Fruit ethylene production genotypes for Md-ACS1 and Md-ACO1 were determined for 60 apple cultivars and 35 advanced breeding
selections. Two alleles for each gene are commonly found in cultivated apple. Earlier studies showed that genotypes homozygous
for the ACS1-2 allele produce less ethylene and have firmer fruit than ACS1-1/2 and ACS1-1/1 genotypes. ACO1 plays a minor
role compared to ACS1, with homozygous ACO1-1 having lower ethylene production. In this study, ACS1-2 and ACO1-1 homozygotes
had firmer fruit at harvest and after 60 days of 0–1°C cold storage compared to other genotypes. These genotypes, ACS1-2/2
and ACO1-1/1, were observed for the following 8 of 95 cultivars/selections: “Delblush”, “Fuji”, “Pacific Beauty”, “Sabina”
and four breeding selections. Cultivars/selections that were homozygous ACS1-2 but not ACO1-1 were: “Ambrosia”, “Aurora Golden
Gala”, “CrimsonCrisp”, “Gala”, “GoldRush”, “Huaguan”, “Pacific Rose, “Pacific Queen”, “Pinova”, “Sansa”, “Sonja”, “Sundance”,
“Zestar”, and 17 breeding selections. Cultivars with the heterozygous ACS1-1/2 genotype were “Arlet”, “Braeburn”, “Cameo”,
“Delicious”, “Delorgue”, “Empire”, “Enterprise”, “Ginger Gold”, “Golden Delicious”, “Granny Smith”, “Honeycrisp”, “Orin”,
“Pink Lady”, “Silken”, “Suncrisp”, “Sundowner”, “Sunrise” and 11 breeding selections. No cultivars were detected homozygous
for both ACS1-1 and ACO1-1, or for both ACS1-2 and ACO1-2. This study is the first large-scale allelic genotyping of both
ethylene synthesis genes for a comprehensive set of apple breeding parents used in an ongoing breeding project. The data reported
here are important for informative selection of parent combinations and marker-assisted selection of progeny for breeding
low ethylene-producing apple cultivars for better storability and improved consumer acceptance. 相似文献

Earliness is a crucial factor of tomato field production in Poland. A. Michalska and B. Kubicki (1978) conducted the first
investigations on tomato earliness at the Department of Plant Genetics and Breeding. The main conclusions from these studies
indicated that it was possible to obtain varieties earlier than ‘New Yorker’ as a result of transgression. The two new varieties
(‘Alfa’ and ‘Akord F1’) proved to be two weeks earlier than ‘New Yorker’. However, earliness was correlated with smaller fruit weight (30–40 g).
At the next stage of experiment some improvement was achieved both in the size of fruit (45 g for ‘Beta’ up to 70 g for ‘Alka’)
as well as colour intensity and good flavour. With the next few years fruit weight was still increased (up to 100 g for ‘Maskarena’)
and a higher level of firmness was obtained (mid firm fruits for ‘Maskarena’ and ‘Delta’). At present, the Polish Research
Centre for Cultivar Testing recommends our varieties ‘Betalux’, ‘Promyk’ and ‘Delta’ as standards. 相似文献

The effects of six basal media on in vitro shoot proliferation of the greek grapevines Vitis vinifera L. cv. ‘Malagouzia’ and ‘Xinomavro’ were investigated. Galzy and Zlenco proved to be the most effective for ‘Malagouzia’
and ‘Xinomavro’, respectively. If only BA was present in the medium, shoot development was poor and the plantlets were chlorotic.
When the medium was supplemented with BA and NAA, growth was enhanced. The best ratio (in μM) of growth regulators was 0.5/0.3
for ‘Malagouzia’, and 0.1/0.03 for ‘Xinomavro’, which resulted in the highest number of microshoots per explant and greatest
proliferation rate. The development of ‘Malagouzia’ and ‘Xinomavro’ explants at 21±2 and 26±2°C was also investigated, revealing
the higher temperature to be more effective. Regarding rooting, 0.5 μM IBA improved root formation at 26°C for ‘Malagouzia’
and 0.5 μM IBA at 21°C for ‘Xinomavro’. Moreover, 0.5 μM IBA resulted in a higher rooting percentage (>95%) and proved to
be more beneficial for the overall morphological appearance of the plantlets of ‘Malagouzia’. After acclimatization, survival
of microshoots cultivated in media with IBA was higher than those in NAA. 相似文献

