Adenovirus is recognized as the most UV-resistant waterborne pathogen of concern to public health microbiologists. The U.S. EPA has stipulated that a UV fluence (dose) of 186 mJ cm−2 is required for 4-log inactivation credit in water treatment. However, all adenovirus inactivation data to date published in the peer-reviewed literature have been based on UV disinfection experiments using UV irradiation at 253.7 nm produced from a conventional low-pressure UV source. The work reported here presents inactivation data for adenovirus based on polychromatic UV sources and details the significant enhancement in inactivation achieved using these polychromatic sources. When full-spectrum, medium-pressure UV lamps were used, 4-log inactivation of adenovirus type 40 is achieved at a UV fluence of less than 60 mJ cm−2 and a surface discharge pulsed UV source required a UV fluence of less than 40 mJ cm−2. The action spectrum for adenovirus type 2 was also developed and partially explains the improved inactivation based on enhancements at wavelengths below 230 nm. Implications for water treatment, public health, and the future of UV regulations for virus disinfection are discussed.  相似文献

Dinitrophenyl phosphotidylethanolamine-containing bilayers have been immobilized on carbon-shadowed support films by UV irradiation of the first monolayer transferred to the support film. The immobilized bilayer is capable of allowing bound protein (anti-DNP antibody) to organize into 2-D arrays in the presence of organic solvents such as acetonitrile and dilute concentrations of detergents such as beta-octyl glucoside. The ability of the bilayers to remain attached to supports under various conditions that include organic solvents and detergents as well as divalent ions is of potential interest in the study of protein crystallization and particularly in the study of membrane proteins.  相似文献

There is indicative epidemiological evidence that exposures of children younger than about 10 years are linked with an increased risk of the development of malignant melanoma as well as non-melanocytic skin cancers later in life. However, an important area of uncertainty relates to lack of knowledge of the sun-sensitivity of children's skin both absolutely and relative to that of adult's skin. For example the thickness of children's skin is very similar to that of adults but due to the nature of the anatomical structure of children's skin, there are indications of children's skin being adversely exposed on the top of the papilla before a significant exposure manifests itself as visible damage to the skin (for example erythema). This might also affect the induction of heavily UV-damaged cells persisting in the basal layer of the epidermis after UV-exposure which are supposed to be keratinocytic epidermal stem cells and may characterize an initiation step of non-melanoncytic skin cancer. For malignant melanoma the number of nevi received in dependence of UV-exposure in childhood is a clear risk factor. Recent data show that the bulge region of hair follicles hosting melanocytic stem cells are located deeper (more protected) in the skin in adults (terminal hair) as compared to pre-pubertal children (vellus hair). This may be an explanation for enhanced risk of malignant melanoma due to UV-exposure in pre-pubertal childhood.  相似文献

Studies were conducted on three herbaceous plant species growing in naturally high solar UV environments in the subalpine of Mauna Kea, Hawaii, USA, to determine if diurnal changes in epidermal UV transmittance (TUV) occur in these species, and to test whether manipulation of the solar radiation regime could alter these diurnal patterns. Additional field studies were conducted at Logan, Utah, USA, to determine if solar UV was causing diurnal TUV changes and to evaluate the relationship between diurnal changes in TUV and UV-absorbing pigments. Under clear skies, TUV, as measured with a UV-A-pulse amplitude modulation fluorometer for leaves of Verbascum thapsus and Oenothera stricta growing in native soils and Vicia faba growing in pots, was highest at predawn and sunset and lowest at midday. These patterns in TUV closely tracked diurnal changes in solar radiation and were the result of correlated changes in fluorescence induced by UV-A and blue radiation but not photochemical efficiency (Fv/Fm) or initial fluorescence yield (Fo). The magnitude of the midday reduction in TUV was greater for young leaves than for older leaves of Verbascum . Imposition of artificial shade eliminated the diurnal changes in TUV in Verbascum , but reduction in solar UV had no effect on diurnal TUV changes in Vicia . In Vicia , the diurnal changes in TUV occurred without detectable changes in the concentration of whole-leaf UV-absorbing compounds. Results suggest that plants actively control diurnal changes in UV shielding, and these changes occur in response to signals other than solar UV; however, the underlying mechanisms responsible for rapid changes in TUV remain unclear.  相似文献

Summary The UV protection and mutation properties of the I group plasmid TP110 have been investigated. It is demonstrated that the genes responsible for these effects are able to complement the deficiency in umuC36 mutants of E. coli, as are the similar genes carried by the B group plasmid R16. Mu-lac inserts into TP110 have been isolated which abolish the UV protection and mutation functions. Restriction mapping of these inserts locates them within a single region of the genome. A comparison of the restriction sites of this region with the muc region of pKM101 reveals very little similarity. Expression of -galactosidase in those Mu-lac inserts in which the lacZ gene is fused to the promoter for the protection and mutation functions is inducible by DNA damaging agents, and induction in mutant strains suggests that these genes are under the direct control of the lexA repressor.  相似文献

