the association between artificial light at night (ALAN) and obesity. Since sleep health is related to both obesity and ALAN, we investigated the association between outdoor ALAN and obesity after adjusting for sleep health. We also investigated the association between outdoor ALAN and sleep health. This cross-sectional survey included 8526 adults, 39–70 years of age, who participated in the Korean Genome and Epidemiology Study. Outdoor ALAN data were obtained from satellite images provided by the US Defense Meteorological Satellite Program. We obtained individual data regarding outdoor ALAN; body mass index; depression; and sleep health including sleep duration, mid-sleep time, and insomnia; and other demographic data including age, sex, educational level, type of residential building, monthly household income, alcohol consumption, smoking status and consumption of caffeine or alcohol before sleep. A logistic regression model was used to investigate the association between outdoor ALAN and obesity. The prevalence of obesity differed significantly according to sex (women 47% versus men 39%, p < 0.001) and outdoor ALAN (high 55% versus low 40%, p < 0.001). Univariate logistic regression analysis revealed a significant association between high outdoor ALAN and obesity (odds ratio [OR] 1.24, 95% confidence interval [CI] 1.14–1.35, p < 0.001). Furthermore, multivariate logistic regression analyses showed that high outdoor ALAN was significantly associated with obesity after adjusting for age and sex (OR 1.25, 95% CI 1.14–1.37, p < 0.001) and even after controlling for various other confounding factors including age, sex, educational level, type of residential building, monthly household income, alcohol consumption, smoking, consumption of caffeine or alcohol before sleep, delayed sleep pattern, short sleep duration and habitual snoring (OR 1.20, 95% CI 1.06–1.36, p = 0.003). The findings of our study provide epidemiological evidence that outdoor ALAN is significantly related to obesity.  相似文献

Ecological light pollution (ELP) is quickly becoming a worldwide concern and can negatively affect aquatic ecosystems. The given intensity and spectrum of a light source can influence how organisms function within their environment. These properties of artificial lighting at night (ALAN) and their impacts on the physiology and behaviour of crayfish were examined in this work. Hemolymph was obtained from crayfish to quantify a physiological response. Behavioural data were measured as the number, duration, and maximum intensity of agonistic fights. Exposure to higher intensities of light and the presence of ultraviolet light induced a behavioural trend, resulting in significantly altered social interactions within both species of crayfish. The number and maximum intensity of lights significantly decreased, whereas the duration of time spent fighting significantly increased. Due to the importance of freshwater environments and the role crayfish play as a keystone species, examining how crayfish are impacted from ALAN is imperative to maintaining the health of aquatic ecosystems.  相似文献