This study examined the thermoregulatory responses of men (group M) and women (group F) to uncompensable heat stress. In
total, 13 M [mean (SD) age 31.8 (4.7) years, mass 82.7 (12.5) kg, height␣1.79␣(0.06) m, surface area to mass ratio 2.46␣(0.18) m2 · kg−1 · 10−2, Dubois surface area 2.01 (0.16) m2, %body fatness 14.6 (3.9)%, V˙O2peak 49.0 (4.8) ml · kg−1 · min−1] and 17 F [23.2 (4.2) years, 62.4 (7.7) kg, 1.65 (0.07) m, 2.71 (0.14) m2 · kg−1 · 10−2, 1.68 (0.13) m2, 20.2 (4.8)%, 43.2 (6.6) ml · kg−1 · min−1, respectively] performed light intermittent exercise (repeated intervals of 15 min of walking at 4.0 km · h−1 followed by 15 min of seated rest) in the heat (40°C, 30% relative humidity) while wearing nuclear, biological, and chemical
protective clothing (0.29 m2 ·°C · W−1 or 1.88 clo, Woodcock vapour permeability coefficient 0.33 i
m). Group F consisted of eight non-users and nine users of oral contraceptives tested during the early follicular phase of
their menstrual cycle. Heart rates were higher for F throughout the session reaching 166.7 (15.9) beats · min−1 at 105 min (n = 13) compared with 145.1 (14.4) beats · min−1 for M. Sweat rates and evaporation rates from the clothing were lower and average skin temperature () was higher for F. The increase in rectal temperature (T
re) was significantly faster for the F, increasing 1.52 (0.29)°C after 105 min compared with an increase of 1.37 (0.29)°C for
M. Tolerance times were significantly longer for M [142.9 (24.5) min] than for F [119.3 (17.3) min]. Partitional calorimetric
estimates of heat storage (S) revealed that although the rate of S was similar between genders [42.1 (6.6) and 46.1 (9.7) W · m−2 for F and M, respectively], S expressed per unit of total mass was significantly lower for F [7.76 (1.44) kJ · kg−1] compared with M [9.45 (1.26) kJ · kg−1]. When subjects were matched for body fatness (n = 8 F and 8 M), tolerance times [124.5 (14.7) and 140.3 (27.4) min for F and M, respectively] and S [8.67 (1.44) and 9.39 (1.05) kJ · kg−1 for F and M, respectively] were not different between the genders. It was concluded that females are at a thermoregulatory
disadvantage compared with males when wearing protective clothing and exercising in a hot environment. This disadvantage can
be attributed to the lower specific heat of adipose versus non-adipose tissue and a higher percentage body fatness.

