Considering the importance of extrafloral nectaries (EFNs) in Fabaceae, the objectives of this research were to analyze (1) the anatomical and histochemical characteristics of the EFNs of Chamaecrista ramosa var. ramosa, C. ramosa var. curvifoliola, C. ramosa var. parvifoliola, and C. ramosa var. lucida and (2) the ultrastructure of the EFNs of C. ramosa var. ramosa. Standard techniques in plant anatomy and transmission electron microscopy were used. The anatomical analyses confirmed the characteristics described for extrafloral nectaries, evidencing three well-defined regions: epidermis, nectariferous, and subnectariferous parenchymas. Carbohydrates, proteins, pectins/mucilages, and lipids were detected by histochemical analyzes in all varieties. The ultrastructure of the EFNs of C. ramosa var. ramosa allowed the observation of microchannels at the external periclinal cell walls of the epidermis covering the secretory region. The nectariferous and subnectariferous parenchyma cells have periplasmic spaces, large plastids containing starch grains and plastoglobules, mitochondria, developed endoplasmic reticulum, large vacuoles with electron-dense contents, and membrane residues may be associated with the vacuole, suggesting the occurrence of autophagic processes. The anatomical, histochemical, and ultrastructural patterns revealed characteristics that confirm the glands of C. ramosa as extrafloral nectaries and suggest the eccrine mechanism of secretion.  相似文献

In plant/parasitic plant interaction, little is known about the host plant response before the establishment of the parasite within the host. In the present work, we focused on host responses to parasitic plant, O. ramosa in the early stage of infection. We used a co-culture system of A. thaliana suspension cells and O. ramosa germinated-seeds to avoid parasite attachment. We showed that O. ramosa induced H2O2 generation and camalexin synthesis by A. thaliana followed by a drastic increase in cell death. We further demonstrated that a heat sensitive diffusible signal is responsible for this cell death. These data indicate that recognition of O. ramosa occurs before the attachment of the parasite and initiates plant defence responses.Key words: Orobanche ramosa, Arabidopsis thaliana, cell death, hydrogen peroxide, secondary metabolism  相似文献

It is well known that dopaminergic genes affect the development of attention deficit hyperactivity disorder (ADHD) in various populations. Many studies have shown that variable number tandem repeats (VNTRs) located within the 3′-untranslated region of DAT1 and in exon 3 of DRD4 are associated with ADHD development; however, these results were inconsistent. Therefore, we investigated the genetic association between two VNTRs and ADHD in Korean children. We determined the VNTRs using PCR. We examined genotype and allele frequency differences between the experimental and control groups, along with the odds ratios, using Chi square and exact tests. We observed a significant association between the children with ADHD and the control group in the 10R/10R genotype of DAT1 VNTRs (p?=?0.025). In addition, the 11R allele of DAT1 VNTRs showed a higher frequency in the control group than in the ADHD group (p?=?0.023). Also, the short repeat (without 11R) and long repeat alleles (including 11R) were associated with ADHD (p?<?0.05). The analysis of DRD4 VNTRs revealed that the 2R allele is associated with ADHD (p?=?0.025). A significant result was also observed in long and short repeats (p?<?0.05). Additionally, ADHD subtypes showed that the DRD4 VNTRs are associated with combined and hyperactive-impulsive subtype groups (p?<?0.05). Therefore, our results suggest that DAT1 VNTRs and DRD4 VNTRs play a role in the genetic etiology of ADHD in Korean children.  相似文献