Bean leaves grown under periodic illumination (56 cycles of 2 min light and 98 min darkness) were subsequently exposed to continuous illumination, and in connection with granum formation and accumulation of the light-harvesting pigment-protein complex thermoluminescence and light-induced shrinkage of thylakoid membranes were studied. Juvenile chloroplasts with large double sheets of thylakoids obtained under periodic light exhibited low temperature spectra of polarized fluorescence yielding fluorescence polarization (FP) values < 1 at 695 nm, characteristic for pheophytin emission. In the course of maturation under continuous light when normal grana appeared and the chlorophyll a/b light-harvesting photosystem II complex was incorporated into the membrane, at 695 nm the relative intensity of fluorescence dropped and FP changed to a value of > 1, suggesting an overlap between the emission of pheophytin and that of the chlorophyll a/b light-harvesting photosystem II complex. Thermoluminescence glow curves recorded with juvenile thylakoids displayed a relatively high proportion of emission at low temperatures (around -10°C) while with mature chloroplasts, more thermoluminescence originated from energetically deeper traps (discharged around 28°C). This means that during thylakoid development the capacity of the membrane to stabilize the separated charges increases, which might be favourable for the ultimate conservation of energy. An in‐depth investigation of how various illumination conditions influence microalgal growth in photobioreactors (PBR) has been presented. Effects of both the light emission spectrum (white and red) and the light incident angle (0° and 60°) on the PBR surface were investigated. The experiments were conducted in two fully controlled lab‐scale PBRs, a torus PBR and a thin flat‐panel PBR for high cell density culture. The results obtained in the torus PBR were used to build the kinetic growth model of Chlorella vulgaris taken as a model species. The PBR model was then applied to the thin flat‐panel PBR, which was run with various illumination conditions. Its detailed representation of local rate of photon absorption under various conditions (spectral calculation of light attenuation, incident angle influence) enabled the model to take into account all the tested conditions with no further adjustment. This allowed a detailed investigation of the coupling between radiation field and photosynthetic growth. Leaves were obtained from 4-week-old seedlings of Lavandula latifolia Medicus grown in vitro. Leaf explants were then cultured on MS medium supplemented with different concentrations and combinations of the auxins IAA or NAA with the cytokinin BA and maintained under three illumination conditions, 16h photoperiod, darkness or darkness followed by a photoperiod, to assess morphogenic responses. Irrespective of illumination conditions, bud regeneration was achieved only in media containing BA or BA/auxin combinations, with the best results being obtained in the presence of BA and 0.06 or 0.6 M IAA or NAA. A photoperiod of 16h appeared to yield the best response in terms of bud regeneration percentage. High auxin concentrations (6.0 or 11.0 M) inhibited bud differentiation, especially when explants were cultured in darkness. On the other hand, low auxin levels and photoperiod improved shoot development. Excised shoots were induced to form roots by transfer to hormone-free MS medium with macronutrients at half strength. Design and analysis of a solar reactor for anaerobic wastewater treatment   总被引:1,自引:0,他引:1  
The aim of this research was to design a solar heated reactor system to enhance the anaerobic treatment of wastewater or biological sludge at temperatures higher than the ambient air temperature. For the proposed reactor system, the solar energy absorbed by flat plate collectors was transferred to a heat storage tank, which continuously supplied an anaerobic-filter reactor with water at a maximum temperature of 35 degrees C. The packed reactor was a metallic cylindrical tank with a peripheral twin-wall enclosure. Inside this enclosure was circulated warm water from the heat storage tank. Furthermore, a mathematical model was developed for the prediction of the temperature distribution within the reactor under steady state conditions. Preliminary results based on model simulations performed with meteorological data from various geographical regions of the world suggested that the proposed solar reactor system could be a promising and environmentally friendly approach for anaerobic treatment of wastewater and biological sludge.  相似文献

An experimental growth system was devised to study the ecophysiological responses of submerged macrophytes to temperature and light. Using the system, a pilot study was conducted to compare responses to light of three representative species common to the littoral zone of Lake Biwa: Vallisneria asiatica Miki var. biwaensis Miki, which is endemic to Japan, Potamogeton maackianus A. Benn., a dominant submerged macrophyte in Lake Biwa, and Elodea nuttallii (Planch.) St. John, one of the most influential submerged exotic macrophytes naturalized in Japan. Different relative growth rate responses and other growth parameters among the species were observed. We ascertained the experimental system to be useful for the comparison of ecophysiological responses of submerged macrophytes.  相似文献

The effects of UV-B radiation generated in the laboratory and as a component of sunlight on the viability and particular biochemical activities of the bacterium Staphylococcus aureus have been examined. UV-B radiation progressively inhibits protein synthesis (assayed as 3H-alanine incorporation) and kills cells. Cell respiration, and RNA and DNA synthesis (3H-uridine and 3H-thymidine incorporation) were not greatly affected by UV-B irradiation. The OH and 1O2-free radical scavengers protected cells against killing and inhibition of protein synthesis by UV-B, suggesting that such radicals mediate the effects of UV-B on this organism. Artificial media are used in physiological studies of microalgae to maintain consistent conditions from one experiment to another and these media must be adapted to the needs of the organism studied. The artificial medium, in this case named diatom artificial medium (DAM), was designed to maintain long-term cultures of Haslea ostrearia and 19 other planktonic microalgae, and to allow physiological studies related to metal metabolism. The biomass and biochemical composition of H. ostrearia grown in the DAM and in a modified Provasoli medium were compared to assess the suitability of this new artificial medium for the culture of this diatom. The DAM provided sufficient nutrients to allow H. ostrearia to grow as efficiently as in the enriched seawater medium, without negative impact on metabolism. The DAM was tested with 19 other microalgae in order to widen its potential use, and 18 of the 19 showed a good adaptation to this medium. The chemical speciation of metals (Cd, Cu, Pb, Zn) was assessed using a speciation mathematical model. The presence of EDTA resulted in the total complexation of the trace metals implying that they were present in a sole chemical species in the DAM.  相似文献