Mating disruption treatments for the tufted apple bud moth (TABM),Platynota idaeusalis (Walker), were tested in small plot trials in apple orchards in Pennsylvania. Treatments were evaluated by fruit injury and
by capture of male TABM in traps baited with synthetic pheromone sources or virgin females. The TABM pheromone is a two component
isomeric blend ofE-11-tetradecen-1-ol (E11-14:OH) andE-11-tetradecenyl acetate (E11-14:Ac). A 50∶50 ratio of these two components was used in standard monitoring septa and in mating
disruption treatments released from either hollow fibers (‘fiber’) or PVC tubes (‘PVC’). Other pheromone blends tested included
a 90∶10 ratio of E11-14: Ac and E11-14:OH (‘EAc’) and its reverse (‘EOH’), mixture ‘EAc’ with 30% of the Z-isomers (‘low AEc’),
and a blend similar to the preceding with 2% Z9-12:Ac (‘generic’). These other blends were released from multi tube tape (‘tape’)
or Shin-Etsu type rope (‘rope’) dispensers. Seasonal dispenser release rate in mg ha−1 h−1 was ca. 30 for the ‘rope’ dispensers, 14 for ‘PVC’ and 6 for ‘fiber’. ‘EAc-tape’ and ‘EOH-tape’ were equally effective in
reducing catches of males in traps baited with synthetic lures and in traps baited with virgin females. Both treatments also
reduced fruit injury. ‘EAc-rope’, ‘fiber’ and ‘PVC’ also were generally effective; whereas, the ‘low EAc’ and ‘generic’ treatments
reduced trap capture less than 90% and did not reduce fruit injury. Dispenser density was positively correlated with reduction
in trap capture for the ‘low EAc-rope’ and ‘genericrope’ treatments. Traps loaded with ‘fiber’ dispensers captured more male
TABM than the other treatments in non-pheromone permeated environments. Trap capture of other tortricids was reduced in pheromone
treatments. ‘EAc-rope’ and the ‘TABM’ treatments provided mean (s.e.) percent reduction in trap catch of 99.5 (0.4) and 42.9
(10.1), respectively, for the redbanded leafroller,Argyrotaenia velutinana (Walker), and 90.4 (6.8) and 90.4 (1.3), respectively, for the obliquebanded leafroller,Choristoneura rosaceana (Harris). 相似文献

Potential uptake and clearance rates of fluorescent microspheres (FM) from 0.25 to 4.05 μm diameter were determined for the
non-loricate ciliate Pseudocohnilembus sp. from Antarctic sea ice. The percentage of ciliate cells that ingested FM after 20 min incubation decreased with increasing
particle diameter. Pseudocohnilembus sp. ingested FM between 0.25 and 4.05 μm in diameter. We offered FM at concentrations less than natural concentrations for
plankton plus detrital material and obtained clearance rates less than those previously reported for bactivorous ciliates.
Clearance rates were 3.6–5.4 nl cell−1 h−1 for FM 0.5 and 1 μm diameter, respectively, but decreased to 1.1 nl cell−1 h−1 for 1.97 μm diameter and 1.4 nl cell−1 h−1 for 4.05-μm-diameter FM. Clearance and uptake rates of FM 0.5 and 1 μm diameter indicate that Pseudocohnilembus sp. principally grazes on bacteria-sized particles. However, it can also ingest organisms as large as nanoplankton and may
graze particles as small as femtoplankton and colloids. This suggests a feeding strategy that may suit the temporal and spatial
changes in food availability in the sea-ice habitat.

Concentrations of selected metals were determined using ICP-MS in canned bonito, sardines and mackerel commercialized in Turkey.
Thirty samples and two different brands were sampled for each fish species. The minimum and maximum concentrations of trace
metals in canned bonito, sardines and mackerel were found as 0.000–34.742, 0.000–89.015, 0.000–28.725 mg/kg for iron, 2.388–26.620,
10.930–41.340, 4.778–29.270 mg/kg for zinc, 0.331–1.548, 0.599–2.242, 0.336–2.884 mg/kg for copper, 0.000–0.065, 0.000–0.113,
0.000–0.115 mg/kg for cadmium, 0.000–0.190, 0.000–0.158, 0.000–0.385 mg/kg for tin, 0.000–0.111, 0.000–0.223, 0.000–0.208 mg/kg
for mercury and 0.000–3.046, 0.000–2.875, 0.000–3.529 mg/kg for lead, respectively. These levels are similar those found in
other studies. Although the samples have concentrations within permissible limits for Zn, Cu, Sn and Hg, some of them contained
Fe, Cd and Pb above these limits. Periodical controls of metals in canned fish are essential both to protect human health