Analysis of mucin genes has identified the presence of several features that may represent important functional domains in mucin glycoproteins. In the central region of each mucin, there are a variable number of tandem repeats (VNTR; minisatellites). However, their genomic levels are unclear because of complex genomic properties. We report here the distribution of VNTR and polymorphic analysis ofMUC8. We searched for VNTR ofMUC8 using the Tandem Repeat Finder program and found nine VNTR motif. Six (MUC8 MS1~MS6) among the nine VNTRs were evaluated in this study. Each VNTR inMUC8 region was analyzed in genomic DNA obtained from 200 unrelated individuals and multi-generational families. All VNTRs (MUC8 MS1, -MS2, -MS3, -MS4,-MS5 and -MS6) were genotyped as polymorphic. The degree of polymorphism within theMUC8-MS5 showed the highest heterozygosity (h = 0.786) in theMUC8 region. In order to perform a segregation analysis of the VNTRs inMUC8, we analyzed genomic DNA obtained from two generations of five families and from three generations of two families. Six of the polymorphic VNTRs were transmitted through meiosis following a Mendelian inheritance, which suggests that polymorphic VNTRs could be useful markers for paternity mapping and DNA fingerprinting.  相似文献

Phelipanche ramosa is a major root-holoparasitic damaging weed characterized by a broad host range, including numerous Fabaceae species. In France, the agricultural threat posed by P. ramosa has increased over two decades due to the appearance of a genetically differentiated pathovar presenting a clear host specificity for oilseed rape. The new pathovar has led to a massive expansion of P. ramosa in oilseed rape fields. The germination rate of P. ramosa seeds is currently known to vary among P. ramosa pathovars and host species. However, only a few studies have investigated whether phylogenetic relatedness among potential host species is a predictor of the ability of these species to induce the seed germination of parasitic weeds by testing for phylogenetic signal. We focused on a set of 12 Fabaceae species and we assessed the rate of induction of seed germination by these species for two pathovars based on in vitro co-cultivation experiments. All Fabaceae species tested induced the germination of P. ramosa seeds. The germination rate of P. ramosa seeds varied between Fabaceae species and tribes studied, while pathovars appeared non-influential. Considering oilseed rape as a reference species, we also highlighted a significant phylogenetic signal. Phylogenetically related species therefore showed more similar rates of induction of seed germination than species drawn at random from a phylogenetic tree. In in vitro conditions, only Lotus corniculatus induced a significantly higher germination rate than oilseed rape, and could potentially be used as a catch crop after confirmation of these results under field conditions.  相似文献

The dramatic spread of root holoparasite Orobanche ramosa L. (branched broomrape) has been observed in the last few years in France in fields of Brassica napus L., Nicotiana tabacum L., and Cannabis sativa L. In the face of this occurrence and considering its tremendous capacity to adapt to its host, the question about variability of O. ramosa populations arises. The virulence of broomrape populations was investigated by cross infestations performed under greenhouse conditions using the three host species mentioned above and broomrape seeds collected from Orobanche plants parasitizing them. After 45 days of co-culture, the intensity and kinetics of infestation were determined. In all cases, parasites developed but differences were observed among associations between the host species and the broomrape population. Differences in virulence of O. ramosa populations and greater affinity between a host species and “its own” broomrape population were demonstrated by statistical analyses (F-test, CFAs). These results led to the concept of host specificity and suggestions about the existence of three different pathovars in O. ramosa. Analysis of genetic variability of the three O. ramosa populations by PCR of RAPD markers confirmed that they may be considered as three distinct pathovars.  相似文献

Pasteuria penetrans, an obligate endospore-forming parasite of Meloidogyne spp. (root knot nematodes), has been identified as a promising agent for biocontrol of these destructive agricultural crop pests. Pasteuria ramosa, an obligate parasite of water fleas (Daphnia spp.), has been shown to modulate cladoceran populations in natural ecosystems. Selected sporulation genes and an epitope associated with the spore envelope of these related species were compared. The sigE and spoIIAA/spoIIAB genes differentiate the two species to a greater extent than 16S rRNA and may serve as probes to differentiate the species. Single-nucleotide variations were observed in several conserved genes of five distinct populations of P. ramosa, and while most of these variations are silent single-nucleotide polymorphisms, a few result in conservative amino acid substitutions. A monoclonal antibody directed against an adhesin epitope present on P. penetrans P20 endospores, previously determined to be specific for Pasteuria spp. associated with several phytopathogenic nematodes, also detects an epitope associated with P. ramosa endospores. Immunoblotting provided patterns that differentiate P. ramosa from other Pasteuria spp. This monoclonal antibody thus provides a probe with which to detect and discriminate endospores of different Pasteuria spp. The presence of a shared adhesin epitope in two species with such ecologically distant hosts suggests that there is an ancient and ecologically significant recognition process in these endospore-forming bacilli that contributes to the virulence of both species in their respective hosts.  相似文献