Photooxidative processes play a role in the pathobiochemistry of various disorders of light-exposed tissue. After irradiation of skin with UV light, erythema (sunburn) is an initial effect suitable for monitoring the direct biological response. Carotenoids are efficient in photoprotection, scavenging singlet oxygen and peroxyl radicals. Intervention studies with supplements or a carotenoid-rich diet documented efficiency in systemic photoprotection, measuring a decreased sensitivity against UV-induced erythema. For successful intervention, treatment with carotenoids is needed for a period of at least ten weeks. An increased consumption of carotenoids may contribute to life-long protection against UV-induced damage.  相似文献

Skin cancer incidence in Croatia is steadily increasing in spite of public and governmental permanently measurements. It is clear that will soon become a major public health problem. The primary cause of skin cancer is believed to be a long exposure to solar ultraviolet (UV) radiation. The future designers of UV protective materials should be able to block totally the ultraviolet radiation. The aim of this paper is to present results of measurements concerning UV protecting ability of garments and sun-screening textiles using transmission spectrophotometer Cary 50 Solarscreen (Varian) according to AS/NZS 4399:1996; to show that standard clothing materials are not always adequate to prevent effect of UV radiation to the human skin; and to suggest the possibilities for its improvement for this purpose.  相似文献

Shoaling behaviour in fish is influenced by numerous factors, such as familiarity, kinship, group size and shoal composition. Grouping decisions are based on both olfactory and visual cues. The visual system of many vertebrates is extended into the ultraviolet (UV) wave range as in three-spined sticklebacks (Gasterosteus aculeatus, L.). We investigated whether the presence or absence of UV wavelengths has an influence on shoaling behaviour in this species. Reproductively non-active three-spined sticklebacks were given the choice between two shoals, equal in numbers of individuals, which could be seen either through a UV-transmitting [UV(+)] or a UV-blocking [UV(-)] filter. Test fish preferred to join the shoal seen under UV(+) conditions. Due to differences in quantal flux between the UV(+) and UV(-) filters used, control experiments with neutral-density optical filters were performed in order to clarify the role of luminance. Here, test fish spent significantly more time near shoals that were seen in a darker environment, suggesting a potential trade-off between UV radiation and lower brightness during shoal choice.To our knowledge, these results demonstrate for the first time that shoaling decisions are influenced by UV wavelengths.  相似文献

It has been recently reported that the pR plasmid enhances the UV survival in E.coli c600. In order to test whether this function may be expressed also in mammalian cells, LTA (tk- aprt-) mouse cells were cotransformed with pR plasmid DNA and ptk1 plasmid as selectable marker. Tk+ transformants were analyzed for their UV survival and for the presence of pR DNA sequences by blot-hybridization. The results show a correlation between the enhanced UV survival and presence of pR DNA sequences in cotransformed LTA mouse cells.  相似文献

Exposure of eukaryotic cells to ultraviolet light results in a temporary inhibition of DNA replication as well as a temporary blockage of DNA fork progression. Recently there has been considerable debate as to whether the (5-6)cyclobutane pyrimidine dimer, the pyrimidine(6-4)pyrimidone lesion or both are responsible for these effects. Using cell lines that repair both of these lesions (CHO AA8), only (6-4) lesions (CHO UV61) or neither (CHO UV5), we have shown that in rodent cells both lesions appear to play a role in both the inhibition of thymidine incorporation and the blockage of DNA fork progression. Specifically, after exposure to 2.5 J/m2, AA8 cells recover normal rates of DNA replication within 5 h after exposure, while UV5 cells exhibit a greater depression in thymidine incorporation for at least 10 h. UV61 cells, on the other hand, show an intermediate response, both with respect to the extent of the initial depression and the rate of recovery of thymidine incorporation. UV61 cells also exhibit an intermediate response with respect to blockage of DNA fork progression. In previous publications we have shown that UV5 cells exhibit extensive blockage of DNA fork progression and only limited recovery of this effect within the first 5 h after exposure to UV. In this report we show that UV61 cells exhibit a more extensive blockage of fork progression than is observed in AA8 cells. These blocks also appear to be removed (or overcome) more slowly than in the AA8 cells, but more rapidly than in UV5 cells. Taken together we conclude that both lesions appear to be involved in the initial depression in thymidine incorporation and the initial blockage of DNA fork progression in rodent cells. These data also indicate that (6-4) lesions may be responsible for the prolonged depression in thymidine incorporation and the prolonged blockage of DNA fork progression observed in UV5 cells.  相似文献

A technique is described which makes it possible to measure the absorption spectrum of phenylalanine powder. Reflectance spectra are used to calculate the amount of light scattered from the samples; thus, the treatment of the data makes it unnecessary to normalize the absorbance to some value known from other work. Further, the unique scattering in the spectral region of an absorption band can be found. The fluorescence quantum yield of phenylalanine powder at 300K is calculated to be 1.0 ± 0.1, a sizeable increase over the value of 0.04 found for aqueous phenylalanine solutions.  相似文献