Low temperature at the booting stage of rice causes male sterility resulting in severe yield loss. Cold tolerance has long
been an important objective in rice breeding. We identified a quantitative trait locus (QTL) for cold tolerance on the long
arm of chromosome 3 from the cold-tolerant breeding line ‘Ukei 840’ by using F2 and BC1F2 populations from crosses between ‘Ukei 840’ and ‘Hitomebore’. The cold tolerance of ‘Ukei 840’ is derived from the Chinese
cultivar ‘Lijiangxintuanheigu’. The effect of this QTL on cold tolerance was confirmed by developing ‘Hitomebore’ chromosome
segment substitution lines having ‘Lijiangxintuanheigu’ alleles on chromosome 3. By producing recombinants in chromosome 3,
the QTL region for cold tolerance was delimited to the region of about 1.2-Mb region between RM3719 and RM7000. All lines
heterozygous for the QTL showed seed fertilities as low as that of ‘Hitomebore’, suggesting that the ‘Lijiangxintuanheigu’
allele for cold tolerance in the QTL region is recessive. Determination of a 1.2-Mb nucleotide sequence of ‘Ukei 840’ and
comparison with the published genomic sequence of ‘Nipponbare’ showed 254 SNPs, of which 11 were in coding regions of genes,
seven in five genes being non-synonymous. SNPs were detected in the 5-kb upstream regions of 89 genes, but no differences
of gene expression levels were detected between alleles of these genes. Although further delimitation is required to identify
the gene responsible for cold tolerance of ‘Lijiangxintuanheigu’, SNP markers developed here will be useful for marker-assisted
selection in a breeding program using ‘Lijiangxintuanheigu’ as a donor of cold tolerance. 相似文献

The river types in Wallonia (Belgium) were defined according to the system B of the European Water Framework Directive (WFD)
taking into account obligatory and optional factors synthesized in three criteria: ‘size’, ‘slope’ and ‘natural region’. Under
the hypothesis that benthic invertebrate assemblages would be specialized according to river type, a set of 627 faunal samples
originating from an 11-year sampling period was tested to characterize river types with faunal assemblages.
A multivariate approach led to gather 23 river types into seven groups exhibiting similar faunal assemblages.
Using biocenotic metrics based on benthic invertebrate assemblages (e.g., the French standard IBGN), type-specific reference
conditions and ecological status class limits were defined for each ‘natural’ river type group. Ecological potential was defined
for heavily modified and for artificial (i.e., man-made canals) types.
An ‘ecological status’ evaluation strategy was therefore developed and applied in the southern – and more natural – part of
Wallonia, where many reference sites were available. In the northern part of Wallonia (i.e., the ‘Loess region’) where no
high quality site was available, the expert judgement took a larger part in the definition of the reference conditions and
of the ecological status class limits, in addition to the calculations. Two independent distribution gradients of taxa assemblages
resulted from multivariate ordination: a first ‘saprobity axis’, as the taxa-sensitivity to organic contamination was increasing
from ‘very resistant’ taxa (mainly located in the ‘Loess region’) to ‘sensitive’ and ‘very sensitive’ taxa (from the river
types belonging to the Condroz, the Famenne, the Arden and the Jurassic regions) and a second axis characterizing the Meuse-specific
faunal assemblage, gathering exotic species and typical limnophilous taxa of large heavily modified rivers. The ecological
status monitoring management system developed in this study – i.e., the definition of faunal river type groups, related reference
conditions and ecological status class limits – represents a proposal to be integrated in the ecological status assessment
of biological elements for the implementation of the WFD and was tested in Wallonia. For the period 2000–2002 involving 349
different sites, the element ‘benthic invertebrate fauna’ was in that way classified ‘high status’ for 31.5% of sites, ‘good
status’ for 31.5% and below ‘good status’ for 37% of sites. The best ecological status (i.e., 100% ‘high’ and ‘good’ status)
was found in river type ‘Arden’s xenotrophic brooks with strong slope’ and in river types 8large rivers with medium slope’.
The worst status was found in river types ‘Loess brooks and rivers with medium slope’. 相似文献