The abundance and structure of the foraminifera community have been investigated. The role of agglutinating and secreting species in bottom sediments of the deep trenches of the Kara Sea has been studied. It has been found that the abundance and dominance of the agglutinated foraminifera Saccorhiza ramosa depend on the depth of the sea and are related to the genetic type of origin of such sediments. A correlation has been revealed between the distribution of S. ramosa and the concentrations of natural radionuclides (226Ra, 232Th, and 210Pb) in bottom sediments. The degree of dominance of S. ramosa in the community of benthic foraminifera increases along with natural radioactivity.  相似文献

The specificity of cholinesterase (ChE) activity to detect the presence of anatoxin-a(s) and sublethal effects of a 7-day exposure to Anabaena spiroides extract containing anatoxin-a(s) were assessed in two freshwater cladoceran species. Activities of ChE of both Pseudosida ramosa and Daphnia magna can be used to indicate the presence of the neurotoxin anatoxin-a(s), but not for the hepatotoxic microcystin. Activity of ChE of P. ramosa, however, performed better as a biomarker of exposure to A. spiroides than that of D. magna. Furthermore, sublethal exposure to A. spiroides extract significantly inhibited the ChE activity in P. ramosa and negatively affected both individual and population endpoints. For D. magna, the inhibition of ChE activity was not related to effects at higher levels of biological organization, since no direct effect was recorded on the individual and population endpoints. The activity of ChE in P. ramosa also proved to be a good predictor of chronic effects of the A. spiroides extract at higher levels of biological organization, since 48-h ChE inhibition was linked to the sublethal effects on the individual and population. These relationships could not be established for D. magna. Since relationships between the effects of A. spiroides extract at different levels of biological organization were species-specific, it can be concluded that the choice of test organism interferes with the accuracy of the environment risk assessment of this neurotoxin and, hence, the use of native species is recommended for its assessment.  相似文献

Phelipanche and Orobanche spp. (broomrapes) are economically important parasitic weeds, causing severe damage to many agricultural crops. However, conventional methods to control these parasitic weeds are often not effective. Targeting molecular and biochemical processes involved in the establishment of the connection between the parasite and the host may offer a new perspective for control. However, progress in the understanding of these processes is hampered by the fact that genetic transformation and regeneration of these parasites is difficult if not impossible due to their specific lifecycle. Phelipanche and Orobanche spp. are holoparasites that need to attach to the roots of a host plant to get their assimilates, nutrients and water to develop and reproduce. The present study describes a highly efficient genetic transformation and regeneration protocol for the root holoparasitic Phelipanche ramosa. We present a new transformation system for P. ramosa using Agrobacterium rhizogenes MSU440 carrying a non-destructive selection marker gene coding for a red fluorescent protein (DsRed1). Using this protocol up to 90% transformation efficiency was obtained. We transformed 4 weeks old P. ramosa calli and transgenic calli expressing DsRed1 were then cultured on host plants. For the first time, we present shoot and flower development of the transgenic parasitic plant P. ramosa after successful connection of transgenic calli with the host plant roots. Moreover, we also present, for the first time, growth and development of P. ramosa shoots and flowers in vitro in the absence of a host plant.  相似文献