Long-term studies of the effects of low-frequency magnetic fields on cells in culture require an incubator that is free of contaminating magnetic fields and that provides automatic control of exposure duration, uniform applied fields, a uniform and well-controlled environmental temperature, and high reliability of the total system. We describe a dual-incubator system in which the enclosed solenoid of either unit can provide a sham exposure while the other generates a magnetic induction of up to 500 microT. Each solenoid-incubator unit contains an enclosed rack that can hold as many as 140 petri dishes for culturing cells; each unit is heated radiantly by circulating warm water. Field-exposure conditions and temperatures are continuously monitored and controlled by a microcomputer.  相似文献

Cultivation of microalgae in closed photobioreactors suffers from high temperatures, whereas in open raceway ponds the optimal temperature is seldom reached. With an integrated device coupling a raceway pond with a suitably sized and positioned closed system (alveolar panel), the heat accumulated in the panel was efficiently transferred to the pond and a near-optimum temperature regimen for Arthrospira platensis was maintained, with no need for any additional cooling device. The productivity obtained in the integrated system was higher than the sum of the productivities (g reactor−1 day−1) of the pond and panel systems operating separately. The structure and function of chloroplast in plant leaves can be affected by acid rain and air pollution. The photosystem II in a plant is considered the primary site where light-induced delayed light emission (DLE) is produced. With the lamina of zijinghua (Bauhinia variegata L.) and soybean (Glycine max (L.) Merr.) as testing models, we studied the effects of artificial acid rain and SO2 on characteristics of DLE by using a home-made weak luminescence detection system. The results show that the changes in DLE intensity of green plants can reflect the changes in chloroplast intactness and function. With proper calibration, DLE may provide an alternative means of evaluating environmental acid stress on plants. The changes in DLE intensity may provide a new approach for the detection of environmental pollution and its impact on the ecosystem.  相似文献

We have succeeded in culturing an axenic biofilm of the green sulfur bacterium Prosthecochloris aestuarii strain CE 2404 in an artificial sandy sediment under visible light (400–700 nm). This simulates the conditions of deep submerged sediments. A five-week incubation period, using a 16-hour light / 8-hour dark regime, was applied in the benthic gradient chamber (BGC). The biofilm was located below the oxygen penetration depth of 1.2 mm, namely between 1.5 and 2.5 mm and the biomass peak was at 2.1 mm depth. This is much shallower compared to previously described artificial mats of P. aestuarii, which were grown in the BGC under near infrared (NIR)-rich light. High resolution time courses of photosynthesis were measured as sulfide photo-oxidation rates and studied under visible light and visible light amended with NIR to assess the effect of light quality. Sulfide photo-oxidation rates were rather low under visible light and strongly stimulated at most depths under full light conditions. However, under the latter conditions the rates decelerated after a maximum rate was reached at 8–10 min, apparently due to diffusional limitation of sulfide supply. It was concluded that the top of the mat was not limited by the photon flux density, while the biomass peak and the bottom of the biofilm were severely light limited under the culture conditions. These results support the hypothesis that a biofilm of P. aestuarii can develop in deep submerged sediments, when the oxygen penetration depth is very shallow. The mechanisms underpinning the ecological impacts of the presence of artificial night lighting remain elusive. One suspected underlying cause is that the presence of light at night (LAN) supresses nocturnal production of melatonin, a key driver of biological rhythm and a potent antioxidant with a proposed role in immune function. Here, we briefly review the evidence for melatonin as the link between LAN and changes in behaviour and physiology. We then present preliminary data supporting the potential for melatonin to act as a recovery agent mitigating the negative effects of LAN in an invertebrate. Adult crickets (Teleogryllus commodus), exposed to constant illumination, were provided with dietary melatonin (concentrations: 0, 10 or 100 µg ml−1) in their drinking water. We then compared survival, lifetime fecundity and, over a 4-week period, immune function (haemocyte concentration, lysozyme-like and phenoloxidase (PO) activity). Melatonin supplementation was able only partially to mitigate the detrimental effects of LAN: it did not improve survival or fecundity or PO activity, but it had a largely dose-dependent positive effect on haemocyte concentration and lysozyme-like activity. We discuss the implications of these relationships, as well as the usefulness of invertebrates as model species for future studies that explore the effects of LAN.  相似文献