The fuel oxygenate, methyl tert-butyl ether (MTBE), although now widely banned or substituted, remains a persistent groundwater contaminant. Multidimensional
compound-specific isotope analysis (CSIA) of carbon and hydrogen is being developed for determining the extent of MTBE loss
due to biodegradation and can also potentially distinguish between different biodegradation pathways. Carbon and hydrogen
isotopic fractionation factors were determined for MTBE degradation in aerobic and anaerobic laboratory cultures. The carbon
isotopic enrichment factor (εC) for aerobic MTBE degradation by a bacterial consortium containing the aerobic MTBE-degrading bacterium, Variovorax paradoxus, was −1.1 ± 0.2‰ and the hydrogen isotope enrichment factor (εH) was −15 ± 2‰. This corresponds to an approximated lambda value (Λ = εH/εC) of 14. Carbon isotope enrichment factors for anaerobic MTBE-degrading enrichment cultures were −7.0 ± 0.2‰ and did not vary
based on the original inoculum source, redox condition of the enrichment, or supplementation with syringic acid as a co-substrate.
The hydrogen enrichment factors of cultures without syringic acid were insignificant, however a strong hydrogen enrichment
factor of −41 ± 3‰ was observed for cultures which were fed syringic acid during MTBE degradation. The Λ = 6 obtained for NYsyr cultures might be diagnostic for the stimulation of anaerobic MTBE degradation by methoxylated compounds
by an as yet unknown pathway and mechanism. The stable-isotope enrichment factors determined in this study will enhance the
use of CSIA for monitoring anaerobic and aerobic MTBE biodegradation in situ. 相似文献

To understand how chemoreceptor organs may extract temporal information from odor plumes, we investigated the frequency filter
properties of lobster chemoreceptor cells. We used rapid stimulation and high-resolution stimulus measurement for accurate
stimulus control and recorded extracellular responses from chemoreceptors in the lobster lateral antennule in situ. We tested
16 hydroxyproline-sensitive cells with a series of ten 100-ms pulses at 10, 100 and 1000 μmol l−1 at stimulation frequencies from 0.5 Hz to 4 Hz. Receptor cell responses could accurately encode 10 μmol l−1, but not 100 or 1000 μmol l−1 pulses, delivered at rates of 4 Hz. Flicker-fusion frequency and synchronization with the stimulus pulse train were concentration
dependent: performance rates above 1 Hz became poorer both with increasing pulse amplitude and frequency. Flicker fusion frequency
was 3 Hz for 100 μmol l−1 pulses and 2 Hz for 1000 μmol l−1 pulses. Individual cells showed differences in their stimulus pulse following capabilities, as measured by the synchronization
coefficient. These individual differences may form a basis for coding temporal features of an odor plume in an across-fiber
pattern.