The mitochondrial control regions (CRs) and flanking sequences of Pelodiscus sinensis, Apalone ferox, Palea steindachneri and Carettochelys insculpta were obtained using Long-PCR with gene-specific primers. The CR lengths of the four species were 1843 bp, 1356 bp, 1725 bp, and 969 bp. The base composition percentages of A+T were 60.5%, 60.7%, 65.7%, 64.7%, respectively. Combined with CR sequences of other three soft-shelled turtles published in GenBank (Pelodiscus sinensis, Korea, AY962573; Dogania subplana, AF366350; Lissemys punctata, EF050073), we compared the CR structures and identified three functional domains (TAS, CD and CSB) in which conserved sequence blocks (TAS, CSB -F, CSB-1, CSB-2 and CSB-3) were also successfully identified according to their sequence similarities to those of other turtles. The variable numbers of tandem repeats (VNTRs 1) with 50–52 bp motif were identified at 5′-end of CR among the five soft-shelled turtles P. sinensis (China), P. sinensis (Korea), A. ferox, P. steindachneri, D. subplana. The copy number of the VNTRs varied from 5 to 15. VNTRs 2 with 2–11 bp motif were identified in the 3′- end of CR among all of the six soft-shelled turtles with variable number of motifs from 4 to 29. Moreover, VNTRs 3 with 6 bp motif were identified between CSB-1 and CSB-2 of CR both in P. sinensis (China) and P. sinensis (Korea), in which the number of motifs varied from 19 to 29. The types and distribution of VNTRs of the six soft-shelled turtles were also discussed. With Alligator mississippiensis as an outgroup, combined with the CR sequences (excluding VNTRs) of other five turtles which were published in GenBank, the molecular phylogenetic trees were constructed using PAUP 4.0b10 and MrBayes ver. 3.0. The results strongly supported the monophyly of Carretochelyidae and Carettochelyidae as sister group to an assemblage of Cryptodira. Our research suggested that the earliest phylogenetic tree splits into three separated basal branches; the Pelomedusidira (Pelomedusa subrufa), the Carettochelyidae (C. insculpta), and an assemblage of Cryptodira and the C. insculpta that might be a representation of distinctive suborder.  相似文献

Aechmea ramosa Mart. ex Schult. f. is an endemic bromeliad of the Brazilian Atlantic Forest. The current habitat degradation of this hotspot biome threatens this species, which besides having an important ecological role, is also of invaluable ornamental interest. Plant tissue culture has been used in mass propagation and conservation of various bromeliads. We have established a micropropagation protocol for A. ramosa var. ramosa using leaf explants grown in MS medium supplemented with 2 μM of 1-naphthaleneacetic acid (NAA) and 2 μM of 6-benzylaminopurine (BAP) that showed higher values of shoot induction. NAA and BAP are associated with the production of proteins involved in stress response modulation, metabolic activity, and cell division, the latter being involved in inducing the differentiation of competent cells. After 120 d of culture, each explant presented 28.9 shoots with an average size of 27.8 mm, with no variation in either Stomatal Index or density of the regenerated shoots. Plantlets measuring above 15-mm height were successfully acclimatized, presenting 100% survival rate. Thus, this protocol can be used for mass propagation of A. ramosa, and to supply demand for the market of ornamental plants. Furthermore, it represents an important tool for the conservation of this species and maintenance of an in vitro germplasm.  相似文献

In France, the fact that the holoparasitic plant Phelipanche ramosa has adapted to oilseed rape over the past two decades is deeply worrying, as it can cause over 80% yield losses. Its distribution area and the range of its host plants, whether they are cultivated plants or weeds, are expanding dramatically. As no natural regression of the infestation has been recorded so far, we undertook a detailed study of the different steps of the biological cycle of the P. ramosa/oilseed rape pathosystem to determine the adaptive traits that favour the parasite's success. We combined experimental approaches involving controlled conditions with in vitro and in pot co-cultivation on the one hand and a field trial on the other hand. These experiments allowed us to determine a 4-mm zone around host roots within which oilseed rape root exudates stimulate the germination of P. ramosa, the early action of host root exudates and the rapid fixation of the parasite on host roots. We also unveiled that P. ramosa was able to tune its biological cycle to that of oilseed rape. The important trophic relationships between the parasite and its host induced phenotypic (dwarfism, leaf chlorosis, silique abortion) as well as agronomical (90% yield losses) consequences on oilseed rape. Our results can constitute a relevant basis for further experimental studies. The research perspectives they open will focus on key-processes of the host–parasite relationship, and more particularly on the trophic relationships that are set up as far as carbon assimilates and minerals such as nitrogen are concerned.  相似文献

Due to the amount of nutrients available in the agroindustrial wastes, these can be converted into high added-value products by the action of microorganisms in solid-state bioprocesses. The aim of this work was to evaluate the growth physiology and lipase production of the fungus Lichtheimia ramosa using the following Brazilian savannah fruit wastes as substrates: bocaiuva (Acrocomia aculeata), pequi (Caryocar brasiliense), guavira (Campomanesia pubescens), araticum (Annona crassiflora) and seriguela (Spondias purpurea). These residues were triturated, homogenized, adjusted to pH 5.0 and 60 % moisture, sterilized and packaged in plastic tray-type bioreactors before inoculation with 10 % (w/v) of L. ramosa pre-culture medium. The cultivations were conducted in a bacteriological incubator at 30 °C for 40 days. Samples were taken every 5 days and fungi and bacteria contents, proximate composition and lipase activity were evaluated. The maximum fungal counting was observed between 25 and 35 days. L. ramosa reached the stationary phase next to 40 days in all substrates. Mesophilic and psicrophilic aerobic bacteria were not detected. Protein enrichment was obtained for all media, being superior in seriguela residues (391.66 %), followed by pequi (160.04 %), araticum (143.31 %), guavira (102.42 %), and bocaiuva (67.88 %). Lipase production was observed in all cultivated media, except in pequi residues that showed decreasing lipase activity. The higher production was observed in guavira (1.12 U/g) followed by araticum (0.58 U/g), seriguela (0.41 U/g) and bocaiuva (0.21 U/g) waste substrates. It was concluded that the studied fruit wastes have been successfully utilized as substrates for protein enrichment and lipase production with L. ramosa.  相似文献

During the last 100 years, the neuston bacterium Nevskia ramosa has been described several times. This bacterium forms conspicuous rosette-like microcolonies at the air-water interface. In this study, pure cultures of Nevskia ramosa were obtained for the first time, from a bog lake (strain Soe1, DSMZ 11499T) and a freshwater ditch (strain OL1, DSMZ 11500). The isolates showed special adaptations to life in the epineuston. They formed hydrophobic surface films with a dull appearance. N. ramosa is sensitive to UV radiation but revealed a very effective photorepair mechanism. Exposure to light at a wavelength of 350 nm after UV treatment raised the number of surviving cells by several orders of magnitude. The isolates grew with a broad range of organic substrates. Surface films were formed only in the absence of combined nitrogen; however, nitrogenase activity was not detected. It appears that during growth at the air-water interface the cells benefit from trapping ammonia from the air. The G+C content of the DNA was 67.8 and 69.0 mol% for strains Soe1 and OL1, respectively. The slight difference was confirmed by enterobacterial repetitive intergenic consensus PCR. The 16S rRNA sequences revealed 99.2% similarity. Thus, both isolates belong to the same species. The phylogenetic analysis indicated that Nevskia is a member of the gamma-subclass Proteobacteria that has no known close relatives.Some morphologically conspicuous bacteria were observed in the 19th century but still have not been isolated in pure culture. In 1892, Famintzin (7) described Nevskia ramosa from the water surface of an aquarium in the botanical garden of St. Petersburg, Russia. The typical microcolonies consist of flat rosettes with a bush-like appearance on the water surface. The rosettes are colonies of dichotomously branched slime stalks with rod-shaped, slightly bent cells in the tips. The cells contain refractile globules, which were presumed to be ethereal oil (7), sulfur globules (12), or fat droplets (3). The slime stalks consist of polysaccharides (3) and sometimes appear to contain iron and aluminum encrustations (11).Enrichments of Nevskia-like cells in lake water supplied with lactate were described by Babenzien (14). He observed the following life cycle of N. ramosa. Young motile cells develop submersed, then adsorb to the water surface, lose the polar flagellum, and form a hyaline slime stalk on the concave side of the cell. When a cell multiplies by binary fission, branching of the stalk occurs. The resulting flat rosette can reach a size of 70 μm in diameter.Since pure cultures have not been available, little is known about the physiology, phylogeny, and ecology of Nevskia. It was assumed that Nevskia is oligocarbophilic (14). Tests with the nitrification inhibitor nitrapyrin gave no indications that the cells oxidize ammonia (16). N. ramosa was assumed to be related to the stalk-forming genera Caulobacter and Gallionella or to the sulfur-oxidizing Thiobacterium. In Bergey’s manual (4) N. ramosa was affiliated with the budding and/or appendaged bacteria.In addition to its conspicuous morphology, the typical habitat of N. ramosa prompted us to initiate the present investigation. The water-air interface is a very special environment, characterized by high surface tension and a relatively high hydrophobicity. Organic compounds and various typical bacteria are enriched in this zone. The living community is called the neuston (18, 21). Depending on whether they adsorb to the underside or the top of the water surface, organisms belong to the hyponeuston or epineuston, respectively. This habitat requires special adaptations with respect to adsorption, substrate uptake, and UV tolerance.In our study we have isolated N. ramosa in pure culture and carried out ecophysiological and phylogenetic characterizations. We found several adaptions to life in the epineuston in this interesting bacterium.  相似文献