This paper elucidates nutrient dynamics in a pine forest, previouslyinvestigated for dry matter dynamics. The nutrient concentrationsin different life forms were of the order: herb > shrub >tree whereas the standing state of nutrients were of the order:tree > shrub > herb. Soil, litter and vegetation respectivelyaccounted for 38·182·2, 2·43·7and 15·458·2 per cent of the total nutrientsin the system. Considerable reductions (5269 per cent)in concentrations of nutrient in needles occurred during senescence.The uptake of nutrients by vegetation, and also by differentcomponents with and without adjustment for internal recycling,has been calculated separately. Annual transfer of litter tothe soil by vegetation was 76·21 N, 6·25 P, 57·24Ca, 14·22 Mg, 19·92 K and 1·92 kg ha1Na. Turnover rate and turnover time for different nutrientsranged between 0·400·64 and 1·562·50year, respectively. Studies have evidenced that zinc metabolism is altered in presence of Down syndrome, and zinc seems to have a relationship
with the metabolic alterations usually present in this syndrome. In this work, the Zn-related nutritional status of adolescents
with Down syndrome was evaluated by means of biochemical parameters and diet. A case–control study was performed in a group
of adolescents with Down syndrome (n = 30) and a control group (n = 32), of both sexes, aged 10 to 19 years. Diet evaluation was accomplished by using a 3-day dietary record, and the analysis
was performed by the NutWin program, version 1.5. Antropometric measurements were performed for evaluation of body composition.
The Zn-related nutritional status of the groups was evaluated by means of zinc concentration determinations in plasma and
erythrocytes, and 24-h urinary zinc excretion, by using the method of atomic absorption spectroscopy. The diet of both groups
presented adequate concentrations of lipids, proteins, carbohydrates, and zinc. The mean values found for zinc concentration
in erythrocytes were 49.2 ± 8.5 μg Zn/g Hb for the Down syndrome group and 35.9 ± 6.1 μg Zn/g Hb for the control group (p = 0.001). The average values found for zinc concentration in plasma were 67.6 ± 25.6 μg/dL for the Down syndrome group and
68.9 ± 22.3 μg/dL for the control group. The mean values found for zinc concentration in urine were 244.3 ± 194.9 μg Zn/24 h
for the Down syndrome group and 200.3 ± 236.4 μg Zn/24 h for the control group. Assessment of body composition revealed overweight
(26.7%) and obesity (6.6%) in the Down syndrome group. In this study, patients with Down syndrome presented altered zinc levels
for some cellular compartments, and the average zinc concentrations were low in plasma and urine and elevated in erythrocytes. 相似文献

The inheritance of resistance to Plum pox virus (PPV) has been studied in 1,178 apricot hybrids. Seven hundred and eighteen F1 hybrids, obtained from controlled crosses
between the susceptible Greek cultivar “Bebecou” and the resistant PPV cultivars of American origin (“Stark Early Orange,”
‘NJA2,” ‘Veecot,” “Sunglo,” “Harlayne,” and “Orangered”) were evaluated for resistance to the PPV-M (Marcus) strain, 8 years
after artificial inoculation. The inheritance of resistance to PPV has been additionally studied for the first time in a BC1 population of 95 apricot hybrids for four vegetative periods. Reaction of each hybrid to PPV-M was scored through visual
symptoms, indexing onto GF-305 and double-antibody sandwich enzyme-linked immunosorbent assay tests. Segregation within the
hybrids, determined by Chi-squared analysis, fits a 1:1 ratio (P ≤ 0.05) of the resistant vs susceptible, indicating that resistance to PPV is controlled by a single dominant gene locus
and that the above six resistant cultivars are heterozygous for the trait. Plants carrying this gene may initially develop
disease symptoms on leaves but eventually recover and no virus can be detected in leaves. Susceptible plants show similar
symptoms initially but remain symptomatic. Inheritance of resistance to PPV also has been studied in 365 F1 hybrids by crossing
the resistant cultivar “Stella” with the susceptible “Bebecou” and the resistant cultivars “Sunglo” and “NJA2,” for 8 years
after inoculation. The segregation ratio was 1:0 (resistant/susceptible) suggesting that “Stella” is homozygous for the resistance

Cocconeis scutellum var.ornata Grun. from three localities of Japan was studied. The striation density in 10 μm showed a marked tendency to increase with
the decrease of the valve length in both raphe and rapheless valves, and this tendency did not vary with locality or environmental
condition. The striation densities of rapheless valves were 4–6 in 10 μm for a valve length of 40μm, 4–6.5 for 30 μm, 6–9
for 20μm and 6.5–11 for 15μm. Those of raphe valves were 10–11 in 10μm for a valve length of 40μm, 10–12 for 30μm, 11–14.5
for 20μm and 12.5–17 for 15μm.
According to the range of changing value in striation density obtained by the present study,C. scutellum var.schmidti Frenguelli andC. japonica Schmidt are identical withC. scutellum var.ornata.