Knowledge of the genetic architecture of pathogen infectivity and host resistance is essential for a mechanistic understanding of coevolutionary processes, yet the genetic basis of these interacting traits remains unknown for most host–pathogen systems. We used a comparative genomic approach to explore the genetic basis of infectivity in Pasteuria ramosa, a Gram-positive bacterial pathogen of planktonic crustaceans that has been established as a model for studies of Red Queen host–pathogen coevolution. We sequenced the genomes of a geographically, phenotypically, and genetically diverse collection of P. ramosa strains and performed a genome-wide association study to identify genetic correlates of infection phenotype. We found multiple polymorphisms within a single gene, Pcl7, that correlate perfectly with one common and widespread infection phenotype. We then confirmed this perfect association via Sanger sequencing in a large and diverse sample set of P. ramosa clones. Pcl7 codes for a collagen-like protein, a class of adhesion proteins known or suspected to be involved in the infection mechanisms of a number of important bacterial pathogens. Consistent with expectations under Red Queen coevolution, sequence variation of Pcl7 shows evidence of balancing selection, including extraordinarily high diversity and absence of geographic structure. Based on structural homology with a collagen-like protein of Bacillus anthracis, we propose a hypothesis for the structure of Pcl7 and the physical location of the phenotype-associated polymorphisms. Our results offer strong evidence for a gene governing infectivity and provide a molecular basis for further study of Red Queen dynamics in this model host–pathogen system.  相似文献

Setaria italica (L.) P. Beauv. (foxtail millet) was originally domesticated in northern China. The time and route of its introduction into South Asia is currently unclear due to the possible confusion with autochthonous Brachiaria ramosa (L.) Stapf. (browntop millet). Geometric morphometrics (GM) offer an alternative to traditional archaeobotanical methods to distinguish between these two small millet species. This study aims at finding a method to securely distinguish among charred caryopses of S. italica and B. ramosa, testing its validity on archaeobotanical assemblages and proposing a new approach for studying the dispersion of S. italica throughout Eurasia. Modern S. italica (n = 35) and B. ramosa (n = 34) caryopses and 15 archaeological specimens from a 5th millennium bp archaeological occupation site in northwestern India were analysed. Archaeological and modern caryopses (before and after charring) were photographed with a Leica EZ4D stereoscope, and TPSdig software was used to scale the photographs and manually apply a configuration of three landmarks and six semi-landmarks onto the contours of the embryos. Multivariate statistics were carried out to analyse the shape differences between modern S. italica and B. ramosa and to classify the archaeological specimens. The results show that the shape of the embryo of both species can be clearly distinguished using a GM-approach, both before and after charring. However, charring tends to smooth the shape differences between the two groups, which may affect the interpretation of archaeobotanical assemblages. The comparison between modern and archaeological caryopses suggests that S. italica was not present in northwestern India during the 5th millennium bp.  相似文献