Summary Anthers and ovaries of six grapevine cultivars (three Vitis vinifera L., two V × Labruscana L. H. Bailey, and one complex hybrid) were extracted from flower buds over 2 yr and cultured on three media reported to promote
somatic embryogenesis in Vitis tissues. The highest percent embryogenesis from the hybrid ‘Chancellor’ and V. vinifera ‘Chardonnay’, ‘Merlot’, and ‘Pinot Noir’ occurred on medium C [Nitsch and Nitsch, 1969, basal medium with 3.0% (w/v) sucrose,
0.01% (w/v) inositol. 0.3% (w/v) Phytagel, 2.5 μM 2.4-dichlorophenoxyacetic acid, 2.5μM β-naphthoxyacetic acid, 5.0μM N-(2-chloro-4-pyridyl)-N′-phenylurea, and 0.05% (w/v) glutamine]. Regardless of the media, the labrusca cultivars ‘Concord’ and ‘Niagara’ produced
soft non-embryogenic callus that was sometimes mixed with well-developed somatic embryos. Nine vinifera genotypes were further
tested for several different years on medium C. Embryogenic cultures suitable for transformation were obtained from all genotypes
in more than 1 yr. The average percent embryogenesis from ovaries was 7-fold higher than from anthers. There was significant
annual variation in percent embryogenesis, demonstrating the need for media comparisons to be replicated for more than one
season. Suspension cultures suitable for use in genetic transformation were initiated from ‘Chardonnay’, ‘Merlot,’ and ‘Pinot
Noir’ pro-embryogenic masses. ‘Chardonnay’ suspension cultures plated and grown under conditions developed for recovery of
plants after biolistic transformation yielded approximately 500 non-transformed embryos per plate after 4 mo. of culture,
with 68.6% of the embryos converting to plants. This is the first reported protocol for embryogenesis from ‘Concord,’ ‘Cabernet
Franc,’ and ‘Pinot Noir’ grapevines. 相似文献

Summary An efficient and simple plant regeneration system via organogenesis from leaf segments of persimmon (Diospyros kaki Thunb.) cultivars ‘Fuyu’ and ‘Nishimurawase’ has been developed. The regeneration capacity was influenced by the culture
vessels, gelling agents, plant growth regulators, and light conditions. Leaf explants taken from in vitro shoots were cultured on a modified Murashige and Skoog medium (MS1/2N), for 16 wk without transfer to fresh medium. Adventious
shoots appeared after 4 and 8 wk in culture of ‘Nishimurawase’ and ‘Fuyu’ tissues, respectively. The culture of leaf explants
in Erlenmeyer flasks with medium containing 4 g l−1 agar enhanced shoot formation in comparison to media with increased agar concentrations. Optimal shoot regeneration was obtained
with 5 mg l−1 (22.8 μM) zeatin and 0.1 mg l−1 (0.05 μM) indole-3-butyric acid (IBA) for ‘Nishimurawase’, and 10 mg l−1 (45.6 μM) zeatin and 0.1 mg l−1 (0.05 μM) IBA for ‘Fuyn’. Shoot regeneration frequencies in both cultivars were 100%, and shoot numbers per explant reached up to
9.2 for ‘Nishimurawase’ and 2.2 for ‘Fuyu’. Dark incubation during the first 4–5 wk was the most effective condition to successfully
influence shoot regeneration in both cultivars. While dark incubation was essential for adventitious shoot formation by ‘Fuyu’,
it was only slightly beneficial to ‘Nishimurawase’. More than 80% of the regenerated shoots rooted within 4 wk on hormone-free
MS1/2N demium after having been dipped for 30 s in 250 mg l−1 (1.1. mM) IBA solution. 相似文献
